Abstract:Modulators of cardiac cyclic adenosine 3’5’-monophosphate (cAMP) and/or its effector protein kinase A (PKA) are used to treat heart disease. Elucidating the mechanisms of endogenous and pharmacological modulators of cAMP, including phosphodiesterase (PDE) inhibitors, is key to predicating alternative drug therapies for heart failure (HF). PDEs regulate confined cAMP/PKA signaling pathways, depending on PDE isoform and localization. PDE3 regulates the SR and sarcolemmal membrane cAMP pools, controlling calcium influx and efflux at these domains. PDE1 regulates sarcolemmal cAMP/PKA signaling. Despite these differences, inhibitors of either increase heart contractility. However, inhibition of PDE3 is pro-arrhythmic, unlike that of PDE1. The regulation of localized cAMP signaling is key to controlled regulation of cell function, with ramifications for HF, and which GPCR lies upstream of PDE1 remains unknown. We hypothesized that the three cAMP-hydrolyzing cardiac PDEs (1, 3, 4) that are found in the human heart would regulate distinct cAMP signaling pathways. Using the cAMP biosensor EPAC-SH187, we performed fluorescence resonance energy transfer experiments in healthy guinea pig adult ventricular myocytes. Cells were treated with inhibitors of PDEs 1, 3, and 4 along with GPCR agonists isoproterenol, glucagon (gcg), and amthamine (amt), respectively for β-adrenergic, gcg and histamine H2 receptors at sub-maximal doses. Cell function and calcium data were obtained using IonOptix. We first asked if PDEi would synergistically regulate pools of cAMP, and found that PDE4i does at pools of cAMP generated by beta-agonism. PDE3i regulates that generated by amt at the H2R, with comparable effects observed in simultaneous or prior H2R stimulation. The combination of these sub-saturating doses exerted strong inotropic and lusitropic effects in paced myocytes. In neither of these domains, PDE1i changed cAMP levels. However, PDE1i intriguingly attenuated the level of gcg-stimulated cAMP production, suggesting a feedback loop mechanism. We conclude that PDEs 1, 3, and 4 regulate distinct pools of cAMP. Importantly, we demonstrate this in a species where the myocardial expression of these PDEs match that of humans as opposed to smaller rodent models.

Abstract:Abstract The aim of this study was to investigate the functional role of phosphodiesterase enzymes (PDE) in the isolated porcine ureter. Distal ureteral strips were mounted in organ baths and pre‐contracted with 5‐HT (100 μM). Upon generation of stable phasic contractions, PDE‐4 and PDE‐5 inhibitors were added cumulatively to separate tissues. PDE‐4 inhibitors, such as rolipram (10 nM and greater) and roflumilast (100 nM and greater), resulted in significant attenuation of ureteral contractile responses, while a higher concentration of piclamilast (1 μM and greater) was required to induce a significant depressant effect. The attenuation effect by rolipram was abolished by SQ22536 (100 μM). PDE‐5 inhibitors, such as sildenafil and tadalafil, were not nearly as effective and were only able to suppress the 5‐HT‐induced contractions at higher concentrations of 1 μM. Rolipram significantly enhanced the depressant effect of forskolin, while sodium nitroprusside‐induced attenuation of contractile responses remained unchanged in the presence of tadalafil. In summary, our study demonstrates that PDE‐4 inhibitors are effective in attenuating 5‐HT‐induced contractility in porcine distal ureteral tissues, while PDE‐5 inhibitors are less effective. These findings suggest that PDE‐4 inhibitors, such as rolipram, may hold promise as potential therapeutic agents for the treatment of ureteral disorders attributable to increased intra‐ureteral pressure.

Pharmacological and physiological assessment of serotonin formation and degradation in isolated preparations from mouse and human hearts

Abstract:Using transgenic (TG) mice that overexpress the human serotonin (5-HT) 4a receptor specifically in cardiomyocytes, we wanted to know whether 5-HT can be formed and degraded in the mammalian heart and whether this can likewise lead to inotropic and chronotropic effects in this TG model. We noted that the 5-HT precursor 5-hydroxy-tryptophan (5-HTP) can exert inotropic and chronotropic effects in cardiac preparations from TG mice but not from wild-type (WT) mice; similar results were found in human atrial preparations as well as in intact TG animals using echocardiography. Moreover, by immunohistochemistry we could detect 5-HT metabolizing enzymes and 5-HT transporters in mouse hearts as well as in human atria. Hence, in the presence of an inhibitor of aromatic l-amino acid decarboxylase, the positive inotropic effects of 5-HTP were absent in TG and isolated human atrial preparations, and, moreover, inhibitors of enzymes involved in 5-HT degradation enhanced the efficacy of 5-HT in TG atria. A releaser of neurotransmitters increased inotropy in the isolated TG atrium, and this effect could be blocked by a 5-HT 4a receptor antagonist. Fluoxetine, an inhibitor of 5-HT uptake, elevated the potency of 5-HT to increase contractility in the TG atrium. In addition, inhibitors of organic cation and monoamine transporters apparently reduced the positive inotropic potency of 5-HT in the TG atrium. Hence, we tentatively conclude that a local production and degradation of 5-HT in the mammalian heart and more specifically in mammalian myocytes probably occurs. Conceivably, this formation of 5-HT and possibly impaired degradation may be clinically relevant in cases of unexplained tachycardia and other arrhythmias. NEW & NOTEWORTHY The present work suggests that inotropically active serotonin (5-HT) can be formed in the mouse and human heart and probably by cardiomyocytes themselves. Moreover, active degradation of 5-HT seems to occur in the mammalian heart. These findings may again increase the interest of researchers for cardiac effects of 5-HT.

Abstract:5-hydroxytryptamine-4 (5-HT(4)) receptors have been proposed to contribute to the generation of atrial fibrillation in human atrial myocytes, but it is unclear if these receptors are present in the hearts of small laboratory animals (e.g. rat). In this study, we examined presence and functionality of 5-HT(4) receptors in auricular myocytes of newborn rats and their possible involvement in regulation of gap junctional intercellular communication (GJIC, responsible for the cell-to-cell propagation of the cardiac excitation). Western-blotting assays showed that 5-HT(4) receptors were present and real-time RT-PCR analysis revealed that 5-HT(4b) was the predominant isoform. Serotonin (1 microM) significantly reduced cAMP concentration unless a selective 5-HT(4) inhibitor (GR113808 or ML10375, both 1 microM) was present. Serotonin also reduced the amplitude of L-type calcium currents and influenced the strength of GJIC without modifying the phosphorylation profiles of the different channel-forming proteins or connexins (Cxs), namely Cx40, Cx43 and Cx45. GJIC was markedly increased when serotonin exposure occurred in presence of a 5-HT(4) inhibitor but strongly reduced when 5-HT(2A) and 5-HT(2B) receptors were inhibited, showing that activation of these receptors antagonistically regulated GJIC. The serotoninergic response was completely abolished when 5-HT(4), 5-HT(2A) and 5-HT(2B) were simultaneously inhibited. A 24 h serotonin exposure strongly reduced Cx40 expression whereas Cx45 was less affected and Cx43 still less. In conclusion, this study revealed that 5-HT(4) (mainly 5-HT(4b)), 5-HT(2A) and 5-HT(2B) receptors coexisted in auricular myocytes of newborn rat, that 5-HT(4) activation reduced cAMP concentration, I(Ca)(L) and intercellular coupling whereas 5-HT(2A) or 5-HT(2B) activation conversely enhanced GJIC.

Pacing-induced regional differences in adenosine receptors mRNA expression in a swine model of dilated cardiomyopathy

Abstract:The adenosinergic system is essential in the mediation of intrinsic protection and myocardial resistance to insult; it may be considered a cardioprotective molecule and adenosine receptors (ARs) represent potential therapeutic targets in the setting of heart failure (HF). The aim of the study was to test whether differences exist between mRNA expression of ARs in the anterior left ventricle (LV) wall (pacing site: PS) compared to the infero septal wall (opposite region: OS) in an experimental model of dilated cardiomyopathy. Cardiac tissue was collected from LV PS and OS of adult male minipigs with pacing-induced HF (n = 10) and from a control group (C, n = 4). ARs and TNF-α mRNA expression was measured by Real Time-PCR and the results were normalized with the three most stably expressed genes (GAPDH, HPRT1, TBP). Immunohistochemistry analysis was also performed. After 3 weeks of pacing higher levels of expression for each analyzed AR were observed in PS except for A(1)R (A(1)R: C = 0.6±0.2, PS = 0.1±0.04, OS = 0.04±0.01, p<0.0001 C vs. PS and OS respectively; A(2A)R: C = 1.04±0.59, PS = 2.62±0.79, OS = 2.99±0.79; A(2B)R: C = 1.2±0.1, PS = 5.59±2.3, OS = 1.59±0.46; A(3)R: C = 0.76±0.18, PS = 8.40±3.38, OS = 4.40±0.83). Significant contractile impairment and myocardial hypoperfusion were observed at PS after three weeks of pacing as compared to OS. TNF-α mRNA expression resulted similar in PS (6.3±2.4) and in OS (5.9±2.7) although higher than in control group (3.4±1.5). ARs expression was mainly detected in cardiomyocytes. This study provided new information on ARs local changes in the setting of LV dysfunction and on the role of these receptors in relation to pacing-induced abnormalities of myocardial perfusion and contraction. These results suggest a possible therapeutic role of adenosine in patients with HF and dyssynchronous LV contraction.
Tegaserod Stimulates 5-HT4 Serotonin Receptors in the Isolated Human Atrium

Abstract:Tegaserod (1-{[(5-methoxy-1H-indol-3-yl)methyliden]amino}-3-pentylguanidine) is a potent agonist at human recombinant 5-HT4 serotonin receptors. Consequently, tegaserod is utilized in the treatment of bowel diseases. The objective of this study was to test the hypothesis that tegaserod stimulates human cardiac atrial 5-HT4-receptors via cyclic adenosine monophosphate (cAMP)-dependent pathways. Tegaserod exerted positive inotropic effects (PIEs) and positive chronotropic effects (PCEs) in isolated left and right atrial preparations, respectively, from mice with cardiac-specific overexpression of the human 5-HT4 serotonin receptor (5-HT4-TG) in a concentration- and time-dependent manner. However, no effect was observed in the hearts of littermates of wild-type mice (WT). Western blot analysis revealed that the expression of 5-HT4 receptors was significantly higher in 5-HT4-TG mice compared to WT mice. The specificity of the signal for the 5-HT4 receptor was confirmed by the absence of the signal in the hearts of 5-HT4 receptor knockout mice. Furthermore, tegaserod increased the force of contraction (at concentrations as low as 10 nM), reduced the time of tension relaxation, and increased the rate of tension development in isolated electrically stimulated (at a rate of 60 beats per minute) human right atrial preparations (HAPs, obtained during open-heart surgery) when administered alone. The potency and efficacy of tegaserod to raise the force of contraction were enhanced in the presence of cilostamide, a phosphodiesterase III inhibitor. The positive inotropic effect of tegaserod in HAPs was found to be attenuated by the 5-HT4 serotonin receptor antagonist GR 125487 (0.1 μM). The efficacy of tegaserod (10 μM) in raising the force of contraction in HAPs was less pronounced than that of serotonin (10 μM) or isoprenaline (1 μM). Tegaserod shifted the concentration–response curve of the force of contraction to serotonin to the right in HAPs, indicating that it is a partial agonist at 5-HT4 serotonin receptors in this model. We propose that the mechanism of action of tegaserod in HAPs involves cAMP-dependent phosphorylation of cardiac regulatory proteins.

The Experimental TASK-1 Potassium Channel Inhibitor A293 Can Be Employed for Rhythm Control of Persistent Atrial Fibrillation in a Translational Large Animal Model

Abstract:Background Upregulation of the two-pore-domain potassium channel TASK-1 (hK 2 P 3.1) was recently described in patients suffering from atrial fibrillation (AF) and resulted in shortening of the atrial action potential. In the human heart, TASK-1 channels facilitate repolarization and are specifically expressed in the atria. In the present study, we tested the antiarrhythmic effects of the experimental ion channel inhibitor A293 that is highly affine for TASK-1 in a porcine large animal model of persistent AF. Methods Persistent AF was induced in German landrace pigs by right atrial burst stimulation via implanted pacemakers using a biofeedback algorithm over 14 days. Electrophysiological and echocardiographic investigations were performed before and after the pharmacological treatment period. A293 was intravenously administered once per day. After a treatment period of 14 days, atrial cardiomyocytes were isolated for patch clamp measurements of currents and atrial action potentials. Hemodynamic consequences of TASK-1 inhibition were measured upon acute A293 treatment. Results In animals with persistent AF, the A293 treatment significantly reduced the AF burden (6.5% vs. 95%; P < 0.001). Intracardiac electrophysiological investigations showed that the atrial effective refractory period was prolonged in A293 treated study animals, whereas, the QRS width, QT interval, and ventricular effective refractory periods remained unchanged. A293 treatment reduced the upregulation of the TASK-1 current as well as the shortening of the action potential duration caused by AF. No central nervous side effects were observed. A mild but significant increase in pulmonary artery pressure was observed upon acute TASK-1 inhibition. Conclusion Pharmacological inhibition of atrial TASK-1 currents exerts in vivo antiarrhythmic effects that can be employed for rhythm control in a porcine model of persistent AF. Care has to be taken as TASK-1 inhibition may increase pulmonary artery pressure levels.

Abstract:Objective: To study the AIDS-related socio-demographic characteristics and high risk behaviors of Burmese HIV-infected individuals, in Dehong prefecture, Yunnan province. Methods: A cross-sectional study with questionnaire interview, was conducted. Results: Features of the 489 HIV-infected individuals appearfed as: 367(75.1%) males, 407(83.2%) illiterate or with primary education, 371(75.9%) from Kachin State or Shan State, 306(62.6%) visited Dehong twice or more per year, 238(48.7%) entered Dehong illegally, 339(69.3%) came to the city looking for jobs or working on cargo transportation, 309(63.2%) of them living in working place/rented house/own house, and 313(64.0%) with the length of stay shorter than six months. Only 36(7.4%) of the interviewees were aware of their HIV sero-positive status, with 23.3% of them (113/485) reported having had unprotected sex experiences in Dehong. Results from the multivariate logistic regression model analysis showed that HIV-infected individuals with the following characteristics: having spouses/regular sexual partners (OR=12.98, 95%CI: 6.53-25.83, P<0.001), living in working place (OR=2.88, 95% CI: 1.09-7.61, P=0.033), living in rented houses (OR=5.36, 95% CI: 1.94-14.84, P=0.001). Those living in their own houses (OR=5.14, 95% CI: 1.36-19.35, P=0.016) were more likely to engage in unprotected sex. 32.0% (156/487) of the interviewees clained that they ever used drugs in Dehong, with 9.7% (47/487) of them sharing syringe. Data also showed that HIV-infected individuals who had the following feartures as: being male (OR=15.61, 95% CI: 6.47-37.69, P<0.001), illiterate (OR=2.89, 95% CI: 1.44-5.84, P=0.003), coming from Kachin State (OR=3.01, 95% CI: 1.50-6.04, P=0.002) or Shan State (OR=5.30, 95% CI: 2.66-10.58, P<0.001), illegal visiters (OR=1.93, 95% CI: 1.18-3.14, P=0.009), living with friends/relatives (OR=5.78, 95% CI: 2.01-16.61, P=0.001), living in working place (OR=6.67, 95% CI: 3.06-14.53, P<0.001), or in rented houses (OR=11.84, 95% CI: 4.76-29.47, P<0.001). The ones who were living in their own houses (OR=6.77, 95%CI: 1.84-24.85, P=0.004) were more likely to engage in drug use in Dehong. Conclusion: Results from our study showed that the Burmese HIV-infected citizens in Dehong prefecture had the following characteristics: high mobility, low awareness of HIV positive status and high prevalence of AIDS-related risky behaviors. These findings called for greater efforts on HIV/AIDS prevention and management in Dehong city.
Inhibition of prejunctional parasympathetic pathways by β3-adrenoceptor agonists in the isolated pig detrusor: comparison with human detrusor studies

Abstract:Adrenergic receptors of the β3-subtype (β3-ADRs) seem to represent a new target for a more effective pharmacological treatment of overactive bladder (OAB), a wide spread urinary disorder. A promising opportunity for OAB therapy might rely on the development of selective β3-ADR agonists, but an appropriate preclinical screening, as well as investigation of their pharmacological mechanism(s), is limited by poor availability of human bladder samples and of translational animal models. In this study, we used the porcine urinary bladder as experimental tool to ascertain the functions of β3-ADRs in the control the parasympathetic motor drive. Tritiated acetylcholine ([3H]-ACh), mainly originated from neural stores, was released by electrical field stimulation (EFS) in epithelium-deprived detrusor strips from pigs bred without estrogens. EFS produced simultaneously [3H]-ACh release and smooth muscle contraction allowing to asses neural (pre-junctional) and myogenic (postjunctional) effects in the same experiment. Isoprenaline and mirabegron produced on the EFS-evoked effects a concentration-dependent inhibition antagonized by L-748,337, a high selective β3-ADR antagonist. The analysis of the resultant pharmacodynamic parameters supports the notion that in pig detrusors, as well as in previously described human detrusors, the activation of inhibitory β3-ADRs can modulate neural parasympathetic pathways. In such inhibitory control, the involvement of membrane K+ channels, mainly of the SK type, seems to play a pivotal role similarly to what previously described in humans. Therefore, the isolated porcine detrusor can provide a suitable experimental tool to study the mechanisms underlying the clinical efficacy of selective β3-ADR compounds for human use.

Abstract:Background: Atrial fibrillation (AF) is the most common clinically reported arrhythmia. It is associated with electrophysiological and structural alterations that promote the maintenance of the disease. The limited longevity of human cardiac samples has historically restricted mechanistic investigation into these remodeling processes of human atrial cardiomyocytes and has hampered antiarrhythmic-therapy development. Objective: We aim to assess if classical AF-associated remodeling events can be elicited in atrial engineered human myocardium (EHM) constructed from atrial subtype-specific induced pluripotent stem cell derived cardiomyocytes (iPSC-CM). Methods: To allow for long-term optical tachypacing at 3 Hz (TP), EHM were generated from cells expressing the light-gated ion-channel f-Chrimson (CRISPR-Cas9 technique). 24-hour or 7 day TP was applied to EHMs. Action potentials (AP) were acquired using the sharp microelectrode impaling technique. Results: 24-hour TP of atrial EHMs resulted in action potential shortening (APD 90 : 215.7±4.9 vs. 243.7±7.3, P <0.01) compared to EHMs paced at 1 Hz. In addition, TP attenuated the response to the M-receptor agonist carbachol. Resting membrane potential was unaffected. In order to more closely simulate persistent AF (clinically diagnosed after 7 consecutive days of AF events), 7 day TP was applied to the EHMs. Similar to 24 hour TP, this produced AP shortening and a blunted response to M-receptor agonists. Interestingly, this also led to hyperpolarization of the resting membrane potential (RMP: -75.1±0.8 vs. -72.5±0.8, P <0.05) compared to EHMs paced at 1 Hz. Conclusion: Classical characteristics of AF-associated remodeling were elicited through TP of EHM. This unique optogenetic technique represents the first in vitro model to investigate the time-dependence of cellular AF-associated remodeling in the long-term. This offers immense potential for drug development studies and personalized approaches for antiarrhythmic therapy.

Abstract:Recent improvements in MR technology have enabled the development of volumetric three-dimensional (3D) imaging methods for fat-suppressed T1-weighted images of the entire upper abdomen with pixel sizes of approximately 2 mm in all dimensions and with acquisition times of less than 25 seconds for breath-hold imaging. When performed with a timing scheme, dynamic contrast-enhanced volumetric imaging of the liver can be performed with selective imaging during the arterial phase and portal venous phase of enhancement. The volumetric data sets can be reconstructed in any oblique plane, enabling improved detection, localization, and characterization of small liver lesions. The combination of high-resolution isotropic pixels and accurate timing also permits angiographic reconstructions of the 3D images, producing MR angiography and venography that can be useful in therapeutic planning, such as for catheter-based interventions or surgical resections or transplantation. Additionally, with use of a hepatobiliary contrast agent such as mangafodipir, T1-weighted volumetric MR cholangiography can be performed in patients with nonobstructed systems for depiction of intrahepatic biliary anatomy.

Abstract:The present study was carried out to determine the role of 5-HT(1A) receptors in the generation and modulation of basic respiratory rhythm. Neonatal (aged 0-3 d) Sprague-Dawley rats of either sex were used. The medulla oblongata slice was prepared and the surgical procedure was performed in the modified Kreb's solution (MKS) with continuous carbogen (95% O2 and 5% CO2), and ended in 3 min. In cold MKS, a 600-700 microm single transverse slice was cut, which was rostral to the edge of area postrema and retained the hypoglossal nerve roots and some parts of the ventral respiratory group. The preparation was quickly transferred to a recording chamber and continuously perfused with carbogen-saturated MKS at a rate of 4-6 mL/min at 27-29 degrees C. Glass adsorb-electrodes containing Ag-AgCl needle were attached to the ventral roots of the hypoglossal nerve. Respiratory rhythmical discharge activity (RRDA) of the rootlets of hypoglossal nerve was recorded. Ten medulla oblongata slice preparations were divided into two groups. In group I, 5-HT(1A) receptor specific agonist (+/-)-8-hydroxy-2-(di-N-propylamino)tetralin hydrobromide (8-OHDPAT, 20 micromol/L) was added into the perfusion solution for 10 min first, after washing out, the 5-HT(1A) antagonist [4-iodo-N-[2-[4-methoxyphenyl]-1-piperazinyl]ethyl]-N-2-pyridynyl-benzamide hydrochloride] (PMPPI, 10 micromol/L) was applied to the perfusion solution for 10 min. In group II, after application of 8-OHDPAT for 10 min, additional PMPPI was added into the perfusion solution for 10 min. The discharges of the rootlets of hypoglossal nerve were recorded. Signals were amplified and band-pass filtered (100-3.3 kHz). Data were sampled (1-10 kHz) and stored in the computer via BL-420 biological signal processing system. Our results showed that 8-OHDPAT increased the respiratory cycle (RC) and expiratory time (TE) as well as reduced the integral amplitude (IA), but the changes of the inspiratory time (TI) were not statistically significant. PMPPI induced a significant decrease in RC, TE and TI, but the changes of IA were not statistically significant. The effect of 8-OHDPAT on the respiratory rhythm was partially reversed by additional application of PMPPI. Taken together with previous results, 5-HT(1A) receptors may play an important role in the modulation of RRDA in the medulla oblongata slice preparation of neonatal rats.

Abstract:Interactions between the polysaccharide chitosan and negatively charged phospholipid liposomes were studied as a function of compositional and environmental conditions. Using isothermal titration calorimetry, different levels of deprotonation of chitosan in acidic solutions were attained with titration of the fully protonated polymer at pH 4.48 into solutions with increasing pH. The process was found to be highly endothermic. We then examined the interaction of the polymer with vesicles in solutions of different pH. Even when partially deprotonated, the chitosan chains retain their affinity to the negatively charged liposomes. However, the stronger adsorption results in lower organization of the chains over the membrane.
Contractile Effects of Amphetamine, Pseudoephedrine, Nor-pseudoephedrine (Cathine), and Cathinone on Atrial Preparations of Mice and Humans

Abstract:Amphetamine derivatives are used worldwide legally or illegally and intoxications may be accompanied by cardiac arrhythmias. Here, we tested contractile effects of cumulative applied (±)-amphetamine, pseudoephedrine, nor-pseudoephedrine (cathine), and cathinone in electrically stimulated (1 Hz) human right atrial preparations (HAP) and mouse left atrial preparations and in spontaneously beating mouse right atrial preparations. In mouse atrial preparations, amphetamine increased force of contraction and beating rate in a concentration- and time-dependent manner, starting at 1 μM in left atrial preparations to 157.1% ± 3.0% and right atrial preparations to 146.6% ± 9.8% at 10 μM, respectively [mean ± standard error of the mean (SEM); n = 5; P < 0.05]. Pseudoephedrine, cathine, or cathinone alone were ineffective in mouse atrial preparations but after pre-incubation with the phosphodiesterase IV inhibitor rolipram (0.1 μM), a positive inotropic effect was noted (mean ± SEM: pseudoephedrine: 112.3% ± 9.8%; cathine: 109.0% ± 4.3%; cathinone: 138.3% ± 21.2%). The effects of all drugs were greatly attenuated by 10 μM cocaine or 10 μM propranolol treatments. However, In HAP, not only amphetamine (to a mean ± SEM of 208% ± 32%) but also pseudoephedrine (to a mean ± SEM of 287% ± 60%), cathine (to a mean ± SEM of 234% ± 52%), and cathinone (to a mean ± SEM of 217% ± 65%) increased force of contraction without the need of phosphodiesterase inhibition. The contractile effects in HAP were attenuated by 10 μM cocaine and antagonized by 10 μM propranolol. We conclude that amphetamine, pseudoephedrine, cathine, and cathinone act probably via release of noradrenaline from cardiac stores as indirect sympathomimetic agents in mouse and more pronounced in human atrial preparations.

Abstract:Aims: We have recently identified a functional pool of intracellular β 1 adrenergic receptors (β 1 ARs) crucial for cardiac function. However, the systematic signaling network of intracellular β 1 ARs remains elusive. Here, we aim to uncover the integrative control of cardiac function, compartmentalized β 1 AR signaling, and potential interface between two critical modulators of catecholamine homeostasis, MAO-A and OCT3. Methods and Results: Cardiac deletion of MAO-A (MAO-A-CKO) elevated myocardium catecholamines, cAMP levels and cardiac ejection fraction by disrupting intracellular catecholamine degradation. Deletion and pharmacological inhibition of MAO-A both enhanced cardiac adrenergic response, with elevated ejection fraction and heart rate. Inhibiting OCT3 by corticosterone prevented the impacts of MAO-A inhibition on cardiac function. Using subcellular anchored Förster resonance energy transfer (FRET) biosensors, we then determined the regulation of compartmentalized β 1 AR/PKA signaling at the sarcoplasmic reticulum (SR) and plasma membrane (PM) microdomains. We also examined local PKA-substrates phosphorylation and myocyte excitation-contraction coupling. MAO-A deletion and inhibitor (MAOi) enhanced local PKA activity at the SR, phosphorylation of phospholamban, Ca 2+ cycling, and contractile response. Whereas overexpression of MAO-A impairs SR-localized PKA activity and phospholamban phosphorylation. In contrast, these modifying of MAO-A did not affect PKA activity at PM and L-type Ca 2+ channel phosphorylation. Inhibiting OCT3 blocked blocking catecholamine transport and diminished the efficacy of MAO-A inhibitor (MAO-A) and MAO-A-CKO to regulate SR-localized β 1 AR signaling. In parallel, OCT3 inhibition and deletion prevented the MAO-A-dependent enhancement of Ca 2+ cycling and cardiac contractile function. Conclusion: Together, we show that MAO-A and OCT3 act in a concert to fine-tune the intracellular catecholamines, activation of SR-localized β 1 AR-PKA signaling, and cardiac fight-or-flight response. Further, we reveal a drug contraindication between anti-inflammatory corticosterone and anti-depressant MAOi in modulating adrenergic regulation in the heart, providing novel perspectives of those drugs in cardiac implications.

Abstract:It has been reported that dipyridamole and some benzodiazepines potentiate the responses to adenosine in peripheral organs and in particular in the guinea pig isolated atria or trachea by inhibition of adenosine uptake and/or metabolism. In this study, we have examined the sensitization of guinea pig isolated trachea to relaxant responses to adenosine produced by dipyridamole, diazepam and 3 compounds chemically unrelated to benzodiazepines but which display selective agonistic activity towards the central (zolpidem and zopiclone) or peripheral (alpidem) type benzodiazpine receptors. In preparations under spontaneous tone and in the absence of adenosine, dipyridamole (10(-5) M) and diazepam (10(-5)-10(-4) M), alpidem (3 x 10(-6) M-10(-5) M) and zopiclone (10(-6)-10(-4) M) induced a relaxation of the airway smooth muscle. In addition, dazepam (10(-4) M) attenuated the phasic response to histamine (10(-5) M). Dipyridamole (10(-5) M) and diazepam (10(-4) M) respectively produced a 56.2 and 32.4-fold potentiation of adenosine relaxant effects. Alpidem (10(-6)-10(-5) M), zolpidem (10(-6)-10(-4) M) and zopiclone (10(-6)-10(-4) M) were without any significant effect on the adenosine concentration-response curves. Moreover, alpidem, zolpidem, and zopiclone did not modify the 2-chloroadenosine dose-response curves nor the diazepam induced sensitization of adenosine-induced relaxation. In conclusion, adenosine sensitization of the guinea pig isolated trachea caused by diazepam might involve a peripheral benzodiazpine receptor subtype coupled to a nucleoside transporter system which is different from those recognized by compounds derived from the imidazopyridine series.

The role of 5-hydroxytryptamine in the control of pulmonary vascular tone in a rabbit model of pulmonary hypertension secondary to left ventricular dysfunction

Abstract:The role of 5-hydroxytryptamine (5-HT) in a rabbit model of pulmonary hypertension (PHT) secondary to left ventricular dysfunction was investigated. Following pulmonary artery catheterisation under anaesthesia, 5-HT (1-400 microg kg(-1) i.v.) was administered before and after either 5-HT(2A) receptor antagonism with ketanserin (0.5 mg kg(-1)) or infusion of the nitric oxide synthase inhibitor, l-NAME (30 micromol min(-1)). Eight week coronary artery ligated rabbits demonstrate increased mean pulmonary arterial pressure (PAP) compared to controls (17.0+/-0.5 versus 12.0+/-0.5 mmHg, P<0.001) accompanied by right ventricular hypertrophy. 5-HT alone produced a greater pulmonary pressor response in rabbits with PHT (increase of 7.5+/-1.2, n=12 c.f. 3.5+/-0.4 mmHg in shams, n=12, P<0.01). Ketanserin had no effect on basal PAP in either PHT or control rabbits but inhibited the response to 5-HT in both groups. The response to 5-HT following l-NAME was increased in both groups and was greater in rabbits with PHT (an increase of 20.1+/-2.9, n=6 c.f. 11.4+/-1.8 mmHg, n=6, P<0.05). These results suggest that the difference shown in the in vivo pulmonary response to exogenous 5-HT is mediated largely through 5-HT(2A) receptors in this model. However, activity of endogenous 5-HT at the 5-HT(2A) receptors is not responsible for maintaining the raised basal PAP through vasoconstriction in PHT rabbits once PHT has developed.

Abstract:1. alpha(1)-Adrenoceptors are known to play an important role in vasoconstriction in response to adrenergic stimulation. However, the functional importance of alpha(1)-adrenoceptor subtypes at the epicardial coronary artery remains unclear. We examined alpha(1)-adrenoceptor subtypes by comparing functional affinities for alpha-adrenoceptor antagonists on noradrenaline (NA)-induced vasoconstriction in porcine denuded right coronary arteries.2. Noradrenaline induced a dose-dependent vasoconstriction in incubated vessel rings. Prazosin and phentolamine were potent and competitive antagonists for NA-induced contraction (pA(2) 10.27 and 9.03, respectively). In contrast, the selective alpha(2)-adrenoceptor antagonist yohimbine had a low affinity (pA(2) 6.13). Two selective alpha(1A)-adrenoceptor antagonists, WB 4101 and 5-methyl urapidil, were potent and competitive antagonists of alpha 1-adrenoceptor-induced contraction (pA(2) 10.67 and 8.90, respectively) and the selective alpha(1D)-adrenoceptor antagonist BMY 7378 had a low affinity (pA(2) 6.06), Noradrenaline-induced contraction was insensitive to the alkylating effects of chlorethylclonidine, These observations indicate that the vasoconstriction is predominantly mediated by the alpha(1A)-adrenoceptor subtype, This was also supported by a good correlation between pA(2) values from the present study and reported binding affinities (pK(i)) of various alpha-adrenoceptor antagonists with cloned human alpha(1A)-adrenoceptors (r = 0.98), but not for alpha(1B)- or alpha(1D)-adrenoceptor subtypes (r = 0.77 and 0.41, respectively).3. Our results indicate that the alpha(1A)-adrenoceptor is the main functional receptor subtype in porcine denuded coronary arteries.

Abstract:Background More than 50% of patients with atrial fibrillation (AF) suffer from sleep disordered breathing (SDB). Obstructive respiratory events contribute to a transient vagally-mediated atrial arrhythmogenic substrate, which is resistant to most available antiarrhythmic drugs. Objective To study the effect of pharmacological inhibition of the G-protein-gated acetylcholine-regulated potassium current ( I K,ACh ) with and without acute autonomic nervous system activation by nicotine in a pig model for obstructive respiratory events. Methods In pigs (n=21) SDB was simulated by applying an intermittent negative upper airway pressure (INAP). AF-inducibility and atrial effective refractory periods (aERP) were determined before and during INAP by a S1S2 atrial pacing-protocol. Pigs were randomised into three groups. Group 1: vehicle (n=4); Group 2: XAF-1407 ( I K,ACh -inhibitor) (n=7); Group 3: Nicotine followed by XAF-1407 (n=10). Results In Group 1, INAP shortened the aERP (ΔaERP: -42.6 ms (p=0.004) and transiently increased AF-inducibility from 0 to 31%. In Group 2, XAF-1407 prolonged the aERP by 25.2 ms (p=0.005) during normal breathing and prevented INAP-induced aERP-shortening (ΔaERP: -3.6 ms, p=0.3) and AF-inducibility. In Group 3, INAP transiently shortened the aERP during nicotine perfusion (ΔaERP: -33.6 ms, p=0.004) and increased AF-inducibility up to 61%, which were both prevented by XAF-1407. Conclusion Simulated obstructive respiratory events transiently shorten the aERP and increase AF-inducibility, which can be prevented by the I K,ACh -inhibitor XAF-1407. XAF-1407 also prevents these arrhythmogenic changes induced by obstructive respiratory events during nicotine perfusion. Whether I K,ACh -channels represent a target for SDB-related AF in humans warrants further study.
