Abstract:Cells accomplish diverse functions using the same molecular building blocks, from setting up cytoplasmic flows to generating mechanical forces. In particular, transitions between these non-equilibrium states are triggered by regulating the expression and activity of cytoskeletal proteins. However, how these proteins set the large-scale mechanics of the cytoskeleton and drive such non-equilibrium phase transitions remain poorly understood. Here, we show that a minimal network of biopolymers, molecular motors, and crosslinkers exhibits two distinct emergent behaviors depending on its composition, spontaneously flowing like an active fluid, or buckling like an active solid. Molecular motors play a dual antagonistic role, fluidizing or stiffening the network depending on the ATP concentration. By combining experiments, continuum theory, and chemical kinetics, we identify how to assemble materials with targeted activity and elasticity by setting the concentrations of each component. Active and elastic stresses can be further manipulated in situ by light-induced pulses of motor activity, controlling the solid-to-fluid transition. These results highlight how cytoskeletal stresses regulate the self-organization of living matter and set the foundations for the rational design and control of active materials.

Abstract:Living systems naturally exhibit internal driving: active, molecular processes drive non-equilibrium phenomena such as metabolism or migration. Active gels constitute a fascinating class of internally driven matter, in which molecular motors exert localized stresses inside polymer networks. There is evidence that network crosslinking is required to allow motors to induce macroscopic contraction. Yet a quantitative understanding of how network connectivity enables contraction is lacking. Here we show experimentally that myosin motors contract crosslinked actin polymer networks to clusters with a scale-free size distribution. This critical behaviour occurs over an unexpectedly broad range of crosslink concentrations. To understand this robustness, we developed a quantitative model of contractile networks that takes into account network restructuring: motors reduce connectivity by forcing crosslinks to unbind. Paradoxically, to coordinate global contractions, motor activity should be low. Otherwise, motors drive initially well-connected networks to a critical state where ruptures form across the entire network.

Active patterning and asymmetric transport in a model actomyosin network

Abstract:Cytoskeletal networks, which are essentially motor-filament assemblies, play a major role in many developmental processes involving structural remodeling and shape changes. These are achieved by nonequilibrium self-organization processes that generate functional patterns and drive intracellular transport. We construct a minimal physical model that incorporates the coupling between nonlinear elastic responses of individual filaments and force-dependent motor action. By performing stochastic simulations we show that the interplay of motor processes, described as driving anti-correlated motion of the network vertices, and the network connectivity, which determines the percolation character of the structure, can indeed capture the dynamical and structural cooperativity which gives rise to diverse patterns observed experimentally. The buckling instability of individual filaments is found to play a key role in localizing collapse events due to local force imbalance. Motor-driven buckling-induced node aggregation provides a dynamic mechanism that stabilizes the two dimensional patterns below the apparent static percolation limit. Coordinated motor action is also shown to suppress random thermal noise on large time scales, the two dimensional configuration that the system starts with thus remaining planar during the structural development. By carrying out similar simulations on a three dimensional anchored network, we find that the myosin-driven isotropic contraction of a well-connected actin network, when combined with mechanical anchoring that confers directionality to the collective motion, may represent a novel mechanism of intracellular transport, as revealed by chromosome translocation in the starfish oocyte.

Abstract:As animals develop, molecules, cells, and cell ensembles move in beautifully orchestrated choreographies. Movement at each of these scales requires generation of mechanical force. In eukaryotic cells, the actomyosin cytoskeleton generates mechanical forces. Continuous advances in in vivo microscopy have enabled visualization and quantitative assessment of actomyosin dynamics and force generation, within and across cells, in living embryos. Recent studies reveal that actomyosin networks can form periodic waves in vivo . Here, we highlight contributions of actomyosin waves to molecular transport, cell movement, and cell coordination in developing embryos.

Abstract:Stress generation by the actin cytoskeleton shapes cells and tissues. Despite impressive progress in live imaging and quantitative physical descriptions of cytoskeletal network dynamics, the connection between processes at molecular scales and cell-scale spatio-temporal patterns is still unclear. Here we review studies reporting acto-myosin clusters of micrometer size and with lifetimes of several minutes in a large number of organisms ranging from fission yeast to humans. Such structures have also been found in reconstituted systems in vitro and in theoretical analysis of cytoskeletal dynamics. We propose that tracking these clusters can serve as a simple readout for characterising living matter. Spatio-temporal patterns of clusters could serve as determinants of morphogenetic processes that play similar roles in diverse organisms.

Abstract:Biological functions in each animal cell depend on coordinated operations of a wide variety of molecular motors. Some of the these motors transport cargo to their respective destinations whereas some others are mobile workshops which synthesize macromolecules while moving on their tracks. Some other motors are designed to function as packers and movers. All these motors require input energy for performing their mechanical works and operate under conditions far from thermodynamic equilibrium. The typical size of these motors and the forces they generate are of the order of nano-meters and pico-Newtons, respectively. They are subjected to random bombardments by the molecules of the surrounding aqueous medium and, therefore, follow noisy trajectories. Because of their small inertia, their movements in the viscous intracellular space exhibits features that are characteristics of hydrodynamics at low Reynold's number. In this article we discuss how theoretical modeling and computer simulations of these machines by physicists are providing insight into their mechanisms which engineers can exploit to design and control artificial nano-motors.

Abstract:Active stresses, which are collectively generated by the motion of energy-consuming rod-like constituents, generate chaotic autonomous flows. Controlling active stresses in space and time is an essential prerequisite for controlling the intrinsically chaotic dynamics of extensile active fluids. We design single-headed kinesin molecular motors that exhibit optically enhanced clustering, and thus enable precise and repeatable spatial and temporal control of extensile active stresses. Such motors enable rapid, reversible switching between flowing and quiescent states. In turn, spatio-temporal patterning of the active stress controls the evolution of the ubiquitous bend-instability of extensile active fluids and determines its critical length dependence. Combining optically controlled clusters with conventional kinesin motors enables one-time switching from contractile to extensile active stresses. These results open a path towards real-time control of the autonomous flows generated by active fluids.

Abstract:Life is driven by awe-inspiring coordinated movements observed in cells and tissues. In each cell, nm-size molecular motor proteins contribute to these movements as they power numerous mechanical processes with precision and complex orchestration. For the multiple functions that an eukaryotic cell accomplish, motility is essential both at molecular and cellular scales. Tissue morphogenesis, cell migration, cell division or cell differentiation are all controlled by the precise action of such nanomotors that work on cytoskeletal tracks using ATP as fuel. The study of motility has a long history and scientists of all disciplines have contributed to its understanding. The first part of this review compares myosin and kinesin motors to describe the principles underlying how motors convert chemical energy into mechanical movement. In a second part, I will describe how sequence differences selected through evolution can lead to distinct force production output despite a common mechanism. Motors within a superfamily can thus carry out distinct functions in cells. Such differences give rise to their individual, specific motility properties, including reversal of directionality or ability to organize cytoskeletal tracks. The power of structural biology to reveal unexpected and surprising structures, with certainty when visualized at atomic resolution, has been a great advantage for this field. The critical insights gained from the structures can be carefully tested with functional experiments, leading to progress in defining the role motors play in cells. Last, I will describe how targeting these motors can be beneficial for human health. Allosteric sites for specific small molecules can act as activators or inhibitors of the force produced by these nanomotors. While frequent sites of mutations in these motors can lead to disease phenotypes, high therapeutic potential of allosteric effectors is now established for heart muscle diseases and should be extended to treat other pathologies.

Abstract:Synthetic materials can change shape in response to stimuli, with mechanisms reported so far based on the induction of disorder in a pre-organized molecular system. By contrast, harnessing molecular motion by transducing the work of molecular machines is energetically more effective and can mediate functional complexity, as exemplified in biological systems. Here, we show that the power strokes operated by a light-driven molecular motor at the nanoscale can be transduced into the repeated back-and-forth swaying motion of a polymer at the macroscopic length scale. The synchronization of molecular motors, as governed by the energetic landscape of the rotary cycle, is essential to this transduction. Combining synchronization in time with orientation in space allows transducing one molecular rotation into one macroscopic swaying motion using a mechanism that shows analogy with reciprocating pumps. Making materials operate through a variety of sophisticated transduction modes will be critical for the field of autonomous soft robots.

Abstract:Active matter systems evade the constraints of thermal equilibrium, leading to the emergence of intriguing collective behavior. A paradigmatic example is given by motor-filament mixtures, where the motion of motor proteins drives alignment and sliding interactions between filaments and their self-organization into macroscopic structures. After defining a microscopic model for these systems, we derive continuum equations, exhibiting the formation of active supramolecular assemblies such as micelles, bilayers, and foams. The transition between these structures is driven by a branching instability, which destabilizes the orientational order within the micelles, leading to the growth of bilayers at high microtubule densities. Additionally, we identify a fingering instability, modulating the shape of the micelle interface at high motor densities. Mobility of Molecular Motors Regulates Contractile Behaviors of Actin Networks

Abstract:Cells generate mechanical forces primarily from interactions between F-actin, cross-linking proteins, myosin motors, and other actin-binding proteins in the cytoskeleton. To understand how molecular interactions between the cytoskeletal elements generate forces, a number of in vitro experiments have been performed but are limited in their ability to accurately reproduce the diversity of motor mobility. In myosin motility assays, myosin heads are fixed on a surface and glide F-actin. By contrast, in reconstituted gels, the motion of both myosin and F-actin is unrestricted. Because only these two extreme conditions have been used, the importance of mobility of motors for network behaviors has remained unclear. In this study, to illuminate the impacts of motor mobility on the contractile behaviors of the actin cytoskeleton, we employed an agent-based computational model based on Brownian dynamics. We find that if motors can bind to only one F-actin like myosin I, networks are most contractile at intermediate mobility. In this case, less motor mobility helps motors stably pull F-actins to generate tensile forces, whereas higher motor mobility allows F-actins to aggregate into larger clustering structures. The optimal intermediate motor mobility depends on the stall force and affinity of motors that are regulated by mechanochemical rates. In addition, we find that the role of motor mobility can vary drastically if motors can bind to a pair of F-actins. A network can exhibit large contraction with high motor mobility because motors bound to antiparallel pairs of F-actins can exert similar forces regardless of their mobility. Results from this study imply that the mobility of molecular motors may critically regulate contractile behaviors of actin networks in cells.
Hierarchical self-organization of cytoskeletal active networks

Abstract:The structural reorganization of the actin cytoskeleton is facilitated through the action of motor proteins that crosslink the actin filaments and transport them relative to each other. Here, we present a combined experimental-computational study that probes the dynamic evolution of mixtures of actin filaments and clusters of myosin motors. While on small spatial and temporal scales the system behaves in a very noisy manner, on larger scales it evolves into several well distinct patterns such as bundles, asters, and networks. These patterns are characterized by junctions with high connectivity, whose formation is possible due to the organization of the motors in "oligoclusters" (intermediate-size aggregates). The simulations reveal that the self-organization process proceeds through a series of hierarchical steps, starting from local microscopic moves and ranging up to the macroscopic large scales where the steady-state structures are formed. Our results shed light into the mechanisms involved in processes like cytokinesis and cellular contractility, where myosin motors organized in clusters operate cooperatively to induce structural organization of cytoskeletal networks.

Spontaneous motion in hierarchically assembled active matter

Abstract:With exquisite precision and reproducibility, cells orchestrate the cooperative action of thousands of nanometer-sized molecular motors to carry out mechanical tasks at much larger length scales, such as cell motility, division and replication. Besides their biological importance, such inherently non-equilibrium processes are an inspiration for developing biomimetic active materials from microscopic components that consume energy to generate continuous motion. Being actively driven, these materials are not constrained by the laws of equilibrium statistical mechanics and can thus exhibit highly sought-after properties such as autonomous motility, internally generated flows and self-organized beating. Starting from extensile microtubule bundles, we hierarchically assemble active analogs of conventional polymer gels, liquid crystals and emulsions. At high enough concentration, microtubules form a percolating active network characterized by internally driven chaotic flows, hydrodynamic instabilities, enhanced transport and fluid mixing. When confined to emulsion droplets, 3D networks spontaneously adsorb onto the droplet surfaces to produce highly active 2D nematic liquid crystals whose streaming flows are controlled by internally generated fractures and self-healing, as well as unbinding and annihilation of oppositely charged disclination defects. The resulting active emulsions exhibit unexpected properties, such as autonomous motility, which are not observed in their passive analogues. Taken together, these observations exemplify how assemblages of animate microscopic objects exhibit collective biomimetic properties that are starkly different from those found in materials assembled from inanimate building blocks, challenging us to develop a theoretical framework that would allow for a systematic engineering of their far-from-equilibrium material properties.
Molecular motors stiffen non-affine semiflexible polymer networks

Abstract:Reconstituted filamentous actin networks with myosin motor proteins form active gels, in which motor proteins generate forces that drive the network far from equilibrium. This motor activity can also strongly affect the network elasticity; experiments have shown a dramatic stiffening in in vitro networks with molecular motors. Here we study the effects of motor generated forces on the mechanics of simulated 2D networks of athermal stiff filaments. We show how heterogeneous internal motor stresses can lead to stiffening in networks that are governed by filament bending modes. The motors are modeled as force dipoles that cause muscle like contractions. These contractions "pull out" the floppy bending modes in the system, which induces a cross-over to a stiffer stretching dominated regime. Through this mechanism, motors can lead to a nonlinear network response, even when the constituent filaments are themselves purely linear. These results have implications for the mechanics of living cells and suggest new design principles for active biomemetic materials with tunable mechanical properties.

Abstract:Significance Single-molecule techniques have elucidated how isolated molecular motors generate piconewton forces with unprecedented detail. However, in diverse biological and synthetic settings, force-generating proteins collectively power nonequilibrium dynamics, including continuous large-scale rearrangements and persistent fluid flows. Characterizing motor-generated forces in these dense and dynamical environments remains a challenge. We assembled a reversible DNA-based force-sensing probe that, by an optical readout, reveals the molecular arrangements and the force loads experienced by kinesin motors. These probes provide insight into motor-generated forces that collectively power the unique dynamics of microtubule-based active nematics, a noteworthy example of an internally driven active matter system. DNA-based force probes can be extended to study forces and stresses in various synthetic systems as well as diverse cellular environments.

Abstract:Living systems are capable of locomotion, reconfiguration, and replication. To perform these tasks, cells spatiotemporally coordinate the interactions of force-generating, "active" molecules that create and manipulate non-equilibrium structures and force fields that span up to millimeter length scales [1-3]. Experimental active matter systems of biological or synthetic molecules are capable of spontaneously organizing into structures [4,5] and generating global flows [6-9]. However, these experimental systems lack the spatiotemporal control found in cells, limiting their utility for studying non-equilibrium phenomena and bioinspired engineering. Here, we uncover non-equilibrium phenomena and principles by optically controlling structures and fluid flow in an engineered system of active biomolecules. Our engineered system consists of purified microtubules and light-activatable motor proteins that crosslink and organize microtubules into distinct structures upon illumination. We develop basic operations, defined as sets of light patterns, to create, move, and merge microtubule structures. By composing these basic operations, we are able to create microtubule networks that span several hundred microns in length and contract at speeds up to an order of magnitude faster than the speed of an individual motor. We manipulate these contractile networks to generate and sculpt persistent fluid flows. The principles of boundary-mediated control we uncover may be used to study emergent cellular structures and forces and to develop programmable active matter devices.

Abstract:Mixtures of microtubules and molecular motors form active materials with diverse dynamical behaviors that vary based on their constituents' molecular properties. We map the non-equilibrium phase diagram of microtubules and tip-accumulating kinesin-4 molecular motors. We find that kinesin-4 can drive either global contractions or turbulent-like extensile dynamics, depending on the concentrations of both microtubules and a bundling agent. We also observe a range of spatially heterogeneous non-equilibrium phases, including finite-sized radial asters, 1D wormlike chains, extended 2D bilayers, and system-spanning 3D active foams. Finally, we describe intricate kinetic pathways that yield microphase separated structures and arise from the inherent frustration between the orientational order of filamentous microtubules and the positional order of tip-accumulating molecular motors. Our work shows that the form of active stresses and phases in cytoskeletal networks are not solely dictated by the properties of individual motors and filaments, but are also contingent on the constituent's concentrations and spatial arrangement.

A discontinuously coupled network of phase oscillators replicate actomyosin cooperation

Abstract:Groups of non-processive myosin motors exhibit complex and non-linear behaviors when binding to actin. These operate at larger scales and time frames than an individual motor, indicating the presence of a strong cooperative disposition. Limits in contemporary microscopy prevent verification of motor-filament binding dynamics, whilst mathematical models rely on continuum abstractions in which cooperativity is implicit and individual motor behavior cannot be separated from the behaviour of the whole. Understanding the fundamental interactions driving the emergent behaviour in actomyosin therefore remains an open question. Here we suggest that the diversity of empirically observed in-vitro oscillations can be explained by a minimal Kuramoto-style phase oscillator model of actomyosin, where cooperativity is orchestrated by the actomyosin geometry and mechanical environment. The model mirrors the irregular and regular saw-tooth oscillations present in in-vitro actomyosin and sarcomeric 'SPOC' experiments with only adjustments of the external mechanical environment, and despite the model's simplicity. Actomyosin-like behaviour thus arises as a generic property of the discontinuous mechanical coupling in an incommensurate architecture, rather than specific to molecular motor reaction kinetics. We demonstrate a range of synchronising behaviours arising from the cooperative motor dynamics that, once synchronised, are stable over a large range of external forces. The nature of the synchronisation patterns allow recruitment of rotors as the external force increases, reducing variance in the backbone's velocity. This is a demonstration of morphological control. Due to interest in this behaviour in contemporary robotics, we build a physical experiment, using electric motors to power our oscillators. Using the experiment we verify both the organisational and control properties of the system. This demonstrates non-biological motors can cooperate similarly to biological motors when working within an actomyosin geometry, suggesting that the actomyosin complex may not depend on motor-specific qualities to achieve its biological function. These findings offer novel insights into synchronising networks of oscillators and have potential applications in emulating actomyosin-like behaviors within contemporary robotics using non-biological motors.

Rigidity percolation and active advection synergize in the actomyosin cortex to drive amoeboid cell motility

Abstract:Spontaneous locomotion is a common feature of most metazoan cells, generally attributed to the properties of actomyosin networks. This force-producing machinery has been studied down to the most minute molecular details, especially in lamellipodium-driven migration. Nevertheless, how actomyosin networks work inside contraction-driven amoeboid cells still lacks unifying principles. Here, using stable motile blebs from HeLa cells as a model amoeboid motile system, we imaged the dynamics of the actin cortex at the single filament level and revealed the co-existence of three distinct rheological phases. We introduce "advected percolation," a process where rigidity percolation and active advection synergize, spatially organizing the actin network's mechanical properties into a minimal and generic locomotion mechanism. Expanding from our observations on simplified systems, we speculate that this model could explain, down to the single actin filament level, how amoeboid cells, such as cancer or immune cells, can propel efficiently through complex 3D environments.
