Abstract:The direct dehydrogenation of ketones has been widely utilized in both academic and industrial settings as the most efficient means of constructing unsaturated ketones and phenols. In contrast to the induction of dehydrogenation at the alpha-site of carbonyl by enol activation, direct and efficient activation of the carbonyl beta-C(sp(3))-H bond to form unsaturated ketones remains unexplored. Herein, we report a polarity-matching-based activation strategy for the carbonyl beta-C(sp(3))-H bond, enabling the photoinduced intramolecular dehydrogenation of ketones without prefunctionalization via the merger of photoredox and cobalt catalysis. This photoinduced mode employs a benign and mild approach to construct unsaturated ketone and phenol skeletons via acceptorless catalytic dehydrogenation, showcasing potential applications in the field of drug synthesis.
Asymmetric Dearomatization of Indoles through Cobalt‐Catalyzed Enantioselective C−H Functionalization Enabled by Photocatalysis

Abstract:Photocatalysis holds a pivotal position in modern organic synthesis, capable of inducing novel reactivities under mild and environmentally friendly reaction conditions. However, the merger of photocatalysis and transition-metal-catalyzed asymmetric C−H activation as an efficient and sustainable method for the construction of chiral molecules remains elusive and challenging. Herein, we develop a cobalt-catalyzed enantioselective C−H activation reaction enabled by visible-light photoredox catalysis, providing a synergistic catalytic strategy for the asymmetric dearomatization of indoles with high levels of enantioselectivity (96% to >99% ee). Mechanistic studies indicate that the excited photocatalyst was quenched by divalent cobalt species in the presence of Salox ligand, leading to the formation of catalytically active chiral Co(III) complex. Moreover, stoichiometric reactions of cobaltacycle intermediate with indole suggest that the irradiation of visible light also play a critical role in the dearomatization step.

Abstract:A photoredox-neutral radical-radical cross-coupling is described for the synthesis of 3-hydroxy-3-alkyloxindoles using isatins and benzyl carboxylic acids as substrates and 2,4,5,6-tetra(9H-carbazol-9-yl)isophthalonitrile (4CzIPN) as the photocatalyst. The method features a broad substrate scope and good functional group tolerance, providing 30 sterically hindered alcohols with moderate to excellent yields. This approach utilizes inexpensive and commercially available starting materials, avoiding the use of transition metals, extra oxidants/reductants, and harsh reaction conditions, showcasing significant applicability and environmental friendliness.

Abstract:The photocatalytic C(sp2)‐H functionalization of quinoxaline derivatives using potassium iodide as photoredox‐mediators has been described. The lower price of potassium iodide compared to standard photocatalysts, readily available substrates, and environmental solvent constitute make this method cost‐effective and interesting. A catalytic amount of KI is applied to the direct activation of C‐H bonds for the construction of heterocyclic compound. This system has simple operation, the characteristics of easy to obtain raw materials, high yield and excellent substrate adaptability provide a new method for the synthesis of C2 substituent quinoxalines.

Abstract:Abstract The Mizoroki-Heck reaction and its reductive analogue are staples of organic synthesis, but the ensuing products often lack a chemical handle for further transformation. Here we report an atom-economical cross-coupling of halopyridines and unactivated alkenes under photoredox catalysis to afford a series of alkene halopyridylation products. This protocol with mild and redox neutral conditions contributes broad substrate scope. As a complement to conventional Heck-type reaction, this radical process avoids the involvement of β -H elimination and thus useful pyridyl and halide groups could be simultaneously and regioselectively incorporated onto alkenes. The success depends on TFA-promoted domino photocatalytic oxidative quenching activation and radical-polar crossover pathway. Plausible mechanism is proposed based on mechanistic investigations. Moreover, the reserved C − X bonds of these products are beneficial for performing further synthetic elaborations.

Abstract:β-Alkoxyketones are important building blocks in organic synthesis. By utilizing CBZ6, with an oxidative potential of -2.16 V (vs the saturated calomel electrode), as a redox-neutral photocatalyst, alkoxyacylation of olefins was accomplished under the irradiation of visible light via a cationic intermediate. It involves the addition of an acyl radical to olefin to form a radical intermediate and the following oxidation of the radical intermediate to the benzyl cationic intermediate that is captured by alkoxy anions. This process provides concise and practical access to the β-functionalized ketones.

Abstract:C(sp3)–H bond functionalization is a powerful strategy for the synthesis of organic compounds due their abundance in simple starting materials. Photoredox catalysis has led to a diverse array of enabling C(sp3)–H activation strategies; however, the direct functionalization of C(sp3)–H to carboxylic acid derivatives remains underexplored. Disclosed herein is the development of a cooperative NHC/photoredox-catalyzed C(sp3)–H esterification transformation. This method enables access to benzylic, 𝛼–heteroatom, and formal β-esterification products in good to excellent yields under mild reaction conditions.

Abstract:A novel ternary EDA complex catalysis strategy has been developed for the photoactivation of oxime esters. By augmenting the electron-donating properties of zwitterions with a base, a ternary EDA complex is formed, facilitating intermolecular charge transfer under visible light. This process enables the single-electron reduction of γ,δ-unsaturated oxime esters, resulting in the homolytic cleavage of the N–O bond. The resulting iminyl radicals then undergo intramolecular cyclization to generate alkyl radicals, enabling C3-alkylation of quinoxalin-2(1 H )-ones in good to excellent yields. Additionally, a novel covalent fluorescent labeling method was established using this mild alkylation method.

Abstract:Transition-metal-catalyzed cross-coupling reactions are a powerful tool to construct carbon–carbon bonds in modern synthetic chemistry. Chromium catalysis is much less developed compared with the widely used palladium and nickel catalysis. Herein, we reported an efficient and flexible chromium-catalyzed radical diacylation of alkenes with the help of visible-light photoredox catalysis, giving access to valuable 1,4-, 1,6-, and 1,7-diones under mild conditions. The synthetic utility of this methodology was proven by converting diones to diverse heterocycles. Furthermore, the same dual catalysis system can be successfully applied to dienes and vinyl cyclopropanes. A possible mechanism of alkene diacylation via dual photoredox/chromium catalysis was proposed according to control experiments and DFT calculations.

Abstract:Recently, the visible-light photoredox decarboxylative couplings of N-(acyloxy)phthalimides (NHPI esters) and its derivatives have become an efficient chemical transformation. Under visible light, the NHPI esters undergo a single-electron transfer (SET) process to afford the corresponding carbon or nitrogen radicals that participate in many chemical transformations. The photoredox decarboxylative couplings have been applied to achieve construction of an array of carbon–carbon and carbon–heteroatom bonds as well as the synthesis of carbocycles and heterocycles. This review categorises photocatalysts, discusses the application and catalysis mechanisms of NHPI esters, and details recent progress in this field.

Abstract:We report the photocatalytic oxidation of α-carbonyl radicals of amides or esters to the corresponding α-carbonyl carbocations through super photoreductant CBZ6 induced redox-neutral photocatalysis. The α-carbonyl radicals are formed by the β-addition of alkyl radicals generated in situ by the photocatalytic fragmentation of N -hydroxyphthalimide esters to the α,β-unsaturated amides and esters. This method enables the α-nucleophilic addition of hydroxyl or alkoxyl radicals to amides and esters without any prefunctionalization.

Abstract:The dearomatization of indoles represents the most efficient approach for accessing highly valued indolines. The inherent nucleophilic reactivity of indoles has dictated indole dearomatization development in both 1e(-) and 2e(-) processes. However, the dearomatization of electron-deficient indoles has been challenging. Herein, we introduce a conceptually distinct photoredox-mediated Giese-type transformation strategy, which is generally used for the conjugate addition of radicals to simple alpha, beta-unsaturated systems, for chemoselectively breaking C=C bonds embedded in the aromatic structure. Moreover, highly diastereoselective addition of challenging neutral radicals has been achieved by Oppolzer camphorsultam chiral auxiliary. Structurally diverse amine-functionalized chiral indolines carrying distinct functional and stereochemical diversity are produced from a wide array of amines as radical precursors. Furthermore, the mild, powerful manifold is capable of the late-stage modification of complex natural products and pharmaceuticals. DFT studies are performed to elucidate the observed stereochemical outcomes.

Abstract:A synergetic catalytic system of organophotocatalyst 4CzIPN, thianthrene, and methanol was developed for visible‐lightinduced Giese reactions of various alkanes and alkenes, upon using alkoxy radicals as hydrogen atom transfer reagents. The reaction can be performed under mild conditions to provide the alkylated products in moderate to excellent yields, avoiding the use of any strong oxidants and transition metal reagents. Different from the known LMCT and PCET processes, the key to success of this approach is the in situ thianthrenation of methanol by thianthrene radical cation to trigger the generation of methoxy radical.

Abstract:C-H functionalization provides unparalleled benefits for the late-stage modification of complex molecules. In recent years, photocatalysis has progressed significantly due to its mild conditions, sustainability and selectivities. Current mechanisms for photocatalytic C-H functionalization primarily involve direct single-electron oxidation and direct hydrogen atom transfer (d-HAT) by radicals, both of which are invalid for the functionalization of highly electron-deficient C(sp3)-H bonds. Here, we developed a cooperative photoenzymatic system consisting of a flavin-dependent ene-reductase (ER) and an exogenous photocatalyst fluorescein (FI) to achieve enantiodivergent functionalization of electron-deficient C(sp3)-H bonds. Mechanistic studies revealed a novel pathway for radical intermediate formation via excited-state FI*-induced single-electron oxidation of carbanions under alkaline conditions. The overall catalytic efficiency is enhanced by the electron transfer (ET) between FMNox and FI radical anion, while the stereoselectivity is controlled by ERs through enantioselective hydrogen atom transfer (HAT). This suggests that thermodynamically unfavorable single-electron oxidation of highly electron-deficient species can occur through ion-to-radical conversion. We anticipate that our novel mode of photoenzymatic catalysis will inspire new pathways for generating free radical intermediates and foster innovative strategies for achieving photoenzymatic new-to-nature reactions.

Abstract:Hydrogen atom transfer (HAT) and photoredox dual catalysis provides a unique opportunity in organic synthesis, enabling the direct activation of C/Si/S–H bonds. However, the activation of O–H bonds of β,γ-unsaturated oximes poses a challenge due to their relatively high redox potential, which exceeds the oxidizing capacity of most currently developed photocatalysts. We here demonstrate that the combination of HAT and photoredox catalysis allows the activation of O–H bond of β,γ-unsaturated oximes. The strategy effectively addresses the oxime's high redox potential and offers a universal pathway for iminoxyl radical formation. Leveraging the versatility of this approach, a diverse array of valuable heterocycles have been synthesized with the use of different radical acceptors. Mechanistic studies confirm a HAT process for the O–H bond activation.

Abstract:Photoredox-catalyzed isomerization of gamma-carbonyl-substituted allylic alcohols to their corresponding carbonyl compounds was achieved for the first time by C-H bond activation. This catalytic redox-neutral process resulted in the synthesis of 1,4-dicarbonyl compounds. Notably, allylic alcohols bearing tetrasubstituted olefins can also be transformed into their corresponding carbonyl compounds. Density functional theory calculations show that the carbonyl group at the gamma-position of allylic alcohols are beneficial to the formation of their corresponding allylic alcohol radicals with high vertical electron affinity, which contributes to the completion of the photoredox catalytic cycle.
