Abstract:Programmed cell death ligand-1 (PD-L1) expression levels in patients' tumors have demonstrated clinical utility across many cancer types and are used to determine treatment eligibility. Several independently developed PD-L1 immunohistochemical (IHC) predictive assays are commercially available and have demonstrated different levels of staining between assays, generating interest in understanding the similarities and differences between assays. Previously, we identified epitopes in the internal and external domains of PD-L1, bound by antibodies in routine clinical use (SP263, SP142, 22C3, and 28-8). Variance in performance of assays utilizing these antibodies, observed following exposure to preanalytical factors such as decalcification, cold ischemia, and duration of fixation, encouraged additional investigation of antibody-binding sites, to understand whether binding site structures/conformations contribute to differential PD-L1 IHC assay staining. We proceeded to further investigate the epitopes on PD-L1 bound by these antibodies, alongside the major clones utilized in laboratory-developed tests (E1L3N, QR1, and 73-10). Characterization of QR1 and 73-10 clones demonstrated that both bind the PD-L1 C-terminal internal domain, similar to SP263/SP142. Our results also demonstrate that under suboptimal decalcification or fixation conditions, the performance of internal domain antibodies is less detrimentally affected than that of external domain antibodies 22C3/28-8. Furthermore, we show that the binding sites of external domain antibodies are susceptible to deglycosylation and conformational structural changes, which directly result in IHC staining reduction or loss. The binding sites of internal domain antibodies were unaffected by deglycosylation or conformational structural change. This study demonstrates that the location and conformation of binding sites, recognized by antibodies employed in PD-L1 diagnostic assays, differ significantly and exhibit differing degrees of robustness. These findings should reinforce the need for vigilance when performing clinical testing with different PD-L1 IHC assays, particularly in the control of cold ischemia and the selection of fixation and decalcification conditions.
[Comparison of two different tissue decalcification methods for immunohistochemistry].

Abstract:OBJECTIVE:To investigate the effects of 1% hydrochloric acid and 10% nitric acid on immunohistochemical staining for several antigens. METHODS:Fifteen cases of focally calcified soft tumors including three cases of thyroid papillary carcinoma, three of breast invasive ductal carcinoma, three of prostate cancer, three of colon adenocarcincoma and three of normal lymph nodes, were selected in this study, and so were nine cases of bone and bone marrow tissue from biopsy and autopsy. Tissue samples were divided into three groups. One group was each soaked into 10% nitric acid for 24, 42 and 48 h, another group was each incubated in 1% hydrochloric acid for 1, 2 and 3 h. The other group which was not treated with acid was regarded as control. Then immunohistochemical staining was conducted in these samples for antigens as follows: ER, PR and Ki-67 in breast invasive ductal carcinoma; TTF-1 in thyroid papillary carcinoma; 34βE12 and P504s in prostate cancer; AE1/AE3, P504s and Ki-67 in colon adenocarcincoma; CD3, CD20 and Ki-67 in lymph nodes; CD3,CD20,CD68,CD235a,MPO,Ki-67,AE1/AE3 and TTF-1 in bone tissue. RESULTS:After the tissues decalcificated in both acid solutions for some time, the numbers of positive cells decreased and the intensity of immunostaining became weaker, which was proportionate to time. Bone tissue was less influenced than calcified soft tissue. Different antigens showed different sensitivity to the same acid treatment. TTF-1 and Ki-67 were the two most vulnerable antigens among them; while ER, CD3 and CD20 were the least influenced. The effects of decalcification on immunostaining of one antigen were the same in different tissues. CONCLUSION:In most cases, both hydrochloric acid and nitric acid decalcification can lower the sensitivity of immunohistochemical staining, but have different effects on different antigens. It is better to use 10% nitric acid for decalcification of relatively large bone tissue and to use 1% hydrochloric acid for bone marrow tissues and bone tissues containing much more soft tissue.
Evaluation of Decalcification Techniques for Rat Femurs Using HE and Immunohistochemical Staining

Abstract:Aim . In routine histopathology, decalcification is an essential step for mineralized tissues. The purpose of this study is to evaluate the effects of different decalcification solutions on the morphological and antigenicity preservation in Sprague Dawley (SD) rat femurs. Materials and Methods . Four different decalcification solutions were employed to remove the mineral substances from rat femurs, including 10% neutral buffered EDTA, 3% nitric acid, 5% nitric acid, and 8% hydrochloric acid/formic acid. Shaking and low temperature were used to process the samples. The stainings of hematoxylin-eosin (HE) and immunohistochemical (IHC) were employed to evaluate the bone morphology and antigenicity. Key Findings . Different decalcification solutions may affect the quality of morphology and the staining of paraffin-embedded sections in pathological examinations. Among four decalcifying solutions, 3% nitric acid is the best decalcifying agent for HE staining. 10% neutral buffered EDTA and 5% nitric acid are the preferred decalcifying agents for IHC staining. Significance . The current study investigated the effects of different decalcifying agents on the preservation of the bone structure and antigenicity, which will help to develop suitable protocols for the analyses of the bony tissue.

Optimization of bleaching and immunostaining techniques on heavily pigmented choroidal tumor specimens

Abstract:Aim:To optimize the method of bleaching and immunostaining on heavily pigmented choroidal tumor specimens,using the samples from an extremely heavy-pigmented choroidal melanoma arising from melanocytoma case.Method:Formalin fixed and paraffin embedded tissue were processed with or without postfixation and bleached by potassium permanganate/oxalic acid at different concentrations,followed by immunohistochemistry for Ki-67.Result:Those sections that were postfixed by paraformldehyde or formalin,then bleached with 1% potassium permanganate/2%oxalic acid showed satisfying result on HE staining.Antigen retrieval and postfixation before bleaching helped to achieve complete bleaching and maintain the antigenicity of tissue and integrity of sections.Conclusion:For extremely heavy-pigmented choroidal tumor,postfixation before bleaching or after antigen retrieval,and processing with higher concentration of potassium permanganate/oxalic acid,result in complete bleaching,good tissue antigenicity and section integrity.
Comparative study of three different decalcifying solutions in bone metastasis specimens with breast cancer

Abstract:Objective To investigate the optimal strategy for immunohistochemical (IHC) staining in bone metastasis specimens from breast cancer.Methods Twenty-eight bone metastases specimens from breast cancers were divided into three groups and subjected to different decalcifying agents (group A-10% nitrate, group B-EDTA decalcification, and group C-imported decalcifying solution RapidCal).The effects of those on HE and IHC staining for Ki-67, ER, PR, GATA3, RANK, RANKL, HER2 and HER2 FISH results were assessed.Results There were no significant differences among three groups in HE morphology and IHC staining.Antigen content in the RapidCal group were all intact;the EDTA group showed a similar staining rate, which was better than the nitrate group (P<0.05).Nitrate group showed marked reduction in nuclear Ki-67 staining, but the loss of cytoplasmic antigens (RANK, RANKL) was less than cell membrane antigen (HER2).For FISH, the RapidCal group and EDTA group showed same results, concordant with IHC staining results.The expression of HER2 protein in the nitric acid group was significantly decreased and chromosome 17 labelling was lost (P<0.05).Conclusions RapidCal treated bone metastases specimens from breast cancer show excellent sample quality in morphological, IHC and FISH results compared with traditional decalcifying agents.Owing to the longer time of EDTA decalcification, the new decalcifying agent RapidCal plays an important role in quality control and clinical application.

Abstract:Objective:To observe the relationship between effects of ganoderma lucidum polysaccharides(GLP) on learning and memory in Alzheimer's disease(AD) rats and apoptosis of the hippocampus cells.Methods: The AD pathological model was made by hippocampal CA1 lesions due to stereotaxic mini-injection of Aβ 25-35,and the learning and memory of rats were observed by Morris water maze;the apoptosis of hippocampal cells were detected by using flow cytometry and electronic microscope.Results: Compared with the control group,GLP was shown to improve learning and memory in rats with bilateral hippocampal lesions,and to decrease apoptosis rate in hippocampal cells significantly.Conclusion: These results suggest that the protective effects of GLP on AD rats may be related to its ability to decrease the apoptosis rate of hippocampal cells.

Abstract:Objective:To observe the effect of fixative,mounting medium,temperature and sterilized method to three waves quantum dots(QDs)served as a mark on the peritoneal macrophages and normal skin of mice.Methods:Three different waves QDs:610nm,523nm and 576nm,and peritoneal macrophages and normal skin of mice as the carriers were utilized to investigate the effects of different fixative,mounting medium,temperature and sterilized method on cells and tissues.Results:There was no obvious toxicity about these three kinds of QDs to peritoneal macrophages and skin tissue of mice,and they could keep no fading in macrophages and skin tissue for a long time(6-72h).In temperature higher than 56℃,QDs would lose the property of fluorescence.Better sterilized method was ~(60)Co irradiation and micro-pore film filtering.The most stable mounting medium was hydro-soluble ClearmountTM and glycerin buffer.Macrophage which swallowed QDs 523,could be fixed by various fixatives.Conclusion:The functions of peritoneal macrophages and normal skin tissue marked by quantum dots with different waves could be affected by fixative,mounting medium,temperature and sterilized method.
Effects of Prefixation and Fixation Times on Apoptosis Detection by in Situ End-Labeling of Fragmented DNA.

Abstract:Objective.-Apoptosis is considered to play an important role in the pathogenesis and progression of neoplasia. An in situ 3'-end DNA labeling (TUNEL) method was recently developed and has been widely used to identify apoptotic cells in tissue sections. However, sometimes the TUNEL method labels many more cells than expected. We investigated the effects of prefixation time and fixation time on the apoptotic index detected by this method.Materials and Methods.-Using the spleen and thymus of rats, the effects of prefixation time (0, 1, 2, 4, 6, 12, 24, 48, and 72 hours) at 4 degrees C and fixation time (6, 12, 24, 48, 72, and 96 hours; i, 2, and 3 weeks) on the apoptotic index were examined by the TUNEL method. Agarose gel electrophoresis of extracted DNA from the specimens of each prefixation time was also performed.Results.-In comparison with control tissue (no prefixation time), which showed scattered positive cells with distinct staining restricted to the nucleus, the splenic tissue unfixed for 2 hours or more and the thymic tissue unfixed for 4 hours or more showed cytoplasmic staining in the positive cells. Moreover, as the prefixation time was prolonged, the number of positive cells gradually increased. Agarose gel analysis of DNA extracted from tissue sections left unfixed longer than 24 hours showed a ladder pattern consisting of multiples of about 200 base pairs.Conclusions.-Two hours was the limit of prefixation time for the precise identification of apoptosis by the TUNEL method. The false-positive cells in tissue sections left unfixed for longer time intervals may have been due to internucleosomal DNA cleavage following necrosis. In contrast, the length of fixation time in buffered formalin seemed to have no effect on the results obtained by this method.
Computer-assisted three-dimensional quantitation of programmed death-ligand 1 in non-small cell lung cancer using tissue clearing technology

Abstract:Abstract Immune checkpoint blockade therapy has revolutionized non-small cell lung cancer treatment. However, not all patients respond to this therapy. Assessing the tumor expression of immune checkpoint molecules, including programmed death-ligand 1 (PD-L1), is the current standard in predicting treatment response. However, the correlation between PD-L1 expression and anti-PD-1/PD-L1 treatment response is not perfect. This is partly caused by tumor heterogeneity and the common practice of assessing PD-L1 expression based on limited biopsy material. To overcome this problem, we developed a novel method that can make formalin-fixed, paraffin-embedded tissue translucent, allowing three-dimensional (3D) imaging. Our protocol can process tissues up to 150 μm in thickness, allowing anti-PD-L1 staining of the entire tissue and producing high resolution 3D images. Compared to a traditional 4 μm section, our 3D image provides 30 times more coverage of the specimen, assessing PD-L1 expression of approximately 10 times more cells. We further developed a computer-assisted PD-L1 quantitation method to analyze these images, and we found marked variation of PD-L1 expression in 3D. In 5 of 33 needle-biopsy-sized specimens (15.2%), the PD-L1 tumor proportion score (TPS) varied by greater than 10% at different depth levels. In 14 cases (42.4%), the TPS at different depth levels fell into different categories (< 1%, 1–49%, or ≥ 50%), which can potentially influence treatment decisions. Importantly, our technology permits recovery of the processed tissue for subsequent analysis, including histology examination, immunohistochemistry, and mutation analysis. In conclusion, our novel method has the potential to increase the accuracy of tumor PD-L1 expression assessment and enable precise deployment of cancer immunotherapy.

Effects of Maxingloushi decoction on immune inflammation and programmed death markers in mice with chronic obstructive pulmonary disease

Abstract:Background: To investigate effects of Maxingloushi decoction on lung inflammation and programmed death markers (programmed death-1 [PD-1], programmed death-ligand 1 [PD-L1]) in the lung tissue, peripheral blood, and bronchoalveolar lavage fluid (BLF) in a mouse model of chronic obstructive pulmonary disease (COPD). Methods: Thirty-six mature male BALB/C mice were randomly divided into normal group (group A, n=6), COPD model group (group B, n=10), Maxingloushi decoction + COPD group (group C, n=10), and PD-1 inhibitor + COPD group (group D, n=10). The COPD model was established by smoke inhalation combined with lipopolysaccharide (LPS). Levels of PD-1 and PD-L1 in plasma and BLF were measured by enzyme-linked immunosorbent assay (ELISA). Histopathological techniques were used to semi-quantitatively analyze the immuno-fluorescence optical density (IOD) value of the lung tissue. Results: In plasma and BLF, the expression of PD-1 in the group B was higher than that in the group A, and the expression of PD-L1 was lower than that in the group A. The expression of PD-1 and PD-L1 in the lung tissue was normalized in the group C in comparison with the group B (P<0.05) and the group D (P<0.05), and inflammatory cell infiltration in the lung tissue was also improved. Conclusions: These findings reveal that COPD causes an immune imbalance in the peripheral blood and lung tissue, and that both Maxingloushi decoction and PD-1 inhibitor treatment can mitigate lung inflammation in COPD by reducing PD-1 expression and increasing PD-L1 expression. The treatment effect of Maxingloushi decoction may be superior to that of PD-1 inhibitor.
The Effect of Postconditioning on Apoptosis-Related Gene Expression of Lung Tissues in Rat Intestinal Ischemia-Reperfusion

Abstract:AIM: To investigate the effect of postconditioning on apoptosis-related gene expression of lung tissues in rat intestinal ischemia-reperfusion( I/R) . METHODS: Male Sprague-Dawley rats were divided into 3 groups which were sham operation group ( sham) ,I/R group and ischemic postconditioning ( IPOST) group. The experimental animals were sacrificed after reperfusion for 6 h. The tissue sections were stained with hematoxylin and eosin and examined under light microscope to observe lung histopathological changes. Lung W/D ratios were measured. Lung tissue samples were taken to detect Bcl-2,Bax and caspase-3 mRNA and protein expression using RT-PCR and Western blotting analysis,respectively. RESULTS: Compared with I/R group,the morphological changes of lung were alleviated and the lung W/D ratios significantly decreased in IPOST group ( P 0. 05) . The mRNA and protein expression of Bcl-2 in IPOST group was obviously higher than that in I/R group ( P 0. 05) ,while the mRNA and protein expression of Bax and caspase-3 in IPOST group was obviously higher than that in I/R group ( P 0. 05) . CONCLUSION: Ischemic postconditioning attenuated intestinal ischemia-reperfusion-induced acute lung injury partly by increasing Bcl-2 mRNA and protein expression,reducing Bax mRNA and protein expression.
Influence of fixation and decalcification on the immunohistochemical staining of cell-specific markers in paraffin-embedded human bone biopsies.

Abstract:A number of fixation and decalcification procedures were evaluated to determine their suitability for immunohistochemistry on trephine samples of bone marrow after paraffin embedding. In particular, the immunoreactivity of antigens characteristic for various hematopoietic cell lines (immunoglobulin heavy and light chains for plasmacytoid cells; elastase for neutrophil myeloid cells; lysozyme, alpha-1-antitrypsin and alpha-1-antichymotrypsin for hystiocytic cells; leukocyte common antigen for lymphocytes; hemoglobin and glycophorin A for erythroid cells; Factor VIII-related antigen for thrombocytoid cells) as well as some antigens specific for epithelial tumors (CEA, 115D8, and keratin) were investigated. Fixation in a mercuric chloride-formaldehyde mixture followed by decalcification in acetic acid-formaldehyde-saline proved to be the best procedure for antigen preservation and retention of morphologic detail. Moreover, there is no need of trypsinization when using this procedure. The only exception was Factor VIII-related antigen in megakaryocytes, which was best demonstrated in trypsin-digested sections of formalin-fixed and acetic acid-decalcified biopsies.

[An Experimental Study on Destruction of K562 and HL60 Induced by 5-Aminolaevulinic Acid-Based Photodynamic Therapy].

Abstract:This experiment was designed to explore the pattern of K562 and HL60 leukemia cells death, the effects on their cell cycle and the cytoplasmic free calcium concentration ([Ca2+]i) induced by 5-aminolaevulinic acid-based photodynamic therapy (ALA-PDT). Under the transmission electron microscope (TEM), two kinds of leukemia cells' ultrastructure were observed. Flow cytometry combined with Annexin V-FITC/PI labeling was used to detect the pattern of K562 and HL60 cells' death induced by ALA-PDT. Flow cytometry combined with PI labeling was used to analyze the change in the cell cycle induced by ALA-PDT, and confocal laser scanning microscopy (CLSM) combining with calcium fluorescence probe was used to detect the change in the cytoplasmic free calcium concentration ([Ca2+]i). Immediately after irradiation, many typical apoptotic bodies were seen in the cells treated. Most of the cells treated were necrotic at 24 hours following irradiation. Flow cytometry analysis suggested that the main patterns of the cells' death were apoptosis immediately after irradiation and necrosis post-apoptosis at 24 hours post irradiation. Immediately and 24 hours after irradiation, the proportion of S phase of K562 was 57. 67% +/- 1.13% and 84.77% +/- 6.20% respectively, and the proportion of S phase of HL60 was 74.60% +/- 7.27% and 84.60% 1.74% respectively. Both [Ca+]i of the treated K562 and HL60 were increased obviously. In the best experiment condition, the initial pattern of the K562 and HL60 leukemia cells' death induced by PDT was apoptosis and the main pattern was necrosis post apoptosis. The two kinds of cells were arrested at S phase by ALA-PDT. During the death of the leukemia cells, the increase in intracellular free calcium concentration could be responsible for the ALA photodynamically induced damage to K562 and HL60 cells.

Abstract:Background: The infiltration and activation of immune cells in the tumor microenvironment (TIME) affect the prognosis of patients with cancer. Tertiary lymphoid structure (TLS) formation favors tumour- infiltrating-lymphocyte (TIL) recruitment and is regarded as an important indicator of good prognosis associated with immunotherapy in patients with tumors. Chemotherapy is currently one of the most commonly used clinical treatment methods. However, there have been no clear report to explore the effects of different types of chemotherapy on TLS formation in the TIME. This study examined the effects of immunogenic cell death (ICD)-inducing chemotherapeutics on immune cells, high-endothelial venules (HEV), and TLSs in mouse melanomas. Methods: Doxorubicin (an ICD inducer), gemcitabine (non-ICD inducer), and a combination of the two drugs was delivered intra-peritoneally to B16F1-loaded C57BL/6 mice. The infiltration of immune cells into tumor tissues was evaluated using flow cytometry. HEV and TLS formation was assessed using immunohistochemistry and multiple fluorescent immunohistochemical staining. Results: Doxorubicin alone, gemcitabine alone, and the two-drug combination all slowed tumor growth, with the combined treatment demonstrating a more pronounced effect. Compared with the control group, the doxorubicin group showed a higher infiltration of CD8 + T cells and tissue-resident memory T cells (T RM ) and an increase in the secretion of interferon-γ, granzyme B, and perforin in CD8 + T subsets and activation of B cells and dendritic cells. Doxorubicin alone and in combination with gemcitabine decreased regulatory T cells in the TIME. Moreover, doxorubicin treatment promoted the formation of HEV and TLS. Doxorubicin treatment also upregulated the expression of programmed cell death protein (PD)-1 in CD8 + T cells and programmed cell death protein ligand (PD-L)1 in tumor cells. Conclusions: These results indicate that doxorubicin with an ICD reaction promotes TLS formation and increases PD-1/PD-L1 expression in tumor tissues. The results demonstrate the development of a therapeutic avenue using combined immune checkpoint therapy.

Abstract:AIM: To investigate the effects of photodynamic therapy with quantum dots-arginine-glycine-aspartic acid (RGD) probe as photosensitizer on the proliferation and apoptosis of pancreatic carcinoma cells.METHODS: Construction of quantum dots-RGD probe as photosensitizer for integrin-targeted photodynamic therapy was accomplished. After cells were treated with photodynamic therapy (PDT), the proliferation of SW1990 cells were measured by methyl thiazolyl tetrazolium assay. Morphologic changes, cell cycle retardance and apoptosis were observed under fluoro-scope and flow cytometry. The expression of myeloid cell leukemia-1 (Mcl-1), protein kinase B (Akt) and tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) mRNA were detected by reverse transcriptionpolymerase chain reaction. The amount of reactive oxygen species were also evaluated by fluorescence probe.RESULTS: The photodynamic therapy with quantum dots-RGD probe as photosensitizer significantly inhibited cell proliferation (P < 0.01). Apoptotic cells and morphologic changes could be found under optical microscope. The FCM revealed PDT group had more significant cell apoptosis rate compared to control cells (F = 130.617, P < 0.01) and cell cycle G(0)/G(1) and S retardance (P < 0.05) compared to control cells. The expression of Mcl-1 and Akt mRNA were down-regulated, while expression of TRAIL mRNA was up-regulated after cells treated with PDT. PDT group had more significant number of cells producing reactive oxygen species compared to control cells (F = 3262.559, P < 0.01).CONCLUSION: The photodynamic therapy with quantum dots-RGD probe as photosensitizer significantly inhibits cell proliferation and increases apoptosis in SW1990 cells. Effects of prolonged formalin fixation on the immunohistochemical detection of infectious agents in formalin-fixed, paraffin-embedded tissues

Abstract:Immunohistochemistry is commonly used to detect and characterize infectious agents in diagnostic pathology. The principal advantage of immunohistochemistry over other antigen detection techniques is the ability to identify antigen within the context of histologic lesions. Although epitope masking attributed to formalin fixation, especially prolonged fixation, has been considered a limiting factor in diagnostic immunohistochemistry, only a few studies have evaluated the immunohistochemical detection of infectious agents following prolonged formalin fixation. Therefore, the goal of this study was to evaluate the effects of prolonged formalin fixation on the immunohistochemical detection of 21 infectious agents. Tissue slices about 5 mm thick were fixed in 10% neutral-buffered formalin, processed, and paraffin embedded at day 1 or 2 and then at approximately weekly intervals. Three pathologists graded immunoreactivity according to a four-tier grading system: negative, weak, moderate, strong. Canine parvoviral immunoreactivity was markedly decreased following 2, 7, and 10 weeks of fixation in myocardium, small intestine, and spleen, respectively. Bovine respiratory syncytial virus immunoreactivity was markedly decreased following 7 weeks of fixation. Bartonella henselae had an abrupt loss of immunoreactivity following 9 weeks of fixation. Despite variation among time points, immunoreactivity remained moderate to strong throughout the study period for the other 18 antigens. These results suggest that prolonged formalin fixation of up to 7 weeks generally does not limit immunohistochemical detection of infectious agents. However, the effects of prolonged fixation depend on the targeted antigen and the selected antibody. The results of this study further validate the utility and reliability of immunohistochemistry in diagnostic pathology.
Effect of Hesperidin Extraction on Cell Proliferation and Apoptosis of Alzheimer's Disease Induced by A&#x03B2;<inf>25&#x2013;35</inf>

Abstract:Objective: To study the effect of hesperidin extracted from the peel of Gannan kumquat on Alzheimer's disease and its possible mechanisms. Methods: Cellular model of Alzheimer's disease was established using PC 12 cell induced by serum starvation method and treated by β-amyloid peptide 25-35 (Aβ 25-35 ). Effect of hesperidin on proliferation of PC12 cell induced by AP 25-35 was detected by the MTT method. Effect of hesperidin on apoptosis of PC12 cell induced by Aβ 25-35 was utilized by the acridine orange/ethidium bromide staining method. Results: PC12 cells stayed at Gο stage induced by serum starvation method in order to simulate the neuron in the state of terminal differentiation. After this, the PC cells induced by serum atarvation were treated by Aβ 25-35 to establish the cellular model of Alzheimer's disease. After synchronized PC12 cells being pretreated with different concentrations of hesperidin (0-200 μmol/L) for 1 h, and then 30 μmol/L Aβ 25-35 for 24 h, it was found that the survival rate increased along with the increasing dosage of hesperidin (F = 995.8, P <;; 0.001); however, there was no significant difference between 100 μmol/L group and 200 μmol/L group (P > 0.05). After synchronized PC12 cells being pretreated with 100 μmol/L hesperidin for 1 h, and then 30 μmol/L Aβ 25-35 for 24h, the cell nuclei were gradually similar to the cell nuclei of control group by by acridine orange/ethidium bromide fluorescent staining.Conclusion: Hesperidin inhibited the cytotoxicity and apoptosis induced by Aβ 25-35 and had protection for neurocyte.
A Comparative Study on the Damage Against Liver in Different Donation after Cardiac Death Rat Models

Abstract:Objective To investigate the damage against liver in different donation after cardiac death rat models and explore the underlying mechanisms. Methods Fifteen rats were equally divided into three groups: potassium chloride injection (PCI), diaphragmatic cutting (DC), apical clipping (AC) group. The PCL group was induced by intravenous injection of 10% potassium chloride solution to cardiac arrest through superficial dorsal vein of the penis. The DC group was made by clipping diaphragm to induce hypotension and hypoxia, and finally cardiac arrest. The AC group was made by clamping top of left and right ventricle of the heart with vascular clamp until cardiac arrest. The time of cardiac arrest was recorded. Serum was collected through postcava after cardiac death to evaluate liver function. Liver samples were collected and HE staining was performed. The level of nitric oxide (NO) and hydrogen peroxide (H2O2) were determined.The mRNA level of Bax, Bcl-2 and Caspase-3 were detected by RT-PCR and the protein level of p-Akt, Bax and Bcl-2 were detected by western blot. One-way ANOVA was used for comparing the time of cardiac arrest、liver function indexes、NO and H2O2 content in liver tissue、transcription levels of apoptosis-related molecules (Bax, Bcl-2, Capase-3) and related protein expressions between different groups. P<0.05 was considered statistically significant. Results The time of cardiac arrest of DC and AC group were (720±137), (1 020±142) s respectively, longer than that in PCI group [(111±10) s, P all<0.05]. The serum levels of ALT in the PCI and DC group were (29±14)、(49±11) U/L, respectively, lower than that in AC group [(71±13) U/L, P all<0.05]. The serum levels of AST、glucose and lactate dehydrogenase in the AC group were (293±61), (26.7±6.6), (2 534±907) U/L, respectively, higher than those in PCI group [(131±67), (15.8±8.5), (1 528±1 170) U/L, P all<0.05]. The result of HE staining showed that the AC and DC groups suffered more serious congestion and red blood cell infiltration than that in PCI group, while there were no significant differences between the AC and DC group. The contents of H2O2 of PCI and DC group were (13.8±3.7), (13.7±1.8) mmol/L, lower than that in AC group [(19.1±4.1) mmol/L, P all <0.05]. The NO content in the AC group was (13.3±3.2) mmol/L, higher than that in DC group [(6.9±0.4) mmol/L, P all <0.05]. The mRNA level of Bax was found higher in DC group than that in PCL group. The expression of Bax in AC group was higher than that in the PCL group. There were marked increase of P-Akt 473 and P-Akt 308 expression in PCL group than AC group (P all<0.05). Conclusion The PCL group was considered the less damage to liver and its mechanism was probably associated with the cell apoptosis reduction and P-Akt pathway activation. Abstract:The disadvantage of 10% EDTA decalcification is a long time-consuming. It needs to identify a quick and straightforward decalcification method when the preparation of lymphedema models using mouse tail which was a sample of bone wrapped in other tissues. In the present study, mouse tail samples were decalcified in 10% EDTA at 25, 37, and 42°C, respectively, with continuous shaking (150 rpm/min). The histologic integrity of samples was evaluated by hematoxylin and eosin staining, and the preservation of antigenicity was tested by either immunohistochemistry or immunofluorescence. The decalcification was distinctly accelerated by temperature. Results of hematoxylin and eosin staining were similar among different temperature groups. Immunohistochemistry and immunofluorescence staining revealed almost no signals in samples decalcified at 42°C for 1 week. Clear signals were detected when samples were decalcified at 37 and 25°C.

Abstract:Objective Arsenic trioxide(As2O3) eluting stent has the effect to inhibit inflammatory reaction in the section of stented coronary arteries in porcine coronary artery injury model.The aim of this study is to observe the effect of As2O3 on apoptosis of T lymphocyte cells and on apoptosis,phenotype and cytokine production of dendritic cells(DCs).Method Human monocytes were purified with Anti-CD14 microbeads and cultured in DMEM containing GM-CSF and IL-4 for 5 days to generate immature DCs.Cells were stimulated by As2O3 for 48 hours,the apoptotic T lymphocyte cells and DCs,phenotypic change of DCs were analyzed by flow cytometry,and the cytokine production of DCs was determined by ELISA.Results As2O3 dose-dependently induced the apoptosis of human T lymphocyte and DCs and inhibited the maturation of DCs below apoptosisinduced dose(3umol/L) by reducing expression of CD83,CD86 and cytokine production of IL10,IL12,IFN-γ.Conclusion The role of As2O3 in T lymphocyte and DCs is associated to the effect of As2O3 eluting stent on inhibition of inflammatory reaction in the section of stented porcine coronary arteries.
