Abstract:The effect of acid hydrolysis (Lintnerisation) and the thermal gelatinisation on the dynamic state of starch and the microscopic distribution of water within native potato starch granules is investigated with proton NMR relaxometry. The NMR relaxation data show several dynamic states of the amylose and amylopectin chains and clearly identify the melting transition during thermal processing. The Lintnerisation process is found to follow first-order kinetics and to shift and broaden the melting transition. The unique value of NMR relaxometry for monitoring the processing response of starch granules is discussed.

Abstract:The evolution of the starch fine structure during growth and its impact on the gelatinization behavior of cassava starch (CS) was investigated by isolating starch from South China 6068 (SC6068) cassava harvested from the 4th to 9th growth period. During growth, the short-range ordered structure, crystallinity as well as particle size distribution of starch were increased. Meanwhile, the starch molecular size and amylopectin (AP) proportion increased, while the proportion of amylose (AM) exhibited a decreasing tendency. The chains of short-AM (X - 100-1000) were mainly significantly reduced, whereas the short and medium-AP chains (X - 6-24) had the most increment in AP. The solubility, thermal stability, shear resistance, and retrogradation resistance of starch were enhanced after gelatinized under the influence of the results mentioned above. This study presented a deeper insight into the variation of starch fine structure during growth and its influence on gelatinization behavior, which would provide a theoretical basis for starch industrial applications.

Abstract:To overcome the drawbacks of dehydration and low gel strength of the gels of L-arginine-treated konjac glucomannan (KGM), thermally irreversible gel was prepared by L-arginine induced KGM-modified starch mixed sol. The effects of the three modified starches (corn modified starch, acetylated bis-starch adipate, and hydroxypropyl distarch phosphate) on the mixed system were investigated by texture analysis, scanning electron microscopy, Low field NMR, Fourier transform infrared spectroscopy and Differential thermal analysis. The results showed a gradual increase in hardness with increasing concentration of modified starch. Differential thermal results indicated that acetylated bis-starch adipate composited gels had the highest thermal decomposition temperature (309 C). The changes in the textural properties and thermal stability of the three mixed gels were mainly attributed to the enhanced hydrogen bonding between starch/KGM/L-arginine, and changes in the microstructure. When the starch concentration was higher, the denser the gel network structure was, the more difficult it was for water trapped in the gel system to precipitate out. The results indicated that acetylated bisstarch adipate was more suitable to be added to the mixed system to prepare thermally irreversible gels.

Abstract:Starch gelatinization is important in food processing and industrial use. Granule swelling and gelatinization temperature of 11 starches from different plants were investigated in situ using hot stage microscopy during heating. The amylose content, swelling power, pasting temperature and thermal property of these starches were also measured. The results showed that hot stage microscopy was suitable for measuring granule swelling and the gelatinization temperature of starch during heating. The sectional area swelling percentage of starch granules measured using hot stage microscopy was significantly positively correlated with the swelling power. The gelatinization temperature measured using hot stage microscopy was significantly positively correlated with the pasting temperature and with the thermal property for all 11 starches. For rice starches with the same crystallinity and similar size, the gelatinization temperature was negatively correlated with the amylose content and positively correlated with the swelling power and the sectional area swelling percentage at 95°C.

Abstract:The digital image analysis, integral optical density (IOD) method, combined with "the model of response difference of crystallite change (MRDCC)" and " the model of gelatinization process (GP)" was applied to further study the gelatinization of corn starch, potato starch, and pea starch in different concentrations of NaCl solutions. It was found that the addition of non-aqueous media not only affected the gelatinization temperature, but also impacted on the whole gelatinization process and effected differently at different stages of gelatinization. NaCl solution of 1, 3 and 4 mol/L (M) rose the GP curve of corn starch while 2 M had little effects on it; for potato starch, it seemed that NaCl gave little efforts on GP curve while 1 M rose the GP curve at the relative biggest degree; for pea starch, it performed two stage gelatinization process in different concentration of NaCl solution, this may due to the resultant force of heat, plasticizer, solute and expansion power and more studies were needed to reveal the gelatinization mechanism. Compared with the loss of crystalline structure (long-ordered structure), the disappearance of the molecular helixcoil structure (short-ordered structure) contributed greater to the DSC enthalpy, which could be deduced from the GP model. Relationship among gelatinization, retrogradation behavior, and impedance characteristics of potato starch

Abstract:In this study, the physicochemical properties of potato starch from different varieties were investigated. Furthermore, the relationships among gelatinization, retrogradation behavior, and impedance characteristics of potato starch gels were evaluated by texture analysis, low-field nuclear magnetic resonance spectroscopy, and electrical impedance spectroscopy. The results indicated amylose content was positively correlated with setback viscosity, and negatively correlated with To and ΔH. In addition, impedance values of potato starch gels differed in a frequency-dependent manner. Notably, higher frequencies resulted in low diffusion of ions in prepared gels, which combined with the concentration of mobile ions in free water, led to a gradual decrease in impedance module. Compared with phase values, impedance module showed high correlation with gelatinization parameters (To, Tp, and Tc) and viscosity parameters (peak temperature and setback viscosity), more notably at frequencies below 100 Hz. In this context, the electric current flowed through mobile ions that interacted with bound water attached to the starch molecules at lower voltage frequencies, and were repressed by the formation of an ordered and compact gel network during retrogradation. Collectively, these results indicate that impedance spectroscopy can be potentially used as an efficient and reliable method to predict gelatinization and retrogradation behavior of potato starch.

Abstract:The influences of various gelatin emulsion additions on gelatinization, rheology, retrogradation and structural properties of waxy rice starch (WRS) were investigated. The gelatin emulsion was uniformly dispersed in the starch pastes, which increased the particle size through agglomerating around the starch powder particles. As gelatin emulsion content increased, the pasting viscosity of WRS pastes significantly increased (P < 0.05), whereas its setback value, gelatinization and retrogradation enthalpy decreased. Rheological measurements indicated that the addition of gelatin emulsion did not change the shear thinning behavior of WRS pastes blends, and the storage modulus and loss modulus were positively correlated with gelatin emulsion content. During the heating stage of the pasting process, the interfacial organization consisting of gelatin undergoes disintegration and melting, reconfiguring the emulsion system. Gelatin dispersed in the interfacial and continuous phases interacts with starch to form hydrogen bonds and inhibit starch rearrangement. The starch-lipid complex formed by the interaction between lipids released from emulsion and starch is capable of destroying the crystalline region. Therefore, after 14 d of storage at 4 C-degrees, the relative crystallinity of WRS decreased by 72.4 % as the gelatin emulsion addition was increased from 0 % to 14 %. The addition of gelatin emulsion induced the formation of steric hindrance effects, which impeded the mobility of water molecules and improved freeze-thaw stability. These results may contribute to the creation of high-quality and innovative water-oil-starch mixture food products that remain fresh for a longer period.
Effect of typical starch on the rheological properties and NMR characterization of myofibrillar protein gel

Abstract:Background: Composite gels were individually prepared from 20 g kg-1 myofibrillar protein (MP) imbedded with typical native starch (potato, tapioca, rice or corn starch) in 0.6 mol L-1 NaCl at pH 6.2. The gel strength, water holding capacity, rheological properties and microstructure of the obtained myofibrillar protein-starch composite gels were evaluated. Results: Tapioca starch improved (P < 0.05) gel strength and water holding capacity of MP composite gel at 80 °C. Rheological properties of MP-starch composites differed significantly with the addition of different types of native starch. Additionally, the promoting effect of starch on the storage modulus of the composite gels positively correlated with the gelatinization properties of different typical starch. Environmental scanning electron microscopy showed that the filling effect of starch on the composite gel was related to the pasting temperature and particle size of typical starch, with almost no particles forming at 80 °C. Moreover, the addition of starch changed the relaxation peak area and increased the relaxation time in nuclear magnetic resonance tests, which suggested that starch could improve the water holding capacity of MP-starch composite gels. Conclusion: Different typical native starch has varied impacts on the gel strength, water holding capacity, rheological properties and microstructure of MP gels, indicating the potential and feasibility of these typical native starches as an addition agent to modify the textural properties in comminuted meat products. © 2019 Society of Chemical Industry.

Abstract:In the present study, intact potato cells as whole potato food model were isolated through soaking with mild acid and alkali solutions, followed by controlled hydrothermal treatment to reach 0–100% degree of gelatinization (DG). The morphological and structural changes of entrapped starch granules as well as in vitro starch digestion properties were investigated. Our results indicated that the changes of cell wall permeability induced by varied DG values dominated the in vitro starch digestion profiles of intact cells. The gelatinization of intracellular potato starch leads to cell wall expansion and high cell wall permeability, resulting in the accessibility changes of digestive enzymes to encapsulated starch substrates through cell wall barrier. This study suggests that starch digestibility of potato foods can be regulated through cell wall permeability induced by intracellular starch gelatinization degree.
Study on Gel Texture Properties of Potato Starch Paste in Ultrasonic Field

Abstract:The gel texture properties of starch paste have great effects on guiding the process of food production. The gel texture properties of potato starch paste in ultrasound field were investigated. The effects of the ultrasonic field condition and the content of potato starch paste on the gel texture properties of potato starch paste were evaluated. The results showed that with the prolonging of the ultrasonic time and the strengthen of sound intensity, the gel texture properties of potato starch paste such as hardness value, fracture ability value, adhesiveness value, gumminess value and chewiness value decreased in ultrasonic field. Moreover, hardness value, adhesiveness value and gumminess value showed a slower decreasing tendency with the increase of the content of potato starch paste.

Abstract:Dynamic changes of rice starch granules selected for different amylose contents were analyzed in excess water while heated in-situ in a small-angle X-ray scattering (SAXS) instrument. Normal rice starch (NS) and rice starch with high amylopectin (HAP) and high amylose (HAM) were used as models. A 1D linear correlation function and a combination power-law and Gaussian function were used to extract the starch lamellar structure parameters and the fractal dimension, ordering and distribution of starch lamellae from SAXS data. For the resulting starch paste/gels, a model of two-correlation length was fitted to afford the correlation length (ξ) for the paste/gel system. The results showed that HAM exhibited higher long period (LP) and thickness of the crystalline layers (dc) values than HAP and NS. However, HAP showed the highest ordering lamellar structure. HAP granules were more thermostable than the amylose containing starches. For the gelatinized starches, HAM showed the highest correlation length values but these notably decreased with increasing temperature indicating strong chain segment interaction. This research reveals essential structural changes in lamellae of rice starch granules and rice starch gel structure, which provides potentially useful in the working of starch-based foods and materials.
Insights into the interaction of CaCl2 and potato starch: Rheological, structural and gel properties

Abstract:High viscosity of starch greatly limit its application in some specific foods, in this work, a novel low-viscosity potato starch (PS) was developed via crosslinking between PS (3 %, w/v) and Ca2+ to investigate the effect of CaCl2 concentration (0.1-5 % CaCl2, w/v) on the rheological behaviors, structural and gel properties of PS. The results showed that peak viscosity (PV), trough viscosity (TV), final viscosity (FV), and breakdown viscosity (BD) of pasting curves of CaCl2-treated PS were significantly reduced compared with the native PS. The CaCl2 treatment also decreased the firmness of the PS gel and increased its pasting temperature (PT) and gelatinization enthalpy (∆H). Moreover, The CaCl2 treatment also led to more organized crystallites in the PS granules as affected by the slight increase in the ratio of 1044/1015 cm-1 in the FT-IR analysis, reduced the homogeneity of ordered structures inside granules as indicated by the increase in conclusion temperature (Tc)-onset temperature (To) in DSC analysis, and decreased relatively crystallinity revealed by XRD analysis. The findings of this study indicated CaCl2-treated PS could serve as food ingredients with reduced paste viscosity and regulated paste stability under shear during heating.

Abstract:The effects RG-І pectin (0.5, 1, 2, 5 and 10%, w/w) on the gelatinization behaviour, gel structure and digestibility of potato starch were characterized by means of rapid viscosity analysis (RVA), rheometry, scanning electron microscopy (SEM) and in vitro digestion. Results indicated that the addition of RG-I pectin significantly decreased the peak and breakdown viscosity and prolonged the pasting temperature of potato starch, but increased the setback viscosity. Meanwhile, the swelling power and amylose leaching of starch granules were restrained because of RG-I pectin adsorption on the starch granules surface. The presence of RG-I pectin also decreased the G’ values of potato starch gel but increased the tan δ values (especially for 1%), indicating that RG-І pectin decreased the 3D network formation of potato starch gel. The microstructure analysis indicated that RG-I pectin decreased the formation of the gel structure, which might place constraints on the relative amylose concentration in the continuous phase. The addition of RG-I pectin also reduced the rapidly digestible starch (RDS) content and increased the slowly digestible starch (SDS) content, while no significant differences of resistant starch (RS) content was observed. Those results indicated that RG-I pectin could affect the physicochemical properties of potato starch and modulate its digestibility and might be a potential option for producing starchy food with lower digestibility.

Abstract:Gelatinization is an important property of starch for biomedical applications. However, studies on the changes in starch granules in terms of morphology, swelling, amylose leaching and so on during gelatinization, which are key to uncovering the starch gelatinization process, have rarely been reported. Herein, changes of cassava and potato starch granules during gelatinization were investigated. It was found that there is a substantial difference in the granule changes during gelatinization between cassava and potato starch. Cassava starch granules remain intact with slight swelling, with approximately 8.5% amylose leaching in water for 30 min at 60 °C. In sharp contrast, potato starch granules swell very well and rapidly, losing much integrity with 51.05% amylose leaching. The gelatinization time and temperature have much greater effects on the changes of potato starch granules than cassava starch granules.

Abstract:Starch gelatinization has been widely studied previously, but there is still a lack of systematical research on the relationship between the degree of starch gelatinization (DSG) and its physicochemical and structural properties. In this study, potato starch samples with DSG ranging from 39.41% to 90.56% were obtained by hydrothermal treatment. The thermal, rheological, and structural properties, as well as the water-binding capacity of samples were investigated. A starch solution with a DSG of 39.41% was partially sedimented at room temperature, while starch with a DSG of 56.11% can form a stable paste with a fine shear-thinning property, as well as samples with a DSG larger than 56.11%. The endothermic enthalpy, gelatinization range, and short-range ordered structure of starch were negatively correlated with DSG, whereas onset gelatinization temperature, apparent viscosity, and water-binding capacity were positively correlated. The viscoelasticity of starch gels was negatively correlated with the DSG after full gelatinization (DSG > 39.41%). Starch granules gradually lose their typical shape and less birefringence can be observed with increasing DSG. Hydrothermal treatment has a more significant effect on the amount of exposed hydroxyl groups than the ordered and amorphous structures of partially gelatinized starch. This study built linear correlations between starch physicochemical properties and the DSG and provided comprehensive insight into the characteristics of partially gelatinized potato starch.

Abstract:In order to reveal the influence of ultrasonic treatment on the gel properties of starch pastes,the gel properties of potato starch paste with variable concentration were investigated using a texture analyzer in different ultrasonic treatment conditions.Then,the microstructure of the potato starch paste after the ultrasonic treatment was observed by means of SEM.The results show that the ultrasonic treatment greatly affects the gel properties of potato starch,and that the gel strength decreases with the prolonging of ultrasonic treatment time and with the increase of ultrasonic intensity,the decreasing degree being positively linear with the concentration of starch paste.Moreover,it is demonstrated that the degradation of gel properties mainly results from the variation of paste microstructure caused by ultrasonic treatment,concretely,after the ultrasonic treatment,the microstructure of starch paste changes from a stronger composite network to a loosen and group-gathering one.
Gelatinization Characteristics of Potato Starch by Microscopic Observation Combined with Target Recognition Technology

Abstract:A novel method of microscopy observation combined with target recognition based on Artificial Neural Network (ANNs) was developed to study the gelatinization characteristics of starch, and the on-line detection of the gelatinization characteristics of potato starch was realized by machine learning algorithm. In this paper, a detector based on deep neural network (starch-SSD) was designed to monitor the morphological changes of potato starch with the increasing of temperature. The starch birefringence characteristics were automatically identified first and then, the number of starches with birefringence characteristics in each micrograph during the process of gelatinization was calculated. The gelatinization temperature of starch and the degree of gelatinization (DG) at different temperatures were determined according to the change of birefringence. Results showed that compared with the traditional methods based on artificial vision observation, the proposed method had the advantages of high precision and fast speed. In addition, this method overcame the uncertainty of artificial subjective judgments and provided a uniform standard for judging the gelatinization characteristics of starch by microscopic. This study would provide the possibility of on-line monitoring during the industrialized production of starch products.

Abstract:Modulation of the starch digestion rate and extent is a major target for increasing nutritional value of potato-based food. In order to understand the effect of controlled gelatinization on the structure and in vitro digestion properties of whole potato food, intact parenchyma cells were isolated as a model through soaking in mild acid and alkali solutions, and then hydrothermally treated at different incubation temperatures in excess water. The morphological and structural changes of entrapped starch granules as well as in vitro starch digestion properties were investigated. Three classes of starch digestion of potato cells are identified: (1) when the temperature of hydrothermal treatment was set at 55 °C or 60 °C, the efficient physical barrier of intact cell walls resulted in a limited starch digestion extent; (2) when the temperature is set at 65 °C, the cell wall structures apparently reduced the digestion rate but only slightly reduced the final extent compared to the starch counterpart, probably because starch swelling weakens the physical barrier; (3) at 70-95 °C, the damaged cell wall still reduced the digestion rate but did not reduce the digestion extent compared to the starch counterpart. The results suggest that controlling hydrothermal processing temperature is a viable approach to develop potato ingredients with enhanced nutritional functionality.

Abstract:A thorough understanding of starch gelatinization is extremely important for precise control of starch functional properties for food processing and human nutrition. Here we reveal the molecular mechanism of starch gelatinization by differential scanning calorimetry (DSC) in conjunction with a protocol using the rapid viscosity analyzer (RVA) to generate material for analysis under conditions that simulated the DSC heating profiles. The results from DSC, FTIR, Raman, X-ray diffraction and small angle X-ray scattering (SAXS) analyses all showed that residual structural order remained in starch that was heated to the DSC endotherm end temperature in starch:water mixtures of 0.5 to 4:1 (v/w). We conclude from this study that the DSC endotherm of starch at a water:starch ratio of 2 to 4 (v/w) does not represent complete starch gelatinization. Probing structural features in potato starch granules at moderate hydration through the modelling of 1H->13C polarization transfer kinetics

Abstract:The structural arrangement of starch polymers in presence of water is known to impact the functional properties of starchy products. In this study, the hydration of potato starch granules was investigated at the molecular level through various 1H->13C polarization transfer solid-state Nuclear Magnetic Resonance (ss-NMR) experiments. The impact of increasing the water content from 12.3 % to 45.9 % was assessed using 13C Cross Polarization Magic Angle Spinning (CPMAS), Variable Contact Time (VCT-CPMAS), Variable Spin Lock (VSL-CPMAS), and T One Rho QUEnching (TORQUE) NMR sequences. Of these, VCT-CPMAS proved to be the most promising. When applied with an optimal number of contact times, it enabled the application of several mathematical models that provided detailed insights into the structuring of protons in the hydrated potato starch granules. At low hydration (12.3 %), the models enabled various structural domains to be distinguished, which we suggest are associated with helical and amorphous structures. At moderate hydration (45.9 %), we tested two fitting models. Two pools of protons were revealed, corresponding to loosely ordered structures on the scale of tens of nanometers. These findings suggest varying water distribution during starch hydration and are likely to indicate variable hydration levels in the multilamellar amorphous structures of starch granules.
