Abstract:For the past 4 decades, intensive molecular studies of mostly leaf mesophyll cell-infecting pathogens and chewing insects have led to compelling models of plant-pathogen and plant-insect interactions. Yet, some of the most devastating pathogens and insect pests live in or feed on the phloem, a systemic tissue belonging to the plant vascular system. Phloem tissues are difficult to study, and phloem-inhabiting pathogens are often impossible to culture, thus limiting our understanding of phloem-insect/pathogen interactions at a molecular level. In this Perspective, we highlight recent literature that reports significant advances in the understanding of phloem interactions with insects and prokaryotic pathogens and attempt to identify critical questions that need attention for future research. It is clear that study of phloem-insect/pathogen interactions represents an exciting frontier of plant science, and influx of new scientific expertise and funding is crucial to achieve faster progress in this important area of research that is integral to global food security.

Abstract:In the past four decades, tremendous progress has been made in understanding how plants respond to microbial colonization and how microbial pathogens and symbionts reprogram plant cellular processes. In contrast, our knowledge of how environmental conditions impact plant-microbe interactions is less understood at the mechanistic level, as most molecular studies are performed under simple and static laboratory conditions. In this review, we highlight research that begins to shed light on the mechanisms by which environmental conditions influence diverse plant-pathogen, plant-symbiont, and plant-microbiota interactions. There is a great need to increase efforts in this important area of research in order to reach a systems-level understanding of plant-microbe interactions that are more reflective of what occurs in nature.

Abstract:The co-existence of plants and microorganisms throughout their evolution resulted in an intertwined ecological network that serves as the basis for the functioning of today's terrestrial ecosystems. The interactions between plants, bacteria, and fungi are often multipartite and occur both within and among kingdoms. Apart from the textbook examples of tight mutualistic associations such as root nodule symbiosis or mycorrhizas, there are also looser interactions, e.g., those between plants and plant growth-promoting bacteria and fungi. Additionally, plants are also targets of microbial pathogens. Thus, intricate mechanisms of how plants and microorganisms recognize each other have evolved, mediated by chemical signals resulting in a differential gene expression and, eventually, a physiological response. This paper aims to review the molecular basis of such signaling, the apparatus of signal sensing and transduction in bacteria and plants are outlined and exemplified. Moreover, examples of recent discoveries in the field of plant–microbe signaling are presented, i.e., quorum quenching and the involvement of small RNAs. Last but not least, suggestions are provided for the potential exploitation of plant–microbe communication networks for human benefit, e.g., in the field of sustainable agriculture, aiming at crop growth promotion and protection against pathogens.

Abstract:Plant pathogens represent a constant and major threat to global food production, with 20%–30% global crop losses estimated, principally in food-deficit areas [<a class="ref-tip" href="#ppat.1007740.ref001">1</a>]. Pesticide use, breeding of resistance genes, and genetic manipulation of plant immune components have helped to mitigate this threat. However, rapid evolution of pathogen resistance and virulence, together with host range expansion and host jumps, contribute to severe disease outbreaks, especially in the context of current agricultural practices [<a class="ref-tip" href="#ppat.1007740.ref002">2</a>]. This underscores the need to reduce the lag time between the appearance of new diseases and development of protective measures effective on a broad range of pathogens and host plants.In this context, microbial products and inoculants for plant protection have recently gained attention thanks to the large efforts made to systematically isolate, identify, and characterize plant-associated microbes that engage in intimate association with healthy plants [<a class="ref-tip" href="#ppat.1007740.ref003">3</a>]. Recent findings indicate that individual- and community-level features provided by plant microbiota members can confer extended immune functions to the plant host. Importantly, the traits lent by "beneficial" microbes strongly depend on the interplay between the soil nutrient status and the plant immune system [<a class="ref-tip" href="#ppat.1007740.ref004">4</a>, <a class="ref-tip" href="#ppat.1007740.ref005">5</a>]. Thus, the successful implementation of microbiota-mediated disease protection will depend on our mechanistic understanding of how microorganisms interact with their hosts and with one another in natural environments. Here, we seek to synthesize the state of this cross-disciplinary field to bridge the gap toward rational design of synthetic microbial communities (SynComs) [<a class="ref-tip" href="#ppat.1007740.ref006">6</a>] with broad, durable, and flexible plant protective activities.

Abstract:Marchantia polymorpha is a bryophyte gaining significance as a model plant in evolutionary studies in recent years. This is attributed to its small-sequenced genome, standardized transformation methodology, global distribution, and easy and rapid in vitro culturing. As an evolutionary model, M. polymorpha contributes to our understanding of the evolution of plant defensive responses and the associated hormonal signaling pathways. Through its interaction with microorganisms, M. polymorpha serves as a valuable source of knowledge, yielding insights into new microbial species and bioactive compounds. Bibliographic analysis involved collecting, reading, and categorizing documents obtained from the Scopus and Web of Science databases using different search terms. The review was based on 30 articles published between 1995 and 2023, with Japanese and Spanish authors emerging as the most prolific contributors in this field. These articles have been grouped into four main themes: antimicrobial metabolites produced by M. polymorpha ; identification and characterization of epiphytic, endophytic, and pathogenic microorganisms; molecular studies of the direct interaction between M. polymorpha and microorganisms; and plant transformation using bacterial vectors. This review highlights the key findings from these articles and identifies potential future research directions.

A growing body of literature reports that plants interact through substrate-borne vibrations with arthropod pests and mutualistic arthropods that provide biological control and pollination services. Here, we propose a new theoretical framework that integrates insights from biological control, the ecology of fear, and plant-borne vibrations, to address plant-insect interactions and explore new, sustainable opportunities to improve plant health and productivity.

Abstract:In their natural environment, plants are part of a rich ecosystem including numerous and diverse microorganisms in the soil. It has been long recognized that some of these microbes, such as mycorrhizal fungi or nitrogen fixing symbiotic bacteria, play important roles in plant performance by improving mineral nutrition. However, the full range of microbes associated with plants and their potential to replace synthetic agricultural inputs has only recently started to be uncovered. In the last few years, a great progress has been made in the knowledge on composition of rhizospheric microbiomes and their dynamics. There is clear evidence that plants shape microbiome structures, most probably by root exudates, and also that bacteria have developed various adaptations to thrive in the rhizospheric niche. The mechanisms of these interactions and the processes driving the alterations in microbiomes are, however, largely unknown. In this review, we focus on the interaction of plants and root associated bacteria enhancing plant mineral nutrition, summarizing the current knowledge in several research fields that can converge to improve our understanding of the molecular mechanisms underpinning this phenomenon.

Abstract:Deciphering plant-microbe interactions is a promising aspect to understand the benefits and the pathogenic effect of microbes and crop improvement. The advancement in sequencing technologies and various 'omics' tool has impressively accelerated the research in biological sciences in this area. The recent and ongoing developments provide a unique approach to describing these intricate interactions and test hypotheses. In the present review, we discuss the role of plant-pathogen interaction in crop improvement. The plant innate immunity has always been an important aspect of research and leads to some interesting information like the adaptation of unique immune mechanisms of plants against pathogens. The development of new techniques in the post - genomic era has greatly enhanced our understanding of the regulation of plant defense mechanisms against pathogens. The present review also provides an overview of beneficial plant-microbe interactions with special reference to <i>Agrobacterium tumefaciens</i>-plant interactions where plant derived signal molecules and plant immune responses are important in pathogenicity and transformation efficiency. The construction of various Genome-scale metabolic models of microorganisms and plants presented a better understanding of all metabolic interactions activated during the interactions. This review also lists the emerging repertoire of phytopathogens and its impact on plant disease resistance. Outline of different aspects of plant-pathogen interactions is presented in this review to bridge the gap between plant microbial ecology and their immune responses.

Abstract:In natural ecosystems, plants interact with biotic components such as microbes, insects, animals and other plants as well. Generally, researchers have focused on each interaction separately, which condenses the significance of the interaction. This limited presentation of the facts masks the collective role of constantly interacting organisms in complex communities disturbing not only plant responses but also the response of organisms for each other in natural ecological settings. Beneficial microorganisms interact with insect herbivores, their predators and pollinators in a bidirectional way through the plant. Fascinatingly, insects employ diverse tactics to protect themselves from parasites or predators. Influences of microbial and insects attack on plants can bring changes in info-chemical frameworks and play a role in the food chain also. After insect herbivory and microbial pathogenesis, plants exhibit intense morpho-physiological and chemical reprogramming that leads to repellence/attraction of attacking organism or its natural enemy. The characterization of such interactions in different ecosystems is receiving due consideration, and underlying molecular and physiological mechanisms must be the point of concentration to unveil the evolution of multifaceted multitrophic interactions. Therefore, we have focused this phenomenon in a more realistic setting by integrating ecology and physiology to portray these multidimensional interfaces. We have shown, in this article, physiological trajectories in plant-microbe and insect relationship and their ecological relevance in nature. We focus and discuss microbial pathogenesis in plants, induced defense and the corresponding behavior of herbivore insects and vice-versa. It is hoped that this review will stimulate interest and zeal in microbes mediated plant-insect interactions along with their ecological consequences and encourage scientists to accept the challenges in this field.

Abstract:Plant-associated microorganisms fulfill important functions for plant growth and health. Direct plant growth promotion by microbes is based on improved nutrient acquisition and hormonal stimulation. Diverse mechanisms are involved in the suppression of plant pathogens, which is often indirectly connected with plant growth. Whereas members of the bacterial genera Azospirillum and Rhizobium are well-studied examples for plant growth promotion, Bacillus, Pseudomonas, Serratia, Stenotrophomonas, and Streptomyces and the fungal genera Ampelomyces, Coniothyrium, and Trichoderma are model organisms to demonstrate influence on plant health. Based on these beneficial plant–microbe interactions, it is possible to develop microbial inoculants for use in agricultural biotechnology. Dependent on their mode of action and effects, these products can be used as biofertilizers, plant strengtheners, phytostimulators, and biopesticides. There is a strong growing market for microbial inoculants worldwide with an annual growth rate of approximately 10%. The use of genomic technologies leads to products with more predictable and consistent effects. The future success of the biological control industry will benefit from interdisciplinary research, e.g., on mass production, formulation, interactions, and signaling with the environment, as well as on innovative business management, product marketing, and education. Altogether, the use of microorganisms and the exploitation of beneficial plant–microbe interactions offer promising and environmentally friendly strategies for conventional and organic agriculture worldwide.

Abstract:Chemical pesticides and fertilizers are used in agricultural production worldwide to prevent damage from plant pathogenic microorganisms, insects, and nematodes, to minimize crop losses and to preserve crop quality. However, the use of chemical pesticides and fertilizers can severely pollute soil, water, and air, posing risks to the environment and human health. Consequently, developing new, alternative, environment-friendly microbial soil treatment interventions for plant protection and crop yield increase has become indispensable. Members of the filamentous fungal genus Trichoderma (Ascomycota, Sordariomycetes, Hypocreales) have long been known as efficient antagonists of plant pathogenic microorganisms based on various beneficial traits and abilities of these fungi. This minireview aims to discuss the advances in the field of Trichoderma-containing multicomponent microbiological inoculants based on recent experimental updates. Trichoderma strains can be combined with each other, with other fungi and/or with beneficial bacteria. The development and field performance of such inoculants will be addressed, focusing on the complementarity, synergy, and compatibility of their microbial components.

Abstract:Plant-microbe interaction research has had a transformative trajectory, from individual microbial isolate studies to comprehensive analyses of plant microbiomes within the broader phytobiome framework. Acknowledging the indispensable role of plant microbiomes in shaping plant health, agriculture, and ecosystem resilience, we underscore the urgent need for sustainable crop production strategies in the face of contemporary challenges. We discuss how the synergies between advancements in 'omics technologies and artificial intelligence can help advance the profound potential of plant microbiomes. Furthermore, we propose a multifaceted approach encompassing translational considerations, transdisciplinary research initiatives, public-private partnerships, regulatory policy development, and pragmatic expectations for the practical application of plant microbiome knowledge across diverse agricultural landscapes. We advocate for strategic collaboration and intentional transdisciplinary efforts to unlock the benefits offered by plant microbiomes and address pressing global issues in food security. By emphasizing a nuanced understanding of plant microbiome complexities and fostering realistic expectations, we encourage the scientific community to navigate the transformative journey from discoveries in the laboratory to field applications. As companies specializing in agricultural microbes and microbiomes undergo shifts, we highlight the necessity of understanding how to approach sustainable agriculture with site-specific management solutions. While cautioning against overpromising, we underscore the excitement of exploring the many impacts of microbiome-plant interactions. Abstract:Plants are constantly interacting with the diverse microbial community as well as insect pests throughout their life cycle. Due to their sessile nature, plants rely solely on the intracellular signaling and reprogramming of cellular events to resist against pathogens. Insect pests are usually dependent on the nutrient-rich fluid obtained from plants or directly consume plant parts to sustain their life cycle. Plants possess a plethora of microbial communities; these microbiomes constantly influence the physiology, growth, development, and immunity in plants. Hence, the phyllosphere and rhizosphere are believed to play a major role in plant-insect interaction. The phyllosphere, rhizosphere, and endosymbiotic microbiome are currently under extensive scientific investigation. Recently, the advancement of metagenomic next-generation sequencing (mNGS) platforms revolutionized plant-associated microbiome analysis and has unveiled many beneficial microbial communities protecting against diverse pathogenic microorganisms and insect pests. Mycorrhiza is also an important component of the rhizosphere, as it may play a role in soil microbiota, thus indirectly influencing the interaction of insects with plants. In this regard, the present review tries to focus on some major insect pests of plants, the molecular mechanism of plant–insect interaction, and the probable role of phyllosphere and rhizosphere microbiome in this plant–insect encounter. This review is believed to open up a new dimension in developing resistance in plants against insect pests.
