Abstract:This pilot study compares the effect on walking speed, in eight subjects with neuromuscular conditions, of wearing Ligaflex ankle-foot orthoses (AFO), Leafspring orthoses and shoes or with shoes alone. Range of motion, muscle strength and sensation were tested in the lower leg. Subjects underwent a standardized timed 10-m walking test five times in each of the orthoses and shoes as a measure of gait efficiency. A self-administered questionnaire was used to seek the subjects' perceptions of their functional difficulties and their opinions about the relative comfort and stability of these orthoses. Subjects had reduced ranges and strength of dorsiflexion and eversion. Some had proprioceptive deficiencies. Mean walking speed was 0.99 m/s (Leafspring) and 1.1 m/s (Ligaflex or shoes) compared to about 1.3 m/s for a normal population. Repeated measures ANOVA revealed that subjects were significantly slower in Leafspring compared to Ligaflex or to shoes. Questionnaire results rated the Leafspring as least comfortable and the Ligaflex most stable. Providing stability may be more important than assisting foot clearance when weakness is restricted to distal muscles. Further research is required to evaluate the comfort and effectiveness of orthoses to compensate for ankle instability in people with neuromuscular conditions.
Alcohol-induced changes of beta-lactoglobulin-retinol-binding stoichiometry

Abstract:It has been demonstrated using CD that ethanol induces important secondary structure changes of beta-lactoglobulin. CD spectra indicate that beta-lactoglobulin secondary structure, which is mainly composed of beta-strands, becomes mostly alpha-helical under the influence of the solvent polarity changes. The midpoint of beta-strand/alpha-helix transition in beta-lactoglobulin is observed at dielectric constant approximately 60 (35% ethanol; v/v). According to CD measurements, the ethanol-dependent secondary structure changes are reversible. The alkylation of lysines epsilon-NH2 in beta-lactoglobulin weakens the central beta-barrel structure, since the beta-strand/alpha-helix transition midpoint of alkylated beta-lactoglobulin is shifted to lower ethanol concentration (25% ethanol; v/v). beta-Lactoglobulin structural changes are triggering the dissociation of the beta-lactoglobulin-retinol complex as judged from complete quenching of its fluorescence in ethanol concentration greater than 30% (v/v). However, in 20% ethanol (v/v), beta-lactoglobulin still retains most of its native secondary structure as shown by CD and, in this condition, one beta-lactoglobulin molecule binds an additional second retinol molecule. This suggests that the highly populated species observed around 20% ethanol (v/v) might represent an intermediate state able to bind two molecules of retinol.
High-pressure effects on beta-lactoglobulin interactions with ligands studied by fluorescence

Abstract:The effects of pressure (0.1 MPa to 400 MPa) on intrinsic fluorescence of beta-lactoglobulin and on its binding of retinol and cis-parinaric acid have been studied at neutral and acid pHs. In neutral pH, fluorescence emission spectra of beta-lactoglobulin tryptophanes are characterized by an irreversible 14 nm red-shift indicating pressure-induced folding changes. The intensity of the fluorescence of retinol in beta-lactoglobulin-retinol complex is enhanced by a pressure increase up to 150 MPa. It decreases at higher pressures and disappears altogether at 300 MPa. beta-Lactoglobulin-retinol complex does not reassociate after decompression at neutral pH. At acid pH condition, the fluorescence quenching by pressure of beta-lactoglobulin tryptophans is coupled with a 2 nm spectral shift and is fully reversible demonstrating almost complete restoration of globulin folding. The evolution of retinol fluorescence in beta-lactoglobulin-retinol complex is also entirely reversible between 0.1 MPa and 400 MPa and the complex never dissociates in the studied pressure range. beta-lactoglobulin-cis-parinaric acid complexes at neutral and acid pH values dissociate irreversibly at 200 MPa and 350 MPa, respectively.
Interaction and Binding Mechanism of Cyanidin-3-o-glucoside to Ovalbumin in Varying Ph Conditions: A Spectroscopic and Molecular Docking Study

Abstract:Egg ovalbumin (OVA) as a prevalent dietary protein and has the potential to serve as a carrier for unstable bioactive compounds, however, understanding their interaction mechanism is the preliminary step. In this work, the interactions between cyanidin-3-O-glucoside (C3G) and OVA in both acidic and neutral pH environment were investigated by spectroscopic methods and molecular docking analysis. The results revealed that fluorescence quenching mechanism of OVA-C3G was predominantly static. The main acting forces were hydrogen bonds and van der Waals forces under varying pH conditions. However, the binding affinity of C3G to OVA was higher in neutral environment than that in acidic condition. The binding of C3G slightly increased the diameter of the complex, resulting in increase of α-helix, decrease of β-turn, random coil, and total main secondary structure. Moreover, the thermostability of C3G was significantly improved after OVA addition, suggesting its promising application in functional foods.
Unraveling the effects of low protein-phenol binding affinity on the structural properties of beta-lactoglobulin

Abstract:Non-covalent interactions of phenolics with proteins cannot always be readily identified, often leading to contradictory results described in the literature. This results in uncertainties as to what extent phenolics can be added to protein solutions (for example for bioactivity studies) without affecting the protein structure. Here, we clarify which tea phenolics (epigallocatechin gallate (EGCG), epicatechin and gallic acid) interact with the whey protein β-lactoglobulin by combining various state-of-the-art-methods. STD-NMR revealed that all rings of EGCG can interact with native β-lactoglobulin, indicating multidentate binding, as confirmed by the small angle X-ray scattering experiments. For epicatechin, unspecific interactions were found only at higher protein:epicatechin molar ratios and only with 1H NMR shift perturbation and FTIR. For gallic acid, none of the methods found evidence for an interaction with β-lactoglobulin. Thus, gallic acid and epicatechin can be added to native BLG, for example as antioxidants without causing modification within wide concentration ranges.

Abstract:Inclusion complexation of all-trans-retinol, retinal and retinoic acid with β-cyclodextrin (β-CD) and heptakis(2,6-O-dimethyl)-β-cyclodextrin (DM-β-CD) were investigated by means of UV-vis spectroscopy. The association constants (Ka) obtained for vitamin A with DM-β-CD is greater than with β-CD. On the other hand, for the same host compoundKa values of retinol, retinal and retinoic acid are very close to each other.

Abstract:The binding interactions of simvastatin (SIM), pravastatin (PRA), fluvastatin (FLU), and pitavastatin (PIT) with bovine serum albumin (BSA) were investigated for determining the affinity of four statins with BSA through multiple spectroscopic and molecular docking methods. The experimental results showed that SIM, PRA, FLU, and PIT statins quenched the intrinsic fluorescence of BSA through a static quenching process and the stable stains-BSA complexes with the binding constants in the order of 10(4)M(-1) at 298K were formed through intermolecular nonbond interaction. The values of H-0, S-0 and G(0) in the binding process of SIM, PRA, FLU, and PIT with BSA were negative at the studied temperature range, suggesting that the binding process of four statins and BSA was spontaneous and the main interaction forces were van der Waals force and hydrogen-bonding interactions. Moreover, the binding of four statins with BSA was enthalpy-driven process due to |H degrees|>|TS degrees| under the studied temperature range. From the results of site marker competitive experiments and molecular docking, subdomain IIIA (site II) was the primary binding site for SIM, PRA, FLU, and PIT on BSA. The results of UV-vis absorption, synchronous fluorescence, 3D fluorescence and FT-IR spectra proved that the slight change in the conformation of BSA, while the significant changes in the conformation of SIM, PRA, FLU, and PIT drug in statin-BSA complexes, indicating that the flexibility of statin molecules plays an important role in increasing the stability of statin-BSA complexes.

Abstract:The interaction of atorvastatin with bovine serum albumin (BSA) was investigated using multi-spectroscopic methods and molecular docking technique for providing important insight into further elucidating the store and transport process of atorvastatin in the body and the mechanism of action and pharmacokinetics. The experimental results revealed that the fluorescence quenching mechanism of BSA induced atorvastatin was a combined dynamic and static quenching. The binding constant and number of binding site of atorvastatin with BSA under simulated physiological conditions (pH=7.4) were 1.41 × 10(5) M(-1) and about 1 at 310K, respectively. The values of the enthalpic change (ΔH(0)), entropic change (ΔS(0)) and Gibbs free energy (ΔG(0)) in the binding process of atorvastatin with BSA at 310K were negative, suggesting that the binding process of atorvastatin and BSA was spontaneous and the main interaction forces were van der Waals force and hydrogen bonding interaction. Moreover, atorvastatin was bound into the subdomain IIA (site I) of BSA, resulting in a slight change of the conformation of BSA.
New Perspective on the Interaction Behavior Between Riboflavin and β Lactoglobulin-β Casein Complex by Biophysical Techniques

Abstract:Riboflavin (RF) is a vitamin that only exists in plants and microorganisms and must be procured externally by humans. On the other hand, there are two major allergic factors in cow's milk, including β-lactoglobulin (βLG) and β-casein (βCN), while their allergic properties can be eliminated by binding to micronutrients. In this regard, we examined the binding process of RF to βLG and βCN in the binary and ternary systems by different spectroscopies such as zeta potential, electric conductivity, and molecular modeling. According to the result of the fluorescence spectrum regarding the interaction of RF with βLG and βCN in binary and ternary systems, an increase in RF concentration declined the fluorescence intensity of three systems and also caused the quenching of proteins. Static quenching plays a pivotal role in the formation of stable interactions. The obtained thermodynamic parameters by Van't Hoff equation ascertained the predominance of hydrogen bonds and van der Waals interaction in all the systems. Considering how the negative value of ΔH0 resulted in the negative value of ΔG0, the systems were assumed to be enthalpy driven. The outcomes of circular dichroism (CD) disclosed that the attachment of RF to the targets of systems increased their a-helix content, which particularly included the binding of RF to βLG that led to the conversion of β-sheet to α-helix content. As indicated by the results of zeta potential, the low concentration of RF contained the dominance of hydrophobic forces in the interactions, whereas the enlargement of this concentration prevailed electrostatic forces. Moreover, conductometry measurements showed an extension in the rate of ionizable groups due to the addition of RF to the systems, which may increase the probability of an interaction between RF, βCN, and βLG in binary and ternary systems. In consistency with the outcomes of molecular dynamics simulation, the data of molecular docking approved the capability of RF in forming strong and stable interactions with βCN and βLG.
Effects of Labeling Thiophilic FRET Dyes on the Stability and Dimerization Process of Β-Lactoglobulin

Abstract:The stability and dimeric state of beta-lactoglobulin (beta-lg) can be dramatically affected by labeling the thiophilic agent to Cys121, whereas the underlining mechanism of such an effect is still unclear. We label a fluorescence-resonance-energy-transfer (FRET) pair of donor (1,5-IAEDANS) and acceptor (5-IAF) dyes to Cys121 of beta-lg monomers to investigate the effect of bulky thiophilic modification on the structure and stability of beta-lg. It is found that the modification dramatically destroys the native structure of beta-lg and results in an obvious increase of the alpha-helical content, coincident with the accumulation of non-native alpha-helical intermediates during its folding process. Importantly, the dimeric state of beta-lg can still be reached whereas its dimerization rate decreases dramatically, allowing us to characterize the dimerization process using the FRET method based on a stopped-flow apparatus. Our results reveal that the dimerization process occurs before the completely folding of individual monomers, providing direct evidence on the cooperativity of folding and binding processes.
Structure-function studies on human retinol-binding protein using site-directed mutagenesis.

Abstract:Retinol-binding protein (RBP) transports vitamin A in the plasma. It consists of eight anti-parallel beta-strands (A to H) that fold to form an orthogonal barrel. The loops connecting the strands A and B, C and D, and E and F form the entrance to the binding site in the barrel. The retinol molecule is found deep inside this barrel. Apart from its specific interaction with retinol, RBP is involved in two other molecular-recognition properties, that is it binds to transthyretin (TTR), another serum protein, and to a cell-surface receptor. Using site-directed mutagenesis, specific changes were made to the loop regions of human RBP and the resultant mutant proteins were tested for their ability to bind to retinol, to TTR and to the RBP receptor. While all the variants retained their ability to bind retinol, that in which residues 92 to 98 of the loop E-F were deleted completely lost its ability to interact with TTR, but retained some binding activity for the receptor. In contrast, the double mutant in which leucine residues at positions 63 and 64 of the loop C-D were changed to arginine and serine respectively partially retained its TTR-binding ability, but completely lost its affinity for the RBP receptor. Mutation of Leu-35 of loop A-B to valine revealed no apparent effect on any of the binding activities of RBP. However, substitution of leucine for proline at position 35 markedly reduced the affinity of the protein for TTR, but showed no apparent change in its receptor-binding activity. These results demonstrate that RBP interacts with both TTR and the receptor via loops C-D and E-F. The binding sites, however, are overlapping rather than identical. RBP also appears to make an additional contact with TTR via its loop A-B. A further implication of these results is that RBP, when bound to TTR, cannot bind simultaneously to the receptor. This observation is consistent with our previously proposed mechanism for delivery of retinol to target tissues [Sivaprasadarao and Findlay (1988) Biochem. J. 255, 571-579], according to which retinol delivery involves specific binding of RBP to the cell-surface receptor, an interaction that triggers release of retinol from RBP to the bound cell rather than internalization of retinol-RBP complex.

Abstract:In this study, 71 phenolic acids and their derivatives were used to investigate the structure-affinity relationship of β-lactoglobulin binding, and the effect of this interaction on antioxidant activity. Based on a fluorescence quenching method, an improved mathematical model was adopted to calculate the binding constants, with a correction for the inner-filter effect. Hydroxylation at the 3-position increased the affinity of the phenolic acids for β-lactoglobulin, while hydroxylation at the 2- or 4-positions had a negative effect. Complete methylation of all hydroxy groups, except at the 3-position, enhanced the binding affinity. Replacing the hydroxy groups with methyl groups at the 2-position also had a positive effect. Hydrogen bonding was one of the binding forces for the interaction. The antioxidant activity of phenolic acid-β-lactoglobulin complexes was higher than that of phenolic acids alone. These findings provide an understanding of the structure-activity relationship of the interaction between β-lactoglobulin and phenolic acids.

The common protein fold of ß-lactoglobulin, retinol-binding protein and bilin-binding protein must be particularly suitable for binding of various hydrophobic ligands of small molecular mass.

Dietary Polyphenols Reduced the Allergenicity of Β-Lactoglobulin Via Non-Covalent Interactions: a Study on the Structure-Allergenicity Relationship

Abstract:Studies showed that complexation of polyphenols with milk allergens reduced their immunogenic potential. However, the relationship between structures of polyphenols and their hypoallergenic effects on milk allergens in association with physiological and conformational changes of the complexes remain unclear. In this study, polyphenols from eight botanical sources were extracted to prepare non-covalent complexes with β-lactoglobulin(β-LG), a major allergen in milk. The dominant phenolic compounds bound to β-LG with a diminished allergenicity were identified to investigate their respective role on the structural and allergenic properties of β-LG. Extracts from Vaccinium fruits and black soybeans were found to have great inhibitory effects on the IgE-and IgG-binding abilities of β-LG. Among the fourteen structure-related phenolic compounds, flavonoids and tannins with larger MWs and multi-hydroxyl substituents, notably rutin, EGCG, and ellagitannins were more potent to elicit changes on the conformational structures of β-LG to decrease the allergenicity of complexed β-LG. Correlation analysis further demonstrated that a destabilized secondary structure and protein depolymerization caused by polyphenol-binding were closely related to the allergenicity property of formed complexes. This study provides insights into the understanding of structure-allergenicity relationship of β-LG-polyphenol interactions and would benefit the development of polyphenol-fortified matrices with hypoallergenic potential.
Comparative Binding Affinities Of Flavonoid Phytochemicals With Bovine Serum Albumin

Abstract:Dietary flavonoids show beneficial effects in the prevention of chronic diseases. However, flavonoid bioavailability is poor, probably due to their interaction with serum albumins. In the current work, the binding interactions of eight related flavonoids, sharing a similar core structure, with bovine serum albumin (BSA) were investigated by fluorescence spectroscopy. The binding affinities of the flavonoids with BSA were in the order hesperetin (K-A = 5.59 x 10(5)) > quercetin (4.94 x 10(5)) > naringenin (3.04 x 10(5)) > isoquercitrin (4.66 x 10(4)) > icariin (3.60 x 10(4)) > rutin (1.65 x 10(4)) > hesperidin (2.50 x 10(3)) > naringin (8.70 x 10(2)). The associations of specific structural components of the flavonoids with their binding properties to BSA were also explored and hydrophobicity, functional group substituents, steric hindrance effects and the spatial arrangements of substituents seem to be the key factors for the affinities of flavonoids towards BSA. The results from the current work contribute to a better understanding of the transport of flavonoids in plasma and helping predict their physiological functions based on their intrinsic structures.
Different interactions driving the binding of soy proteins (7S/11S) and flavonoids (quercetin/rutin): Alterations in the conformational and functional properties of soy proteins

Abstract:The purpose of this research was to comparatively investigate the interactions between bioactive flavonoids (quercetin and rutin) and two predominant soy proteins (β-conglycinin and glycinin), and the structural and functional properties of their complexes. The binding affinities of quercetin/rutin toward 7S/11S were structure-dependent, in that rutin had a higher binding affinity than that of quercetin, and 11S exhibited higher affinity toward quercetin/rutin than that of 7S. The interactions in the 7S/11S-quercetin complexes were driven by van der Waals forces and hydrogen-bonding interactions, whereas the 7S/11S-rutin complexes exhibited hydrophobic interactions. Binding to quercetin or rutin altered the secondary structures (decrease in the α-helix and random coil contents and increase in the β-sheet content), decreased the surface hydrophobicity and thermal stability, and enhanced the antioxidant capacity of 7S and 11S. These findings provide valuable information that can facilitate the design of custom-tailored protein-flavonoid macromolecules.

Abstract:Clarification of the interaction mechanisms between proteins and bioactive components is important to develop effective carriers for encapsulation and protection of bioactive components. Bovine serum albumin (BSA), a globular protein in serum and milk, contains multiple sites to bind a variety of low-molecular-weight molecules, forming protein-monoligand complexes. In this study, the interactions of BSA with retinol, resveratrol, and/or (-)-epigallocatechin-3-gallate (EGCG) were investigated by using fluorescence, circular dichroism, and molecular docking techniques. BSA-triligand complexes were successfully formed when added in the sequence of retinol, resveratrol, and EGCG. The stability of these bioactive components was improved in the complexes relative to free ones. The complexes provided a better protective effect on retinol and resveratrol than did BSA-monoligand complexes, in which the presence of EGCG played an important role.

Comparison of Sensitization Between Beta-Lactoglobulin and Its Hydrolysates.

Abstract:BACKGROUNDBovine beta-lactoglobulin is one of the first foreign antigens encountered by some newborn children, and it has been described as the main allergenic protein in cow's milk even when present at low concentration.OBJECTIVEEnzymatic hydrolysis has been identified to be a very effective way to reduce the sensitization of beta-lg compared to other treatments. The aim of this study was to explore whether enzymatic hydrolysis could reduce the allergenicity of native bovine beta-lg.METHODSbeta-lg was hydrolyzed by trypsin. Twenty four BALB/c mice were divided into three groups and orally sensitized by native bovine beta-lg and its hydrolysates five times at weekly intervals.RESULTSDuring the sensitization period, many serious systemic anaphylactic symptoms were observed in mice sensitized by native beta-lg compared to hydrolysates of beta-lg. Mice sensitized by hydrolysates of beta-lg showed a significantly lower spleen lymphocyte proliferation level than intact beta-lg. The beta-lg-specific IgE antibody levels in serum and intestinal fluid samples induced by native beta-lg were significantly elevated. Plasma histamine levels were also evaluated and showed the same trend as IgE. Moreover, the hydrolysates of beta-lg significantly up-regulated IFN-gamma and IL-10 production and down-regulated IL-4 and IL-5 secretions by murine splenocytes.CONCLUSIONThese results suggested that enzymatic hydrolysis could partly reduce the allergenicity of beta-lg.

Abstract:The interactions of beta-lactoglobulin (BLG) with anionic surfactant sodium decylsulfonate (C10SO3), cationic surfactant decyltriethylammonium bromide (C10NE), and the mixtures of cationic-anionic surfactants (C10NE-C10SO3) were investigated by circular dichroism (CD) and fluorescence methods. At pH 7.0, C10NE and the C10NE-rich surfactant mixtures of C10NE-C10SO3 could form precipitates with BLG, while C10SO3, equimolar mixtures of C10NE-C10SO3, or C10SO3-rich mixtures of C10NE-C10SO3 form homogeneous solutions with BLG. CD observed that both C10NE and C10SO3 could change the BLG structure. The effects of the mixtures of C10NE-C10SO3 on BLG structure depended on the ratio of C10NE to C10SO3. The C10NE-rich or the C10SO3-rich mixtures of C10NE-C10SO3 could significantly affect BLG structure, while the equimolar mixtures of C10NE-C10SO3 exhibited weaker interaction with BLG. Fluorescence measurements showed that both C10NE and C10SO3 could induce the enhancement of fluorescence of BLG, and C10NE enhanced the BLG fluorescence more than C10SO3 did. The effect of the mixtures of C10NE-C10SO3 on the fluorescence of BLG became stronger with the increase of the molar fraction of C10NE in C10NE-C10SO3 mixtures.
Retinoid Affinity Biotinylation Reveals A Retinoid Binding Role for Rpe65

Abstract:Retinal pigment epithelial (RPE) membranes contain the full biochemical apparatus capable of processing all-trans-retinol (vitamin A) into 11-cis-retinal, the visual chromophore. As many of these proteins are integral membrane proteins and resistant to traditional methods of identification, alternate methods of identifying these proteins are sought. The approach described here involves affinity biotinylation with alkali cleavable linkers. A vitamin A containing affinity-labeling haloacetate is described which facilitates the identification of retinoid binding proteins (RBPs). Treatment of crude bovine RPE membranes with (3R)-3-[boc-lys(biotinyl)-O]-all-trans-retinol chloroacetate 1 in the low micromolar range led to the specific labeling of RPE65 and lecithin retinol acyltransferase (LRAT). Only RPE65 is labeled at 5 microM 1 at 4 degrees C. Labeled RPE65 was readily isolated by binding the labeled protein to avidin-containing beads, followed by cleavage of the protein from the beads at pH 11. Trypsin digestion of RPE65 modified by 1, followed by mass spectrometry, demonstrates that C231 and C448 are alkylated by 1. These studies validate the approach that was used, and furthermore demonstrate that RPE65, a major membrane-associated protein of the RPE, is a RBP.
