Abstract:Objective:To establish a method of bromomethyl phenol by spectrophotmetry and RP-HPLC method to the determination of the all contents of hyoscymine alkaloides and the main contents of atropine sulfuric acid in hy- oscyamus extract tablets.Methods:Hyoscymine alkaloides reacts with bromomethyl phenol to produce a colorful ion- ic couple at a certain pH solution and determined by spectrophotmetry,The contents of atropine sulfuric acid in hy- oscyamus extract tablets was measured by RP-HPLC,a column of Capcell Pak C_(18)and Kromasil C_(18)(250 mm× 4.6 mm,5 μm),with mobile phase of methanol-acetic acid buffer solution,flow rate 1.0 mL·min~(-1),detection wavelength of 210 nm,column temperature 30 ℃.Results:The calibration curves showed good linearity in the range of 0-16 μg·mL~(-1)for hyoscymine alkaloides,con'elation coefficients were 0.9997.The average recoveries(n=9) were 100.9%,RSD was 1.9%.The calibration curves showed good linearity in the range of 4.0-20 μg·mL~(-1)for atropine sulfuric acid,correlation coefficients were 0.9994.The average recoveries(n=9)were 100.2%;RSD was 1.1%.Conclusion:Two accurate and reproducible methods can be used for the quality control of hyoscyamus ex- tract tablets.
Validated derivative and ratio derivative spectrophotometric methods for the simultaneous determination of levocetirizine dihydrochloride and ambroxol hydrochloride in pharmaceutical dosage form

Abstract:Three simple, precise, accurate and validated derivative spectrophotometric methods have been developed for the simultaneous determination of levocetirizine dihydrochloride (LCD) and ambroxol hydrochloride (ABH) in bulk powder and in pharmaceutical formulations. The first method is a first derivative spectrophotometric method ((1)D) using a zero-crossing technique of measurement at 210.4 nm for LCD and at 220.0 nm for ABH. The second method employs a second derivative spectrophotometry ((2)D) where the measurements were carried out at 242.0 and 224.4 nm for LCD and ABH, respectively. In the third method, the first derivative of the ratio spectra was calculated and the first derivative of the ratio amplitudes at 222.8 and 247.2 nm was selected for the determination of LCD and ABH, respectively. Calibration graphs were established in the ranges of 1.0-20.0 μg mL(-1) for LCD and 4.0-20.0 μg mL(-1) for ABH using derivative and ratio first derivative spectrophotometric methods with good correlation coefficients. The developed methods have been successfully applied to the simultaneous determination of both drugs in commercial tablet dosage form.

Abstract:An indirect sensitive and selective spectrophotometric method has been developed for the determination of cephalexin monohydrate (CEM) ceftriaxone sodium (CFX) and cefotaxim sodium (CEF) in bulk and pharmaceutical formulation. The method is based on the oxidation of (CEM), (CFX) and (CEF) in hydrochloric acid medium with known excess on N-Bromosuccinimde and subsequent determination of unreacted oxidant by decolorization of Evans blue dye (EB) and measure the absorbance of residual dye at 608 nm. Calibration curves of evans blue dye in the presence of drugs were rectilinear over the ranges 1.0-9.0 , 1.0-8.0 and 1.0-9.0 µg/ml with molar absorptivity 2.75×104 , 9.28×104 and 7.81×104 l.mol-1.cm-1 and average recoveries 98.97, 102.08 and 100.08% for CEM, CFX and CEF respectively with RSD of less than 3.29%. The method was free from interference of many excipients and additives commonly found in pharmaceutical formulations. The developed method was successfully applied for determination of the studied drugs in their pharmaceutical formulation resulted in a good agreement with certified value and standard addition procedure.
Spectrophotometric determination of trazodone, amineptine and amitriptyline hydrochlorides through ion-pair formation using methyl orange and bromocresol green reagents

Abstract:A simple and rapid extraction spectrophotometric procedure has been developed for the determination of tricyclic anti-depressant drugs such as trazodone (TZH), amineptine (APH) and amitriptyline (ATPH) hydrochlorides in pure form and in different dosage forms. The method involves the formation of intense yellow ion-pairs between these drugs under investigation and methyl orange (MO) and bromocresol green (BCG) reagents followed by their extraction with 1,2-dichloroethane and quantitative microdetermination at 420 and 410 nm using MO or BCG, respectively. The optimum experimental conditions for the ion-pairs formation are established. The method permits the determination of TZH, APH and ATPH over a concentration range of 2-50, 2-50 and 1-25 microg ml(-1) for TZH, APH and ATPH, using MO and 1-25 microg ml(-1) for TZH, APH and ATPH, using BCG, respectively. The Sandell sensitivity (S) is found to be 0.106, 0.1071 and 0.0907 g cm(-2) for TZH, APH and ATPH, respectively, using MO reagent and 0.0788, 0.0661 and 0.0494 g cm(-2) for TZH, APH and ATPH, respectively, using BCG. The method is applicable for the assay of the investigated drugs in different dosage forms and the results are in good agreement with those obtained by the official method.
Facile approaches for determination of Bromhexine Hydrochloride and its active metabolite Ambroxol Hydrochloride using Eosin Y

Abstract:New validated Spectroscopic methods were developed to assay Bromhexine Hydrochloride and its active metabolite Ambroxol Hydrochloride separately in pure form and pharmaceutical formulations. The spectrophotometric assay (method I) shows complex formation between each of the drugs and Eosin Y at 540nm at pH 3.6 and 3.4mL of 4×10-4M Eosin for Bromhexine and Ambroxol. The Spectrofluorimetric assay (method II) depends on quenching eosin native fluorescence by the studied drugs, which measured at 540nm after excitation at 302nm. The spectrophotometric absorbance-concentration plot is rectilinear over the ranges (1.0-5.0) and (1.0-10.0) μg/mL for bromhexine and ambroxol with LOD of 0.31 and 0.14μg/mL and LOQ of 0.94 and 0.42μg/mL for the two drugs respectively. The fluorometric-concentration plot is linear along the range (1.0-5.0) μg/mL and (1-10) μg/mL for the two drugs respectively with LOD of 0.13μg/mL and 0.22μg/mL and LOQ of 0.4μg/mL and 0.65μg/mL for the two drugs, respectively. Developed assays have been validated in agreement with ICH recommendations and they were used in the analysis of commercial drug formulations containing the two mucolytic drugs and the results were matching with those obtained by the comparison method.
Spectrophotometric determination of trazodone, amineptine and amitriptyline hydrochlorides through ion-pair formation with molybdenum and thiocyanate

Abstract:Extraction spectrophotometric method has been developed for the determination of tricyclic drugs such as trazodone (TZH), amineptine (APH) and amitriptyline (ATPH) hydrochlorides in pure form and in the dosage forms coming from different Egyptian markets. The method based on the formation of ion-pairs between these drugs under investigation and inorganic complex of Mo(V)-thiocyanate followed by its extraction with methylene chloride. The optimum conditions for the ion-pairs formation are established. The method permits the determination of TZH, APH and ATPH over the concentration range of 2-28, 2-32 and 1-30 microg ml(-1), respectively. The Sandell sensitivity (S) is found to be 0.105, 0.138 and 0.118 g cm(-2) for TZH, APH and ATPH, respectively. The SD is found to be 0.16-0.377, 0.12-0.259 and 0.091-0.286 and the R.S.D. are 0.14-0.55, 0.12-0.399 and 0.095-0.485 for TZH, APH and ATPH, respectively. The method is applicable for the assay of the investigated drugs in different dosage forms and the results are in good agreement with those obtained by the official method.

Abstract:Abstract A spectrophotometric procedure for the determination of sertraline hydrochloride (Sert) and/or clidinium bromide (Clid) in bulk sample and in dosage forms was developed. The purpose of this work was to develop a rapid, simple, inexpensive, precise, and accurate visible spectrophotometric method. The procedure is based on formation of an ion-pair complex by their reaction with bromocresol green (BCG), bromophenol blue (BPB), and bromothymol blue (BTB) in buffered aqueous solution at pH 3. The colored products are extracted into a polar solvent and measured spectrophotometrically at the optimum λmax for each complex. Optimization of different experimental conditions is described. Regression analysis of Beer-Lambert plots showed good correlation in the concentration range of 1–30 μg mL−1. The apparent molar absorptivity, Sandell sensitivity, detection and quantification limits were calculated. For more accurate analysis, Ringbom optimum concentration range of 2–27 μg mL−1 was used. The developed methods were successfully applied for the determination of sertraline hydrochloride and clidinium bromide in bulk in pharmaceutical formulations without any interference from common excipients. The procedure has the advantage of being highly sensitive and simple for the determination of the studied drugs, weak UV-absorbing compounds.

Abstract:A straightforward and efficient spectrum technique was created using Ortho- chloranil as the electron acceptor (-acceptor) in a charge transfer (CT) complex formation reaction to determine the concentration of famotidine (FMD) in solutions. Compared to the double-distilled blank solution, the reaction result detected a definite violet colour at a maximum absorption wavelength of 546 nm, For concentrations range 2—28 μg/ml, the technique demonstrated excellent compliance with Beer-Law and Lambert's, as evidenced by its molar absorptivity of 2159.648 L mol −1 cm –1 . Lower detection limits of 0.3024 μg/ml and 1.471 μg/ml, respectively, were discovered. The complexes of famotidine and Ortho- chloranil were found to have a 2:1 stoichiometry. Additionally, the suggested approach effectively estimated famotidine concentrations in pharmaceutical formulations, particularly in tablet form.

Abstract:Two fluorescence‐based techniques to determine Ambroxol (AMX) and Guaifenesin (GFN) in their isolated forms or combined in pharmaceutical formulations without interference from each other. Ambroxol hydrochloride (AMX) and guaifenesin (GFN) are approved drugs utilized to treat coughs through their potent mucolytic and expectorant properties. Due to their massive, combined administration in many illnesses, there is a persistent need for their concurrent estimation in different pharmaceutical formulations. Two sensitive, environmentally friendly spectrofluorimetric methods were developed. AMX was determined using the first method (I) without interference from GFN. This method depends on the quenching of Erythrosine B (EB) native fluorescence at 552 nm after excitation at 527 nm due to the formation of a non‐fluorescent AMX‐EB ion‐pair complex in Britton–Robinson buffer (BRB) solution pH (3.5). The concentration plot is linear over the 0.25–5.0 μg/mL range, with a mean percent found value of 99.74%. Method (II) depends on measuring the native fluorescence of aqueous GFN solution at two analytical wavelengths, either 300 or 600 nm, after excitation at 274 nm. Relative fluorescence intensity (RFI)–concentration plots are linear over the ranges of 0.02–0.5 and 0.1–2.0 μg/ml, with mean percent found at 99.96% and 99.91% at dual wavelengths, respectively. The proposed methods were successfully applied to assay both drugs in raw materials and different single and combined pharmaceutical formulations. These methods have been thoroughly validated following International Committee on Harmonisation (ICH) guidelines. National Environmental Methods Index, Analytical Eco‐Scale, and Green Analytical Procedure Index were used to prove greenness, thereby enhancing their applicability. The proposed techniques provide straightforward, precise, and cost‐effective solutions for routine formulation analysis in quality control laboratories.

Abstract:Objective: To develop and validate simple, sensitive, precise and free of organic solvents method for the determination of sildenafil (SIL) and vardenafil (VAR) in bulk and pharmaceutical formulation.Methods: The method is based on the reaction of studied drugs with a mixture of potassium iodide and potassium iodate in an aqueous medium at (25±2 °C) to form yellow coloured triiodide ions (I3-) within 45 min. The reaction is followed spectrophotometrically by measuring the absorbance at 288, 351 nm and 285, 351 nm for sildenafil and vardenafil respectively.Results: The effects of analytical parameters on the reported systems were investigated. Beer's law of SIL was obeyed in the range of (0.4-12) μg ml-1and (0.6-16) μg ml-1. Molar absorptivity was found to be (67.659 ×103) lmol/cmand (37.955×103) lmol/cm at 288 nm, 351 nm respectively. Beer's law of VAR was obeyed in the range of (0.2-13) μg/mland (0.5-40) μg/ml. Moreover, molar absorptivity’s were found to be (68.719 ×103) l mol-1 cm-1and (26.691×103) l mol-1 cm-1 at 285 nm, 351 nm respectively.Conclusion: The proposed method has been applied to determine the components in dosage forms with an average recovery of 98.15% to 103.45% and the results have been found in good agreement with those results obtained by the reference methods.

Abstract:A simple, fast, sensitive and selective spectrophotometric method has been developed based on the oxidative coupling reaction process, which depends on the determination of clonazepam (CZP) drug using an oxidizing agent sodium periodate (NaIO4) in the presence of a reagent, phenylephrine hydrochloride (PH-HCL), that uses the pink color of CZP at a wavelength of 495 nm. Where several factors aff ecting the color intensity and absorbance were studied, including the eff ect of color stability time, eff ect of the volume of the reagent, the eff ect of the volume of the oxidizing agent, order of addition and temperature. The calibration curve found to obey Lambert beer at range concentration (2–20 mg/L), while the limit of detection (LoD) (8. 9*10-2), limit of quantitation (LoQ) (2.6*10-2) and molar absorbance is 1.4 × 102 liters mol-1cm-1. The method was applied to pharmaceutical preparations (Tablet) and was found to characterize the best precision and accuracy. The standard methods not need any control of temperature and also not aff ected by interferences and henceforth successfully utilized to determine CZP in pharmaceutical preparations

Abstract:In the present study, two drugs for treating acromegaly are investigated which are either synthetic growth hormone inhibiting hormone (GHIH) or dopamine receptor agonist. Octreotide (OCT) and bromocriptine (BCT) were quantified with a quick, simple, and sensitive spectrophotometric approach in their authentic forms and commercial dosage forms. This approach was based upon formation of ion pair complex between erythrosine B and investigated drugs in an aqueous buffered solution. For OCT determination the higher sensitivity was obtained using ΔA at 525 nm. On the other hand, BCT was estimated using the absorbance at 556 nm. Under optimal conditions for the reaction, construction of calibration curves were performed within concentration range of 0.4-4 µg ml-1 for OCT and 1-9 µg ml-1 for BCT with with low detection limits (0.094 and 0.235 µg ml-1) and low quantitation limits (0.29 and 0.71 µg ml-1) for OCT and BCT respectively. The developed approach was assessed for linearity, accuracy, precision, limits of detection, and limits of quantitation in compliance with ICH validation recommendations. The suggested approaches demonstrated good linearity, as evidenced by determination coefficients (r2) of 0.999 with a slope 0.14 for OCT and 0.9989 with a slope 0.06 for BCT. Additionally, the environmental friendliness of the investigated method was assessed with some of the recent green analytical chemistry metrics.

Abstract:Abstract A simple, rapid, sensitive and economic spectrofluorimetric method has been developed and validated for determination of some β-adrenergic blocking agents namely; betaxolol hydrochloride (BTX), carvedilol (CAR), labetalol hydrochloride (LBT), nebivolol hydrochloride (NEB) and propranolol hydrochloride (PRO). The method is based on the quenching effect of the cited drugs on the fluorescence intensity of eosin Y at pH 3.4 (acetate buffer). The fluorescence quenching is due to the formation of an ion-pair complex and was measured without extraction at 545 nm (λex. 301.5 nm). The factors affecting the formation of the ion-pair complex were carefully studied and optimized. Under the optimal conditions, the linear ranges for the relationship between the fluorescence quenching value and the concentration of the investigated drugs were 100-2500, 150-2500 and 50-2250 ng mL -1 for (BTX, CAR), (LBT, NEB) and (PRO) respectively. The method was validated according to ICH guidelines and was applied for determination of the cited drugs in pharmaceutical dosage forms with excellent recoveries. In addition, content uniformity testing of some commercial dosage forms was checked by the proposed method.

Abstract:Two sensitive and selective spectrophotometric and spectrofluoimetric procedures were developed for the determination of thonzylamine hydrochloride (THAH) in tablets and nasal drops. The methods are based on König reaction which resulted in an orange-yellow fluorescent product. The orange-yellow product of the interaction between the dicyclohexylcarbodiimide (DCC), THAH and dimethylbarbituric acid (DMBA) showed an absorption maximum at 492 nm, a first-derivative signal at 494 nm and a fluorescence emission peak at 518 nm (lambda(ex)=492 nm). The orange-yellow color was found to be stable for at least 2 h. The reaction conditions were studied and optimized. The reaction obeys Beer's law over the ranges 8-20 and 0.2-2.0 microg ml(-1) for the derivative spectrophotometric and fluorimetric measurements, respectively. The detection limits were found to be 0.29 and 0.018 microg ml(-1) for the spectrophotometric and fluorimetric measurements, respectively. The proposed methods were applied to the analysis of pharmaceutical formulations containing THAH, either alone or in combination with naphazoline nitrate.

Abstract:Four sensitive and fast analytical approaches relied on ion pairing with eosin Y were built up and evaluated using spectroscopy for determination of Alcaftadine and Olopatadine hydrochloride with high sensitivity and selectivity. Two spectrofluorimetric techniques were employed to observe the quenching effect of Alcaftadine or Olopatadine hydrochloride on the intrinsic fluorescence of eosin Y in a 0.1 M acetate buffer solution at pH 3.8 and 3.3 for Alcaftadine and Olopatadine hydrochloride, respectively. Those methods are considered the first spectrofluorimetric methods for Alcaftadine and Olopatadine hydrochloride assay. The fluorescence quenching effect was linear with concentration ranging from 150 to 2000 and 200 to 2000 ng mL −1 for Alcaftadine and Olopatadine hydrochloride, respectively. In the two spectrophotometric techniques, the absorbance of the produced ion-pair was monitored at 548 and 547 nm in aqueous buffered solution at pH 3.8 and 3.3 for Alcaftadine and Olopatadine hydrochloride, respectively. Beer's law was obeyed in the concentrations range of 0.8–8.0 and 1.0–10.0 μg mL −1 . The four techniques were evaluated in accordance with ICH requirements and were effectively used to analyze dosage forms with a high percent recovery.

Abstract:Three simple, sensitive, precise, and rapid spectrophotometric methods are developed and optimized for the assay of vardenafil in pharmaceutical formulations. The procedures are ascertained on the oxidation of vardenafil with a slight excess of N-bromosuccinimide (NBS), and the unreacted oxidant is estimated by evaluating the amount of the unconsumed NBS by reacting with a fixed amount of safranin (method A), crystal violet (method B), and aniline blue (method C) in an acidic medium by measuring the absorbance at 530, 600, and 610 nm, respectively. In all the methods, the amount of NBS consumed corresponds to the amount of the drug. Beer's law range, Sandell's sensitivity, and the corresponding molar absorptivity values of 1.1104 × 10 4 , 1.0305 × 10 4 , and 6.217 × 10 3 L/mol × cm for methods A, B, and C are calculated. The limits of detection and quantification are evaluated. The methods are also evaluated statistically by means of Student's t-test and F-test. The results show excellent agreement and insignificant difference between the proposed methods and the reference method. The developed methods are successfully applied to the assay of Vardenafil in pure and dosage forms, and no interference is observed from common excipients present in pharmaceutical formulations.

Abstract:The study aims to find out the effectiveness of the combination of citral and curcumin as antifungal compounds. Lemongrass (Cymbopogon citratus) is a valuable family of grass known due to its flavoring, medicinal, and fragrance application. Haldi (Turmeric) scientifically known as Curcuma longa belongs to familthey Zingiberaceae. Its polyphenolic compound curcumin has been showing a variety of antifungal investigations due to extensive traditional uses and very low side effects.Turmeric has beenutilized in traditional medicine for various diseases counting diabetes, hepatitis, hemorrhoids, hysteria, indigestion, skin disease, inflammation, anorexia, hepatic disorders, cough, and sinusitis. In this formulation substudy combination of two bioactive oils is considered to form an effective antifungal spray preparation. The spray preparation is helpful to achieve fast absorption of the drugs through the transdermal way of drug administration. The effectiveness and activity rate of spray preparation is more beneficial. Lemongrass oil and curcumin are dissolved in ethanol to form a stable, safe, and effective spray formulation.
A novel green spectrofluorimetric method for simultaneous determination of antazoline and tetryzoline in their ophthalmic formulation

Abstract:A novel spectrofluorimetric method has been developed for determination of antazoline (ANT) and tetryzoline (TET) in their pharmaceutical formulation. A combined application of synchronous spectrofluorimetry and second derivative mathematical treatment was developed. The proposed method depends on reacting the cited drugs with dansyl chloride (DNS‐Cl) being a suitable derivatizing agent generating highly fluorescent derivatives. A novel spectrofluorimetric method has been developed for determination of antazoline (ANT) and tetryzoline (TET) in their pharmaceutical formulation. A combined application of synchronous spectrofluorimetry and second derivative mathematical treatment was developed. The proposed method depends on reacting the cited drugs with dansyl chloride (DNS‐Cl) being a suitable derivatizing agent generating highly fluorescent derivatives measured at emission wavelengths of 703.0 and 642.0 nm after excitation wavelengths of 350.0 and 320.0 nm for ANT and TET, respectively. The joint use of synchronous spectrofluorimetry with second derivative mathematical treatment is for the first time to be developed and optimized in aid of using fluorescence data manager software generating second derivative peak amplitudes at 556.5 nm for ANT and 516.7 nm for TET. Linear responses have been represented over a wide range of concentration (0.5–12.0 μg/mL for ANT and 0.5–10.0 μg/mL for TET). Additionally, statistical comparison of the developed method with the official ones has been carried out where no significant difference was found. Additionally, greenness profile assessment was accomplished by means of four metric tools. Indeed, the method developed is found to be precise, sensitive, and discriminating to assess the cited drugs for regular analysis.

Abstract:Two simple and sensitive spectrophotometric methods (A and B) for the determination of raloxifene hydrochloride in bulk samples and pharmaceutical formulations are described. Method A is based on the oxidation of the drug with ferric chloride and coupling with potassium ferric cyanide. Method B is based on reduction of the drug with Fehling's reagent. Bluish green color formed in method A absorbs at 735 nm and brown color produced in method B absorbs at 430 nm.
