Abstract:OBJECTIVE To assess the experimental and clinical data regarding the effects of electromagnetic fields (EMFs) on fracture non-union. DATA SOURCES The English language literature regarding EMFs on fracture non-union were searched using MEDLINE, Web of Science and Embase, for the period January 2006 to June 2011. The search terms were electromagnetic fields and non-union/bone marrow stem cells (BMSCs)/bone. STUDY SELECTION Articles were included in the review if they were related to the use of EMFs on BMSCs or bone tissue. Papers without full manuscripts available were excluded. RESULTS The basic and clinical research in this field, while somewhat limited, supports the insightful application of EMFs to ameliorate disability due to fracture non-union. CONCLUSIONS Further basic and clinical research to validate the use of EMFs in facilitating function and bone reparative processes in fracture non-union is required.

Abstract:Recently, magnetic fields (MFs) have received major attention due to their potential therapeutic applications and biological effects. This review provides a comprehensive analysis of the cellular and molecular impacts of MFs, with a focus on both in vitro and in vivo studies. We investigate the mechanisms by which MFs influence cell behavior, including modifications in gene expression, protein synthesis, and cellular signaling pathways. The interaction of MFs with cellular components such as ion channels, membranes, and the cytoskeleton is analyzed, along with their effects on cellular processes like proliferation, differentiation, and apoptosis. Molecular insights are offered into how MFs modulate oxidative stress and inflammatory responses, which are pivotal in various pathological conditions. Furthermore, we explore the therapeutic potential of MFs in regenerative medicine, cancer treatment, and neurodegenerative diseases. By synthesizing current findings, this article aims to elucidate the complex bioeffects of MFs, thereby facilitating their optimized application in medical and biotechnological fields.

The Effects of 50 Hz Magnetic Field–exposed Cell Culture Medium on Cellular Functions in FL Cells

Abstract:Abstract Although extremely low frequency magnetic fields (ELF-MFs) have been classified as a possible carcinogen for humans by the International Agency for Research on Cancer (IARC), their biological effects and underlying mechanisms are still unclear. Our previous study indicated that ELF-MF exposure influenced the relative permittivity of the saline solution, suggesting that the MF exposure altered physical properties of the solution. To explore the biophysical mechanism of ELF-MF–induced biological effects, this study examined the effects of 50 Hz sinusoidal MF at 0–4.0 mT on the permittivity of culture medium with phase-interrogation surface plasmon resonance (SPR) sensing. Then, the biological effects of MF pre-exposed culture medium on cell viability, the mitogen-activated protein kinase (MAPK) signaling pathways, oxidative stress, and genetic stabilities were analyzed using Cell Counting Kit-8, western blot, flow cytometry, γH2AX foci formation, and comet assay. The results showed that SPR signals were decreased under MF exposure in a time- and dose-dependent manner, and the decreased SPR signals were reversible when the exposure was drawn off. However, MF pre-exposed culture medium did not significantly change cell viability, intracellular reactive oxygen species level, activation of the MARK signaling pathways, or genetic stabilities in human amniotic epithelial cells (FL cells). In conclusion, our data suggest that the relative permittivity of culture medium was influenced by 50 Hz MF exposure, but this change did not affect the biological processes in FL cells.

Abstract:The way that living cells respond to non-ionizing electromagnetic fields (EMF), including static/extremely-low frequency and radiofrequency electromagnetic fields, fits the pattern of 'cellular stress response' – a mechanism manifest at the cellular level intended to preserve the entire organism. It is a set pattern of cellular and molecular responses to environmental stressors, such as heat, ionizing radiation, oxidation, etc. It is triggered by cellular macromolecular damage (in proteins, lipids, and DNA) with the goal of repairing and returning cell functions to homeostasis. The pattern is independent of the type of stressor encountered. It involves cell cycle arrest, induction of specific molecular mechanisms for repair, damage removal, cell proliferation, and cell death if damage is too great. This response could be triggered by EMF-induced alternation in oxidative processes in cells. The concept that biological response to EMF is a 'cellular stress response' explains many observed effects of EMF, such as nonlinear dose- and time-dependency, increased and decreased risks of cancer and neurodegenerative diseases, enhanced nerve regeneration, and bone healing. These responses could be either detrimental or beneficial to health, depending on the duration and intensity of the exposure, as well as specific aspects of the living organism being exposed. A corollary to electromagnetic hypersensitivity syndrome (EHS) could be an inappropriate response of the hippocampus/limbic system to EMF, involving glucocorticoids on the hypothalamic-pituitary-adrenal axis.

Abstract:Emerging evidence indicates that the cellular electromagnetic field regulates the fundamental physics of cell biology. The electromagnetic oscillations and synchronization of biomolecules triggered by the internal and external pulses serve as the physical basis of the cellular electromagnetic field. Recent studies have indicated that centrosomes, a small organelle in eukaryotic cells that organize spindle microtubules during mitosis, also function as a nano-electronic generator in cells. Additionally, cellular electromagnetic fields are defined by cell types and correlated to the epigenetic status of the cell. These interactions between tissue-specific electromagnetic fields and chromatin fibers of progenitor cells regulate cell differentiation and organ sizes. The same mechanism is implicated in the regulation of tissue homeostasis and morphological adaptation in evolution. Intercellular electromagnetic interactions also regulate the migratory behaviors of cells and the morphogenesis programs of neural circuits. The process is closely linked with centrosome function and intercellular communication of the electromagnetic fields of microtubule filaments. Clearly, more and more evidence has shown the importance of cellular electromagnetic fields in regulatory processes. Furthermore, a detailed understanding of the physical nature of the inter- and intracellular electromagnetic interactions will better our understanding of fundamental biological questions and a wide range of biological processes.

Abstract:A general theory explaining how electromagnetic waves affect cells and biological systems has not been completely accepted yet; nevertheless, extremely low-frequency electromagnetic fields (ELF-EMFs) can interfere with and modify several molecular cellular processes. The therapeutic effect of EMFs has been investigated in several clinical conditions with promising results: in this context a better understanding of mechanisms by which ELF-EMF influences cellular events is necessary and it could lead to more extended and specific clinical applications in different pathological conditions. This paper develops a thermodynamic model to explain how ELF-EMF directly interferes with the cellular membrane, inducing a biological response related to a cellular energy conversion and modification of flows across cell membranes. Indeed, energy, irreversibly consumed by cellular metabolism, is converted into entropy variation. The proposed thermodynamic model views living systems as adaptative open systems, analysing the changes in energy and matter moving in and out of the cell.

Biological effects of low power nonionizing radiation: A Narrative Review

Abstract:Background and controlled electromagnetic radiation (EMR) on biological cells and tissues induces thermal, non-thermal, and dielectric property change. After EMR interaction with cells/tissues the resulting signal is used for imaging, bio-molecular response, and photo-biomodulation studies at infrared regime, and for therapeutic use. We attempt to present a review of current literature with a focus to present compilation of published experimental results for each regime viz. microwave (extremely low frequency, ELF to 3 GHz), to cellular communication frequencies (100 KHz to 300 GHz), millimeter wave (300 GHz- 1 THz), and the infra-red band extending up to 461 THz. A unique graphical representation of frequency effects and their relevant significance in detection of direct biological effects, therapeutic applications and biophysical interpretation is presented. A total of seventy research papers from peer-reviewed journals were used to compile a mixture of useful information, all presented in a narrative style. Out of the Journal articles used for this paper, 63 journal articles were published between 2000 to 2020. Physical, biological, and therapeutic mechanisms of thermal, non-thermal and complex dielectric effects of EMR on cells are all explained in relevant sections of this paper. A broad up to date review for the EMR range KHz-NIR (kilohertz to near infra-red) is prepared. Published reports indicate that number of biological cell irradiation impact studies fall off rapidly beyond a few THz EMR, leading to relatively a smaller number of studies in FIR and NIR bands covering most of the thermal effects and microthermal effects, and rotation-vibration effects.

Abstract:With the wide applications of magnetic fields (MFs) in medicine, researchers from different disciplines have gained interest in understanding the effect of various types of MFs on living cells and organisms. In this paper, we mainly focus on the immunological and physical aspects of the immune responses and their mechanisms under different types of MFs. Immune cells were slightly affected by low-frequency alternating MFs but were strongly influenced by moderate-intensity MFs and high-gradient MFs (HGMFs). Larger immune cells, such as macrophages, were more sensitive to HGMFs, which biased the cell polarization into the anti-inflammatory M2 phenotype. Subject to the gradient forces of varying directions and strength, the elongated M2 macrophage also remodeled the cytoskeleton with actin polymerization and changed the membrane receptors and ion channel gating. These alterations were very similar to changes caused by the small GTPase RhoA interference in macrophage. Regulation of iron metabolism may also contribute to the MF effects in macrophages. High MFs were found to regulate the iron content in monocyte-/macrophage-derived osteoclasts by affecting the expression of iron-regulation genes. On the other hand, paramagnetic nanoparticles (NPs) combined with external MFs play an important role in T-cell immunity. Paramagnetic NP-coated T-cells can cluster their T-cell receptors (TCRs) by using an external MF, thus increasing the cell–cell contact and communication followed by enhanced tumor killing capacity. The external MF can also guide the adoptively transferred magnetic NP-coated T-cells to their target sites <i>in vivo</i>, thus dramatically increasing the efficiency of cell therapy. Additionally, iron oxide NPs for ferroptosis-based cancer therapy and other MF-related therapeutic applications with obstacles were also addressed. Furthermore, for a profound understanding of the effect of MFs on immune cells, multidisciplinary research involving both experimental research and theoretical modeling is essential.

Abstract:Time‐varying electromagnetic fields (EMF) can induce some physiological effects in neuronal tissues, which have been explored in many applications such as transcranial magnetic stimulation. Although transmembrane potentials and induced currents have already been the subjects of many theoretical studies, most previous works about this topic are mainly completed by utilizing Maxwell's equations, often by solving a Laplace equation. In previous studies, cells were often considered to be three‐compartment models with different electroconductivities in different regions (three compartments are often intracellular regions, membrane, and extracellular regions). However, models like that did not take dynamic ion channels into consideration. Therefore, one cannot obtain concrete ionic current changes such as potassium current change or sodium current change by these models. The aim of the present work is to present a new and more detailed model for calculating transmembrane potentials and ionic currents induced by time‐varying EMF. Equations used in the present paper originate from Nernst‐Plank equations, which are ionic current‐related equations. The main work is to calculate ionic current changes induced by EMF exposure, and then transmembrane potential changes are calculated with Hodgkin‐Huxley model. Abstract:In a number of recently published experimental studies from our research group, the positive impact of the magnetic stimuli (static/pulsed) on cell functionality modulation or bactericidal effect, in vitro has been established. In order to develop the theoretical understanding towards such magnetobiological effects, the present study is aimed to present two quantitative models to determine magnetic Maxwell stresses as well as pressure acting on the cell membrane, under the influence of time varying magnetic field. The model predicts that magnetic field induced stress on the cell/bacterial is dependent on the conductivity properties of extracellular region and is determined to be of significantly lower magnitude to cause any significant effect. However, the force on the cell/bacteria due to electric field is more significant and was used to determine the membrane tension that can cause membrane poration. With a known critical membrane tension for cells, the field parameters necessary to cause membrane rupture were determined. Based on the experimental results and theoretically predicted values, the field parameters can be classified into three regimes, wherein the magnetic fields cause no effect or result in biophysical stimulation or induce cell death due to membrane damage. Taken together, this work provides some quantitative insights into the impact of magnetic field on the biological system.

Abstract:There have been a great number of investigations about the influence of weak magnetic fields on biological systems, such as isolated cells and whole organisms. This is also a subject of considerable medical concern since old epidemiologic observations have indicated a possible tumorigenic effect of these fields. Their mechanism of action, however, is not firmly established. A large number of biological effects of electromagnetic fields have been attributed either to the production of reactive oxygen species (ROS) or to the entrance Ca2+ in the cell. A new biochemical pathway is proposed that covers these two possibilities: the primary effect of the magnetic field would be by the mechanism of radical pairs resulting in the production of ROS; these could activate the ion channels TRPM2 producing cellular inflow of Ca2+, which would induce the calcium dependent effects. Thus, a large number of biological effects observed up to the present could be explained.

Abstract:Biological cells, characterized by complex and dynamic structures, demand precise models for comprehensive understanding, especially when subjected to external factors such as electric fields (EF) for manipulation or treatment. This interaction is integral to technologies like pulsed electric fields (PEF), inducing reversible and irreversible structural variations. Our study explores both simplified and sophisticated equivalent circuit models for biological cells under the influence of an external EF, covering diverse cell structures from single- to double-shell configurations. The paper highlights challenges in circuit modeling, specifically addressing the incorporation of reversible or irreversible pores in the membrane during external EF interactions, emphasizing the need for further research to refine technical aspects in this field. Additionally, we review a comparative analysis of the performance and applicability of the proposed circuit models, providing insights into their strengths and limitations. This contributes to a deeper insight of the complexities associated with modeling biological cells under external EF influences, paving the way for enhanced applications in medical and technological domains in future.&#xD.

Abstract:The electromagnetic fields have a great influence on the behaviour of all the living systems. The as low as reasonably achievable (ALARA) principle imposes, in case of long exposures to low (i.e. power systems) or high frequency (i.e. microwave systems or cell phones) fields, some limitations to the radiated fields by the industrial equipment. On the other hand, some benefits can be taken from the effects of the electromagnetic fields on the living being: the hyperthermal technique is well known for the treatment of the cancer. Either we want to be protected from the fields, or we want to take benefit of the positive effects of these fields, all the effects thermal as well as genetic have to be well known. Like in any industrial application, the electromagnetic field computation allows a better knowledge of the phenomena, and an optimised design. Hence, there is a very important challenge for the techniques of computation of electromagnetic fields. The major difficulties that appear are: (1) related to the material properties – the “material” (the human body) has very unusual properties (magnetic permeability, electric permittivity, electric conductivity), these properties are not well known and depend on the activity of the person, and this material is an active material at the cell scale; (2) related to the coupling phenomena – the problem is actually a coupled problem: the thermal effect is one of the major effects and it is affected by the blood circulation; (3) related to the geometry – the geometry is complex and one has to take into account the environment. The problems that we have to face with are – the identification of the properties of the “material”, the coupled problem solution and the representation of the simulated phenomena.

Abstract:Electromagnetic medical devices refer to a category of equipment used for medical diagnosis and treatment by utilizing electric field, magnetic field, or electromagnetic field. Electromagnetic biological effects including the thermal effects and non-thermal effectsare the main principle and the bases of assessment for innovative electromagnetic medical devices. Physical factors influencing the biological effects include electromagnetic energy as well as permittivity and permeability of organisms. Thermal effects are evidenced by temperature rise. The cells do not suffer from obvious injury, such as the reversible and/or irreversible damage injury caused by the process of coagulation, drying, carbonization or vaporization, which make it able to be applied in high-frequency therapy, microwave tumor therapy, radiofrequency ablation, electric surgery,etc. Non-thermal effects is evidenced bybiochemical and physiological response in the molecules, cells, tissues, organs and body with no temperature rise in the organism when exposed to the electromagnetic field, which can be applied to pacemakers, heart defibrillation, extracorporeal shock wave lithotripsy, and so on. In recent years, research has carried out in both the basic principles and equipment of electromagnetic medical devices. Some electromagnetic medical devices were developed, such as electromagnetic-phoresis apparatuses, transdermal drug delivery, and irreversible electroporation tumor therapy. Effective means of safety control of electromagnetic medical device is to establish the electromagnetic limit standard and to implement according to the standard based on human physiological integrity.

Abstract:Magnetic fields are widely used in medical diagnostics because of their superior non-invasive properties. In addition, with the widespread use of magnetic fields in transportation and other areas, their potential hazards to human health and the assessment of their safety have attracted considerable attention. The effects of magnetic fields on living organisms have a long history. The biological effects of magnetic field exposure in mice and rats depend on the magnetic field strength, exposure time, and direction; depending on these and potentially other factors, magnetic fields can cause a series of neurobiological effects. We reviewed global research on the neurobiological effects of magnetic fields from recent years to provide an overview and insights into the underlying mechanisms. This review focuses on the biological effects of static and dynamic magnetic fields of different frequencies and intensities on animals and nerve cells and their mechanisms of action.

Abstract:A number of effects of weak combined (static and alternating) magnetic fields with an alternating component of tens and hundreds nT at a collinear static field of 42 μT, which is equivalent to the geomagnetic field, have been found: activation of fission and regeneration of planarians Dugesia tigrina, inhibition of the growth of the Ehrlich ascites carcinoma in mice, stimulation of the production of the tumor necrosis factor by macrophages, decrease in the protection of chromatin against the action of DNase 1, and enhancement of protein hydrolysis in systems in vivo and in vitro. The frequency and amplitude ranges for the alternating component of weak combined magnetic fields have been determined at which it affects various biological systems. Thus, the optimal amplitude at a frequency of 4.4 Hz is 100 nT (effective value); at a frequency of 16.5 Hz, the range of effective amplitudes is broader, 150–300 nT; and at a frequency of 1 (0.5) Hz, it is 300 nT. The sum of close frequencies (e.g., 16 and 17 Hz) produces a similar biological effect as the product of the modulating (0.5 Hz) and carrying frequencies (16.5 Hz), which is explained by the ratio A = A0sinω1t + A0sinω2t = 2A0sin(ω1 + ω2)t/2cos(ω1–ω2)t/2. The efficiency of magnetic signals with pulsations (the sum of close frequencies) is more pronounced than that of sinusoidal frequencies. These data may indicate the presence of several receptors of weak magnetic fields in biological systems and, as a consequence, a higher efficiency of the effect at the simultaneous adjustment to these frequencies by the field. Even with consideration of these facts, the mechanism of the biological action of weak combined magnetic fields remains still poorly understood.

Abstract:The interaction of static magnetic fields (SMFs) with living organisms is a rapidly growing field of investigation. The magnetic fields (MFs) effect observed with radical pair recombination is one of the well-known mechanisms by which MFs interact with biological systems. Exposure to SMF can increase the activity, concentration, and life time of paramagnetic free radicals, which might cause oxidative stress, genetic mutation, and/or apoptosis. Current evidence suggests that cell proliferation can be influenced by a treatment with both SMFs and anticancer drugs. It has been recently found that SMFs can enhance the anticancer effect of chemotherapeutic drugs; this may provide a new strategy for cancer therapy. This review focuses on our own data and other data from the literature of SMFs bioeffects. Three main areas of investigation have been covered: free radical generation and oxidative stress, apoptosis and genotoxicity, and cancer. After an introduction on SMF classification and medical applications, the basic phenomena to understand the bioeffects are described. The scientific literature is summarized, integrated, and critically analyzed with the help of authoritative reviews by recognized experts; international safety guidelines are also cited.

EMF can be classified into extremely low-frequency electromagnetic field, low-frequency magnetic field, static moderate magnetic field, strong electromagnetic field, alternating magnetic field, and magnetic nanoparticles. Each type of EMF influences melanoma development differently, and the detailed influence of each specific type of EMF on melanoma is reviewed. Furthermore, EMF influences melanoma cell polarity and hence affects drug uptake. In this review, the impacts of EMF on the effectiveness of drugs used to treat melanoma are listed according to drug types, with detailed effects according to the types of EMF and specific melanoma cell lines. EMF also impacts clinical therapies of melanoma, including localized magnetic hyperthermia, focalized thermotherapy, proton radiation treatment, nanostructure heating magnetic hyperthermia, radiation therapy, Polycaprolactone-Fe3O4 fiber mat-based bandage, and optune therapy. Above all, EMF has huge potential in melanoma treatment.

Abstract:All organisms on Earth live in the weak but ubiquitous geomagnetic field. Human beings are also exposed to magnetic fields generated by multiple sources, ranging from permanent magnets to magnetic resonance imaging (MRI) in hospitals. It has been shown that different magnetic fields can generate various effects on different tissues and cells. Among them, stem cells appear to be one of the most sensitive cell types to magnetic fields, which are the fundamental units of regenerative therapies. In this review, we focus on the bioeffects of static magnetic fields (SMFs), which are related to regenerative medicine. Most reports in the literature focus on the influence of SMF on bone regeneration, wound healing, and stem cell production. Multiple aspects of the cellular events, including gene expression, cell signaling pathways, reactive oxygen species, inflammation, and cytoskeleton, have been shown to be affected by SMFs. Although no consensus yet, current evidence indicates that moderate and high SMFs could serve as a promising physical tool to promote bone regeneration, wound healing, neural differentiation, and dental regeneration. All in vivo studies of SMFs on bone regeneration and wound healing have shown beneficial effects, which unravel the great potential of SMFs in these aspects. More mechanistic studies, magnetic field parameter optimization, and clinical investigations on human bodies will be imperative for the successful clinical applications of SMFs in regenerative medicine.
