Abstract:Abstract Ribosome biogenesis is an energetically expensive program that is dictated by nutrient availability. Here we report that nutrient deprivation severely impairs precursor ribosomal RNA (pre-rRNA) processing and leads to the accumulation of unprocessed rRNAs. Upon nutrient restoration, pre-rRNAs stored under starvation are processed into mature rRNAs that are utilized for ribosome biogenesis. Failure to accumulate pre-rRNAs under nutrient stress leads to perturbed ribosome assembly upon nutrient restoration and subsequent apoptosis via uL5/uL18-mediated activation of p53. Restoration of glutamine alone activates p53 by triggering uL5/uL18 translation. Induction of uL5/uL18 protein synthesis by glutamine is dependent on the translation factor eukaryotic elongation factor 2 (eEF2), which is in turn dependent on Raf/MEK/ERK signaling. Depriving cells of glutamine prevents the activation of p53 by rRNA synthesis inhibitors. Our data reveals a mechanism that tumor cells can exploit to suppress p53-mediated apoptosis during fluctuations in environmental nutrient availability.

What problem does this paper attempt to address?
The paper aims to explore how nutrient deprivation (specifically glutamine deficiency) affects the resistance mechanisms of solid tumor cells to ribosomal RNA (rRNA) synthesis inhibitors. ### Main Issues: 1. **Impact of Nutrient Deprivation on rRNA Synthesis**: The study found that nutrient deprivation severely impairs the processing of precursor rRNA (pre-rRNA) and leads to the accumulation of unprocessed rRNA. When nutrient supply is restored, these accumulated pre-rRNAs are further processed into mature rRNA for ribosome biogenesis. 2. **Role of Glutamine**: Glutamine plays a crucial role in this process. The addition of glutamine alone can activate p53, triggering the translation of uL5/uL18 proteins, thereby inhibiting MDM2 and leading to p53 stabilization. Glutamine inhibits eEF2K by activating the Ras/Raf/MEK/ERK signaling pathway, allowing overall protein translation. 3. **p53-Mediated Apoptosis**: If sufficient pre-rRNA fails to accumulate under nutrient stress, it will lead to ribosome assembly blockage, which in turn triggers cell apoptosis through uL5/uL18-mediated p53 activation. 4. **Glutamine Deficiency and Drug Resistance**: Glutamine deficiency can prevent rRNA synthesis inhibitors from activating p53, indicating that solid tumors with glutamine deficiency may have intrinsic resistance to small molecule drugs targeting Pol I. ### Summary: The study reveals how solid tumor cells utilize pre-rRNA accumulation to adapt to fluctuating nutrient environments and explains the critical role of glutamine in this process. This provides a new perspective on understanding the adaptation mechanisms of tumor cells under nutrient stress and helps in developing therapeutic strategies targeting specific metabolic pathways.
