What problem does this paper attempt to address?
The paper aims to present the latest research progress on ion channel functions and their dysfunctions in diseases. Specifically, the paper explores the following aspects: 1. **Functions and Diversity of Ion Channels**: Ion channels play a crucial role in maintaining normal physiological balance, participating in signal transduction, transepithelial transport, regulation of nerve function, and maintenance of cell volume and ion concentration. The research focuses not only on proteins expressed in humans but also on ion channels expressed in other animal phyla to better understand the relationship between the structure and function of these key transporters. 2. **Channelopathies**: This is a heterogeneous group of diseases caused by genetic or acquired factors (such as drugs and toxins), mainly due to ion channel dysfunctions. The most common is the dysfunction caused by gene mutations encoding ion channels. The paper emphasizes the importance of the ability to distinguish whether gene sequence changes are benign (polymorphism) or pathogenic. 3. **Roles of Ion Channels in Specific Diseases**: - **Epilepsy**: The research points out the roles of potassium channel subfamily T member 1 (KCNT1) and α4β2 neuronal nicotinic acetylcholine receptor in epilepsy. Specific gene mutations in these two channels lead to an imbalance in nerve excitability. - **Neuropathic Pain**: The research explores the potential of the natural product Narirutin to reverse peripheral neuralgia by blocking the voltage - gated sodium channel Nav1.7. - **Cystic Fibrosis (CF)**: Besides pulmonary dysfunction, CF also involves gastrointestinal dysfunction, which affects the nutritional status of patients and, in turn, their morbidity and mortality. The research investigates the changes in the essential fatty acid status in a CF rat model and its impact on metabolic enzyme and fatty acid transporter genes. 4. **Development of Research Methods**: Traditional electrophysiological methods are less efficient in analyzing ion channel mutations, while the development of high - throughput sequencing and proteomics techniques allows for detailed analysis of molecular mutations related to diseases, which may help improve the diagnostic accuracy of ion channelopathies related to genetic mutations. In conclusion, by reviewing the latest research progress, this paper aims to deepen the understanding of the relationship between ion channel structure and function and explore new treatment methods and technical means to deal with various diseases caused by ion channel dysfunctions.
