Abstract:The present review was attempted to analyze the multiple channels of basal ganglia-thalamocortical connections, and the connections of their related nuclei. The prefrontal and motor areas consist of a number of modules, which seem to provide multiple subloops of the basal ganglia-thalamocortical connections in subhuman primates. There may be a great degree of convergence of the limbic, associative and motor loops at the level of the striatum, substantia nigra, pallidum, and the subthalamic nucleus as well as the pedunculopontine nucleus. Nigral dopaminergic neurons receive limbic input directly as well as indirectly through the striosomes in the striatum. Dopamine contributes to behavioral learning by signaling motivation and reinforcement. The pedunculopontine nucleus might be involved in behavioral state control, learning and reinforcement processes, locomotion and autonomic functions. Each subdivision of the motor areas receives a mixed and weighted transthalamic input from both the cerebellum and basal ganglia. In particular, based on the author's data, the hand/arm motor area and adjacent premotor area receive strong superficial basal ganglia-thalamocortical projections as well as the deep cerebello-thalamocortical projections. These areas, have very dense corticocotrical connections with other cortical areas, receive polymodal afferents from the parietal and temporal cortices, and integrated information, via multiple routes, from the prefrontal cortex. The author suggests that the ventrolateral part of the caudal medial pallidal segment (GPi) and the ventromedial part of the GPi are linked directly to these areas by ways of the oral part of ventral lateral nucleus (VLo) and the ventral part of the parvicellular part of ventral anterior nucleus (VApc), respectively. These connections are thought to be involved in the acquisition and coordination of motor sequences.
Basal ganglia-thalamocortical circuits: parallel substrates for motor, oculomotor, "prefrontal" and "limbic" functions

Abstract:The central theme of the "segregated circuits" hypothesis is that structural convergence and functional integration occurs within, rather than between, each of the identified circuits. Admittedly, the anatomical evidence upon which this scheme is based remains incomplete. The hypothesis continues to be predicated largely on comparisons of anterograde and retrograde labeling studies carried out in different sets of animals. Only in the case of the "motor" circuit has evidence for the continuity of the loop been demonstrated directly in individual subjects; for the other circuits, such continuity is inferred from comparisons of data on different components of each circuit obtained in separate experiments. Because of the marked compression of pathways leading from cortex through basal ganglia to thalamus, comparisons of projection topography across experimental subjects may be hazardous. Definitive tests of the hypothesis of maintained segregation await additional double- and multiple-label tract-tracing experiments wherein the continuity of one circuit, or the segregation of adjacent circuits, can be examined directly in individual subjects. It is worthy of note, however, that the few studies to date that have employed this methodology have generated results consistent with the segregated circuits hypothesis. Moreover, single cell recordings in behaving animals have shown striking preservation of functional specificity at the level of individual neurons throughout the "motor" and "oculomotor" circuits. It is difficult to imagine how such functional specificity could be maintained in the absence of strict topographic specificity within the sequential projections that comprise these two circuits. This is not to say, however, that we expect the internal structure of functional channels (e.g., the "arm" channel within the "motor" circuit) to have cable-like, point-to-point topography. When the grain of analysis is sufficiently fine, anatomical studies have shown repeatedly that the terminal fields of internuclear projections (e.g., to striatum, pallidum, nigra, thalamus, etc.) often appear patchy and highly divergent, suggesting that neighboring groups of projection cells tend to influence interdigitating clusters of postsynaptic neurons. While more intricate and complex than simple point-to-point topography, however, this type arrangement should also be capable of maintaining functional specificity. As discussed briefly above, it is not yet clear to what extent the inputs to the "motor" circuit from the different precentral motor fields (e.g., MC, SMA, APA) are integrated in their passage through the circuit. Abstract:Abstract The cortico–basal ganglia–thalamo–cortical loop is one of the fundamental network motifs in the brain. Revealing its structural and functional organization is critical to understanding cognition, sensorimotor behaviour, and the natural history of many neurological and neuropsychiatric disorders. Classically, this network is conceptualized to contain three information channels: motor, limbic and associative 1–4 . Yet this three-channel view cannot explain the myriad functions of the basal ganglia. We previously subdivided the dorsal striatum into 29 functional domains on the basis of the topography of inputs from the entire cortex 5 . Here we map the multi-synaptic output pathways of these striatal domains through the globus pallidus external part (GPe), substantia nigra reticular part (SNr), thalamic nuclei and cortex. Accordingly, we identify 14 SNr and 36 GPe domains and a direct cortico-SNr projection. The striatonigral direct pathway displays a greater convergence of striatal inputs than the more parallel striatopallidal indirect pathway, although direct and indirect pathways originating from the same striatal domain ultimately converge onto the same postsynaptic SNr neurons. Following the SNr outputs, we delineate six domains in the parafascicular and ventromedial thalamic nuclei. Subsequently, we identify six parallel cortico–basal ganglia–thalamic subnetworks that sequentially transduce specific subsets of cortical information through every elemental node of the cortico–basal ganglia–thalamic loop. Thalamic domains relay this output back to the originating corticostriatal neurons of each subnetwork in a bona fide closed loop.

Abstract:The striatum receives inputs from different areas of the cerebral cortex, including association cortical areas far on in the hierarchy of cortical information processing as well as the sensori-motor cortex, and has connections via the globus pallidus and substantia nigra to the thalamus and thence to premotor and prefrontal cortical areas. Recordings of the activity of neurons in different parts of the striatum of primates show that they have the following properties: 1) neurons in much of the putamen, which receives inputs from the sensori-motor cortex, have activity related to movements; 2) neurons in the caudate nucleus, which receives from the association cortex, have activity related for example to environmental stimuli which signal preparation for or initiation of behavioral responses; 3) neurons in the tail of the caudate nucleus, which receives strongly from the inferior temporal visual cortex, respond when a patterned visual stimulus changes; 4) some neurons in the posterior ventral putamen, which receives from the inferior temporal visual cortex and the prefrontal cortex, respond in a visual short term memory task, delayed match to sample. The neurons responded in the delay period, or differentially to match and non-match stimuli. These neurons did not respond in an auditory delayed match to sample task, so that their activity was not related to movement per se, but was instead more closely related to visual inputs relevant to a memory task; 5) some neurons in the ventral striatum (including the nucleus accumbens), which receives from limbic structures such as the amygdala and hippocampus, respond to stimuli associated with reinforcement or to novel stimuli.It is concluded that there is considerable segregation of function within the striatum. It is suggested that there is an opportunity for inputs which originate from different parts of the cerebral cortex to interact, via a first stage of convergence in the striatum, and by a further stage of convergence on the dendrites of single neurons in the globus pallidus and substantia nigra; and that both these parts of the basal ganglia may learn associations between the different signals they receive. The result of this convergence and learning is that the basal ganglia provide a way for cortical areas far on in the hierarchy of information processing to become linked during motor learning to particular sequences of movements, and thus to be involved in the execution of motor programs.
An Intelligent System for Retrieving Economic Information from Corporate Websites

Abstract:The prompt availability of up-to-date economic indicators is crucial to monitor the economy and to steer the design of policies for promoting business innovation and raising firm competitiveness. Economic indicators usually suffer important lags since they are commonly obtained from official databases or from interviews to a sample of agents, thus limiting the representative ness and usefulness of the information. In a context in which the presence of companies in the World Wide Web is almost an obligation to succeed, corporate websites are connected, in some way, to the firm economic activity. On the basis of this relation, this paper proposes an intelligent system that analyzes corporate websites to produce web indicators related to the economic activity of the firms. This system has been successfully implemented and applied to infer company size characteristics from data gathered from corporate websites. Our results show that relatively large companies provide web content in a foreign language and use proprietary web servers.

Abstract:Basal ganglia and the cerebellum are part of a densely interconnected network. While both subcortical structures process information in basically segregated loops that primarily interact in the neocortex, direct subcortical interaction has been recently confirmed by neuroanatomical studies using viral transneuronal tracers in non-human primate brains. The thalamus is thought to be the main relay station of both projection systems. Yet, our understanding of subcortical basal ganglia and cerebellar interconnectivity within the human thalamus is rather sparse, primarily due to limitation in the acquisition of in vivo tracing. Consequently, we strive to characterize projections of both systems and their potential overlap within the human thalamus by diffusion MRI and tractography. Our analysis revealed a decreasing anterior-to-posterior gradient for pallido-thalamic connections in: (1) the ventral-anterior thalamus, (2) the intralaminar nuclei, and (3) midline regions. Conversely, we found a decreasing posterior-to-anterior gradient for dentato-thalamic projections predominantly in: (1) the ventral-lateral and posterior nucleus; (2) dorsal parts of the intralaminar nuclei and the subparafascicular nucleus, and (3) the medioventral and lateral mediodorsal nucleus. A considerable overlap of connectivity pattern was apparent in intralaminar nuclei and midline regions. Notably, pallidal and cerebellar projections were both hemispherically lateralized to the left thalamus. While strikingly consistent with findings from transneuronal studies in non-human primates as well as with pre-existing anatomical studies on developmentally expressed markers or pathological human brains, our assessment provides distinctive connectional fingerprints that illustrate the anatomical substrate of integrated functional networks between basal ganglia and the cerebellum. Thereby, our findings furnish useful implications for cerebellar contributions to the clinical symptomatology of movement disorders.

Abstract:The neurobiological and functional organization of the prefrontal cortex and the striatum is reviewed in the context of parallel, functionally segregated anatomical 'loops'. Although cortical input converges to some extent within the striatum, particular striatal sectors project back to a subset of their cortical inputs via relays in the globus pallidus and thalamus. The control of striatal outflow by direct and indirect pathways and their modulation by striatal dopamine are described, and recent attempts to provide neurocomputational models of the striatum briefly reviewed. The possible functions of corticostriatal loops in the formation, maintenance, and shifting of cognitive set, as well as in reversal learning, are investigated using a paradigm related to the Wisconsin Card-Sorting Test (WCST) in variants for patients with frontal lobe lesions or basal ganglia disorders, for monkeys with selective and excitotoxic lesions, and for normal humans in functional imaging studies.
The Striatum and Subthalamic Nucleus as Independent and Collaborative Structures in Motor Control

Abstract:The striatum and the subthalamic nucleus (STN) are two separate input structures into the basal ganglia (BG). Accordingly, research to date has primarily focused on the distinct roles of these structures in motor control and cognition, often through investigation of Parkinson's disease (PD). Both structures are divided into sensorimotor, associative, and limbic subdivisions based on cortical connectivity. The more recent discovery of the STN as an input structure into the BG drives comparison of these two structures and their respective roles in cognition and motor control. This review compares the role of the striatum and STN in motor response inhibition and execution, competing motor programs, feedback based learning, and response planning. Through comparison, it is found that the striatum and STN have highly independent roles in motor control but also collaborate in order to execute desired actions. There is also the possibility that inhibition or activation of one of these structures indirectly contributes to the function of other connected anatomical structures. Both structures contribute to selective motor response inhibition, which forms the basis of many tasks, but the STN additionally contributes to global inhibition through the hyperdirect pathway. Research is warranted on the functional connectivity of the network for inhibition involving the rIFG, preSMA, striatum, and STN.

A cortico-subcortical loop for motor control via the pontine reticular formation

Abstract:MMovement and locomotion are controlled by large neuronal circuits like the cortex-basal ganglia (BG)-thalamus loop. Inhibitory output of the BG loop can directly control movement via specialized connections with the brainstem. Whether other parallel loops with similar logic exist is presently unclear. Here we demonstrate that glycine transporter 2-positive (GlyT2+) cells of the pontine reticular formation (PRF) receive cortical inputs and in turn innervate the thalamus. Thalamus-projecting GlyT2+ cells innervate subcortical regions distinct from BG targets. Cortical cells co-innervate PRF/GlyT2+ cells and thalamus as in the BG loops. Cortex exerts strong excitatory control on PRF/GlyT2+ cells and these neurons powerfully inhibit their thalamic targets. Activation of thalamus projecting PRF/GlyT2+ cells leads to contralateral turning. These results demonstrate that the PRF is part of a cortico-subcortical loop that regulates motor activity parallel to the BG circuits. The cortico-PRF-thalamus loop can synergistically control turning with the BG loops via distinct descending pathways.

A biologically constrained model of the whole basal ganglia addressing the paradoxes of connections and selection

Abstract:The basal ganglia nuclei form a complex network of nuclei often assumed to perform selection, yet their individual roles and how they influence each other is still largely unclear. In particular, the ties between the external and internal parts of the globus pallidus are paradoxical, as anatomical data suggest a potent inhibitory projection between them while electrophys-iological recordings indicate that they have similar activities. Here we introduce a theoretical study that reconciles both views on the intra-pallidal projection, by providing a plausible characterization of the relationship between the external and internal globus pallidus. Specifically, we developed a mean-field model of the whole basal ganglia, whose parameterization is optimized to respect best a collection of numerous anatomical and electrophysiological data. We first obtained models respecting all our constraints, hence anatomical and electrophysiological data on the intrapallidal projection are globally consistent. This model furthermore predicts that both aforementioned views about the intra-pallidal projection may be reconciled when this projection is weakly inhibitory, thus making it possible to support similar neural activity in both nuclei and for the entire basal ganglia to select between actions. Second, we predicts that afferent projections are substantially unbalanced towards the external segment, as it receives the strongest excitation from STN and the weakest inhibition from the striatum. Finally, our study strongly suggest that the intrapallidal connection pattern is not focused but diffuse, as this latter pattern is more efficient for the overall selection performed in the basal ganglia.

Abstract:Corticospinal neurons (CSNs) represent the direct cortical outputs to the spinal cord and play important roles in motor control across different species. However, their organizational principle remains unclear. By using a retrograde labeling system, we defined the requirement of CSNs in the execution of a skilled forelimb food-pellet retrieval task in mice. In vivo imaging of CSN activity during performance revealed the sequential activation of topographically ordered functional ensembles with moderate local mixing. Region-specific manipulations indicate that CSNs from caudal or rostral forelimb area control reaching or grasping, respectively, and both are required in the transitional pronation step. These region-specific CSNs terminate in different spinal levels and locations, therefore preferentially connecting with the premotor neurons of muscles engaged in different steps of the task. Together, our findings suggest that spatially defined groups of CSNs encode different movement modules, providing a logic for parallel-ordered corticospinal circuits to orchestrate multistep motor skills.
Neuronal connections of direct and indirect pathways for stable value memory in caudal basal ganglia

Abstract:Direct and indirect pathways in the basal ganglia work together for controlling behavior. However, it is still a controversial topic whether these pathways are segregated or merged with each other. To address this issue, we studied the connections of these two pathways in the caudal parts of the basal ganglia of rhesus monkeys using anatomical tracers. Our previous studies showed that the caudal basal ganglia control saccades by conveying long-term values (stable values) of many visual objects toward the superior colliculus. In experiment 1, we injected a tracer in the caudate tail (CDt), and found local dense plexuses of axon terminals in the caudal-dorsal-lateral part of substantia nigra pars reticulata (cdlSNr) and the caudal-ventral part of globus pallidus externus (cvGPe). These anterograde projections may correspond to the direct and indirect pathways, respectively. To verify this in experiment 2, we injected different tracers into cdlSNr and cvGPe, and found many retrogradely labeled neurons in CDt and, in addition, the caudal-ventral part of the putamen (cvPut). These cdlSNr-projecting and cvGPe-projecting neurons were found intermingled in both CDt and cvPut (which we call "striatum tail"). A small but significant proportion of neurons (<15%) were double-labeled, indicating that they projected to both cdlSNr and cvGPe. These anatomical results suggest that stable value signals (good vs. bad) are sent from the striatum tail to cdlSNr and cvGPe in a biased (but not exclusive) manner. These connections may play an important role in biasing saccades toward higher valued objects and away from lower valued objects.

Abstract:The functional architecture of the basal ganglia make an interesting comparison with the neocortex, for the basal ganglia do not use excitatory recurrent collaterals between the neurons. Instead the much evolutionarily older basal ganglia use direct inhibition between the neurons at the striatal and at the pallidal / nigral stages of processing. In comparison with cortex, this is a much safer method of selection, and of ensuring that only one output is selected, which is important in motor systems so that contradictory outputs are not attempted, for there is no risk of positive feedback becoming uncontrolled and runaway with the striatal direct mutual inhibition by neurons. The basal ganglia do have regular principles of internal connectivity between the neurons which lead to hypotheses about how it performs its computations, and this emphasizes the importance of building on the details of the anatomical and functional connectivity of neurons and networks of neurons to develop theories of how brain systems operate. This comparison helps to underscore the adaptive value of the excitatory recurrent collaterals in the cerebral cortex, for though subject to runaway excitation and instability, they provide the way that the cortex uses to implement short-term memory by continuing firing maintained by the positive feedback, and which provides a computational basis for planning ahead; for completion of a long-term memory from a partial retrieval cue; for decision-making in which the result of the decision can be maintained `on-line’ by continuing attractor-related firing, to guide actions performed on the basis of the decision; and for associating together semantic items which provide the basis for semantic structures in language.
Striosomes Target Nigral Dopamine-Containing Neurons via Direct-D1 and Indirect-D2 Pathways Paralleling Classic Direct-Indirect Basal Ganglia Systems

Abstract:The classic output pathways of the basal ganglia are known as the direct-D1 and indirect-D2, or Go/No-Go, pathways. Balance of the activity in these canonical direct-indirect pathways is considered a core requirement for normal movement control, and their imbalance is a major etiologic factor in movement disorders including Parkinsons disease. We present evidence for a conceptually equivalent parallel system of direct-D1 and indirect-D2 pathways that arise from striatal projection neurons (SPNs) of the striosome compartment rather than from the matrix. These striosomal direct (S-D1) and indirect (S-D2) pathways, as a pair, target dopamine-containing neurons of the substantia nigra (SNpc) instead of the motor output nuclei of the basal ganglia. The novel anatomically and functionally distinct indirect-D2 striosomal pathway targets dopaminergic SNpc cells indirectly via a core region of the external pallidum (GPe). We demonstrate that these S-D1 and S-D2 pathways oppositely modulate striatal dopamine release in freely behaving mice under open-field conditions and oppositely modulate locomotor and other movements. These S-D1 and S-D2 pathways further exhibit different, time-dependent responses during performance of a probabilistic decision-making maze task and respond differently to rewarding and aversive stimuli. These contrasts depend on mediolateral and anteroposterior striatal locations of the SPNs as are the classic direct and indirect pathways. The effects of S-D1 and S-D2 stimulation on striatal dopamine release and voluntary locomotion are nearly opposite. The parallelism of the direct-indirect circuit design motifs of the striosomal S-D and S-D2 circuits and canonical matrix M-D1 and M-D2, and their contrasting behavioral effects, call for a major reformulation of the classic direct-indirect pathway model of basal ganglia function. Given that some striosomes receive limbic and association cortical inputs, the S-D1 and S-D2 circuits likely influence motivation for action and behavioral learning, complementing and possibly reorienting the motoric activities of the canonical matrix pathways. At a fundamental level, these findings suggest a unifying framework for aligning two sets of circuits that share the organizational motif of opponent D1 and D2 regulation, but that have different outputs and can even have opposite polarities in their targets and effects, albeit conditioned by striatal topography. Our findings further delineate a potentially therapeutically important set of pathways influencing dopamine, including a D2 receptor-linked S-D2 pathway likely unknowingly targeted by administration of many therapeutic drugs including those for Parkinsons disease. The novel parallel pathway model that we propose here could help to account for the normally integrated modulatory influence of the basal ganglia on motivation for actions as well as the actions themselves.

Pathways from the Superior Colliculus to the Basal Ganglia

Abstract:The present work aims to review the structural organization of the mammalian superior colliculus (SC), the putative pathways connecting the SC and the basal ganglia, and their role in organizing complex behavioral output. First, we review how the complex intrinsic connections between the SC's laminae projections allow for the construction of spatially aligned, visual-multisensory maps of the surrounding environment. Moreover, we present a summary of the sensory-motor inputs of the SC, including a description of the integration of multi-sensory inputs relevant to behavioral control. We further examine the major descending outputs toward the brainstem and spinal cord. As the central piece of this review, we provide a thorough analysis covering the putative interactions between the SC and the basal ganglia. To this end, we explore the diverse thalamic routes by which information from the SC may reach the striatum, including the pathways through the lateral posterior, parafascicular, and rostral intralaminar thalamic nuclei. We also examine the interactions between the SC and subthalamic nucleus, representing an additional pathway for the tectal modulation of the basal ganglia. Moreover, we discuss how information from the SC might also be relayed to the basal ganglia through midbrain tectonigral and tectotegmental projections directed at the substantia nigra compacta and ventrotegmental area, respectively, influencing the dopaminergic outflow to the dorsal and ventral striatum. We highlight the vast interplay between the SC and the basal ganglia and raise several missing points that warrant being addressed in future studies.
Basal Ganglia and Thalamic Contributions to Language Function: Insights from A Parallel Distributed Processing Perspective

Abstract:Cerebral representations are encoded as patterns of activity involving billions of neurons. Parallel distributed processing (PDP) across these neuronal populations provides the basis for a number of emergent properties: 1) processing occurs and knowledge (long term memories) is stored (as synaptic connection strengths) in exactly the same networks; 2) networks have the capacity for setting into stable attractor states corresponding to concepts, symbols, implicit rules, or data transformations; 3) networks provide the scaffold for the acquisition of knowledge but knowledge is acquired through experience; 4) PDP networks are adept at incorporating the statistical regularities of experience as well as frequency and age of acquisition effects; 5) networks enable content-addressable memory; 6) because knowledge is distributed throughout networks, they exhibit the property of graceful degradation; 7) networks intrinsically provide the capacity for inference. This paper details the features of the basal ganglia and thalamic systems (recurrent and distributed connectivity) that support PDP. The PDP lens and an understanding of the attractor trench dynamics of the basal ganglia provide a natural explanation for the peculiar dysfunctions of Parkinson's disease and the mechanisms by which dopamine deficiency is causal. The PDP lens, coupled with the fact that the basal ganglia of humans bears strong homology to the basal ganglia of lampreys and the central complex of arthropods, reveals that the fundamental function of the basal ganglia is computational and involves the reduction of the vast dimensionality of a complex multi-dimensional array of sensorimotor input into the optimal choice from a small repertoire of behavioral options — the essence of reactive intention (automatic responses to sensory input). There is strong evidence that the sensorimotor basal ganglia make no contributions to cognitive or motor function in humans but can cause serious dysfunction when pathological. It appears that humans, through the course of evolution, have developed cortical capacities (working memory and volitional and reactive attention) for managing sensory input, however complex, that obviate the need for the basal ganglia. The functions of the dorsal tier thalamus, however, even viewed with an understanding of the properties of population encoded representations, remain somewhat more obscure. Possibilities include the enabling of attractor state constellations that optimize function by taking advantage of simultaneous input from multiple cortical areas; selective engagement of cortical representations; and support of the gamma frequency synchrony that enables binding of the multiple network representations that comprise a full concept representation.

Abstract:Many tasks require rapid and fine-tuned adjustment of motor performance based on incoming sensory information. This process of sensorimotor adaptation engages two parallel subcortico–cortical neural circuits, involving the cerebellum and basal ganglia, respectively1,2,3,4,5,6,7,8,9,10. How these distributed circuits are functionally coordinated has not been shown in humans. The cerebellum and basal ganglia show very similar convergence of input–output organization11,12, which presents an ideal neuroimaging model for the study of parallel processing at a systems level13. Here we used functional magnetic resonance imaging to measure the temporal coherence of brain activity during a tactile discrimination task. We found that, whereas the prefrontal cortex maintained a high level of activation, output activities in the cerebellum and basal ganglia showed different phasic patterns. Moreover, cerebellar activity significantly correlated with the activity of the supplementary motor area but not with that of the primary motor cortex; in contrast, basal ganglia activity was more strongly associated with the activity of the primary motor cortex than with that of the supplementary motor area. These results demonstrate temporally partitioned activity in the cerebellum and basal ganglia, implicating functional independence in the parallel subcortical outputs. This further supports the idea of task-related dynamic reconfiguration of large-scale neural networks14,15.
Synaptic integration of somatosensory and motor cortical inputs onto spiny projection neurons of mice caudoputamen

Abstract:By pairing dual genetic labeling and large volume serial electron microscopy, we demonstrate that L5 inputs from somatosensory and motor cortices frequently converge onto single spiny projection neurons in the mouse caudoputamen. In detailing the features of this convergence, we find that cortical L5 terminals largely avoid local interneurons but that inputs to spiny projection neurons have morphological features of driver‐type synapses, which we confirm using an optogenetic approach in acute slices. These findings indicate that integration of sensory and motor information in spiny projection neurons is common for regions of the striatum. The basal ganglia play pivotal roles in motor control and cognitive functioning. These nuclei are embedded in an anatomical loop: cortex to basal ganglia to thalamus back to cortex. We focus here on an essential synapse for descending control, from cortical layer 5 (L5) onto the GABAergic spiny projection neurons (SPNs) of the caudoputamen (CP). We employed genetic labeling to distinguish L5 neurons from somatosensory (S1) and motor (M1) cortices in large volume serial electron microscopy and electrophysiology datasets to better detail these inputs. First, M1 and S1 synapses showed a strong preference to innervate the spines of SPNs and rarely contacted aspiny cells, which are likely to be interneurons. Second, L5 inputs commonly converge from both areas onto single SPNs. Third, compared to unlabeled terminals in CP, those labeled from M1 and S1 show ultrastructural hallmarks of strong driver synapses: They innervate larger spines that were more likely to contain a spine apparatus, more often had embedded mitochondria, and more often contacted multiple targets. Finally, these inputs also demonstrated driver‐like functional properties: SPNs responded to optogenetic activation from S1 and M1 with large EPSP/Cs that depressed and were dependent on ionotropic but not metabotropic receptors. Together, our findings suggest that individual SPNs integrate driver input from multiple cortical areas with implications for how the basal ganglia relay cortical input to provide inhibitory innervation of motor thalamus.
