Abstract:Seed borne mycoflora associated with ten commercial varieties of wheat viz. Blue silver, Faisalabad 85, Manthar-3, Pak 81, Parwaz 94, Pirsabaq 2005, Punjnad-1, Sariab-92, Sh-2002 and Wafaq-2001 was investigated through standard blotter paper and agar plate method by using Mann-Whitney U test. At least eleven fungal genera were recovered from seeds. The most frequently isolated fungi were Bipolaris sorokiniana (11.125%), Aspergillus flavus (9.825%), Alternaria alternata (7.15%) and Aspergillus niger (6.225%). It is apparent from the present investigation that all commercial wheat verities tested were contaminated by fungi. The rolled paper method was used to find out the effect of seed borne fungi on seed germination. Seeds of Pak 81, Wafaq-2001 and Blue silver were germinated in high proportion with variable number of normal and abnormal seedlings than the seeds of other varieties tested. The fungi associated with seeds of wheat cause dire diseases in wheat reducing the germination capacity.
Anti-inflammatory, analgesic and antipyretic activities of loxoprofen sodium given intramuscularly in animals

Abstract:The evaluation of the anti-inflammatory, analgesic and antipyretic activities of loxoprofen sodium given in intramuscular route was investigated as compared to oral application in rats and mice. The intramuscular ED50 values of loxoprofen sodium in carrageenan edema and vascular permeability tests are 1.15 and 7.8 mg/kg, respectively, which represent more potent than in case of oral application. Its therapeutic effects in adjuvant arthritis were shown at 6 mg/kg i.m. and 3mg/kg p.o. Analgesic effect was shown to be more potent as given intramuscularly. Similar potency of antipyretic effects was shown in both administration routes. Considerably weak gastric damages were observed in intramuscular application.

Abstract:Herbal products are believed to be safe as they are naturally occurring, however, this is a misconception. Currently, there are many herbal products which are not manufactured under the Good Manufacturing Practice (GMP) guidelines and hence the quality of the product is not controlled. Quality can be assessed by evaluation of data on levels of either active constituent(s) or chemical markers indicative of origin of plant material. Assessment of published data on levels of quality of herbal medicinal products revealed that only a percentage of the total number of products investigated were found to contain the levels of markers as stated on the label, and others had widely varying levels of constituents. Thus few products complied with acceptable quality control standards. Herbal products are increasingly self-prescribed for the treatment of minor ailments. Patients relying on herbal products for therapeutic effects may expose themselves to either low doses of active constituents causing insufficient effects or alternatively take higher levels than expected, with the increased risk of toxicity or adverse effects, or be affected by the inadvertent inclusion of unexpected components with associated potential health risks.
Loxoprofen enhances intestinal barrier function via generation of its active metabolite by carbonyl reductase 1 in differentiated Caco-2 cells

Abstract:Nonsteroidal anti-inflammatory drugs (NSAIDs) are used worldwide as antipyretic analgesics and agents for rheumatoid arthritis and osteoarthritis, but known to cause damage to the gastrointestinal mucosae as their serious adverse effects. Few studies showed the impairment of intestinal epithelial barrier function (EBF) by high concentrations (0.5-1 mM) of NSAIDs, but the underlying mechanism is not fully understood. This study is aimed at clarifying effects at a low concentration (50 μM) of three NSAIDs, loxoprofen (Lox), ibuprofen and indomethacin, on intestinal EBF using human intestinal epithelial-like Caco-2 cells. Among those NSAIDs, Lox increased the transepithelial electric resistance (TER) value, decreased the paracellular Lucifer yellow CH (LYCH) permeability, and upregulated claudin (CLDN)-1, -3 and -5, indicating that low doses of Lox enhanced EBF through increasing expression of CLDNs. Lox is known to be metabolized to a pharmacologically active metabolite, (2S,1'R,2'S)-loxoprofen alcohol (Lox-RS), by carbonyl reductase 1 (CBR1), which is highly expressed in human intestine. CBR1 was expressed in the Caco-2 cells, and the pretreatment with a CBR1 inhibitor suppressed both the Lox-evoked CLDN upregulation and EBF enhancement. In addition, the treatment of the cells with Lox-RS resulted in higher TER value and lower LYCH permeability than those with Lox. Thus, Lox-RS synthesized by CBR1 may greatly contribute to the improving efficacy of Lox on the barrier function. Since EBF is decreased in inflammatory bowel disease, we finally examined the effect of Lox on EBF using the Caco-2/THP-1 co-culture system, which is used as an in vitro inflammatory bowel disease model. Lox significantly recovered EBF which was impaired by inflammatory cytokines secreted from THP-1 macrophages. These in vitro observations suggest that Lox enhances intestinal EBF, for which the metabolism of Lox to Lox-RS by CBR1 has an important role.

Abstract:Loxoprofen (Loxonin®, Loxonin® Pap, Loxonin® Tape) is a prodrug-type NSAID that is available in several formulations, including 60 mg tablets, 100 mg hydrogel patches and 50 or 100 mg tape. In active comparator-controlled trials, oral loxoprofen therapy (ranging from 2 days to 6 weeks’ duration depending on the pain type) provided analgesic efficacy that generally did not significantly differ from that of celecoxib for postoperative pain or frozen shoulder, ibuprofen for knee osteoarthritis or naproxen for lumbar pain. In double-blind, double-dummy, multicentre trials, loxoprofen hydrogel patches were noninferior to oral loxoprofen with regard to rates of final overall symptomatic improvement over 1–4 weeks in patients with knee osteoarthritis, myalgia or trauma-induced swelling and pain. Loxoprofen hydrogel patches were also noninferior to other commercially available patches (ketoprofen and indometacin) over 2 or 4 weeks in patients with knee osteoarthritis or myalgia in open-label studies. Oral and topical loxoprofen were generally well tolerated in clinical trials. Thus, loxoprofen is a useful analgesic option for patients with pain and inflammation, with topical loxoprofen potentially reducing the risk of gastrointestinal, cardiovascular and renal complications associated with oral NSAID use.

A new test for evaluating nonsteroidal anti-inflammatory drugs in vitro: inhibition of prostaglandin E2 production in minced intestinal tissue

Abstract:A new method for evaluating the inhibitory effect of nonsteroidal anti-inflammatory drugs on prostaglandin E2 production is presented. Minced rat intestinal tissue was incubated with a nonsteroidal anti-inflammatory drug and homogenized. After centrifugation, prostaglandin E2 in the supernatant was measured by radioimmunoassay. The production of prostaglandin E2 was decreased by nonsteroidal anti-inflammatory drugs, in a concentration-dependent manner, the order of potency being diclofenac greater than loxoprofen greater than indomethacin greater than ibuprofen greater than TA-847 (imidazole derivative). Loxoprofen, a prodrug, inhibited the prostaglandin E2 production. In a classical enzyme assay with sheep seminal vesicle microsomal prostaglandin endoperoxide synthase, the order of inhibitory potency was TA-847 greater than diclofenac greater than indomethacin greater than ibuprofen. The inhibitory effect of loxoprofen was very weak. On the other hand, the potency order of these nonsteroidal anti-inflammatory drugs, with regard to their anti-inflammatory effect as determined in the rat carrageenin-induced paw edema model, was loxoprofen greater than indomethacin greater than diclofenac greater than ibuprofen greater than TA-847. The results of the new method with intestinal tissue showed thus a good correlation with the anti-inflammatory activity in vivo.
Comparison of pharmacokinetics between loxoprofen and its derivative with lower ulcerogenic activity, fluoro-loxoprofen

Abstract:We recently reported that, compared to loxoprofen (LOX, an non-steroidal anti-inflammatory drug), the LOX derivative fluoro-loxoprofen (F-LOX) is less ulcerogenic but has similar anti-inflammatory activity. Our previous in vitro studies suggested that both LOX and F-LOX are pro-drugs, the active metabolites of which are their trans-alcohol forms. In this study, we compared the pharmacokinetics of F-LOX and LOX in rats. Overall, the pharmacokinetic characteristics of F-LOX, including the formation of metabolites in vivo and in vitro, were comparable to those of LOX. However, F-LOX disappeared from the plasma more rapidly than LOX, which could potentially explain its lower ulcerogenicity. However, we showed that F-LOX produced fewer gastric lesions than LOX, even when a higher plasma concentration of F-LOX was maintained. Similar to LOX, F-LOX was readily metabolized to its trans- and cis-alcohol forms, with a higher level of the trans-alcohol form being observed after oral or intravenous administration of the drug. The preferential formation of the trans-alcohol form was also observed after incubation of F-LOX with rat liver homogenates in vitro. These results suggest that, similar to LOX, F-LOX acts as a pro-drug and that there is a metabolic system that selectively produces its active metabolite.
Dose-related anti-inflammatory/analgesic effects of lornoxicam: A spinal c-Fos protein study in the rat

Abstract:Abstract.Objective: To evaluate the anti-inflammatory/analgesic effects of lornoxicam in the carrageenan model of inflammatory nociception.¶Material and Methods: Three hours after intraplantar carrageenan (6 mg/150 μl of saline), we assessed the effects of pre-administered lornoxicam (0.1, 0.3, 1, 3 and 9 mg/kg i.v., n = 10 rats for each group) on both the peripheral oedema and number of c-Fos-protein-like immunoreactive (c-Fos-LI) neurons in the lumbar L4-L5 segments, in the awake rat.¶Results: Lornoxicam dose-relatedly reduced both the carrageenan evoked oedema (r = 0.63 and r = 0.53 for paw and ankle diameter respectively; p<0.001 for both) and total number of spinal c-Fos-LI neurons (r = 0.79; p<0.001), with the strongest effect corresponding to a 75 ± 2% reduction of the number of c-Fos-LI neurons (p<0.001) for the highest dose (9 mg/kg), and a 45 ± 3% reduction (p<0.001) for the low dose of 0.3 mg/kg. Reductions of both the peripheral oedema and spinal c-Fos expression were correlated (r = 0.74 and r = 0.57 for the paw and ankle diameter respectively; p<0.001 for both).¶Conclusions: Our results demonstrate that lornoxicam reduces in parallel both the carrageenan-evoked oedema and spinal c-Fos expression, with clear evidence for a potent effect of low doses of lornoxicam. Correlated reductions in c-Fos expression and paw oedema suggest a predominantly peripheral site of action of lornoxicam.

Abstract:An invitation from Eduardo Marban, on the occasion of the 50th anniversary of Circulation Research , provides an opportunity to reflect on my understanding of the most important advance in regulation of cardiac metabolism. By the early 1950s, the major findings were the observation by Locke and Rosenheim in 1907 that glucose is used as a fuel by the isolated perfused heart and the observation by Bing and coworkers in 1954 that normally beating human hearts prefer fatty acid to any other substrate for oxidation. Bing’s in situ experiments involved measurements of the myocardial extraction of various substrates and oxygen and of coronary flow. Although there is no substitute for studying metabolism of the heart working in a normal environment, these studies do not allow for the exploration of individual enzymatic reactions. The most important advance in understanding the regulation of cardiac metabolism was the discovery, in the post–World War II era, of the pathways of glycolysis and fatty acid metabolism including the role of membrane transport in glucose uptake. Although heart muscle slices and homogenates were used for studies of glycolysis, some or all of the cardiac muscle cells were disrupted and prevented rigorous studies of membrane permeability. I began my research career in 1953 to test the hypothesis that the effect of insulin on glycolysis in muscle is due to acceleration of glucose transport into the cell. After completing medical school at Johns Hopkins and a residency in Obstetrics and Gynecology at Vanderbilt, I joined the laboratory of Charles R. Park, Professor of Physiology at Vanderbilt. My long-term goal was to define the mechanisms of placental transport of nutrients. Abstract:1. Loxoprofen (LX), is a prodrug of the pharmacologically active form, trans-alcohol metabolite (trans-OH form), which shows very potent analgesic effect. In this study, the pharmacokinetics and metabolism of [(14)C]LX-derived radioactivity after dermal application of [(14)C]LX gel (LX-G) to rats were evaluated. 2. The area under concentration-time curve (AUC0-∞) of radioactivity in the plasma after the dermal application was 13.6% of that of the oral administration (p < 0.05). 3. After the dermal application, the radioactivity remained in the skin and skeletal muscle at the treated site for 168 h, whereas the AUC0-168 h of the radioactivity concentration in every tissue examined except the treated site was statistically lower than that after the oral administration (p < 0.05). 4. The trans-OH form was observed at high levels in the treated skin site at 0.5 h. Metabolite profiles in plasma, non-treated skin site and urine after the dermal application were comparable with those after the oral administration. 5. Renal excretion was the main route of elimination after the dermal application. 6. In conclusion, compared to the oral administration, the dermal application of [(14)C]LX-G showed lower systemic and tissue exposure with higher exposure in the therapeutic target site. The radioactivity revealed similar metabolite profiles in both administration routes.

Abstract:We report on space-selective growth of a second-harmonic-generation beta-BaB(2)O(4) (BBO) crystal inside a BaO-Al(2)O(3)-B(2)O(3) glass sample at the focal point of an 800-nm femtosecond laser beam. A spherical heated region was formed during the focused laser irradiation through observation with an optical microscope. We moved the heated region by changing the position of the focal point of the laser beam relative to the glass sample. We grew BBO crystal continuously in the glass sample by adjusting the moving speed of the heated zone. Our results demonstrate that functional crystals can be formed three dimensionally in glasses by use of a nonresonant ultrashort pulsed laser.
Analgesic effect of S (+)-flurbiprofen plaster in a rat model of knee arthritis: analysis of gait and synovial fluid prostaglandin E2 levels

Abstract:Objectives: We developed S (+)-flurbiprofen plaster (SFPP), a novel NSAID patch containing S (+)-flurbiprofen (SFP), a potent cyclooxygenase (COX) inhibitor. The purpose of this study was to assess efficacy of SFPP by analysing its effect on the gait disturbance and measuring the prostaglandin E2 (PGE2 ) production in synovial fluid in a rat model of knee arthritis. Methods: Knee inflammation was induced in rats by intra-articular injection of a yeast suspension. Subsequently, an NSAID patch containing SFP, ketoprofen or loxoprofen was applied over the affected knee. Gait was assessed at 2, 4 and 6 h after application of the patch. The PGE2 concentration in the synovial fluid was measured after the gait assessment. Key findings: Application of SFPP (0.125, 0.25, 0.5 or 1 mg/sheet) was followed by a decrease in the visual gait score at all the doses examined. In the case of the other two NSAID patches, only the ketoprofen patch (1 or 2 mg/sheet) and loxoprofen patch (5 mg/sheet) produced a decrease in the visual gait score. All of the NSAID patches decreased the PGE2 production in the synovial fluid. Conclusions: These results suggest the potential usefulness of SFPP as an analgesic patch in patients with inflammatory joint pain.
Anti-inflammatory Effects and Gastrointestinal Safety of NNU-hdpa, a Novel Dual COX/5-LOX Inhibitor.

Abstract:Nonsteroidal anti-inflammatory drugs (NSAIDs) are associated with a risk of serious adverse events. Now, the development of dual inhibitors of cyclooxygenase (COX) and 5-lipoxygenase (5-LOX) has become a hot area in searching for safer NSAIDs. NNU-hdpa, 2-(4-hydroxylphenyl)-3-(3,5-dihydroxylphenyl) propenoic acid, a newly synthesized compound, is expected to have COX/5-LOX dual inhibition with an improved gastrointestinal profile. In this study, NNU-hdpa was subjected to in vitro and in vivo experiment protocols. In vitro COX/5-LOX inhibition assays showed that NNU-hdpa exhibits a dual inhibitory activity against the COX and 5-LOX enzymes. Anti-inflammatory activity in vivo was evaluated using two animal edema model tests. Pretreatment with NNU-hdpa (p.o.) dose-dependently inhibited the xylene-induced ear edema in mice and carrageenan-induced paw edema in rats respectively. In gastric lesion test, NNU-hdpa was gastric-sparing in that it elicited markedly fewer stomach lesions as compared to the stomach lesions caused by aspirin in rats. In further studies, NNU-hdpa was found to significantly inhibit the productions of PGE2 and LTB4 in LPS-challenged RAW 264.7, which is parallel to its prevention of the nuclear translocation of the NF-κB p50 and p65 subunits. These data indicate that NNU-hdpa comprises a novel class of dual inhibitors of COX and 5-LOX having therapeutic potential with an enhanced gastric safety profile.
[Persistent infection of pig embryo kidney cell cultures caused by a variant of tick-borne encephalitis virus].

Abstract:The results of modelling and study of persistent infection of pig embryo kidney cell cultures (PEK) with MF variant of tick-borne encephalitis (TBE) virus are presented. In the life of chronically infected cultures periods of formation and stabilization were observed which were characterized by different types of relationships between the persisting virus and the cells. By the characters under study the persisting virus did not differ significantly from the initial MF variant. Inoculation of PEK and BHK-21 cell monolayers with samples of DNA from chronically infected cultures permitted to isolate transfected agents. Their identification by the neutralization tests and the indirect immunofluorescence procedure confirmed that they belonged to TBE virus. It is suggested that the mechanism of TBE virus persistence in PEK-MF system is based on the integration of TBE virus genome with DNA of chronically infected PEK cells.
Paeonol Inhibits Il-1 Beta Induced Expression Of Inos, Cox-2, And Mmps Through Nf-Kappa B Activation: An In Vitro And In Vivo Study

Abstract:Introduction: Paeonol (2'-hydroxy-4'-methoxyacetophenone) is a common traditional Chinese remedy that possesses diverse biological activities, including anti-inflammatory properties. Osteoarthritis (OA) is a degenerative joint disease with an inflammatory component that drives the degradation of cartilage extracellular matrix. In the present study, we investigated the anti-inflammatory properties of paeonol in chondrocytes. Methods: We assessed the effects of paeonol on MMPs, iNOS and COX-2 mRNA expression by quantitative real-time PCR and western blot in IL-1 beta-induced rabbit chondrocytes and evaluated the in vivo effects of paeonol in an experimental OA model in rabbits. The NO production was determined by Griess method, the PGE(2) production was detected by ELISA. Western blot and PCR were performed to investigate the protein level of inhibitor of I kappa B-alpha and the translocation of NF-kappa B. Results: We found that paeonol inhibited the production of PGE(2) and NO induced by IL-1 beta and significantly decreased IL-1 beta-stimulated gene expression and production of MMPs, iNOS and COX-2 in rabbit chondrocytes. Paeonol inhibited IL-1 beta-mediated activation of NF-kappa B by suppressing degradation of I kappa B alpha in the cytoplasm. In rabbits, morphological and histological analyses determined that paeonol decreased cartilage degradation. Downregulation of MMP-1, MMP-3, MMP-13, iNOS and COX-2 expression and up-regulation of TIMP-1 expression were also detected in paeonol-treated cartilage compared with the control group, confirming these findings in an in vivo model. Conclusions: These findings suggest that paeonol may be a potential agent in the treatment of OA.

Abstract:Epidemiological surveillance of Hepatitis B surface antigen in the city of Hyderabad at our Institute shows that the incidence of HBsAg is considerably high among the general population. In a comparative evaluation of the two techniques used, RPHA and ELISA, the latter emerged as a reliable one with high specificity and sensitivity. Screening of all blood samples by ELISA is imperative for uniform reporting of the incidence. Non risk groups like voluntary blood donors, cardiac patients serve as potential reservoirs of HBV infection.

Abstract:Here we report the inhibitory effects of nine non-steroidal anti-inflammatory drugs (NSAIDs) on soybean 15-lipoxygenase (15-LOX) enzyme (EC 1.13.11.12) by three different methods; UV-absorbance, colorimetric and chemiluminescence methods. Only two drugs, Ibuprofen and Ketoprofen, exhibited enzyme inhibition by UV-absorbance method but none of the drug showed inhibition through colorimetric method. Chemiluminescence method was found highly sensitive for the identification of 15-LOX inhibitors and it was more sensitive and several fold faster than the other methods. All tested drugs showed 15-LOX-inhibition with IC50 values ranging from 3.52 ± 0.08 to 62.6 ± 2.15 µM by chemiluminescence method. Naproxen was the most active inhibitor (IC50 3.52 ± 0.08 µM) followed by Aspirin (IC50 4.62 ± 0.11 µM) and Acetaminophen (IC50 6.52 ± 0.14 µM). Ketoprofen, Diclofenac and Mefenamic acid showed moderate inhibitory profiles (IC50 24.8 ± 0.24 to 39.62 ± 0.27 µM). Piroxicam and Tenoxicam were the least active inhibitors with IC50 values of 62.6 ± 2.15 µM and 49.5 ± 1.13 µM, respectively. These findings are supported by expression analysis, molecular docking studies and density functional theory calculations. The expression analysis and flow cytometry apoptosis analysis were carried out using mononuclear cells (MNCs) which express both human 15-LOX and 5-LOX. Selected NSAIDs did not affect the cytotoxic activity of MNCs at IC50 concentrations and the cell death showed dose dependent effect. However, MNCs apoptosis increased only at the higher concentrations, demonstrating that these drugs may not induce loss of immunity in septic and other inflammatory conditions at the acceptable inhibitory concentrations. The data collectively suggests that NSAIDs not only inhibit COX enzymes as reported in the literature but soybean 15-LOX and MNCs LOXs are also inhibited at differential values. A comparison of the metabolomics studies of arachidonic acid pathway after inhibition of either COX or LOX enzymes may reconfirm these findings.

Abstract:The arachidonic acid metabolizing enzymes cyclooxygenase-2 (COX-2) and 5-lipoxygenase (5-LOX) have been considered to be associated with the occurrence and development of a variety of diseases, including inflammatory. Compared to classical NSAIDs, the dual COX-2/5-LOX inhibition is an effective strategy for designing compounds with more effective biological activities, fewer side effects, and a broader anti-inflammatory spectrum. In this study, we designed and synthesized a series of novel indole and indazole arylamide benzoic acid analogues as dual COX-2 / 5-LOX inhibitors and evaluated their anti-inflammatory properties in vivo. Compounds 7f and 7n showed significant anti-inflammatory activity in a xylene-induced mouse model of auricular edema. Furthermore, 7f and 7n exhibited moderate COX-2 inhibitory activity (IC 50 = 537 and 321.5 nM) than celecoxib (IC 50 = 10.04 nM) in vitro, among which 7n had higher COX-2 selectivity activity (selectivity index (COX-1/COX-2) = 7.89) and moderate 5-LOX inhibitory activity (IC 50 = 222.1 nM). Compared to zileuton (IC 50 = 36.46 nM), compound 7f was identified as the most potent 5-LOX inhibitor (IC 50 = 77.37 nM). According to the biological results, compounds 7f and 7n have better inhibitory activities on the production of NO and PGE 2 in LPS-induced RAW 264.7 cell macrophages than celecoxib and indomethacin. As demonstrated by docking studies, 7f and 7n have stronger interactions with key residues in the active pocket of COX-1 or COX-2, which is consistent with the activity results. Based on these results, further research into safer and more effective anti-inflammatory drugs might be possible using indole arylamide benzoic acid analogs.
