Abstract:Carbamazepine (CBZ) is an anticonvulsant with very low water solubility, presenting as a white crystalline powder with poor mechanical properties and is hard to bend. To enhance CBZ's physicochemical properties, such as water solubility and mechanical properties, we selected six cocrystal coformers (CCFs): nicotinamide (NIC), benzamide (BZM), salicylic acid (SCA), fumaric acid (FMA), trimesic acid (TMA) and hesperetin (HPE). Six CBZ cocrystals were successfully prepared using the solution method. Fourier transform infrared spectroscopy (FT-IR), powder X-ray diffraction (PXRD), differential scanning calorimetry (DSC) and single crystal X-ray diffraction (SCXRD), were used to characterize the crystal structures and gain comprehensive insights into the six cocrystals. The mechanical, fluorescence and solubility properties of the six cocrystals were tested. The results reveal that most of the prepared cocrystals exhibit improved water solubility and mechanical properties when compared to CBZ. Among them, the dissolution rate of cocrystals excluded CBZ-HPE has increased by an average of 3 or 4 times compared to CBZ, while CBZ-HPE exhibit superior mechanical properties. Moreover, all the six cocrystals possess better fluorescence performance than CBZ. We thoroughly evaluated the mechanical properties of the cocrystals through both experimental and theoretical approaches. This work provides a new direction for studying drug cocrystals to improve the physicochemical properties of drugs.

Abstract:ABSTRACT: Poor physicochemical properties like solubility, dissolution profile, stability, etc. of drugs remain as an obstacle to formulation development. In the last two decades, pharmaceutical cocrystal gaining lots of interest from researchers as these physicochemical, mechanical, and pharmacokinetic properties can be altered without compromising the therapeutic effect. The multi-component systems wherein one component is essentially the Active Pharmaceutical Ingredient and another may be either one more drug or coformer are termed as pharmaceutical cocrystal. Cocrystal is a novel, promising, and emerging approach to encountering such problems and improving the performance of pharmaceuticals. The present article describes the types, rationale of design, synthesis methods, and characterization of pharmaceutical cocrystals. In addition to this, challenges before and after synthesizing cocrystal along with currently approved formulations based on the pharmaceutical cocrystal approach and cocrystals under clinical investigation are highlighted.
Enhancing Stability and Formulation Capability of Fungicides by Cocrystallization Through a Novel Multistep Slurry Conversion Process

Abstract:Solid agrochemicals with poor thermal stability pose a huge challenge to their production and application. Herein, two low-melting-point fungicides, pyraclostrobin (ZJ) and flusilazole (FZ), were designed to form cocrystals with the high melting point fungicide of thiophanate-methyl (TM), respectively. An infrequent multistep slurry conversion crystallization process was discovered and studied, which accelerated the formation of cocrystals, and the mechanism was revealed. Oiling-out of FZ and ZJ was the key process to multistep slurry conversion crystallization. The heterogeneous system possessed the advantage of high energy, which accelerated the synthesis of cocrystals. Furthermore, the variations of physicochemical properties containing thermal stability, tap density, formulation ability, and storage stability of both the parent fungicides and cocrystals were analyzed and compared. Packing coefficient and crystal structure analysis revealed the structure-property relationships of cocrystals. The single-crystal structure showed the strong N-H center dot center dot center dot N hydrogen bonds between the FZ and TM, and N-H center dot center dot center dot O and N-H center dot center dot center dot S interactions in the ZJ-TM cocrystal. Strong hydrogen bonds stabilize the structures of cocrystals, which enables ZJ-TM and FZ-TM to be made into a greener water suspension concentrate, instead of an emulsifiable concentrate with pollution.

Abstract:This study investigates the cocrystallization of secnidazole (SNZ), a potent antimicrobial drug, with several coformers, overcoming the inherent stability challenges posed by its hemihydrate form to produce novel solid states. We successfully synthesized and characterized three novel cocrystals with carvacrol (CAR), fumaric acid (FMA), and 3,5-dinitrobenzoic acid (DNZ), employing synchrotron single crystal analysis to elucidate their structures. The application of Hirshfeld surface and 2D fingerprint plots further enriched our understanding of the unique packing arrangements within these cocrystals. Moreover, preliminary powder X-ray diffraction (PXRD) patterns hinted at the emergence of additional novel solid forms with salicylic acid (SLC), oxalic acid (OXA), succinic acid (SUC), citric acid (CTR), and thymol (THY). Supported by literature and a thorough analysis of molecular interactions, we propose the existence of pharmaceutical cocrystals among these newly identified solid forms. Our findings not only augment the repertoire of SNZ based pharmaceuticals but also accentuates the strategic utilization of supramolecular synthons, offering significant advancements in the understanding of novel solid form formation, and the critical role of hydrogen bonding in these processes.

Abstract:Here we described a co-crystal strategy for the solidification of liquid 2,3-dimethylpyrazine (2,3-DMP), 2,5-dimethylpyrazine (2,5-DMP), 2,6-dimethylpyrazine (2,6-DMP) with the poly-substituted salicylic acids [3,5,6-trichlorosalicylic acid (TCSA) and 3,5-dinitrosalicylic acid (DNSA)] under identical conditions. Those newly grown co-crystals (2,3-DMP:TCSA = 1:1 for 1; 2,5-DMP:TCSA = 1:2 for 2; 2,6-DMP:DNSA = 1:1 for 3) were characterized by single-crystal X-ray diffraction. The structure determination shows that strong O-H⋯N hydrogen bonds play an important role in connection of pyrazine derivatives and poly-substituted salicylic acids. Moreover, aromatic cycle stacking interactions play a crucial role in the 3D supramolecular network. Hirshfeld surfaces and fingerprint plots of the reported co-crystals were analyzed, which showed that O-H⋯N, C-H⋯O, H⋯H, H⋯Cl and aromatic cycle stacking intermolecular interactions are the main stabilizing factors of the co-crystal structures. The melting points studies showed that the co-crystal strategy is effective for solidification of liquid pyrazine derivatives.

Abstract:In this study, the existing set of carbamazepine (CBZ) cocrystals was extended through the successful combination of the drug with the positional isomers of acetamidobenzoic acid. The structural and energetic features of the CBZ cocrystals with 3- and 4-acetamidobenzoic acids were elucidated via single-crystal X-ray diffraction followed by QTAIMC analysis. The ability of three fundamentally different virtual screening methods to predict the correct cocrystallization outcome for CBZ was assessed based on the new experimental results obtained in this study and data available in the literature. It was found that the hydrogen bond propensity model performed the worst in distinguishing positive and negative results of CBZ cocrystallization experiments with 87 coformers, attaining an accuracy value lower than random guessing. The method that utilizes molecular electrostatic potential maps and the machine learning approach named CCGNet exhibited comparable results in terms of prediction metrics, albeit the latter resulted in superior specificity and overall accuracy while requiring no time-consuming DFT computations. In addition, formation thermodynamic parameters for the newly obtained CBZ cocrystals with 3- and 4-acetamidobenzoic acids were evaluated using temperature dependences of the cocrystallization Gibbs energy. The cocrystallization reactions between CBZ and the selected coformers were found to be enthalpy-driven, with entropy terms being statistically different from zero. The observed difference in dissolution behavior of the cocrystals in aqueous media was thought to be caused by variations in their thermodynamic stability.

Abstract:Spherical crystallization is widely employed in pharmaceutical industry to produce spherical particles that exhibit improved micrometric performances of active pharmaceutical ingredients (APIs), including particle size, morphology, flowability, and compressibility. Especially for APIs with poor powder performance and low solubility, spherical co-crystallization offers the potential to enhance both dissolution properties and tablet manufacturability by combining the API and cocrystal coformer (CCF) into a spherical particle. In this study, we aimed to enhance the physicochemical properties of carbamazepine (CBZ) and hesperetin (HPE) by synthesizing a novel cocrystal with the stoichiometric ratio of 1:1. Crystal structure of the CBZ-HPE cocrystal was determined using single crystal X-ray diffraction (SCXRD). To further improve the poor powder performances such as low flowability and compressibility resulting from the needle morphology of the cocrystals, we designed three spherical crystallization strategies: common spherical agglomeration, oil-out-induced spherical agglomeration and spherulitic growth method. The spherical particles of CBZ-HPE cocrystals were characterized by powder X-ray diffraction (PXRD), polarized light microscopy (PLM), differential scanning calorimetry (DSC), scanning electron microscopy (SEM), thermogravimetric analysis (TGA), particle size instrumentation, and Fourier transform infrared spectroscopy (FTIR) to provide information about crystal structure, particle morphology, particle size and circularity distribution. The results revealed significant differences in particle morphology, flowability, particle size and dissolution rate among the various spherical crystallization strategies. Theoretical calculations elucidated three spherical crystallization mechanisms: spherical agglomeration (SA), oil-out-induced spherical agglomeration (OOSA) and spherulite growth (SG). This study offers both experimental and theoretical foundations for the application of different spherical crystallization techniques in the pharmaceutical industry.

Abstract:Single-crystal X-ray diffraction analysis is a vital and precise method to identify the structures and absolute configurations of molecules at the atomic level. However, this method is not suitable for liquid molecules. Here, we report a new and easy method to tackle this challenge via merging supramolecular chemistry and crystal engineering. A glycoluril-derived molecular clip as a molecule catcher is designed and synthesized which forms cocrystals with four different liquid molecules within 24 hours, leading to the confirmation of the structures of these liquid molecules. This work is expected to open new opportunities for the structural identification of liquid molecules.

Abstract:Cocrystal engineering for bioavailability enhancement is still a serendipitous process, which requires systematic solution crystallization, in vivo predictive dissolution, and biopharmaceutical simulation. The purpose of this study is to improve bioavailability through cocrystal engineering, with a focus on understanding the mechanism of supersaturation maintenance during dissolution. BI 639667 is a poorly soluble and fast precipitating compound. The ability of its cocrystal with salicylic acid (SA) to maintain supersaturation was assessed using a modified two-step dissolution method, and its bioavailability was determined in rats. Biopharmaceutical simulation and dissolution modeling were performed to predict the in vivo performance in humans. Conductor-like screening model for realistic solvents and molecular dynamics (MD) simulation were used to study the interactions between the active pharmaceutical ingredient (API) and coformers in vacuum and aqueous media, respectively. A high melting cocrystal with SA, crystallized in dichloromethane, showed moderate solubility enhancement with prolonged supersaturation during dissolution, which was able to enhance bioavailability with reduced C max, compared to amorphous dispersion. The radial distribution function (RDF) between the API and selected coformers was calculated using MD simulation to determine the mean distance between the API and coformer molecules in aqueous media. The result suggests that SA could better compete against water for interactions with the API and could penetrate the API molecular clusters to inhibit nucleation. Thus, the RDF by MD simulation may be used to determine the disruptive effect of water on the interaction between the API and coformer and improve cocrystal engineering for bioavailability enhancement.

Abstract:Miconazole shows low oral bioavailability in humans due to poor aqueous solubility, although it has demonstrated various pharmacological activities such as antifungal, anti-tubercular and anti-tumor effects. Cocrystal/salt formation is one of the effective methods for solving this problem. In this study, different methods (liquid-assisted grinding, slurrying and lyophilization) were used to investigate their impact on the formation of the miconazole multicomponent crystals with succinic, maleic and dl-tartaric acids. The solid state of the prepared powder was characterized by differential scanning calorimetry, powder X-ray diffraction and scanning electron microscopy. It was found that lyophilization not only promotes partial amorphization of both salts but also allows obtaining a new polymorph of the miconazole salt with dl-tartaric acid. The lyophilized salts compared with the same samples prepared by two other methods showed better dissolution rates but low stability during the studies due to rapid recrystallization. Overall, it was determined that the preparation method of multicomponent crystals affects the solid-state characteristics and miconazole physicochemical properties significantly. The in vivo studies revealed that the miconazole multicomponent crystals indicated the higher peak blood concentration and area under the curve from 0 to 32 h values 2.4-, 2.9- and 4.6-fold higher than the pure drug. Therefore, this study demonstrated that multicomponent crystals are promising formulations for enhancing the oral bioavailability of poorly soluble compounds.

A novel solubilization technique for poorly soluble drugs through the integration of nanocrystal and cocrystal technologies

Abstract:The aim of the present study was to develop a novel solubilization technique consisting of a nano-cocrystal suspension by integrating cocrystal and nanocrystal formulation technologies to maximize solubilization over current solubilizing technologies. Monodisperse carbamazepine-saccharin, indomethacin-saccharin, and furosemide-caffeine nano-cocrystal suspensions, as well as a furosemide-cytosine nano-salt suspension, were successfully prepared with particle sizes of less than 300nm by wet milling with the stabilizers hydroxypropyl methylcellulose and sodium dodecyl sulfate. Interestingly, the properties of resultant nano-cocrystal suspensions were dramatically changed depending on the physicochemical and structural properties of the cocrystals. In the formulation optimization, the concentration and ratio of the stabilizers also influenced the zeta potentials and particles sizes of the resultant nano-cocrystal suspensions. Raman spectroscopic analysis revealed that the crystalline structures of the cocrystals were maintained in the nanosuspensions, and were physically stable for at least one month. Furthermore, their dissolution profiles were significantly improved over current solubilization-enabling technologies, nanocrystals, and cocrystals. In the present study, we demonstrated that nano-cocrystal formulations can be a new promising option for solubilization techniques to improve the absorption of poorly soluble drugs, and can expand the development potential of poorly soluble candidates in the pharmaceutical industry.

Abstract:Highlights • 1:1 SMZ-ASA cocrystal produced using evaporation, liquid assisted grinding, and slurry crystallization methods. • Ternary phase diagrams are established at 25 °C in acetonitrile and water. • Slurry conversion rate determined using in-situ Raman measurements and compared with off-line PXRD analysis. Successful production of 1:1 sulfamethazine-acetylsalicylic acid (SMZ-ASA) cocrystal was achieved through slow solvent evaporation, liquid-assisted grinding, and slurry conversion method. SCXRD, PXRD, DSC, FTIR, and Raman spectroscopy were employed to characterize the cocrystal. Ternary phase diagrams (TPD) for SMZ and ASA in acetonitrile (ACN) and deionized water (DIW) has been constructed at 25 °C. Using TPD of the incongruent system, slurry compositions for stable production of cocrystal was determined in both the solvents. The cocrystal conversion process in slurry was monitored using in-situ Raman spectroscopy. Intermittent sampling was also carried out to determine the purity of the solid phase from the slurry using offline PXRD. In-situ Raman and offline PXRD measurements confirmed fast conversion of the pure coformer crystals to the cocrystal in ACN, within a span of 5 minutes. However, the conversion in DIW was much slower and the in-situ Raman measurements significantly underpredicted the transformation time in comparison to offline PXRD analysis. The study highlights the utilization of TPD for developing the cocrystallization process and the need for multiple characterization techniques for monitoring cocrystallization. Abstract:Cymoxanil (CX), an essential fungicide, effectively controls fungal diseases but its environmental instability often leads to decreased efficacy and frequent applications, increasing costs and environmental risks. We propose to address this issue by cocrystal engineering, particularly, utilizing organic small acids as coformers to enhance the stability of CX in alkaline and neutral conditions. This study reports the synthesis of two CX cocrystals with succinic acid (SA) and salicylic acid (SCA). The prepared cocrystals were evaluated using X-ray diffraction, FT-IR spectroscopy, differential scanning calorimetry, thermogravimetric analysis, chemical stability experiment, and solubility and dissolution measurements. Hirshfeld surface analysis was applied to quantify the molecular interactions in CX–SA and CX–SCA cocrystals. Notably, the cocrystals preserve high CX residuals in PBS buffer at pH 5.8 and 8.0 and in pure water after 20 days, demonstrating their ability to enhance chemical stability of CX. The findings related to solubility and dissolution rates further underscore the utility of the cocrystals. This work may facilitate the design of a practical system through the cocrystallization of APIs that exhibit instability or other challenges.

Abstract:Drug-drug cocrystals are formulated to produce combined medication, not just to modulate active pharmaceutical ingredient (API) properties. Nano-crystals adjust the pharmacokinetic properties and enhance the dissolution of APIs. Nano-cocrystals seem to enhance API properties by combining the benefits of both technologies. Despite the promising opportunities of nano-sized cocrystals, the research at the interface of nano-technology and cocrystals has, however, been described to be in its infancy. In this study, high-pressure homogenization (HPH) and high-power ultrasound were used to prepare nano-sized cocrystals of 4-aminosalysilic acid and sulfamethazine in order to establish differences between the two methods in terms of cocrystal size, morphology, polymorphic form, and dissolution rate enhancement. It was found that both methods resulted in the formation of form I cocrystals with a high degree of crystallinity. HPH yielded nano-sized cocrystals, while those prepared by high-power ultrasound were in the micro-size range. Furthermore, HPH produced smaller-size cocrystals with a narrow size distribution when a higher pressure was used. Cocrystals appeared to be needle-like when prepared by HPH compared to those prepared by high-power ultrasound, which had a different morphology. The highest dissolution enhancement was observed in cocrystals prepared by HPH; however, both micro- and nano-sized cocrystals enhanced the dissolution of sulfamethazine.
Polymorph selection of pharmaceutical cocrystals via bench-top and continuous production techniques

Abstract:Polymorphism can be a valuable tool as well as an impediment in the development and approval of pharmaceuticals, providing an opportunity to tune active pharmaceutical ingredient (API) physicochemical properties. The control of polymorphism in cocrystalline systems and other multicomponent forms remains underexplored. The study herein aims to investigate the potential of several techniques, liquid-assisted grinding (LAG), solvent evaporation (SE), supercritical enhanced atomization (SEA) and electrospraying, to control the cocrystal polymorphic outcome of three cocrystals: isonicotinamide-citric acid (IsoCa), ethenzamide-saccharin (EthSac) and ethenzamide-gentisic acid (EthGa). Solvent selection employing LAG and SE showed little effect on polymorphic outcome. Electrospraying and SEA primarily produced the α form of IsoCa, with process parameter variations leading to the β form during SEA, and a mixture of α and γ from electrospraying. Electrospraying led to the stable form I of EthSac, while SEA could produce pure form II, and a mixture. Electrospraying produced the form I of EthGa while SEA could produce form II, with an unknown polymorphic impurity. Density functional theory (DFT) computed electron density (ED) maps of cocrystal polymorph binary systems further rationalised the polymorphic predominance observed through the electrospraying. Ultimately this study provides a general road map for polymorph selection via atomization-based methodologies.

Abstract:Pharmaceutical cocrystals consist of an active pharmaceutical ingredient and a coformer in a fixed stoichiometric ratio. They can improve the manufacturability and properties of a drug powder. However, the manufacture of cocrystals is often more complicated compared to crystals of an active ingredient due to the possible formation of different solid-state forms. Cocrystallization through anti-solvent addition is a commonly used technique. However, the risk of nucleation of multiple solid-state forms is often high due to the high supersaturation levels achieved during anti-solvent crystallization. Therefore, intensifying a solution-mediated phase transformation is important. In this work, the ability of tubular flow crystallizers to intensify the phase transformation process in the carbamazepine-saccharin cocrystal system is characterized and compared to a conventional stirred tank crystallizer. The influence of the flow rate, tubular crystallizer type, and ratio of residence time in the tube to that in the tank are documented. The phase transformation process is substantially faster with the tubular crystallizers compared to that in a stirred tank crystallizer, which we explain through increased nucleation rates. We show that such intensification can be achieved either by recirculating the slurry through a static mixer or by installing a tubular crystallizer upstream of a stirred tank crystallizer.

Abstract:A novel chemical structure named pharmaceutical cocrystal was synthesized with the purpose to enhance the solubility of levofloxacin (LVFX). Nicotinic acid (NA) was selected as cocrystal coformer (CCF). The cocrystal was successfully obtained by solvent evaporation method combined with ultrasonic technology. The crystal structure of LVFX-NA is triclinic with a P1 space group, unit cell parameters a = 7.1392 Å, b = 10.0188 Å, c = 17.5394 Å, α=86.109 (7), β=86.082 (6), γ=73.809 (7), z = 2, R1=0.0518, ωR2=0.1425. The supramolecular structure of LVFX-NA is mainly controlled by OH···N and CH···O intermolecular hydrogen bond. Moreover, intramolecular hydrogen bonds CH···F have also been found in the cocrystal structural units. The obtained cocrystal exhibits unique properties of thermoanalysis, Fourier transform infrared (FT-IR) spectrum and X-ray powder diffraction (PXRD) spectrum, demonstrated the new formed crystal material. Equilibrium solubility measurement revealed that LVFX-NA exhibited improved aqueous solubility, that is 5.02 times higher than the original LVFX.

Effect of functional group position in co-formers and solvent on cocrystal polymorphism/stoichiomorphism: a case study

Abstract:In recent years, there has been an increase in the number of reports on cocrystal polymorphism and stoichiomorphism. However, the research on the factors that influence these phenomena is limited. Herein, picolinamide (PAM), nicotinamide (NAM), and isonicotinamide (INA) were selected as co-formers to form multicomponent solids with 4-chloro-3-sulfamoylbenzoic acid (CSBA). Six new cocrystal forms of CSBA were discovered and their crystal structures were determined. It was found that PAM and NAM can only form one cocrystal with CSBA, while INA can form up to four cocrystals, including both cocrystal polymorphism and stoichiomorphism. Molecular electrostatic potential analysis and crystal structure analysis showed that the functional group position of PAM limited the diversity of cocrystal synthons, while the lattice energy limited the diversity of cocrystal synthons when NAM acted as a co-former. Only INA was not subject to these restrictions when forming cocrystals. Finally, the influence of solvents on cocrystals was illustrated by determining the ternary phase diagrams. The mechanism of two similar solvents, ethyl acetate and acetone, controlling the crystallization of cocrystal polymorphism was analyzed by molecular simulations.

Abstract:Crystal habit and crystal form are critical elements in determining product properties and functions. In this work, we developed a microfluidic antisolvent crystallization technique to rapidly screen and accurately control the solid form and crystal habit of triphenylmethanol (Ph 3 COH). This advanced technique separates the primary mixing of solutions from crystal formation (nucleation and growth) by introducing the microfluidic device, avoiding clogging in microchannels to obtain high-quality crystals. The results show that we can achieve controllable preparation of pure 2Ph 3 COH·DMSO (DMSO solvate), pure Ph 3 COH (form β ), and mixed crystals with different mass ratios. Moreover, the microscale can prompt the DMSO solvate to grow into hexagonal sheet-like and bulk crystals. We can regulate the aspect ratio of hexagonal sheet-like crystals in binary solvents and control the crystal habit of the form β to transition between long needle-like shapes and short hexagonal prisms in DMF-H 2 O. Meanwhile, we revealed that the solvent ratio, the antisolvent flow rate, and the initial concentration of Ph 3 COH are the main factors affecting the solid form selectivity and morphology transition. Such a novel method would be considered as a promising technique to be extended to screen and control key crystallization parameters of other substances.

Otherwise, a stable form of the parent drug will be recrystallized on the dissolving cocrystal surface and/or in the bulk solution during the cocrystal dissolution process, negating the solubility advantage.
