Abstract:ABSTRACT:Free radicals are the unstable electron-deficient species that reacts with different molecules to gain stability and to eliminate their unpaired condition. Antioxidant molecules neutralizes the free radicals by donating their electrons and inhibits the unwanted oxidative reactions in biological system. The imbalance between antioxidants and free radicals generated oxidative stress which leads to severe impairment of the biological systems. The purpose of the present review is to highlight the beneficial role of naturally occurring antioxidant systems in minimizing the damage and maintaining the homeostasis in the biological system. Enzymatic and non-enzymatic antioxidants are the major classes of natural antioxidants which executes diverse functions in the biological system to provide defense against the destructive accumulating effects of ROS/RNS. Superoxide dismutase (SOD) are responsible for providing first line of defense to thebiological system, by converting the superoxide anion radical (O oˉ) into hydrogen 2 peroxide (H O ) which eventually converted into water and oxygen. Non-enzymatic 2 2 antioxidants either endogenous or exogenous provides numerous crucial mechanisms to quench the ROS/RNS in the biological system. Endogenous antioxidants inhibits lipid peroxidation in the cellular membranes while the exogenous antioxidants acts as chain breakers and terminates the oxidative chain reaction. It is significant to sustain the level of exogenous antioxidants in the body through diet so that the normal biological processes can be maintained at optimal levels.

Abstract:Reactive oxygen species (ROS) and reactive nitrogen species (RNS) are well recognized for playing a dual role, since they can be either deleterious or beneficial to biological systems. An imbalance between ROS production and elimination is termed oxidative stress, a critical factor and common denominator of many chronic diseases such as cancer, cardiovascular diseases, metabolic diseases, neurological disorders (Alzheimer's and Parkinson's diseases), and other disorders. To counteract the harmful effects of ROS, organisms have evolved a complex, three-line antioxidant defense system. The first-line defense mechanism is the most efficient and involves antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx). This line of defense plays an irreplaceable role in the dismutation of superoxide radicals (O 2 •− ) and hydrogen peroxide (H 2 O 2 ). The removal of superoxide radicals by SOD prevents the formation of the much more damaging peroxynitrite ONOO − (O 2 •− + NO • → ONOO − ) and maintains the physiologically relevant level of nitric oxide (NO • ), an important molecule in neurotransmission, inflammation, and vasodilation. The second-line antioxidant defense pathway involves exogenous diet-derived small-molecule antioxidants. The third-line antioxidant defense is ensured by the repair or removal of oxidized proteins and other biomolecules by a variety of enzyme systems. This review briefly discusses the endogenous (mitochondria, NADPH, xanthine oxidase (XO), Fenton reaction) and exogenous (e.g., smoking, radiation, drugs, pollution) sources of ROS (superoxide radical, hydrogen peroxide, hydroxyl radical, peroxyl radical, hypochlorous acid, peroxynitrite). Attention has been given to the first-line antioxidant defense system provided by SOD, CAT, and GPx. The chemical and molecular mechanisms of antioxidant enzymes, enzyme-related diseases (cancer, cardiovascular, lung, metabolic, and neurological diseases), and the role of enzymes (e.g., GPx4) in cellular processes such as ferroptosis are discussed. Potential therapeutic applications of enzyme mimics and recent progress in metal-based (copper, iron, cobalt, molybdenum, cerium) and nonmetal (carbon)-based nanomaterials with enzyme-like activities (nanozymes) are also discussed. Moreover, attention has been given to the mechanisms of action of low-molecular-weight antioxidants (vitamin C (ascorbate), vitamin E (alpha-tocopherol), carotenoids (e.g., β-carotene, lycopene, lutein), flavonoids (e.g., quercetin, anthocyanins, epicatechin), and glutathione (GSH)), the activation of transcription factors such as Nrf2, and the protection against chronic diseases. Given that there is a discrepancy between preclinical and clinical studies, approaches that may result in greater pharmacological and clinical success of low-molecular-weight antioxidant therapies are also subject to discussion.

Abstract:An imbalance between oxidants and antioxidants in favour of the oxidants, potentially leading to damage, is termed 'oxidative stress'. Oxidants are formed as a normal product of aerobic metabolism but can be produced at elevated rates under pathophysiological conditions. Antioxidant defense involves several strategies, both enzymatic and non-enzymatic. In the lipid phase, tocopherols and carotenes as well as oxy-carotenoids are of interest, as are vitamin A and ubiquinols. In the aqueous phase, there are ascorbate, glutathione and other compounds. In addition to the cytosol, the nuclear and mitochondrial matrices and extracellular fluids are protected. Overall, these low molecular mass antioxidant molecules add significantly to the defense provided by the enzymes superoxide dismutase, catalase and glutathione peroxidases.

Abstract:The definition of antioxidants, given in 1995 by Halliwell and Gutteridge, stated that an antioxidant is “any substance that, when present at low concentrations compared with that of an oxidizable substrate, significantly delays or inhibits oxidation of that substrate” [1]. In 2007, Halliwell gave a more specific definition, stating that an antioxidant is “any substance that delays, prevents or removes oxidative damage to a target molecule” [2]. Oxidation reactions produce free radicals that can start multiple chain reactions that eventually cause damage or death to the cell. Antioxidants remove these free-radical intermediates by being oxidized themselves, and inhibit other oxidation reactions, thus stopping the harmful chain reactions. Such oxidative processes are dangerous for all living cells, especially those in proximity to sites where active oxygen is released by photosynthesis. Spontaneous oxidation causes food rancidity and spoilage of medicines. Furthermore, oxidative stress is an important part of many human diseases that can occur, inter alia, due to a lack of appropriate nutrition and exercise, air pollution, smoking, and more, leading to lethal diseases, such as cancer. Therefore, it is imperative to include antioxidants in our diets. Due to the fact that synthetically produced antioxidants are currently used in the food and pharmaceutical industries in order to prolong product shelf life, there is currently a strong trend to search for large, available, and efficient natural sources of antioxidants to replace the synthetic ones, thus minimizing damage to our cells.

Abstract:Since the last few years, the growing interest in the use of natural and synthetic antioxidants as functional food ingredients and dietary supplements, is observed. The imbalance between the number of antioxidants and free radicals is the cause of oxidative damages of proteins, lipids, and DNA. The aim of the study was the review of recent developments in antioxidants. One of the crucial issues in food technology, medicine, and biotechnology is the excess free radicals reduction to obtain healthy food. The major problem is receiving more effective antioxidants. The study aimed to analyze the properties of efficient antioxidants and a better understanding of the molecular mechanism of antioxidant processes. Our researches and sparing literature data prove that the ligand antioxidant properties complexed by selected metals may significantly affect the free radical neutralization. According to our preliminary observation, this efficiency is improved mainly by the metals of high ion potential, e.g., Fe(III), Cr(III), Ln(III), Y(III). The complexes of delocalized electronic charge are better antioxidants. Experimental literature results of antioxidant assays, such as diphenylpicrylhydrazyl (DPPH) and ferric reducing activity power assay (FRAP), were compared to thermodynamic parameters obtained with computational methods. The mechanisms of free radicals creation were described based on the experimental literature data. Changes in HOMO energy distribution in phenolic acids with an increasing number of hydroxyl groups were observed. The antioxidant properties of flavonoids are strongly dependent on the hydroxyl group position and the catechol moiety. The number of methoxy groups in the phenolic acid molecules influences antioxidant activity. The use of synchrotron techniques in the antioxidants electronic structure analysis was proposed.

Scavenging of reactive oxygen species as the mechanism of drug action

Abstract:Reactive oxygen species (ROS) are generated when oxygen is supplied in excess and/or its reduction is insufficient. The best explored ROS are superoxide anions, hydroxyl radicals and hydrogen peroxide. The first two are free radicals. ROS are harmful for the living cells and are implicated in a variety of pathological processes and diseases. Drugs used in the treatment of these states are either stimulators of endogenous defense mechanisms against ROS or inhibitors of ROS formation. Six groups of anti-ROS substances have been described in this paper. 1) Antioxidant substances used in substitutive therapy such as enzymes (e.g. superoxide dismutase), substances containing thiol groups and vitamins (A, E, P, C). 2) Chelating agents (e.g. desferoxamine), which lower the level of prooxidative transition metal ions. 3) Inhibitors of superoxide ions generation by stimulated cells or xanthine oxidase. Such mechanism of action was described for xanthine oxidase inhibitor-allopurinol. 4) Superoxide scavengers. Many known drugs were investigated for this activity, but the best documentation was presented for flavonoids. 5) Substances which eliminate hydrogen peroxide, mainly glutathione and its precursors. 6) Scavengers of hydroxyl radicals. Studies of the above activity were conducted mainly using an unspecific method--estimation of malondialdehyde generated during the action of hydroxyl radicals on lipids or on desoxyribose. Inhibition of malondialdehyde formation was described for many drugs of plant and synthetic origin.

Abstract:Free radicals and oxidants play a dual role as both toxic and beneficial compounds, since they can be either harmful or helpful to the body. They are produced either from normal cell metabolisms in situ or from external sources (pollution, cigarette smoke, radiation, medication). When an overload of free radicals cannot gradually be destroyed, their accumulation in the body generates a phenomenon called oxidative stress. This process plays a major part in the development of chronic and degenerative illness such as cancer, autoimmune disorders, aging, cataract, rheumatoid arthritis, cardiovascular and neurodegenerative diseases. The human body has several mechanisms to counteract oxidative stress by producing antioxidants, which are either naturally produced in situ, or externally supplied through foods and/or supplements. This mini-review deals with the taxonomy, the mechanisms of formation and catabolism of the free radicals, it examines their beneficial and deleterious effects on cellular activities, it highlights the potential role of the antioxidants in preventing and repairing damages caused by oxidative stress, and it discusses the antioxidant supplementation in health maintenance.

Abstract:Oxidative stress is a disturbance of the balance between the production of reactive oxygen species (ROS) and antioxidants. Oxidative stress is caused by the presence of any of a number of reactive oxygen species, which the cell is unable to counterbalance. The result is damage to one or more biomolecules including DNA, RNA, proteins and lipids. Oxidative stress has been implicated in the natural aging process as well as a variety of disease states, such as neoplastic, metabolic, neurological etc., accompanied by different complications. Risk factors of generation of oxidative stress are oxidizing species, induced by pathologies include alcohol consumption, cigarette smoking, diet, gender, geographic location specifically at high altitude and occupation. ROS are composed of superoxide, hydroxyl, peroxyl, hydroperoxyl and alkoxyl radicals, hydrogen peroxide and singlet oxygen and ozone. These compounds produced endogenous in reaction of autooxidation in respiratory chaine of bioobjects. Among exogenous sources of ROS can be listed exposure of pollutants, toxins, heavy metals, drugs with different chemical origin and effects, radiation, electromagnetic fields, alcohol, cigarette smoke, stresses, allergies, dietary factors, temperature and microscopic form of life, such as bacteria, yeasts, viruses etc. The oxidative stress in biological systems is often characterized by increase in the formation of radicals; decrease in small-molecular-weight and lipid soluble antioxidants; disturbance in cellular redox balance; oxidative damage to cellular components (biomacromolecules). The presence of oxidative stress may be tested in one of three ways: direct measurement of the ROS; measurement of the resulting damage to biomolecules; and detection of antioxidant levels. Directly measuring ROS might seem the preferred method, but many reactive oxygen species are extremely unstable and difficult to measure directly. Many markers of damage are extremely stable and therefore provide a more reliable method to measure oxidative stress. Another approach is to measure the levels of antioxidant enzymes and other redox molecules which serve to counterbalance ROS generated in the cell. At the same time, it must emphasize that oxidative stress not only has a cytotoxic effect, but also plays an important role in the modulation of messengers that regulate essential cell membrane functions, which are vital for survival. For prevention of oxidative stress cells produce or uptake antioxidants – substance significantly delays or prevents oxidation of that substrate. Antioxidants may be enzymatic and non-enzymatic in nature in which enzymatic system directly or indirectly help in defence against the ROS. Antioxidants are involved in the prevention of oxidants and ROS formation; exhibits scavenger of ROS; and repairs the oxidized molecules through sources like dietary or consecutive antioxidants. Among non-enzymatic antioxidants distinguished glutathione, α-tocopherol, ascorbic acid, beta-carotene, and uric acid; these are mostly considered to be chain-breaking antioxidants in that they interrupt the auto-catalytic spread of radical reactions. Among enzymatic antioxidants most known superoxide dismutase, catalase, glutathione-SH peroxidase. The significant correlation found between ROS, parameters of oxidative stress and pathology indicate that there is a need in finding of measures of prevention of endo- and exogenous factors provoke their generation. There is a need to continue to explore the relationship between free radicals, pathological processes and the complications of them, and to elucidate the mechanisms by which increased oxidative stress accelerates the development of complications, in an effort to expand treatment options. Improvement of complications control seems to be a beneficial factor to decrease oxidative stress. For a better investigation of oxidative stress, it would be wise to supplement the clinical research by determination of special products typical for oxidative stress that let to understand mechanism of some pathological processes more clearly.

Abstract:Annotation. Under the conditions of entry of toxic substances into the body and activation of redox processes, compounds of natural origin with antioxidant properties are widely used to correct metabolic disorders. The purpose of the work is to conduct an analysis and systematization of basic and modern scientific sources of literature on the role of the glutathione system in the development of pathologies. For the scientific review, data from the scientometric databases PubMed, ScienceDirect, Web of Science, Google Scholar, etc. were used, and the analysis of scientific textbooks and monographs was carried out. Among water-soluble antioxidants, low- and high-molecular compounds containing SH-groups are popular in medicine, mono-, di- and tricarboxylic acids and other anions. Glutathione, a thiol-containing tripeptide, is the main endogenous non-enzymatic antioxidant that exerts cytoprotective and detoxifying properties. Various epithelial cells – enterocytes, endotheliocytes, alveolar cells of the lungs, epithelium of the proximal renal tubules, absorb and use glutathione most intensively, in particular, taken orally, so the idea of using glutathione in diseases of organs that actively participate in detoxification processes: kidneys, liver, lungs, as well as under the conditions of systemic diseases accompanied by endothelial dysfunction and oxidative stress – diabetes, atherosclerosis, nephropathy. Under physiological conditions, the body maintains a dynamic balance between the content of oxidants, which stimulate the processes of free radical oxidation of biomolecules, and the activity of antioxidant systems. But under the conditions of damage to any link of the living system, increased formation of reactive oxygen species (ROS) and inhibition of antioxidant systems contributes to the activation of free radical oxidation processes, the intensity of which depends on the concentration of oxygen in the tissues and on the enzymatic and non-enzymatic systems of antioxidant protection, which prevents the destruction of cells by active forms of oxygen.

Abstract:Free radicals have acquired growing importance in the fields of biology and medicine. They are produced during many different endogenous and exogenous processes. Mitochondria are the main source of endogenous reactive oxygen species (ROS) produced at cell level. The overproduction of free radicals can damage macromolecules such as nucleic acids, proteins and lipids. This leads to tissue damage in various chronic and degenerative diseases. Antioxidants play a crucial role in the body’s defense against free radicals. This review concerns the main properties of free radicals, their sources and deleterious effects. It highlights the potential role of the dietary supplementation of antioxidants and discusses unsolved problems regarding antioxidant supplements in the prevention and therapy of diseases.

Abstract:In recent years, there has been a great deal of attention toward the field of free radical chemistry. Free radicals reactive oxygen species and reactive nitrogen species are generated by our body by various endogenous systems, exposure to different physiochemical conditions or pathological states. A balance between free radicals and antioxidants is necessary for proper physiological function. If free radicals overwhelm the body's ability to regulate them, a condition known as oxidative stress ensues. Free radicals thus adversely alter lipids, proteins, and DNA and trigger a number of human diseases. Hence application of external source of antioxidants can assist in coping this oxidative stress. Synthetic antioxidants such as butylated hydroxytoluene and butylated hydroxyanisole have recently been reported to be dangerous for human health. Thus, the search for effective, nontoxic natural compounds with antioxidative activity has been intensified in recent years. The present review provides a brief overview on oxidative stress mediated cellular damages and role of dietary antioxidants as functional foods in the management of human diseases.

Abstract:Free radicals (FRs), also known as reactive oxygen species (ROS), are usually established in the body when adequate oxygen depletion occurs. Oxidative stress and the establishment of FRs in the body are mainly caused by high metabolic activity, the need for rapid growth, inadequate flock management, exposure to viral and bacterial microorganisms, and adverse environmental conditions. Furthermore, FRs can also be produced during the activity of phagocytes when they depend on the action of ROS to kill the engulfed pathogen. FRs have very adverse effects on all cells, particularly the cells of the immune system. They are extremely erratic and reactive molecules that directly harm DNA, cellular proteins, lipids, and carbohydrates within cells. Antioxidants are substances that can eliminate and neutralize FRs within the body and free the body from the oxidative stress that occurs due to the accumulation of FRs. Many vitamins and minerals support the activity and effect of the immune system in fighting against microbes and cancer, which mostly depend on their antioxidant elements to diminish the negative impact of FRs in the body. Examples are vitamin C, vitamin E, superoxide dismutase, selenium, glycine, cofactors of glutathione peroxidase, manganese, essential oils, and phenolic compounds.

Abstract:The body encloses a complex antioxidant defence grid that relies on endogenous enzymatic and non-enzymatic antioxidants. These molecules collectively act against free radicals to resist their damaging effects to vital biomolecules and ultimately body tissues. Based on their response to general free radical invasion, they can be categorized into first, second, third and even fourth line defense antioxidants. The role and effectiveness of the first line defense antioxidants which basically include superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) is important and indispensable in the entire defense strategy of antioxidants, especially in reference to super oxide anion radical (*O2) which is perpetually generated in normal body metabolism, particularly through the mitochondrial energy production pathway (MEPP). A lot has been published concerning antioxidants and their significance in preventing oxidative stress and the attendant cellular damage, howbeit with paucity of awareness on the fundamental role of SOD, CAT and GPX. The present review tends to articulate important information on SOD, CAT and GPX as first line defense antioxidant enzymes.

Abstract:Reactive oxygen species (ROS) are produced by living cells as normal cellular metabolic byproduct. Under excessive stress conditions, cells will produce numerous ROS, and the living organisms eventually evolve series of response mechanisms to adapt to the ROS exposure as well as utilize it as the signaling molecules. ROS molecules would trigger oxidative stress in a feedback mechanism involving many biological processes, such as apoptosis, necrosis and autophagy. Growing evidences have suggested that ROS play a critical role as the signaling molecules throughout the entire cell death pathway. Overwhelming production of ROS can destroy organelles structure and bio-molecules, which lead to inflammatory response that is a known underpinning mechanism for the development of diabetes and cancer. Cytochrome P450 enzymes (CYP) are regarded as the markers of oxidative stress, can transform toxic metabolites into ROS, such as superoxide anion, hydrogen peroxide and hydroxyl radical which might cause injury of cells. Accordingly, cells have evolved a balanced system to neutralize the extra ROS, namely antioxidant systems that consist of enzymatic antioxidants such as superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidases (GPxs), thioredoxin (Trx) as well as the non-enzymatic antioxidants which collectively reduce oxidative state. Herein, we review the recent novel findings of cellular processes induced by ROS, and summarize the roles of cellular endogenous antioxidant systems as well as natural anti-oxidative compounds in several human diseases caused by ROS in order to illustrate the vital role of antioxidants in prevention against oxidative stress.

Abstract:Due to the negative impact of reactive species (including free radicals) on humans and animals, the investigations to find effective substances (antioxidants), which protect living organisms against their damaging influence are carried out throughout the world. As most widespread synthetic antioxidants are suspected of having a noxious effect on the human body, more and more attention is paid to natural antioxidant compounds found in plants (especially phenolic compounds). The aim of this paper is to present the data about antioxidant activity of polyphenolic compounds with the emphasis on the main factors having influence on their antioxidant activity: chemical structure, ability to form hydrogen bonds, capability of metal ions chelation and reduction, adduct formation, kinetic solvents effect, mechanism of antioxidant reaction, capability of antioxidant enzyme activation and reduction potential.

Abstract:Reactive oxygen species (ROS) are produced during oxidative metabolism in aerobic organisms. Under normal conditions, ROS production and elimination are in a relatively balanced state. However, under internal or external environmental stress, such as high glucose levels or UV radiation, ROS production can increase significantly, leading to oxidative stress. Excess ROS production not only damages biomolecules but is also closely associated with the pathogenesis of many diseases, such as skin photoaging, diabetes, and cancer. Antioxidant peptides (AOPs) are naturally occurring or artificially designed peptides that can reduce the levels of ROS and other pro‐oxidants, thus showing great potential in the treatment of oxidative stress‐related diseases. In this review, we discussed ROS production and its role in inducing oxidative stress‐related diseases in humans. Additionally, we discussed the sources, mechanism of action, and evaluation methods of AOPs and provided directions for future studies on AOPs.

Abstract:Cytochrome P450 enzyme system is a clan of heme oxidases that is substantial in oxidative metabolism and activation of drugs in the liver and extrahepatic sites of the body. Compounds that are metabolized through CYP450 can fabricate reactive intermediates that can deplete oxygen at molecular level and yield reactive oxygen species (ROS). Such detrimental events taking place in the body are defied and compensated by body cells via diverse defense mechanisms. Defensive mechanisms may include the induction of free radical removers, for instance, some vitamins (C and E), bilirubin, thiols, some lipophilic scavengers such as ubiquinol and enzymes, e.g., glutathione peroxidases, superoxide dismutase, or catalases to sophisticate and embellish DNA repair mechanisms. In the circumstances, when the defense is inadequate to combat these free radicals, ROS hold the charge and oxidative stress is an obvious incident to happen. The resulting oxidative stress then bows the seeds of idiosyncratic drug toxicity by damaging the cells, proteins, and DNA. Consequently, by the study of CYP-controlled modified drug metabolism and resulting in oxidative stress, plenty of malfunctions can be prevented. Ample studies have intensified the contribution of hepatic metabolism, directing toward the complex diseases. The motive of this review is to highlight the oxidative stress due to ROS that are produced during CYP-induced drug metabolism and detoxification with antioxidant therapy. The writing will also elaborate on the unexplored aspects of this field that are yet to be considered.

Abstract:The present review aims to high light on the oxidative stress, and prevention by internal antioxidants and external antioxidants by some natural products possessing antioxidant properties. Oxidative stress occurs when the balance between reactive oxygen species (ROS) formation and detoxification favors an increase in ROS levels, leading to disturbed cellular function. ROS causes damage to cellular macromolecules causing lipid peroxidation, nucleic acid, and protein alterations. Their formation is considered as a pathobiochemical mechanism involved in the initiation or progression phase of various diseases such as atherosclerosis, ischemic heart diseases, diabetes, and initiation of carcinogenesis or liver diseases. In order to maintain proper cell signaling, it is likely that a number of radical scavenging enzymes maintain a threshold level of ROS inside the cell. However, when the level of ROS exceeds this threshold, an increase in ROS production may lead to excessive signals to the cell, in addition to direct damage to key components in signaling pathways. ROS can also irreversibly damage essential macromolecules. Protein-bound thiol and non-protein-thiol are the major cytosolic low molecular weight sulfhydryl compound that acts as a cellular reducing and a protective reagent against numerous toxic substances including most inorganic pollutants, through the –SH group. Hence, thiol is often the first line of defense against oxidative stress. Flavonoids have been found to play important roles in the non-enzymatic protection against oxidative stress, especially in the case of cancer. Flavonoids have occurred widely in tea, fruit, red wine, vegetables, and cocoas. Flavonoids, including flavones, flavanone, flavonols, and isoflavones, are polyphenolic compounds which are widespread in foods and beverages, and possess a wide range of biological activities, of which anti-oxidation has been extensively explored. It can be concluded that oxidative stress causes irreversible damage in cellular macromolecules that leads to initiation of various diseases such as atherosclerosis, ischemic heart diseases, liver diseases, diabetes, and initiation of carcinogenesis. Antioxidants inhibit reactive oxygen species production and scavenging of free radicals. Therefore, the review recommends that high consumption of natural foods that are rich in antioxidants will provide more protection against toxic agents and related diseases.
