Abstract:Spodoptera frugiperda (JE Smith) represents the first documented case of field-evolved resistance to a genetically engineered crop expressing an insecticidal protein from Bacillus thuringiensis (Bt). In this case it was Cry1F-expressing maize (Mycogen 2A517). The ladybird beetle, Coleomegilla maculata , is a common and abundant predator that suppresses pest populations in maize and many other cropping systems. Its larvae and adults are polyphagous, feeding on aphids, thrips, lepidopteran eggs and larvae, as well as plant tissues. Thus, C. maculata may be exposed to Bt proteins expressed in genetically engineered crops by several pathways. Using Cry1F-resistant S. frugiperda larvae as prey, we evaluated the potential impact of Cry1F-expressing maize on several fitness parameters of C. maculata over two generations. Using Cry1F resistant prey removed any potential prey-mediated effects. Duration of larval and pupal stages, adult weight and female fecundity of C. maculata were not different when they were fed resistant S. frugiperda larvae reared on either Bt or control maize leaves during both generations. ELISA and insect-sensitive bioassays showed C. maculata were exposed to bioactive Cry1F protein. The insecticidal protein had no effect on C. maculata larvae, even though larvae contained 20–32 ng of Cry1F/g by fresh weight. Over all, our results demonstrated that the Cry1F protein did not affect important fitness parameters of one of S. frugiperda ’s major predators and that Cry1F protein did not accumulate but was strongly diluted when transferred during trophic interactions.

Abstract:Geocoris punctipes (Say) and Orius insidiosus (Say) are generalist predators found in a wide range of crops, including cotton (Gossypium hirsutum L.) and maize (Zea mays L.), where they provide important biological control services by feeding on an array of pests, including eggs and small larvae of caterpillars. A high percentage of cotton and maize in the United States and several other countries are transgenic cultivars that produce one or more of the insecticidal Cry proteins of Bacillus thuringiensis Berliner (Bt). Here we quantify effects of three Cry proteins on the life history of these predators over two generations when they are exposed to these Cry proteins indirectly through their prey. To eliminate the confounding prey quality effects that can be introduced by Bt-susceptible prey, we used Cry1Ac/Cry2Ab-resistant Trichoplusia ni (Hübner) and Cry1 F-resistant Spodoptera frugiperda (J.E. Smith) in a series of tri-trophic studies. Survival, development, adult mass, fecundity, and fertility were similar when predators consumed larvae feeding on Cry1Ac/Cry2Ab cotton or Cry1 F maize compared with prey feeding on isogenic or near-isogenic cotton or maize. Repeated exposure of the same initial cohort over a second generation also resulted in no differences in life-history traits when feeding on non-Bt- or Bt-fed prey. Enzyme-linked immunosorbent assay showed that predators were exposed to Bt Cry proteins from their prey and that these proteins became increasingly diluted as they moved up the food chain. Results show a clear lack of effect of three common and widespread Cry proteins on these two important predator species. The use of resistant insects to eliminate prey quality effects provides a robust and meaningful assessment of exposure and hazard.

Abstract:The high-dose/refuge strategy has been the primary approach for resistance management in transgenic crops engineered with Bacillus thuringiensis toxins. However, there are continuing pressures from growers to reduce the size of Bt toxin-free refugia, which typically suffer higher damage from pests. One complementary approach is to release male transgenic insects with a female-specific self-limiting gene. This technology can reduce population sizes and slow the evolution of resistance by introgressing susceptible genes through males. Theory predicts that it could be used to facilitate smaller refugia or reverse the evolution of resistance. In this study, we used experimental evolution with caged insect populations to investigate the compatibility of the self-limiting system and the high-dose/refuge strategy in mitigating the evolution of resistance in diamondback moth, Plutella xylostella. The benefits of the self-limiting system were clearer at smaller refuge size, particularly when refugia were inadequate to prevent the evolution of resistance. We found that transgenic males in caged mesocosms could suppress population size and delay resistance development with 10% refugia and 4%-15% initial resistance allele frequency. Fitness costs in hemizygous transgenic insects are particularly important for introgressing susceptible alleles into target populations. Fitness costs of the self-limiting gene in this study (P. xylostella OX4139 line L) were incompletely dominant, and reduced fecundity and male mating competitiveness. The experimental evolution approach used here illustrates some of the benefits and pitfalls of combining mass release of self-limiting insects and the high-dose/refuge strategy, but does indicate that they can be complementary.

Abstract:Transgenic cotton plants producing Bacillus thuringiensis (Bt) (Berliner) insecticidal proteins have contributed to the management of key lepidopteran pests. Larval high dispersal rates between non-Bt and Bt plants in landscapes with seed contamination can speed the evolution of insect resistance to Bt plants. We evaluated the effect of Bt and non-Bt cotton plants on the larval dispersal pattern and survival of susceptible, heterozygous and Cry1F-resistant Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) genotypes in pure and contaminated artificial micro-landscape. A computer model was used to analyse the consequences of S. frugiperda larval dispersal behaviour on resistance evolution in refuge areas with different contamination levels and migration of different adult genotype combinations. The biological data from artificial micro-landscape experiments were used in simulations of macro-landscape scales. The Cry1F-resistant genotype avoided non-Bt cotton. The heterozygote had a similar larval dispersal behaviour as the susceptible genotype when non-Bt cotton was the central plant. Our simulations provide evidence that in refuge areas contaminated with Bt cotton plants, the evolution of resistance may be > 75-fold faster in relation to a contamination-free refuge. In conclusion, S. frugiperda resistance management practices on regional scales with contamination-free refuges are important to prevent loss in different crops.

Abstract:Abstract The polyphagous pest Helicoverpa zea (Lepidoptera: Noctuidae) has evolved practical resistance to transgenic corn and cotton producing Cry1 and Cry2 crystal proteins from Bacillus thuringiensis (Bt) in several regions of the United States. However, the Bt vegetative insecticidal protein Vip3Aa produced by Bt corn and cotton remains effective against this pest. To advance knowledge of resistance to Vip3Aa, we selected a strain of H. zea for resistance to Vip3Aa in the laboratory. After 28 generations of continuous selection, the resistance ratio was 267 for the selected strain (GA-R3) relative to a strain not selected with Vip3Aa (GA). Resistance was autosomal and almost completely recessive at a concentration killing all individuals from GA. Declines in resistance in heterogeneous strains containing a mixture of susceptible and resistant individuals reared in the absence of Vip3Aa indicate a fitness cost was associated with resistance. Previously reported cases of laboratory-selected resistance to Vip3Aa in lepidopteran pests often show partially or completely recessive resistance at high concentrations and fitness costs. Abundant refuges of non-Bt host plants can maximize the benefits of such costs for sustaining the efficacy of Vip3Aa against target pests.

Genome evolution in an agricultural pest following adoption of transgenic crops

Abstract:Abstract Replacement of synthetic insecticides with transgenic crops for pest management has been both economically and environmentally beneficial. These benefits have often eroded as pests evolved resistance to transgenic crops, but a broad understanding of the timing and complexity of adaptive changes which lead to field-evolved resistance in pest species is lacking. Wild populations of Helicoverpa zea , a major lepidopteran crop pest and the target of transgenic Cry toxin-expressing cotton and corn, have recently evolved widespread, damaging levels of resistance. Here, we quantified patterns of genomic change in wild H. zea collected between 2002 and 2017 when adoption rates of Cry-expressing crops expanded in North America. Using a combination of genomic approaches, we identified significant temporal changes in allele frequency throughout the genomes of field-collected H. zea . Many of these changes occurred concurrently with increasingly damaging levels of resistance to Cry toxins between 2012 and 2016, in a pattern consistent with polygenic selection. Surprisingly, none of the eleven previously described Cry resistance genes showed signatures of selection in wild H. zea . Furthermore, we observed evidence of a very strong selective sweep in one region of the H. zea genome, yet this strongest change was not additively associated with Cry resistance. This first, whole genome analysis of field-collected specimens to study evolution of Cry resistance demonstrates the potential and need for a more holistic approach to examining pest adaptation to changing agricultural practices. Significance Statement Evolution of pest resistance to management approaches in agricultural landscapes is common and results in economic losses. Early detection of pest resistance evolution prior to significant crop damage would benefit the agricultural community. It has been hypothesized that new genomic approaches could track molecular signals of emerging resistance problems and trigger efforts to pre-empt widespread damage. We tested this hypothesis by quantifying genomic changes in the pest Helicoverpa zea over a 15 year period concurrent with commercialization and subsequent loss of efficacy of transgenic Bt-expressing crops. Our results demonstrate the complex nature of evolution in agricultural ecosystems and provide insight into the potential for and pitfalls associated with use of genomic approaches for resistance monitoring. We discuss approaches for improvement.
Spatio-Temporal Variation in Landscape Composition May Speed Resistance Evolution of Pests to Bt Crops

Abstract:Transgenic crops that express insecticide genes from Bacillus thuringiensis (Bt) are used worldwide against moth and beetle pests. Because these engineered plants can kill over 95% of susceptible larvae, they can rapidly select for resistance. Here, we use a model for a pyramid two-toxin Bt crop to explore the consequences of spatio-temporal variation in the area of Bt crop and non-Bt refuge habitat. We show that variability over time in the proportion of suitable non-Bt breeding habitat, Q, or in the total area of Bt and suitable non-Bt habitat, K, can increase the overall rate of resistance evolution by causing short-term surges of intense selection. These surges can be exacerbated when temporal variation in Q and/or K cause high larval densities in refuges that increase density-dependent mortality; this will give resistant larvae in Bt fields a relative advantage over susceptible larvae that largely depend on refuges. We address the effects of spatio-temporal variation in a management setting for two bollworm pests of cotton, Helicoverpa armigera and H. punctigera, and field data on landscape crop distributions from Australia. Even a small proportion of Bt fields available to egg-laying females when refuges are sparse may result in high exposure to Bt for just a single generation per year and cause a surge in selection. Therefore, rapid resistance evolution can occur when Bt crops are rare rather than common in the landscape. These results highlight the need to understand spatio-temporal fluctuations in the landscape composition of Bt crops and non-Bt habitats in order to design effective resistance management strategies.

Abstract:High dispersal of Lepidoptera larvae between non-Bt and Bt cotton plants can favour the evolution of insect resistance; however, information on host acceptance of neonates in tropical transgenic crops is scarce. Therefore, the purposes of this study were as follows: (i) to investigate the feeding behaviour of susceptible and Cry1F-resistant strains of Spodoptera frugiperda (J.E. Smith) on Bt and non-Bt cotton (Gossypium hirsutum L.) varieties and (ii) to understand the possible effects of cotton field contamination on the dispersal and infestation capacity of S. frugiperda larvae by using an individual-based model. The main results of this paper are as follows: (1) the highest post-feeding larval dispersal of the Cry1F-resistant strain occurred at an exposure time of 18–24 h; (2) via video tracking assays, we found that the least distance moved was by larvae resistant to Cry1F on non-Bt cotton; and (3) the model indicated differences in mobility capacity between Bt and non-Bt cotton. We conclude that resistant neonates exhibit sedentary behaviour. Our report represents the first findings concerning the fitness cost of larval behaviour traits of S. frugiperda associated with Cry1F resistance in Brazilian populations.

Abstract:Ecological impact of transgene flow into populations of wild/weedy relatives is associated with fitness effects in hybrid progeny. Most studies assessing fitness effects focus essentially on early-generation hybrid progeny. However, whether the transgenes remain effective and durable in advanced generations of hybrid progeny remains unclear. We conducted a common garden experiment with F5–F7 hybrid progeny derived from crosses between insect-resistant transgenic (Bt/CpTI) rice and weedy rice, to examine their insect resistance and fitness effects of transgenes on progeny. Hybrid progeny were grown under different insect pressures and cultivation modes where insect damage and fitness-related traits were measured in the same growth season. Plants with transgenes showed significantly lower insect damage (10% vs. 32%) and higher fecundity (551 vs. 392 seeds/plant) than those without transgenes in F5–F7 populations, suggesting the efficacy of transgenes for insect resistance. Fitness benefits of the transgenes were similar among the F5–F7 populations, indicating the stability of transgenic effects. A positive correlation between insect index and fecundity change was detected, stressing the important role of ambient insect pressures in assessing fitness effects caused by insect-resistance transgenes. Our results have important implications for assessing ecological impacts caused by transgene flow to wild/weedy relatives. For cost-effectiveness, the experimental estimation of fitness effects is probably sufficient based on data from hybrids in early generations. Given that fitness effects of insect-resistance transgenes are associated with ambient insect pressure, ecological risk assessment on transgene flow should consider this variable in experimental design, reasonably reflecting actual situations in wild/weedy populations.

Abstract:The corn earworm/bollworm, Helicoverpa zea (Boddie), is a pest species that is targeted by both Bacillus thuringiensis (Bt) maize and cotton in the United States. Cry1Ab and Vip3Aa20 are two common Bt toxins that are expressed in transgenic maize. The objective of this study was to determine the resistance allele frequency (RAF) to Cry1Ab and Vip3Aa20 in H. zea populations that were collected during 2018 and 2019 from four southeastern U.S. states: Louisiana, Mississippi, Georgia, and South Carolina. By using a group-mating approach, 104 F-2 iso-lines of H. zea were established from field collections with most iso-lines (85) from Louisiana. These F-2 iso-lines were screened for resistance alleles to Cry1Ab and Vip3Aa20, respectively. There was no correlation in larval survivorship between Cry1Ab and Vip3Aa20 when the iso-lines were exposed to these two toxins. RAF to Cry1Ab maize was high (0.256) and the RAFs were similar between Louisiana and the other three states and between the two sampling years. In contrast, no functional major resistance allele (RA) that allowed resistant insects to survive on Vip3Aa20 maize was detected and the expected RAF of major RAs with 95% probability was estimated to 0 to 0.0073. However, functional minor RAs to Vip3Aa20 maize were not uncommon; the estimated RAF for minor alleles was 0.028. The results provide further evidence that field resistance to Cry1Ab maize in H. zea has widely occurred, while major RAs to Vip3Aa20 maize are uncommon in the southeastern U.S. region. Information that was generated from this study should be useful in resistance monitoring and refinement of resistance management strategies to preserve Vip3A susceptibility in H. zea.

Abstract:Abstract Under ideal conditions, widely adopted transgenic crop pyramids producing two or more distinct insecticidal proteins from Bacillus thuringiensis (Bt) that kill the same pest can substantially delay evolution of resistance by pests. However, deviations from ideal conditions diminish the advantages of such pyramids. Here, we tested the hypothesis that changes in maturing cotton producing Cry1Ac and Cry2Ab affect evolution of resistance in Helicoverpa zea (Boddie) (Lepidoptera: Noctuidae), a pest with low inherent susceptibility to both toxins. In terminal leaves of field-grown Bt cotton, the concentration of both toxins was significantly higher for young, squaring plants than for old, fruiting plants. We used laboratory bioassays with plant material from field-grown cotton to test H. zea larvae from a strain selected for resistance to Cry1Ac in the laboratory, its more susceptible parent strain, and their F1 progeny. On young Bt cotton, no individuals survived to pupation. On old Bt cotton, survival to pupation was significantly higher for the lab-selected strain and the F1 progeny relative to the unselected parent strain, indicating dominant inheritance of resistance. Redundant killing, the extent to which insects resistant to one toxin are killed by another toxin in a pyramid, was complete on young Bt cotton, but not on old Bt cotton. No significant fitness costs associated with resistance were detected on young or old non-Bt cotton. Incorporation of empirical data into simulations indicates the observed increased selection for resistance on old Bt cotton could accelerate evolution of resistance to cotton producing Cry1Ac and Cry2Ab in H. zea.

Abstract:Abstract Fitness costs associated with insect resistance to insecticides can be exploited to implement resistance management programs. However, most of these studies are restricted to evaluating biological traits on artificial diets. Here, we investigated the fitness cost associated with chlorantraniliprole in Spodoptera frugiperda (J.E. Smith) feeding on corn, soybean, and cotton plants. We used a near-isogenic strain of S. frugiperda resistant to chlorantraniliprole (Iso-RR), a susceptible strain (SS), and heterozygotes strains (H1 and H2) to evaluate several biological and population growth parameters. Larval survival of the Iso-RR strain was on average 90% on corn, 65% on soybean, and 57% on cotton plants. Development time of the larval stage also differed among host plants, Iso-RR strain took on average 14, 17, and 26 days to reach the pupal stage on corn, soybean, and cotton plants respectively. Net reproductive rate, intrinsic rate of population increase, and finite rate of population increase were higher for Iso-RR strain feeding on corn plants than other host plants. The relative fitness, based on the intrinsic rate of population increase, of S. frugiperda resistant strain on corn, soybean, and cotton plants were 1.04, 0.85, and 0.88, respectively. Therefore, no fitness cost was observed for S. frugiperda feeding on corn plants, but a significant fitness cost was observed when this pest fed on soybean and cotton plants. We showed that the food source influences the fitness cost of S. frugiperda resistant to diamide. Such information may help to implement resistance management strategies based on each crop.

Abstract:The corn earworm, Helicoverpa zea (Boddie) (Lepidoptera: Noctuidae), preferentially oviposits and feeds on ears of corn (Zea mays L.) and can be managed using transgenic hybrids that produce insecticidal proteins from the bacterium Bacillus thuringiensis (Bt). Concentrations of Bt proteins can vary spatially and temporally in plant tissues, creating a heterogeneous environment that can increase the risk of resistance development. We planted small-plot trials of nine Bt and non-Bt corn hybrids in South Carolina in 2016 and 2017 and investigated the development, survival, feeding injury, and feeding behavior in corn ear tissues. ELISA was used to quantify the concentrations of Cry1F and Cry2Ab2 in young silk, old silk, maternal tip tissue, kernels, and husk. Cry1F and Cry2Ab2 significantly varied with silk age and both proteins were generally highest in the silk and tip tissue. Hybrids with pyramided proteins significantly reduced feeding injury to the silk, tip, and kernel ear tissues, which was less apparent with single Bt protein hybrids. The pyramided hybrid expressing Vip3A incurred no injury to either the ear tip or kernels, and only eight 1st instar larvae were collected in the silk of 520 sampled ears. Age of larvae significantly varied among ear tissues but not between hybrids. Depending on hybrid family, mean larval instar in the silk, tip, and kernels was 1st or 2nd, 3rd, and 5th, respectively. Instar-specific feeding penetrance into corn ears increased with age but did not differ between hybrids. We characterized the instar- and tissue-specific feeding behavior of H. zea larvae but did not detect differences in feeding behavior between Bt and non-Bt hybrids. Implications for resistance management strategies such as seed mixtures are discussed.

Most studies assessing fitness effects focus essentially on early-generation hybrid progeny. However, whether the transgenes remain effective and durable in advanced generations of hybrid progeny remains unclear. We conducted a common garden experiment with F5–F7hybrid progeny derived from crosses between insect-resistant transgenic(Bt/Cp TI) rice and weedy rice, to examine their insect resistance and fitness effects of transgenes on progeny. Hybrid progeny were grown under different insect pressures and cultivation modes where insect damage and fitness-related traits were measured in the same growth season. Plants with transgenes showed significantly lower insect damage(10 % vs.32 %) and higher fecundity(551 vs. 392 seeds/plant) than those without transgenes in F5–F7populations, suggesting the efficacy of transgenes for insect resistance. Fitness benefits of the transgenes were similar among the F5–F7populations, indicating the stability of transgenic effects. A positive correlation between insect index and fecundity change was detected, stressing the important role of ambient insect pressures in assessing fitness effects caused by insect-resistance transgenes. Our results have importantimplications for assessing ecological impacts caused by transgene flow to wild/weedy relatives. For cost-effectiveness, the experimental estimation of fitness effects is probably sufficient based on data from hybrids in early generations. Given that fitness effects of insect-resistance transgenes are associated with ambient insect pressure,ecological risk assessment on transgene flow should consider this variable in experimental design, reasonably reflecting actual situations in wild/weedy populations.

Abstract:Abstract Diabroticite corn rootworms are significant maize, Zea mays L., pests that have evolved resistance to cultural and chemical management methods. Transgenic maize synthesizing a rootwormtoxic protein originating from the bacterium Bacillus thuringiensis Berliner is an important new management technology. Its use requires a refuge of nontransgenic maize to delay evolution of resistance by generating susceptible beetles capable of copulating with any resistant individuals surviving on the transgenic crop. The evolution of such resistance may concurrently influence a variety of fitness-related characters. Here, we examined multiple mating by female Diabrotica barberi Smith & Lawrence (Coleoptera: Chrysomelidae) in relation to the following fitness-related traits: female longevity, fecundity, and body size of males and females at the first copulation. For the first copulation, large and small males were reciprocally paired to large and small females for comparison with average-sized beetles. Once-mated females were then maintained under conditions conducive to oviposition and paired with averaged-sized males at weekly intervals until death. Although female age at second copulation varied significantly with size category, female age at first copulation, longevity and fecundity did not. Females that mated multiple times lived longer and laid more eggs than did those that mated once. The results suggest that resistance management models may need to consider those females deriving fitness benefits from a second mating.

Abstract:BackgroundHelicoverpa armigera and Helicoverpa zea are major caterpillar pests of Old and New World agriculture, respectively. Both, particularly H. armigera, are extremely polyphagous, and H. armigera has developed resistance to many insecticides. Here we use comparative genomics, transcriptomics and resequencing to elucidate the genetic basis for their properties as pests.ResultsWe find that, prior to their divergence about 1.5 Mya, the H. armigera/H. zea lineage had accumulated up to more than 100 more members of specific detoxification and digestion gene families and more than 100 extra gustatory receptor genes, compared to other lepidopterans with narrower host ranges. The two genomes remain very similar in gene content and order, but H. armigera is more polymorphic overall, and H. zea has lost several detoxification genes, as well as about 50 gustatory receptor genes. It also lacks certain genes and alleles conferring insecticide resistance found in H. armigera. Non-synonymous sites in the expanded gene families above are rapidly diverging, both between paralogues and between orthologues in the two species. Whole genome transcriptomic analyses of H. armigera larvae show widely divergent responses to different host plants, including responses among many of the duplicated detoxification and digestion genes.ConclusionsThe extreme polyphagy of the two heliothines is associated with extensive amplification and neofunctionalisation of genes involved in host finding and use, coupled with versatile transcriptional responses on different hosts. H. armigera’s invasion of the Americas in recent years means that hybridisation could generate populations that are both locally adapted and insecticide resistant.

Abstract:The emergence of Bt resistance among key insect pests (especially Noctuidae) is a major threat to the deployment of transgenic (Bt) cotton crops, used to combat these pests, worldwide. Refuge crops are, therefore, commonly grown in association with Bt cotton to generate large numbers of Bt‐susceptible insects, thus reducing the risk of Bt resistance. In Australian Bt cotton production systems, where the primary pests are Helicoverpa moths, refuge crops are mandatory, and pigeon pea and non‐Bt cotton are the refuge options available to growers. When Bt cotton was first deployed (in the mid 1990s), pigeon pea was assessed as having twice the capacity to produce Helicoverpa moths as unsprayed, non‐Bt cotton. Only half the amount (in area) of pigeon pea was, therefore, required to be grown as a refuge compared with unsprayed, non‐Bt cotton. The agronomy, insecticide use, varieties of Bt cotton (now based on two Bt genes (Bollgard II®), compared with the original single gene (Ingard®) varieties) and farmer profit margins have, however, changed radically since then. This has led to questioning of the continued relativities and best management practices involving these refuge crops. This paper assesses the performance records of pigeon pea and non‐Bt cotton refuges in the cotton production systems of eastern Australia since 1996. Pupae abundance is used as a surrogate for moth production. Pigeon pea has maintained its substantial superiority over unsprayed, non‐Bt cotton as a refuge generator of Helicoverpa throughout both the Ingard and Bollgard II eras. This is supported by field counts of both live pupae and empty pupal cases left behind in the soil when moths have emerged. There was some evidence that Helicoverpa production has decreased in time in both pigeon pea and cotton refuges. The incidence of parasitism of pupae has increased from the Ingard to the Bollgard II era. This latter change may, at least in part, explain a reduction in Helicoverpa productivity from refuges in recent years. There was no evidence to support a difference in parasitism of Helicoverpa between the two refuge crop types, but a temporal shift in parasite community (Tachinidae becoming more common) was apparent.

Abstract:The density of Helicoverpa armigera (Hübner) populations on Bacillus thuringiensis Berliner (Bt) transgenic cotton, corn, peanut, and soybean; differences in its development on Bt cotton and common (nontransgenic) cotton; and the potential for mating among populations from Bt cotton fields and other crop fields were investigated in the suburbs of Xinxiang City (Henan Province) and Langfang City (Hebei Province) in the southern and northern parts of north China, respectively. Although development of H. armigera on Bt cotton was much slower than on common cotton, there was a still high probability of mating between populations from Bt cotton and other sources due to the scattered emergence pattern of H. armigera adults, and overlap of the second and third generations. In a cotton and corn growing region, early and late planted corn provided suitable refugia for the third and fourth generations of H. armigera, but not for the second generation. In a cotton and soybean/ peanut mix system, noncotton crops provided a natural refugia from the second- to fourth-generation H. armigera, but function of the refuge would closely depend on the proportion of Bt cotton. Consequently, it may be necessary to compensate the original mixed cropping patterns in different areas for delaying resistance development of H. armigera to Bt cotton.

Abstract:In order to explore the stabilities of resistance to Bacillus thuringiensis(Bt) Cry1Ac toxin in cotton bollworm Helicoverpa armigera(Hübner) and its changes of relative fitness,the resistance regressions of Cry1Ac-resistant strain after transferring to normal artificial diet and its resistance recovers after re-selection were determined using bioassay test,the differences of relative fitness among susceptible,resistant and regressive strains were also compared by measuring their biological characters.The results showed that the resistance of resistant strain(BtR,which had high resistance to Cry1Ac) decreased very rapidly when lost selection pressure,the resistance ratio reduced from 3626.67-fold to 1436.67-fold after 4 generations,the resistance ratio reached to less than 10-fold till 12th generation,after that it kept a relative stable low resistance level(BtR-D,resistance ratio was 6.67-fold).While the H.armigera in this BtR-D strain was re-selected using Cry1Ac,the resistance regained quickly and the resistance ratio in-creased to 1123.33-fold after reselection for 5 generations.Compared with susceptible strain(96S),the fitness of BtR was obviously lower,the relative fitness was only 0.33.However,after continuously reared on normal artificial diet for 14 generations,the fitness obviously increased,the relative fitness was 0.87,the major features were egg hatchability and survival rate of larvae were enhanced.
