Abstract:Intracerebral hemorrhage (ICH) will be accompanied by the overload of iron and reactive oxygen species (ROS) following hematoma clearance. Although deferoxamine (DFO) has been widely utilized as a clinical first-line siderophore to remove the iron overload, the ROS-inducing damage still greatly limits the therapeutic effect of DFO. To address this issue, we designed and fabricated a series of dual-functional macromolecular nanoscavengers featuring high-density DFO units and catechol moieties. Note that the former units could effectively remove the iron overload, while the latter ones could efficiently deplete the ROS. The resulting nanoscavengers efficiently down-regulate the iron and ROS levels as well as significantly reduce the cell death in both iron-overloaded RAW 264.7 cells and the ICH mice model. This work suggests a novel clue for the ICH-ameliorated iron-depleting interventional therapeutic regimen. The Supporting Information is available free of charge at <a class="ext-link" href="/doi/10.1021/acsami.1c00491?goto=supporting-info">https://pubs.acs.org/doi/10.1021/acsami.1c00491</a>.Details of material preparation; hemocompatibility of nanoscavengers <i>in vitro</i>; synthesis scheme of poly(DFO-PEGm)n; ESI-MS and HPLC spectra of Cat–DFO, UV–vis spectra of 3,4-dihydroxyhydrocinnamic acid, DFO, Cat-DFO, P0, P1, P2, and P3; FT-IR spectra of Cat-DFO, P0, and P3; 1H NMR spectra of P1, P2, and P3; average size and zeta potential value of P3 in different media obtained by DLS; UV-Vis spectra of P3 solution with different concentrations before and after binding with Fe2+ for 12 h; UV-Vis spectra of the bottom filtrate of DFO and P3 after binding with Fe2+ for 12 h; RBC deformability of the precipitate of RBC suspension incubated with P3 and H2O for 1 h at different shear stress; PT of PPP after incubation with DFO, P1, P2, and P3 with different concentrations; cell viability of RAW 264.7 cells after incubating with DFO, P1, P2, and P3 of different concentrations for 48 h using the alamarBlue assay; flow cytometry result of RAW 264.7 cells stimulated with different concentrations of Fe2+, FAC (Fe3+) for 0.5 h using DCFH-DA as the fluorescent probe; bodyweight of normal mice, ICH mice treated with saline, DFO, and P3 nanoscavenger; and instrument for ICH mouse model construction (<a class="ext-link" href="/doi/suppl/10.1021/acsami.1c00491/suppl_file/am1c00491_si_001.pdf">PDF</a>)This article has not yet been cited by other publications.
