Abstract:A phytochemical investigation on the roots and stems of Litsea cubeba led to the isolation of seven isoquinolone alkaloids. By spectroscopic analysis and comparison of their 1H and 13C-NMR data with those in literatures, these alkaloids were identified as (+)-norboldine (1), (+)-boldine (2), (+)-reticuline (3), (+)-laurotetanine (4), (+)-isoboldine (5), (+)-N-methyl-laurotetanine (6), and berberine (7), respectively. Among them, 7 was isolated from the genus for the first time. The evaluation of these compounds showed weak anti-inflammatory activity against NO production in RAW 267.4 and BV-2 cells.

Abstract:A new bisbenzylisoquinoline, lancifoliaine (1), together with seven known alkaloids--N-allyllaurolitsine (2), reticuline (3), actinodaphnine, norboldine, pallidine, cassythicine and boldine--were isolated from the stem bark of Litsea lancifolia (Lauraceae). In addition to that of lancifoliaine, complete ¹³C-NMR data of N-allyl-laurolitsine (2) was also reported. The alkaloidal structures were elucidated by means of high field 1D- and 2D-NMR IR, UV, and LCMS-IT-TOF spectral data. N-Allyllaurolitsine (2) showed a moderate vasorelaxant activity on isolated rat aorta.

Abstract:Phytochemical investigation on the stems of Alstonia scholaris (L.) R. Br. led to the isolation and identification of 30 natural products, including sixteen alkaloids (1–16), nine triterpenoids (17–25), two phenols (26–27), and three lignans (28–30). Their structures were established by comprehensive spectroscopic analyses together with the comparison with reported data. Compound 1 was a new indole alkaloid. Compound 24 was firstly isolated from Apocynaceae family, compounds 22–23 and 26–30 were obtained from the genus Alstonia for the first time, and this is the first report of the presence of 11 and 16 in A. scholaris. The chemotaxonomic significance of these isolated compounds was also discussed.

Abstract:The study aims to investigate the Isolation, Characterization & Antihypertensive Life of Natural Alkaloids out of certain Selected Plants. The Alstonia scholaris papers used in this study are generally available in the tropics and can be obtained in Asia. The plant sample was verified by the pharmacognosy and pharmacology department. The powdered leaves of Alstonia scholaris (500 gm) are macerated using 1% HCl (pH 2) at space temperature overnight. After that, the combination was produced alkaline by putting 25% NH4OH solution (pH 9). The combination's color changed from the red wine to the black. The alkaline mixture was then bounced satisfactorily and purified using Whatman filter paper. Four fractions (15-19) were collected from column chromatography. All the fractions have shown the same Rf value in the TLC fingerprint, therefore they are incorporated established on TLC analysis generated in Hexane: Ethyl acetate (14:6). Nitric oxide synthase inhibitor, i.e. N-nitro-L-arginine methyl ester was used to produce hypertension in rats in (40 mg/ml/kg, i.p.). Every day, it is solubilized in 0.9 per cent NaCl solution. Colourless powder compound was obtained (yield 0.4%) and having MP 132-1340 C. Rf value in (Hexane: Ethyl acetate,65:35) at 0.55, UV-Vis λmax in methanol: (nm) 297, IR (KBr), m 913 (N-H bending), 1260 (C-N Stretching), 1396 (C-N), 1165, 1259 (-C-O- stretching) 1396, 1464 (C=C, Ar.), 2831, 2928 (C-H, Aliphatic) and 3564, 3315 (N-H Stretching). The 1H NMR spectrum also portrayed the distinctive peaks for various chemical compounds. The peak of 7.28-8.85 ppm was due to multiple aromatic protons. The 6.94-7.04 ppm peaks were characteristic of ethylene amino protons, and the 1.57-2 ppm peaks were allocated to alcohol protons. L-NAME significantly elevated MABP, SBP, and DBP in pentobarbital-anesthetized rats but not HR. The mean arterial blood pressure, systolic blood pressure and diastolic blood pressure of pentobarbital-anaesthetized L-NAME caused hypertensive rats do not alter after a single intragastric injection of the isolated alkaloid. Finally, isolated alkaloids from Alstonia scholaris supplement had antihypertensive properties in hypertensive rats.

Abstract:Eight alkaloids, including a new compound, (+)-norboldine acetate (1), and seven known ones, (+)-norboldine (2), (+)-boldine (3), (+)-laurotetanine (4), (+)- N-methyllaurotetanine (5), (+)-reticuline (6), (–)-pronuciferine (7), and pallidine (8) were isolated from the roots of Lindera aggregata. The structures of these alkaloids were determined by spectroscopic and chemical methods, especially 2D NMR techniques, which also allowed the first full NMR assignments of alkaloids 2, 4 and 5. Among them, the 1D NMR chemical shifts of (+)-norboldine (2) showed a remarkable environmental sensitive behavior. All the alkaloids were tested in cytotoxicity assays against L1210 and K562 tumor cell lines; only (+)-norboldine (2) showed weak activity against the L1210 cell line.

Abstract:Two novel aporphine-derived alkaloids, aporaloids A and B (1 and 2), together with eight known biogenetically related alkaloids (3−10), two known isoquinoline alkaloids (3 and 4), and six known aporphinoid alkaloids (5–10) were isolated from the stems of Fissistigma glaucescens. Their structures were elucidated using comprehensive spectroscopic methods. Compounds 1 and 2 represent the rare example of a six-membered lactone ring aporphine-derived alkaloids from natural products. The inhibitory activities of all compounds against four cancer cell lines were evaluated. Aporaloids A and B (1 and 2) showed broad spectrum cytotoxic activities.

Abstract:A new monoterpene and a new lignan, named litsecols A and B (1 and 2), respectively, together with nine known compounds (3-11), were isolated in a continuous investigation on the roots and stems of Litsea cubeba. Their structures were elucidated on the basis of extensive spectroscopic data analysis, and the absolute configuration of 1 was resolved by X-ray diffraction analysis. Compounds 2-5 and 7-9 showed significant inhibitory activity against nitric oxide (NO) production in lipopolysaccharide (LPS)-induced murine microglial (Bv-2) cell line. Compounds 10 and 11 exhibited significant neuroprotective effect against hydrogen peroxide-induced oxidative damage in rat adrenal pheochromocytoma (PC12) cell line.

Abstract:Two new aminoethylstilbene isoquinoline alkaloids denoted litsine B (1) and Litsine C (2) were isolated from an ethanol extract from the root bark of Litsea glutinosa. The structures of the new chemical entities were established using 1D and 2D NMR spectroscopy in combination with high resolution mass spectrometry. Both compounds were tested for their effect on glucose consumption in HepG2 cells at different concentrations. The results showed that compound 2 significantly increased the glucose uptake.

Abstract:The petroleum ether, chloroform and ethyl acetate soluble fractions of methanolic extract of Litsea lancifolia Roxb. leaves were subjected to different pharmacological screenings to explore its potential as anti-oxidant, antimicrobial, peripheral analgesic, hypoglycemic and CNS depressing agent. The ethyl acetate soluble fraction showed highest total phenolic content and free radical scavenging activity compared to the standard, acetyl salicylic acid. Potential antimicrobial activity was shown against P. aeruginosa (23.50 mm), E. coli (22.33 mm), B. cereus (18 mm) and S. paratyphi (18 mm). The crude extract demonstrated significant peripheral analgesic (p < 0.01) activity with 69.45 and 77.96% inhibition of acetic acid-induced writhing at 100 and 200 mg/kg b.w., respectively. The crude methanolic extract also showed significant hypoglycemic activity (p < 0.01) at a dose of 500 mg/kg/day on the 7th day of treatment. All the organic soluble fractions exhibited noteworthy (p < 0.001) CNS depressant activity. Taken together, the plant can be considered as a good material for further chemical investigation to isolate the bioactive constituents.

Abstract:Abstract Lime flower (Tiliae flos) is traditionally used either for treatment of the common cold or to relieve symptoms of mental stress. Recently, the presence of a new class of piperidine and dihydro-pyrrole alkaloids from lime flower has been described. The present study aimed to investigate the pharmacological activity of hydroacetonic lime flower extracts, alkaloid-enriched lime flower fractions, and isolated alkaloids on the murine airway smooth muscle and the cholinergic system. While a hydroacetonic lime flower extract did not show any pharmacological activity, enriched Tilia alkaloid fractions potentiated acetylcholine-induced contractions of the trachea by ~ 30%, showing characteristics comparable to galanthamine. Effects were abrogated by atropine, indicating an involvement of muscarinic receptors. The dihydro-pyrrole alkaloid tiliine A, the piperidine alkaloid tiliamine B, and the acetylated piperidine alkaloid tilacetine A were characterized as acetylcholinesterase inhibitors. The positive control galanthamine (IC50 = 2.0 μM, 95% CI 1.7 to 2.2 μM) was approximately 100 times more potent compared to tiliine A (IC50 = 237 μM, 95% CI 207 to 258 μM) and tiliamine B (IC50 = 172 μM, 95% CI 158 to 187 μM). Neither DNA synthesis of HepG2 liver cells, HaCaT keratinocytes, and Caco-2 intestinal epithelial cells nor cell viability of primary human fibroblasts was reduced by the alkaloids. The indirect cholinergic activity of the alkaloids might explain some aspects of the traditional use of lime flowers and may extend the portfolio of compounds with regard to diseases involving parasympathetic malfunction or central cholinergic imbalance.

Abstract:Two previously undescribed monoterpenoid indole alkaloids, N -formylyunnanensine ( 1 ) and scholaphylline ( 2 ), along with eight known alkaloids, namely, 19,20- E -vallesamine ( 3 ), 19,20- Z -vallesamine ( 4 ), 19,20- E -vallesamine N -oxide ( 5 ), 6,7- seco -19,20α-epoxyangustilobine B ( 6 ), 16 R -19,20- E -isositsirikine ( 7 ), N -demethylalstogustine N -oxide ( 8 ), E -vallesiachotamine ( 9 ), and Z -vallesiachotamine ( 10 ), were isolated from the bark of Alstonia scholaris. Alkaloid 1 was determined to be the N -formyl derivative of the known alkaloid, yunnanensine, comprising a pair of inseparable E / Z -formamide rotamers, while scholaphylline ( 2 ) was identified as the first member of the seco stemmadenine-yunnanensine type bisindole alkaloid. The structures of these alkaloids and their absolute configurations were established based on detailed analysis of the spectroscopic data in conjunction with the TDDFT-ECD method. A possible biogenetic pathway to 1 and 2 , involving stemmadenine as the primary precursor, was proposed. Scholaphylline ( 2 ) showed modest cytotoxicity against Panc1 (IC 50 27.7 ± 5.1 μM), MDA-MB-231 (IC 50 32.0 ± 2.2 μM), and MDA-MB-468 (IC 50 34.1 ± 3.3 μM) cancer cell lines, while N -formylyunnanensine ( 1 ) did not show appreciable cytotoxicity against any of the tested cell lines.

Abstract:Lysiphyllum binatum (Blanco) de Wit in the Fabaceae family, despite its traditional medicinal uses, has not been the subject of prior scientific inquiry into its chemical and biological profile. The dichloromethane and MeOH extracts of its roots exhibited notably similar antioxidant activity, while the dichloromethane extract of the vine stems showed aromatase inhibition. This study aimed to identify the bioactive components responsible for these activities. Chemical investigation of the roots led to the isolation of six new metabolites, named lysiphans A-F (1-6), along with eight known compounds (7-14). The vine stem yielded lysiphan C (3) and compound 7, as well as five known isolates (15-19). The structures of these metabolites were determined through NMR spectral analysis, HRESIMS, quantum chemical calculations of NMR and ECD spectra, and Mosher's modifications to establish their absolute configurations. The biogenetic relationships between the new compounds were proposed. Several of the isolates were evaluated for their antioxidant, anti-aromatase, and cytotoxic properties. Lysiphan B (2) exhibited significant antioxidant activity, with an IC50 value of 28.8 ± 0.4 μM in the diphenyl picrylhydrazyl radical (DPPH) assay, 3.5 ± 0.2 μM in the xanthine/xanthine oxidase (XXO) assay, and 1.5 ± 0.0 ORAC units in oxygen radical absorbance capacity (ORAC) assay. Additionally, compounds 12, 13, and 16 exhibited very strong aromatase inhibitory activity with IC50 values of 0.3 ± 0.2, 4.7 ± 0.1, and 0.9 ± 0.2 µM, respectively. Compound 16 also demonstrated strong ORAC activity of 1.9 ± 0.1 ORAC units.

Abstract:Three undescribed aporphine alkaloids laurodionine B (1), illigerine A (2), and N-formyl-laurolitsine (3) were isolated from the methanolic extracts of the Chinese medicinal plant, Illigera aromatica, together with three known analogues (4-6). The chemical structures of 1-6 were identified by spectroscopic methods including 1D and 2D NMR (H-1, C-13, COSY, HSQC, and HMBC) and high resolution mass spectrometry (HRESIMS). Compounds 1-3 showed moderate inhibitory activities in vitro against two cultured tumor cell lines, Hela and SMMC7721, with IC50 values of 32.42-62.90 mu M. Only compound 1 had in vitro cytotoxic activity against Bcap37 cells, with the IC50 value of 90.61 mu M.

Abstract:Aim Investigation of alkaloids from the leaves of Brazilian medicinal plant Croton hemiargyerius var.gymnodiscus. Methods Silica gel column chromatography was used repeatedly for the isolation and purification, and their structures were identified by extensive spectroscopy and comparison of the chemical and physical data with those of authentic samples reported in literature. Results Twelve alkaloids were isolated and their structures were identified. Conclusion Four new alkaloids named hemiargines A (1), B (5), C (6) and D (7), together with eight known alkaloids namely isocorydine (2), corydine (3), norcorydine (4), salutaridine (8), glaucine (9), tetrahydropalmatrubine (10), xylopinoine (11), and norlaudanosine (12) were isolated.

Abstract:A pair of new enantiomeric alkaloids (+)- 1 and (–)- 1 , along with eight known compounds ( 2–9 ) were isolated from the stem of Tinospora sinensis (Lour.) Merr. Their structures were elucidated on the basis of comprehensive analysis of HRESIMS and NMR spectroscopic data as well as CD spectroscopy methods. In addition, the isolated compounds were evaluated for their anti-inflammatory activity. Compounds (+)- 1 and (–)- 1 inhibited the release of NO from RAW264.7 cells induced by LPS with the IC 50 values of 0.34 ± 0.02 μM and 3.37 ± 0.24 μM, respectively.

Abstract:Phytochemical investigation of Nauclea latifolia Smith. (Rubiaceae), an African ethnomedicinal plant, yielded a new indolo[2,3-a]quinolizidine alkaloid, nauclealatimide (1) , two previously unknown lignans named nauclealignan A (2) and B (3) , as well rhemaneolignan B (4) and spicatolignan A (5) from the hydromethanolic extract obtained after ultrasonication of the stem bark. Their chemical structures were elucidated using IR, NMR (1D and 2D) and HRESIMS spectral evidence as well as comparisons with existing literature data.

Abstract:The methanol stem bark extract of A. boonei and methanol seed extract of P. nitida , were subjected to purification using chromatographic techniques. A. boonei yielded loganic acid ( 1 ), sweroside ( 2 ) and secoxyloganin ( 3 ), while P. nitida afforded (1) , akuammidine ( 4 ), akuammicine ( 5 ) and alstonine ( 6 ). The structures of the compounds were elucidated based on their nuclear magnetic resonance (NMR), high-resolution mass spectrometry (HRMS) profiles and comparison with literature data. The antibacterial activities of the compounds were evaluated using the disc diffusion assay with chloramphenicol as the positive control. Alstonine ( 6 ) demonstrated weak activity against Pseudomonas aeruginosa and Streptococcus agalactiae with zones of inhibition of 9.3 ± 0.6 and 10.0 ± 0.0 mm, respectively. This is the first report of sweroside ( 2 ) and secoxyloganin ( 3 ) in A. boonei .
