Abstract:Introduction: Gastric cancer is the second most common cause of cancer-related mortality worldwide. 5-fluorouracil (5-FU) is a commonly used anti-cancer drug. Various polyunsaturated fatty acids (PUFAs) are known to have tumoricidal action both in vitro and in vivo. Though PUFAs are known to augment the cytotoxic action of anti-cancer drugs, the exact mechanism is not clear.Material and methods: The human gastric cancer cell line MGC (undifferentiated) and human gastric cancer cell line SGC (semi-differentiated) were either 5-FU alone or a combination of 5-FU + PUFAs and their proliferation, and ability to secrete tumor necrosis factor-alpha (TNF-alpha) and vascular endothelial growth factor (VEGF) and lipid metabolism-related factors lipoprotein lipase (LPL), peroxisome proliferator-activated-gamma (PPAR-gamma), and CCAAT enhancer-binding protein (C/EBP) were investigated and analyzed.Results: It was noted that combined treatment of 5-FU + PUFAs on gastric carcinoma (MGC and SGC) cells produced a significant growth inhibitory action compared with either agent alone by inhibiting the production of TNF-alpha and VEGF and a simultaneous increase in the expression of LPL, PPAR-gamma, and C/EBP.Conclusions: Based on the results of the present study, it is concluded that PUFAs enhance the tumoricidal action of the anti-cancer drug 5-FU by acting on anti-angiogenic factors and enzymes involved in lipid metabolism.
[RAD001 Reverses Radio-Resistance and Exerts Synergistic Tumor Inhibition with 5-Fluorouracil].

Abstract:OBJECTIVE To explore the in vitro synergistic anti-tumor efficacy of mammalian target of rampamycin (mTOR) inhibitor (RAD001) and 5-fluorouracil (5-FU) for radio-resistant tumors. METHODS Radio-resistant cells of human pancreatic cancer cell and human colon cancer cell were established. The expression profiles of VEGF (vascular endothelial growth factor) and TP (thymidine phosphorylase) were compared between parental and radio-resistant tumor cells. The tumor proliferation was analyzed after 5-FU alone or in combination with a mTOR inhibitor. RESULTS After several cycles of radiation induction (3 Gy), the radio-resistant cells of human pancreatic cancer (AsPCres) and colon cancer (HT29res) were established. There was a higher expression of VEGF in radio-resistant tumor cells than their parental cells. They were 1215 ± 67 pg/ml in AsPCres and 689 ± 25 pg/ml in HT29res respectively (P < 0.01). The up-regulation of TP was observed in both AsPC-res and HT29-res. The combined therapy of 5-FU plus a mTOR inhibitor might exert synergistic tumor inhibition. CONCLUSION RAD001 decreases the radiation-induced expression of VEGF in tumor. And the post-radiation up-regulation of TP promotes the efficacy of 5-FU. The combined therapy of RAD001 and 5-FU may inhibit synergistically the growth of radio-resistant tumors.
THE SYNERGISTIC ANTI-PROLIFERATIVE EFFECT OF CURCUMIN AND FLUOROURACIL ON HUMAN COLON CANCER CELL LINE HT-29

Abstract: Objective : To demonstrate whether the suppression of cyclooxygenase-2 (COX-2) expression by curcumin can increase the anti-colon-cancer effectiveness of fluorouracil (FU). Method:The combination effect was quantitatively determined by using the method of median-effect principle and the combination index. The COX-2 expression inhibition after treatment of the combination of curcumin and FU was also evaluated by Western blot analysis. Results: The IC50 values in the HT-29 cells for curcumin and FU were 15.9±1.96, and 17.3±1.85 μmol/L respectively. When curcumin and FU were jointly used, the synergistic inhibition of growth was quantitatively demonstrated at higher effect levels. And this synergism was associated with decreased expression of COX-2 protein. Conclusion: Curcumin can enhance the chemotherapy effect of FU, and this combined treatment may have clinical implications in the treatment of colon cancer.
Combined Effect of 5-Fluorouracil and Recombinant Tumor Necrosis Factor Against Human Gastric Carcinoma Cell Lines.

Abstract:This study was performed to investigate the direct effects of recombinant human tumor necrosis factor (rH-TNF) and recombinant human interleukin-2 (rH-IL-2), either alone or in combination, on the cytotoxicity of 5-FU measured by MTT assay against human gastric adenocarcinoma cell lines (MKN-28 and MKN-45), and also to determine the optimal schedule for their combination. The antitumor activity of rH-TNF was enhanced more than 42% by 10(2) U/ml of rH-IL-2. The enhancing effects of rH-TNF and rH-IL-2 on the cytotoxicity of 5-FU were evaluated in terms of Modification Index(MI), the MI value at 10 U/ml rH-TNF was 1.6; the MI at the same concentration of rH-TNF in the presence of 10(2) U/ml of rH-IL-2 was 2.1. These results demonstrated that the antitumor effect of 5-FU was enhanced 1.6 times by 10 U/ml of rH-TNF and further enhanced by the combined use of rH-TNF and rH-IL-2. The combined effect of equal concentrations of 5-FU and rH-TNF was superior or equivalent to that of 5-FU or rH-TNF alone. The sequence of 5-FU followed by rH-TNF and rH-IL-2 showed a higher inhibitory effect than the reverse sequence. This sequence combination seems worthy of further consideration for the treatment of gastric cancer.
Melatonin Synergizes the Chemotherapeutic Effect of 5‐fluorouracil in Colon Cancer by Suppressing PI3K/AKT and NF‐κB/iNOS Signaling Pathways

Abstract:5-Fluorouracil (5-FU) is one of the most commonly used chemotherapeutic agents in colon cancer treatment, but has a narrow therapeutic index limited by its toxicity. Melatonin exerts antitumor activity in various cancers, but it has never been combined with 5-FU as an anticolon cancer treatment to improve the chemotherapeutic effect of 5-FU. In this study, we assessed such combinational use in colon cancer and investigated whether melatonin could synergize the antitumor effect of 5-FU. We found that melatonin significantly enhanced the 5-FU-mediated inhibition of cell proliferation, colony formation, cell migration and invasion in colon cancer cells. We also found that melatonin synergized with 5-FU to promote the activation of the caspase/PARP-dependent apoptosis pathway and induce cell cycle arrest. Further mechanism study demonstrated that melatonin synergized the antitumor effect of 5-FU by targeting the PI3K/AKT and NF-B/inducible nitric oxide synthase (iNOS) signaling. Melatonin in combination with 5-FU markedly suppressed the phosphorylation of PI3K, AKT, IKK, IB, and p65 proteins, promoted the translocation of NF-B p50/p65 from the nuclei to cytoplasm, abrogated their binding to the iNOS promoter, and thereby enhanced the inhibition of iNOS signaling. In addition, pretreatment with a PI3K- or iNOS-specific inhibitor synergized the antitumor effects of 5-FU and melatonin. Finally, we verified in a xenograft mouse model that melatonin and 5-FU exerted synergistic antitumor effect by inhibiting the AKT and iNOS signaling pathways. Collectively, our study demonstrated that melatonin synergized the chemotherapeutic effect of 5-FU in colon cancer through simultaneous suppression of multiple signaling pathways.
Synergistic effect of 5-fluorouracil and the flavanoid oroxylin A on HepG2 human hepatocellular carcinoma and on H22 transplanted mice

Abstract:Aim: To investigate the synergistic inhibitory effects of the combination of 5-fluorouracil (5-FU) with the natural flavanoid oroxylin A on human hepatocellular carcinoma cells HepG2 in vitro and on transplanted murine hepatoma 22 (H22) tumors in vivo and the preliminary mechanisms. Methods: The inhibitory effects of 5-FU combined with the natural flavanoid oroxylin A in vitro were detected by MTT assay and the effects in vivo were investigated by transplanted H22 mice model. DAPI staining and Annexin V/propidium iodide (PI) double staining were used to detect the cell morphological changes and apoptosis. The mRNA levels of thymidine synthetase (TS) and dihydropyrimidine dehydrogenase (DPD) in HepG2 cells after oroxylin A and 5-FU combination treatment were observed by quantitative real-time PCR. Western blotting assay was used to reveal the expressions of apoptotic-inducing proteins P53, cleaved PARP, COX-2, Bcl-2, and pro-caspase3. Results: Oroxylin A in combination with 5-FU presented synergistic effect (CI<1) on HepG2 cells in vitro when the inhibitory rate was higher than 7.5%. The inhibitory rate on H22 murine solid tumor in vivo in the combination group was higher than monotherapy. 5-FU combined with oroxylin A exerted stronger apoptotic induction in HepG2 cells than either single drug treatment. Quantitative real-time PCR discovered the downregulation of TS mRNA and DPD mRNA in HepG2 cells after combination treatment. Western blotting assay revealed oroxylin A enhanced 5-FU-induced apoptosis in HepG2 cells by elevating the expressions of apoptotic-inducing proteins P53 and cleaved PARP and decreasing the expression of apoptotic-inhibitory proteins COX-2, Bcl-2, and pro-caspase3. Conclusion: The anti-hepatocellular carcinoma effects in vitro and in vivo of 5-FU and oroxylin A combinations were synergistic and oroxylin A increased the sensitivity of HepG2 cells to 5-FU by modulating the metabolic enzymes of 5-FU and apoptotic-related proteins.
Luteolin synergizes the antitumor effects of 5-fluorouracil against human hepatocellular carcinoma cells through apoptosis induction and metabolism

Abstract:Aims: Some compounds derived from Chinese medicine have demonstrated great prospective roles in sensitization to chemotherapy. This study aimed to investigate the combination of luteolin and 5-fluorouracil on proliferations of hepatocellular carcinoma cells and the potential mechanisms. Main methods: The antitumor effects of luteolin, 5-fluorouracil, and their combinations were detected by 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium/phenazine methosulfate assay, and isobole method was used to evaluate drug combinations. CellTiter-Blue and Caspase-Glo 3/7 assay were used for assessment of cell viability and apoptosis after treatment with luteolin, 5-fluorouracil and their combinations. Cell cycle distributions and apoptosis were detected by PI staining, Hoechst 33342 staining and FITC-Annexin V/PI staining. Bcl-2, bax, p53 and PARP expressions were determined by Western blot. Furthermore, mRNA levels of 5-fluorouracil metabolism related enzymes were detected by RT-PCR. Key findings: Drug combination study showed that luteolin could synergize the antitumor effects of 5-fluorouracil at different dose ratios (luteolin: 5-fluorouracil=10:1, 20:1, 40:1) against HepG2 and Bel7402 cells. Cell viability and cell apoptosis analysis showed that the synergistic growth inhibition caused by combined luteolin and 5-fluorouracil was closely related to apoptosis. Further mechanism studies showed that the synergistic effects of drug combinations were related with enhanced bax/bcl-2 ratios and p53 expressions, and induced PARP cleavage. Also, combined luteolin and 5-fluorouracil could significantly decrease the dihydropyrimidine dehydrogenase. Significance: These results showed that luteolin could synergize the antitumor effects of 5-fluorouracil on HepG2 and Bel7402 cells, which might be related with apoptosis and regulation of 5-fluorouracil metabolism.

Abstract:Dimethoxycurcumin (DMC) is a lipophilic analog of curcumin, an effective treatment for colon cancer, which has greater chemical and metabolic stability. Chemotherapy treatments, such as 5-fluorouracil (5-Fu), play a key role in the current management of colon cancer. In this study, we investigated the antitumor efficacy of DMC in combination with 5-Fu in SW480 and SW620 colon cancer cells. CCK-8 assay was used to evaluate the inhibitory effect of DMC and 5-Fu on cancer cells proliferation, and the combination index was calculated. The influence of DMC and 5-Fu on cell cycle, apoptosis, reactive oxygen species (ROS) production, and mitochondrial membrane potential in SW480 and SW620 cells was determined using flow cytometry, and the related signaling pathways were detected by western blot. Transmission electron microscopy was used to observe endoplasmic reticulum expansion. DMC- and/or 5-Fu-induced apoptosis, stimulated G0/G1 phase arrest, increased ROS levels, decreased mitochondrial membrane potential, and enhanced endoplasmic reticulum expansion. The induction of apoptosis is involved in the increasing of Bax and cytochrome c and decreasing of Bcl2 expressions. Increased production of ROS was accompanied by upregulation of CHOP and Noxa. Combination therapy of DMC and 5-Fu had increased efficacy on the above pathways compared with either drug alone. Based on the calculated IC50 , combination treatment with DMC and 5-Fu had an additive antitumor effect in both cell lines. Combined treatment with DMC and 5-Fu led to an additive antitumor effect in colon cancer cells that was related to apoptosis induction, G0/G1 phase arrest, increased ROS production, decreased mitochondrial membrane potential, and enhanced endoplasmic reticulum expansion.

Abstract:Objective:To assess the synergistic interaction between CLFF (Chong Lou Fu Fang) and 5-fluorouracil (5-FU) in gastric cancer cells,and to investigate the influence of CLFF on thymidylate synthase gene expression and cell apoptosis.Methods:The cell proliferation capacity was determined by MTT assay.Interaction between CLFF and 5-FU in human gastric cancer SGC-7901 and BGC-823 cells was evaluated using the combination index (CI) method based on median-effect principle.Apoptosis rates of cancer cells were evaluated by means of flow cytometry.Expression of thymidylate synthase gene with or without CLFF treatment were measured by real time quantitative PCR.Results:CLFF had a synergistic effect on the cytotoxicity of 5-FU in both two gastric cancer cell lines.The combination of CLFF and 5-FU could also induce apoptosis in a synergistic manner.TS mRNA levels in both cells were markedly suppressed at (0.30±0.03) and (0.46±0.03)fold,respectively,by the presentation of CLFF for 24h.Compared with the levels before CLFF treatment,the difference was significant.Conclusion:The combination of CLFF and 5-FU showed synergistic antitumor activity in both two gastric cancer cell lines.The possible mechanisms might be the synergistic apoptotic effect and the downregulation of thymidylate synthase gene.

Anti-Cancer Agents in Medicinal Chemistry

Abstract:BACKGROUNDA Chinese folk medicine plant Pleurospermum lindleyanum possesses pharmacological activities of heat-clearing, detoxifying and preventing from hepatopathy, coronary heart disease, hypertension, and high altitude sickness. We isolated and characterized its constituents to investigate its synergistic effects against human hepatoma SMMC-7721 cells. Objective: The aim of this study was to explore the synergistic anti-cancer activities of isolates from P. lindleyanum with 5-FU on hepatoma SMMC-7721 cells in vitro and their primary mechanisms. Methods: Sequential chromatographic techniques were conducted for the isolation studies. The isolates structures were established by spectroscopic analysis as well as X-ray crystallographic diffraction. Growth inhibition was detected by MTT assay. The isobologram method was used to assess the effect of drug combinations. Flow cytometry and western blot were used to examine apoptosis and protein expression. Results: A new coumarin (16), along with sixteen known compounds, were isolated from the whole plant of P. lindleyanum and their structures were elucidated by spectroscopic methods. Four coumarins (2, 3, 5, and 16), two flavonoids (8 and 9) and three phytosterols and triterpenes (12-14) were found to synergistically enhance the inhibitory effect of 5-FU against SMMC-7721 cells. Among them, compounds 3 and 16 exhibited the best synergistic effects with IC50 of 5-FU reduced by 16-fold and 22-fold possessing the minimum Combination Index (CI) 0.34 and 0.27. The mechanism of action of combinations might be through synergistic arresting for the cell cycle at G1 phases and the induction of apoptosis. Moreover, western blotting and molecular docking revealed that compounds 3 or 5 might promote 5-FU-induced apoptosis by regulating the expression of Caspase 9 and PARP. Conclusion: Constituents from P. lindleyanum may improve the treatment effectiveness of 5-FU against hepatocellular carcinoma cells.
Enhancement of anti-tumor effects of 5-fluorouracil on hepatocellular carcinoma by low-intensity ultrasound

Abstract:Background Hepatocellular carcinoma (HCC) accounts for 75 % of liver cancers and is the second most lethal cancer, associated with its multiple etiologies, poor prognosis and resistance to chemotherapy drugs. Chemotherapy treatment on HCC suffers low efficacy of drug uptake and can produce a range of side effects. Here we report an investigation on the effect of a combined treatment on human hepatocellular carcinoma BEL-7402 cells using low-intensity ultrasound (US) and 5-fluorouracil (5-FU). Methods The uptake of 5-FU was measured by the high-performance liquid chromatography (HPLC). DNA damage was detected by the comet assay. MTT assay was used to examine cell viability. Intracellular reactive oxygen species (ROS) and mitochondrial membrane potential (Δψm) were respectively detected by the fluorescent probes DCFH-DA or JC-1. Endogenous apoptosis-associated proteins were analyzed by the western blot and immunohistochemistry. Histopathological changes were evaluated by the hematoxylin and eosin (H&E) staining. Cell apoptosis was evaluated by the TUNEL and flow cytometry assays. Cell proliferation was measured using the immunohistochemical staining of PCNA. Results Our results showed that low-intensity US (1.1 MHz, 1.0 W/cm 2 , 10 % duty cycle) significantly enhanced the uptake of 5-FU, 5-FU-mediated DNA damage and reactive oxygen species (ROS) generation. The increased ROS production up-regulated the p53 protein level, which led to the up-regulation of Bax and down-regulation of Bcl-2. The enhancement of ROS generation and the activation of the apoptosis-associated proteins further triggered the collapse of mitochondrial membrane potential, released cytochrome c from mitochondria into cytosol and activated the mitochondria-caspase pathway, and cell apoptosis. Such enhanced effects could be partially blocked by the ROS scavenger N-acetylcysteine (NAC). Overall, low-intensity US combined with 5-FU led to an effective inhibition of tumor growth and prolonged overall survival of BEL-7402 HCC-bearing nude mice by more than 15 % compared with 5-FU treatment alone. Conclusions Our results showed that low-intensity ultrasound combined with 5-FU produced much enhanced synergistic anti-tumor effects via enhanced ROS production in treating HCC.
Enhanced Anticancer Activity of 5-Fu in Combination with Bestatin: Evidence in Human Tumor-Derived Cell Lines and an H22 Tumor-Bearing Mouse

Abstract:The clinical use of 5-fluorouracil (5-FU) is increasingly limited by low response rates, adverse reactions, and toxicity. A drug combination offers a new strategy for appropriate use of 5-FU. Bestatin, an aminopeptidase N (APN) inhibitor, has been used as an adjuvant chemotherapy drug because of its actions to suppress tumorigenesis and invasion. The current study evaluated the anticancer efficacy of 5-FU plus Bestatin at the cellular and animal level. The combination killed more colonic cancer, hepatic carcinoma, and ovarian cancer cells and fewer nonmalignant human embryonic kidney (HEK293) and Chinese hamster ovary (CHO) cells than 5-FU or Bestatin alone. Moreover, 41.58% of ES-2 and 20.86% of PLC/PRF/5 cell apoptosis was caused by the combination of the two, while 5-FU caused apoptosis of 20.86% of ES-2 cells and 8.85% of PLC/PRF/5 cells. The cell cycle was arrested in the S and G0/G1 phases when a combination of the two was used. In an experiment involving mice bearing tumors, a combination of the two had a rate of tumor inhibition of 61.98%, while 5-FU alone had a rate of tumor inhibition of just 49.17%. In addition, the combination of the two was safer than either drug alone and did not cause weight loss or death. In conclusion, combining 5-FU and Bestatin could enhance the anticancer activity of 5-FU and decrease its cytotoxicity. These results suggest that 5-FU plus Bestatin has greater efficacy as a tumor therapy.
Parthenolide Augments the Anticancer Effects of 5-Fluorouracil on Cell Apoptosis and Decreased Nf-Kappa B Activity in A Gastric Cancer Cell Line

Abstract:5-Fluorouracil (5-FU) is not well tolerated by some patients, because of its severe side effects. The aim of the present study was to investigate the effect of parthenolide (PTL) in augmenting the anti-tumor effect of 5-FU in gastric cancer cells KATO III and to determine if parthenolide can be used to decrease the dose of 5-FU. The mechanism underlying the anti-tumor effects of 5-FU was also investigated. Using flow cytometry, we analyzed apoptosis of KATO III cells. Growth proliferation of the cell line was assayed by MTT. PGE(2) protein level was detected by ELISA and western blot analysis was used to detect the expression of the NF-kappa B/p65, COX-2, caspase-3 and caspase-9 proteins. We found that either 5-FU or PTL alone induced apoptosis of KATO III cells in a concentration-dependent manner; however, combined treatment with both 5-FU and PTL significantly improved apoptosis. MTT data demonstrated that KATO III cells were more sensitive to 5-FU treatment in the presence of PTL. The combination of 5-FU with PTL also resulted in a significant reduction of PGE(2) secretion, compared with the control. Expression of NF-kappa B/p65 and COX-2 was inhibited, whereas, caspase-3 and caspase-9 protein were upregulated. In conclusion, it is rational to include the NF-kappa B inhibitor PTL in regular 5-FU-based chemotherapy, which may not only allow a reduction in the dose of 5-FU to prevent adverse effect, but also enhance the chemotherapeutic effect of 5-FU in gastric cancer.
α‐Phellandrene enhances the apoptosis of HT‐29 cells induced by 5‐fluorouracil by modulating the mitochondria‐dependent pathway

Abstract:α‐Phellandrene (α‐PA), a natural constituent of herbs, inhibits cancer cell viability and proliferation. 5‐Fluorouracil (5‐FU) is a frequently utilized chemotherapeutic medicine for the treatment of colon cancer, which works by triggering cancer cell apoptosis. The present study examined how the combination of α‐PA and 5‐FU affects the suppression of human colon cancer cells by promoting apoptosis. The impact of this treatment on cell viability, apoptosis, and the expression levels of Bcl‐2 family members, caspase family members and mitochondria‐related molecules in HT‐29 cells was assessed by the MTT assay, immunocytochemistry, western blotting and quantitative PCR. The combination of 5‐FU and α‐PA had a synergistic inhibitory effect on cell viability, as determined by assessing the combination index value. Bax protein expression levels were higher in the 50, 100 or 250 μM α‐PA combined with 5‐FU groups compared with those in the 5‐FU alone group (P<0.05). By contrast, Bcl‐2 protein expression levels and mitochondrial membrane potential (MMP, ΔΨm) were lower in the 100 or 250 μM α‐PA combined with 5‐FU groups than those in the 5‐FU alone group (P<0.05). In addition, hexokinase‐2 (HK‐2) protein expression levels were lower in the 50, 100 or 250 μM α‐PA combined with 5‐FU groups than those in the 5‐FU alone group (P<0.05). Compared with 5‐FU alone, after HT‐29 cells were treated with 50, 100 or 250 μM α‐PA combined with 5‐FU, the mRNA expression levels of extrinsic‐induced apoptotic molecules, including caspase‐8 and Bid, were higher (P<0.05). Treatment with 50, 100 or 250 μM α‐PA combined with 5‐FU also increased the mRNA expression levels of cytochrome c, caspase‐9 and caspase‐3, regulating intrinsic apoptosis (P<0.05). These results showed that α‐PA and 5‐FU had a synergistic effect on reducing the viability of human colon cancer HT‐29 cells by inducing extrinsic and intrinsic apoptosis pathways. The mechanism by which apoptosis is induced may involve the intrinsic apoptosis pathway that activates the mitochondria‐dependent pathway, including regulating the expression levels of Bcl‐2 family members, including Bax, Bcl‐2 and Bid, regulating MMP and HK‐2 expression levels, and increasing the expression of caspase cascade molecules, including caspase‐9 and caspase‐3. In addition, it may involve the extrinsic apoptosis pathway that activates caspase‐8 and caspase‐3 leading to apoptosis.

Weekly Paclitaxel and High-Dose 5-Fluorouracil Plus Leucovorin in Hormone-Refractory Prostate Cancer: in Vitro Combined Effects and a Phase II Trial

Abstract:Purpose: Paclitaxel and 5-fluorouracil have been used to treat hormone-refractory prostate cancer with some success. In vitro data suggest that the combined cytotoxicity may be sequence dependent. Thus, we explored the combined effects of the 2 agents, both in vitro and in vivo.Patients and Methods: The combined cytotoxicity of paclitaxel and 5-fluorouracil, and the possible schedule dependence were studied in vitro using PC-3 and DU 145 cells and the microculture tetrazolium assay. There were 23 patients with hormone-refractory prostate cancer treated with the regimen T-HDFL: paclitaxel 90 mg/m(2) intravenously I hour on days I and 8; 5-fluorouracil 2000 mg/m2; and leucovorin 300 mg/m(2) intravenous 24-hour infusion on days 2 and 9, which repeated every 21 days. The allowed percentage of bone marrow irradiation was 50%.Results: Significant synergistic cytotoxicity was seen only when paclitaxel was given 24 hours before 5-fluorouracil. With the T-HDFL regimen, 11 (52%) of the 21 evaluable patients had >= 50% reduction of prostate-specific antigen, lasting for 6 weeks. Of the 7 patients with measurable disease, 2 had a partial response. Median overall survival was 14.1 months. Grade III/IV leukopenia occurred in 2 patients. There was no treatment-related death. Toxicities were well tolerated.Conclusions: The combined cytotoxicity of paclitaxel and 5-fluorouracil is schedule dependent. It is feasible to administer weekly paclitaxel and high-dose 5-fluorouracil infusions in patients with hormone-refractory prostate cancer. Herein, we found that H1 (a derivative of Tetrandrine) exerted a potent anti-MDR effect on growth of 5-FU resistant HCC cells (Be17402/5FU). The resistant fold (RF) of 5-FU is over 160-fold, while the RF of H1 is only 4.8-fold in Be17402/5-FU cells. Further studies demonstrated that blockage of STAT3/MCL-1 signaling and induction of PUMA is responsible to anti-MDR activity of H1. Moreover, combination of H1 and 5-FU (Ratio = 1:2) could synergistically induce apoptosis of Be17402/5-FU cells. Co-treatment of H1 enhances the suppression of p-STAT3 and MCL-1, and significantly increases PUMA expression. Finally, the combination of H1 and 5-FU results in an increase of cleaved PARP. Taken together, H1 effectively improve the cytotoxic effect of 5-FU against Be17402/5-FU cells via blocking STAT3/MCL-1 pathway and inducing PUMA. Our findings suggested that combination 5-FU with anti-MDR agents might present a novel strategy to enhance the therapeutic efficacy of 5-FU in resistant HCC. (C) 2019 Elsevier Inc. Synergistic antitumor effect of puerarin combined with 5-fluorouracil on gastric carcinoma

Abstract:Combination chemotherapy is a crucial method in the treatment of gastric cancer. The aim of the present study was to investigate the inhibitory effects of puerarin and 5‑fluorouracil (5‑FU) on BGC‑823 gastric cancer cells in vitro and in vivo. The in vitro growth inhibition of puerarin or 5‑FU alone or combined on BGC‑823 cells was determined using a cell counting kit 8 (CCK‑8) on living cells. Apoptotic morphological features and proteins expression levels were detected by Hoechst 33258 staining, an Annexin V/propidium iodide apoptosis kit and western blot analysis, respectively. Tumor xenografts were established in nude mice and the inhibitory effects and side effects were detected. Results of the CCK‑8, Hoechst 33258 staining and flow cytometry revealed that the combined treatment was more effective than the separate treatments. The tumor volume was 90.65% of that of the controls and the mean tumor weight was only 0.125 g at the end of the experiment in the combination group compared with the control group (0.822 g). In addition, it was determined that liver and renal toxicity did not increase in combined treatment. These findings showed that puerarin and 5‑FU produced a significant synergic effect on gastric cancer cells, while there was no increase in side effects.

Abstract:It was found that aerobic strategies combined with multiple nutrient limitations produced greater quantities of polyhydroxyalkanoates (PHAs) than strategies relying on oxygen limitation (either microaerophilic or anaerobic/aerobic). This was true both for a synthetic wastewater composed of acetic and propionic acid, and also for a nutrient deficient industrial wastewater. PHA/substrate yields were shown to be comparable to axenic systems for many operating strategies analyzed, and it was found that PHA composition could be affected by process operational conditions. The molecular weight and melting point of the PHA produced were found to be in a desirable range with respect to material properties, which have not been well studied in the previous literature for mixed cultures (Salehizadeh and Van Loodsrecht, 2004). The effects of process staging, multiple treatment cycles, and inocula source were also addressed.
Experience of using local flaps to cover open lower limb injuries at an Indian trauma center

Abstract:Background: Optimal care of open, high-velocity, lower limb injury requires surgical skills in debridement, skeletal stabilization, and in providing appropriate soft tissue cover. Timely coordination between orthopedic and plastic surgeons, though ideal, is often difficult. In our center, orthopedic surgeons undertake comprehensive treatment of open fractures including soft tissue cover. We reviewed the results of the local flaps of lower limb, done by orthopedic surgeons. Materials and Methods: We retrospectively reviewed the results of the lower limb flaps done between January 2005 and December 2006. All flaps done at and below the level of knee were included. Results: There were 105 patients with 120 flaps during this period. Two patients with two flaps were lost to follow-up. The average age was 32 years. Sixty-four patients had Type IIIB Gustilo and Anderson injuries. Thirty-nine patients had isolated soft tissue injuries. The indications for flaps were exposed bone, tendon, and joint in 45, 11, and 12, respectively, or a combination in 35 patients. The flaps done were 51 reverse sural artery, 35 gastrocnemius, 25 local fasciocutaneous, and seven foot flaps. The flap dimensions ranged from 2 × 2 to 30 × 15 cm. Ninety-three flaps (79%) healed primarily. Among 25 flaps (21%) with necrosis, 14 flaps required secondary split skin graft for healing, while the other nine flaps healed without further surgery. Conclusion: Appropriate soft tissue cover provided by orthopedic surgeons can help in providing independent, composite care of lower limb injuries.
