Abstract:The blue-green algae Anabaena flos-aquae and Nostoc muscorum may reduce acetylene to ethylene most actively at p O 2 levels below 0.2 atm. High p O 2 levels inhibit acetylene reduction, nitrogen fixation, respiration and 14 CO 2 fixation in A . flos-aquae . The effect is not solely via an inhibition of nitrogenase activity because inhibition of 14 CO 2 fixation by a species of Phormidium which does not fix nitrogen, and by A . flos-aquae grown on combined nitrogen also occurs. The inhibition of acetylene reduction in Anabaena is reversible in short-term experiments, the rate of recovery being rather similar irrespective of the p O 2 level to which the alga was subjected initially. 3(3,4-dichlorophenyl)-1-1-dimethyl urea (DCMU) at a concentration of 3 x 10 -5 M, which completely inhibits oxygen evolution and acetylene reduction but not respiration by aerobically grown Anabaena cultures also inhibits acetylene reduction in Na 2 S-grown cultures. Salicylaldoxime at a concentration of 10 -4 M which partially inhibits electron flow from photosystem II inhibits acetylene reduction to a greater extent under aerobic conditions than in the presence of Na 2 S. The presence of Na 2 S also results in the removal of free oxygen from the medium in the pH range at which Anabaena normally grows. The data suggest that under our conditions (1) the photolysis of water, or photosystem II is essential for the growth of A. flos-aquae in the presence of H 2 S ; (2) H 2 S may provide electrons when reducing power from the photolysis of water is reduced but not inhibited completely; (3) H 2 S prevents an accumulation of oxygen in the medium during photosynthesis. These physiological findings from the laboratory may help to explain why blue-green algae live not only in aerobic environments but also in habitats where reducing conditions may prevail.
Chloroplast Intragenic Suppression Enhances the Low CO2/O2 Specificity of Mutant Ribulose-bisphosphate Carboxylase/Oxygenase

Abstract:The competition between CO2 and O2 at the active site of ribulose-1,5-bisphosphate carboxylase/oxygenase limits net CO2 fixation in photosynthesis. In the green alga Chlamydomonas reinhardtii, a mutation in the chloroplast large-subunit gene reduces the CO2/O2 specificity of the enzyme by 37% and causes valine-331 to be replaced by alanine. Revertant selection identified an intragenic suppressor mutation that increases the CO2/O2 specificity of the mutant enzyme by 33%. This second-site mutation causes threonine-342 to be replaced by isoleucine. The complementing amino acid substitutions flank a catalytically essential lysyl residue at position 334. It thus appears that a number of amino acid residues can influence the CO2/O2 specificity of this bifunctional enzyme. The well defined chloroplast genetics of C. reinhardtii allows the interactions of these residues to be investigated.

Abstract:To maintain CO2 fixation in the Calvin Benson-Bassham cycle, multi-step regulation of the chloroplast ATP synthase (CF1Fo) is crucial to balance the ATP output of photosynthesis with protection of the apparatus. A well-studied mechanism is thiol modulation; a light/dark regulation through reversible cleavage of a disulfide in the CF1Fo γ-subunit. The disulfide hampers ATP synthesis and hydrolysis reactions in dark-adapted CF1Fo from land plants by increasing the required transmembrane electrochemical proton gradient (ΔμH+). Here, we show in Chlamydomonas reinhardtii that algal CF1Fo is differently regulated in vivo. A specific hairpin structure in the γ-subunit redox domain disconnects activity regulation from disulfide formation in the dark. Electrochromic shift measurements suggested that the hairpin kept wild type CF1Fo active whereas the enzyme was switched off in algal mutant cells expressing a plant-like hairpin structure. The hairpin segment swap resulted in an elevated ΔμH+ threshold to activate plant-like CF1Fo, increased by ~1.4 photosystem (PS) I charge separations. The resulting dark-equilibrated ΔμH+ dropped in the mutants by ~2.7 PSI charge separation equivalents. Photobioreactor experiments showed no phenotypes in autotrophic aerated mutant cultures. In contrast, chlorophyll fluorescence measurements under heterotrophic dark conditions point to a reduced plastoquinone pool in cells with the plant-like CF1Fo as the result of bioenergetic bottlenecks. Our results suggest that the lifestyle of Chlamydomonas reinhardtii requires a specific CF1Fo dark regulation that partakes in metabolic coupling between the chloroplast and acetate-fueled mitochondria.

Abstract:Membrane inlet mass spectrometry indicated massive light-dependent cycling of inorganic carbon between the medium and the cells of various phytoplankton species representing the main groups of aquatic primary producers. These included diatoms, symbiotic and free living dinoflagellates, a coccolithophorid, a green alga and filamentous and single cell cyanobacteria. These organisms could maintain an ambient CO2 concentration substantially above or below that expected at chemical equilibrium with HCO3−. The coccolithophorid Emiliania huxleyishifted from net CO2 uptake to net CO2 efflux with rising light intensity. Differing responses of CO2 uptake and CO2 fixation to changing light intensity supported the notion that these two processes are not compulsorily linked. Simultaneous measurements of CO2 and O2 exchange and of the fluorescence parameters in Synechococcus sp. strain PCC 7942, showed that CO2 uptake can serve as a sensitive probe of the energy status of the photosynthetic reaction centers. However, during transitions in light intensity, changes in CO2 uptake did not accord with those expected from fluorescence change. Quantification of the net fluxes of CO2, HCO3− and of photosynthesis at steady-state revealed that substantial HCO3− efflux accompanied CO2 uptake and fixation in the case of `CO2 users'. On the other hand, `HCO3− users' were characterized by a rate of net CO2 uptake below that of CO2 fixation. The results support the notion that entities associated with the CCM function not only in raising the CO2 concentration at the site of Rubisco; they may also serve as a means of diminishing photodynamic damage by dissipating excess light energy.

Abstract:When grown under nitrogen limitation, pronounced chlororespiratory activity develops together with an altered composition of thylakoid membranes in Chlamydomonas reinhardtii. Relative to control cultures, the flash-inhibited, chlororespiration-dependent O2 consumption signal increases 10-fold. Also augmented is the light-sensitive respiratory activity responsible for the "Kok effect," reflecting competitive inhibition of chlororespiratory electron transport by photosystem I. Fluorescence measurements show that the thylakoid plastoquinone pool is extensively reduced in dark-adapted, N-limited cells. Thylakoids of N-limited cells have reduced amounts of cytochrome b6, cytochrome f, and light-harvesting complexes. However, thylakoid-bound NADH-PQ oxidoreductase, with major subunits of 51 kDa and 17 kDa, is increased 7-fold and two novel cytochromes of 34 and 12.5 kDa are highly abundant. Thus, components of photosynthetic and chlororespiratory electron transport pathways are differentially regulated by N availability.
Proton channel inactivation results in loss of chloroplast NDH complex activity

Abstract:The plastidal photosynthetic complex I (formerly NAD(P)H dehydrogenase-like complex, NDH) remains enigmatic in its function within the electron transport chain of higher plants. While the NDH complex shares high homology with complex I, a key component of the respiratory electron transport chain, electron transport rates through the NDH complex in thylakoids are relatively low. In this study, we took a structure-function approach and mutated the plastid genome-encoded ndhF gene to abolish the NdhF proton channel of the NDH complex. These mutations led to loss of plastoquinone reductase activity, indicating tight coupling between the proton and electron transfer reactions within NDH. Additionally, loss of the transverse helix of NdhF led to loss of the NDH complex, suggesting that this region of the NdhF subunit is required for complex stability. In agreement with previous studies using ndh knockout mutants, loss of NDH complex activity did not result in measurable changes in rates of steady-state cyclic electron flow. However, all mutants displayed a shift in the sensitivity of pH-dependent feedback regulation of the photosystem II antennae to total protonmotive force (pmf), indicating a defect in either stromal redox state or pmf distribution into DeltapH and DeltaPsi.

[Investigation of the relation between pigment composition and CO2 exchange in the blue-green alga anacystis nidulans]

Abstract:Changes in culture conditions caused strong changes in the pigment composition in the blue-green alga Anacystis nidulans. Under "normal" illumination (white light; 0.6·10(3) erg/cm(2)·sec) the relation between the amounts of chlorophyll a and phycocyanin was 1:6.6. In a high light intensity (20.8·10(3) erg/cm(2)·sec) the phycocyanin content was reduced and the relations thus changed to 1:1.9. Growing the algae in red light of high intensity (20·10(3) erg/cm(2)·sec) increased the phycocyanin content; the chlorophyll a: phycocyanin relation was then 1:12.1.The action spectrum of apparent photosynthesis showed a minimum at 473 nm in all three cultures. The maximum of photosynthesis in low light cultures fell in the absorption region of phycocyanin at 621 nm. The action spectrum of the red light culture showed a reduced rate of photosynthesis in the same region. The strong light culture had an action spectrum similar to that of the red light culture with a maximum at 651 nm. The differing action spectrum of the low light culture may be a result of interruption in the energy transfer from phycocyanin to chlorophyll a within pigment system II.The transients of CO2 exchange are independent of the pigment composition. Two different types of transients were found depending on the wavelength of the incident light. In red light of 550-650 nm a higher stationary rate was reached after a maximum of photosynthesis at the beginning of the illumination period. In blue and far red light a lower rate was found after the first maximum. Following a illumination period in blue or far red light a CO2 evolution in the dark was observed. On the other hand, this CO2 evolution was not found after illumination with red light. These effects are possiblt caused by a decarboxylation reaction (photorespiration) which occurs only in blue and far red light.

Abstract:Polypeptides encoded by plastid ndhgenes form a complex (Ndh) which could reduce plastoquinone with NADH. Through a terminal oxidase, reduced plastoquinone would be oxidized in chlororespiration. However, isolated Ndh complex has low activity with plastoquinone and no terminal oxidase has been found in chloroplasts, thus the function of Ndh complex is unknown. Alternatively, thylakoid hydroquinone peroxidase could oxidize reduced plastoquinone with H2O2. By immunoaffinity chromatography, we have purified the plastid Ndh complex of barley (Hordeum vulgareL.) to investigate the electron donor and acceptor specificity. A detergent-containing system was reconstructed with thylakoid Ndh complex and peroxidase which oxidized NADH with H2O2 in a plastoquinone-dependent process. This system and the increases of thylakoid Ndh complex and peroxidase activities under photooxidative stress suggest that the chlororespiratory process consists of the sequence of reactions catalyzed by Ndh complex, peroxidase (acting on reduced plastoquinone), superoxide dismutase, and the non-enzymic one-electron transfer from reduced iron-sulfur protein (FeSP) to O2. When FeSP is a component of cytochrome b 6·fcomplex or of the same Ndh complex, O2 may be reduced with NADH, without requirement of light. Chlororespiration consumes reactive species of oxygen and, eventually, may decrease their production by lowering O2 concentration in chloroplasts. The common plastoquinone pool with photosynthetic electron transport suggests that chlororespiratory reactions may poise reduced and oxidized forms of the intermediates of cyclic electron transport under highly fluctuating light intensities.

Energy metabolism and intracellular pH regulation reveal different physiological acclimation mechanisms of Chlorella strains to high concentrations of CO 2

Abstract:The intolerance of high CO 2 in the exhaust gas is the "bottleneck" limiting the wide application of microalgae for CO 2 biosequestration. Around this topic, we selected high-CO 2 -tolerant (LAMB 33 and 31) and nontolerant (LAMB 122) Chlorella strains to study their different energy metabolisms and cytoplasmic pH regulations in response to high CO 2 . Under 40 % CO 2 , LAMB 33 and 31 both showed elevated ATP synthesis, accelerated ATP consumption and fast cytoplasmic pH regulation while exhibiting different acclimating strategies therein: chloroplast acclimations were reflected by high chlorophyll contents in 33 but photosystem transitions in 31; faster mitochondrial acclimations occurred in 33 than in 31; cellular organic carbon mainly flowed to monosaccharide synthesis for 33 but to monosaccharide and protein synthesis for 31; and cytoplasmic pH regulation was attributed to V-ATPase in 31 but not in 33. All the above metabolic processes gradually collapsed in 122, leading to growth inhibition. Our study identified different metabolic acclimation strategies among Chlorella strains to high CO 2 and provided new traits for breeding microalgae for CO 2 biosequestration.

Abstract:It has been shown repeatedly that exposure to elevated atmospheric CO 2 causes an increased C/N ratio of plant biomass that could result from either increased carbon or – in relation to C acquisition - reduced nitrogen assimilation. Possible reasons for diminished nitrogen assimilation are controversial, but an impact of reduced photorespiration at elevated CO 2 has frequently been implied. Using a mutant defective in peroxisomal hydroxy-pyruvate reductase ( hpr1-1 ) that is hampered in photorespiratory turnover, we show that indeed, photorespiration stimulates the glutamine-synthetase 2 (GS) / glutamine-oxoglutarate-aminotransferase (GOGAT) cycle, which channels ammonia into amino acid synthesis. However, mathematical flux simulations demonstrated that nitrate assimilation was not reduced at elevated CO 2 , pointing to a dilution of nitrogen containing compounds by assimilated carbon at elevated CO 2 . The massive growth reduction in the hpr1-1 mutant does not appear to result from nitrogen starvation. Model simulations yield evidence for a loss of cellular energy that is consumed in supporting high flux through the GS/GOGAT cycle that results from inefficient removal of photorespiratory intermediates. This causes a futile cycling of glycolate and hydroxy-pyruvate. In addition to that, accumulation of serine and glycine as well as carboxylates in the mutant creates a metabolic imbalance that could contribute to growth reduction.

Abstract:In freshwaters, algal species are exposed to different inorganic nitrogen (Ni) sources whose incorporation varies in biochemical energy demand. We hypothesized that due to the lesser energy requirement of ammonium ( <math><msubsup><mtext>NH</mtext> <mrow><mn>4</mn></mrow> <mo>+</mo></msubsup> </math> )-use, in contrast to nitrate ( <math><msubsup><mtext>NO</mtext> <mrow><mn>3</mn></mrow> <mo>-</mo></msubsup> </math> )-use, more energy remains for other metabolic processes, especially under CO2- and phosphorus (Pi) limiting conditions. Therefore, we tested differences in cell characteristics of the green alga Chlamydomonas acidophila grown on <math><msubsup><mtext>NH</mtext> <mrow><mn>4</mn></mrow> <mo>+</mo></msubsup> </math> or <math><msubsup><mtext>NO</mtext> <mrow><mn>3</mn></mrow> <mo>-</mo></msubsup> </math> under covariation of CO2 and Pi-supply in order to determine limitations, in a full-factorial design. As expected, results revealed higher carbon fixation rates for <math><msubsup><mtext>NH</mtext> <mrow><mn>4</mn></mrow> <mo>+</mo></msubsup> </math> -grown cells compared to growth with <math><msubsup><mtext>NO</mtext> <mrow><mn>3</mn></mrow> <mo>-</mo></msubsup> </math> under low CO2 conditions. <math><msubsup><mtext>NO</mtext> <mrow><mn>3</mn></mrow> <mo>-</mo></msubsup> </math> -grown cells accumulated more of the nine analyzed amino acids, especially under Pi-limited conditions, compared to cells provided with <math><msubsup><mtext>NH</mtext> <mrow><mn>4</mn></mrow> <mo>+</mo></msubsup> </math> . This is probably due to a slower protein synthesis in cells provided with <math><msubsup><mtext>NO</mtext> <mrow><mn>3</mn></mrow> <mo>-</mo></msubsup> </math> . In contrast to our expectations, compared to <math><msubsup><mtext>NH</mtext> <mrow><mn>4</mn></mrow> <mo>+</mo></msubsup> </math> -grown cells <math><msubsup><mtext>NO</mtext> <mrow><mn>3</mn></mrow> <mo>-</mo></msubsup> </math> -grown cells had higher photosynthetic efficiency under Pi-limitation. In conclusion, growth on the Ni-source <math><msubsup><mtext>NH</mtext> <mrow><mn>4</mn></mrow> <mo>+</mo></msubsup> </math> did not result in a clearly enhanced Ci-assimilation, as it was highly dependent on Pi and CO2 conditions (replete or limited). Results are potentially connected to the fact that C. acidophila is able to use only CO2 as its inorganic carbon (Ci) source.

Effect of the mitochondrial membrane potential on the absorbance of the reduced form of cytochrome c oxidase

Abstract:The effect of mitochondrial membrane potential (ΔΨm) on the absorbance of the reduced cytochrome c oxidase (COX) was evaluated in isolated rabbit heart mitochondria using integrating sphere optical spectroscopy. Maximal reduction of the mitochondrial cytochromes was achieved by either blowing nitrogen to remove oxygen, or by adding cyanide. Gradual depolarization of ΔΨm by adding increasing concentrations of uncoupler resulted in an increase of up to 50 % in the absorbance of cytochrome aa3 under nitrogen saturation, and of 25 % with cyanide. Cytochrome aa3 absorbance increases were also observed in the presence of cyanide with apyrase (20 %) or oligomycin (12 %). The bL heme absorbance also decreased as expected from ΔΨm depolarization. A ~ 1 nm red shift in the peak wavelength of cytochrome aa3 was observed under anoxic conditions as ΔΨm was depolarized. Importantly, cytochrome c and c1 absorbances remained constant at levels corresponding to full reduction under all experimental manipulations of ΔΨm, especially with cyanide. These data suggest that ΔΨm-dependent changes in the absorbance of reduced COX were due to a variable extinction coefficient of heme a and/or a3 as a function of ΔΨm. A similar increase in the reduced cytochrome aa3 absorbance without changes in cytochrome c and c1 was observed in the perfused rabbit heart when decreasing ΔΨm with uncoupler. Our results imply that COX absorbance in its fully reduced state does not simply reflect the oxygen tension but also the ΔΨm. This may prove useful in monitoring ΔΨm under anoxic or ischemic conditions in intact tissue.

Abstract:As global temperatures climb to historic highs, the far-reaching effects of climate change have impacted agricultural nutrient availability. This has extended to low latitude oceans, where a deficit in both nitrogen and phosphorus stores has led to dramatic decreases in carbon sequestration in oceanic phytoplankton. Although Chlamydomonas reinhardtii, a freshwater model green alga, has shown drastic systems-level alterations following nitrogen deprivation, the mechanisms through which these alterations are triggered and regulated are not fully understood. This study examined the role of reversible oxidative signaling in the nitrogen stress response of C. reinhardtii. Using oxidized cysteine resin-assisted capture enrichment coupled with label-free quantitative proteomics, 7889 unique oxidized cysteine thiol identifiers were quantified, with 231 significantly changing peptides from 184 proteins following 2 h of nitrogen deprivation. These results demonstrate that the cellular response to nitrogen assimilation, photosynthesis, pigment biosynthesis, and lipid metabolism are regulated by reversible oxidation. An enhanced role of non-damaging oxidative pathways is observed throughout the photosynthetic apparatus that provides a framework for further analysis in phototrophs.

A chloroplast pump model for the CO2 concentrating mechanism in the diatom Phaeodactylum tricornutum

Abstract:Prior analysis of inorganic carbon (Ci) fluxes in the diatom Phaeodactylum tricornutum has indicated that transport of Ci into the chloroplast from the cytoplasm is the major Ci flux in the cell and the primary driving force for the CO2 concentrating mechanism (CCM). This flux drives the accumulation of Ci in the chloroplast stroma and generates a CO2 deficit in the cytoplasm, inducing CO2 influx into the cell. Here, the “chloroplast pump” model of the CCM in P. tricornutum is formalized and its consistency with data on CO2 and HCO3− uptake rates, carbonic anhydrase (CA) activity, intracellular Ci concentration, intracellular pH, and RubisCO characteristics is assessed. The chloroplast pump model can account for the major features of the data. Analysis of photosynthetic and Ci uptake rates as a function of external Ci concentration shows that the model has the most difficulty obtaining sufficiently low cytoplasmic CO2 concentrations to support observed CO2 uptake rates at low external Ci concentrations and achieving high rates of photosynthesis. There are multiple ways in which model parameters can be varied, within a plausible range, to match measured rates of photosynthesis and CO2 uptake. To increase CO2 uptake rates, CA activity can be increased, kinetic characteristics of the putative chloroplast pump can be enhanced to increase HCO3− export, or the cytoplasmic pH can be raised. To increase the photosynthetic rate, the permeability of the pyrenoid to CO2 can be reduced or RubisCO content can be increased.

Abstract:Some cyanobacteria, but not all, experience an induction of alternative electron flow (AEF) during CO2-limited photosynthesis. For example, Synechocystis sp. PCC 6803 (S. 6803) exhibits AEF, but Synechococcus elongatus sp. PCC 7942 does not. This difference is due to the presence of flavodiiron 2 and 4 proteins (FLV2/4) in S. 6803, which catalyze electron donation to O2. In this study, we observed a low-[CO2] induced AEF in the marine cyanobacterium Synechococcus sp. PCC 7002 that lacks FLV2/4. The AEF shows high affinity for O2, compared with AEF mediated by FLV2/4 in S. 6803, and can proceed under extreme low [O2] (about a few µM O2). Further, the transition from CO2-saturated to CO2-limited photosynthesis leads a preferential excitation of PSI to PSII and increased non-photochemical quenching of chlorophyll fluorescence. We found that the model green alga Chlamydomonas reinhardtii also has an O2-dependent AEF showing the same affinity for O2 as that in S. 7002. These data represent the diverse molecular mechanisms to drive AEF in cyanobacteria and green algae. In this paper, we further discuss the diversity, the evolution, and the physiological function of strategy to CO2-limitation in cyanobacterial and green algal photosynthesis.

Abstract:Upon illumination, photosystem I in chloroplasts catalyzes light-driven electron transport from plastocyanin to ferredoxin, followed by the reduction of NADP+ to NADPH by ferredoxin:NADP+ reductase for CO2 fixation. At the beginning of photosynthesis, NADP+ supply control is dominated by de novo NADP+ synthesis rather than being recycled from the Calvin cycle. Importantly, ferredoxin distributes electrons to NADP+ as well as to thioredoxins for light-dependent regulatory mechanisms, to cyclic electron flow for more adenosine triphosphate (ATP) production, and to several metabolites for reductive reactions. We previously demonstrated that the NADP+ synthesis activity and the amount of the NADP pool size, namely the sum of NADP+ and NADPH, varies depending on the light conditions and the ferredoxin-thioredoxin system. In addition, the regulatory mechanism of cytoplasmic NAD+ supply is also involved in the chloroplastic NADP+ supply control because NAD+ is an essential precursor for NADP+ synthesis. In this mini-review, we summarize the most recent advances on our understanding of the regulatory mechanisms of NADP+ production, focusing on the interactions, crosstalk, and co-regulation between chloroplasts and the cytoplasm at the level of NAD+ metabolism and molecular transport.

Abstract:The activity of the molecular motor enzyme, chloroplast ATP synthase, is regulated in a redox-dependent manner. The γ subunit, CF1-γ, is the central shaft of this enzyme complex and possesses the redox-active cysteine pair, which is reduced by thioredoxin (Trx). In light conditions, Trx transfers the reducing equivalent obtained from the photosynthetic electron transfer system to the CF1-γ. Previous studies showed that the light-dependent reduction of CF1-γ is more rapid than those of other Trx target proteins in the stroma. Although there are multiple Trx isoforms in chloroplasts, it is not well understood as to which chloroplast Trx isoform primarily contributes to the reduction of CF1-γ, especially under physiological conditions. We therefore performed direct assessment of the CF1-γ reduction capacity of each of the Trx isoforms. The kinetic analysis of the reduction process showed no significant difference in the reduction efficiency between two major chloroplast Trxs, namely Trx-f and Trx-m. Based on the thorough analyses of the CF1-γ redox dynamics in Arabidopsis thaliana Trx mutant plants, we found that lack of Trx-f or Trx-m had no significant impact on the in vivo light-dependent reduction of CF1-γ. The results showed that CF1-γ can accept the reducing power from both Trx-f and Trx-m in chloroplasts.

Abstract:The local density of electronic states N(E) has been measured on C{sub 36} thin films using scanning tunneling spectroscopy (STS). A 0.8thinspthinspeV energy gap and sharp features in N(E) with widths as small as 0.2thinspthinspeV can be ascribed to the discrete molecular energy levels predicted for the C{sub 36} molecule, broadened by intermolecular interactions. However, a more complete agreement between the experimental results and theoretical predictions occurs if the system is modeled as covalently bonded C{sub 36} dimers and trimers. These results confirm the molecular nature of C{sub 36} as well as its higher reactivity as compared to larger size fullerenes. {copyright} {ital 1998} {ital The American Physical Society}
