Abstract:Herein, Cu(NO3)(2)/acetonitrile was proven to be an efficient oxidation system in the oxidation of 5-HMF to DFF in an aqueous solution. After optimization of the reaction conditions, the DFF yield of 97.0% was obtained under the conditions of 1.5 mmol 5-HMF, 1.5 mmol Cu(NO3)(2), 0.48 ml acetonitrile, 10 ml H2O, 80 degrees C, and 18 h. The oxidation system could be recycled at least three times by adding HNO3, which avoided the risk of production of Cu-containing waste. The formation of Cu(CH3CN)(4)(+) in the aqueous solution played an important role in enabling the oxidation of 5-HMF via the redox cycle of Cu2+/Cu+. To further improve the reaction efficiency, a reactive-extraction process was developed by introducing toluene as the organic phase due to its excellent extraction capacity for DFF. As a result, the reaction time was shortened to 8 h, and the highest DFF yield of 98.9% was achieved.
The metabolism and toxicity of furosemide in the Wistar rat and CD-1 mouse: a chemical and biochemical definition of the toxicophore

Abstract:Furosemide, a loop diuretic, causes hepatic necrosis in mice. Previous evidence suggested hepatotoxicity arises from metabolic bioactivation to a chemically reactive metabolite that binds to hepatic proteins. To define the nature of the toxic metabolite, we examined the relationship between furosemide metabolism in CD-1 mice and Wistar rats. Furosemide (1.21 mmol/kg) was shown to cause toxicity in mice, but not rats, at 24 h, without resulting in glutathione depletion. In vivo covalent binding to hepatic protein was 6-fold higher in the mouse (1.57 +/- 0.98 nmol equivalent bound/mg protein) than rat (0.26 +/- 0.13 nmol equivalent bound/mg protein). In vivo covalent binding to mouse hepatic protein was reduced 14-fold by a predose of the cytochrome P450 (P450) inhibitor, 1-aminobenzotriazole (ABT; 0.11 +/- 0.04 nmol equivalent bound/mg protein), which also reduced hepatotoxicity. Administration of [(14)C]furosemide to bile duct-cannulated rats demonstrated turnover to glutathione conjugate (8.8 +/- 2.8%), gamma-ketocarboxylic acid metabolite (22.1 +/- 3.3%), N-dealkylated metabolite (21.1 +/- 2.9%), and furosemide glucuronide (12.8 +/- 1.8%). Furosemide-glutathione conjugate was not observed in bile from mice dosed with [(14)C]furosemide. The novel gamma-ketocarboxylic acid, identified by nuclear magnetic resonance spectroscopy, indicates bioactivation of the furan ring. Formation of gamma-ketocarboxylic acid was P450-dependent. In mouse liver microsomes, a gamma-ketoenal furosemide metabolite was trapped, forming an N-acetylcysteine/N-acetyl lysine furosemide adduct. Furosemide (1 mM, 6 h) became irreversibly bound to primary mouse and rat hepatocytes, 0.73 +/- 0.1 and 2.44 +/- 0.3 nmol equivalent bound/mg protein, respectively, which was significantly reduced in the presence of ABT, 0.11 +/- 0.03 and 0.21 +/- 0.1 nmol equivalent bound/mg protein, respectively. Furan rings are part of new chemical entities, and mechanisms underlying species differences in toxicity are important to understand to decrease the drug attrition rate.
Impact of the induced organic anion transporter 1 (Oat1) renal expression by furosemide on the pharmacokinetics of organic anions

Abstract:Aim: Furosemide is a loop diuretic. Different authors demonstrated that continuous administration of furosemide modulates the expression of organic anion transporters. This study was undertaken to simultaneously evaluate the effects of furosemide pretreatment on organic anion transporter 1 (Oat1) and multidrug resistance protein 2 (Mrp2) renal expressions, on p-aminohippurate (PAH) pharmacokinetics and on renal and urinary PAH levels in rats. Methods: Male Wistar rats were treated with furosemide (6 mg/100 g body weight per day, subcutaneously, 4 days) (treated group) or saline (control group). On the fifth day, PAH was administered as a bolus infusion in the femoral vein, and plasma samples were obtained from femoral artery at different time points. PAH levels in renal tissue and urine were also assessed. Renal Oat1 and Mrp2 expressions were evaluated by western blotting. Results: Furosemide pretreatment increased both the expression of Oat1 and Mrp2. PAH plasma concentrations decreased following a biexponential function. The furosemide-treated group showed higher PAH plasma levels, a lower systemic clearance and elimination rate constant from the peripheral compartment, indicating that PAH renal elimination was decreased. PAH levels in renal tissue were significantly elevated and in urine appeared to be significantly lower as compared with control animals. Conclusions: Furosemide pretreatment caused a significant decrease of PAH renal elimination, despite Oat1 and Mrp2 augmented renal expression. The goal of the present study is the addition of important information in the wide gap of knowledge that exists about drug-drug interactions. Because of furosemide worldwide use, the data obtained are interesting and useful in terms of translation to clinical practice.
Effects of cytochrome P450 inducers and inhibitors on the pharmacokinetics of intravenous furosemide in rats: involvement of CYP2C11, 2E1, 3A1 and 3A2 in furosemide metabolism

Abstract:Objectives: It has been reported that the non-renal clearance of furosemide was significantly faster in rats pretreated with phenobarbital but was not altered in rats pretreated with 3-methylcholanthrene. However, no studies on other cytochrome P450 (CYP) isozymes have yet been reported in rats. Method: Furosemide 20 mg/kg was administered intravenously to rats pretreated with various CYP inducers--3-methylcholanthrene, orphenadrine citrate and isoniazid, inducers of CYP1A1/2, 2B1/2 and 2E1, respectively, in rats--and inhibitors--SKF-525A (a non-specific inhibitor of CYP isozymes), sulfaphenazole, cimetidine, quinine hydrochloride and troleandomycin, inhibitors of CYP2C6, 2C11, 2D and 3A1/2, respectively, in rats. Key findings: The non-renal clearance of furosemide was significantly faster (55.9% increase) in rats pretreated with isoniazid, but slower in those pretreated with cimetidine or troleandomycin (38.5% and 22.7% decreases, respectively), than controls. After incubation of furosemide with baculovirus-infected insect cells expressing CYP2C11, 2E1, 3A1 or 3A2, furosemide was metabolized via CYP2C11, 2E1, 3A1 and 3A2. Conclusions: These findings could help explain possible pharmacokinetic changes of furosemide in various rat disease models (where CYP2C11, 2E1, 3A1 and/or CYP3A2 are altered) and drug-drug interactions between furosemide and other drugs (mainly metabolized via CYP2C11, 2E1, 3A1 and/or 3A2).

Abstract:Furosemide (FRSM) is a cardiovascular drug with high concentration (up to μg L −1 ) in the surface water environment, which are not properly removed by traditional wastewater treatment plants, resulting in negative impact on organisms. In this study, the use of characteristic electron beam (EB) irradiation degraded FRSM with high efficiency. The degradation process precisely conformed to the pseudo first-order kinetic model (R 2 > 0.99). The radical scavenger experiments indicated that •OH was the main free radical for the degradation of FRSM. Adding oxidants such as H 2 O 2 (1 mM) had a positive effect on the degradation of 150 mg L −1 FRSM, while adding more than 10 mM H 2 O 2 resulted in the negative effect. The addition of K 2 S 2 O 8 enhanced the degradation and mineralization of FRSM, with a mineralization rate of 57.1% under 1.5 kGy, which was 2.14 times that of the control group. The degradation of FRSM by EB irradiation in actual water was studied, and the results showed that the presence of NO3− and CO32− (10 mM) inhibited the degradation of FRSM, while the presence of SO32−, SO42− and Cl− slightly accelerated the degradation. The degradation effect of adding tap water and river water was similar to that of the control group. Moreover, transformation products (TPs) were identified (11 types) and FRSM was mainly oxidized and ultimately converted into small molecule acids, inorganic ions and CO 2 . Based on toxicity assessment, most TPs have much lower acute and chronic toxicity than maternal FRSM, suggesting EB irradiation is an alternative advanced treatment process to degrade FRSM in wastewater.

Abstract:This study discloses a benign-by-design synthetic protocol for furosemide using a biomass-derived active pharmaceutical ingredient. The optimized protocol, designed for efficiency, was implemented within an hour using a microwave reactor and a range of environmentally friendly solvents. Investigations were carried out to assess the influence of additives and reaction media on the synthesis process. Green metrics were measured to propose an environmentally sound protocol aiming to minimize waste generation. These findings advance the development of a sustainable and efficient synthesis route for furosemide.

Abstract:The occurrence of pharmaceuticals and pesticides in water is of concern due to their hazardous effect on the environment. Such micropollutants deteriorate environmental quality and can induce antimicrobial resistance in pathogenic bacteria. Thus, removal of these emerging contaminants is a necessity and thus requires a clear understanding of the effective treatment and mechanistic approach utilized in degradation and removal. Frequently, treatment systems are optimized for the removal of contaminants while the formation of transformation products and their removal are not targeted. We utilized our established iron powder catalyzed Fe (0) Fenton Oxidation treatment to study the degradation of emerging contaminants — sulfamethoxazole, gabapentin, diuron, and triazine class pesticides. We used state-of-the-art mass spectrometry to establish the common oxidative mechanistic approach after identifying the transformation products formed during the Fe (0) Fenton Oxidation processes. Fenton transformed product was produced after 60 min reaction time when treatment was initiated after adding the optimized H2O2 and iron powder Fe (0)) in a micropollutant solution adjusted to pH 5. About 5-to-7 transformed products of each micropollutant were identified and predicted to have low toxicity risk to aquatic organisms. The oxidative degradation pathway of micropollutants illustrates the shared oxidative mechanism where the aliphatic chain of the heteroatom: Sulphur (R—S—R as thiosulphate), functional groups (R—C=O, R—COOR, R—NH2, R— NH=O—N) and alkyl chain present in the target compound was suggested as the prime region of hydroxyl radicle (•OH) attack. The tandem mass spectrometry estimated the evolution of sulfamethoxazole-transformed product formation and removal during the treatment. The informed optimization of Fe (0) Fenton treatment, confirmed through MS2 was established at 40 min reaction time at which both the sulfamethoxazole and its transformed product were reduced to trace levels (ngL-1).

3D Model Retargeting Using Offset Statistics

Abstract:Texture synthesis is a versatile tool for creating and editing 2D images. However, applying it to 3D content creation is difficult due to the higher demand of model accuracy and the large search space that also contains many implausible shapes. Our paper explores offset statistics for 3D shape retargeting. We observe that the offset histograms between similar 3D features are sparse, in particular for man-made objects such as buildings and furniture. We employ sparse offset statistics to improve 3D shape retargeting (i.e., Rescaling in different directions). We employ a graph-cut texture synthesis method that iteratively stitches model fragments shifted by the detected sparse offsets. The offsets reveal important structural redundancy which leads to more plausible results and more efficient optimization. Our method is fully automatic, while intuitive user control can be incorporated for interactive modeling in real-time. We empirically evaluate the sparsity of offset statistics across a wide range of subjects, and show our statistics based retargeting significantly improves quality and efficiency over conventional MRF models.

Abstract:The degradation in water of furosemide (FUR), a widely used diuretic drug, was herein reported. The method entails an integrated approach based on the hybridisation of hydrodynamic cavitation (HC) with electrical discharge (ED) plasma technology. This dynamic duo could increase the production of oxidising compounds in water, in particular hydroxyl radicals (OH radicals), by triggering the rapid homolytic decomposition of water molecules and avoiding the addition of external oxidants. This study clearly emphasises the effectiveness of an integrated approach to improve the degradation of pollutants in wastewater originating from active pharmaceutical ingredients (APIs). The results of HC/ED-assisted FUR degradation in the presence of radical scavengers highlight the predominant role of the radical oxidation mechanism at the gas–liquid interface of the cavitation bubble during HC/ED treatment. A comparative analysis of the three technologies—HC alone, HC/ED and UV alone—emphasised the promising potential of hybrid HC/ED as a scalable industrial technology. This is demonstrated by the higher degradation rates (100%, 10 min) when treating large volumes (5L) of wastewater contaminated with FUR (50 mg/L), even in the presence of other APIs.

Electrochemical oxidation of Florfenicol in aqueous solution with mixed metal oxide electrode: Operational factors, reaction by-products and toxicity evaluation

Abstract:Veterinary antibiotics have become an emerging pollutant in water and wastewater sources due to excess usage, toxicity and resistance to traditional water and wastewater treatment. The present study explored the degradation of a model antibiotic- Florfenicol (FF) using electrochemical oxidation (EO) with Ti-RuO2/IrO2 anode. The anode material was characterized using SEM-EDS studies expressing stable structure and optimal interaction of the neighboring metal oxides with each other. The EDS results showed the presence of Ru, Ir, Ti, O and C elements with 6.44%, 2.57%, 9.61%, 52.74% and 28.64% atomic weight percentages, respectively. Optimization studies revealed pH 5, 30 mA cm-2 current density and 0.05 M Na2SO4 for 5 mg L-1 FF achieved 90% TOC removal within 360 min treatment time. The degradation followed pseudo-first order kinetics. LC-Q-TOF-MS studies revealed six predominant byproducts illustrating hydroxylation, deflourination, and dechlorination to be the major degradation mechanisms during the electrochemical oxidation of FF. Ion chromatography studies revealed an increase in Cl-, F- and NO3- ions as treatment time progressed with Cl- decreasing after the initial phase of the treatment. Toxicity studies using Zebrafish (Danio rerio) embryo showed the treated sample to be toxic inducing developmental disorders such as pericardial edema, yolk sac edema, spinal curvature and tail malformation at 96 h post fertilization (hpf). Compared to control, delayed hatching and coagulation were observed in treated embryos. Overall, this study sets the stage for understanding the effect of mixed metal oxide (MMO) anodes on the degradation of veterinary antibiotic-polluted water and wastewater sources using electrochemical oxidation.
Degradation of Losartan in Fresh Urine by Sonochemical and Photochemical Advanced Oxidation Processes

Abstract:In this work, the degradation of the pharmaceutical losartan, in simulated fresh urine (which was considered because urine is the main excretion route for this compound) by sonochemistry and UVC/H2O2 individually, was studied. Initially, special attention was paid to the degrading action of the processes. Then, theoretical analyses on Fukui function indices, to determine electron-rich regions on the pharmaceutical susceptible to attacks by the hydroxyl radical, were performed. Afterward, the ability of the processes to mineralize losartan and remove the phyto-toxicity was tested. It was found that in the sonochemical treatment, hydroxyl radicals played the main degrading role. In turn, in UVC/H2O2, both the light and hydroxyl radical eliminated the target contaminant. The sonochemical system showed the lowest interference for the elimination of losartan in the fresh urine. It was established that atoms in the imidazole of the contaminant were the moieties most prone to primary transformations by radicals. This was coincident with the initial degradation products coming from the processes action. Although both processes exhibited low mineralizing ability toward losartan, the sonochemical treatment converted losartan into nonphytotoxic products. This research presents relevant results on the elimination of a representative pharmaceutical in fresh urine by two advanced oxidation processes.

Acidic Permanganate Oxidation of Sulfamethoxazole by Stepwise Electron-Proton Transfer.

Abstract:Permanganate is a versatile chemical oxidant, and has undergone a dramatic evolution toward deep insight into its reaction mechanism. However, the hydrogen abstraction of the N-H bond by permanganate remains unclear. We studied the permanganate oxidation of the emerging micropollutant sulfamethoxazole in acidic aqueous solution. The reaction followed autocatalytic kinetics and demonstrated first-order with respect to each reactant. The presence of HMnO4 accelerated the reaction rate, which was four orders of magnitude higher than that of MnO4-. Based on the identified products, the rate-limiting step was determined to be simple N-H bond oxidation by metal-oxo species permanganate. The mechanism was then studied computationally by density functional theory (DFT) using ammonia as the simplest model. Results showed that the N-H bond oxidation by MnO4- (32.86 kcal/mol) was a concerted mechanism similar to that of C-H bond oxidation, whereas HMnO4 oxidation of the N-H bond (10.44 kcal/mol) was a stepwise electron-proton transfer. This reminds us that coordination of Bronsted acid could not only produce the stronger electrophile but also change the reaction mode by avoiding the bond cleavage in electron transfer process. Abstract:Fosfomycin (FOS) as a widely used antibiotic has been found in abundance throughout the environment, but little effort has been devoted to its treatment. In this study, we systemically looked into the degradation of FOS by ultraviolet-activated persulfate (UV/PS) in aqueous solutions. Our findings demonstrated that FOS can be degraded efficiently under the UV/PS, e.g., >90 % of FOS was degraded with 19,200 mJ cm(- 2 )of UV irradiance and 20 mu M of PS. HO center dot was the dominant radical responsible for FOS degradation. FOS degradation increased as PS dosage increased, and higher degradation efficiency was observed at neutral pH. Natural water constitutes either promoted (e.g., Cu2+, Fe3+, and SO2/4 ) or inhibited (e.g., humic acid, HCO3, and CO2/3 ) FOS degradation to varying degrees. Hydroxyl substitution, C-P bond cleavage, and coupling reactions were the major degradation pathways for FOS degradation. Finally, the toxicity evaluation revealed that FOS was toxic to E. coli and S. aureus, but the toxicity of the intermediate products of FOS to E. coli and S. aureus rapidly decreased over time after UV/PS treatment. Therefore, these findings provided a fundamental understanding of the transformation process of FOS and supplied useful information for the environmental elimination of FOS contamination and its toxicity.
Isolation and characterization of novel degradation products in Fenoverine by LC-QTOF-MS/MS, LC-MS n and 2D-NMR

Abstract:This study investigates the degradation profile of Fenoverine (FEN), an antispasmodic drug, following the International Conference on Harmonization (ICH) Q1A (R2) guidelines. FEN is stable in the presence of light, humidity, and thermal stress; however, it is highly susceptible to oxidative stress (0.1% H2O2), alkaline stress (0.1 N NaOH), and low susceptibility to acid (1N HCl). Seven degradation products (DPs) were identified using a mass-compatible ultra-high-performance liquid chromatography (UPLC) method: five oxidative degradation products (FOD1 to FOD5) and two alkaline degradation products (FAD1 and FAD2). All DPs, except FOD4, were isolated through preparative HPLC and structurally characterized using high-resolution mass fragmentation, multistage fragmentation, and multidimensional nuclear magnetic resonance (NMR) spectroscopy. Notably, FOD3, FOD4, and FOD5 were identified as isomeric oxidative degradation products (M + 16 Da). Atmospheric pressure chemical ionization (APCI) mass spectrometry and NMR studies pinpointed the oxidation sites, distinguishing FOD3 as FEN's N-oxide, FOD4 as its hydroxyl product, and FOD5 as the sulfoxide. To the best of our knowledge, the discussion of FOD1 to FOD5 and FAD1 in the context of this study appears to be unique and not documented in prior literature. These degradation products were successfully separated from FEN using an Acquity UPLC BEH300 C18 column with 1.7 μm particles as the stationary phase and a mobile phase of 10 mM ammonium acetate in water (pH adjusted to 9.0), with acetonitrile and methanol (80:20, v/v) as co-solvents. In-silico toxicity predictions were conducted, which revealed no structural alerts indicative of mutagenicity for any of the seven degradation products (FOD1 to FOD5, FAD1, and FAD2).

Abstract:Substituted 2,5-dimethoxy-2,5-dihydrofurans ( cis / trans isomer mixtures) were obtained via the electrochemical oxidation of the corresponding furans in an undivided cell in methanol in the presence of bromides as mediators. The method is suitable for furans containing electron-donating groups.

Theoretical Insights into the Transformation Mechanism and Eco-Toxicity Effects of 5-Fluorouracil by O3 and ·OH in Waters

Abstract:The existences of non-biodegradable 5-Fluorouracil (5-FU) in aquatic systems and its potential toxic effects on aquatic organisms have caused widespread concern. In this work, two typical oxidants (O3 and ·OH) were selected to investigate the mechanism, kinetics, and the potential eco-toxicology assessment of 5-FU using computational chemistry methods. Results show that 5-FU can be degraded rapidly by O3 and ·OH, which subsequently undergoes ring-opening, decomposition, defluorination, and hydroxylation steps. The rate constants of initiation reactions are suppressed as the temperature increases. The half-lives of 5-FU determined by O3 and ·OH are on the order of seconds in the advanced oxidation processes (AOPs) system. Fifteen structures of transformation products were identified by theoretical calculation, including five experimental products. The toxicity assessment results show that the acute and chronic toxicities of the degradation process to aquatic organisms gradually decreased, but the developmental toxicity and mutagenicity of several products on human still exist. In addition, the main products have been found to decompose into some small molecules (NO, Formic acid, Acetic acid, etc.). This result could help to reveal the transformation behaviors and risk assessment of 5-FU in aquatic environments, and further design the experimental and industrial infrastructure.
Oxidation of Flumequine in Aqueous Solution by UV-activated Peroxymonosulfate: Kinetics, Water Matrix Effects, Degradation Products and Reaction Pathways.

Abstract:The degradation of flumequine (FLU) in aqueous solution by ultraviolet (UV)-activated peroxymonosulfate (PMS) was investigated in this work. Under the conditions of [PMS](0):[FLU](0) =1:1, T = 25 +/- 2 degrees C, pH = 7.0 +/- 0.1, nearly complete removal of FLU was achieved after 60 min. The effects of various operating parameters, including oxidant doses, pH, the presence of typical ions (NH4+, Mg2+, Fe3+, Cl-, No-3(-), HCO3-) and humic acid were evaluated. It was found that the pseudo-first-order rate constants of FLU degradation increased with increasing PMS dosage and decreasing solution pH. The presence of Mg2+ could accelerate FLU removal, while Fe3+, HCO3-, NO3- and HA inhibited the reaction. Moreover, the degradation of FLU in different water matrices were also explored, and the removal followed the order of Tap water> Ultrapure water> River water> Secondary clarifier effluent. According to the control and radical quenching experiment results, direct photolysis and reactive radicals (SO4 center dot(-) and HO center dot) contributed mainly to FLU degradation in the UV/PMS system. Initial FLU molecule underwent reactions such as hydroxylation, hydroxyl substitution, demethylation, decarboxylation/decarbonylation and ring opening, leading to the formation of nineteen oxidation products. The effective degradation by UV/PMS suggests a feasible technology for treating FLU in waters and wastewaters. (C) 2019 Elsevier Ltd. All rights reserved.

Abstract:The reactivity of furoin with various photogenerated reactive species has been studied. The oxidation of furoin by hydroxyl radicals, produced by photolysis of dilute hydrogen peroxide solutions at 253.7 nm, leads to 5‐hydroxyfuranone, furaldehyde and furoic acid. It was found k5 = 1.3 × 1010 M−1.s−1. The oxygenation of furoin by singlet oxygen, obtained by rose bengal photosensitization, gives rise to 5‐hydroxyfuranone and 1‐furanyl‐2‐dihydroxyethanone with k2 = 1.8 × 108 M−1.s−1. Superoxide anion generated by photolysis of concentrated hydrogen peroxide solutions and RO2 obtained by photolysis of 4,4'‐azobis‐4 cyanovaleric acid in the presence of oxygen reacts with furoin with a very low rate constant. Thus, furoin can be used as a test molecule for hydroxyl radicals and singlet oxygen involvements in aqueous phase photoinduced transformations.
