Detoxification of Tobacco-Specific Nitrosamines in Humans: Association with UGT2B10 Haplotypes.

Abstract:Fifth AACR International Conference on Frontiers in Cancer Prevention Research, Nov 12-15, 2006 A211 The nicotine-derived tobacco-specific nitrosamine (TSNA), 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK), is one of-the most potent and abundant pro-carcinogens found in tobacco and tobacco smoke, and glucuronidation of its major metabolite, 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNAL), by UDP-glucuronosyltransferases (UGTs) is an important mechanism for NNK detoxification. Although several UGTs have been identified as being active NNAL-glucuronidating enzymes, no studies have as yet been performed on UGT2B10, a hepatic enzyme also found in lung. In the present study, we examined the glucuronidation activity of UGT2B10 against NNAL as well as other TSNAs and assessed whether individual UGT2B10 haplotypes are associated with differences in liver microsomal NNAL glucuronidation activities in vitro . HPLC analysis demonstrated that UGT2B10 exhibited significant N -glucuronidating activity against NNAL and other TSNAs. Kinetic analysis demonstrated that UGT2B10-over-expressing cell homogenates exhibits 9.2-fold higher levels of activity against NNAL and between 4-19-fold higher activity against other TSNAs including N'-nitrosonornicotine (NNN), N'-nitrosoanabasine (NAB), and N'-nitrosoanatabine (NAT) as compared to the other known N -glucuronidating enzyme, UGT1A4. Four common (>2% prevalence) haplotypes (termed A , B, C, and D ) were identified in one block of high linkage disequilibrium (LD; D'>0.80) by HapMap screening of the entire UGT2B10 locus, and three haplotype-tagging SNPs were chosen in the present study to distinguish the four haplotypes. The frequencies of haplotypes A, B, C , and D were 63%, 19%, 11%, and 6% in 73 subjects from whom liver specimens were obtained. The C hapalotype was significantly associated with low NNAL N -glucuronidation activity in liver microsomal specimens. Specimens with at least one copy of the C haplotype exhibited a significant ( P <0.001) 44% reduction in NNAL- N -glucuronidation activity as compared to all other subjects. In addition, the C/ C diplotype exhibited a significant ( P <0.001) 11-fold reduction in NNAL- N -glucuronidation activity as compared to subjects without a C haplotype. UGT2B10 expression levels in liver tissues, determined by real time-PCR on cDNA, were not significantly associated with haplotypes. A potentially-functional non-synonomous novel SNP and a synonomous novel SNP were discovered in the UGT2B10 coding region in individuals with a C haplotype. These data suggest that UGT2B10 plays a major role in the glucuronidation and detoxification of a major class of tobacco carcinogens, and that the UGT2B10 haplotype C may be strongly associated with altered TSNA detoxification in smokers.
