Abstract:In this study, we synthesized PVA-g-poly(AMPS) nanogels with the aim of enhancing the solubility and dissolution of ticagrelor (TGR). Ticagrelor, a noncompetitive, reversible P2Y12 receptor antagonist, is prescribed to treat acute coronary syndrome. Ticagrelor has restricted oral bioavailability (≈36%) because of its poor solubility and permeability. The free radical polymerization methodology was employed to synthesize nanogels with varied concentrations of poly(vinyl alcohol) (polymer),...

What problem does this paper attempt to address?
The main problem that this paper attempts to solve is to improve the solubility and oral bioavailability of the drug Ticagrelor. Ticagrelor is a non - competitive and reversible P2Y12 receptor antagonist used for treating acute coronary syndrome (ACS), but due to its poor water - solubility and permeability, its oral bioavailability is only about 36%. Therefore, researchers synthesize PVA - g - poly(AMPS) nanogels to enhance the solubility and release characteristics of Ticagrelor, thereby improving its bioavailability and therapeutic effect. ### Research Background - **Problem Raising**: Many drugs have low oral bioavailability due to poor water - solubility, which affects the therapeutic effect. As an important drug for treating cardiovascular diseases, Ticagrelor has a particularly prominent problem of poor water - solubility. - **Existing Solutions**: Currently, there are various methods for improving the solubility of drugs, such as amorphous solid dispersions, nanogels, solid - lipid nanoparticles, etc. Among them, nanogels have attracted much attention because of their high hydrophilicity, high swelling property, high drug - loading efficiency and good biocompatibility. ### Research Purposes - **Synthesis of PVA - g - poly(AMPS) Nanogels**: Synthesize nanogels composed of different concentrations of polyvinyl alcohol (PVA), 2 - acrylamido - 2 - methylpropanesulfonic acid (AMPS) and cross - linker N,N - methylenebisacrylamide (MBA) by free - radical polymerization method. - **Characterization and Evaluation**: Conduct physical and chemical characterizations of the synthesized nanogels, including particle size analysis, ζ - potential measurement, swelling study, drug - loading efficiency, dissolution study, Fourier transform infrared spectroscopy (FTIR), powder X - ray diffraction (PXRD), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC) and scanning electron microscopy (SEM), etc. - **Toxicological Evaluation**: Evaluate the biocompatibility and safety of nanogels through cell viability assay (MTT test) and acute oral toxicity experiment. ### Main Results - **Particle Size and ζ - Potential**: The particle size of the nanogel of the optimized formula (PA5) is 45.86 nm, the polydispersity index (PDI) is 0.41, and the ζ - potential is - 7.04 mV, indicating that it has good colloidal stability. - **Drug - Loading Efficiency**: The drug - loading efficiency of the PA5 formula reaches 85.1%. - **Dissolution and Solubility**: In vitro dissolution studies show that the release rate of Ticagrelor from the nanogel reaches 99.91%, and the solubility is increased by 8.2 times. - **Thermal Stability and Crystalline Properties**: FTIR, XRD and TGA/DSC analyses confirm the thermal stability and amorphous structure of the nanogels. - **Biocompatibility**: Cell viability assay and acute oral toxicity experiment verify the biocompatibility and safety of the nanogels. ### Conclusion As an effective drug - delivery system, PVA - g - poly(AMPS) nanogels can significantly improve the solubility and oral bioavailability of Ticagrelor, have good biocompatibility and safety, and are expected to play an important role in clinical applications.
