Abstract:In this study, we have biochemically characterized the high-stearic sunflower (Helianthus annuus L.) mutant CAS-14. This mutant displays an abnormal fatty acid composition along the length of the seed when grown at high temperatures. Thus, at the expense of oleate and linoleate, CAS-14 seeds present an increasing axial gradient of stearic acid from the embryo to the distal extreme of the seed. The accumulation of oil in this mutant was initially characterized by analysing the incorporation of radiolabelled acetate, a fatty acid synthetic precursor, into the developing seed tissues in vivo. These experiments indicated that the mutant phenotype was associated with a decrease in the soluble stearoyl-acyl carrier protein desaturase (SAD) activity, as later confirmed when assessing this activity in cell-free extracts from developing sunflower kernels. Furthermore, SAD enzyme gene transcription was also examined in this tissue, identifying the coordinated decrease in the transcription of the sad17 and sad6 genes as underlying the decrease in enzyme activity. On the basis of these results and those previously obtained on the inheritance of the CAS-14 trait, we discuss the possible regulatory mechanisms acting on plant soluble desaturases.
Temperature-related non-homogeneous fatty acid desaturation in sunflower (Helianthus annuus L.) seeds

Abstract:The fatty acid compositions of half-seeds and whole seeds of the temperature-dependent high-stearic-acid sunflower (Helianthus annuus L.) mutant CAS-14 were unexpectedly different. We found that there is a longitudinal gradient starting from the embryo up to the end of the cotyledon. The stearic acid content varied from 9.7 to 34.6% in seeds produced in a growth chamber (39/24 degrees C; day/night), and from 14.0 to 34.4% in seeds produced in the field during the summer season (35-40 degrees C in daylight and 20-25 degrees C at night). The gradient occurs throughout seed formation, and is due to a spatial and non-temporal regulation of stearic acid desaturation. A similar temperature-regulated behaviour, but for oleic and linoleic acid contents, was found in normal sunflower seeds. Since the deposition of oil bodies was homogeneous during seed formation, seeds showed the gradient throughout their development. This non-homogeneous distribution must be due to differences in the enzymatic pathway of de-novo fatty acid desaturation along the seed, resembling a morphogen gradient. Other high-stearic-acid mutant lines, such as CAS-3, did not show any gradient. This is the first time that a gradient and an inheritable maternal control of the fatty acid composition have been found in oilseeds.
Temperature effect on a high stearic acid sunflower mutant

Abstract:Vegetable oil with elevated saturated fatty acid content may be useful for producing solid fat without hydrogenation or transesterification. Under the nutritional point of view stearic acid is preferred to other saturated fatty acids because of its neutral effect on serum cholesterol lipoproteins. Selection of a very high stearic acid sunflower (Helianthus annuus L.) line (CAS-14), with up to a 37.3% of stearic acid in the seed oil, and the relationship between the expression of this character and the growth temperature are presented. The mutant was selected from the M(2) progeny of 3000 mutagenized seeds (4 mM sodium azide mutagenesis treatment) by analysing the fatty acid composition of half-seed by gas liquid chromatography. In order to genetically fix the mutant character, plants were grown at high day/night temperatures during seed formation. We found that temperatures higher than 30/20 degrees C are required for good expression of the phenotype, the maximum stearic acid content being obtained at 39/24 degrees C. This behaviour is totally opposed to that observed in normal and previously isolated high-stearic acid sunflower lines that contain more stearic acid at low temperature. Thus, a new type of temperature regulation on the stearate desaturation must occur. This line is the sunflower mutant with the highest stearic acid content reported so far.
Molecular analysis of the high stearic acid content in sunflower mutant CAS-14

Abstract:Increasing the stearic acid content to improve sunflower (Helianthus annuus L.) oil quality is a desirable breeding objective for food-processing applications. CAS-14 is a sunflower mutant line with a high stearic acid content in its seed oil (>35% vs. <6% in currently grown sunflower hybrids), which is controlled by the Es3 gene. However, the expression of the high stearic acid character in CAS-14 is strongly influenced by temperature during seed maturation and it is not uniform along the seed. The objectives of this study were (1) to identify PCR-based molecular markers linked to the Es3 gene from CAS-14, (2) to map this gene on the sunflower genetic map, and (3) to characterize the interaction between CAS-14 and CAS-3, a sunflower high stearic acid (about 26%) mutant line with the Es1 and Es2 genes determining this trait. Two F2 mapping populations were developed from crosses between CAS-14 and P21, a nuclear male sterile line with the Ms11 gene controlling this character, and between CAS-14 and CAS-3. One hundred and thirty-three individuals from P21xCAS-14, and 164 individuals from CAS-3xCAS-14 were phenotyped in F2 and F3 seed generations for fatty acid composition using gas-liquid chromatography, and they were then genotyped with microsatellite [simple sequence repeat (SSR)] and insertion-deletion (INDEL) markers. Bulk segregant analysis in the P21xCAS-14 population identified two markers on LG 8 putatively linked to Es3. A large linkage group was identified using additional markers mapping to LG 8. Es3 mapped to the distal half of LG 8 and was flanked by the SSR markers ORS243 and ORS1161 at genetic distances of 0.5, and 3.9 cM, respectively. The Ms11 gene was also mapped to LG 8 and genetic distance between this gene and Es3 was found to be 7.4 cM. In the CAS-3xCAS-14 population, two QTLs were identified on LG 1 and LG 8, which underlie the Es1 gene from CAS-3 and the Es3 gene from CAS-14, respectively. A significant epistatic interaction between these two QTLs was found. Results from this study provided a basis for determining CAS-14 efficient breeding strategies.

Abstract:The extraction and refinement of rosa laevigata's seed oil investigates the physicochemical properties of seed oil.The ingredients and contents of fatty acids in the seed oil are determined with GC-MS.The result shows that the oil yield of rosa laevigata's seed oil is 5.24%;the acid value is 0.36 mgKOH / g;the peroxide value is 46.10 mmol / kg;the iodine value is 160.90 gI2/100 g;and the saponification value is 193.76 mgKOH / g.Five fatty acids are identified in the rasa laevigata's seed oil.The content of unsaturated fatty acid is 86.10% which includes 43.99% linoleic acid and 42.11% α-linolenic acid.But the saturated fatty acid is 13.90% which is composed of palmitic acid,stearic acid and arachidic acid.The seed oil has plentiful unsaturated fatty acids and it is a potential source for nutritional and healthy functions.
Extraction Optimization by Response Surface Methodology: Purification and Characterization of Phytosterol from Sugarcane (saccharum Officinarum L.) Rind.

Abstract:A green, simple, and effective method for the extraction of sugarcane lipids from sugarcane rind was investigated by response surface methodology. The optimum conditions of technological progress obtained through response surface methodology were as follows: liquid‐to‐solid ratio 7.94: 1 mL/g, extraction temperature 50°C and extraction time 5.98 h. The practical sugarcane lipids extraction yield was 6.55 ± 0.28%, which was in good consistence with the predicted extraction yield of 6.47%. The results showed that the sugarcane lipids extraction yield obtained in optimum conditions increased by 1.16∼7.28‐fold compared to the yields obtained in single‐factor experiments. After saponification and SPE steps, the nonsaponifiable fraction of sugarcane lipids was analyzed by gas chromatography with mass spectrometry and high‐performance liquid chromatography. β‐Sitosterol, stigmasterol, and campesterol were the prevailing phytosterols in the sample, while fucosterol, gramisterol, stigmast‐7‐en‐3‐ol, (3β,5α,24S)‐, stigmasta‐4,6,22‐trien‐3α‐ol, and cholest‐8(14)‐en‐3β‐ol acetate were also identified as minor steroids. Furthermore, the content of β‐sitosterol and a mixture of campesterol and stigmasterol (quantified by high‐performance liquid chromatography) was 44.18 mg/100 g dry weight and 43.20 mg stigmasterol/100 g dry weight, respectively. Our results indicate that sugarcane rind is a good source of phytosterol.
Characterization of the seed and leaf lipids of high and low linolenic acid flax genotypes

Abstract:The total seed lipids of four flax (Linum usitatissimum) genotypes, differing markedly in their acyl composition, were extracted and fractionated using column, preparative, and thin-layer chromatography. In the total lipid extract of seeds, the lower linolenate content of the cultivar Glenelg (39.1% compared to that of cv. Croxton (50.5%) was associated with a higher oleate content. Further reductions in linolenate content in the induced mutants of cv. Glenelg, M1722 (17.2%) and "Zero" (1.9%) were accompanied by equivalent increases in linoleate but only minor increases in oleate. Similar changes were observed in the major triacylglycerol fraction of the simple lipids (fatty acid esters of glycerol and sterols), but there was considerable heterogeneity for acyl composition in the minor simple lipid components, including both diacylglycerols and sterol esters, and the complex lipids (glycolipids and phospholipids). The induced mutations substantially reduced linolenate content of all lipid fractions but in no case was it eliminated. Maturation of "Zero" seed at 15/10 degrees C (compared to 24/19 degrees C) increased linoleate and decreased stearate and oleate contents in all lipid fractions. In contrast to seed lipids, the acyl composition of the leaf lipids of the mutant genotypes was the same as those of their parent.
Characterization of seed fatty acid accumulation in DELLA mutant lines of Arabidopsis

Abstract:Arabidopsis seeds accumulate considerable fatty acids (FA/s) from embryo until maturity. The environmental stimuli and the regulatory factors that determine FA accumulation, however, remain unclear. In this study, we investigated the role of DELLAs, the negative regulatory factors in the gibberellin transduction pathway, in seed oil formation in Arabidopsis. Seed morphology, weight-per-thousand seeds, oil content, and FA composition were compared among the seeds of the DELLA ga1 - 3 rga - t2 rgl2 - 1 ( triple ), ga1 - 3 rga - t2 rgl1 - 1rgl2 - 1 ( quadruple ), ga1 - 3 gai - t6 rga - t2 rgl1 - 1 rgl 2 - 1 ( penta ), and wild type (WT), Landsberg erecta (L er ). Significant differences between the WT and the penta mutant for the above traits were observed. Compared with WT, the penta seeds were characterized by a decrease of 22 % seed oil in term of μg/mg, an increment of 10 % seed oil in term of FA/seed, and an alteration of FA composition. In general, the dynamics of FA accumulation in the penta and WT developing seeds (2–16 days after anthesis, DAA) were not obviously different. However, there was higher FA accumulation in penta seeds from 6 DAA compared with WT. RNA gel blotting indicated that the fatty acid desaturase genes were expressed in seeds as well as in leaves. The different transcript levels of the genes between the penta mutant and WT unveiled a part of the reasons accounting for the differences observed. To our knowledge, this is the first report on the effect of DELLAs on the dynamic process of seed oil formation of a flowering plant.
Identification and integrated analysis of mRNAs, lncRNAs, and microRNAs of developing seeds in high oleic acid sunflower (Helianthus annuus L.)

Abstract:High unsaturated fatty acid content of sunflower seed is desirable for human-healthy high oleic acid oil production. Knowledge of lipid metabolic regulatory mechanisms is key to improving oil content and quality. This study is expected to explore regulatory mechanisms of lipid metabolism in high oleic acid seeds. First, we analyzed the oil contents and detected the fatty acid compositions in developing sunflower seeds with high oleic acid content. Next, high-throughput sequencing (HTS) of seed RNA samples from three key seed developmental stages relevant to oil content and quality was performed. Pairwise comparisons of differentially expressed mRNAs, long non-coding RNAs (lncRNAs) and microRNAs (miRNAs) yielded respective differentially expressed transcripts as follows: 52293, 120625, 95 (L7d vs L22d); 57398, 123485, 127 (L7 vs L37d); 15417, 9554, 77 (L22d vs L37d). 15652 novel mRNAs, 123888 novel lncRNAs, and 98 novel miRNAs were identified, which enriched the RNA library of sunflower. Furthermore, gene functions were predicted using GO and KEGG analyses. These prediction analysis revealed that differentially expressed ncRNAs and mRNAs were mainly included in fatty acid metabolism and oil accumulation. Moreover, 9072 pairs of competing endogenous RNA (ceRNA) relationships comprised of interacting lncRNAs, miRNAs, and mRNAs. Pathway network analysis of differentially expressed lipid metabolism-associated ceRNAs revealed several important enzymes including glycerol-3-phosphate O-acyltransferase, linoleate 9S-lipoxygenase, acyl-CoA oxidase, and others that were enriched in fatty acid synthesis and oil accumulation, which highlighted competitive interactions in lipid metabolism. This study will elucidate potential regulatory mechanisms of lipid metabolism in sunflower seeds.

Macrolipidomic Profiling of Vegetable Oils: The Analysis of Sunflower Oils with Different Oleic Acid Content

Abstract:Conventional breeding techniques and genetic modifications have made it possible to alter the composition of vegetable oils. In recent years, the field of lipidomics has rapidly evolved due to technological developments in mass spectrometry. "Macrolipidomics" is an approach dedicated to detailed characterization of the most abundant lipids of a sample and has the potential to be useful for the profiling of commercial seed oils. Seed oils are composed largely of triacylglycerols (TAG) with various fatty acyls that can result in a number of isobaric and isomeric TAG species in each sample. Comprehensive methods for fatty acyl TAG characterization are still scarce. In this chapter, we describe the steps required to process and analyze different sunflower oils with altered oleic acid content to generate quantitative data for discrete fatty acyl species of TAG molecules. We utilized a dual ultra-high-performance liquid chromatography (UHPLC) serial coupling setup and untargeted tandem mass spectrometry (MS/MS) to quantitate 23 common TAG species in three sunflower oils containing 40% (low), 60% (mid), and 85% (high) oleic acid by weight.
Functional Characterization of Lysophosphatidylcholine: Acyl-CoA Acyltransferase Genes From Sunflower (Helianthus annuus L.)

Abstract:Lysophosphatidylcholine acyltransferase (LPCAT, EC 2.3.1.23) is an evolutionarily conserved key enzyme in the Lands cycle that catalyzes acylation of lysophosphatidylcholine (LPC) to produce phosphatidylcholine (PC), the main phospholipid in cellular membranes. In this study, three LPCAT genes from sunflower were identified and the corresponding proteins characterized. These HaLPCAT genes encoded functionally active enzymes that were able to complement a deficient yeast mutant. Moreover, enzymatic assays were carried out using microsomal preparations of the yeast cells. When acyl specificities were measured in the forward reaction, these enzymes exhibited a substrate preference for unsaturated acyl-CoAs, especially for linolenoyl-CoA, while in the reverse reaction, linoleoyl or linolenoyl acyl groups were transferred from PC to acyl-CoA to a similar extent. Expression levels of LPCAT genes were studied revealing distinct tissue-specific expression patterns. In summary, this study suggests that the combined forward and reverse reactions catalyzed by sunflower LPCATs facilitate acyl-exchange between the sn-2 position of PC and the acyl-CoA pool. Sunflower LPCATs displayed different characteristics, which could point to different functionalities, favoring the enrichment of seed triacylglycerols (TAGs) with polyunsaturated fatty acid (PUFA).

Abstract:As a crucial oil crop, sunflower ( Helianthus annuus L.) has a high content of unsaturated fatty acids (FAs). The unsaturated FAs found in sunflower oil mainly include oleic acid (OA, C18:1) and linoleic acid (LOA, C18:2). OA's antioxidant activity makes sunflower oil with high health value and suitable for storage. However, molecular mechanisms underlying differences in OA accumulation between high- and low-oleate sunflower varieties in seed development remain unexplored. Here, to identify key OA synthesis-related proteins, isobaric tag for relative and absolute quantitation (iTRAQ) was performed. Ultimately, 592 differentially expressed proteins (DEPs) (233 upregulated, 359 downregulated) were identified between high-oleate ('L-1-OL-1') and low-oleate ('86–1') maintainer lines. KEGG enrichment analysis of DEPs identified biosynthesis of unsaturated FAs as the most highly enriched biological pathway. Subsequently, combined transcriptome and proteome analysis results suggested that several proteins in this pathway might influence final seed OA content, including FAD2 (Δ12 fatty acid desaturase). Notably, FAD2 expression was significantly downregulated in 'L-1-OL-1' versus '86–1' seeds, with results aligning with FAD2 mRNA expression results determined via qRT-PCR analysis. These findings and understanding of OA synthesis mechanisms could help improve oil quality through breeding and germplasm-based efforts in sunflower.

Abstract:Abstract Background The investigation of molecular mechanisms involved in lipid metabolism plays a critical role for the genetic engineering of safflower ( Carthamus tinctorius L.) to increase the oil accumulation level or to change the oil composition. Although transcript sequences are currently available for the leaves and flowers of safflower, a wide range scan of temporal transcripts at different stages of seed development has not been conducted for safflower. Results In this study, temporal transcriptome sequencing was executed at 10, 14, 18, and 22 days after flowering (DAF) to uncover the molecular networks concerned in the biosynthesis of unsaturated fatty acids (USFAs). The results revealed that the biosynthesis of fatty acids is a dominant cellular process from 10 to 14 DAF, while degradation mainly happens after 18 DAF. Significant expression changes of two genes, stearoyl-[acyl-carrier-protein] 9-desaturase gene ( SAD ) from 10 to 14 DAF and oleate desaturase ( FAD2–1 ) from 14 to 18 DAF, were detected at the transcriptomic levels, and the temporal expression patterns revealed by the transcriptomic analysis were confirmed using quantitative real-time PCR experiments. In addition, 13 candidate transcription factors (TFs) involved in regulating the expression level of the FAD2–1 gene were identified. Conclusions These results create a link between fatty acid biosynthesis and gene expression at different developmental stages of the seeds, provide insight into the underlying lipid metabolism, and meanwhile lay an important foundation for the genetic engineering of safflower varieties. We have identified novel candidate genes, including TFs, that are worthy of further exploration.

Abstract:In plants, changes in the levels of oleic acid (18:1), a major monounsaturated fatty acid (FA), results in the alteration of salicylic acid (SA)- and jasmonic acid (JA)-mediated defense responses. This is evident in the Arabidopsisssi2/fab2 mutant, which encodes a defective stearoyl-acyl carrier protein-desaturase (S-ACP-DES) and consequently accumulates high levels of stearic acid (18:0) and low levels of 18:1. In addition to SSI2, the Arabidopsis genome encodes six S-ACP-DES-like enzymes, the native expression levels of which are unable to compensate for a loss-of-function mutation in ssi2. The presence of low levels of 18:1 in the fab2 null mutant indicates that one or more S-ACP-DES isozymes contribute to the 18:1 pool. Biochemical assays show that in addition to SSI2, four other isozymes are capable of desaturating 18:0-ACP but with greatly reduced specific activities, which likely explains the inability of these SSI2 isozymes to substitute for a defective ssi2. Lines containing T-DNA insertions in S-ACP-DES1 and S-ACP-DES4 show that they are altered in their lipid profile but contain normal 18:1 levels. However, overexpression of the S-ACP-DES1 isoform in ssi2 plants results in restoration of 18:1 levels and thereby rescues all ssi2-associated phenotypes. Thus, high expression of a low specific activity S-ACP-DES is required to compensate for a mutation in ssi2. Transcript level of S-ACP-DES isoforms is reduced in high 18:1-containing plants. Enzyme activities of the desaturase isoforms in a 5-fold excess of 18:1-ACP show product inhibition of up to 73%. Together these data indicate that 18:1 levels are regulated at both transcriptional and post-translational levels.

Genetic enhancement of palmitic acid accumulation in cotton seed oil through RNAi down-regulation of ghKAS2 encoding β-ketoacyl-ACP synthase II (KASII)

Abstract:Palmitic acid (C16:0) already makes up approximately 25% of the total fatty acids in the conventional cotton seed oil. However, further enhancements in palmitic acid content at the expense of the predominant unsaturated fatty acids would provide increased oxidative stability of cotton seed oil and also impart the high melting point required for making margarine, shortening and confectionary products free of trans fatty acids. Seed-specific RNAi-mediated down-regulation of β-ketoacyl-ACP synthase II (KASII) catalysing the elongation of palmitoyl-ACP to stearoyl-ACP has succeeded in dramatically increasing the C16 fatty acid content of cotton seed oil to well beyond its natural limits, reaching up to 65% of total fatty acids. The elevated C16 levels were comprised of predominantly palmitic acid (C16:0, 51%) and to a lesser extent palmitoleic acid (C16:1, 11%) and hexadecadienoic acid (C16:2, 3%), and were stably inherited. Despite of the dramatic alteration of fatty acid composition and a slight yet significant reduction in oil content in these high-palmitic (HP) lines, seed germination remained unaffected. Regiochemical analysis of triacylglycerols (TAG) showed that the increased levels of palmitic acid mainly occurred at the outer positions, while C16:1 and C16:2 were predominantly found in the sn-2 position in both TAG and phosphatidylcholine. Crossing the HP line with previously created high-oleic (HO) and high-stearic (HS) genotypes demonstrated that HP and HO traits could be achieved simultaneously; however, elevation of stearic acid was hindered in the presence of high level of palmitic acid.

Abstract:Main conclusionThe enzymes HaKCS1 and HaKCS2 are expressed in sunflower seeds and contribute to elongation of C18 fatty acids, resulting in the C20–C24 fatty acids in sunflower oil.Most plant fatty acids are produced by plastidial soluble fatty acid synthases that produce fatty acids of up to 18 carbon atoms. However, further acyl chain elongations can take place in the endoplasmic reticulum, catalysed by membrane-bound synthases that act on acyl-CoAs. The condensing enzymes of these complexes are the ketoacyl-CoA synthase (KCSs), responsible for the synthesis of very long chain fatty acids (VLCFAs) and their derivatives in plants, these including waxes and cuticle hydrocarbons, as well as fatty aldehydes. Sunflower seeds accumulate oil that contains around 2–3% of VLCFAs and studies of the fatty acid elongase activity in developing sunflower embryos indicate that two different KCS isoforms drive the synthesis of these fatty acids. Here, two cDNAs encoding distinct KCSs were amplified from RNAs extracted from developing sunflower embryos and named HaKCS1 and HaKCS2. These genes are expressed in developing seeds during the period of oil accumulation and they are clear candidates to condition sunflower oil synthesis. These two KCS cDNAs complement a yeast elongase null mutant and when expressed in yeast, they alter the host’s fatty acid profile, proving the encoded KCSs are functional. The structure of these enzymes was modelled and their contribution to the presence of VLCFAs in sunflower oil is discussed based on the results obtained.

Abstract:Acyl-CoA-binding proteins (ACBP) bind to long-chain acyl-CoA esters and phospholipids, enhancing the activity of different acyltransferases in animals and plants. Nevertheless, the role of these proteins in the synthesis of triacylglycerols (TAGs) remains unclear. Here, we cloned a cDNA encoding HaACBP1, a Class II ACBP from sunflower (Helianthus annuus), one of the world's most important oilseed crop plants. Transcriptome analysis of this gene revealed strong expression in developing seeds from 16 to 30 days after flowering. The recombinant protein (rHaACBP1) was expressed in Escherichia coli and purified to be studied by in vitro isothermal titration calorimetry and for phospholipid binding. Its high affinity for saturated palmitoyl-CoA (16:0-CoA; KD 0.11 μM) and stearoyl-CoA (18:0-CoA; KD 0.13 μM) esters suggests that rHaACBP1 could act in acyl-CoA transfer pathways that involve saturated acyl derivatives. Furthermore, rHaACBP1 also binds to both oleoyl-CoA (18:1-CoA; KD 6.4 μM) and linoleoyl-CoA (18:2-CoA; KD 21.4 μM) esters, the main acyl-CoA substrates used to synthesise the TAGs that accumulate in sunflower seeds. Interestingly, rHaACBP1 also appears to bind to different species of phosphatidylcholines (dioleoyl-PC and dilinoleoyl-PC), glycerolipids that are also involved in TAG synthesis, and while it interacts with dioleoyl-PA, this is less prominent than its binding to the PC derivative. Expression of rHaACBP in yeast alters its fatty acid composition, as well as the composition and size of the host acyl-CoA pool. These results suggest that HaACBP1 may potentially fulfil a role in the transport and trafficking of acyl-CoAs during sunflower seed development.

Abstract:Camellia oleifera is a major tree species for producing edible oil. Its seed oil is well known for the high level of oleic acids; however, little is known regarding the molecular mechanism of lipid biosynthesis in C. oleifera. Here, we measured the oil contents and fatty acid (FA) compositions at four developmental stages and investigated the global gene expression profiles through transcriptomics sequencing. We identified differentially-expressed genes (DEGs) among the developmental stages and found that the distribution of numbers of DEGs was associated with the accumulation pattern of seed oil. Gene Ontology (GO) enrichment analysis revealed some critical biological processes related to oil accumulation, including lipid metabolism and phosphatidylcholine metabolism. Furthermore, we investigated the expression patterns of lipid biosynthesis genes. We showed that most of the genes were identified with single or multiple copies, and some had correlated profiles along oil accumulation. We proposed that the higher levels of stearoyl-ACP desaturases (SADs) coupled with lower activities of fatty acid desaturase 2 (FAD2) might be responsive to the boost of oleic acid at the late stage of C. oleifera seeds' development. This work presents a comprehensive transcriptomics study of C. oleifera seeds and uncovers valuable DEGs that are associated with the seed oil accumulation.

Abstract:Long and very long chain fatty alcohols are produced from their corresponding acyl-CoAs through the activity of fatty acyl reductases (FARs). Fatty alcohols are important components of the cuticle that protects aerial plant organs, and they are metabolic intermediates in the synthesis of the wax esters in the hull of sunflower ( Helianthus annuus ) seeds. Genes encoding 4 different FARs (named Ha FAR2, Ha FAR3, Ha FAR4 and Ha FAR5) were identified using BLAST, and studies showed that four of the genes were expressed in seed hulls. In this study, the structure and location of sunflower FAR proteins were determined. They were also expressed exogenously in Saccharomyces cerevisiae to evaluate their substrate specificity based on the fatty alcohols synthesized by the transformed yeasts. Three of the four enzymes tested showed activity in yeast. Ha FAR3 produced C18, C20 and C22 saturated alcohols, whereas Ha FAR4 and Ha FAR5 produced C24 and C26 saturated alcohols. The involvement of these genes in the synthesis of sunflower seed wax esters was addressed by considering the results obtained.

Abstract:Abstract Background Cottonseed is one of the major sources of vegetable oil. Analysis of the dynamic changes of fatty acid components and the genes regulating the composition of fatty acids of cottonseed oil is of great significance for understanding the biological processes underlying biosynthesis of fatty acids and for genetic improving the oil nutritional qualities. Results In this study, we investigated the dynamic relationship of 13 fatty acid components at 12 developmental time points of cottonseed ( Gossypium hirsutum L.) and generated cottonseed transcriptome of the 12 time points. At 5–15 day post anthesis (DPA), the contents of polyunsaturated linolenic acid (C18:3n-3) and saturated stearic acid (C18:0) were higher, while linoleic acid (C18:2n-6) was mainly synthesized after 15 DPA. Using 5 DPA as a reference, 15,647 non-redundant differentially expressed genes were identified in 10–60 DPA cottonseed. Co-expression gene network analysis identified six modules containing 3275 genes significantly associated with middle-late seed developmental stages and enriched with genes related to the linoleic acid metabolic pathway and α-linolenic acid metabolism. Genes ( Gh_D03G0588 and Gh_A02G1788 ) encoding stearoyl-ACP desaturase were identified as hub genes and significantly up-regulated at 25 DPA. They seemed to play a decisive role in determining the ratio of saturated fatty acids to unsaturated fatty acids. FAD2 genes ( Gh_A13G1850 and Gh_D13G2238 ) were highly expressed at 25–50 DPA, eventually leading to the high content of C18:2n-6 in cottonseed. The content of C18:3n-3 was significantly decreased from 5 DPA (7.44%) to 25 DPA (0.11%) and correlated with the expression characteristics of Gh_A09G0848 and Gh_D09G0870 . Conclusions These results contribute to our understanding on the relationship between the accumulation pattern of fatty acid components and the expression characteristics of key genes involved in fatty acid biosynthesis during the entire period of cottonseed development.
