Abstract:Stress fibers are contractile bundles in the cytoskeleton that stabilize cell structure by exerting traction forces on the extracellular matrix. Individual stress fibers are molecular bundles composed of parallel actin and myosin filaments linked by various actin-binding proteins, which are organized end-on-end in a sarcomere-like pattern within an elongated three-dimensional network. While measurements of single stress fibers in living cells show that they behave like tensed viscoelastic fibers, precisely how this mechanical behavior arises from this complex supramolecular arrangement of protein components remains unclear. Here we show that computationally modeling a stress fiber as a multi-modular tensegrity network can predict several key behaviors of stress fibers measured in living cells, including viscoelastic retraction, fiber splaying after severing, non-uniform contraction, and elliptical strain of a puncture wound within the fiber. The tensegrity model can also explain how they simultaneously experience passive tension and generate active contraction forces; in contrast, a tensed cable net model predicts some, but not all, of these properties. Thus, tensegrity models may provide a useful link between molecular and cellular scale mechanical behaviors and represent a new handle on multi-scale modeling of living materials.
A unified multiscale mechanical model for soft collagenous tissues with regular fiber arrangement

Abstract:In this paper the mechanical response of soft collagenous tissues with regular fiber arrangement (RSCTs) is described by means of a nanoscale model and a two-step micro-macro homogenization technique. The non-linear collagen constitutive behavior is modeled at the nanoscale by a novel approach accounting for entropic mechanisms as well as stretching effects occurring in collagen molecules. Crimped fibers are reduced to equivalent straight ones at the microscale and the constitutive response of RSCTs at the macroscale is formulated by homogenizing a fiber reinforced material. This approach has been applied to different RSCTs (tendon, periodontal ligament and aortic media), resulting effective and accurate as proved by the excellent agreement with available experimental data. The model is based on few parameters, directly related to histological and morphological evidences and whose sensitivity has been widely investigated. Applications to simulation of some physiopathological mechanisms are also proposed, providing confirmation of clinical evidences and quantitative indications helpful for clinical practice.

Abstract:A novel constitutive model that describes stress relaxation in transversely isotropic soft collagenous tissues such as ligaments and tendons is presented. The model is formulated within the nonlinear integral representation framework proposed by Pipkin and Rogers (J. Mech. Phys. Solids. 16:59–72, 1968). It represents a departure from existing models in biomechanics since it describes not only the strain dependent stress relaxation behavior of collagenous tissues but also their finite strains and transverse isotropy. Axial stress–stretch data and stress relaxation data at different axial stretches are collected on rat tail tendon fascicles in order to compute the model parameters. Toward this end, the rat tail tendon fascicles are assumed to be incompressible and undergo an isochoric axisymmetric deformation. A comparison with the experimental data proves that, unlike the quasi-linear viscoelastic model (Fung, Biomechanics: Mechanics of Living Tissues. Springer, New York, 1993) the constitutive law can capture the observed nonlinearities in the stress relaxation response of rat tail tendon fascicles.

Abstract:Recently, we introduced an efficient discrete fiber dispersion model for characterizing the mechanical behavior of soft fibrous tissues, and we also extended that model to consider microscale collagen fiber recruitment, softening and damage. However, the viscous behavior of collagen fibers were not considered in that study. The goal of this study is to further extend the discrete fiber dispersion model to consider both elastic and viscous behavior of collagen fibers and ground substance such that the strain rate-dependent behavior of soft fibrous tissues can be characterized. We first reformulated the 3D discrete fiber dispersion model to a 2D version for tissues with planar fiber dispersion. Then, we introduced viscous parts to the model by adding Maxwell elements to the elastic parts of the strain–energy function. We implemented the proposed model in a finite element program and illustrated it with three numerical examples. The computational solutions of the first two examples agree well with our previous published results. In the last example, we characterized the rate-dependent behavior of aortic valve tissue with the proposed model. In conclusion, the proposed model is capable of characterizing the viscoelastic behavior of collagen fibers and ground substance. Future studies with patient-specific boundary conditions are necessary to verify this method.

Abstract:Collagen is a ubiquitous protein with remarkable mechanical properties. It is highly elastic, shows large fracture strength and enables substantial energy dissipation during deformation. Most of the connective tissue in humans consists of collagen fibrils composed of a staggered array of tropocollagen molecules, which are connected by intermolecular cross-links. In this study, we report a three-dimensional coarse-grained model of collagen and analyze the influence of enzymatic cross-links on the mechanics of collagen fibrils. Two representatives immature and mature cross-links are implemented in the mesoscale model using a bottom-up approach. By varying the number, type and mechanical properties of cross-links in the fibrils and performing tensile test on the models, we systematically investigate the deformation mechanisms of cross-linked collagen fibrils. We find that cross-linked fibrils exhibit a three phase behavior, which agrees closer with experimental results than what was obtained using previous models. The fibril mechanical response is characterized by: (i) an initial elastic deformation corresponding to the collagen molecule uncoiling, (ii) a linear regime dominated by molecule sliding and (iii) the second stiffer elastic regime related to the stretching of the backbone of the tropocollagen molecules until the fibril ruptures. Our results suggest that both cross-link density and type dictate the stiffness of large deformation regime by increasing the number of interconnected molecules while cross-links mechanical properties determine the failure strain and strength of the fibril. These findings reveal that cross-links play an essential role in creating an interconnected fibrillar material of tunable toughness and strength.

Abstract:Knowledge of the highly nonlinear behavior of crimped collagen fibers has a great importance for medical diagnosis. This behavior can be associated with the gradual straightening of fibers and the amount of fibers that bears load. To model this phenomenon, we propose an explicit scalar potential, based on the Weibull distribution, in terms of the fiber bundle strength. The total strain is given in terms of the stress and is defined by sum of the strain of crimped fibers, arising from the proposed potential, and the strain of completely straightened fibers by using the Hookean model. Besides three physically meaningful parameters, further two new parameters that have intuitive interpretations were considered. The proposed model was compared with the Freed-Rajagopal model and the Holzapfel-Gasser-Ogden model for verifying its ability to fit the measured data from bovine tendon samples under uniaxial tension. Our model better described the highly nonlinear behavior of the tendon. Experimental data from literature were also used for validation of the parameters of the proposed model. The estimated parameters were consistent with those found in the literature. The effectiveness of the proposed model and Freed-Rajagopal model in describing the stress-strain curve with different initial range of strain with the zero-stress state was also investigated. Unlike the Freed-Rajagopal model, the proposed model fits well in all cases, including those with wide range of strain.

Abstract:The aim of this work is to develop a 3D finite elements model to study the nanomechanical behaviour of mineralized collagen microfibrils, which consists of three phases, (i) collagen phase formed by five tropocollagen (TC) molecules linked together with cross links, (ii) a mineral phase (Hydroxyapatite) and (iii) impure mineral phase, and to investigate the important role of individual properties of every constituent. The mechanical and the geometrical properties (TC molecule diameter) of both tropocollagen and mineral were taken into consideration as well as cross-links, which was represented by spring elements with adjusted properties based on experimental data. In the present paper an equivalent homogenised model was developed to assess the whole microfibril mechanical properties (Young's modulus and Poisson's ratio) under varying mechanical properties of each phase. In this study both equivalent Young's modulus and Poisson's ratio which were expressed as functions of Young's modulus of each phase were obtained under tensile load with symmetric and periodic boundary conditions.

Abstract:Some gold(III)-dithiocarbamato derivatives of either single amino acids or oligopeptides have shown promise as potential anticancer agents, but their capability to interact with biologically relevant macromolecules is still poorly understood. We investigated the affinity of the representative complex [Au(III)Br2(dtc-Sar-OCH3)] (dtc: dithiocarbamate; Sar: sarcosine (N-methylglycine)) with selected model molecules for histidine-, methionine-, and cysteine-rich proteins (that is, 1-methylimidazole, dimethylsulfide, and N-acetyl-L-cysteine, respectively). In particular, detailed mono- and multinuclear NMR studies, in combination with multiple (13)C/(15)N enrichments, allowed interactions to be followed over time and indicated somewhat unexpected reaction pathways. Whereas dimethylsulfide proved to be unreactive, a sudden multistep redox reaction occurred in the presence of the other potential sulfur donor, N-acetyl-L-cysteine (confirmed if glutathione was used instead). On the other hand, 1-methylimidazole underwent an unprecedented acid-base reaction with the gold(III) complex, rather than the expected coordination to the metal center by replacing, for instance, a bromide. Our results are discussed herein and compared with the data available in the literature on related complexes; our findings confirm that the peculiar reactivity of gold(III)-dithiocarbamato complexes can lead to novel reaction pathways and, therefore, to new cytotoxic mechanisms in cancer cells.
Modeling of mechanosensitive remodeling processes in collagen fibers

Abstract:Abstract A large number of biochemical processes such as enzymatic collagen degradation are involved in the remodeling of biological soft tissue constituents. Experiments have found that moderate strain may stabilize fibrillar collagen to enzymatic degradation. This strain stabilization is crucial for the homeostatic balance during the tissue remodeling process in healthy soft tissue. This article studies a material model applicable to the modeling of collagenous soft tissue. The ground substance properties are taken to be time‐independent, while the mechanical behavior of the fibers is modeled in terms of the Helmholtz free energy of the underlying protofibers along with a survival kernel that accounts for how the fiber density is dependent upon the deformation of the overall material. The examples illustrate the consequences of a compressively induced mechanical disruption to the homeostatic fiber remodeling, and they show the differences in the re‐approach to homeostasis when either the stress or strain remains constant.
A model for one-dimensional morphoelasticity and its application to fibroblast-populated collagen lattices

Abstract:The mechanical behaviour of solid biological tissues has long been described using models based on classical continuum mechanics. However, the classical continuum theories of elasticity and viscoelasticity cannot easily capture the continual remodelling and associated structural changes of biological tissues. Furthermore, models drawn from plasticity theory are difficult to apply and interpret in this context, where there is no equivalent of a yield stress or flow rule. In this work, we describe a novel one-dimensional mathematical model of tissue remodelling based on the multiplicative decomposition of the deformation gradient. We express the mechanical effects of remodelling as an evolution equation for the 'effective strain', a measure of the difference between the current state and a hypothetical mechanically-relaxed state of the tissue. This morphoelastic model combines the simplicity and interpretability of classical viscoelastic models with the versatility of plasticity theory. A novel feature of our model is that while most models describe growth as a continuous quantity, here we begin with discrete cells and develop a continuum representation of lattice remodelling based on an appropriate limit of the behaviour of discrete cells. To demonstrate the utility of our approach, we use this framework to capture qualitative aspects of the continual remodelling observed in fibroblast-populated collagen lattices, in particular its contraction and its subsequent sudden re-expansion when remodelling is interrupted.

Abstract:The tympanic membrane (TM), located at the end of the ear canal, is a collagenous multi-layer soft tissue membrane with fibers highly aligned in radial and circumferential orientations. This unique multi-layer fiber ultrastructure makes TM’s mechanical behavior display both anisotropy and nonlinearity, which is important in sound transmission. However, the constitutive model of TM which includes both features has not been proposed. In this study, we develop a fiber-reinforced mesoscale constitutive model of TM which captures both anisotropic and nonlinear elastic mechanical behaviors. The TM is considered a continuum fiber-reinforced composite with two families of collagen fibers. Its overall properties are built up by integrating its heterogeneous material properties through the thickness. The homogenized mechanical properties are assumed to be uniformly distributed through TM’s thickness and superposed by three uncoupled elastic contributions of radial collagen fibers, circumferential collagen fibers, and an equivalent isotropic matrix. The model is calibrated using literature data through the inverse method. Simulation results indicate that specific collagen fibers alignment is responsible for the significant spatial and directional variation of deformation of the TM strip. With the appropriate strength criteria related to fiber deformation, the anisotropic localized failure mode of the TM strip observed in the experiment can be captured. The nonlinear nature and rotation of collagen fiber bundles are the origin of the nonlinear mechanical behavior of TM strips under uniaxial loading. The mesoscale constitutive model offers a different perspective on TM’s anisotropic and nonlinear elastic mechanical behavior. This research improves our understanding of the mechanical behavior of the TM and could help biomimetic graft development.
Local micromorphic non-affine anisotropy for materials incorporating elastically bonded fibers

Abstract:There has been increasing experimental evidence of non-affine elastic deformation mechanisms in biological soft tissues. These observations call for novel constitutive models which are able to describe the dominant underlying micro-structural kinematics aspects, in particular relative motion characteristics of different phases. This paper proposes a flexible and modular framework based on a micromorphic continuum encompassing matrix and fiber phases. It features in addition to the displacement field so-called director fields which can independently deform and intrinsically carry orientational information. Accordingly, the fibrous constituents can be naturally associated with the micromorphic directors and their non-affine motion within the bulk material can be efficiently captured. Furthermore, constitutive relations can be formulated based on kinematics quantities specifically linked to the material response of the matrix, the fibres and their mutual interactions. Associated stress quantities are naturally derived from a micromorphic variational principle featuring dedicated governing equations for displacement and director fields. This aspect of the framework is crucial for the truly non-affine elastic deformation description. In contrast to conventional micromorphic approaches, any non-local higher-order material behaviour is excluded, thus significantly reducing the number of material parameters to a range typically found in related classical approaches. In the context of biological soft tissue modeling, the potential and applicability of the formulation is studied for a number of academic examples featuring anisotropic fiber-reinforced composite material composition to elucidate the micromorphic material response as compared with the one obtained using a classical continuum mechanics approach.

Multiscale characterization of the mechanics of curved fibered structures with application to biological materials

Abstract:Curved fibered structures are ubiquitous in nature and this organization is found in the majority of biological tissues. Indeed, the mechanical behavior of these materials is of pivotal importance in biomechanics and mechanobiology fields. In this paper, we develop a multiscale formulation to characterize the macroscopic mechanical nonlinear behavior from the microstructure of fibered matrices. From the analysis of the mechanics of a randomly curved single fiber, a fibered matrix model is built to determine the macroscopic behavior following a homogenization approach. The model is tested for tensile, compression and shear loads in a number of applications reminiscent to collagen extracellular matrices. However, any other fibered microstructures can be studied following the proposed formulation. The presented approach naturally recovers instabilities at compression as well as the strain stiffening regime, which are observed experimentally in the mechanical behavior of collagen matrices. Indeed, it was found that the bending energy associated to fiber unrolling, is the most important source of energy developed by fibers for the analyzed cases in tensile and shear in all deformation regions (except the strain stiffening region), whereas bending energy dominates at compression too during buckling. The proposed computational framework can also be used to perform multiscale simulations in the referred applications. As a result, the developed methodology may be an interesting and complementary tool to characterize the nonlinear behavior and evolution of curved fibered structures present in biology and engineered materials.

Hyperelastic modelling of arterial layers with distributed collagen fibre orientations

Abstract:Constitutive relations are fundamental to the solution of problems in continuum mechanics, and are required in the study of, for example, mechanically dominated clinical interventions involving soft biological tissues. Structural continuum constitutive models of arterial layers integrate information about the tissue morphology and therefore allow investigation of the interrelation between structure and function in response to mechanical loading. Collagen fibres are key ingredients in the structure of arteries. In the media (the middle layer of the artery wall) they are arranged in two helically distributed families with a small pitch and very little dispersion in their orientation (i.e. they are aligned quite close to the circumferential direction). By contrast, in the adventitial and intimal layers, the orientation of the collagen fibres is dispersed, as shown by polarized light microscopy of stained arterial tissue. As a result, continuum models that do not account for the dispersion are not able to capture accurately the stress–strain response of these layers. The purpose of this paper, therefore, is to develop a structural continuum framework that is able to represent the dispersion of the collagen fibre orientation. This then allows the development of a new hyperelastic free-energy function that is particularly suited for representing the anisotropic elastic properties of adventitial and intimal layers of arterial walls, and is a generalization of the fibre-reinforced structural model introduced by Holzapfel & Gasser (Holzapfel & Gasser 2001 Comput. Meth. Appl. Mech. Eng . 190 , 4379–4403) and Holzapfel et al . (Holzapfel et al . 2000 J. Elast . 61 , 1–48). The model incorporates an additional scalar structure parameter that characterizes the dispersed collagen orientation. An efficient finite element implementation of the model is then presented and numerical examples show that the dispersion of the orientation of collagen fibres in the adventitia of human iliac arteries has a significant effect on their mechanical response.

Evaluation of Cerebrospinal Fluid in Cavalier King Charles Spaniel Dogs Diagnosed with Chiari-Like Malformation with or without Concurrent Syringomyelia

Abstract:Syringomyelia is a common clinical problem in the Cavalier King Charles Spaniel dog population. The underlying pathophysiology of the development and progression of syringes is currently unknown. The primary aim of the current study was to determine whether development of syringomyelia is accompanied by alterations in cerebrospinal fluid composition. Patient records of Cavalier King Charles Spaniels with Chiari-like malformations, with or without concurrent syringomyelia, that had magnetic resonance imaging and cerebrospinal fluid analysis between December 2004 and December 2009, were retrospectively reviewed. Total nucleated cell count per microliter, with differential count, red blood cell count per microliter, and protein concentration (g/l), were reviewed. Data were analyzed with the Mann–Whitney U-test. Spearman rank test was used to test association between cerebrospinal fluid composition and syrinx size, and the Fisher exact test was used to compare the presence of macrophages, reactive lymphocytes, and myelin. A P value of ≤0.05 was considered significant for statistical tests. Dogs with syringomyelia had a higher total nucleated cell count (syringomyelia: 4/μl [range, 0–15/μl] vs. without syringomyelia: 2/μl [0–8/μl]; P = 0.0047), higher protein concentration (syringomyelia: 0.26 g/l [0.07–0.42 g/l] vs. without syringomyelia: 0.2 g/l [0.12–0.39 g/l]; P = 0.039), and an increased neutrophil percentage (syringomyelia: 0% [0–37%] vs. without syringomyelia: 0% [0–21%]; P = 0.0203) than those with the Chiari-like malformation alone. There was a positive correlation between total nucleated cell count and syrinx size (r = 0.51; P = 0.0068). Development of syringomyelia is accompanied by alterations in cerebrospinal fluid composition. The pathogenesis of syringomyelia has not been fully elucidated; therefore, the exact origin of cerebrospinal fluid changes remains unclear.
Mechanical model for a collagen fibril pair in extracellular matrix

Abstract:In this paper, we model the mechanics of a collagen pair in the connective tissue extracellular matrix that exists in abundance throughout animals, including the human body. This connective tissue comprises repeated units of two main structures, namely collagens as well as axial, parallel and regular anionic glycosaminoglycan between collagens. The collagen fibril can be modeled by Hooke's law whereas anionic glycosaminoglycan behaves more like a rubber-band rod and as such can be better modeled by the worm-like chain model. While both computer simulations and continuum mechanics models have been investigated the behavior of this connective tissue typically, authors either assume a simple form of the molecular potential energy or entirely ignore the microscopic structure of the connective tissue. Here, we apply basic physical methodologies and simple applied mathematical modeling techniques to describe the collagen pair quantitatively. We find that the growth of fibrils is intimately related to the maximum length of the anionic glycosaminoglycan and the relative displacement of two adjacent fibrils, which in return is closely related to the effectiveness of anionic glycosaminoglycan in transmitting forces between fibrils. These reveal the importance of the anionic glycosaminoglycan in maintaining the structural shape of the connective tissue extracellular matrix and eventually the shape modulus of human tissues. We also find that some macroscopic properties, like the maximum molecular energy and the breaking fraction of the collagen, are also related to the microscopic characteristics of the anionic glycosaminoglycan.
Physically based 3D finite element model of a single mineralized collagen microfibril

Abstract:Mineralized collagen microfibrils in human bone provide its mechanical properties (stiffness, elasticity, ductility, energy dissipation and strength). However, detailed 3D finite element models describing the mechanical behavior of the mineralized collagen microfibrils are still lacking. In the current work, we developed a 3D finite element model of the mineralized collagen microfibril that incorporates the physical 3D structural details. The model components consist of five tropocollagen molecules, mineral hydroxyapatite and intermolecular cross-links joining primarily the ends of the tropocollagen molecules. Dimension, arrangement and mechanical behavior of the constituents are based on previously published experimental and theoretical data. Tensile and compressive loads were applied to the microfibril under different conditions (hydrated and dehydrated collagen) to investigate the relationship between the structure and the mechanical behavior of the mineralized collagen microfibril (stress-strain curve and elastic modulus). The computational results match the experimental available data well, and provide insight into the role of the phases and morphology on the microfibril behavior. Our predicted results show that the mechanical properties of collagen microfibrils arise due to their structure and properties. The proposed 3D finite element model of mineralized collagen microfibril contributes toward the investigation of the bottom-up structure-property relationships in human bone.

Abstract:Progressive stiffening of collagen tissue by bioapatite mineral is important physiologically, but the details of this stiffening are uncertain. Unresolved questions about the details of the accommodation of bioapatite within and upon collagen's hierarchical structure have posed a central hurdle, but recent microscopy data resolve several major questions. These data suggest how collagen accommodates bioapatite at the lowest relevant hierarchical level (collagen fibrils), and suggest several possibilities for the progressive accommodation of bioapatite at higher hierarchical length scales (fibres and tissue). We developed approximations for the stiffening of collagen across spatial hierarchies based upon these data, and connected models across hierarchies levels to estimate mineralization-dependent tissue-level mechanics. In the five possible sequences of mineralization studied, percolation of the bioapatite phase proved to be an important determinant of the degree of stiffening by bioapatite. The models were applied to study one important instance of partially mineralized tissue, which occurs at the attachment of tendon to bone. All sequences of mineralization considered reproduced experimental observations of a region of tissue between tendon and bone that is more compliant than either tendon or bone, but the size and nature of this region depended strongly upon the sequence of mineralization. These models and observations have implications for engineered tissue scaffolds at the attachment of tendon to bone, bone development and graded biomimetic attachment of dissimilar hierarchical materials in general.
A Finite-Strain Micromechanical Model for the Hyperelasticity of Tendons and Ligaments with Crimped Fibers

Abstract:The hyperelasticity of soft biological tissues such as tendons and ligaments is closely related to their microstructures consisting of wavy fibers embedded in a soft matrix. Large deformation of such composites involves both microstructural evolution and material nonlinearity, posing a challenge of theoretical analysis. In this paper, we propose a finite-strain micromechanical method to predict the hyperelastic constitutive relation of tendons and ligaments reinforced with crimped fibers. The Mori–Tanaka method is extended to account for the interaction of wavy fibers embedded in a soft matrix under large deformation. It is found that the tension–shear coupling mechanism induced by fiber–matrix interaction is crucial for the nonlinear behavior of tendons and ligaments. The hyperelasticity of such composites relies strongly on the initial crimp shape, Young's modulus, and volume fraction of fibers. This model is validated by finite element simulations of a unit cell model with crimped fibers. Furthermore, an explicit expression based on dimensional analysis is derived for the hyperelastic constitutive relation in terms of microstructural and material parameters, and it fits well with experimental data of tendons at different ages. This study can not only deepen the understanding of the mechanical behaviors of such soft tissues of tendons and ligaments, but also help the optimal design of biomimetic composites with superior elasticity.
Acupressure and Preoperative Parental Anxiety: A Pilot Study

Abstract:In this randomized sham-controlled study we examined the anxiolytic and sedative effects of acupressure on parents in the preoperative holding area before their children’s surgery. Sixty-one parents received acupressure either at the Yintang point (midpoint between the two eyebrows) or at a sham point. Anxiety (as measured by the Stait-Trait Anxiety Inventory), arterial blood pressure, and heart rate were assessed before and after the intervention and a Bispectral Index monitor was used to continuously monitor hypnotic sedation levels. Repeated-measures analysis of variance showed that parents in the acupressure group reported significantly less anxiety at 20 min postintervention as compared with parents in the sham group (37 ± 10 versus 45 ± 13, P = 0.03). Bispectral Index values, heart rate, and arterial blood pressure, however, did not differ between the two study groups (P = not significant). We conclude that acupressure at the Yintang point may be used as a treatment for parental preoperative anxiety. Future studies are needed to quantify the magnitude and duration of the anxiolytic effect.
