Abstract:Objective To study the expression of vascular endothelial growth factor receptor KDR in gastric cancer and its clinical value.Methods Collected 60 specimens from gastric cancer patients.The expression of KDR in gastric cancer tissues and their adjacent normal tissues were detected with SP immunohistochemistry.Results The positive rate of KDR expression in gastric cancer tissue was higher than that in normal tissue(55.0% vs 18.3%).There was significant difference in the expression of KDR between the gastric cancer tissue and their adjacent normal tissue(P<0.01).Besides,the expression rate of KDR in gastric cancer tissue was correlated with gastric cancer TNM stages(P<0.05).Conclusion KDR is highly expressed in gastric cancer tissues,and it is closely related to the TNM staging of gastric cancer.To detect the expression of KDR in gastric cancer tissues may be helpful to evaluate the prognosis of patients with gastric cancer.
Membrane-bound Ligand-Dependent Bystander Effect of the TNF-related Apoptosis-Inducing Ligand (TRAIL)

Abstract:AIM: To investigate the mechanism of bystander effect caused by TRAIL gene.METHODS: Bystander target cells were transduced with an adenovector expressing the lacZ gene(Ad/Lac-Z),while effector cells were transduced with an adenovector expressing the green fluorescent protein(GFP)/TRAIL fusion gene(Ad/g-TRAIL).Effector and target cells were cocultured in the same well with or without effector and target cell contact using a Transwell plate.RESULTS: A series of different cancer cell lines of different tissue origin were tested for the bystander effect,including human colon cancer cell lines DLD1,human lung cancer cell line A549,human hepatoma cell line Hep G2,human breast cancer cell line MDA-MB231 and human ovarian cancer cell line DOV13.No detectable soluble TRAIL(s-TRAIL) was found from the TRAIL-expressing cell cultures in all of the cell lines.Target cells were killed only when effector and target cells were mixed.The bystander effect and apoptosis induction of TRAIL was dramatically reduced if cells were seeded at a very low density.FACS analysis showed a high apoptosis percentage in high density group in DOV13,morphologically,the effector cells were surrounded by the apoptotic bystander cells.CONCLUSION: The bystander effect of the TRAIL gene is mainly mediated by membrane-bound TRAIL on the surface of transduced cells through cellcell contacts.

Synergistic Anti-Gastric Cancer Effect and Mechanism of Newcastle Disease Virus in Combination with Gastric Cancer Antigen Loaded Dendritic Cells.

Abstract:Objective To investigate the anti-gastric cancer effect and mechanism of cytokine induced killer cells pulsed with gastric cancer antigen loaded dendritic cells(Ag-DC-CIK) in combination with Newcasatle Diease virus(NDV).Methods CIK cells and DC cells were induced and cultured by mononuclear cells from peripheral blood of healthy adult.Human gastric cancer cells SGC-7901 were cultured in vitro and the antigen suspension of gastric cancer cells prepared by repetitive freeze thaw method was used to allergize DC cells.Model of gastric cancer were established in 24 mice and divided into PBS group,NDV group,Ag-DC-CIK group and NDV+ Ag-DC-CIK group on average.Then PBS,NDV,Ag-DC-CIK,NDV+ Ag-DC-CIK were injected into tumors of which respectively.The volume of tumor was measured to calculate the tumor inhibitory rate,the necrosis area scores were noted after HE stain. Results Compared with the NDV group or Ag-DC-CIK group,the average volume of tumor in NDV+Ag-DC-CIK group reduced,while the tumor inhibitory rare and the score of tumor necrosis area increased.Conclusion CIK pulsed with gastric cancer antigen loaded DC combined with NDV have a synergistic effect to kill gastric cancer cells.The mechanism is that NDV can enhance the antigenicity of gastric cancer cells,so as to afford more targes for the CIK cells.
Coexpression of vascular endothelial growth factor and its receptor KDR on gastric adenocarcinoma MGC803 cell line and stimulation of exogenous VEGF 165 to MGC803 cells

Abstract:Vascular endothelial growth factor (VEGF), also known as vascular permeability factor (VPF), is an angiogenic factor playing an important role in tumor growth. VEGF/VPF interacts with endothelial cells by way of two high-affinity receptor tyrosine kinases: flt-1 and KDR. The vast majority of published studies have described expression of the VPF/VEGF receptors specifically in endothelial cells. To elucidate the further function of VEGF in solid tumor development, the coexpression of VEGF and KDR in gastric adenocarcinoma MGC803 cell lines was shown by reverse transcription polymerase chain reaction (RT-PCR). The MGC803 tumor cells could also be strongly immunostained for KDR by immunocytochemistry. It was further demonstrated that exogenous VEGF 165 can stimulate the MGC803 cell growth in both dose-dependent and time-dependent manners by 3 H-thymidine incorporation. Furthermore, anti-VEGF 165 monoclonal antibody and anti-KDR monoclonal antibody could dose-dependently block the VEGF 165 -induced cell growth. These results provided new evidence that VEGF could cause autocrine stimulation to the proliferation of gastric adenocarcinoma cells.
Effective Inhibition Of Cancer Cells By Recombinant Adenovirus Expressing Egfr-Targeting Artificial Microrna And Reversed-Caspase-3

Abstract:The EGFR-targeting cancer therapies are commonly facing drug resistance, mostly due to mutations. Gene therapy with artificial microRNA targeting EGFR conserved sequence may avoid such problem. In this study, we constructed a recombinant adenovirus expressing EGFR-targeting artificial microRNA and active revCASP3 (Ad-EC), under the control of tumor-specific SLPI promoter, and evaluated its inhibitory effect on HEP-2 cancer cells bothin vitroandin vivo. MTT assay showed that cell growth inhibition rate at 72h was 44.0% in Ad-EC group at MOI 50, while the rate was 7.7% in the control virus Ad-GFP group and 3.6% in Cetuximab (500 mu g/ml) group respectively. Flow cytometry analysis revealed the late apoptotic cells rate was 36.1% in Ad-EC group, significantly higher than 6.5% of Ad-GFP group (p< 0.001). When Ad-EC (MOI 50) was combined with CDDP (0.25 mu g/ml), late apoptotic cells rate increased to 61.2%, significantly higher than each monotherapy group (P <0.001). The real-time xCELLigence system recorded an effective cell growth inhibition in Ad-EC and CDDP groups, and more enhanced effect in Ad-EC plus CDDP group. Western blot revealed that Ad-EC could inhibit the activation of AKT pathway and ERK1/2 pathway, while Cetuximab had the AKT pathway over-activated.In vivoexperiments with HEP-2 xenograft in nude mice confirmed the tumor inhibition in Ad-EC, CDDP and Ad-EC plus CDDP groups compared with PBS group (P <0.01). Collectively, these data support the effective inhibition of cancer cells by this novel gene therapy strategy.

Abstract:Objective To study on the effects of epidermal growth factor(EGF) in intervention of gastric mucosa atrophy in early stage.Methods Fifteen rats administered by gastric gavage with 60% alcohol,20mmol/L deoxycholic acid Na-salt pure (DOC-Na) and 0.1% ammonia for 24 weeks were randomly assigned to three groups,rats in intervention group were injected subcutaneously with 10μg/kg weight EGF,one group was injected with normal saline and other group was used as control.Results Comparing to other two groups inflammation grades at gastric body and antrum in EGF group were lower,thickness of lamina propria mucosa,percentage of pyloric gland area to total lamina propria at antrum and number of parietal cell in one intact oxyntic gland at gastric body were higher.Conclusion EGF could intervene gastric mucosa from atrophy in early stage.
The Inhibitory Effect of CD/5-FC System on the Prostate Cancer Cell DU145 and RM-1 in Vitro

Abstract:Objective To study the inhibitory effect of CD/5 FC system on the prostate cancer cell DU145,RM 1 in vitro. Methods In vitro prostate cancer cells DU145 and RM 1 were infected by recombinant adenoviral vector and then the inhibitory effect and the bystander effect of CD/5 FC system were observed. Results CD/5 FC system had a remarkable inhibitory effect on prostate cancer cell DU145 and RM 1 in vitro, the inhibition rates being higher than 97% when infected by AdCMVCD after 4 days added with 15.5mmol/L 5 FC. A strong bystander effect could also be observed, more than 75% tumor cells being killed when only 10% were infected. Conclusions CD/5 FC system may serve as an effective gene therapeutic method.The existence of bystander effect makes itself available in the use of tumor treatment in vivo.
Experimental Study of EGFR Monoclonal Antibody Combined with CIK Cells for the Treatment of Gastric Cancer

Abstract:Background: Monoclonal antibody targeted drugs and cellular immunotherapy have shown great potential in the treatment of malignant tumors,but the therapeutic effect of their combined use on gastric cancer is still unclear.Aims: To investigate the therapeutic effect of epidermal growth factor receptor (EGFR) monoclonal antibody (cetuximab) combined with cytokine-induced killer cells (CIK cells) on gastric cancer by in vitro and in vivo experiments.Methods: The over-expression of EGFR in human gastric adenocarcinoma cell line SGC7901 was confirmed by immunocytochemistry assay,and the effect of EGFR monoclonal antibody on the binding between CIK cells and SGC7901 cells was investigated. The experiment was divided into three groups: EGFR monoclonal antibody combined with CIK cells group,CIK cells group and EGFR monoclonal antibody group.MTT assay was used to measure the killing rate against SGC7901 cells in each group,and the tumor inhibition rate was assessed by the xenograft growth in nude mice.Results: EGFR monoclonal antibody did not significantly improve the binding rate of CIK cells with SGC7901 cells.Compared with CIK cells group and EGFR monoclonal antibody group,both the killing rate and the tumor inhibition rate were significantly higher in EGFR monoclonal antibody combined with CIK cells group (P0.05).Conclusions: EGFR monoclonal antibody combined with CIK cells demonstrates greater therapeutic effect on gastric cancer when compared with their single use.

Results: We found that the use of YN968D1 in CD8 T cells could reduce the expression levels of inhibitory checkpoints, such as Lag-3, PD-1, and Tim3, escalate the production of IFN-γ and IL-2 and promote the cytotoxicity of T cells dramatically in vitro. The transfer of YN968D1-treated T cells achieved better tumor control compared to DMSO-treated T cells or control in both peritoneal dissemination and subcutaneous gastric cancer mouse models. Conclusion: Our results indicate that YN968D1 can enhance the T cell-mediated antitumor immunity.
In vitro antitumor effect of CD, TK fusion suicide gene induced by adenovirus vector

Abstract:Objective To construct a recombinant adenovirus and observe the antitumor effect of the double suicide gene therapy to three different colorectal cancer cell lines. Methods Cytosine deami-nase (CD) and herpes simplex virus thymidine kinase (HSV-tk) fusion gene were transfected into LoVo colon cancer cells, HCT-8 colon cancer cells and HR-834813 rectal cancer cells via an adenovirous vector. Clony forming test and MTT method were used to evaluate the antitumor effect of HSV-tk/GCV (ganci-clovir) system and CD/5-FC(5-fluorocytosine) system against tumor cells. The bystander effect was also observed. Results Western blot analysis demonstrated the fusion protein could be effectively expressed in these three kinds of transfected tumor cells. The plating efficiency of transfection groups and control group was 94, 93, 95 and 96, 92, 94 in the absence of 5-FC and GCV, respectively. There was no statistical significance between them, P 0.05. In the presence of 5-FC(80 mg/L)and GCV(0. 8 mg/L), the survival rate of tumor cells was reduced to 2.1 % , 4. 3 % and 3.2%, significantly different from that before administration ( P 0.01) . Strong bystander effect could be obtained when the proportion of transfected cells in the mixing group was 5 % and the cell killing ratio was increased to 22.2 % , 34.8 % and 40.4 % respectively. Conclusion Significant antitumor effects could be obtained with the combined use of CD, TK fusion gene and double prodrugs.

Abstract:Objective To evaluate the anti-cervical carcinoma cells effects of oncolytic adenovirus d/1520 and Ad-Endo in vitro,and investigate the influence of d/1520 on the Endostatin expression directed by Ad-Endo in cervical carcinoma cells.Methods After cervical carcinoma cells were infected with d/1520 or/and Ad-Endo,the cytopathic effects (CPE) were observed,and the cytotoxicities were measured by MTT assay.Endestatin concentrations were measured by ELISA.Results d/1520 can replicate in cervical carcinoma cells and induce cytopathie effects; In MTT assay,Ad-Endo hardly inhibited the growth of cervical carcinoma cells,while d/1520 inhibited efficiently the proliferation of cervical cancer cells.And the inhibitory rates in Ad-Endo + d/1520 group were higher than that in d/1520 group.Ad-Endo expressed Endestatin protein in cervical cancer cells,while Endostatin concentrations in Ad-Endo+dl1520 group were much higer than those in Ad-Endo group.Conclusions dl1520 can kill cervical carcinoma cells in vitro,and this eytotoxieity can be enhanced by combining with Ad-Endo.The expression of Endostatin directed by Ad-Endo in cervical cancer cells can be dramatically increased by dl1520.

Abstract:PurposeTo study the therapeutic and side effect of CIKs plus DCs on advanced gastric cancers.MethodsPBMCs from patients were cultured in vitro and grew into CIKs and DCs.DCs were co-cultured with CIKs after pulsed with tumor antigens.The cells were returned after 14 days′culturing.ResultsCIK cell-therapy can alleviate symptoms and improve immune functions of the patients.ConclusionCIKs cell-therapy can be an effective adjuvant instrument of the routine therapy of advanced gastric cancers.

Abstract:Previous studies have found that Bifidobacterium infantis-mediated herpes simplex virus-TK/ganciclovir (BF-TK/GCV) reduces the expression of VEGF and CD146, implying tumor metastasis inhibition. However, the mechanism by which BF-TK/GCV inhibits tumor metastasis is not fully studied. Here, we comprehensively identified and quantified protein expression profiling for the first time in gastric cancer (GC) cells MKN−45 upon BF-TK/GCV treatment using quantitative proteomics. A total of 159 and 72 differential expression proteins (DEPs) were significantly changed in the BF-TK/GCV/BF-TK and BF-TK/GCV/BF/GCV comparative analysis. Kyoto encyclopedia of genes and genomes (KEGG) pathway analysis enriched some metastasis-related pathways such as gap junction and cell adhesion molecules pathways. Moreover, the transwell assay proved that BF-TK/GCV inhibited the invasion and migration of tumor cells. Furthermore, immunohistochemistry (IHC) demonstrated that BF-TK/GCV reduced the expression of HIF−1α, mTOR, NF-κB1-p105, VCAM1, MMP13, CXCL12, ATG16, and CEBPB, which were associated with tumor metastasis. In summary, BF-TK/GCV inhibited tumor metastasis, which deepened and expanded the understanding of the antitumor mechanism of BF-TK/GCV.

Therapeutic effect of endostatin-vascular endothelial growth inhibitor recombinant adenoviruses on gastric carcinoma in nude mice

Abstract:AIM: To investigate the effects of recombinant adenoviruses carrying the endostatin-VEGI151 fusion gene AIerapy on gastric carcinoma in nude mice. METHODS: Human gastric carcinoma SGC7901 cells (5×106) were injected sc into the dorsal midline of nude mice (3 weeks old), and 500 μL (1012 TCID50/L) of recombinant adenoviruses AdCA13-hENDO-VEGI151 or AdLacZ was given respectively sc every other day for ten times. Twenty-four hours after the last injection, tumor size, inhibition rate, expression of target gene, proliferating cell nuclear antigen label index (PCNA LI), apoptotic index (AI), and intratumoral microvessel density (MVD) were evaluated respectively after the mice were sacrificed. RESULTS: Compared with the control group AdLacZ (2 356± 1 140 mm 3), the average tumor size of group AdCA13-hENDO-VEGI151, was reduced in size(328±156 mm3, F=12.42, P=0.0125) with an inhibition rate of 86.1 %. The adenoviruses carriyring the target gene could be expressed in gastric carcinoma. The PCNA LI of the treated mice (0.13±0.09 %) was lower than the oontol (1.40±0.53 %, F=22.30, P=0.0 033). The AI was higher in the treated mice (5.09±0.25 %) than in the control (0.61±0.67 %, F=155.13, P=0.0 001). The MVD was lower in the treated mice (0.06±0.03 %) than in the control (1.09±0.76 %, F=7.38, P=0.0 348). CONCLUSION: The recombinant adenovirus carrying fusion gene of hENDO-VEGI151 can express the fuion protein in gastric carcinoma in nude mice and inhibit the tumor growth.

Abstract:AIM: The expression of vascular endothelial growth factor (VEGF) and its receptors KDR and Flt-1 by gastric carcinoma tissues and different gastric carcinoma cell lines was detected to elucidate the molecular mechanism of this growth factor in promoting tumor growth. METHODS: The expression of VEGF, Flt-1 and KDR was determined by reverse transcription-polymerase chain reaction (RT-PCR) in gastric cancer cell lines RF-1, RF-48, AGS-1, NCI-N87, NCI-SNU-1, NCI-SNU-5, NCI-SNU-16 and KATO-III. The expression of Flt-1 and KDR in paraffin-embedded specimens of gastric cancer was determined by immunohistochemistry. The 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay was used to assess the role of VEGF in tumor cell proliferation. RESULTS: All 8 gastric cancer cell lines analyzed expressed VEGF(121) and VEGF(165) and six of them expressed both Flt-1 and KDR, while cell line NCI-SNU-5 expressed Flt-1 only and cell line KATOIII expressed neither Flt-1 nor KDR. The gastric carcinoma tissues expressed Flt-1 and KDR widely, with the positive rate of expression of Flt-1 and KDR being 84.6% and 70% respectively. The exogenous VEGF stimulated the growth of KDR-positive cell lines NCI-N87 and AGS-1 in a dose-dependent manner but exhibited no effect on the growth of KDR-negative cell line NCI-N87. CONCLUSION: VEGF and its receptors KDR and Flt-1 were expressed widely in gastric carcinoma cells and the VEGF stimulated KDR-positive tumor cell growth directly. These results suggest that VEGF may play a role in promoting tumor growth and metastasis by participating in both paracrine and autocrine pathways.
The Adenovirus Vector-mediated Expression of EMC6 Inhibits Migration of Gastric Cancer Cells via Down-regulating the FAK Signaling Pathway

Abstract:Endoplasmic reticulum membrane protein complex subunit 6 (EMC6),also known as transmembrane protein 93 (TMEM93),is a novel human autophagy-related molecule identified in our laboratory.The EMC6 gene is conserved and expressed widely in a variety of tissues and organs.However,under abnormal conditions the expression of EMC6 was reduced or absent e.g.in gastric cancer tissues.In this report,we constructed a recombinant adenovirus 5-EMC6 (Ad5-EMC6) vector and evaluated its expression and anti-tumor activities in BGC823 and SGC7901 gastric cancer cells.Colony formation analysis revealed that the adenovirus increased the production of EMC6 protein and inhibited tumor cell growth.The ability of migration and invasion was also reduced in cells infected by Ad5-EMC6.Further studies suggested that Ad5-EMC6 could decrease the phosphorylation levels of FAK at Y576/577,as well as the levels of MMP2 and MMP9 proteins.Data obtained from an animal model of peritoneal metastasis proved that the number of metastatic nodules in the peritoneal cavity was significantly decreased or absent in the Ad5-EMC6 group compared with the Ad5-Null group.Collectively,these data suggested that Ad5-EMC6 mediated the inhibition of the FAK signaling pathway,and it may be one of the mechanisms of its anti-tumor activity.
The Clinical Study on Application of Using a Novel Blockade Technique for Gastric Cancer to Decrease Blood-Borne Metastasis of Cancer Cells

Abstract:Objective To evaluate the effect of a novel blockade technique for gastric cancer on blood-borne metastasis of gastric cancer cells to portal vein. Methods Twenty-three cases of gastric cancer were divided into routine operation group(8 cases intraoperatively without blockade technique) and blockade group (15 cases with blockade technique). Blood samples from portal vein pre- and intraoperatively, as well as gastroepiploic vein limited within the blockade area were obtained to detect CK19 mRNA expression by using RT-PCR technique. Results Before the dissection of gastric lesion, the overall positive rate of CK19 mRNA expression in portal vein blood is 34.7%(9/23), including 37.5%(3/8) in routine operation group and 33.3%(5/15) in blockade group. While the course of tumor resection, those positive rates were 87.5%(7/8) in routine operation group and 6.7%(1/15) in blockade group respectively( P 0.05). CK19 mRNA expression in the right gastroepiploic venous blood limited within the blocking area was all positive in 15 cases of blockade group. Conclusion This blockade technique can be used effectively to block the intraoperative spread of gastric cancer cells, thus prevent blood-borne metastasis due to operative manipulation.

Abstract:Background: Radiation resistance remains the leading cause of radiotherapy (RT) failure. The development of tumor-specific targeted sensitizers is key to overcoming radiation resistance. Our early data showed that cancer cell penetration was simulated by internalizing arginine-glycine-aspartic acid (iRGD), and the irradiation efficacy was improved. The present study aims to design and fabricate iRGD-modified red blood cell (RBCs) for tumor targeting and RT enhancement, and to evaluate its safety and efficacy in vivo.Methods: 1,2-Distearoyl-sn-glycero-3-phosphoethanolamine-poly ethylene glycol-iRGD (DSPE-PEG-iRGD) was used to modify RBCs by a lipid-insertion method without direct chemical bioconjugation. Fluorescent dyes were used to trace the functional RBCs through confocal microscopy examination. In vitro stability evaluation was performed using cell culture medium incubation for 48 h followed by fluorescence decay assay. Furthermore, a subcutaneous cancer cell mouse model was constructed with MKN-45 cells for target efficacy and RT enhancement evaluation with DSPE-PEG-iRGD-modified RBCs (RBC-iRGD).Results: Successful construction of RBC-iRGD was verified by the presence of the yellow fluorescence, and an approximately 108 iRGD molecules were labeled on a single RBC. The final RBC-iRGD showed good stability without any hemolytic effects in the cell culture medium. Moreover, higher fluorescence intensity and decreased liver and spleen accumulation could be observed in RBC-iRGD compared to RBC + iRGD in vivo. The RBC-iRGD exerted enhanced radiosensitivity in subcutaneous gastric tumor mice.Conclusions: The RBC-iRGD exerted good tumor-targeting efficacy and favorable effects for RT enhancement in vivo.
