Abstract:Six types of sensilla on the antenna of Opogona sacchari were found by scanning electron microscopy. They are trichoid sensillum, sensillun basiconicum, sensillum chaeticum, terminal hair, sensillum coeloconicum, sensilla squamiformia. Location of sensilla squamiformia was different to other insects, there may be one sensilla squamiformia in each sensilla group at top view, but in fact there are 4 to 6 sensilla squamiformia in each group at lateral view.

Abstract:Antennal surface structure of the Japanese tusser, Antheraea yamamai was observed by scanning electron microscope. The antennae of male and female were bipectinate with scape, pedicel and 33--37 flagellar segments. There were two pairs of equal long branch on eacli flagellar segment of male (Fig.1), but a pair of longer and a pair of shorter on female's (Fig.2).Seven types of sensillum, namely sensillum trichodeum, sensillum cheaticum, sensillum coeloconicum, sensillum campaniformlum, sensillum styloconicum, Bohm's bristle and sensillum squamiformium wer found on the antenna both of male and female. The sensillum trichodeum could be subdivided into long and short according to the shape and surface substructure. The long sensillum trichodeum had a diameter of 4.8μm at its base. It was reratively stright and slightly curved at the tip (Fig.3). The short sensillum trichodeum was 2.5--3.6 μm in diameter at the base. It was shorter, strongly bend down near its base and run parallel with the surface of the antenna (Fig.4). The surface of long and short sensilla trichodea were covered with annular ridges (Fig.5).

Abstract:The crystal structures of the human androgen receptor (hAR) and human progesterone receptor ligand-binding domains in complex with the same ligand metribolone (R1881) have been determined. Both three-dimensional structures show the typical nuclear receptor fold. The change of two residues in the ligand-binding pocket between the human progesterone receptor and hAR is most likely the source for the specificity of R1881 to the hAR. The structural implications of the 14 known mutations in the ligand-binding pocket of the hAR ligand-binding domains associated with either prostate cancer or the partial or complete androgen receptor insensitivity syndrome were analyzed. The effects of most of these mutants could be explained on the basis of the crystal structure.

Abstract:Agasicles hygrophila Selman and Vogt (Coleoptera: Chrysomelidae) is the key natural enemy of Alternanthera philoxeroides (Mart.) Griseb, an invasive weed worldwide. To understand the morphology of A. hygrophila and further explore the specific host localization mechanism, scanning electron microscopy was used to observe and study the morphological characteristics of sensilla on the head appendages, tarsi, and external genital segments of A. hygrophila. Twelve types and forty-six subtypes of sensilla were observed. These contain various types of head appendices, including sensilla chaetica, sensilla trichodea, sensilla basiconca, sensilla coeloconica, sensilla styloconica, Böhm bristles, sensilla campaniform, sensilla terminal, sensilla dome, sensilla digit-like, sensilla aperture, and many subtypes. A new type of sensor was reported for the first time, which may be related to host plant recognition. This sensor was located on the distal segment of the maxillary palps of A. hygrophila and was named as sensilla petal-shaped based on its morphological characteristics. Sensilla chaetica, sensilla trichodea, and sensilla basiconca are also found on the tarsi and external genital segments. In addition, sensilla basiconica 4, sensilla coeloconica 1 and 2, sensilla styloconica 2, Böhm bristles 2, and sensilla campaniform 1 were only found in females. On the contrary, sensilla styloconica 3, sensilla coeloconica 3, and sensilla dome were only found in males. Numbers and sizes of the sensilla were also different between males and females. The potential functions related to structure were discussed in comparison with previous investigations on beetles and other monophagous insects. Our results provide a microscopic morphological basis for further research on the localization and recognition mechanism of A. hygrophila and its obligate host.

Abstract:The antennal sensilla of both genders of macropterous and brachypterous adults of the small brown planthopper, Laodelphax striatellus (Fallen) (Hemiptera: Delphacidae) were examined using light and scanning electron microscopy. Scanning electron microscopy revealed seven types of antennal sensilla in adult L. striatellus which were not evenly distributed on all antennal segments. Sensilla chaetica, a sensillum campaniformium and a Bohm bristle were found on the scape. Sensilla chaetica, sensilla trichodea, sensilla placodea which always present as plaque organs, sensilla basiconica and a sensillum campaniformium were present on the pedicel. Three sensilla basiconica and one sensillum coeloconicum containing two sensory pegs were located on the swollen sensory region of the basal flagellum. Pores observed on the surface of s. trichodea and s. placodea suggest these organs probably play a role in olfaction, whereas the aporous s. chaetica with flexible sockets probably function as mechanoreceptors. The aporous s. basiconica with inflexible sockets are probable to be thermo-hygroreceptors while the Bohm bristle and s. campaniformia may act as antennal proprioceptors. The function of s. coeloconicum remains uncertain. The numerical dominance of antennal olfactory receptors suggests olfaction is an important function of the antenna in L. striatellus. Although a small degree of sexual/wing dimorphism was observed in the numbers of sensilla and in the length and width of antennae and antennal segments, the basic shape and structure of the antennae and antennal sensilla did not differ between the gender or wing form in L. striatellus. Abstract:The Angoumois grain moth, Sitotroga cerealella, is a serious pest of stored grains worldwide. Presently, the best effective control against the moth is to disrupt the sexual communication between sexes. Sexual communication in moths includes two processes in which females produce and release pheromones from the sex pheromone gland and males detect and respond to them with a relatively sophisticated olfactory system in their antennae. To better understand these processes, we studied the ultrastructure of antennal and ovipositor sensilla of S. cerealella and determined the location of the female sex pheromone gland. Seven types of antennal sensilla were identified on both sexes: sensilla trichodea, sensilla chaetica, sensilla coeloconica, sensilla styloconica, sensilla auricillica, sensilla squamiformia and Bӧhm bristles. Of these sensilla, the sensilla trichodea were significantly more abundant on male antennae than on those of females, suggesting that these sensilla may detect the sex pheromones. On the ovipositor, only sensilla chaetica of various lengths were found. The sexual gland was an eversible sac of glandular epithelium that was situated dorsally in the intersegmental membrane between the 8th and 9th abdominal segments. These results will lead to a better understanding of mate finding with sex pheromones for this worldwide pest species.

Abstract:Cotesia gregalis Yang et Wei (Hymenoptera: Braconidae) is a gregarious koinobiont endoparasitic wasp attacking the larvae of fall webworm, Hyphantria cunea, an important invasive insect pest in China. To better understand the parasitic wasps' mating and parasitic behaviors, we examined the morphology of the antennae of adult C. gregalis, as well as the type, number, and distribution of antennal sensilla, via scanning electron microscopy. The antennae of female and male C. gregalis are filiform and comprise a scape, pedicel, and 16 flagellomeres. The female antennae are significantly shorter than those of male. A total of nine morphological types of antennal sensilla (mechanoreceptor and chemoreceptor) are presented in both sexes, including four mechanoreceptors (sensilla chaetica [two subtypes], sensilla trichodea and Böhm bristles); five chemoreceptors (sensilla basiconica [two subtypes], sensilla placodea, sensilla styloconica, and sensilla coelocapitula). There is no difference in the type and distribution of antennal sensilla between males and females, but the number and length of some antennal sensilla show sexual dimorphism. The functional morphology of the sensilla of C. gregalis is discussed by comparison with other parasitic wasps. These findings provide foundation for further research on the chemical communication and host localization mechanisms of C. gregalis. RESEARCH HIGHLIGHTS: The first report of morphology and distribution pattern of the antennal sensilla in C. gregalis is discussed. A total of seven main types and nine antennal sensilla subtypes are observed in male and female C. gregalis. The type and distribution of antennal sensilla in males and females are identical; however, the number and length of certain antennal sensilla show sexual dimorphism.
Morphology and distribution of sensilla on the antennae and ovipositor of the Hymenopteran parasitoid Pseudotorymus jaapiellae Yang et Chen

Abstract:Pseudotorymus jaapiellae is an important ectoparasitoid of the larvae of Gephyraulus lycantha, a serious gall-forming pest that devastates wolfberry, Lycium barbarum, in Northwest China. To provide requisite background for our ongoing research on the mechanisms of P. jaapiellae's host location and subsequent oviposition, we used scanning electron microscopy to describe the external morphologies and distributions of sensilla on their antennae and ovipositors. The geniculate antennae of both male and female P. jaapiellae were each composed of a scape with a basal radiculum, a pedicel, an anellum and a flagellum. We identified nine morphological sensilla types on the antennae of both sexes, including three sensilla trichodea (ST), one sensillum basiconicum (SB), two sensilla chaetica (SCh), one sensillum placodeum (SP), one sensillum coeloconicum (SCo), and one sensillum campaniformia (SCa). Females had significantly more ST I and SP than males had, but males had more ST III than did females. We observed six types of sensilla on the ovipositor, including three ST, one SB, and two SCa. ST II, ST IV and SB II were on the sheath, whereas ST V and the SCa were on the stylus. Finally, the possible biological functions of these sensilla were discussed according to their morphology and ultrastructure. These results provide an important basis for further study on chemical communication between P. jaapiellae and their host, and contribute to the development of a biological control program for G. lycantha, using the parasitoid, P. jaapiellae.

Abstract:Ooencyrtus phongi (Hymenoptera: Encyrtidae) is an important egg parasitoid of the litchi stink bug, Tessaratoma papillosa (Drury) (Hemiptera: Tessaratomidae). Antennae of parasitic Hymenoptera are important sensory organs and play an important role in host location, host discrimination, courtship, and mating behavior. In this article, we examined the external morphology of the antennal sensilla of female and male of O. phongi using scanning electron microscopy. Twelve morphological sensillar types were recognized in both sexes, including the placoid sensilla, basiconic sensilla, two types of sensilla trichodea, and eight types of sensilla chaetica. Major differences were found between the sexes, in number, distribution, shape, structure, and size of the identified sensilla. These results are discussed in relation to the possible role of these sensilla in the host location behavior of O. phongi.

Abstract:Antennal sensilla were compared in parthenogenetic females and bisexual adults of rice water weevil, Lissorhoptrus oryzophilus Kuschel (Coleoptera: Curculionidae), using scanning electron micrographs. The antenna of parthenogenetic females and of both sexes of bisexual reproductive weevils consisted of the scape, pedicel, and 6 flagellar segments. Females, regardless of mode of reproduction, had longer antennal segments than males. The 3 adult forms/genders can be correctly classified into their respective groups with a set of discriminant mathematical functions based on 4 morphological measurements of length or maximum width of segments of the antennae. For both sexes, 6 morphological sensilla types were found on the antennae, including hydrofuge scales I (hsI), trichoid sensilla (ts), 2 types of squamiform sensilla (ssI, ssII), tuft hairs (th), and Böhm bristles (Bb). No remarkable sexual differences in the types or distribution of sensilla were found on the antennae among parthenogenetic and both sexes of bisexual reproductive weevils, except for hsI. Both the length and basal diameter of hsI of females were significantly greater than those of the males. The hsI of the parthenogenetic females were significantly longer than those of the bisexual females. The numbers of branches of hsI on the sixth flagellum of the parthenogenetic females were significantly more than on both sexes of bisexual adults. The numbers of branches of hsI of the females were significantly more than those of the males. From the information gained from this scanning electronic microscope study of antennae, we can separate rice water weevils by gender and reproductive mode.
Identification and Characterization of Chemosensory Genes in the Antennal Transcriptome of Spodoptera Exigua

Abstract:Chemical senses are crucial for insect behaviors such as host preference, mate choice and oviposition site selection. Various protein families are involved in these processes, including odorant receptors (ORs), ionotropic receptors (IRs), gustatory receptors (GRs), chemosensory proteins (CSPs), odorant binding proteins (OBPs) and sensory neuron membrane proteins (SNMPs). To better understand the olfactory mechanism in Spodoptera exigua, we conducted transcriptome analysis of adult antennae and identified a total of 157 candidate chemosensory genes encoding 51 ORs, 20 IRs, 7 GRs, 32 CSPs, 45 OBPs and 2 SNMPs. Quantitative real time PCR (qPCR) analysis of the tissue- and sex-specific expression profiles of ORs, GRs and IRs revealed that these genes could be detected in at least one tissue tested. SexiOR6, 11, 13 and 16, which were predicted to be pheromone receptors based on phylogenetic analysis, exhibited male-specific antennae expression. SexiOR18, 26, 28, 30, 34, 39, and 40 exhibited female-biased expression. SexiGR1, SexiGR2 and SexiGR3, are predicted carbon dioxide receptors, and the former was expressed specifically in antennae, and the latter two were expressed both in antennae and labial palps. SexiIRs had diverse expression profiles. SexiIR8a and SexiIR25a were quite conserved and expressed at high levels. This work will greatly facilitate the understanding of olfactory system in S. exigua and provides valuable information for further functional studies of the chemoreception mechanism in Lepidopteran moths.
Antennal sensilla and ovipositor morphology of the European birch sawfly Arge pullata Zadd (Hymenoptera: Tenthredinidae, Argidae)

Abstract:Arge pullata Zadd is an important phytophagous pest that damages red birch Betula albo-sinensis in Hubei Province, South China. Massive ecological and economic losses have been caused by this species, which threatens the ecological security of the Shennongjia Nature Reserve. To investigate the mechanoreception, chemoreception, and oviposition processes of A. pullata, scanning electron microscopy and optical confocal microscopy were used to reveal the typology, morphology, and distribution of ovipositor and antennal sensilla. The results show that A. pullata has clavate antennae and eight types of sensilla in total, including sensilla chaetica, sensilla trichodea (types 1-3), sensilla basiconica, sensilla coeloconica (types 1 and 2), and Böhm's bristles. Sensilla trichodea type 1 distributed only on male antennae; the densities of sensilla trichodea type 2 and sensilla basiconica differed between the sexes. The binding pattern of ovipositor valvulae was discovered, and one type of sensilla chaetica, two types of sensory pits, and tooth-like cones as well as two types of microtrichia were found in the ovipositor. Based on morphological evidence and research on Hymenoptera, putative functions are suggested to increase our understanding of the mechanisms by which this species finds hosts and mates, and how oviposition takes place.
Loss of HCN channel mediated Ih current following seizures accounts for movement dysfunction

Abstract:Hyperpolarisation-activated, cyclic nucleotide-gated (HCN) channel mutations are linked to disorders characterized by the occurrence of recurrent seizures (epilepsies) and HCN channel dysfunction has also been observed following evoked seizures in otherwise typical brains. Whether HCN channels contribute to the behavioral co-morbidities associated with epilepsy has received limited attention, despite extensive work to show that they serve as key regulators of neuronal excitability. A recent article, co-authored by us, provides evidence that experimental seizures disrupt HCN channel function and that this type of disruption leads to long-term impairments in skilled motor behavior. Given that interictal motor impairments are observed following experimental seizures and clinical epilepsy, this new study suggests an intriguing possibility that HCN channels represent a novel therapeutic target to treat co-morbidities of this disorder. HCN channels provide a diverse set of contributions to brain excitability. At the level of individual neurons, the current mediated by HCN channels, referred to by many names including Ih, affects integration of synaptic input as well as patterns of action potential firing. It has previously been shown that HCN channels serve as a mechanism to restrict spatial firing fields within entorhinal cortex. HCN channels also restrict hippocampal-dependent spatial memory. In our recent work, we sought to examine the role of HCN channels for networks located in motor cortex. The study manipulated HCN channels using 3 separate approaches. Repeated experimental seizures were used as they reduce Ih in layer 5 pyramidal cells that make up the cortical spinal tract. The pharmacological blocker ZD7288 was locally applied within motor cortex and global HCN1 knockout (HCN1) mice were used as a genetic strategy. In order to examine network function of motor cortex, in vivo studies were performed using standard intracortical microstimulation (ICMS) to systematically measure evoked forelimb movement responses across sites within neocortex. With short-train ICMS parameters, stimulation at individual sites within motor cortex predominantly results in responses on the contralateral side of the body that are characterized by simple flexion or extension across a single joint. With repeated seizures there was a substantial increase in the number of stimulation sites that exhibited complex forelimb movement responses rather than the simple flexion or extension movements. These new complex forelimb responses occurred as combinations of simple movement responses and were present contralateral to stimulation and often bilaterally. This result was also seen after direct application of ZD7228 and in HCN1 mice. Additionally, no further change in ICMS responses occurred when the effects of ZD7228 were tested in HCN1 mice. Since experimental seizures had previously been shown to impair skilled motor behavior, additional experiments tested whether genetic or pharmacological manipulation of HCN channels also affected skilled motor behavior in awake behaving rodents. Abstract:The woodwasp Sirex noctilio Fabricius is a major quarantine pest that was reported in China in 2013 and mostly damages Pinus sylvestris var. mongolica. Reverse chemical ecology, which uses chemical lures to catch or block insects from mating is the classic way to control forestry pests. This indicates that insect sensilla play a crucial role in detecting external chemical and physical stimuli. Nonetheless, the categorization and distribution of sensilla on the antennae and ovipositor of S. noctilio are insufficiently specific. In this paper, scanning electron microscopy (SEM) was used to observe the ultrastructure of the sensilla of S. noctilio on the antenna and ovipositor. It was found that the types and distribution of sensilla on the antennae of S. noctilio male and female are consistent, and six types of sensilla are found: sensilla trichodea (ST), sensilla chaetica (SC), Böhm bristles (BB), sensilla basiconica (SB), sensilla ampullacea (SA), and contact chemoreceptors (CC). Besides, there are five types of sensilla on the female ovipositor. In addition to ST, SC and BB, two more types of sensilla are also found: sensilla cavity (SCa) and sensilla coeloconica (SCo). Through identification of the morphology and distribution of the sensilla, the functions of different sensilla in the mating and host selection mechanisms of S. noctilio are proposed, thereby establishing a foundation for S. noctilio chemical communication research.

Abstract:As part of an ongoing process of curriculum development, a new course was developed at the University of British Columbia (UBC) to address problems such as lack of student enjoyment of the learning process, poor student preparedness for clinical treatment planning, and underdeveloped critical thinking skills in students beginning their clinical experience. The new course utilized a hybrid problem-based learning (PBL) format that provided students with an overview in lecture format while encouraging active learning in small group tutorials and seminars. Half the second-year class was randomly selected to participate in the new course, while the other half received the standard lectures on the subject. An outcomes assessment examined whether the aforementioned problems had been addressed in the new course. Course participants completed a post-course evaluation, and all students completed a self-assessment of their preparedness and progress in treatment planning and diagnostic. Clinical instructors, who were blind to the identity of the new course participants, were asked to independently assess each student using the same criteria. Results indicate that students who participated in the hybrid-PBL course enjoyed the learning process and later rated themselves as being better prepared and improving more in the areas of treatment planning and diagnostic records than their counterparts, although the latter results were not routinely statistically significant (p < 0.1). Instructor evaluations supported these differences between the two groups.

Abstract:The antennal sensilla play an important role in many behavioral activities of insects. The fungivorous beetle Triplax ainonia Lewis (Erotylidae) is an important pest which prefers to feed on Pleurotus mushrooms. In order to clarify the types, number, and distribution of the antennal sensilla of male and female T . ainonia , scanning electron microscopy was used. The results showed that there were five sensillum types on the antennae of adults male and female, including Böhm's bristles (BB), sensilla chaetica (three subtypes: SC 1, SC 2, and SC 3), sensilla basiconica (three subtypes: SB 1, SB 2, and SB 3), sensilla trichodea (ST), and sensilla styloconica (SS). Among all the sensilla, the number of SB 2 was the most abundant in both sexes. We found that there was no sexually dimorphic in the sensillum types, but there were differences in the number, lengths, and diameters of some sensilla between males and females. Based on the information of the morphology and distribution of the sensilla, the potential functions of the antennal sensilla of T . ainonia adults were discussed. The results of this study provide a basis for further study on the behavioral ecology and electrophysiology of the fungivore beetles of the Erotylidae.

Abstract:Tea geometrid Ectropis obliqua Prout (Lepidoptera: Geometridae) is a devastating defoliator throughout the tea plantations in China. To explore the putative functions of chemosensilla involved in host selection, the morphology and ultrastructure of antennal and tarsal sensilla in E. obliqua moths were visualized by scanning and transmission electron microscopy. Eight types (including 15 subtypes) of sensilla were identified. On antennae, sensilla trichodea (STR I and STR II) were the most abundant sensillum type characterized by porous cuticle and dendrites in the sensillum lymph. Sensilla chaetica (SCH I–SCH III) possessed a terminal pore and thick nonporous wall, with dendrites in the inner lymph cavity. Three subtypes of sensilla basiconica (SBA I–SBA III) and one type of sensilla auricillica were equipped with thin porous cuticular walls and multiple dendrites in the lymph. Sensilla styloconica were equipped with two or three dendrites that lacked cuticular pores. Sensilla coeloconica possessed four to seven dendrites and were double walled with spoke channels. Böhm bristles and sensilla squamiformia were also observed on the antennae. On tarsi, two subtypes of sensilla chaetica (SCH IV and SCH V) comprised the majority of chemosensilla, and are believed to function in gustatory perception. We summarize the validated functions of related sensilla, and propose potential functions of the corresponding sensilla in E. obliqua. These findings provide a working basis for investigating sensillum function and sensory mechanisms in this pest species.
