Abstract:Virulence bioassays of the 5 isolates of Beauveria bassiana from different species of insects against Takecallis taiwanus (Takahashi) were carried out. The result showed that all 5 isolates tested were capable of infecting T. taiwanus(Takahashi). The time-dose-mortality model (namely complementary log-log model) was used for the analysis of the data obtained from the bioassay. Time-dependent lg(LT_50) and dose-dependent LC_50 were estimated respectively. For B_12, Bxs, REBb01, Zhe-B and F-263, the lg(LC_50) by the 9~th day after inoculation were estimated to be 4.054 9, 4.462 0, 4.626 1, 4.929 0 and 5.158 0 conidia·mL~-1 respectively, and the LT_50 at an inoculate dose of 1×10~6 conidia·mL~-1 were estimated to be 3.63, 3.70, 4.31, 4.34 and 5.20 days respectively. Based on three-dimension figures of cumulative mortality of T. taiwanus(Takahashi) with the changes of time and dose, virulence of each isolate under different time and dose were evaluated integrally. The virulence of 5 isolates in a descending order was B_12, Bxs, REBb01, and followed by Zhe-B and F-263.
Virulence of Beauveria bassiana on reproduction of the aphid Takecallis taiwanus

Abstract:Virulence of three isolates of Beauveria bassiana REBb01,B_(12) and Bxs,on reproduction of the aphid,Takecallis taiwanus,was studied.Due to reduced survival,within 10 d of inoculation the pathogen reduced infection fecundity of T.taiwanus.However,for an individual aphid infected with the pathogen,the number of its progeny was similar to that of aphid without infection.After inoculation of B.bassiana at different doses,the rate of increment for T.taiwanus number was no scientific decrease.

Abstract:Abstract Objective This study was aimed to assess the enzymatic activity and pathogenicity potential of Beauveria bassiana and Metarhizium anisopliae against whiteflies in Ethiopia. Results The data showed that Beauveria bassiana AAUMB-29, AAUMFB-77, and AAUEB-59 generated the highest chitinase (EI = 3.41), lipase (EI = 4.45), and protease activities (EI = 5.44) respectively. The pathogenicity study of isolates on whitefly nymphs and adults indicated significant variation (P < 0.05) with mortality ranging from 71.67 to 98.33% and 60 to 100% against Bemisia tabaci and Trialeurodes vaporariorum nymphs respectively. The mortality of adults was between 58 and 94.27% against B. tabaci and 59.03 to 95.37% against T.vaporariorum. The result also showed that AAUMB-29, AAUMFB-77, and AAUDM-43 were the most virulent with LC 50 values of 2.7 × 10 4 , 5.3 × 10 4 , and 5.4 × 10 4 conidia/ml against nymphs of B. tabaci , and with LC 50 values 6.8 × 10 4 , 8.2 × 10 4 , and 7.2 × 10 4 conidia/ml against nymphs of T. vaporariorum , respectively. The B. bassiana AAUMB-29, B. bassiana AAUMFB-77, and M. anisopliae AAUDM-43 induced the highest whitefly mortality than other isolates. These isolates can be recommended for further tests under field conditions to fully realize their potential as effective biocontrol agents against whitefly pests in tomato.
[Evaluation for Enhanced Aphidicidal Activity of a Noval Emulsifiable Formulation of Beauveria Bassiana Conidia].

Abstract:A noval emulsifiable formulation (EF) of Beauveria bassiana SG8702 containing 10(10) conidia/mL was prepared by suspending dry conidia in mineral oil-based liquid containing emulsifier, stabilizer and UV protectant (emulsion), and then bioassayed on newly merged adults of the green peach aphid, Myzus percisae, in parallel to the unformulated conidia (UC). For inoculation, five gradient concentrations of aqueous EF and UC suspensions (5 x 10(5) - 1 x 10(8) conidia/mL) were separately sprayed on to aphids on detached cabbage leaves (30 - 40 adults/leaf) using a Micro Ulva hand-held sprayer, generating EF dosages of 1.5, 6.3, 32.6, 87.6 and 701.1 conidia/mm2, and UC dosages of 2.8, 9.2, 52.7, 114.8 and 1005.9 conidia/mm2, respectively. All dosage treatments including controls for EF (100-fold dilution of the emulsion) and UC (water spray) were replicated three times (aphid colonies on three leaves), and maintained in Petri dishes at 23 degrees C and 12 : 12 (L: D) for 8-day daily records. Cadavers, whenever found, were moved into a moist chamber to facilitate fungal outgrowths for verification of B. bassiana infection. The resulting data fit very well to time-dose-mortality model with EF having a larger dose effect (1.05 +/- 0.10) than UC (0.82 +/- 0.11) and also faster time effect. The estimates of the dose- and time-effect parameters were then used to compute lethal concentrations (LC50) and time lengths (LT50). The estimates of LC50 with 95% confidence intervals for both EF and UC were 9.0 (6.9 - 11.6) and 634 (263 - 1532) conidia/mm2 on day 4 after spray, and 3.3 (2.4 - 4.6) and 5.3 (3.2 - 8.6) on day 7, apparently decreasing with increasing time lengths after spray. The estimates of LT50 for EF and UC were 4.9 d and 7.2 d at 5 conidia/mm2, 4.0 d and 5.6 d at 10 conidia/mm2, and 3.2 d and 4.5 d at 100 conidia/mm2, indicating a trend of decrease with increasing counts of conidia attached to the leaves. The results indicate that the emulsifiable formulation of B. bassiana conidia had greater aphidicidal activity than the unformulated conidia. Finally, problems with evaluation of mycoinsecticides and other microbial formulations are discussed and technical standards are recommended.
Lethal effect of Beauveria bassiana, Metarhizium anisopliae, and Paecilomyces fumosoroseus on the eggs of Tetranychus cinnabarinus (Acari: Tetranychidae) with a description of a mite egg bioassay system

Abstract:Fungal biocontrol agents, including 10 isolates of Beauveria bassiana, Metarhizium anisopliae, and Paecilomyces fumosoroseus were bioassayed for their lethal effects on the eggs of the carmine spider mite, Tetranychus cinnabarinus. The eggs were naturally laid on detached fava bean leaves on agar in petri dishes at 25°C under a photophase of 12:12 L:D. Suspensions of aerial conidia produced on rice were sprayed onto the leaves, yielding three conidial concentrations (No. conidia/mm2) from low to high for each of the fungal isolates with 3 or 4 replicates (30–65 eggs/leaf or replicate) at each concentration. During a 12-day observation period after spray, the infected eggs shrunk in shape, then turned orange-brown for B. bassiana and P. fumosoroseus or dark-gray for M. anisopliae, and eventually had outgrowths of the sprayed fungus when maintained under moist conditions. Egg mortalities caused by fungal infection varied over time but had no change from day 8 or 9 onward. Differences of the mortalities on day 9 were significant among the fungal species or isolates tested. Based on the LC50 estimates determined by the concentration–mortality relationships of all isolates from probit analysis, B. bassiana Bb2860 with an LC50 of 548conidia/mm2 was highly infectious to T. cinnabarinus eggs, followed by P. fumosoroseus Pfr116 (848conidia/mm2) and Pfr153 (913conidia/mm2). Two M. anisopliae isolates with LC50s of 1717 and 1985conidia/mm2 were inferior to the three isolates. The other isolates were poor or very poor in ovicidal activity. Our results confirmed the ovicidal activity of the three fungal species and suggested the feasibility to search for more ovicidal isolates from fungal species that may serve as biocontrol agents against spider mites such as T. cinnabarinus. The mite egg bioassay system developed in this study will be useful towards this goal.
Evaluation Of The Time-Concentration-Mortality Responses Of Plutella Xylostella Larvae To The Interaction Of Beauveria Bassiana With A Nereistoxin Analogue Insecticide

Abstract:Six bioassays were conducted to evaluate the interaction between Beauveria bassiana SGS702 and a nereistoxin analogue insecticide, diammonium S,S'-(2-dimethylaminotrimethylene)di(thiosulfate), which is highly compatible with the fungal biocontrol agent against diamondback moth Plutella xylostella (L.). Second-instar larvae were exposed to sprays of B. bassiana alone (assay 1) at concentrations of 21-38, 157-232 and 822-1133 conidia mm(-2) or together with the insecticide at the low application rates of 5, 10, 25, 50 and 100 mu g AI ml(-1) (assays 2-6), and then maintained at 25 degrees C and 12:12 h light:dark photoperiod for daily monitoring of mortality for 8 days. Based on the modelling of the resultant time-concentration-mortality data sets, the fungal agent was highly virulent to P. xylostella with an LC50 decreasing from 269 conidia mm-2 on day 4 to 107 on day 8. Lower lethal concentrations or shorter median lethal times resulted from fungal sprays including the tested chemical rates; the latter never caused higher mortalities than the fungal treatments alone. The fungal action over 3-7 days after spray was significantly enhanced by including in the fungal sprays the chemical at rates of >= 25 mu g ml(-1) for 2.6- to 1756-fold reduction of LC50 values, >= 50 mu g ml(-1) for 4- to 274-fold reduction of LC70 values and 100 mu g ml(-1) for 9- to 33-fold reduction of LC90 values respectively. These rates were equivalent to 5-20% of the chemical rate labelled for field application. The fungal and chemical interaction outlined above highlights the feasibility of combined formulation or application of B. bassiana and the chemical insecticide for P. xylostella control. Abstract:An aphid-derived isolate of Beauveria bassiana (SGBB8601) and Verticillium lecanii (DNVL8701) were bioassayed against six species of globally distributed cereal-infesting aphids under controlled conditions. Aphids were immersed in conidial suspensions at concentrations ranging from 104 to 108 conidia/ml, each concentration being replicated six times with 20 aphids treated per replicate. Pathogenicity of both fungal species was confirmed on all the aphids tested, but in most cases, B. bassiana was more virulent than V. lecanii . The LC50 values for B. bassiana and V. lecanii were 8.2 × 104 and 4.1 × 105 conidia/ml, respectively, on Diuraphis noxia ; 2.1 × 105 and 9.8 × 105 on Schizaphis graminum ; 3.3 × 105 and 7.0 × 105 on Metapolophium dirhodum ; 1.1 × 105 and 1.3 × 106 on Sitobion avenae ; 2.1 × 106 and 8.9 × 105 on Rhopalosiphum maidis ; and 3.3 × 106 and 6.9 × 106 on R. padi . Statistical analysis indicated that there were substantial differences in susceptibility to the two pathogens and in virulence of each fungus to different aphid species. The LT50 values at all concentrations varied between the pathogens and among the aphid hosts with B. bassiana tending to kill aphids more rapidly. The mortality data were probit transformed, and a third-order polynomial model was fit to the data to describe responses of each host to conidial concentration and time after treatment. The potential for use of B. bassiana for aphid control is discussed.
Compatibility of Ten Acaricides with Beauveria Bassiana and Enhancement of Fungal Infection to Tetranychus Cinnabarinus (acari : Tetranychidae) Eggs by Sublethal Application Rates of Pyridaben

Abstract:The compatibilities of 10 acaricides with Beaveria bassiana SG8702 varied significantly in 24 h liquid cultures including three chemical rates. Pyridaben, hexythiazox and propargite were found highly compatible with the fungus even at field application levels. Four bioassays were performed with Tetranychus cinnabarinus eggs using a non-touch leaf method. The <= 18 h-old eggs laid on detached fava bean leaves were exposed to gradient conidial sprays of B. bassiana alone in Assay 1 or together with the pyridaben rates of 0.05, 1.0 and 2.5 a.i. mu g/ml in Assays 2-4, respectively. The percentages of hatched eggs decreased with the increase in conidial concentrations and increased over post-spray days at a given concentration but had no more change from Day 9 onwards. Final egg mortalities reached 65.4, 72.3, 77.7 and 85.3% at similar to 1,000 conidia/mm(2) in Assays 1-4. Ovicidal activity of B. bassiana [LC50 with 95% CI: 546 (406-818) conidia/mm(2)] was enhanced significantly by fungal sprays including pyridaben [LC(50)s: 322 (220-527), 199 (144-282), and 75 (48-103) conidia/mm(2)]. The effects of both agents and their interactions on the hatch rate of the mite eggs over post-spray days were illustrated by modeling the time-concentration-hatch relationship. The results indicate an alternative to utilize interactions of both agents against spider mites.
Comparative Tolerances of Various Beauveria Bassiana Isolates to UV-B Irradiation with a Description of a Modeling Method to Assess Lethal Dose.

Abstract:The tolerances of 20 Beauveria bassiana isolates derived from host insects worldwide to UV-B irradiation were assessed quantitatively in multi-dose bioassays. Conidial suspensions of the isolates smeared on glass slides were exposed to the gradient UV-B doses of 0.1–1.6 J cm−2 (D), which generated from 0.75 to 10.17 min irradiation of weighted 312-nm wavelength at 2.0–2.61 mW cm−2. Irradiated conidia were then incubated for 24 h at 25°C under saturated humidity. The ratio of germination at each dose over that in the blank control was defined as survival index (I s). For all isolates, the I s − D observations fit well with the survival model I s = 1/[1 + exp(a + bD)] (0.94 ≤ r 2 ≤ 0.99) generated widely spanned lethal doses of 0.154–0.928, 0.240–1.139, and 0.383–1.493 J cm−2 for their losses of 50%, 75%, and 95% viabilities, respectively. These were far below the solar UV-B dose of 2.439 J cm−2 measured in a sunny day during the summer. The large variation of UV-B tolerance among the isolates indicates a necessity to select UV-tolerant candidates for formulations applied to insect control during summer. The highly efficient bioassay method was developed to measure accurately the UV-B tolerances of fungal biocontrol agents as lethal doses.
Time-Concentration-Mortality Modeling of the Synergistic Interaction of Beauveria Bassiana and Imidacloprid Against Nilaparvata Lugens

Abstract:Interactions between the entomopathogenic fungus, Beauveria bassiana SG8702, and imidacloprid on Nilaparvata lugens (Homoptera: Delphacidae) were studied in laboratory bioassays by spraying suspensions of unformulated conidia (assay 1) and aqueous dilutions of emulsifiable conidia formulation alone (assay 2) or together with imidacloprid at the sub-lethal rates of 0.5, 1.0 and 2.0 mu g ml(-1) (assays 3-5). Each assay consisted of five conidia concentrations plus an appropriate control and three replicates, each including 30-40 third-instar nymphs, so as to generate time-concentration-mortality data for modeling analysis. A mineral oil-based emulsion used to formulate B bassiana slightly enhanced fungal activity but had no significant impact on the background mortality of N lugens. On the basis of LC50 estimates and associated variances on days 4-12 after spraying, synergistic interactions of both agents or formulations were determined by estimating relative potencies of assay 2 over assay 1 (1.2-9.0), assay 3 over assay 2 (1.3-1.7), assay 4 over assay 2 (7.5-9.6), assay 5 over assay 2 (22.7-101), assay 4 over assay 3 (3.8-5.8), assay 5 over assay 3 (16.1-61.0), and assay 5 over assay 4 (3.0-10.5). The time-concentration-mortality modeling method was not only mathematically but also biologically robust to evaluate the interactions of B bassiana and imidacloprid on N lugens. Compared with their counterparts, enhanced fungal formulations displayed consistently earlier or greater activities against the pest species based on LC50 and LT50 estimates determined from their time-concentration-mortality relationships. Diversity study of Beauveria bassiana species for finding the most virulent strain to manage Bemisia tabaci in cotton

Abstract:Beauveria bassiana (Bal.-Criv.) is an important entomopathogenic fungus being used for the management of various agricultural pests worldwide. However, all strains of B. bassiana may not be effective against whitefly, Bemisia tabaci , or other pests, and strains show diversity in their growth, sporulation, virulence features, and overall bioefficacy. Thus, to select the most effective strain, a comprehensive way needs to be devised. We studied the diversity among the 102 strains of B. bassiana isolated from 19 insect species based on their physiological features, virulence, and molecular phylogeny, to identify promising ones for the management of B. tabaci . Strains showed diversity in mycelial growth, conidial production, and their virulence against B. tabaci nymphs. The highest nymphal mortality (2nd and 3rd instar) was recorded with MTCC-4511 (95.1%), MTCC-6289 (93.8%), and MTCC-4565 (89.9%) at a concentration of 1 × 10 6 conidia ml −1 under polyhouse conditions. The highest bioefficacy index (BI) was in MTCC-4511 (78.3%), MTCC-4565 (68.2%), and MTCC-4543 (62.1%). MTCC-4511, MTCC-4565, and MTCC-4543 clustered with positive loading of eigenvalues for the first two principal components and the cluster analysis also corresponded well with PCA (principal component analysis) (nymphal mortality and BI). The molecular phylogeny could not draw any distinct relationship between physiological features, the virulence of B. bassiana strains with the host and location. The BI, PCA, and square Euclidean distance cluster were found the most useful tools for selecting potential entomopathogenic strains. The selected strains could be utilized for the management of the B. tabaci nymphal population in the field through the development of effective formulations.

Due to the high toxicity of chemical pesticides on human health and the environment, scientists focused on developing alternative safe methods to control this insect, including the use of biocontrol agents. Beauveria bassiana is one of the common fungi used as a bio-control agent. This entomopathogenic fungus was cultivated on Petri dishes containing PDA. The spore suspension of this fungus was prepared using sterilized distilled water, and three concentrations were used: 20×106 , 10×106 and 5×106 spores/ml. The effectiveness of these three concentrations was tested on larvae and pupae of the studied insect. The larval mortality rate ranged from 73.33% at the highest concentration to 33.33% at the lowest concentration used. Pupal mortality reached 66.66% at the highest concentration and 16.66% at the lowest concentration of spore suspension. Mortality rate was directly proportional to the spore suspension concentration of the entomopathogenic fungus. Abstract:Abstract Background The diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), is a noxious pest of cruciferous crops all over the world causing serious economic damage. Management of insect pest generally depends on chemical control; however, due to development of resistance against all types of insecticides, alternative approaches especially utilization of a microbial agent is inevitable. Results Potential of 2 entomopathogenic fungi (EPF), viz., Beauveria bassiana and Metarhizium anisopliae , was evaluated against 2 nd and 3 rd larval instars of P. xylostella by adopting leaf dip and direct spraying methods under laboratory conditions. Significant mortality rate was achieved by each fungus under adopted methodologies. However, B. bassiana was found to be more effective in both conditions than M. anisopliae . Highest mean corrected mortality (77.80%) was recorded, when spores of B. bassiana were sprayed on the 2 nd instar larvae (LC 50 =1.78×10 4 /ml) after the 6th day of treatment. Similarly, incase of M. anisopliae LC 50 for the 2 nd instar at the same methodology was 2.78×10 4 /ml with a mortality percentage of 70.0%. Offspring sex ratio was non-significantly related to treatment concentration and methodology, except for the control. Conclusion Beauveria bassiana and M. anisopliae had potential to suppress P. xylostella infestations when applied appropriately. Present findings suggested that B. bassiana and M. anisopliae when sprayed on immatures of host insect had more effect as compared to leaf dip procedure. Furthermore, no significant effect of concentrations was observed on sex ratio.

Time-specific Infection Rate of Beauveria Bassiana on Myzus Persicae after Topical Inoculation of Conidial Suspension

Abstract:Conidial suspension(5 x 10(6) conidia.ml-1) of the entomopathogenic fungus, Beauveria bassiana BBSG8702 was used to inoculate newly emerged apterous adults of the green peach aphid (Myzus persicae) on detached cabbage leaves in order to measure time-specific infection rate of the fungal agent. The aphids of conidia attached were disinfected using 0.2% chlorothalonil solution at intervals of 8-56 h after inoculation and then maintained at 10 and 20 degrees C for daily observation. There was a significant difference (P < 0.05) in mortality attributed to the fungal infection among the time intervals within 56 h at 10 degrees C after inoculation or within 40 h at 20 degrees C. Compared with the control (not treated with 0.2% chlorothalonil after inoculation), time-specific infection rate at 10 degrees C was estimated as 22.9%, 48.8%, 64.9%, 80.4%, 72.7%, and 98.3% within 8 h, 16 h, 24 h, 32 h, 40 h, and 56 h after inoculation, respectively. The same estimates at 20 degrees C were 31.6%, 48.8%, 58.6%, 86.9%, 97.2%, and 98.7%, respectively. The results indicate that the first 24 h after inoculation was crucial to the infection of B. bassiana to M. persicae. Effective infection with the first 24 h accumulated to 59-65% at 10-20 degrees C.
Histopathological and Molecular Insights into the Ovicidal Activities of Two Entomopathogenic Fungi Against Two-Spotted Spider Mite

Abstract:Entomopathogenic fungi can infect and kill spider mite eggs but their ovicidal activities are poorly understood. Here we gain histopathogenical and molecular insights into the ovicidal activities of Beauveria bassiana and Isaria fumosorosea against the two-spotted spider mite, Tetranychus urticae. Scanning electronic microscopy indicated successful adhesion and germination of fungal conidia on egg shell at 24h post-spray (HPS). Germ tubes of both fungi could penetrate into egg shell with penetration pegs at 48 HPS. Interestingly, the germ tubes of B. bassiana may elongate on egg surface to locate appropriate sites for penetration, acting as 'searching' hyphae. Aside from the normal penetration, the germ tubes of I. fumosorosea can be completely or partially embedded into egg shell for a distance of extension, forming shell humps. Light microscopy of ultrathin sections of infected eggs showed shrunken (affected) or disrupted embryos at 48-96 HPS despite little effect on egg cleavage at 24 HPS. However, distinguishable hyphal cells were hardly found inside the embryos lacking oxygen although fungal outgrowths were abundant on unhatched (killed) eggs. In PCR with specific probes, the 18S rDNA signals of B. bassiana (412 bp) and I. fumosorosea (454 bp) in the DNA extracts from surface-cleaned mite eggs increased at 0-96 HPS, confirming fungal colonization in the infected eggs. We consider that the colonization on shell surface and underside could rely upon extending hyphae for uptake of egg nutrition, resulting in embryo disruption. Our observations add knowledge to microbial control of spider mites.
Evaluation of Interactive Efficacy of Two Mycoinsecticides and Low Application Rate Imidacloprid in Controlling Greenhouse Whitefly Trialeurodes Vaporariorum (homoptera: Aleyrodidae)

Abstract:The interactive effects of pure emulsifiable formulations (10(10) conidia.ml-1) of Beauveria bassiana and Paecilomyces fumosoroseus conidia and 1% and 3% of imidacloprid 10% WP were tested for controlling greenhouse whitefly, Trialeurodes vaporariorum, on lettuce grown in four polyethylene film-covered greenhouses (5 m x 100 m) in Kunming, Yunnan. Six fungal treatments, two imidacloprid ones, and one water-spray control were included in the experiment, with each being replicated in three plots (7 m x 5 m.plot-1). For each of the treatments, 1000-fold aqueous dilution (10(7) conidia.ml-1) was sprayed twice at a 15-d interval, with the first spray given on day 11 after lettuce transplantation. Counts of live and dead whiteflies from each treatment were made on the initial day, and then, once every five days. During a 30-d period of observation from August 20, the two applications of the fungal formulations alone or those containing low rates of imidacloprid 10% WP effectively protected the lettuce from damage by T. vaporariorum, and generated relative efficacies. Whitefly density decreased > 95%, which was significantly higher than that attributed to the low application rates of imidacloprid 10% WP. Based on the estimations of relative efficacy and percent density decrease, P. fumosoroseus treatments controlled the whiteflies better than B. bassiana ones, and the effects of both fungi on the pest population were apparently enhanced with the quantity of imidacloprid added. The differences among the fungal treatments in the two estimations were larger during the first 10 days, but decreased thereafter, becoming insignificant on day 10 after the second spray. The results indicated that the fungal formulations tested in this study are of great potential for practical use in whitefly control, and their efficacy could be enhanced with low rates of imidacloprid.
Isolation, purification and identification of biological compounds from Beauveria sp. and their evaluation as insecticidal effectiveness against Bemisia tabaci

Abstract:Bemisia tabaci is one of the most notorious agricultural pests in the world. A vicious circle among insect resistance, dose increased, environment and human body impaired as the overuse of synthetic pesticides are becoming increasingly evident. Entomopathogenic Beauveria sp. is known as an effective natural enemy to control B. tabaci. Therefore, this study aimed to purify and identify the biological compounds from Beauveria sp. LY2 via extensive chromatographic techniques, NMR and MS and evaluated for their insecticidal activities against B. tabaci via contact and feeding assay. The outcome identified that one new cerebroside, cerebroside F (1), nine known compounds, cerebroside B (2), bassiatin (3), methyl 1,4-dihydro-4-oxo-2-quinolinecarboxylate (4), cerevisterol (5), 9-hydroxycerevisterol (6), 6-dehydrocerevisterol (7), (22E,24R)-ergosta-8(14),22-diene-3β,5α,6β,7α-tetrol (8), melithasterol B (9) and ergosterol peroxide (10) were isolated. Among the known compounds, methyl 1,4-dihydro-4-oxo- 2-quinolinecarboxylate (4) was isolated from natural origin for the first time. It is demonstrable from the results that compounds 3, 4 and 7 strongly featured insecticidal activities against B. tabaci, being the LC50 value as 10.59, 19.05, 26.59 μg/mL respectively in contact as well as 11.42, 5.66, 5.65 μg/mL respectively in feeding experiment. Moreover, no adverse effect on plant growth/height or phytotoxicity was observed on pepper, cucumber, tomato and cotton. The data from the current study has provided the foundation for the use of newly purified compounds against Bemisia tabaci as an alternative to synthetic chemical compounds.

Abstract:The virulence of six isolates of Beauveria bassiana (Balsamo) Vuillemin to the Russian wheat aphid, Diuraphis noxia (Mordvilko), was bioassayed. All the isolates tested were capable of infecting D. noxia , but their virulence, determined by LC50's and LT50's, varied. An aphid-derived isolate had the lowest LC50, 0.57 × 105 conidia/ml, and among the shortest LT50's at all five concentrations of conidial suspension. Two isolates from nonhomopteran hosts and one from an homopteran also displayed significant virulence and were very similar to one another, whereas two Homoptera-derived isolates were significantly less virulent. Log-transformed concentrations and post-treatment time (days) were correlated with probit-transformed mortality induced by each of the isolates. The resulting models, using third-order polynomials, gave good predictions of mortality within the range of concentrations tested. Colony growth rates for four isolates were relatively similar. Differences in virulence among the tested isolates are discussed in conjunction with their geographic origins, insect hosts, and growth and sporulation characteristics.

Abstract:Entomopathogenic fungi, with their cosmopolitan existence and rich diversity, present a sustainable solution towards integrated pest management programs. Due to their eco-friendliness and bio-persistence, these entomopathogens are preferred to kill insects at various stages of their life cycle. The Cowpea seed beetle, Callosobruchus maculatus (F.) (Coleoptera: Bruchidae), is an important bruchid pest in cowpea seed storages. The used pesticides against this pest have caused the occurrence of resistant populations and direct toxicity to the users. The objective of this study was to evaluate the mortality effects of two entomopathogenic fungi isolates obtained from fungi isolated from adults of C. maculatus collection, Beauveria bassiana and Isaria fumosorosea, against the larval stages of C. maculatus under laboratory conditions. The mortality percentage of 2 nd instar larval mortality peaked at 100% when the B. bassiana spores' concentration was 4*10 5 . On the other hand, the effect of I. fumosorosea spores' concentration on 2 nd instar larvae peaked at 76.67% when the spores' concentration was 4*10 5 , while it peaked at 73.33% when the I. fumosorosea extract concentration was 100%. For 4 th instar larvae, the results had shown the same pattern of effects by B. bassiana and I. fumosorosea spores and extract, with mortality percentage peaked at 96.67% when the B. bassiana spores' concentration was 4*10 5 , while it peaked at 83.33% when the B. bassiana extract concentration was 100%. In contrast, the effect of I. fumosorosea extract concentration on 4 th instar larval mortality peaked at 70% when the extract concentration was 4*10 5 , while it peaked at 73.33% when the I. fumosorosea extract concentration was 100%. For larvae, the relationship between the mortality percentage and (spores or extract) concentration, with increasing duration of exposure to the fungus spores, was positive, whereas the larval mortality increased when the fungus, spores or extract, concentration was raised.
