Abstract:In this work, dual-column capillary microextraction (CME) system consisting of N-(2-aminoethyl)-3-aminopropyltrimethoxysilane (AAPTS)-silica coated capillary (C1) and 3-mercaptopropyl trimethoxysilane (MPTS)-silica coated capillary (C2) was developed for sequential separation/preconcentration of arsenite [As(III)], arsenate [As(V)], monomethylarsonic acid [MMA(V)] and dimethylarsinic acid [DMA(V)] in the extracts of human hair followed by electrothermal vaporization inductively coupled plasma mass spectrometry (ETV-ICP-MS) detection with iridium as permanent modifier. Various experimental parameters affecting the dual-column microextraction of different As species had been investigated in detail. It was found that at pH 9, As(V) and MMA could be quantitatively retained by C1 and only As(III) could be quantitatively retained by C2. With the aid of valve switching, As(V)/MMA(V) retained on C1 and As(III) retained on C2 could be sequentially desorbed by 10 microl of 0.01 mol l(-1) HNO(3) [for As(V)], 0.1 mol l(-1) HNO(3) [for MMA(V)] and 0.2 mol l(-1) HNO(3)-3% thiourea (m/v) [for As(III)], respectively, the eluents were immediately introduced into the Ir-coated graphite tubes for further ETV-ICP-MS detection. With two-step ETV pyrolysis program, Cl(-) in the sample matrix could be in situ removed, and the total As in the human hair extracts or digested solution could be interference-free, determined by ETV-ICP-MS. DMA(V) in the human hair extracts was obtained by subtraction of total As in the human hair extracts from other three As species. Under the optimized conditions, the detection limits (3 sigma) of the method were 3.9 pg ml(-1) for As(III), 2.7 pg ml(-1) for As(V), 2.6 pg ml(-1) for MMA(V) and 124 pg ml(-1) for total As with the relative standard deviations less than 7.0% (C = 0.1 ng ml(-1), n = 7), and the enrichment factor was 286, 262 and 260 for As(III), As(V) and MMA(V), respectively. The developed method was successfully applied for the speciation of arsenic in the extracts of human hair.

Abstract:The current paper describes a method for determining As(III) and As(V) concentrations in wastewater samples. The approach is based on the extraction of As(III) from water samples using a non-polar organic solvent in a 10-12 M HCl medium. The extract contains As(III), while the raffinate contains As(V). Distilled water can be used to re-extract As(III) from the organic solvent. ICP-MS was used to analyze As concentrations in the reextract and the raffinate. The average recovery of arsenic by the proposed method is 99.6 %. It was calculated as the sum of As(III) and As(V) concentrations. The collected data demonstrated high repeatability. The standard addition procedure demonstrated the selectivity and accuracy of the employed method. The relative error of recovery of the standard As(III) addition is about 2 %, which is similar to the RSD found under repeatability conditions.

Abstract:Multi-elemental speciation analysis in a single run reduces costs, waste and time, and also sample consumption, which is favorable to a large set of instable samples. This work demonstrates mercury and arsenic speciation in a single run using high performance liquid chromatography with inductively coupled plasma mass spectrometric detection (HPLC/ICP-MS). Thiol compounds in mobile phases interfered with the elution of arsenite because of coordination action, which was overcome by gradient elution (elution of As(III) by thiol-free mobile phases at first). Four arsenic forms (As(III), DMA, MMA and As(V)) and three mercury species (Hg(II), MeHg and EtHg) were separated and detected by HPLC/ICP-MS using 4 mM tetrabutylammonium hydroxide (TBAH) (pH 6.0) in the first 1.0 min and 4 mM TBAH + 20 mM L-cysteine (pH 6.0) in the next 9.0 min. The detection limits were obtained at 0.078 for As(III), 0.081 for DMA, 0.078 for MMA and 0.15 mu g L-1 for As(V), and 0.016 for Hg(II), 0.027 for MeHg and 0.032 mu g L-1 for EtHg, and the reproducibilities (50 mu g L-1 for each As-species and 10 mu g L-1 for each Hg-species) were all lower than 3.7%. The proposed method was successfully applied for speciation analyses of arsenic and mercury in five lotus roots, which indicated DMA (3.5-7.9 mu g kg(-1)) as the only arsenic species and MeHg (0.6-1.0 mu g kg(-1)) as the only mercury species. Analyses of two certified reference materials demonstrated good agreement between determined values and certified values. All results prove its advantages including high sensitivity and high analytical efficiency, which are beneficial for routine speciation analyses of arsenic and mercury in a single run in environmental and food fields.

Abstract:Abstract An on-line method capable of the separation of arsenic species was developed for the speciation of arsenite As(III), arsenate As(V), monomethylarsenic (MMA) and dimethylarsenic acid (DMA) in biological samples. The method is based on the combination of high-performance liquid chromatograph (HPLC) for separation, UV photo oxidation for sample digestion and hydride generation atomic fluorescence spectrometry (HGAFS) for sensitive detection. The best separation results were obtained with an anion-exchange AS11 column protected by an AG11 guard column, and gradient elution with NaH2PO4 and water as mobile phase. The on-line UV photo oxidation with 1.5% K2S2O8 in 0.2 mol L–1 NaOH in an 8 m PTFE coil for 40 s ensures the digestion of organoarsenic compounds. Detection limits for the four species were in the range of 0.11–0.15 ng (20 μL injected). Procedures were validated by analysis of the certified reference materials GBW09103 freeze-dried human urine and the results were in good agreement with the certified values of total arsenic concentration. The method has been successfully applied to speciation studies of blood arsenic species with no need of sample pretreatment. Speciation of arsenic in blood samples collected from two patients after the ingestion of realgar-containing drug reveals slight increase of arsenite and DMA, resulting from the digestion of realgar.

Abstract:In this paper, a new extraction method based on countercurrent liquid–liquid microextraction (CLLME) has been developed for the extraction and preconcentration of inorganic arsenic (iAs) in plasma and urine samples prior to their analysis by electrothermal atomic absorption spectrometry (ETAAS). In this method, firstly, 5 ml of water is added to the extraction vessel. Then 30.0 μl of the extracting solvent is added to it in order for the extracting solvent to be placed in the narrow-necked vessel. In total, 10 ml of a standard solution or a pretreated real sample is added to the sample container and it is connected to the extraction vessel via a connector. While opening the embedded valve at the bottom of the sample container and the one in the extraction vessel, the sample solution flows into the extracting solvent with the same flow rate, leading to the successful extraction of metal ligand into the extracting organic solvent. Under the optimum conditions, calibration curves are linear in the range of 0.1–50 μg l−1, and limit of detections (LODs) are in the range of 0.03–0.05 μg l−1. The enhancement factor and enrichment factor were in the range of 220–240 and 198–212, respectively. Repeatability (intra-day) and reproducibility (inter-day) of method based on seven replicate measurements of 5.0 μg l−1 of arsenic were in the range of 2.3–3.5% and 4.0–5.7%, respectively. The applicability of the proposed CLLME and ETAAS methods was demonstrated by analyzing the iAs in spiked urine and plasma samples. The obtained recoveries of the arsenic in the range of 92–107% indicated the excellent capability of the developed method for speciation of arsenic from plasma and urine samples.Graphical Abstractᅟ

Abstract:This work establishes a hyphenated methodology coupling HPLC with ICP-MS for simultaneous speciation analysis of arsenic, mercury and lead for the first time. Four arsenicals (As(III), DMA, MMA and As(V)), four mercurials (Hg(II), MeHg, EtHg and PhHg) and three lead compounds (Pb(II), TML and TEL) were simultaneously analyzed within only 8 min with acceptable resolution (2.0-8.2 for As, 1.6-6.1 for Hg and 2.7-4.0 for Pb). The detection limits were 0.036-0.20 for As-species, 0.023-0.041 for Hg-species, and 0.0076-0.14 mu g L-1 for Pb-species. The developed method was applied for the measurement of five lotus seed samples, indicating the presence of DMA (19.6-28.2 mu g kg(-1)), TML (1.4-2.9 mu g kg(-1)), MeHg (1.2-4.8 mu g kg(-1)) and EtHg (0.8-2.2 mu g kg(-1)). This method provides a promising tool for studying the toxic, metabolic and bioavailable behaviors of arsenic, mercury and lead.

Abstract:<p>Arsenic (As) is a ubiquitous element in the environment while its toxicity and chemical properties are highly associated with its chemical form. High quality As speciation data in environmental matrices are thus desirable for understanding its biogeochemistry. Here we present a simple and novel head space-single drop microextraction-spectro-pipette (HS-SDME-SP) method for the on-site measurement of As species in natural waters. Arsine was generated and separated from the sample matrix by borohydride-based hydride generation, which was further extracted into a stable, hanging 5 μL microdroplet on the tip of a spectro-pipette. With the spectro-pipette, the microvolume of extractant is easily handled while gas–liquid absorption, redox and chromogenic reactions and <em>in situ</em> absorbance measurement can be carried out in a single step. The speciation analysis of As(III) and As(V) was achieved by utilizing their different pH requirement for reduction and arsine generation in the hydride generation process. Although the proposed method is a simple sensor based assay, it is sensitive, robust and reliable; the limit of detection (3 × S/N) was 0.1 nM, RSDs for 25 nM As(III) were < 2.0% (n = 10) and results were comparable with the commonly used hydride generation atomic fluorescence spectrometry (HG-AFS) method and in good agreement with two certified reference materials (surface freshwater and seawater). The proposed method is easy to operate, the device is light and portable (1.1 kg) and is practical for the measurement of trace inorganic As species in the field.</p>

Abstract:Magnetic dispersive microsolid phase extraction (MDMSPE) was coupled with dispersive liquid–liquid microextraction (DLLME) for the separation and preconcentration of arsenic(V) and arsenic(III) prior to graphite furnace atomic absorption spectrometry detection. In the first step, magnetic ZnFe2O4 nanotubes were employed for adsorbing arsenic(V). The adsorbent was separated from the aqueous phase by the use of an external magnetic field. Next, arsenic(III) in the upper aqueous phase from MDMSPE was enriched by DLLME. Samples were prepared with artificial gastric juice to avoid inter-conversion of the determined species. The parameters affecting the separation and preconcentration of the determined species were optimized. The detection limits of this protocol were determined to be 1.3 and 2.0 pg mL−1 for arsenic(III) and arsenic(V) with relative standard deviations of 4.5% and 5.6% for a concentration of 1.0 ng mL−1 (n = 9). The enrichment factors were equal to 175-fold for arsenic(III) and 163-fold for arsenic(V). The developed protocol was employed for the determination of arsenic(III) and arsenic(V) in dairy products without preoxidation or prereduction. The recoveries of spiked samples were from 90.8% to 105%. A milk powder certified reference material was analyzed by this approach and the measured results were in good agreement with the certified value.

Abstract:A new method based on dispersive liquid liquid microextraction (DLLME) combined with electrothermal vaporization inductively coupled plasma mass spectrometry (ETV-ICP-MS) was developed for the simultaneous speciation of inorganic arsenic (As), selenium (Se) and tellurium (Te) with sodium diethyldithiocarbamate (DDTC) as both chelating reagent and chemical modifier. As(III), Se(IV) and Te(IV) were transformed into DDTC-chelates at pH 7 and extracted into the fine droplets formed by injecting the binary solution of bromobenzene (extraction solvent) and methanol (dispersive solvent) into the sample solution. After phase separation by centrifugation, As(III), Se(IV) and Te(IV) preconcentrated in the organic phase were determined by ETV-ICP-MS. Total inorganic As, Se and Te were obtained by reducing As(V), Se(VI) and Te(VI) to As(III), Se(IV) and Te(IV) with L-cysteine, which were then subjected to the same DLLME-ETV-ICP-MS process. The concentration of As(V), Se(VI), Te(VI) were calculated by subtracting the concentration of As(III), Se(IV) and Te(IV) from the total inorganic As, Se and Te, respectively. The main factors affecting the microextraction efficiency and the vaporization behavior of target species were investigated in detail. Under the optimal conditions, the limits of detection were 2.5, 8.6 and 0.56 ng L(-1) for As(III), Se(IV) and Te(IV), respectively, with the relative standard deviations (n=7) of 8.5-9.7%. The developed method was applied to the speciation of inorganic As, Se and Te in Certified Reference Materials of GSBZ50004-88, GBW(E)080395 and GBW(E)080548 environmental waters, and the determined values are in good agreement with the certified values. The method was also successfully applied to the simultaneous speciation of inorganic As, Se and Te in different environmental water samples with the recoveries in the range of 86.3-107% for the spiked samples.

Abstract:Nanoliter high-performance liquid chromatography shows low consumption of solvents and samples, offering one of the best choices for arsenic speciation in precious samples in combination with inuctively coupled plasma mass spectrometry. A systematic investigation on coupling nanoliter high-performance liquid chromatography to inductively coupled plasma mass spectrometry from instrument design to injected sample volume and mobile phase was performed in this study. Nanoflow mobile phase was delivered by flow splitting using a conventional high-pressure pump with reuse of mobile phase waste. Dead volume was minimized to 60 nL for the sheathless interface based on the previously developed nanonebulizer. Capillary columns for nanoliter high-performance liquid chromatography were found to be sensitive to sample loading volume. An apparent difference was also found between the mobile phases for nanoliter and conventional high-performance liquid chromatography. Baseline separation of arsenite, arsenate, monomethylarsenic, and dimethylarsenic was achieved within 11 min on a 15 cm C-18 capillary column and within 12 min on a 25 cm strong anion exchange column. Detection limits of 0.9-1.8 mu g/L were obtained with precisions variable in the range of 1.6-4.2%. A good agreement between determined and certified values of a certified reference material of human urine (GBW 09115) validated its accuracy along with good recoveries (87-102%).

Abstract:Toxicity, mobility, bioavailability and biofunctions of chromium, cadmium, mercury and lead are heavily dependent upon their specific chemical forms, leading to a high demand to metal speciation analysis rather than total quantification. Simultaneous speciation analysis of multiple metal(loid)s is attractive to a large sample capacity containing unstable analytes due to its economic and environmental advantages over the conventional single elemental strategies. In this work, an analytical method integrating online solid phase extraction into high performance liquid chromatography interfaced with inductively coupled plasma mass spectrometry (ICP-MS) to simultaneously preconcentrate and quantify Cr, Cd, Hg and Pb forms in pg L-1 levels in water was developed. Cr (III + VI), Cd(II), Hg(II), Pb(II), methylmercury (MeHg), ethylmercury (EtHg), and trimethyl (TML) and triethyl lead (TEL) were captured by the C18 adsorbent (equilibrated with 10 mL of 1.0 mM 2-hydroxyethanethiol at 10 mL min-1) and eluted by mobile phase (5.0 mM Cys at pH 2.0), then completely separated on the C18 column within 8.0 min and eventually determined by ICP-MS. Low limits of detection (0.001-0.007 ng L-1) and quantification (0.003-0.023 ng L-1), good relative standard deviations (<4%) and high enrichment factors (827-2656 folds) were obtained with good linearities. Three reference materials of total cadmium (GBW08602), total mercury (GBW08603) and total lead (GBW08601) in water were analyzed by the developed method to validate the accuracy with good agreement with certified values and satisfactory recoveries (92-100%). This method was proved feasible by the determination of Cr, Cd, Hg and Pb compounds in drinking water, river water, pond water and tap water.

Abstract:Direct determination of arsenic species in real samples is challenging due to their trace concentration and spectral interferences by coexisting ions. Herein, we proposed an ultrasound-assisted dispersive solid phase microextraction (DSPME) procedure for the analyses of the trace inorganic arsenic. The hydrothermally synthesized cadmium sulfide nanoparticles (CdS NPs) completely adsorbed both arsenic species within 20 s at the initial arsenic concentration of 100 µg L-1. The detection limit (3 S/m) of the proposed method was found to be 0.5 ± 0.2 and 0.8 ± 0.2 ng L-1 for As(III) and As(V), respectively. The accuracy of the method against the systematic and constant errors was confirmed by the analysis of the Standard Reference Material (SRM) (>95% recovery with <5% RSD). The Student's t-test values were found to be less than the critical Student's t value at a 95% confidence level. The method was successfully employed for the determination of arsenic in food samples.

Abstract:A simple and fast method of low-density solvent based dispersive liquid-liquid microextraction (LDS-DLLME) followed by high-performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICP-MS) has been developed for the speciation analysis of organoarsenic and inorganic arsenic in water samples. The low-density solvent (octanol) was given as the organic phase and injected into the aqueous sample (donor phase) with methanol as the disperser. With As (V), As (III), p-APAA, 4-HPAA, ROX and PAA as target species, factors of LDS-DLLME including pH value, anionic carriers, elution conditions and extractant, were studied in detail. Besides, volumes of solvents were further optimised by response surface methodology. Under the optimal conditions, the limits of detection for four phenylarsenics and arsenate were in the range of 0.001–0.039 μg L−1. The relative standard deviations (RSDs) were 3.6–9.4% and the enrichment factors varied from 6.2 to 70.8. The proposed method of LDS-DLLME-HPLC-ICP-MS was satisfactorily applied to the determination of six arsenic compounds in water samples with recoveries of 81.8–111.7% for the spiked lake water samples.

Abstract:A novel method for speciation analysis of inorganic arsenic was developed by on-line hyphenating microchip capillary electrophoresis (chip-CE) with hydride generation atomic fluorescence spectrometry (HG-AFS). Baseline separation of As(III) and As(V) was achieved within 54s by the chip-CE in a 90mm long channel at 2500V using a mixture of 25mmoll−1 H3BO3 and 0.4mmoll−1 CTAB (pH 8.9) as electrolyte buffer. The precisions (RSD, n=5) ranged from 1.9 to 1.4% for migration time, 2.1 to 2.7% for peak area, and 1.8 to 2.3% for peak height for the two arsenic species at 3.0mgl−1 (as As) level. The detection limits (3σ) for As(III) and As(V) based on peak height measurement were 76 and 112μgl−1 (as As), respectively. The recoveries of the spikes (1mgl−1 (as As) of As(III) and As(V)) in four locally collected water samples ranged from 93.7 to 106%.
Off-line separation and determination of inorganic arsenic species in natural water by high resolution inductively coupled plasma mass spectrometry with hydride generation combined with reaction of arsenic(V) and L-cysteine

Abstract:A method is presented for the determination of inorganic arsenic species by hydride generation high resolution inductively coupled plasma mass spectrometry. In the presence of 1.5mg/ml of L-cysteine, arsenic(V) is indirectly determined using the difference between As(III) concentrations determined after 2 and 8min of reaction time, whereas the As(III) concentration is obtained by taking the difference between the concentration of arsenic(III) determined after 8min and the concentration of arsenic(III) converted from arsenic(V) after 8min. 25 fold amounts of arsenic(III) and 100 fold amounts of arsenic(V) hardly affected the determination of 1ng/ml arsenic(V) and 100ng/l arsenic(III), respectively. Interferences from transition metal ions were removed by using a Chelex 100 resin. The detection limits for arsenic(III) and arsenic(V) were 11.8 and 39.7ng/l, respectively. The proposed method for the simultaneous determination of inorganic arsenic species was applied to the determination of As(III) and As(V) in spiked and Japanese standard river water samples. Agreement within statistical uncertainty was obtained in all instances.

Abstract:It has been challenging to analyze inorganic arsenic (iAs) with anion exchange HPLC-Electrospray Ionization-Mass spectrometry (HPLC-ESI-MS), because arsenite (As(III)) is difficult to retain on column and the salts in mobile phase causes ionization suppression of iAs. To address these issues, a method has been developed involving the determination of arsenate (As(V)) with mixed mode HPLC-ESI-MS and the conversion of As(III) to As(V) for total iAs. As(V) was separated from other chemicals on Newcrom B, a bi-modal HPLC column involving anion exchange and reverse phase interaction. The elution employed a two-dimensional gradient, including a formic acid gradient to elute As(V) and a concurrent alcohol gradient to elute organic anions used in sample preparations. As(V) was detected by Selected Ion Recording (SIR) in negative mode at m/z = 141 with a QDa (single quad) detector. As(III) was quantitatively converted to As(V) by mCPBA oxidation and measured for total iAs. By replacing salt with formic acid in elution, the ionization efficiency for As(V) was greatly enhanced in ESI interface. The limit of detection (LOD) for As(V) and As(III) were 0.0263 μM (1.97 ppb) and 0.0398 μM (2.99 ppb), respectively. The linear range was 0.05-1 μM. The method has been used to characterize iAs speciation change in the solution and precipitation in a simulated iron-rich groundwater caused by air exposure.

Abstract:In order to better distinguish the different toxic inorganic and organic forms of arsenic (As) exposure in individuals, we have developed and validated a simple and robust analytical method for determining the following six As species in human urine: arsenous (III) acid (As-III), As (V) acid, monomethylarsonic acid, dimethylarsinic acid, arsenobetaine (AsB), and arsenocholine. In this method, human urine is diluted using a pH 5.8 buffer, separation is performed using an anion exchange column with isocratic HPLC, and detection is achieved using inductively coupled plasma-MS. The method uses a single mobile phase consisting of low concentrations of both phosphate buffer (5 mM) and ammonium nitrate salt (5 mM) at pH 9.0; this minimizes the column equilibration time and overcomes challenges with separation between AsB and As-III. In addition, As-III oxidation is prevented by degassing the sample preparation buffer at pH 5.8, degassing the mobile phase online at pH 9.0, and by the use of low temperature (-70 °C) and flip-cap airtight tubes for long term storage of samples. The method was validated using externally provided reference samples. Results were in agreement with target values at varying concentrations and successfully passed external performance test criteria. Internal QC samples were prepared and repeatedly analyzed to assess the method's long-term precision, and further analyses were completed on anonymous donor urine to assess the quality of the method's baseline separation. Results from analyses of external reference samples agreed with target values at varying concentrations, and results from precision studies yielded absolute CV values of 3-14% and recovery from 82 to 115% for the six As species. Analysis of anonymous donor urine confirmed the well-resolved baseline separation capabilities of the method for real participant samples.
