Abstract:Combining hydrophobic charge-induction chromatography(HCIC) and expanded bed adsorption(EBA), a novel efficient bioseparation process was developed to purify the immunoglobulin IgY from poultry blood. New hydrophobic charge-induction expanded bed adsorbent S-MMI was prepared with 2-mercapto-1-methyl imidazole(MMI) as the HCIC ligands coupled to quartz-densified agarose matrix for EBA. S-MMI had high ligand density(about 105 -mol-g-1) and the saturated adsorption capacity for IgY reached 71.26 mg·ml-1 at pH 7. S-MMI could form perfectly classified fluidization in expanded bed with high stability. Chicken plasma was precipitated with caprylic acid and the supernatant was used as the feedstock for IgY separation. The loading and elution conditions were optimized by packed bed chromatography. The effects of expansion factors in expanded bed were investigated and the separation conditions were optimized. Finally, IgY was purified from pretreated chicken plasma by EBA with high purity of 98.9% and yield of 80.5%. The combination of HCIC and EBA could achieve efficient separation of immunoglobulin from crude feedstock, which provided a new way for the comprehensive utilization of poultry blood resource.
Purification of Immunoglobulin of Egg Yolk with Hydrophobic Charge Induction Chromatography: Comparison of Operation Modes with Packed Bed and Expanded Bed

Abstract:Immunoglobulin of egg yolk (IgY) was separated from fresh hen eggs by three steps, water dilution, salt-out precipitation, and hydrophobic charge induction chromatography (HCIC). In the purification step, HCIC was used as a novel technology and two chromatographic operation modes, packed bed and expanded bed, were compared to improve the separation efficiency. For packed bed mode, IgY were separated with the purity of 92% and the recovery of 58.4% at the operation condition of pH 7.5 and sample (total protein concentration of 11.6 mg/mL, IgY purity of 31%) loading of 0.2 mL/mL adsorbent. For expanded bed mode, a similar purity of 90% and the recovery of 59.5% were found at the fluid velocity of 500 cm/h and same sample loading of 0.24 mL/mL adsorbent at pH 7.5. The results demonstrated that HCIC in expanded bed mode is a potential platform technology for antibody separation both in high efficiency and low cost.

Abstract:Hydrophobic charge induction chromatography(HCIC)was used to separate immunoglobulin IgY from chicken plasma.The plasma was pretreated with caprylic acid to precipitate some impurities,and the supernatant was used directly as the feedstock for separation and purification of immunoglobulin IgY by HCIC.Two resins,commercial MEP HyperCel and home-made Bestarose-DVS-MMI,were used as HCIC medium and investigated.Operation conditions were optimized,including pH value of sample loading and elution,as well as sample loading volume.It was found that Bestarose-DVS-MMI with 2-mercapto-1-methyl-imidazole as the ligand was better than MEP HyperCel.The results indicated that IgY could be separated with high purity(above 92% with HPLC analysis)and yield about 95%.The mechanism of HCIC was discussed based on comparison of ligand structure and surface potential.The results demonstrated that HCIC can be efficiently used for IgY purification from chicken plasma,which can be a new approach for the immunoglobulin separation from the plasma.

Abstract:Hydrophobic charge-induction chromatography (HCIC) is a novel bioseparation technique, especially for antibody purification. In this study, HCIC was used for the purification of goose immunoglobulin IgY(ΔFc). IgY(ΔFc) is a kind of unique immunoglobulin existing in waterfowl, which natively lacks CH3 and CH4 domains of heavy chains and has some particular therapeutic applications. A new HCIC ligand, 2-mercapto-1-methyl-imidazole (MMI), was coupled to the divinyl-sulfone-activated agarose matrix to prepare the HCIC gel. The adsorption isotherms of IgY(ΔFc) were investigated at different pHs. The saturated capacity of IgY(ΔFc) at pH 5.0 could reach 187.5mg/ml gel, and the pH-dependent adsorption behaviors were found. After the goose plasma was pre-treated with caprylic acid to precipitate some impurities, the supernatant could be directly loaded onto the HCIC column for IgY(ΔFc) separation. The operation conditions were optimized, including the loading pH, elution pH and the loading volume. High separation efficiency was obtained with final purity of 98.6% and the yield of 85.0%. The results indicate that the new process with HCIC can be a promising technology for the cost-effective separation of IgY(ΔFc) from plasma.
Adsorption Behaviors of Avian Immunoglobulins and Purification of Immunoglobulin Y from Chicken Serum with Mixed-Mode Resins

Abstract:The importance of immunoglobulin Y(IgY) as a specific antibody equivalent to mammalian immunoglobulin G(IgG) is well recognized. However, production of highly purified IgY is still difficult due to the lack of specific purification methods. In this study, adsorption behaviors of Ig Y on four mixed-mode resins with functional ligands of 4-mercatoethyl-pyridine(MEP), 2-mercapto-1-methyli-midazole(MMI), 5-aminobenzi-midazole(ABI) and tryptophan-5-aminobenzi-midazole(W-ABI) were evaluated. The results showed that high adsorption ratio were found at p H 6.0–7.0 with little adsorption under acidic conditions. The resin with ABI ligand was then used to separate IgY from immunized chicken serum. An efficient process with Ig Y purity of 95% and recovery of 90% was developed after optimization of loading and elution p H and injection volume. The biological activity of the purified Ig Y was fully maintained. These results indicated that mixed-mode chromatography with specially-designed ligands has great potential for the separation of Ig Y from crude feedstock.

Abstract:Soy protein hydrolysates with lower molecular weight were enzymatically prepared by several commercially available proteases (Alcalase 2.4L, Flavourzyme, Trypsin, Papain, Protease A and Peptidase R) with protein recovery varied from 42.59% to 79.87%. Relative content of positively charged peptides was determined on SP Sephadex C-25 using gradient sodium chloride solution as eluents. Immunomodulating properties were evaluated by measuring their effect on in vitro proliferation of murine spleen lymphocytes and phagocytic activity of peritoneal macrophages. The results showed that soy protein hydrolysates (SPHs) prepared with Alcalase and insoluble soy protein (InSP), preferable to other enzymes and soy proteins, have the highest immunomodulating activity and the optimum conditions were determined as follows: E/S=2% (Alcalase), 60 degrees C, pH 8.0, InSP concentration 6% and 225min. Positive correlations were obtained between the immunomodulating activity and content of positively charged peptides. The results suggested that lower molecular weight and positively charged peptides released from soy protein were effective in stimulating immunomodulating activity, thus provided insights into the preparation of potent immunomodulating products.
Extraction and Purification of IgY against Aeromonas Hydrophila from Egg Yolk

Abstract:Different methods of extracting(water dilution and Pectin) and purifying(saturated ammonium sulfate and PEG 6 000) were conducted to get IgY against Aeromonas Hydrophila. The results showed that remaining ratio of fat and IgY were 32.69% and 68.34% in suspension of egg yolk extracted with 10-floder diluted water at pH 5.0 and the effect was better. Remaining immunocompetence ratio of IgY was 63.5% and 64.1% purified by 12% PEG 6 000(w/v) and 33% saturated ammonium sulfate (v/v) respectively. SDS-PAGE indicated the amount of non-IgY protein significantly decreased by water dilution and Pectin methods,and there were no other proteins except IgY in supernatants with PEG 6 000 and saturated ammonium sulfate as precipitants. Thin paper showed water dilution and Pectin methods provided a simple,rapid and efficient means of extracting crude IgY,and PEG 6 000 and saturated ammonium sulfate were suitable to purify IgY in suspension.

Abstract:Chicken egg whites contain hundreds of proteins, and are widely used in the food, biological and pharmaceutical industries. It is highly significant to study the separation and purification of egg white proteins. This review first describes the structures and functional properties of several major active proteins in egg whites, including ovalbumin, ovotransferrin, ovomucoid, lysozyme, ovomucin, ovomacroglobulin and avidin. Then, the common techniques (including precipitation, chromatography and membrane separation) and some novel approaches (including electrophoresis, membrane chromatography, aqueous two-phase system and molecular imprinting technology) for the separation and purification of egg white proteins broadly reported in the current research are introduced. In addition, several co-purification methods for simultaneous separation of multiple proteins from egg whites have been developed to improve raw material utilization and reduce costs. In this paper, the reported techniques in the last decade for the separation and purification of chicken egg white proteins are reviewed, discussed and prospected, aiming to provide a reference for further research on egg proteins in the future.

Crude Extraction and Purification of IgY Against Aeromonas Hydrophila and Its Antibacterial Activity in Vitro

Abstract:Using water-dilution to extract crude IgY from egg yolk of hens immunized with an inactivated Aeromonas hydrophila and then introducing saturated ammonium sulfate solution to purify the IgY from crude extract, this study evaluated the immunocompetence recovery and antibacterial activity of IgY in vitro. The results showed that the immunocompetence recovery were 78.34% in the crude extraction suspension with 10×distilled water dilution at pH 5.0 and 64.1% in purification suspension with 33%(v/v) saturated ammonium sulfate solution as precipitates. The purified IgY was of high purity as shown by SDS-PAGE. The crude extraction of IgY abolished the growth of A. hydrophila at a concentration of 1.0 mg/mL within 18 hours. In conclusion, the specific IgY from immunized hens could provide a novel alternative approach to prevention and control of diseases caused by A. hydrophila infection in several aquatic species.

Abstract:Pepsin, trypsin and proteinase K were used in the present study to hydrolyse the proteins from whole eggs, yolks or whites, and the resulting hydrolysates were characterised in terms of antioxidant and IgE-binding properties, using a combination of in vitro and in silico methods. Based on the degree of hydrolysis (DH) results, the egg yolk proteins are better substrates for all the tested enzymes (DH of 6.2–20.1%) compared to those from egg whites (DH of 2.0–4.4%). The SDS-PAGE analysis indicated that pepsin and proteinase K were more efficient compared to trypsin in breaking the intramolecular peptide bonds of the high molecular weight egg proteins. For all the tested substrates, enzyme-assisted hydrolysis resulted in a significant increase in antioxidant activity, suggesting that many bioactive peptides are encrypted in inactive forms in the parent proteins. The hydrolysates obtained with proteinase K exhibited the highest DPPH radical scavenging activity (124–311 μM Trolox/g protein) and the lowest residual IgE-binding capacity. The bioinformatics tools revealed that proteinase K is able to break the integrity of the main linear IgE-binding epitopes from ovalbumin and ovomucoid. It can be concluded that proteinase K is a promising tool for modulating the intrinsic properties of egg proteins.

Abstract:Lecthin from duck eggs was extracted by using ethanol.The optimum extraction conditions were determined by orthogonal experiment and obtained 80% rough lecthin from duck egg yolk.Rough lecthin purification was done by Zncl2 precipitator and recovery of refined lecthin was 90%.The TLC results showed one clear PC spot.

Abstract:Poor rehydration is one of the key factors affecting the functional properties of egg white powder (EWP). Reducing rehydrated precipitates is important for the processing and application of EWP. In this study, effects of ultrasonic pretreatment on the physicochemical and functional properties of EWP rehydration solutions were studied with the aim of revealing the mechanism of ultrasonic pretreatment to improve rehydration. Compared with freeze-dried EWP (FD) and spray-dried EWP (SD), the percentage of ultrasonic pretreated FD (UFD) and ultrasonic pretreated SD (USD) rehydrated precipitates decreased by 13.0 % and 5.6 %, respectively, after ultrasonic pretreatment (0.25 W/mL for 10 min); and the average particle sizes of UFD and USD solutions decreased by 22.5 % and 15.5 %, respectively. Fourier transform infrared spectroscopy showed that ultrasonic pretreatment caused higher β-sheet content in the protein secondary structure of UFD rehydrated precipitates (49.2 %). Quantitative proteomic analysis revealed a decrease in the abundance of major egg white proteins (ovalbumin, ovotransferrin, ovomucoid and ovomucin) in the rehydrated precipitates of UFD, except for lysozyme. It was also shown that lysozyme-centered aggregates were disrupted in the rehydrated precipitates of UFD. Our research suggests that ultrasonic pretreatment improves EWP rehydration by reducing the interactions between high abundance proteins as well as improving the solubility of high abundance proteins.

Abstract:Egg yolk is an excellent protein source for the production of bioactive peptides. However, the recent method need to remove lipid first which involves wastage and pollution of organic reagents. Therefore, the process of directly using oily yolk powder to prepare egg yolk peptides has attracted much attention. This study developed a one-step process to simultaneously extract oil and hydrolyze proteins based on an ultrasound coupled sodium sulfite pretreatment (UCSSP) assisted enzymatic hydrolysis for egg yolk powder. Results showed that UCSSP increased the oil extraction rate from zero to 75 % with 59.35 g/L of soluble protein and 33.99 g/L of peptide. Further analysis of the underlying mechanism demonstrated that ultrasound pre-treatment could change the secondary structure of EYP while sodium sulfite pre-treatment softened the protein and induced more hydrophobic groups exposed, thus inducing more lipoprotein released for hydrolysis. In addition, the proportion of peptides ranging from 180 Da to 3000 Da in the UCSSP group increased from 31.19 % before to 79.74 %, which was 31.27 % and 6.16 % higher than that of UP and SP. Furthermore, the obtained peptides showed obvious activities in uric acid-lowering, anti-obesity and antioxidant with 56.24 % inhibition in XOD activity and close antioxidant activity to vitamin C, implying it a potential health product.
A pivotal peptide (Ile-Leu-Lys-Pro) with high ACE- inhibitory activity from duck egg white: identification and molecular docking

Abstract:Salted duck egg white was desalted and hydrolyzed to produce angiotensin I-converting enzyme (ACE) inhibitory peptides. Single factor test and response surface design were performed to determine the best hydrolysis conditions: enzyme dosage 11875.99 U/g, substrate concentration 33.04 g/L and hydrolysis time 4 h. The fraction V (MW < 1 kDa), which exhibited the strongest ACE inhibitory activity, was characterized by HPLC-ESI-MS/MS. Eighty-three peptides were identified, and among them Ile-Leu-Lys-Pro, Ile-Asn-Ser-Trp, Ile-Arg and His-Pro-Ala were synthesized for further research. Ile-Leu-Lys-Pro exhibited the highest ACE inhibitory activity (IC50: 0.355 mM). The molecular docking studies revealed that nine amino acids contributed to stabilize the docking complex. The ACE inhibition of Ile-Leu-Lys-Pro and Ile-Asn-Ser-Trp were mainly attributed to Ile in N-terminal. The residues Glu362 and Ala332 were the important binding sites in molecular docking. This research expands the understanding of ACE inhibitory peptides from duck egg white as well as highlights an opportunity for recycling an otherwise discarded byproduct.

Simple Two-step Chromatographic Method for Purification of Ovomacroglobulin

Abstract:In the early part of last century, the ability of chicken egg white to inhibit trypsin was recognized and was attributed to ovomucoid. Since then, at last four different proteins of the avian egg white were known to possess proteinase inhibitory activity i.e., ovomucoid, ovoinhibitor, cystatin and ovomacroglobulin. Different from ovomucoid and ovoinhibitor, ovomacroglobulin inhibits proteinases of all four mechanistic classes. Besides, it has many kinds of biological activities, as antibacterial, inhibiting sepsis, accelerate wound healing and antiinflammatory. It completely inhibited the activity of Serratia and Pseudomonas and reduced corneal destruction in experimental keratitis in rabbits. Ovomacroglobulin can inhibit the generation of bradykinin through preventing bacterial protease activates Hageman factor-kinin system. More noteworthy, ovomacroglobulin are highly homologous with α2-macroglobulin (α2M) , so, it may be it has more important biological activities that like α2M. However, the purification of ovomacroglobulin is difficult because of its low content in egg white (only 0.5 % of egg white protein), which limited its research in bioactivity. In the previous research, Nagase et al. purified ovomacroglobulin by polyethylene glycol precipitation from egg white following by chromatography on Ultragel AcA 34, DEAE-cellulose and Sephacryl S-300 columns. Comparative Study on Foaming Properties of Egg White with Yolk Fractions and Their Hydrolysates

Abstract:As an excellent foaming agent, egg white protein (EWP) is always contaminated by egg yolk in the industrial processing, therefore, decreasing its foaming properties. The aim of this study was to simulate the industrial EWP (egg white protein with 0.5% w/w of egg yolk) and characterize their foaming and structural properties when hydrolyzed by two types of esterase (lipase and phospholipase A2). Results showed that egg yolk plasma might have been the main fraction, which led to the poor foaming properties of the contaminated egg white protein compared with egg yolk granules. After hydrolyzation, both foamability and foam stability of investigated systems thereof (egg white protein with egg yolk, egg white protein with egg yolk plasma, and egg white protein with egg yolk granules) increased significantly compared with unhydrolyzed ones. However, phospholipids A2 (PLP) seemed to be more effective on increasing their foaming properties as compared to those systems hydrolyzed by lipase (LP). The schematic diagrams of yolk fractions were proposed to explain the aggregation and dispersed behavior exposed in their changes of structures after hydrolysis, suggesting the aggregated effects of LP on yolk plasma and destructive effects of PLP on yolk granules, which may directly influence their foaming properties.

Hydrolysis of Duck Egg White and Effect of Hydrolysates on Erythrocyte Immune Function in Rats

Abstract:In this study,the duck egg white was desugarized by yeast,then hydrolyzed by alkaline protease and animal proteolysis compoud enzymes.The orthogonal experiment was designed to see the best hydrolysis condition of animal pro-teolysis compoud enzymes.The results showed that with 5% substrate concentration,the best condition of hydrolysis was E/S 0.7%,pH8.0,temperature 54℃,reactiontime 4 hours,the DH value was 92.65%.The hydrolysates didn’t need to be debittered,desalinated and decolorated.And the effect of hydrolysates on erythrocyte immune function was studied.It was found that the rosette rates of red blood cell C3b receptor of experiment goup were significantly higher than control group,but the rosette rates of red blood cell immune complexes of the two groups were not significantly different.The conclusion was that the hydrolysates could enhance erythrocyte immune function in rats.

Abstract:Avian egg albumen participates in embryonic development by providing essential nutrients as well as antimicrobial protection. Although various biological functions of egg white proteins were suggested during embryogenesis, global changes of these proteins under incubation conditions remained uninvestigated. This study presents a proteomic analysis on the change of egg white proteins during the first week of embryonic development. By using 2-DE, together with MALDI-TOF MS/MS, thirty protein spots representing eight proteins were identified showing significant changes in abundance during incubation. An accelerating degradation of ovalbumin was observed in a wide range of molecular weight. In addition, four protein complexes were predicted according to the detected molecular weight increase. Among these speculated protein complexes, an ovalbumin spot coupled with RNA-binding protein was detected. The absence of these protein complexes before incubation, followed by the constant increase in abundance during incubation indicates conceivable pivotal roles in embryonic development. To better understand the function of the proteins identified in this study, discrepancies of egg white protein changes between fertilized and unfertilized chicken eggs were additionally demonstrated. These findings will provide insight into the embryogenesis process to improve our knowledge of egg white proteins in regulating and supporting early embryonic development.

Abstract:The antioxidant peptide was prepared from hydrolyzed egg of Apostichopus japonicus. Ultra-filtration, high-speed counter-current chromatography and gel filtration were used in peptide purification. In each step of purification, meanwhile, peptide was tested with its antioxidant ability. The purity and molecular weight was tested by using Tricine-sodium dodecyl sulphate-polyacrylamide gel electrophoresis. Results showed that the purified peptide obtained a sharply enhancing ability to scavenge hydroxyl radical from 37 to 89.82 U/mL. The purity of peptide was conformed and molecular weight was estimated about 30 KDa. It would be promising for purified peptide to be a good functional food resource.
