Abstract:Effects of temperature, storage time and their combination on germination of aspen (Populus tomentosa) seeds were investigated. Aspen seeds were germinated at 5 to 30 degrees C at 5 degrees C intervals after storage for a period of time under 28 degrees C and 75% relative humidity. The effect of temperature on aspen seed germination could not be effectively described by the thermal time (TT) model, which underestimated the germination rate at 5 degrees C and poorly predicted the time courses of germination at 10, 20, 25 and 30 degrees C. A modified TT model (MTT) which assumed a two-phased linear relationship between germination rate and temperature was more accurate in predicting the germination rate and percentage and had a higher likelihood of being correct than the TT model. The maximum lifetime threshold (MLT) model accurately described the effect of storage time on seed germination across all the germination temperatures. An aging thermal time (ATT) model combining both the TT and MLT models was developed to describe the effect of both temperature and storage time on seed germination. When the ATT model was applied to germination data across all the temperatures and storage times, it produced a relatively poor fit. Adjusting the ATT model to separately fit germination data at low and high temperatures in the suboptimal range increased the models accuracy for predicting seed germination. Both the MLT and ATT models indicate that germination of aspen seeds have distinct physiological responses to temperature within a suboptimal range.

Abstract:Cistus species have seeds with hard coats which impose physical seed dormancy that can be released after seed scarification. In fire-prone habitats, the break of physical seed dormancy is usually related to the heat produced during fires. It is commonly accepted that most hard-seeded species, including those of the genus Cistus, are able to germinate under a wide range of temperatures in light as well as in darkness, once the seed becomes permeable. However, although many studies have focused on the release of physical dormancy only, a few have done so on the effect of environmental factors once dormancy is released. In this research, through a factorial experiment, we analysed the effects of light (light and darkness) and a range of temperatures (10, 15, 20, 25 and 30°C) on the seed germination of eight Cistus species after a heat shock. On average, almost 60% of the seeds did not germinate despite being viable, and this lack of germination increased with higher temperatures during the treatment. Although an idiosyncratic germination response emerged, temperature had a significant effect in all the species, reaching the highest levels of germination between 10 and 20°C. Light interacted with temperature in four cases by increasing the germination, especially under the least favourable temperatures. Environmental factors, such as temperature and light, appear to modulate the germination of the studied Cistus species after the release of physical seed dormancy.

Abstract:Low temperature germination (LTG) is one of crucial agronomic traits for field-grown rapeseed in the Yangtze River basin, where delayed direct sowing frequently exposes seeds to cold stress during germination. Despite its importance, the genetic basis underlying rapeseed germination under different temperatures remains largely unexplored. Here we screened out two cultivars with different LTG performance (JY1621 and JY1605) and conducted transcriptome sequencing at five key timepoints under different temperatures. Comparative analysis among multiple groups of differentially expressed genes (DEGs) revealed sets of early and late temperature response germination (ETRG and LTRG) genes, as well as cold-tolerant (CDT) and temperature-insensitive (TPI) candidate regulators at different germination stages. Furthermore, we performed a genome-wide association study (GWAS) using germination index (GI) of 273 rapeseed accessions and identified 24 significant loci associated with the germination trait under the normal temperature. Through integrated analysis of transcriptome sequencing and GWAS, we identified a series of candidate genes involved in temperature-dependent germination. To further verify the results, we analyzed expression patterns and haplotype distributions of BnaA3.CYP77A4and BnaA3.NAC078 among the genetic population. This study performed a comprehensive multi-omics analysis on temperature-dependent germination and provides insights for low-temperature germination breeding of rapeseed.

This work aimed the study the physiological conditioning of Urochloa humidicola cv. Llanero, using two lots of dormant seeds conditioned by imbibition with water, commercial bioregulator (auxin, gibberellin - 0.144 mM and cytokinin), gibberellin (0.144 and 1.44 mM) or 5% ethanol. Germination, dormancy, SOD activity, MDA and H2O2 content were evaluated. In both lots, germination percentage was positively influenced by the commercial bioregulator without affecting the viability. In the first, the germination, using a bioregulator or ethanol, was higher; in the second, the highest germination was from the treated seeds concerning the control and water treatment. The unconditioned seeds presented a higher H2O2 and protein content in both lots. The MDA content in the first lot was higher in the non-conditioned seeds and conditioned with ethanol. In the second lot, the water conditioned seeds had higher content. In both lots, the bioregulator conditioned seeds showed higher SOD activity. The H2O2 content was related to antioxidation activation. It was impossible to identify the oxidative window of germination in which H2O2 would be a signal, releasing germination. The conditioning of seeds with any product showed superior physiological quality than the unconditioned or conditioned with water.

Abstract:We provided a gene pool of moderate size for selecting or manipulating the candidate genes that favour the acquisition of seed dormancy, shedding light on the elevation of seed oil content in oilseed rape by blocking lipid degradation in developing seeds. In oilseed rape, the association between the germination potential of premature seeds and the final level of seed lipids, and the underlying mechanism, is elusive. Here, we investigated phenotypic differences in the germination percentage of premature seeds in a collection of oilseed rape cultivars. We compared the dynamic lipid accumulation between the deep-, moderate- and non-dormant genotypes and compared the transcriptomes of the seeds at 40 days after pollination between multiple pairs of deep- and non-dormant genotypes. We identified a wide range of differences in germination percentage of premature seeds and the association between the germination potential and the change of fatty acid content at late stage of seed maturation. The comparisons of transcriptomes between deep- and non-dormant seeds revealed the genetic basis for the dormant difference, e.g. the different expression levels of the genes involved in gibberellic and abscisic acid biosynthesis and/or signalling, fatty acid metabolic pathways, and the structure of seed cell wall. Abstract:Anticipating the harvest period of soybean crops can impact on the post-harvest processes. This study aimed to evaluate early soybean harvest associated drying and storage conditions on the physicochemical soybean quality using of mathematical modeling and multivariate analysis. The soybeans were harvested with a moisture content of 18 and 23% (d.b.) and subjected to drying in a continuous dryer at 80, 100, and 120 °C. The drying kinetics and volumetric shrinkage modeling were evaluated. Posteriorly, the soybean was stored at different packages and temperatures for 8 months to evaluate the physicochemical properties. After standardizing the variables, the data were submitted to cluster analysis. For this, we use Euclidean distance and Ward's hierarchical method. Then defining the groups, we constructed a graph containing the dispersion of the values of the variables and their respective Pearson correlations for each group. The mathematical models proved suitable to describe the drying kinetics. Besides, the effective diffusivity obtained was 4.9 × 10-10 m2 s-1 promoting a volumetric shrinkage of the grains and influencing the reduction of physicochemical quality. It was observed that soybean harvested at 23% moisture, dried at 80 °C, and stored at a temperature below 23 °C maintained its oil content (25.89%), crude protein (35.69%), and lipid acidity (5.54 mL). In addition, it is to note that these correlations' magnitude was substantially more remarkable for the treatments allocated to the G2 group. Furthermore, the electrical conductivity was negatively correlated with all the physicochemical variables evaluated. Besides this, the correlation between crude protein and oil yield was positive and of high magnitude, regardless of the group formed. In conclusion, the early harvest of soybeans reduced losses in the field and increased the grain flow on the storage units. The low-temperature drying and the use of packaging technology close to environmental temperatures conserved the grain quality.

Abstract:Dormancy in seeds is a key persistence mechanism for many flowering plants. Physically dormant (PY) seeds have water impermeable seed coats, and in fire-prone systems a common mechanism for dormancy release is fire-induced soil heating. However, the thermal thresholds innate to seeds with PY may be influenced by vegetation, climate, and fire regimes, varying substantially between populations of the same species. To investigate intraspecific variation of thermal thresholds in PY seeds, we sampled obligate seeding Acacia pulchella (Fabaceae) which produces PY seeds. Sampling was undertaken from 13 populations across a climate gradient of rainfall and temperature, and between two vegetation communities in fire-prone Mediterranean-type ecosystems of south-west Western Australia. To test a range of weather and fire-induced soil heating dormancy-break scenarios, we conducted dry heat shock experiments between 40 and 140 °C for 10 min and scored germination for 16 weeks. We created population-specific thermal performance curves and extracted the dormancy release temperature at which 50 % of the seeds had germinated (DRT50), the optimum dormancy-breaking temperature to stimulate maximum germination (T0), and the lethal temperature at which 50 % of the seeds were killed (LT50). Generalised linear models were used to examine relationships between thermal thresholds and possible vegetation, climate, and fire regime drivers of intraspecific variation in seed traits. We found that thermal thresholds differed between vegetation communities, with thresholds consistently higher in forest-type ecosystems compared to open woodland, and the influence of climate varied significantly between the two communities. Seeds from Jarrah Forest populations had a DRT50 16.0 °C higher, a T0 9.7 °C higher, and LT50 7.8 °C higher than seeds from Banksia woodlands. A high rate of non-dormancy was identified in one population that had lost fire in its system and displayed significant germination after both summer and fire-related temperatures. The PY thermal thresholds modelled here provide insight into the strong influence of variable soil heating as a function of vegetation and fuel dynamics in fire-prone environments. Our findings highlight the significant intraspecific variation for this species and suggest that fire-induced soil heating generated by vegetation characteristics may be an overlooked element of fire regimes shaping seed traits.
Quantitative Description of the Effect of Stratification on Dormancy Release of Grape Seeds in Response to Various Temperatures and Water Contents

Abstract:The effect of stratification on dormancy release of grape seeds crossing from the sub- to the supraoptimal range of temperatures and water contents was analysed by modified threshold models. The stratification impacted on dormancy release in three different ways: (i) dormancy was consistently released with prolonged stratification time when stratified at temperatures of <15 °C; (ii) at 15 °C and 20 °C, the stratification effect initially increased, and then decreased with extended time; and (iii) stratification at 25 °C only reduced germinable seeds. These behaviours indicated that stratification could not only release primary dormancy but also induce secondary dormancy in grape seed. The rate of dormancy release changed linearly in two phases, while induction increased exponentially with increasing temperature. The thermal time approaches effectively quantified dormancy release only at suboptimal temperature, but a quantitative method to integrate the occurrence of dormancy release and induction at the same time could describe it well at either sub- or supraoptimal temperatures. The regression with the percentage of germinable seeds versus stratification temperature or water content within both the sub- and supraoptimal range revealed how the optimal temperature (Tso) and water content (Wso) for stratification changed. The Tso moved from 10.6 °C to 5.3 °C with prolonged time, while Wso declined from >0.40 g H2O g DW−1 at 5 °C to ∼0.23 g H2O g DW−1 at 30 °C. Dormancy release in grape seeds can occur across a very wide range of conditions, which has important implications for their ability to adapt to a changeable environment in the wild.
One Way to Achieve Germination: Common Molecular Mechanism Induced by Ethylene and After-Ripening in Sunflower Seeds

Abstract:Dormancy is an adaptive trait that blocks seed germination until the environmental conditions become favorable for subsequent vegetative plant growth. Seed dormancy is defined as the inability to germinate in favorable conditions. Dormancy is alleviated during after-ripening, a dry storage period, during which dormant (D) seeds unable to germinate become non-dormant (ND), able to germinate in a wide range of environmental conditions. The treatment of dormant seeds with ethylene (D/ET) promotes seed germination, and abscisic acid (ABA) treatment reduces non-dormant (ND/ABA) seed germination in sunflowers (Helianthus annuus). Metabolomic and transcriptomic studies have been performed during imbibition to compare germinating seeds (ND and D/ET) and low-germinating seeds (D and ND/ABA). A PCA analysis of the metabolites content showed that imbibition did not trigger a significant change during the first hours (3 and 15 h). The metabolic changes associated with germination capacity occurred at 24 h and were related to hexoses, as their content was higher in ND and D/ET and was reduced by ABA treatment. At the transcriptional level, a large number of genes were altered oppositely in germinating, compared to the low-germinating seeds. The metabolomic and transcriptomic results were integrated in the interpretation of the processes involved in germination. Our results show that ethylene treatment triggers molecular changes comparable to that of after-ripening treatment, concerning sugar metabolism and ABA signaling inhibition.

Abstract:Sapium sebiferum seeds were harvested from the marked donor individual at five developmental stages of maturity to study the changes in fresh weight, dry weight, moisture content, germination percentage, endogenous inhibitor activity, and plant growth regulators (PGRs) content. Moisture content of seeds declined from 46.98% (collected on September 1) to 12.00% (collected on November 1). Fresh weight decreased while dry weight increased significantly during seed development. Germination inhibitor activity in the seed coat and endosperm increased during seed maturation and at the same developmental stage, and inhibitor activity in the seed coat was higher than that in the endosperm. Excised embryos were capable of germinating for seeds collected on September 16, and maximum germination percentage was observed for seeds collected on October 16. With increasing seed maturation, gibberellins (GAs) content decreased while abscisic acid (ABA) content increased in both the stratified and untreated seeds. Compared to the untreated controls, the stratified seeds were consistently found to have lower ABA content and higher GAs content throughout different developmental stages. During the course of seed development, the GAs : ABA ratio was found to steadily decrease in both the stratified and untreated seeds, with an exceptional observation for seeds collected on October 1 that underwent stratification. With increasing seed maturity, the germination of untreated seeds initially increased but declined after October 1. Stratified seeds showed the same trend, with higher germination percentage than the control at all developmental stages. Results of this study showed that seeds harvested in September were immature and incapable of germinating, whereas those harvested in November were fully mature but showed strong dormancy. This study showed that seeds are ready to harvest on October 1, and they will be naturally dispersed in November. Thus we propose to harvest seeds before the middle of October for rapid and efficient germination.

Abstract:Introduction: The seeds of many plants are dormant and unable to germinate at maturity, but gain the ability to germinate through after-ripening during dry storage. The hormone abscisic acid (ABA) stimulates seed dormancy, whereas gibberellin A (GA) stimulates dormancy loss and germination. Methods: To determine whether dry after-ripening alters the potential to accumulate ABA and GA, hormone levels were measured during an after-ripening time course in dry and imbibing ungerminated seeds of wildtype Landsberg erecta (L er ) and of the highly dormant GA-insensitive mutant sleepy1-2 ( sly1-2 ). Results: The elevated sly1-2 dormancy was associated with lower rather than higher ABA levels. L er germination increased with 2-4 weeks of after-ripening whereas sly1-2 required 21 months to after-ripen. Increasing germination capacity with after-ripening was associated with increasing GA 4 levels in imbibing sly1-2 and wild-type L er seeds. During the same 12 hr imbibition period, after-ripening also resulted in increased ABA levels. Discussion: The decreased ABA levels with after-ripening in other studies occurred later in imbibition, just before germination. This suggests a model where GA acts first, stimulating germination before ABA levels decline, and ABA acts as the final checkpoint preventing germination until processes essential to survival, like DNA repair and activation of respiration, are completed. Overexpression of the GA receptor GID1b ( GA INSENSITIVE DWARF1b ) was associated with increased germination of sly1-2 but decreased germination of wildtype L er. This reduction of L er germination was not associated with increased ABA levels. Apparently, GID1b is a positive regulator of germination in one context, but a negative regulator in the other.

Abstract:ContextThe use of native species in ecological restoration is highly recommended but, in practice, it is often impaired by knowledge gaps in the germination ecology of suitable species.AimsThis study aimed to assess the role of storage conditions and seed source on the germination of three Mediterranean shrub species with contrasting types of dormancy.MethodsRipe fruits were harvested at two or three distant locations in mainland Portugal. Seeds were subjected to three treatments consisting in different storage conditions: cold storage at low and high moisture conditions, plus a control. Five replicates of up to 30 seeds were placed under constant temperature conditions and germination was monitored weekly during 14 weeks.ResultsThe effect of cold storage at high moisture on germination differed between the three species and seed source played a significant role in the germination of all three species. In the case of the species with dormancy, the observed differences in germination could reflect changes in the species’ dormancy degree or sensitivity to dormancy breaking factors across their geographical range. In the case of Pistacia lentiscus (no dormancy), the results suggested a possible adaptation of the northern seed source to high moisture conditions.ConclusionsThe observed differences between species agreed well with their dormancy types, and the seed source-related differences could be adaptive features, as they seemed related with local climate conditions.

Abstract:This study aimed to define a methodology that, when combined with temperature, can overcome the dormancy and also to determine the adequate substrate and light conditions for germination of Colubrina glandulosa Perkins, contributing to seed analysis and restoration projects. In the experiment of dormancy and temperature, seeds were immersed in sulfuric acid for 30, 60, 90, 120 and 150 minutes and set to germinate at temperatures of 20-30 °C, 25 °C and 30 °C. The used substrates were sand, vermiculite, Tropstrato® substrate, coconut powder, blotting paper and paper towel. As for the light supplied, the seeds were subjected to four luminosity regimes: white light, far-red light, red light and light absence. Temperatures of 20-30 °C and 30 °C achieved better results, with exposure in sulfuric acid for 120 and 150 minutes. The substrates sand and vermiculite were the most suitable. The species germinated in all light regimes treatments used.

Abstract:The germination process and seedling development are the determining steps in the plant lifecycle that are the most sensitive to adverse environmental conditions. Therefore, this study was conducted to explore the effects of temperature and osmotic potential on germination responses using threshold models and to establish an optimal priming protocol for improving tolerance responses against osmotic stress in early growth stages. The results demonstrated that osmotic stress of − 0.8 MPa significantly influenced the extent, timing, and speed of seed germination. In addition, priming treatments led to an enhanced performance of early growth stages in response to osmotic stress. Based on thermal-time and hydro-time models, the predicted physiological parameters of the median thermal-time at sub-optimal temperature ( h), the median ceiling temperature for 50% germination (T c(50) = 39.29 °C), the common base temperature (T b = 7.88 °C), the constant thermal-time at supra-optimal temperature ( = 805.96 °C h), the threshold water potential (Ψ b(50) = − 1.13 MPa), and the hydro-time constant ( = 56.09 MPa h) quantitatively describe the tolerance threshold of the germination process under different osmotic and temperature conditions. The results also showed that the efficiency of seed treatments depended on the priming conditions, including temperature, duration, and also concentration of the priming agent. However, the treatments of gibberellic acid (5 days, 10 °C, 100 ppm), salicylic acid (5 days, 10 °C, 50 ppm), calcium chloride (3 days, 10 °C, 10 mM), potassium nitrate (3 days, 10 °C, 100 mM), and hydro-priming (3 days, 10 °C) were optimal protocols of each priming method, resulting in an increased seed vigor under osmotic stress. Hence, the predicted biological parameters could easily be applied to determine the physiological changes of germination under environmental factors over time. Also, results suggest that recommended osmo-, hydro-, and hormonal-priming treatments could be efficient methods for ameliorating the osmotic tolerance in the post-priming stages of this plant, especially in arid lands.

Abstract:A human nose‐mimetic diagnosis system that can distinguish the odor of a lung cancer biomarker, heptanal, from human blood is presented. Selective recognition of the biomarker is mimicked in the human olfactory system. A specific olfactory receptor recognizing the chemical biomarker is first selected through screening a library of human olfactory receptors (hORs). The selected hOR is expressed on the membrane of human embryonic kidney (HEK)‐293 cells. Nanovesicles containing the hOR on the membrane are produced from these cells, and are then used for the functionalization of single‐walled carbon nanotubes. This strategy allows the development of a sensitive and selective nanovesicle‐based bioelectronic nose (NvBN). The NvBN is able to selectively detect heptanal at a concentration as low as 1 × 10−14 m, a sufficient level to distinguish the blood of a lung cancer patient from the blood of a healthy person. In actual experiments, NvBN could detect an extremely small increase in the amount of heptanal from human blood plasma without any pretreatment processes. This result offers a rapid and easy method to analyze chemical biomarkers from human blood in real‐time and to diagnose lung cancer.
Effects of H2SO4, GA3, and cold stratification on the water content, coat composition, and dormancy release of Tilia miqueliana seeds

Abstract:Introduction: Tilia miqueliana is an endemic species whose population is declining. The permeability barrier and mechanical constraint of the pericarp (seed coat) are important causes of its seed dormancy. Although there has been considerable research on this subject, questions remain regarding how the permeability barrier and mechanical constraint of the seed coat are eliminated during dormancy release and how water enters the seed. Therefore, protect the species by improving its germination/breaking the dormancy in the laboratory is urgent. Methods: In this study, the changes in the cellular structure, mechanical properties, and components of the Tilia miqueliana seed coat after an H 2 SO 4 -gibberellic acid (GA 3 ) treatment were analyzed during dormancy release. Various analyses (e.g., magnetic resonance imaging, scanning electron microscopy, and paraffin section detection) revealed the water gap and water channel. Results: The H 2 SO 4 treatment eliminated the blockage at the micropyle and hilum of the seeds. Water entered the seeds through the water gap (micropyle) rather than through the hilum or seed coat, after which it dispersed along the radicle, hypocotyl, and cotyledon to the endosperm. During the cold stratification period, the cellular structure was damaged and an increasing number of holes appeared on the inner and outer surfaces of the seed coat. Vickers hardness tests showed that GA 3 decreased the seed coat hardness. Additionally, the seed coat lignin and total phenol contents continuously decreased during the cold stratification period. Notably, the Liquid chromatography–mass spectrometry (LC–MS) analysis of the seed coat detected polyethylene glycol (osmoregulator), which may have destabilized the water potential balance inside and outside the seed and increased the water content to levels required for germination, ultimately accelerating seed dormancy release. Discussion: This sophisticated and multi-level study reveals how H 2 SO 4 and GA 3 eliminate the permeability barrier and mechanical constraints of the seed coat during dormancy release of Tilia miqueliana seeds. This will be beneficial to artificially assist the natural regeneration and population expansion of Tilia miqueliana .

Abstract:During seed development, plant hormones are involved in processes such as the accumulation of reserves, cellular activity and physiological responses. The present study aimed to analyze levels of abscisic acid, gibberellins, auxins, salicylic acid and their role in the physiological responses during development of <i>A. angustifolia</i> seeds in two years of production. Seeds were collected in the stages cotyledonary, II, III and IV during 2 years and submitted to germination, tetrazolium and electrical conductivity tests. The embryonic axes and cotyledons were extracted from the seeds and submitted to hormones analyses. The results demonstrated that environmental conditions during development affected the time when seeds reached physiological maturity and hormone levels. In 2015, seed development occurred under conditions of lower rainfall, cold hours and insolation, providing the formation of larger seeds, favoring a higher abscisic acid synthesis and accumulation, anticipating/shortening seed maturation. The results highlight the antagonistic relationship between abscisic acid and auxins, which acts as a regulatory factor for faster germination. In conclusion, changes in the levels of abscisic acid, gibberellins, auxins, salicylic acid and their role in physiological responses during seed development are linked to climatic conditions during maturation.

Factors affecting the dormancy and germination of bleeding heart [Lamprocapnos spectabilis (L.) Fukuhara] seeds

Abstract:Information on the optimal conditions to promote the germination of Lamprocapnos spectabilis (L.) Fukuhara seeds is limited; consequently, this study was conducted to establish the requirements to break seed dormancy and promote germination. The selected seeds had morphophysiological dormancy and had not begun embryo development. To study the dormancy breaking and embryo development processes, seeds were subjected to constant or changing temperature treatments during moist stratification. High temperature and humidity resulted in vigorous embryo growth, with the longest embryos occurring after 1 month of incubation at 20 °C. At 4 °C, the seeds required incubation period of at least 3 months to germinate. Embryo growth and germination were higher with changing high and low temperatures than under a constant temperature, and changing temperatures also considerably changed the endogenous hormone levels, embryo development and germination. Bioactive gibberellin (GA) content was higher in seeds incubated at 20 °C for 1 month, then at 4 °C for 2 months. The content of endogenous abscisic acid in seeds subjected to the same treatment decreased by 97.6% compared with that of the untreated seeds. Embryo growth and seed germination require changing high and low temperatures; however, exogenous GA3 could substitute for high temperatures, as it also causes accelerated germination. In this study, the seeds of L. spectabilis were identified as an intermediate simple type, a sub-level of morphophysiologically dormant seeds.

Abstract:Background: Seed dormancy, defined as the incapability of a viable seed to germinate under favourable conditions, is an important trait in nature and agriculture. Despite extensive research on dormancy and germination, many questions about the molecular mechanisms controlling these traits remain unanswered, likely due to its genetic complexity and the large environmental effects which are characteristic of these quantitative traits. To boost research towards revealing mechanisms in the control of seed dormancy and germination we depend on the identification of genes controlling those traits. Methods: We used transcriptome analysis combined with a reverse genetics approach to identify genes that are prominent for dormancy maintenance and germination in imbibed seeds of Arabidopsis thaliana. Comparative transcriptomics analysis was employed on freshly harvested (dormant) and after-ripened (AR; non-dormant) 24-h imbibed seeds of four different DELAY OF GERMINATION near isogenic lines (DOGNILs) and the Landsberg erecta (Ler) wild type with varying levels of primary dormancy. T-DNA knock-out lines of the identified genes were phenotypically investigated for their effect on dormancy and AR. Results: We identified conserved sets of 46 and 25 genes which displayed higher expression in seeds of all dormant and all after-ripened DOGNILs and Ler, respectively. Knock-out mutants in these genes showed dormancy and germination related phenotypes. Conclusions: Most of the identified genes had not been implicated in seed dormancy or germination. This research will be useful to further decipher the molecular mechanisms by which these important ecological and commercial traits are regulated.
