Abstract:Luminogens with aggregation-induced emission feature (AIEgens) are highly demanded because of its excellent performance in fluorescent bioprobes. In this work, three AIEgens based on tetraphenylethylene isoquinolinium (TPE-IQ) derivatives by introducing different electron donor and sterically hindered groups to replace the phenyl ring of the isoquinolinium core are prepared in a one-pot reaction with a high yield and characterized systematically. By the analysis of photophysical properties and theoretical calculations, the structure-property relationships among them become clear. Although they all inherit AIE feature as expected, these similar structural AIEgens present different organelle-targets, which is attributed to their different cellular uptake way and energy barrier to cell membrane by detailed experiments. In addition, these TPE-IQ-based AIEgens are potential candidates for photodynamic therapy originating from their activities of generating reactive oxygen species under visible light irradiation.

Abstract:Fluorescent probes have been widely investigated for their features of rapid response, easy operation and high sensitivity. Among them, reaction-based fluorescent probes, for their unique reaction-based nature, guarantee them with excellent selectivity, effectively avoiding the possible interference from other chemical and biological species in physiological environment. Conventional reaction-based fluorescent probes are aggregation-caused quenching (ACQ) fluorophores. The application of these kinds of probes are limited for their poor photostability and narrow Stokes shifts. Compared with ACQ fluorophores, aggregation-induced emission (AIE) fluorophores become emissive in aggregation states with higher signal-to-noise ratio, better photostability and larger Stokes shifts. In this review, we summarize the latest developed reaction-based AIE-active probes, including the design principle and application in various sensing systems and give an outlook for the future development of this kind of promising fluorescent probes.

Abstract:As a sensitive, selective, and non-invasive method, fluorescence bioimaging stands out in biological and pathological studies to visualize biological processes on a microscale. However, the initial photophysical property of conventional organic fluorophores leads to poor photostability and low signal-to-noise ratio, which largely hinder their performance in the long-term monitoring and tracing of intracellular process. In contrast, AIE luminogens (AIEgens) exhibit the opposite photophysical property that overcomes the barrier to facilitate real-time and long-term intracellular dynamics visualization. This work summarizes the recent development of AIE probes to unveil intracellular processes, covering from intracellular microenvironment monitoring to tracing intracellular bioprocess. As fluorescent bioprobes with AIE features depict unlimited potential in bioimaging studies, we hope this review can offer more inspiration and directions to the future design and development of AIEgens for visualizing and monitoring diversified intracellular dynamics.

Abstract:The design of novel nanomaterials and nanoprobes used to diagnose and treat innumerable diseases has exhibited great potential in biomedical fields, which further promoted the development of advanced imaging and therapy techniques for tumors, including computed tomography, radiotherapy, chemother-apy, fluorescent bioimaging, photoacoustic (PA) bioimaging, and phototherapy. PA imaging and photody-namic/photothermal therapy (PDT/PTT) show clear advantages for monitoring and treating tumors due to their high efficiency, spatial specificity, low side effects, and easy operation. Aggregate-induced emission luminous (AIEgens)-based nanoprobes and nanomaterials attracted widespread attention and exhibited a wide range of applications for PA imaging and PDT/PTT treatments in the last years. In this review, we present the advances in the rational design and facile synthesis of various functional AIEgens with near-infrared emission for PA imaging and phototherapy applications. The modifications of AIE cores with various substances such as polymers, biomolecules, nanoparticles, and others for the creation of smart AIEgens through chemical conjugation, nanoprecipitation, and self-assembly are demonstrated and discussed. These designed AIEgens have exhibited enhanced PA imaging efficiency and high perfor-mance for combined imaging-guided PDT/PTT treatment of tumors. This topic-focused review is helpful for the readers to understand potential strategies for the design and preparation of function-specific AIEgens, and could promote the development of PA imaging and phototherapy techniques for practical detection, diagnosis, and treatment of cancers.(c) 2022 Elsevier B.V. Photodiagnosis and Photodynamic Therapy

Abstract:Photodynamic therapy (PDT) is an established clinical treatment technology with which utilizes excitation light of a specific wavelength to excite photosensitizers (PSs) for generating reactive oxygen species (ROS) which lead to the cancer cell death. Over the past decades of PDT research, apparent progress have been made in the development of PSs. However, many inherent characteristics of traditional PSs have caused many various problems in PDT, such as low treatment efficiency at aggregation state and shallow treatment depth. In allusion to these problems, aggregation-induced emission (AIE)-based PSs have been reported in recent years. Here, the article reviews the design strategy and the biomedical applications of AIE PSs in detail, which begins with a summary of traditional PSs for a comparison between traditional PSs and AIE PSs. Subsequently, the different functional AIE PSs in photodynamic cancer cells ablation and image-guided therapy are discussed in detail taking controllable excitation wavelength, stimulus response and PDT/photothermal therapy synergistic effect as examples. These studies have demonstrated the great potential of AIE PSs as effective theranostic agents in recent years. And the review provides references for the development of new PSs and hopefully spur more research interest in AIE PSs for future research.

Abstract:Fluorescence imaging‐guided photodynamic therapy (PDT) has attracted extensive attention due to its potential of real‐time monitoring the lesion locations and visualizing the treatment process with high sensitivity and resolution. Aggregation‐induced emission luminogens (AIEgens) show enhanced fluorescence and reactive oxygen species (ROS) generation after cellular uptake, giving them significant advantages in bioimaging and PDT applications. However, most AIEgens are unfavorable for the application in organisms due to their severe hydrophobicity. Anion‐π+ type AIEgens carry intrinsic charges that can effectively alleviate their hydrophobicity and improve their binding capability to cells, which is expected to enhance the bioimaging quality and PDT performance. This concept summarizes the applications of anion‐π+ type AIEgens in fluorescence imaging, fluorescence imaging‐guided photodynamic anticancer and antimicrobial therapy in recent years, hoping to provide some new ideas for the construction of robust photosensitizers. Finally, the current problems and future challenges of anion‐π+ AIEgens are discussed.

Abstract:The development of novel photosensitizers with aggregation-induced emission (AIE) characteristics has aroused tremendous interest, because it could combine efficient bioimaging with precise photodynamic therapy, which would thus dramatically promote applications in biomedical treatment. Among various AIE luminogens (AIEgens), heterocycle-containing molecules are highly promising for this usage because of their high photostability and tunable electronic properties. In this work, a pyrazine-containing AIEgen with a dicyanopyrazine moiety as an electron acceptor and a triphenylamine unit as an electron donor was chosen for study. The V-shaped donor-π-acceptor-π-donor structure of the AIEgen endowed its nanoparticles with excellent nonlinear optical properties for two-photon cell imaging under near-infrared laser excitation. Also, under the same excitation, the nanoparticles could produce reactive oxygen species and further kill the cells efficiently and accurately. The present work thus presents a pyrazine-containing AIEgen as a new photosensitizer for imaging-guided two-photon photodynamic therapy and gives more opportunities for deep-tissue treatment of cancer in future research.

Abstract:Fluorescent probes with aggregation-induced emission (AIE) property have become fascinating and vital in biological fields due to their bright fluorescence in the solid state. In contrast traditional AIE materials are obscured by the off-target effects and the lack of spatial and temporal control. Photoactivatable materials with AIE characteristics, whose physicochemical behaviors can be remotely activated by light, provide great potentials in biochemical information acquisition with high spatial and temporal resolution. By using AIE-featured photoactivatable fluorescent probes, we can accurately analyze the targets of interest. For example, where, when, and to what extent a process is started or stopped by manipulating the non-invasive light accurately. Thus, many researchers are enthusiastic about developing AIE-featured photoactivatable materials and mainly focus on developing novel molecules by rational molecular structure design, and exploring advanced applications by appropriate molecular functionalization. In this review, we summarize the recent achievements of photoactivatable materials with AIE characteristics from the aspects involving inherent mechanism of photoactivity, molecular design strategy and the corresponding applications in biological fields. The biological applications are highlighted and discussed, including photoactivatable bioimaging, diagnosis, and photo-controlled therapy. Abstract:Luminescent bioprobes are powerful analytical means for biosensing and optical imaging. Luminogens featured with aggregation-induced emission (AIE) attributes have emerged as ideal building blocks for high-performance bioprobes. Bioprobes constructed with AIE luminogens have been identified to be a novel class of FL light-up probing tools. In contrast to conventional bioprobes based on the luminophores with aggregation-caused quenching (ACQ) effect, the AIE-based bioprobes enjoy diverse superiorities, such as lower background, higher signal-to-noise ratio and sensitivity, better accuracy, and more outstanding resistance to photobleaching. AIE-based bioprobes have been tailored for a vast variety of purposes ranging from biospecies sensing to bioimaging to theranostics (i.e., image-guided therapies). In this review, recent five years' advances in AIE-based bioprobes are briefly overviewed in a perspective distinct from other reviews, focusing on the most appealing trends and progresses in this flourishing research field. There are altogether 11 trends outlined, which have been classified into four aspects: the probe composition and form (bioconjugtes, nanoprobes), the output signal of probe (far-red/near-infrared luminescence, two/three-photon excited fluorescence, phosphorescence), the modality and functionality of probing system (dual-modality, dual/multifunctionality), the probing object and application outlet (specific organelles, cancer cells, bacteria, real samples). Typical examples of each trend are presented and specifically demonstrated. Some important prospects and challenges are pointed out as well in the hope of intriguing more interests from researchers working in diverse areas into this exciting research field.

Abstract:In recent years, with increasing demands for in vivo fluorescence imaging and photodynamic therapy, light in the second near-infrared window (NIR-II; 1000–1700 nm) has attracted tremendous interest because it offers numerous merits, including deep penetration, minimal phototoxicity, diminished tissue autofluorescence, and reduced tissue absorption and scattering. Among the diverse types of nanoparticles, organic nanoprobes with aggregation-induced emission (AIE) characteristics have emerged as a better option owing to their ultra-brightness, excellent photostability, low cytotoxicity, and tailorable optical properties. Recent efforts in the AIE realm have revealed advancements in molecular design for long wavelength absorption and multiphoton excitation, which efficiently modulate the working optical region to the NIR-II region. In this review, the current status of AIE nanoprobes in the NIR-II window is summarized. Starting with molecular design strategies, recent efforts in fluorescence imaging and photodynamic therapy are then discussed. Finally, perspectives and challenges in this newly emerging field are given. This review hopes to encourage more innovative ideas in material design and biomedical applications for promoting AIE nanoprobes for future clinical translation.

Abstract:Phototheranostics based on luminogens with aggregation-induced emission (AIE) characteristics is captivating increasing research interest nowadays. However, AIE luminogens are inherently featured by inferior absorption coefficients (ε) resulting from the distorted molecular geometry. Besides, molecular innovation of long-wavelength light-excitable AIE luminogens with highly efficient phototheranostic outputs is an appealing yet significantly challenging task. Herein, on the basis of a fused-ring electron acceptor–donator–acceptor (A–D–A) type molecule (IDT) with aggregation-caused quenching (ACQ) properties, molecular engineering smoothly proceeds and successfully yields a novel AIE luminogen (IDT-TPE) via simply modifying tetraphenylethene (TPE) moieties on the sides of IDT backbone. The AIE tendency endows IDT-TPE nanoparticles with enhanced fluorescence brightness and far superior fluorescence imaging performance to IDT nanoparticles for mice tumors. Moreover, IDT-TPE nanoparticles exhibit near-infrared light-excitable features with a high ε of 8.9 × 104 m−1 cm−1, which is roughly an order of magnitude higher than that of most previously reported AIE luminogens. Combining with their reactive oxygen species generation capability and extremely high photothermal conversion efficiency (59.7%), IDT-TPE nanoparticles actualize unprecedented performance in multimodal phototheranostics. This study thus brings useful insights into the development of versatile phototheranostic materials with great potential for practical cancer theranostics.

Abstract:Fluorescent light-up probes comprising a tetraphenylethene unit with aggregation-induced emission (AIE) characteristics and a water-soluble peptide have been designed and synthesized which provide cell membrane and nuclear permeability to live cells. This strategy has offered new opportunities for the development of probes with light-up ability and good signal-to-noise ratio. The selectivity or targeting specificity is determined by the peptide sequence, i.e. a nuclear localization signal that leads to nucleus imaging and a cell biomarker targeting peptide that offers specific light-up imaging of HT-29 cells.

Abstract:Three-photon absorption (3 PA) in the near IR region is among the most prominent nonlinear optical (NLO) effects and has attractive applications in chemical/biological sensing and imaging. Yet, rationally constructed molecules with small molecular weight and reasonable 3 PA cross-section has been rarely reported. Herein, we designed a novel three-photon absorption photostable luminogen (namely X1) with enhanced aggregation induced emission (AIE) and the ability to achieve multi-photon imaging with femtosecond laser excitation. X1 was constructed from diaminobenzene and diethylamino salicylaldehyde forming a novel di-Schiff base. It possesses a large conjugated delocalization which exhibits large three-photon absorption (3 PA) cross-section values. We also showed that by using a suitable delivery vector, X1 compound could applied as a live cell imaging probe thus providing a valuable tool to study lipid droplets/lysosome interaction in depth tissues.

Abstract:High-quality fluorescence bioimaging requires organic exogenous contrast agents with biocompatibility, brightness, and photostability. The recently discovered and rapidly developed aggregation-induced emission luminogens (AIEgens) are excellent candidates because they exhibit enhanced emission upon the restriction of intramolecular motions (RIM) effect. AIEgens are individually lit up once bound with biomolecules or influenced by the surrounding bio-environment, and abundant biological information can be acquired on the basis of high-sensitivity fluorescence imaging. In addition, AIEgen-incorporated nanoparticles possess bright fluorescence and improved photostability, which is beneficial to long-term bioimaging with high contrast and spatial resolution. In this review, we summarize the latest advances in AIEgen-based fluorescence bioimaging, as well as the relevant applications in theranostics.

Abstract:The severe hypoxia in solid tumors and the vicious aggregation‐caused fluorescence quenching (ACQ) of conventional photosensitizers (PSs) have limited the application of fluorescence imaging‐guided photodynamic therapy (PDT), although this therapy has obvious advantages in terms of its precise spatial–temporal control and noninvasive character. PSs featuring type I reactive oxygen species (ROS) based on free radicals and novel aggregation‐induced emission (AIE) characteristics (AIE‐PSs) could offer valuable opportunities to resolve the above problems, but molecular engineering methods are rare in previous reports. Herein, a strategy is proposed for generating stronger intramolecular charge transfer in electron‐rich anion‐π+ AIE‐active luminogens (AIEgens) to help suppress nonradiative internal conversion and to promote radiative and intersystem crossing to boost free radical generation. Systematic and detailed experimental and theoretical calculations prove the proposal herein: the electron‐donating abilities are enhanced in collaborative donors, and the AIE‐PSs exhibit higher performance in near‐infrared fluorescence imaging‐guided cancer PDT in vitro/vivo. This work serves as an important reference for the design of AIE‐active free radical generators to overcome the ACQ and tumor hypoxia challenges in PDT.

Abstract:Far-red and near-infrared (NIR) fluorescent materials possessing the characteristics of strong two-photon absorption and aggregation-induced emission (AIE) as well as specific targeting capability are much-sought-after for bioimaging and therapeutic applications due to their deep penetration depth and high resolution. Herein, a series of dipolar far-red and NIR AIE luminogens with a strong push-pull effect are designed and synthesized. The obtained fluorophores display bright far-red and NIR solid-state fluorescence with a high quantum yield of up to 30%, large Stokes shifts of up to 244 nm, and large two-photon absorption cross-sections of up to 887 GM. A total of three neutral AIEgens show specific lipid droplet (LD)-targeting capability, while the one with cationic and lipophilic characteristics tends to target the mitochondria specifically. All of the molecules demonstrate good biocompatibility, high brightness, and superior photostability. They also serve as efficient two-photon fluorescence-imaging agents for the clear visualization of LDs or mitochondria in living cells and tissues with deep tissue penetration (up to 150 μm) and high contrast. These AIEgens can efficiently generate singlet oxygen upon light irradiation for the photodynamic ablation of cancer cells. All of these intriguing results prove that these far-red and NIR AIEgens are excellent candidates for the two-photon fluorescence imaging of LDs or mitochondria and organelle-targeting photodynamic cancer therapy.
