Abstract:Rh(III)-catalyzed C(sp(3)) - H bond aminocarbonylation of 8-methylquinolines and isocyanates has been realized under mild conditions. This approach is applicable to different aryl and alkyl isocyanates, leading to the synthesis of various alpha-quinolinyl amide compounds in moderate to excellent yields. A plausible mechanism for this transformation is proposed according to the experimental results obtained.

Abstract:It is shown here for the first time that diethyl azodicarboxylate promotes dehydrogenation of tertiaryamines to afford enamines, which subsequently take place in tandem reactions with sulfonyl azides to give the N-sulfonyl amidine derivatives. A number of different substituted tertiaryamines and sulfonyl azides can successfully be coupled, and several functionalized groups are tolerated in this system. The reaction described here is mild, general, and efficient, thus providing an extremely preferable method for synthesis of a variety of N-sulfonyl amidine derivatives.

Abstract:Traditional coupling reactions generally employ metal organic compounds, boron reagents or halogenated compounds as reaction starting materials under the condition of noble metal-catalyst, such as Suzuki reaction and Heck reaction. Although these reactions have been widely used in synthesis, due to the need for prefunctionalization of reactants, they do not have high atomic economy and step economy. Especially for some substrates that are not easy to introduce functional groups, the application of such reactions is limited. In recent years, with the development of organic chemistry, direct C-H bond functionalization has become a research hotspot in organic synthesis. This kind of reaction can overcome the deficiency of traditional coupling reaction, making this kind of reaction more attractive. Amides are very important compounds, which are widely found in natural products and drugs. Through literature investigation, it was found that some functionalization reactions of C(sp(3))-H bond adjacent to the nitrogen atom of amides had already reported. Due to the facile synthesis, stability and low toxicity of cinnamic acid compounds, they are also used in organic synthesis to obtain desired product. When we studied the decarboxylation reaction of cinnamic acid compounds, we found that the decarboxylative coupling product of cinnamic acid with amides could be obtained by using amide as solvent, tert-butyl hydroperoxide (TBHP) as oxidant and cinnamic acid as starting material without the use of metal catalyst. The reaction mechanism was also proposed. First, homolysis of tert-butyl alcohol peroxide generated tertiary butyl oxygen free radical and hydroxyl free radicals, the oxidation of C(sp(3))-H bonds adjacent to the nitrogen atom of N,N-dimethylacetamide offered carbon free radical, which adds to the a-position of the double bond of cinnamic acid to produce intermediate A. A hydroxyl radical, generated from homolysis of TBHP, then combined with A to generate B, and B was further oxidized to C. Finally, C decarboxylated easily to generate the desired product.

Abstract:Palladium-catalyzed Suzuki-Miyaura cross-coupling or aryl halides is widely employed in the synthesis of many important molecules in synthetic chemistry, including pharmaceuticals, polymers and functional materials. Herein, we disclose the first palladium-catalyzed decarbonylative Suzuki-Miyaura cross-coupling of amides for the synthesis of biaryls through the selective activation of the N-C(O) bond of amides. This new method relies on the precise sequence engineering of the catalytic cycle, wherein decarbonylation occurs prior to the transmetallation step. The reaction is compatible with a wide range of boronic acids and amides, providing valuable biaryls in high yields (>60 examples). DFT studies support a mechanism involving oxidative addition, decarbonylation and transmetallation and provide insight into high N-C(O) bond activation selectivity. Most crucially, the reaction establishes the use of palladium catalysis in the biaryl Suzuki-Miyaura cross-coupling of the amide bond and should enable the design of a wide variety of cross-coupling methods in which palladium rivals the traditional biaryl synthesis from aryl halides and pseudohalides.

Abstract:A new strategy for the sequential formation of aryl and amidyl C-N bonds is reported. Using trichloroethoxysulfonyl azide as a bifunctional nitrogen source, Ir-catalyzed aryl C-H sulfonamidation and subsequent desulfonative amide formation proceed effectively without any need of oxidants or coupling reagents. This protocol is suitable for readily available benzamides and stable carboxylates including primary, secondary, and tertiary alkyl, alkenyl, and phenyl carboxylates, thereby providing a direct and efficient method for the synthesis of biologically and chemically useful N-arylamides.

Modification Structure of Cinnamaldehyde with Primary Amines by Reflux and Sonication Methods in the Presence of Sulfuric Acid as a Catalyst

Abstract:Cinnamon is one of the most valuable natural resources that sustains life and exists freely in nature. Cinnamaldehyde is the primary compound in cinnamon oil. It has a unique structure that contains a benzene ring, an aldehyde group, and an unsaturated double bond. Cinnamaldehyde has been structurally modified to improve biological activity. In this research, cinnamaldehyde and nitrophenyl amines were reacted with sulfuric acid as a catalyst by refluxing and sonication. UV-Vis Spectroscopy, FT-IR, 1H-NMR, and 13C-NMR were used to validate the chemical structures. Thin-layer chromatography (TLC) revealed a new single spot formed by the reaction of cinnamaldehyde and 4-amino-2-nitrophenol. By refluxing for 2 hours or sonicating for 30 minutes, a novel imine chemical, 4-nitro-2-((3-phenylallylidene)amino)phenol, was effectively synthesized with a yield of 75.21% or 83.71%, respectively. This imine was obtained as a dark red powder with a melting point of 237 °C. Meanwhile, only sonication produced a novel product from the reaction of cinnamaldehyde and 4-nitroaniline. However, the structural elucidation has not yet been performed because the yield was so low. Surprisingly, there was no reaction between cinnamaldehyde and 2,4-dinitroaniline. It was most likely owing to the amine’s bulky structure and the presence of two nitro groups in the amine as electron-withdrawing groups that reduced the nucleophilicity of the amine. We demonstrated that sonication is a suitable approach for imine synthesis, as it is commonly utilized in organic compound synthesis protocols.

Abstract:An efficient and straightforward protocol for accessing a new series of functionalized 4‐aminocoumarins from PIDA/I2‐mediated decarboxylative C4‐amination of coumarin‐3‐carboxylic acids via direct Csp2–H dehydrogenative C–N cross‐coupling with secondary amines under ambient conditions has been accomplished. The notable advantages of this protocol are the tolerance of diverse functional groups, mild reaction conditions at ambient temperature, moderate to good yields, short reaction times (in minutes), high regioselectivity, gram‐scale applicability, and eco‐friendliness. This is the first report of decarboxylative Csp2‐H cross‐dehydrogenative C‐N coupling of coumarin‐3‐carboxylic acids for synthesizing 4‐aminocoumarins.

Abstract:In this communication, we report a photocatalyzed amidation strategy from carboxylic acids and tertiary amines through C-N bond cleavage. A wide scope of structurally diverse carboxylic acids participate smoothly in the reaction, providing the desired tertiary amides with moderate-to-good yields (34 examples, up to 93% yield). This amidation strategy provides an alternative way to address the regioselectivity between nucleophilic functional groups, thus complementing the functional group compatibility of classical amidation protocols. Its synthetic robustness is also proved by the late-stage modification of several complex molecules and gram-scale applications.

Abstract:An operationally simple and metal-free approach is described for the synthesis of pyrazole-tethered thioamide and amide conjugates. The thioamides were generated by employing a three-component reaction of diverse pyrazole C-3/4/5 carbaldehydes, secondary amines, and elemental sulfur in a single synthetic operation. The advantages of this developed protocol refer to the broad substrate scope, metal-free and easy to perform reaction conditions. Moreover, the pyrazole C-3/5-linked amide conjugates were also synthesized via an oxidative amination of pyrazole carbaldehydes and 2-aminopyridines using hydrogen peroxide as an oxidant.

Abstract:A convenient synthetic approach for the synthesis of alpha-oxo-acetamidines via copper-catalyzed C(sp(3))-H amidination has been developed. This approach allows the direct amidination of three C(sp(3))-H bonds without cyclization.Methyl ketones and primary and secondary amines are tolerant and afford the corresponding products in moderate to good yields. Abstract:Herein, we describe a two‐component methodology developed for the synthesis of pyrimidine derivatives using tertiary alkylamines and amidines. Tertiary alkylamines serve as dual C2 synthons through copper‐catalyzed aerobic difunctionalization of the α,β‐C(sp3)‐H bonds. The process operates under mild conditions and uses atmospheric oxygen as the oxidant. Notably, this methodology yields up to 85% and shows a broad substrate scope. Mechanistic studies indicate that the annulation proceeds via a radical‐mediated oxidation and C‒N bond coupling process. This approach provides a pathway for synthesizing various heterocycles by employing tertiary alkylamines as dual C2 synthons.

Abstract:Decarboxylation reactions have been widely explored in recent ten years, and decarboxylative cascade reaction of cinnamic acids has also attracted much attention. Generally, this class of reaction includes two processes: radical addition and decarboxylation. The result of reaction can introduce kinds of functional groups on the benzene ring, such as halogen atom, nitro-group and trifluoromethyl. Following the continuous studies of our group on oxidative cascade reaction, especially the oxidative reaction in the absence of metal, herein we disclosed a decarboxylative oxidative cascade reaction of cinnamic acids under metal-free conditions. We employed K 2 CO 3 as base, tert -butyl hydroperoxide (TBHP) as oxidant and DMSO/H 2 O as co-solvent, and cinnamic acids could be converted to propiophenone derivatives in moderate yield. To get an insight into the mechanism of this process, several controlled experiments were conducted. First, DMSO- d 6 was employed under the standard conditions and no d -methyl was detected in the 1 H NMR spectrum of product. It demonstrated that the methyl of product was derived from tert- butyl hydroperoxide. Subsequently, the reaction was carried out under nitrogen and oxygen atmosphere, and it was found that higher reaction efficiency was obtained in N 2 . The result indicated that methyl radical was easily quenched by oxygen. On the basis of experiment results, we proposed a plausible mechanism. First, tert -butyl hydroperoxide yielded methyl radical, then the reaction underwent radical addition and decarboxylation to generate the desired product. The reaction featured that tert -butyl hydroperoxide was used as oxidant and methylation reagent in this process, and the reaction proceeded under metal-free and aqueous phase condition. Therefore, it met the requirement of green chemistry. Meanwhile, the operation of reaction was also simple, for example, to a DMSO/H 2 O (0.5 mL/0.5 mL) solution of cinnamic acids (0.2 mmol) were successively added K 2 CO 3 (0.3 mmol), TBHP (0.8 mmol). The reaction mixture was stirred at 100 ˚ C. Upon the completion, the desired product was purified by silica gel column chromatography.
