Abstract:Epigallocatechin-3-gallate (EGCG) has a promising therapeutic effect for ulcerative colitis (UC), but the treatment mechanism has yet to be fully elucidated. The aim of the present study was to investigate the mechanism of EGCG in the treatment of UC. Experimental colitis mouse models were prepared. The mice were randomly divided into four groups: Normal control, model (MD), 50 mg/kg/day EGCG treatment and 100 mg/kg/day EGCG treatment. The daily disease activity index (DAI) of the mice was recorded, changes in the organizational structure of the colon were observed and the spleen index (SI) was measured. In addition, levels of interleukin (IL)-6, IL-10, IL-17 and transforming growth factor (TGF)-β1 in the plasma and hypoxia-inducible factor (HIF)-1α and signal transducer and activator of transcription (STAT) 3 protein expression in colon tissues were evaluated. Compared with the MD group, the mice in the two EGCG treatment groups exhibited decreased DAIs and SIs and an attenuation in the colonic tissue erosion. EGCG could reduce the release of IL-6 and IL-17 and regulate the mouse splenic regulatory T-cell (Treg)/T helper 17 cell (Th17) ratio, while increasing the plasma levels of IL-10 and TGF-β1 and decreasing the HIF-1α and STAT3 protein expression in the colon. The experiments confirmed that EGCG treated mice with experimental colitis by inhibiting the release of IL-6 and regulating the body Treg/Th17 balance.
Quantification of neural reflex and muscular intrinsic contributions to parkinsonian rigidity

Abstract:Parkinson's disease (PD) is a progressive neurodegenerative disease characterized by rigidity, bradykinesia, resting tremor, and postural instability. Rigidity, defined as an increased resistance to passive movement of a joint, progresses faster than other motor signs in PD. Rigidity is attributable to both exaggerated neural reflex and altered muscle mechanical properties. However, little is known about the contributions of individual components to rigidity. Further, there is no evidence regarding the effects of dopaminergic medication on individual components. Objectives of this study were to quantify the contributions of neural reflexes and intrinsic muscle properties to rigidity and investigate the effects of medication on each contributing component. Joint torque and muscle activities of the wrist in 14 patients and 14 controls were measured during externally induced movements. Each subject with PD was tested in Off- and On-medication states. A system identification technique was applied to differentiate and quantify the neural reflex and intrinsic mechanical components. A mixed model of ANOVA was performed to compare the differences between the two components of rigidity for both groups, and to compare between the Off- and On-medication states for patients. The results showed that reflex and intrinsic components are comparable (p > 0.05), and both are enhanced in subjects with PD than in the controls (p < 0.05). Medication decreased the reflex component of rigidity (p < 0.01). It is concluded that both reflex and intrinsic factors are responsible for rigidity. Present findings are clinically significant as they may provide guidance in development of effective therapeutic interventions.

Abstract:Endometrial stromal sarcomas (ESS) in the female genital tract are uncommon and are usually of uterine origin. Extra-uterine ESS are extremely rare. Only 86 cases have been reported in the literature. More than 50% of the cases were associated with pre-existing endometriosis. We report a case of extrauterine low grade ESS associated with extensive endometriosis of the pelvis. We suspected that the tumor was a primary ovarian carcinoma due to elevated serum CA-125. Agile in public sector: Case study of dairy farm management projects

Abstract:Agile software development approach seems to work well in an organization where processes change on a regular basis. Conversely, adopting Agile practice in public sector, where work procedures have to follow a well-defined processes, faces several challenges. In this paper, we investigated how Agile methods could be adopted for practice in a public sector context using two projects of different sizes and complexity as our case studies. Our results show that certain Agile methods were able to work well in a public sector with some additional procedures required, while the main challenges of applying Agile methods in public sector are the change request management and the procurement process.
Measurement of peripheral muscle resistance in rats with chronic ischemia-induced paraplegia or morphine-induced rigidity using a semi-automated computer-controlled muscle resistance meter

Abstract:In experimental and clinical studies, an objective assessment of peripheral muscle resistance represents one of the key elements in determining the efficacy of therapeutic manipulations (e.g. pharmacological, surgical) aimed to ameliorate clinical signs of spasticity and/or rigidity. In the present study, we characterize a newly developed limb flexion resistance meter which permits a semi-automated, computer-controlled measurement of peripheral muscle resistance (PMR) in the lower extremities during a forced flexion of the ankle in the awake rat. Ischemic paraplegia was induced in Sprague-Dawley rats by transient aortic occlusion (10 min) in combination with systemic hypotension (40 mm Hg). After ischemia the presence of spasticity component was determined by the presence of an exaggerated EMG activity recorded from gastrocnemius muscle after nociceptive or proprioceptive afferent activation and by velocity-dependent increase in muscle resistance. Rigidity was induced by high dose (30 mg/kg, i.p.) of morphine. Animals with defined ischemic spasticity or morphine-induced rigidity were then placed into a plastic restrainer and a hind paw attached by a tape to a metal plate driven by a computer-controlled stepping motor equipped with a resistance transducer. The resistance of the ankle to rotation was measured under several testing paradigms: (i) variable degree of ankle flexion (40 degrees, 50 degrees, and 60 degrees), (ii) variable speed/rate of ankle flexion (2, 3, and 4 sec), (iii) the effect of inhalation anesthesia, (iv) the effect of intrathecal baclofen, (v) the effect of dorsal L2-L5 rhizotomy, or (vi) systemic naloxone treatment. In animals with ischemic paraplegia an increased EMG response after peripheral nociceptive or proprioceptive activation was measured. In control animals average muscle resistance was 78 mN and was significantly increased in animals with ischemic spasticity (981-7900 mN). In ischemic-spastic animals a significant increase in measured muscle resistance was seen after increased velocity (4 > 3 > 2 sec) and the angle (40 degrees > 50 degrees > 60 degrees) of the ankle rotation. In spastic animals, deep halothane anesthesia, intrathecal baclofen or dorsal rhizotomy decreased muscle resistance to 39-80% of pretreatment values. Systemic treatment with morphine induced muscle rigidity and corresponding increase in muscle resistance. Morphine-induced increase in muscle resistance was independent on the velocity of the ankle rotation and was reversed by naloxone. These data show that by using this system it is possible to objectively measure the degree of peripheral muscle resistance. The use of this system may represent a simple and effective experimental tool in screening new pharmacological compounds and/or surgical manipulations targeted to modulate spasticity and/or rigidity after a variety of neurological disorders such as spinal cord traumatic or ischemic injury, multiple sclerosis, cerebral palsy, or Parkinson's disease.
Amplitude- and velocity-dependency of rigidity measured at the wrist in Parkinson's disease

Abstract:Objective: Quantify the effects of increased amplitude and rate of muscle stretch on parkinsonian rigidity. Methods: Eighteen subjects with Parkinson's disease participated in this study. Subjects' tested hand was passively displaced through 60° and 90° ranges of wrist flexion and extension at velocities of 50°/s and 280°/s in both treated and untreated conditions. Joint angular position, resistance torque, and surface electromyography (EMG) of the wrist flexors and extensors were recorded. Rigidity was quantified by normalized work scores and normalized angular impulses for flexion and extension, separately. Reflex responses of stretched and shortened muscles were quantified by mean EMG and EMG ratio. A series of ANOVAs was performed to determine the effect of amplitude, velocity and medication on selected variables. Results: Both work scores and angular impulses revealed that the larger displacement amplitude and the higher velocity were associated with significantly greater rigidity, increased EMG ratio and mean EMG of stretched muscles. Dopaminergic medication was not associated with a reduction in rigidity. Conclusions: Parkinsonian rigidity is modulated by the amplitude and rate of muscle stretch. Significance: These findings shed light on the biomechanical underpinnings and physiological characteristics of rigidity and may inform clinical rigidity assessment in Parkinson's disease.

Abstract:We propose a novel system that analyzes the components of rigidity in Parkinson's disease (PD) usually perceived by physicians, in a very simple and systematic way for routine clinical practice. Our system is composed of two compact force sensors, a gyroscope, and EMG surface electrodes. Muscle tone was assessed in 24 healthy elderly subjects and 27 PD patients by passive extension and flexion of the elbow joint with ramp-and-hold trajectory. Torque and angle data in the dynamic phase were used to calculate "elastic coefficients" in extension and flexion, and the mean value of torque in each dynamic phase at each joint angle (defined as "Bias") was also calculated. The muscle activity index in the static phase (EMG Index) was obtained for both biceps brachii (BB) and triceps brachii (TB) muscles. "Elastic coefficients," sum of the "difference of Bias" and "EMG Index" for BB all correlated well with UPDRS score. Based on our results, Parkinsonian rigidity consists of both an "elastic" component and a "difference of Bias" component, and neurologists may assess greater rigidity when either one or both components are high in value. The EMG index was useful for differentiating PD patients with slight rigidity from healthy elderly adults.

Abstract:Background: Parkinson's disease (PD) is a neurodegenerative disorder that features motor and non-motor deficits. The use of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced dopamine neuron degeneration has been widely practiced to produce reliable animal models of PD. However, most previous preclinical studies focused on motor dysfunction, and few non-motor symptoms were evaluated. Thus far, there is a lack of comprehensive investigations of the non-motor symptoms in animal models. Objectives: In this study, we aim to use a battery of behavioral methods to evaluate non-motor symptoms in MPTP-induced non-human primate PD models. Methods: Cognitive function, sleep, and psychiatric behaviors were evaluated in MPTP-treated cynomolgus monkeys. The tests consisted of a delayed matching-to-sample (DMTS) task, the use of a physical activity monitor (PAM), an apathy feeding task (AFT), the human intruder test (HIT), novel fruit test (NFT), and predator confrontation test (PCT). In addition, we tested whether the dopamine receptor agonist pramipexole (PPX) can improve these non-motor symptoms. Results: The present results show that the MPTP-treated monkeys exhibited cognitive deficits, abnormal sleep, and anxiety-like behaviors when compared to the control monkeys. These symptoms were relieved partially by PPX. Conclusions: These results suggest that MPTP-induced PD monkeys displayed non-motor symptoms that were similar to those found in PD patients. PPX treatment showed moderate therapeutic effects on these non-motor symptoms. This battery of behavioral tests may provide a valuable model for future preclinical research.
Analysis of interactive effect of stretch reflex and shortening reaction on rigidity in Parkinson's disease

Abstract:Objective: To examine the correlation between rigidity and interaction of stretch reflex and shortening reaction during passive movements of the wrist and to compare this correlation with that between rigidity and stretch reflex alone. Methods: Twelve subjects with Parkinson's disease participated in the study in Off-medication and On-medication states. A servomotor imposed wrist flexion and extension within +/-30 degrees at velocities 50 and 280 degrees/s, while joint torque and EMG of the wrist flexors and extensors were recorded. Rigidity was quantified by integrating torque with joint angle, i.e., objective rigidity (OR) score, for extension and flexion, respectively. The interaction between EMG responses was estimated by calculating a ratio of normalized EMG in stretched to shortened muscles for both movements. Results: The OR scores were more strongly correlated with the EMG ratios than with EMG of the stretched muscles alone. The strongest and significant correlation was found between the OR score and EMG ratio during the extension at high velocity in the Off-medication (r=0.792, p=0.002). Conclusions: Both stretch reflex and shortening reaction are important determinants of rigidity. Significance: Study findings provide new insight into mechanistic descriptions of rigidity and have implications for development and evaluation of interventions.
Improved methods for electroacupuncture and electromyographic recordings in normal and parkinsonian rhesus monkeys

Abstract:Although acupuncture has been widely and routinely used in healthcare in the USA, its use has been based more on empirical observation than on scientific knowledge. Therefore, there is a great need for better understanding the underlying mechanism(s) of action. A great body of evidence supports that nonhuman primates are a candidate for studying human diseases. However, the use of nonhuman primates in neurophysiological, neuroimaging and neurochemical studies is extremely challenging, especially under fully conscious, alert conditions. In the present study, we developed a protocol for safely performing acupuncture, electroacupuncture (EA) and electromyography (EMG) in both normal nonhuman primates and animals with parkinsonian-like symptoms. Four normal and four hemiparkinsonian middle-aged rhesus monkeys were extensively trained, behaviorally monitored, and received both EA and EMG for several months. The results demonstrated that (1) all rhesus monkeys used in the study could be trained for procedures including EA and EMG; (2) all animals tolerated the procedures involving needle/electrode insertion; (3) EA procedures used in the study did not adversely alter the animal's locomotor activities; rather, MPTP-treated animals showed a significant improvement in movement speed; and (4) EMG detected significant differences in muscle activity between the arms with and without MPTP-induced rigidity. Our results support that rhesus monkeys can be used as an experimental animal model to study EA and that EMG has the potential to be used to objectively assess the effects of antiparkinsonian therapies. The results also indicate that animals, especially those with parkinsonian-like symptoms, could benefit from long-term EA stimulations.
A Cooperative Video Streaming System over the Integrated Cellular and DSRC Networks

Abstract:Due to the advance of wireless and mobile technologies, vehicular networks are widely discussed in recent years. When a vehicle requests a video stream from the Internet using the cellular network, i.e., 3G or 3.5G network, it may not have enough bandwidth to receive good quality of video. In order to resolve this problem, the vehicle can ask neighboring vehicles for the help of downloading the video stream. This scenario is called cooperative streaming in vehicular communications. The vehicle can communicate with neighboring vehicles using a short-range wireless technique, e.g., Dedicated Short Range Communications (DSRC). Since the topology of vehicle networks changes rapidly, two critical issues in the moving vehicle-based cooperative streaming are (1) how to form a cooperative group dynamically and (2) how to schedule neighboring vehicles to help the video downloading process. The simulation results reveal that the improvement of video quality is proportional to (1) high traffic flow, (2) high probability of willingness to share bandwidth and (3) low relative speed.
Association of Selective Serotonin Reuptake Inhibitor Use and Acute Angle-Closure Glaucoma.

Abstract:OBJECTIVE Selective serotonin reuptake inhibitors (SSRIs) are the most widely used antidepressant medications for treating patients with depression; however, ocular complication has been noted occasionally. This study investigated the relationship between recent SSRI use and the risk of acute angle-closure glaucoma (AACG) in the ethnic Chinese population in Taiwan. METHODS In this case-control study that involved using data from the Taiwan National Health Insurance database for the period 2000-2011, we recruited 1,465 patients with newly diagnosed AACG as case participants and 5,712 persons without glaucoma who were matched according to sex, age, and index year as controls. Immediate SSRI users were defined as patients who received at least 1 prescription for SSRIs within 7 days before the date of AACG diagnosis. Patients who received no SSRI prescriptions were defined as nonusers. Odds ratios (ORs) and 95% confidence intervals (CIs) were used to evaluate the risk of AACG associated with SSRI use. RESULTS After adjustment for confounding factors including non-SSRI antidepressant use and all comorbidities, the multivariable logistic regression model revealed that the adjusted OR of AACG was 5.80 for immediate SSRI users (95% CI, 1.89-17.9) when compared with nonusers. Further analysis, with nonusers as reference, resulted in an adjusted OR of 8.53 (95% CI, 1.65-44.0) for participants with a mean daily SSRI dose exceeding 20 mg. CONCLUSIONS Patients immediately using SSRIs have a 5.80-fold increased risk of AACG. Before prescribing SSRIs, clinicians should be aware of the potential AACG risks among elderly patients with depression.
Research and development of a portable device to quantify muscle tone in patients with Parkinsons disease

Abstract:Parkinson's disease (PD) is a progressive, degenerative condition that is characterised by tremor, bradykinesia, cogwheel rigidity and postural instability. Currently, only subjective tests are employed and we aim to develop a portable device to quantify muscle tone in patients with movement disorders, in particular, Parkinson's disease. A servo-motor robotic arm was developed to rhythmically flex and extend the subjects arm and their response was measured using continuous electromyography (EMG) sampled at 4kHz. Surface electrodes were attached over the biceps of the more severely affected arm and a reference electrode attached to the back of the contra-lateral hand. EMG data was normalized using the rest mean EMG level and then segmented into epochs according to the position data. EMG recorded from normal subjects remained flat throughout the trial with only very small fluctuations in amplitude about the mean. In contrast, biceps activity in parkinsonian patients tended to increase with flexion and as expected there were large variations in amplitude about the baseline activity. A fast fourier transform was also performed and spectra obtained from a typical Parkinsonian patient showed two peaks at approximately 6 Hz and 8 Hz, consistent with previously published data. In conclusion, the results clearly differentiate between normal and parkinsonian cases and to some extent severity. Quantification of disease severity might be possible given a broader range and greater number of patients.
Pharmacological validation of behavioural measures of akinesia and dyskinesia in a rat model of Parkinson's disease

Abstract:In an attempt to define clinically relevant models of akinesia and dyskinesia in 6-hydroxydopamine (6-OHDA)-lesioned rats, we have examined the effects of drugs with high (L-DOPA) vs. low (bromocriptine) dyskinesiogenic potential in Parkinson's disease on three types of motor performance, namely: (i) abnormal involuntary movements (AIMs) (ii) rotational behaviour, and (iii) spontaneous forelimb use (cylinder test). Rats with unilateral 6-OHDA lesions received single daily i.p. injections of L-DOPA or bromocriptine at therapeutic doses. During 3 weeks of treatment, L-DOPA but not bromocriptine induced increasingly severe AIMs affecting the limb, trunk and orofacial region. Rotational behaviour was induced to a much higher extent by bromocriptine than L-DOPA. In the cylinder test, the two drugs initially improved the performance of the parkinsonian limb to a similar extent. However, L-DOPA-treated animals showed declining levels of performance in this test because the drug-induced AIMs interfered with physiological limb use, and gradually replaced all normal motor activities. L-DOPA-induced axial, limb and orolingual AIM scores were significantly reduced by the acute administration of compounds that have antidyskinetic efficacy in parkinsonian patients and/or nonhuman primates (-91%, yohimbine 10 mg/kg; -19%, naloxone 4-8 mg/kg; -37%, 5-methoxy 5-N,N-dimethyl-tryptamine 2 mg/kg; -30%, clozapine 8 mg/kg; -50%, amantadine 40 mg/kg). L-DOPA-induced rotation was, however, not affected. The present results demonstrate that 6-OHDA-lesioned rats do exhibit motor deficits that share essential functional similarities with parkinsonian akinesia or dyskinesia. Such deficits can be quantified using novel and relatively simple testing procedures, whereas rotometry cannot discriminate between dyskinetic and antiakinetic effects of antiparkinsonian treatments.

Abstract:INTRODUCTION:Complete and specific ablation of a single dopaminergic (DA) pathway is a critical step to distinguish the roles of DA pathways in vivo. However, this kind of technique has not been reported in non-human primates. This study aimed to establish a lesioning method with a complete and specific ablation.METHOD:A carefully designed infusion route based on a MRI stereotactic technique was developed to deliver the highly selective dopaminergic toxin 1-methyl-4-phenylpyridinium (MPP+) unilaterally into multiple sites of compact part of substantia nigra (SNc) and striatum in monkeys. The nigrostriatal DA pathway was selected because lesioning of this pathway may induce symptoms that are suitable for evaluation. The pathological, behavioral, neuropharmacological, and clinical laboratorial data were collected to evaluate the lesioning effects.RESULT:Pathological examination revealed a complete ablation of tyrosine hydroxylase positive (TH+) neurons in the SNc, while preserving intact TH+ neurons in the ventral tegmental area (VTA) nearby. TH+ projections in the striatum were also unilaterally lost. The monkeys displayed stable (>28 weeks) rotations and symptoms which were expected with loss of DA neurons in the SNc, with rest tremor being an exception. No item implied the presence of a severe side effect caused by the operation or the intracerebral MPP+ infusion. The results suggested that rest tremor may not directly rely on the nigrostriatal pathway.CONCLUSION:Taken together, in addition to providing a specific nigrostriatal DA lesioned model, this method, combined with brain stimulation or other techniques, can be applied as a powerful tool for the complete lesion of any desired DA pathway in order to study its specific functions in the brain.

Abstract:Triacylglycerol (TG) synthesis rates of low-oxidative fast-twitch white and high-oxidative fast- and slow-twitch red skeletal muscle fiber sections of adult rats were measured by the incorporation of perfusate-derived palmitate into the neutral lipid fraction by use of a perfused hindquarter preparation under high-flow conditions. The perfusion medium consisted of 95% O2-5% CO2 Krebs-Henseleit buffer, pH 7.4, containing 5 g/100 ml bovine serum albumin, 100 microU/ml insulin, 5 mmol/l glucose, amino acids, and added fatty acids (FA), including 0.1 microCi/ml [3H]palmitate. FA incorporation was linear with time. TG synthesis rates correlated (r > or = 0.90) with the oxidative capacity of each of the different fiber type sections and increased in proportion to the perfusate FA concentration (0.25, 0.5, 1.0, 1.5, and 2.0 mmol/l). TG synthesis rates among different muscle fiber sections were related (r > or = 0.90) to perfusate flow during high-flow conditions; however, this was not causal, because TG synthesis rates within each muscle fiber section were independent of flow rate when experimentally varied over an approximately threefold range. Thus the relatively high TG synthesis rates observed in the high-oxidative muscle sections are not uniquely related to their high-flow capacities but are inherent to the TG synthesis process, probably events associated with FA uptake and/or capacity of the TG synthesis pathway.

Abstract:Acupuncture has increasingly been used as an alternative therapy for treatment of Parkinson's disease (PD). However, the efficacy of acupunture for PD still remains unclear. The present study was designed to objectively and safely monitor anti-parkinsonian effects of electroacupuncture (EA) and brain activity in nonhuman primates modeling human PD. Six middle-aged rhesus monkeys were extensively studied by a computerized behavioral testing battery and by pharmacological MRI (phMRI) scans with specific dopaminergic drug stimulations. All animals were evaluated for behavior and phMRI responses under normal, parkinsonian, parkinsonian with EA treatment and parkinsonian after EA treatment conditions. Stable parkinsonian features were observed in all animals prior to entering the EA study and positive responses to levodopa (L-dopa) challenge were also seen in all animals. The results demonstrated that chronic EA treatments could significantly improve the movement speed and the fine motor performance time during the period of EA treatments, and the effectiveness of EA could be detected even 3 months after the EA treatment. The phMRI data revealed that chronic EA treatments could alter neuronal activity in the striatum, primary motor cortex (M1), cingulate gyrus and global pallidus externa (GPe) in the ipsilateral hemisphere to MPTP lesions. As seen in the changes of parkinsonian features, the residual effects of phMRI responses to apomorphine (APO) challenge could also be found in the aforementioned areas. The results strongly suggest that anti-parkinsonian effects of EA can be objectively assessed, and the method used in the present study could be translated into the human clinic with some minor modifications.
