Abstract:Developing sensory neurons require neurotrophic support for survival, neurite outgrowth and myelination. The low-density lipoprotein receptor-related protein-1 (LRP1) transactivates Trk receptors and thereby functions as a putative neurotrophin. Herein, we show that LRP1 is abundantly expressed in developing dorsal root ganglia (DRG) and that LRP1-dependent cell signaling supports survival, neurite extension and receptivity to Schwann cells even in the absence of neurotrophins. Cultured embryonic DRG neurons (E15) were treated with previously characterized LRP1 ligands, LRP1-receptor binding domain of α2-macroglobulin (RBD), hemopexin domain of MMP-9 (PEX) or controls (GST) for two weeks. These structurally diverse LRP1 ligands significantly activated and sustained extracellular signal-regulated kinases (ERK1/2) 5-fold (p<0.05), increased expression of growth-associated protein-43(GAP43) 15-fold (P<0.01), and increased neurite outgrowth 20-fold (P<0.01). Primary sensory neurons treated with LRP1 ligands survived > 2 weeks in vitro, to an extent equaling NGF, a finding associated with canonical signaling mechanisms and blockade of caspase-3 cleavage. LRP1 ligand-induced survival and sprouting were blocked by co-incubation with the LRP1 antagonist, receptor associated protein (RAP), whereas RAP had no effect on NGF-induced activity. Site directed mutagenesis of the LRP1 ligand, RBD, in which Lys(1370) and Lys(1374) are converted to alanine to preclude LRP1 binding, were ineffective in promoting cell signaling, survival or inducing neurite extension in primary sensory neurons, confirming LRP1 specificity. Furthermore, LRP1-induced neurite sprouting was mediated by Src-family kinase (SFK) activation, suggesting transactivation of Trk receptors. Co-cultures of primary embryonic neurons and Schwann cells showed that LRP1 agonists promoted axonal receptivity to myelination to Schwann cells. Collectively, these findings identify LRP1 as a novel and perhaps essential trophic molecule for sensory neuronal survival and development.

Abstract:During recent years there has been much progress in interpreting the histo- and cytomorphology of lymphoreticular proliferations by means of enzyme cytochemical (cell typing by hydrolytic enzymes) and immunological (B- and T-cell differentiation using surface markers and functional tests) methods. Applying these methods in skin biopsies from 101 patients clinically suspected of having cutaneous lymphomas, patterns of lymphoreticular infiltrations in the skin have been elaborated. Based primarily on the 'Kiel' classification, low-grade and high-grade malignant lymphomas, pseudolymphomas and 'histiocytic lymphomas' can be differentiated in the skin; however, there still remain some hitherto unclassifiable lymphoreticular proliferations. In the low-grade malignant lymphomas of the skin, mycosis fungoides, Sézary's syndrome and Pagetoid reticulosis histologically display a pattern which is typical for T-cell infiltrations in the skin, whereas most of the 'malignant reticuloses', including immunocytoma, show a B-cell pattern. Erythrocyte antibody complement rosette fixation on cryostat sections is positive only in cutaneous pseudolymphomas whereas no fixation is seen in malignant B-cell lymphomas of the skin.
Phase‐resolved Faraday rotation in pulsars

Abstract:We have detected significant rotation measure (RM) variations for nine bright pulsars, as a function of pulse longitude. An additional sample of 10 pulsars showed a rather constant RM with phase, yet a small degree of RM fluctuation is visible in at least three of those cases. In all cases, we have found that the rotation of the polarization position angle across our 1.4 GHz observing band is consistent with the λ 2 law of interstellar Faraday rotation. We provide for the first time convincing evidence that RM variations across the pulse are largely due to interstellar scattering, although we cannot exclude that magnetospheric Faraday rotation may still have a minor contribution; alternative explanations of this phenomenon, like erroneous de-dispersion and the presence of non-orthogonal polarization modes, are excluded. If the observed, phase-resolved RM variations are common amongst pulsars, then many of the previously measured pulsar RMs may be in error by as much as a few tens of rad m ―2 .

Abstract:The inhalation of small quantities of carbon-11 labeled carbon monoxide results in a useful red cell label to identify vascular structures in the body and quantitate red cell concentration in tissues. Transverse section images are obtained using PC-II, the Massachusetts General Hospital (MGH) Positron Camera. Attenuation correction is performed either by means of transmission data or by analytical methods. Sets of transverse section images give information on red cell distribution in head and torso. Such images may be useful in identifying vascular abnormalities or in quantitating red cell physiology.
An Invariant for Wheel-Generated Motions and the Logic of its Determination

Abstract:Observers appear to perceive the paths of abstract centers of point-light configurations in making judgments about movement. For configurations on rolling wheels a metric was derived that described the relative vertical motion of this point. It was hypothesized that the smaller the metric the more the stimulus should appear to move in a wheel-like manner with linear translation. In two experiments observers viewed pairs of stimuli and were asked to select either the event that appeared most wheel-like or the one that hopped the most. Viewers consistently selected the stimulus with the smaller metric as being more wheel-like, with a frequency that increased with the difference between metrics. The inverse of this pattern was found for those observers requested to select the stimulus that hopped most. In a second set of two experiments observers drew the translationl paths of these stimuli. Their drawings corresponded to the motion paths of configural centroids. Together, these results strongly suggest that observers perceive the translational component of the motion of the configurations as the path described by their centroids, or geometric centers. We propose that this description of the stimulus event is determined by the logical ordering of information extraction provided by the perceptual system, and discuss this logic and cases where it seems evident.

Abstract:Abstract Diagnosing cervical spine injury in children can be difficult because the clinical examination can be unreliable, and evidence-based consensus guidelines for cervical spine injury evaluation in children have not been established. However, the consequences of cervical spine injuries are significant. Therefore, practitioners should understand common patterns of cervical spine injury in children, the evidence and indications for cervical spine imaging, and which imaging modalities to use. Herein, we review the epidemiology and unique anatomical features of pediatric cervical spine injury. In addition, we will summarize current practice for clearance and imaging of the pediatric cervical spine in trauma.

Abstract:LDL Receptor-related Protein-1 (LRP1/CD91) binds diverse ligands, many of which activate cell-signaling. Herein, we compared three LRP1 ligands that inhibit inflammatory responses triggered by lipopolysaccharide (LPS), including: enzymatically-inactive tissue-type plasminogen activator (EI-tPA); activated α2-macroglobulin (α2M); and S-PrP, a soluble derivative of nonpathogenic cellular prion protein (PrPC). In bone marrow-derived macrophages, the N-methyl-D-aspartate receptor was essential for all three LRP1 ligands to activate cell-signaling and inhibit LPS-induced cytokine expression. Intact lipid rafts also were essential. Only α2M absolutely required LRP1. LRP1 decreased the EI-tPA concentration required to activate cell-signaling and antagonize LPS but was not essential, mimicking its role as a S-PrP co-receptor. Membrane-anchored PrPC also functioned as a co-receptor for EI-tPA and α2M, decreasing the ligand concentration required for cell-signaling and LPS antagonism; however, when the concentration of EI-tPA or α2M was sufficiently increased, cell-signaling and LPS antagonism occurred independently of PrPC. S-PrP is the only LRP1 ligand in this group that activated cell-signaling independently of membrane-anchored PrPC. EI-tPA, α2M, and S-PrP inhibited LPS-induced LRP1 shedding from macrophages, a process that converts LRP1 into a pro-inflammatory product. Differences in the co-receptors required for anti-inflammatory activity may explain why LRP1 ligands vary in ability to target macrophages in different differentiation states.
BDNF-induced recruitment of TrkB receptor into neuronal lipid rafts

Abstract:Brain-derived neurotrophic factor (BDNF) plays an important role in synaptic plasticity but the underlying signaling mechanisms remain unknown. Here, we show that BDNF rapidly recruits full-length TrkB (TrkB-FL) receptor into cholesterol-rich lipid rafts from nonraft regions of neuronal plasma membranes. Translocation of TrkB-FL was blocked by Trk inhibitors, suggesting a role of TrkB tyrosine kinase in the translocation. Disruption of lipid rafts by depleting cholesterol from cell surface blocked the ligand-induced translocation. Moreover, disruption of lipid rafts prevented potentiating effects of BDNF on transmitter release in cultured neurons and synaptic response to tetanus in hippocampal slices. In contrast, lipid rafts are not required for BDNF regulation of neuronal survival. Thus, ligand-induced TrkB translocation into lipid rafts may represent a signaling mechanism selective for synaptic modulation by BDNF in the central nervous system.
LRP1 Triggers Pro-inflammatory Cell-signaling in Response to Extracellular Tau Independently of the NMDA Receptor

Abstract:In Alzheimer's Disease (AD) and other neurodegenerative diseases, microtubule-associated protein Tau forms abnormal intracellular aggregates. The mechanisms by which Tau may promote AD progression remain incompletely understood. Injured and dying neurons release Tau into the extracellular spaces in the CNS. The released Tau may be taken up by receptors in the LDL Receptor gene family, including Low Density Lipoprotein Receptor-related Protein-1 (LRP1), which is expressed by microglia, astrocytes, and neurons. This process may be important for clearing Tau from extracellular spaces but may also promote the seeding of Tau aggregates in new cells. Our laboratory has shown that endocytosis of LRP1 ligands is coupled to the activation of cell-signaling and regulation of inflammation. Notably, different LRP1 ligands can induce either pro-inflammatory or anti-inflammatory responses, depending on the co-receptors that function with LRP1. Here, we demonstrate that in cultured macrophages, microglia, and astrocytes, extracellular Tau induces an LRP1-dependent pro-inflammatory response, characterized by NFκB activation and expression of pro-inflammatory cytokines. Unlike other LRP1 ligands that elicit anti-inflammatory responses, the response to Tau occurs independently of the NMDA receptor. When LRP1 is deleted or silenced, macrophages, microglia, and astrocytes do not respond to Tau, whereas when is deleted or the NMDA-R is pharmacologically inhibited, the responses remain unchanged. Because we have evidence that LRP1 in microglia expresses anti-inflammatory activity in response to ligands other than Tau, understanding the role of LRP1 in microglia and astrocytes in Alzheimer's Disease and other neuroinflammatory processes is an important future goal.

Abstract:ABSTRACT Proper control of epidermal growth factor receptor (EGFR) signaling is important for maintaining cellular homeostasis. Given that EGFR signaling occurs at the plasma membrane and endosomes following internalization, endosomal trafficking of EGFR spatiotemporally regulates EGFR signaling. In this process, leucine-rich repeat kinase 1 (LRRK1) has multiple roles in kinase activity-dependent transport of EGFR-containing endosomes and kinase-independent sorting of EGFR into the intraluminal vesicles (ILVs) of multivesicular bodies. Active, phosphorylated EGFR inactivates the LRRK1 kinase activity by phosphorylating Y944. In this study, we demonstrate that LRRK1 facilitates EGFR dephosphorylation by PTP1B (also known as PTPN1), an endoplasmic reticulum (ER)-localized protein tyrosine phosphatase, at the ER–endosome contact site, after which EGFR is sorted into the ILVs of endosomes. LRRK1 is required for the PTP1B–EGFR interaction in response to EGF stimulation, resulting in the downregulation of EGFR signaling. Furthermore, PTP1B activates LRRK1 by dephosphorylating pY944 on the contact site, which promotes the transport of EGFR-containing endosomes to the perinuclear region. These findings provide evidence that the ER–endosome contact site functions as a hub for LRRK1-dependent signaling that regulates EGFR trafficking.

Abstract:Main Points Neuronal PACSIN1 is released in PNS injury and functions as a Schwann cell LRP1 ligand. PACSIN1 binding to LRP1 activates Schwann cell signaling to promote survival and migration. PACSIN1 signaling in Schwann cells requires TrkC transactivation. Schwann cells (SCs) undergo phenotypic transformation and then orchestrate nerve repair following PNS injury. The ligands and receptors that activate and sustain SC transformation remain incompletely understood. Proteins released by injured axons represent important candidates for activating the SC Repair Program. The low‐density lipoprotein receptor‐related protein‐1 (LRP1) is acutely up‐regulated in SCs in response to injury, activating c‐Jun, and promoting SC survival. To identify novel LRP1 ligands released in PNS injury, we applied a discovery‐based approach in which extracellular proteins in the injured nerve were captured using Fc‐fusion proteins containing the ligand‐binding motifs of LRP1 (CCR2 and CCR4). An intracellular neuron‐specific protein, Protein Kinase C and Casein Kinase Substrate in Neurons (PACSIN1) was identified and validated as an LRP1 ligand. Recombinant PACSIN1 activated c‐Jun and ERK1/2 in cultured SCs. Silencing Lrp1 or inhibiting the LRP1 cell‐signaling co‐receptor, the NMDA‐R, blocked the effects of PACSIN1 on c‐Jun and ERK1/2 phosphorylation. Intraneural injection of PACSIN1 into crush‐injured sciatic nerves activated c‐Jun in wild‐type mice, but not in mice in which Lrp1 is conditionally deleted in SCs. Transcriptome profiling of SCs revealed that PACSIN1 mediates gene expression events consistent with transformation to the repair phenotype. PACSIN1 promoted SC migration and viability following the TNFα challenge. When Src family kinases were pharmacologically inhibited or the receptor tyrosine kinase, TrkC, was genetically silenced or pharmacologically inhibited, PACSIN1 failed to induce cell signaling and prevent SC death. Collectively, these studies demonstrate that PACSIN1 is a novel axon‐derived LRP1 ligand that activates SC repair signaling by transactivating TrkC.

Abstract:The low-density lipoprotein related protein receptor 1 (LRP1), also known as CD91 or α-Macroglobulin-receptor, is a transmembrane receptor that interacts with more than 40 known ligands. It plays an important biological role as receptor of morphogens, extracellular matrix molecules, cytokines, proteases, protease inhibitors and pathogens. In the CNS, it has primarily been studied as a receptor and clearance agent of pathogenic factors such as Aβ-peptide and, lately, Tau protein that is relevant for tissue homeostasis and protection against neurodegenerative processes. Recently, it was found that LRP1 expresses the Lewis-X (Lex) carbohydrate motif and is expressed in the neural stem cell compartment. The removal of Lrp1 from the cortical radial glia compartment generates a strong phenotype with severe motor deficits, seizures and a reduced life span. The present review discusses approaches that have been taken to address the neurodevelopmental significance of LRP1 by creating novel, lineage-specific constitutive or conditional knockout mouse lines. Deficits in the stem cell compartment may be at the root of severe CNS pathologies.

[Spinal cord compression as the presenting form of non-Hodgkin's lymphoma].

Abstract:Spinal cord compression as first presentation of non-Hodgkin's lymphoma (NHL) is an uncommon event. Diagnosis of NHL usually is performed on a laminectomy specimen. Spinal cord compression has been reported in 5% of patients with solid tumors. Although this clinical picture has been considered as very unusual among NHL patients, some series regarding descompresive laminectomy indicate that NHL was the underlying cause in 15% of these cases. We report two cases of patients with NHL who presented paraparesia secondary to spinal cord compression due to lymphomatous mass in the epidural region which was showed by magnetic resonance imaging. The emergency laminectomy and treatment with chemotherapy allowed the clinical recuperation of both patients.

Abstract:Brain-derived neurotrophic factor (BDNF) binds to its cell surface receptor TrkB to regulate differentiation, development, synaptic plasticity, and functional maintenance of neuronal cells. Binding of BDNF triggers TrkB dimerization and autophosphorylation, which provides docking sites for adaptor proteins to recruit and activate downstream signaling molecules. The molecular mechanisms underlying BDNF-TrkB endocytic trafficking crucial for spatiotemporal control of signaling pathways remain to be elucidated. Here we show that retrolinkin, a transmembrane protein, interacts with endophilin A1 and mediates BDNF-activated TrkB (pTrk) trafficking and signaling in CNS neurons. We find that activated TrkB colocalizes and interacts with the early endosome marker APPL1. Both retrolinkin and endophilin A1 are required for BDNF-induced dendrite development and acute extracellular signal-regulated kinase activation from early endosomes. Suppression of retrolinkin expression not only blocks BDNF-triggered TrkB internalization, but also prevents recruitment of endophilin A1 to pTrk vesicles trafficking through APPL1-positive endosomes. These findings reveal a novel mechanism for BDNF-TrkB to regulate signaling both in time and space through a specific membrane trafficking pathway.
Clathrin-dependent Endocytosis Is Required for TrkB-dependent Akt-mediated Neuronal Protection and Dendritic Growth

Abstract:Endocytosis of Trk ((t) under bar ropomyosin-(r) under bar elated (k) under bar inase) receptors is critical for neurotrophin signal transduction and biological functions. However, the mechanism governing endocytosis of TrkB ((t) under bar ropomyosin-(r) under bar elated (k) under bar inase (B) under bar) and the specific contributions of TrkB endocytosis to downstream signaling are unknown. In this study, we report that blocking clathrin, dynamin, or AP2 in cultured neurons of the central nervous system inhibited brain-derived neurotrophic factor (BDNF)-induced activation of Akt but not ERK. Treating neurons with the clathrin inhibitor monodansylcadaverine or a peptide that blocks dynamin function specifically abrogated Akt pathway activation in response to BDNF but did not affect the response of other downstream effectors or the up-regulation of immediate early genes neuropeptide Y and activity-regulated cytoskeleton-associated protein. Similar effects were found in neurons expressing small interfering RNA to silence AP2 or a dominant negative form of dynamin that inhibits clathrin-mediated endocytosis. In PC12 cells, ERK but not Akt activation required TrkA endocytosis following stimulation with nerve growth factor, whereas the opposite was true when TrkA-expressing neurons were stimulated with nerve growth factor in the central nervous system. Thus, the specific effects of internalized Trk receptors probably depend on the presence of cell type-specific modulators of neurotrophin signaling and not on differences inherent to Trk receptors themselves. Endocytosis-dependent activation of Akt in neurons was found to be critical for BDNF-supported survival and dendrite outgrowth. Together, these results demonstrate the functional requirement of clathrin-and dynamin-dependent endocytosis in generating the full intracellular response of neurons to BDNF in the central nervous system.

Abstract:The low-density lipoprotein receptor-related protein 1 (LRP1) is a multifunctional endocytic receptor involved in the metabolism of various extracellular ligands, including proteinases, that play critical roles in tumor invasion. Although several studies have shown an increased expression of LRP1 in cancer cells, its function in tumor development and progression remains largely unclear. Here, we reveal a novel mechanism by which LRP1 induces the expression of matrix metalloproteinase 2 (MMP2) and MMP9 and thereby promotes the migration and invasion of human glioblastoma U87 cells. Knockdown of LRP1 expression greatly decreased U87 cell migration and invasion, which was rescued by the forced expression of a functional LRP1 minireceptor. Inhibition of ligand binding to LRP1 by a specific antagonist, receptor-associated protein, also led to reduced cancer cell migration and invasion. Because MMPs play critical roles in cancer cell migration and invasion, we examined the expression of several MMPs and found that the expression of functional MMP2 and MMP9 was selectively decreased in LRP1 knockdown cells. More importantly, decreased cell migration and invasion of LRP1 knockdown cells were completely rescued by exogenous expression of MMP2 or MMP9, suggesting that these MMPs are likely downstream targets of LRP1-mediated signaling. We further show that the level of phosphorylated extracellular signal-regulated kinase (ERK) was significantly decreased in LRP1-silenced cells, suggesting that ERK is a potential mediator of LRP1-regulated MMP2 and MMP9 expression in U87 cells. Together, our data strongly suggest that LRP1 promotes glioblastoma cell migration and invasion by regulating the expression and function of MMP2 and MMP9 perhaps via an ERK-dependent signaling pathway.
Regulation of TrkB receptor tyrosine kinase and its internalization by neuronal activity and Ca2+ influx.

Abstract:Internalization of the neurotrophin-Trk receptor complex is critical for many aspects of neurotrophin functions. The mechanisms governing the internalization process are unknown. Here, we report that neuronal activity facilitates the internalization of the receptor for brain-derived neurotrophic factor, TrkB, by potentiating its tyrosine kinase activity. Using three independent approaches, we show that electric stimulation of hippocampal neurons markedly enhances TrkB internalization. Electric stimulation also potentiates TrkB tyrosine kinase activity. The activity-dependent enhancement of TrkB internalization and its tyrosine kinase requires Ca2+ influx through N-methyl-d-aspartate receptors and Ca2+ channels. Inhibition of internalization had no effect on TrkB kinase, but inhibition of TrkB kinase prevents the modulation of TrkB internalization, suggesting a critical role of the tyrosine kinase in the activity-dependent receptor endocytosis. These results demonstrate an activity- and Ca2+-dependent modulation of TrkB tyrosine kinase and its internalization, and they provide new insights into the cell biology of tyrosine kinase receptors.
