Abstract:In this paper, we proposed a novel non-invasive electronic nose for detection and diagnosis of lung cancer based on an electronic nose instrument which includes a gas extraction and capillary column to concentrate, desorb and separate volatile organic compounds (VOCs) in patients' breath, respectively. Surface Acoustic Wave (SAW) gas sensors was used to detect chemical compounds. The specific VOCs exhaled by lung cancer cells in the microenvironment are proven the source biomarkers of lung cancer. The clinical experimental results show that this kind of novel electronic nose is effective for recognition of lung cancer patents and healthy persons and will be also possible non-invasive method to diagnose of lung cancer at early stage.

Abstract:A new method for lung cancer pathology analysis and early diagnosis in cell level was presented to find the special volatile organic compounds (VOCs) in the metabolic product of lung cancer cells. A gas detection system using solid phase micro-extraction combined with gas chromatography (SPME-GC) was designed to detect the VOCs existing in the culture medium of several target cells. Lung tissues and their side tissues from 13 lung cancer patients were cultured and observed by electron microscopy, besides, different kinds of cells including bronchial epithelial cells, taste bud cells, osteogenic cells and lipocytes from rats were cultured as control group. The results show that there are special VOCs in the culture medium of lung cancer cells, which are the metabolic product of lung cancer cells. The special VOCs can be viewed as biomarkers of lung cancer, so provide basis for electronic nose for non-invasive detection and diagnosis of lung cancer.

Abstract:It has been reported that detection of exhaled breath condensate (EBC) is available for studies of pulmonary diseases, especially lung disease. In order to detect lung cancer (LC) at early stage, a point-of-care testing system suitable for measurement of tumor markers in EBC is developed. The assay, based on gold nanoparticle sandwich immunoassay and subsequent gold staining, was performed on a Love-wave sensor packaged inside a chip cartridge. Benefit from high sensitivity of Love-wave sensor, oriented immobilization of coating antibodies and immunogold staining enhancement, the present immunosensor could provide a sensitive, specific and rapid measurement. Carcinoembryonic antigen (CEA), neuron specific enolase (NSE) and squamous cell carcinoma antigen (SCC) in EBC collected from 17 patients with LC and 13 healthy volunteers were detected by this system. Results were compared with commercial chemiluminescence immunoassay and showed high correlation between two methods. Additionally, it revealed significantly statistical differences existing between two groups of subjects. These results indicate that the present system is suitable for detection of tumor markers in EBC and could be used as assistant tools for early detection of LC.

Abstract:Abstract Lung cancer has a high mortality rate largely due to late-stage diagnosis. Detecting protein and genetic biomarkers through electrochemical immunosensors enables non-invasive early diagnosis. This review discusses recent advances in electrochemical immunosensors for detecting clinically relevant lung cancer biomarkers. The use of nanomaterials like graphene, carbon nanotubes, metal nanoparticles, and conducting polymers in sensor fabrication improves electron transfer kinetics, enhances signal transduction, and allows higher antibody loading. Smart surface immobilization strategies optimize antibody orientation and binding capacity. Amplification approaches utilizing nanomaterials, enzymes, polymers, dendrimers, and DNA nanostructures are applied to enhance output signal per binding event. Various electroanalytical techniques including amperometry, potentiometry, impedance spectroscopy, and voltammetry are employed for quantitative monitoring. Recent immunosensors showcase low detection limits and wide linear ranges for measurement of major biomarkers like carcinoembryonic antigen, neuron specific enolase, and cytokeratin fragment 21-1. Emerging biomarkers such as microRNAs and circulating tumor cells have also been targeted. However, reproducibility, selectivity, multiplexing, and integration with point-of-care platforms need improvement for widespread clinical translation. Overall, electrochemical immunosensors hold immense potential for sensitive, affordable lung cancer diagnosis if ongoing efforts can address current limitations.

Abstract:The potential for non-invasive lung cancer (LC) diagnosis based on molecular, cellular and volatile biomarkers has been attracting increasing attention, with the development of advanced techniques and methodologies. It is standard practice to tailor the treatments of LC for certain specific genetic alterations, including the epidermal growth factor receptor, anaplastic lymphoma kinase and BRAF genes. Despite these advances, little is known about the internal mechanisms of different types of biomarkers and the involvement of their related biochemical pathways during the development of LC. The development of faster and more effective techniques is essential for the identification of different biomarkers. The present review summarizes some of the latest methods used for detecting molecular, cellular and volatile biomarkers in LC and their potential use in clinical diagnosis and targeted therapy.

Abstract:Lung cancer is the most commonly diagnosed of all cancers and one of the leading causes of cancer deaths among men and women worldwide, causing 1.5 million deaths every year. Despite developments in cancer treatment technologies and new pharmaceutical products, high mortality and morbidity remain major challenges for researchers. More than 75% of lung cancer patients are diagnosed in advanced stages, leading to poor prognosis. Lung cancer is a multistep process associated with genetic and epigenetic abnormalities. Rapid, accurate, precise, and reliable detection of lung cancer biomarkers in biological fluids is essential for risk assessment for a given individual and mortality reduction. Traditional diagnostic tools are not sensitive enough to detect and diagnose lung cancer in the early stages. Therefore, the development of novel bioanalytical methods for early-stage screening and diagnosis is extremely important. Recently, biosensors have gained tremendous attention as an alternative to conventional methods because of their robustness, high sensitivity, inexpensiveness, and easy handling and deployment in point-of-care testing. This review provides an overview of the conventional methods currently used for lung cancer screening, classification, diagnosis, and prognosis, providing updates on research and developments in biosensor technology for the detection of lung cancer biomarkers in biological samples. Finally, it comments on recent advances and potential future challenges in the field of biosensors in the context of lung cancer diagnosis and point-of-care applications.

Abstract:Electrochemical biosensors have emerged as powerful tools for the ultrasensitive detection of lung cancer biomarkers like carcinoembryonic antigen (CEA), neuron-specific enolase (NSE), and alpha fetoprotein (AFP). This review comprehensively discusses the progress and potential of nanocomposite-based electrochemical biosensors for early lung cancer diagnosis and prognosis. By integrating nanomaterials like graphene, metal nanoparticles, and conducting polymers, these sensors have achieved clinically relevant detection limits in the fg/mL to pg/mL range. We highlight the key role of nanomaterial functionalization in enhancing sensitivity, specificity, and antifouling properties. This review also examines challenges related to reproducibility and clinical translation, emphasizing the need for standardization of fabrication protocols and robust validation studies. With the rapid growth in understanding lung cancer biomarkers and innovations in sensor design, nanocomposite electrochemical biosensors hold immense potential for point-of-care lung cancer screening and personalized therapy guidance. Realizing this goal will require strategic collaboration among material scientists, engineers, and clinicians to address technical and practical hurdles. Overall, this work provides valuable insight for developing next-generation smart diagnostic devices to combat the high mortality of lung cancer.

Abstract:Lung cancer remains a major global health problem with high mortality rates. Early diagnosis can significantly improve prognosis. Hence alternative diagnostic platforms facilitating rapid, noninvasive, point-of-care screening are needed. Nanomaterial-assisted electrochemical biosensors present a promising technology. Unique properties of nanomaterials like high electrical conductivity, catalytic activity, and large surface area enhance achieved detection limits and kinetics. Recent works have combined nanomaterials like graphene, metal nanoparticles and conducting polymers with recognition elements like antibodies and aptamers. Considerable progress has focused on improving analytical parameters including sensitivity, linear ranges, reproducibility and response times. Initial embodiments also demonstrate multiplexed detection, microfluidic integration and smartphone connectivity towards viable clinical practice. However considerable efforts on large sample size validation, standardization of testing protocols and viable commercialization pathways integrating regulatory guidelines and reimbursement ecosystem partnerships remain before such lab-on-chip platforms can enter mainstream and guide therapeutic interventions through affordable decentralized personalized monitoring.

Abstract:Lung cancer remains a major driver of global morbidity and mortality, and diagnosing lung tumors early in their development is vital to maximizing treatment efficacy and patient survival. Several biomarkers, including CYFRA 21-1, NSE, ProGRP, CEA, and miRNA, have been identified as reliable indicators for early lung cancer detection and monitoring treatment progress. However, the minute changes in the levels of these biomarkers during the early stages of disease necessitate advanced detection platforms. In this space, electrochemical biosensors have currently emerged as robust tools for early lung cancer screening and diagnosis owing to their low costs, rapid responses, and superior sensitivity and selectivity. This review provides an up-to-date overview of the application of electrochemiluminescence, photoelectrochemical, and other electrochemical analytical strategies for detecting lung cancer-associated protein biomarkers, and miRNA. This review compares these techniques to provide a concise overview of the principles underlying these electrochemical analytical methods, the preparation of their components, and the performance of the resulting biosensors. Lastly, a discussion of the challenges and opportunities associated with electrochemical biosensors detection of lung cancer-associated biomarkers are provided.

Abstract:Lung cancer remains a global health concern, demanding the development of noninvasive, prompt, selective, and point-of-care diagnostic tools. Correspondingly, breath analysis using nanobiosensors has emerged as a promising noninvasive nose-on-chip technique for the early detection of lung cancer through monitoring diversified biomarkers such as volatile organic compounds/gases in exhaled breath. This comprehensive review summarizes the state-of-the-art breath-based lung cancer diagnosis employing chemiresistive-module nanobiosensors supported by theoretical findings. It unveils the fundamental mechanisms and biological basis of breath biomarker generation associated with lung cancer, technological advancements, and clinical implementation of nanobiosensor-based breath analysis. It explores the merits, challenges, and potential alternate solutions in implementing these nanobiosensors in clinical settings, including standardization, biocompatibility/toxicity analysis, green and sustainable technologies, life-cycle assessment, and scheming regulatory modalities. It highlights nanobiosensors' role in facilitating precise, real-time, and on-site detection of lung cancer through breath analysis, leading to improved patient outcomes, enhanced clinical management, and remote personalized monitoring. Additionally, integrating these biosensors with artificial intelligence, machine learning, Internet-of-things, bioinformatics, and omics technologies is discussed, providing insights into the prospects of intelligent nose-on-chip lung cancer sniffing nanobiosensors. Overall, this review consolidates knowledge on breathomic biosensor-based lung cancer screening, shedding light on its significance and potential applications in advancing state-of-the-art medical diagnostics to reduce the burden on hospitals and save human lives.

Abstract:As one of the major public health problems, cancers seriously threaten the human health. Among them, lung cancer is considered to be one of the most life-threatening malignancies. Therefore, developing early diagnosis technology and timely treatment for lung cancer is urgent. Recent research has witnessed that measuring changes of biomarkers expressed in lung cancer has practical significance. Meanwhile, we note that bioimaging with organic fluorescent probes plays an important role for its high sensitivity, real-time analysis and simplicity of operation. In the past years, kinds of organic fluorescent probes targeting lung cancer related biomarker have been developed. Herein, we summarize the research progress of organic fluorescent probes for the detection of lung cancer related biomarkers in this review, along with their design principle, luminescence mechanism and bioimaging application. Additionally, we put forward some challenges and future prospects from our perspective.
Chemiresistive Sensor Array with Nanostructured Interfaces for Detection of Human Breaths with Simulated Lung Cancer Breath VOCs

Abstract:Timely screening of lung cancer represents a challenging task for early diagnosis and treatment, which calls for reliable, low-cost, and noninvasive detection tools. One type of promising tools for early-stage cancer detection is breath analyzers or sensors that detect breath volatile organic compounds (VOCs) as biomarkers in exhaled breaths. However, a major challenge is the lack of effective integration of the different sensor system components toward the desired portability, sensitivity, selectivity, and durability for many of the current breath sensors. In this report, we demonstrate herein a portable and wireless breath sensor testing system integrated with sensor electronics, breath sampling, data processing, and sensor arrays derived from nanoparticle-structured chemiresistive sensing interfaces for detection of VOCs relevant to lung cancer biomarkers in human breaths. In addition to showing the sensor viability for the targeted application by theoretical simulations of chemiresistive sensor array responses to the simulated VOCs in human breaths, the sensor system was tested experimentally with different combinations of VOCs and human breath samples spiked with lung cancer-specific VOCs. The sensor array exhibits high sensitivity to lung cancer VOC biomarkers and mixtures, with a limit of detection as low as 6 ppb. The results from testing the sensor array system in detecting breath samples with simulated lung cancer VOC constituents have demonstrated an excellent recognition rate in discriminating healthy human breath samples and those with lung cancer VOCs. The recognition statistics were analyzed, showing the potential viability and optimization toward achieving the desired sensitivity, selectivity, and accuracy in the breath screening of lung cancer.

Abstract:Cancer is a malignant disease that endangers human life, especially owing to its high fatality rate; therefore, rapid and accurate early screening is needed to effectively improve the survival rate. Compared with traditional cancer detection methods, electrochemical biosensors that recognize cancer biomarkers in blood have the advantages of low invasiveness, fast diagnosis, and low cost. However, there is always a trade-off between sensitivity and selectivity, which limits the detection of trace amounts of biomarkers produced in the early stages. To address this issue, an increasing number of nanomaterials with simultaneous improvements in both sensitivity and selectivity have recently been reported. In this review, different categories of state-of-the-art electrochemical biosensors and their operating principles are introduced, and their respective advantages and disadvantages are described. Furthermore, the review discusses the existing detection strategies and performance of nanomaterial-based cancer biosensors for biomarker recognition, providing overall guidance for the material selection of different biomarkers. Finally, the main challenges involving existing electrochemical cancer biosensors are evaluated to present the future development prospects of nanomaterials and detection strategies.

Abstract:Lung cancer is one of the most common neoplasms and the leading cause of cancer-related deaths worldwide. Despite recent progress in understanding the pathomechanisms of lung cancer, it is frequently associated with late diagnosis, high incidence of metastases and poor response to treatment. Thus, there is extensive research in the field of biomarkers that aims to optimize management of lung cancer. The aim of this study was to review the current perspectives of a wide spectrum of circulating molecules that seem promising as new potential biomarkers of lung cancer. Among these, biochemical (active proteins), immunological (immunocompetent cells, cytokines, chemokines, and antibodies) and genetic (circulating tumor DNA, cell-free DNA and microRNA) markers are presented and discussed. The use of these markers would support the early detection of lung cancer and might be used for predicting disease progression, response of the disease to targeted therapies, monitoring the course of treatment, and developing individualized diagnostic and therapeutic strategies. Special attention was given to potential markers of nervous system involvement in the course of lung cancer, due to its prevalence and devastating impact. Limitations of the potential biomarkers are also outlined and future directions of investigations in this field highlighted, with the aim of improving the accuracy and practical utility of these biomarkers.

Abstract:Lung cancer is the most prevalent severe illness in both sexes and all ages and the leading cause of cancer-related deaths globally. Late-stage diagnosis is the primary cause of its high mortality rate. Therefore, the management of lung cancer needs early-stage screening. Breath analysis is a non-invasive, low-cost, and user-friendly approach to diagnosing lung cancer. Among the various types of breath sensors, MOS gas sensors are preferred due to their high gas responses, fast response times, robustness, and lower price. This review focuses on the critical role of MOS gas sensors in detecting VOCs in lung cancer patients’ exhaled breath. It introduces the basic working mechanism of MOS gas-sensitive materials, summarizes some high-performance MOS materials suitable for detecting potential lung cancer biomarkers and provides performance enhancement strategies. The review also briefly introduces the sensor array and its pattern recognition algorithm. Finally, we discuss the challenges in developing MOS gas sensors for lung cancer screening and present the prospect of using the e-nose for large-scale early lung cancer screening.

Advances in Clinical Chemistry

Abstract:Lung cancer is the second most–frequently occurring cancer and the leading cause of cancer–associated deaths worldwide. Non–small cell lung cancer (NSCLC), the most common type of lung cancer is often diagnosed in middle or advanced stages and have poor prognosis. Diagnosis of disease at an early stage is a key factor for improving prognosis and reducing mortality, whereas, the currently used diagnostic tools are not sufficiently sensitive for early–stage NSCLC. The emergence of liquid biopsy has ushered in a new era of diagnosis and management of cancers, including NSCLC, since analysis of circulating tumor–derived components, such as cell–free DNA (cfDNA), circulating tumor cells (CTCs), cell–free RNAs (cfRNAs), exosomes, tumor–educated platelets (TEPs), proteins, and metabolites in blood or other biofluids can enable early cancer detection, treatment selection, therapy monitoring and prognosis assessment. There have been great advances in liquid biopsy of NSCLC in the past few years. Hence, this chapter introduces the latest advances on the clinical application of cfDNA, CTCs, cfRNAs and exosomes, with a particular focus on their application as early markers in the diagnosis, treatment and prognosis of NSCLC.
