Abstract:Many studies support the cardioprotective effects of folic acid (FA). We aimed to evaluate the utility of FA supplementation in preventing the development of atherosclerotic in low‐density lipoprotein receptor‐deficient (LDLR−/−) mice and to elucidate the molecular processes underlying this effect. LDLR−/− mice were randomly distributed into four groups: control group, HF group, HF + FA group and the HF + RAPA group. vascular smooth muscle cells (VSMCs) were divided into the following four groups: control group, PDGF group, PDGF + FA group and PDGF + FA + RAPA group. Blood lipid levels, oxidative stress and inflammatory cytokines were measured. Atherosclerosis severity was evaluated with oil red O staining. Haematoxylin and eosin (H&E) staining was used to assess atherosclerosis progression. Immunohistochemical staining was performed with antismooth muscle α‐actin (α‐SMA) antibodies and anti‐osteopontin (OPN) antibodies that demonstrate VSMC dedifferentiation. The protein expression of α‐SMA, OPN and mechanistic target of rapamycin (mTOR)/p70S6K signalling was detected by Western blot analysis. FA and rapamycin reduced serum levels of total cholesterol, triacylglycerol, LDL, inhibiting oxidative stress and the inflammatory response. Oil red O and H&E staining demonstrated that FA and rapamycin inhibited atherosclerosis. FA and rapamycin treatment inhibited VSMC dedifferentiation in vitro and in vivo, and FA and rapamycin attenuated the mTOR/p70S6K signalling pathway. Our findings suggest that FA attenuates atherosclerosis development and inhibits VSMC dedifferentiation in high‐fat‐fed LDLR−/− mice by reduced lipid levels and inhibiting oxidative stress and the inflammatory response through mTOR/p70S6K signalling pathway.

Abstract:Abstract Background/Introduction Dilated cardiomyopathy (DCM) is a leading cause of heart failure (HF) in young and middle aged individuals worldwide. Alterations in mitochondrial dynamics (fusion/fission) and quality control (mitophagy) are critical for normal cardiac function. Imbalanced mitochondrial dynamics towards fission has been shown to contribute to DCM development in mice with ablation of the mitochondrial protease YME1L, which results in altered OPA1 processing precluding mitochondrial fusion. In these mice, the DCM phenotype includes a metabolic reprogramming of cardiomyocytes, shifting from fatty acid (FA) oxidation towards glucose. High fat diet (HFD) in these mice results in reversal of this metabolic reprogramming and rescue from DCM despite the genetic defect (lack of YME1) still present. These results opened a new venue for dietary approaches as a therapeutic strategy in HF. To understand whether the fat itself contained in the HFD was responsible of HF rescue, we studied the trajectories of cardiac function in mice with altered mitochondrial dynamics under HFD, caloric restriction and control chow. Methods Cardiomyocyte-specific Yme1l knock-out (cYKO) male mice (N=10–15) known to develop DCM phenotype at 20–25 weeks of age were randomized to 3 different diets (1: HFD containing >30% crude fat; 2: non-fat diet containing <0.5% crude fat, and 3: control diet containing 10% crude fat) at 10 weeks of age. Every 10 weeks, cardiac function was tested by echocardiography. At 30 weeks, animals underwent a multitracer microPET/CT (glucose and FA uptake) to evaluate myocardial substrate utilization. At the end of the study, animals were sacrificed and hearts were harvested for histological and further tissue analyses. Results As expected, HFD was associated with a better left ventricular ejection fraction (LVEF; %) at 30 weeks: 37.97±8.59 vs. 29.16±8.9 in control diet, p=0.04. This functional improvement was associated with an increase in FA uptake on microPET/CT. Non-fat diet was associated with even higher LVEF at 30 weeks: 44.94±11 vs. 29.16±8.9 in control diet, p=0.0001, as well as smaller LV diameter (mm): 3.58±0.29 vs. 4.3±0.39 in control diet, p<0.0001 (A). Lifespan of mice on non-fat diet was significantly prolonged (70% extension compared to control) (B). Enhanced mitochondrial clearance (mitophagy) mediated by Parkin, together with an increase in autophagy related protein as LC3B was identified in the non-fat diet group (C). Conclusion Different dietary approaches have been shown to be beneficial in the prevention of DCM in mice with altered mitochondrial dynamics. Caloric restriction (non-fat diet) is able to prevent DCM, reverse metabolic reprogramming, increase mitochondrial quality control and ultimately prolong lifespan in mice with genetic defect associated with altered mitochondrial dynamics. Interventions aiming at enhancing cardiac mitophagy might represent a new therapeutic option in DCM.

Short Communication: Taurine Long-Term Treatment Prevents the Development of Cardiac Hypertrophy, and Premature Death in Hereditary Cardiomyopathy of the Hamster Is Sex-Independent

Abstract:Recently, we reported that during the hypertrophic phase (230 days old) of hereditary cardiomyopathy of the hamster (HCMH), short-term treatment (20 days) with 250 mg/kg/day of taurine prevents the development of hypertrophy in males but not in females. However, the mortality rate in non-treated animals was higher in females than in males. To verify whether the sex-dependency effect of taurine is due to the difference in the disease's progression, we treated the 230-day-old animals for a longer time period of 122 days. Our results showed that long-term treatment with low and high concentrations of taurine significantly prevents cardiac hypertrophy and early death in HCMH males (p < 0.0001 and p < 0.05, respectively) and females (p < 0.01 and p < 0.0001, respectively). Our results demonstrate that the reported sex dependency of short-term treatments with taurine is due to a higher degree of heart remodeling in females when compared to males and not to sex dependency. In addition, sex-dependency studies should consider the differences between the male and female progression of the disease. Thus, long-term taurine therapies are recommended to prevent remodeling and early death in hereditary cardiomyopathy.

Abstract:Aromatase activity during gonadotropin action in vivo and in vitro was examined in purified Leydig cells to further define the early effects of LH and for elucidation of the enzymatic processes involved in the development of late lesions of androgen biosynthetic pathway. Aromatase was measured by the tritiated water release method using [1 beta-3H]testosterone as substrate. The enzyme activity was proportional to the amount of cells (0.1-1.0 X 10(6] incubated, increased with the incubation time (2-60 min), was inhibited by androstatriendione (ED50, 5.0 microM), and showed a Km for testosterone of 1.69 microM. Aromatase activity was stimulated (10-20%; P less than 0.05) 1 h after treatment of rats with a single sc dose of 5 micrograms hCG. This activation preceded the late steroidogenic lesion at the site of 17 alpha-hydroxylase and 17,20-desmolase activity by 2-5 h. A RIA of improved sensitivity (0.5 pg) was developed to detect the very low cellular and secretory levels of estradiol. The testicular contents of testosterone and estradiol showed small increases (P less than 0.05) within 40 and 60 min after hCG treatment, respectively. Testicular testosterone levels reached a peak by 1 h after injection and preceded the peak level of estradiol formation by 2 h. After in vitro treatment of cultured Leydig cells with 100 ng hCG, the aromatase activity was significantly increased within 30 min (P less than 0.05) and then returned to control levels for up to 16 h of culture. A similar temporal pattern for enzyme activation was observed after treatment of cultures with 8-bromo-cAMP or forskolin (23-27% above control; P less than 0.05), while cholera toxin stimulated aromatase activity at 2 h. Net testosterone accumulation in the incubation medium increased 30 min after the hCG treatment and reached a plateau by 4 h. A small but significant increase in estradiol levels (P less than 0.05) was also observed at 30 min, remaining constant until 120 min, which was followed by a sharp rise parallel to that of testosterone. These results suggest that the estradiol-mediated desensitization of the Leydig cell observed after hCG administration is consistent with an early cAMP-dependent activation of aromatase and a further rise in estradiol formation due to increased substrate availability.
Effect of 6-Month Feeding with a Diet Enriched in EPA + DHA from Fish Meat on the Blood Metabolomic Profile of Dogs with Myxomatous Mitral Valve Disease

Abstract:Animal nutrition plays an important role in the therapy of many diseases, including heart failure. The aim was to assess whether 6 months of feeding an AEP + ADH enriched diet (from fish meat) in dogs suffering from heart failure due to mitral degeneration impacts the dogs’ metabolic profile and clinical status. Twenty small breed dogs were included: 50% were in stage B2 of MMVD and 50%, in stage C according to ACVIM. Dogs were randomly divided into two groups. One group receiving a standard diet, the second one a diet enriched with EPA + DHA (from fish meat). All dogs continued to receive appropriate therapy throughout the study. Control examinations were performed at the start of the study, after 3 and 6 months of appropriate feeding. Examinations included ECG, ECHO, blood hemathology and biochemistry, morphometric measurements, body fat index and subcutaneous fat tissue thickness. Serum samples were analyzed with a high-performance liquid chromatography system. Data were analyzed using the Progenesis QI (PQI, Non-linear Dynamics). The results showed no differences in clinical, cardiological, haematological and biochemical parameters between the two study groups. An effect on the metabolomic profile following a continued diet enriched in DHA + EPA (from fish meat) was more pronounced with time. After 6 months of feeding the diete enriched with DHA + EPA (from fish meat), there was a favorable reduction in glycerophosphocholine and xanthine levels, but an adverse increase in lactate and furvan and a decrease in alanine was not stopped.

Abstract:Abstract Chronic consumption of a high-fat diet (HFD) has profound effects on brain aging, which is mainly characterized by cognitive decline, inflammatory responses, and neurovascular damage. Alisol A (AA) is a triterpenoid with therapeutic potential for metabolic diseases, but whether it has a neuroprotective effect against brain aging caused by a HFD has not been investigated. Six-month-old male C57BL6/J mice were exposed to a HFD with or without AA treatment for 12 weeks. Behavioral tasks were used to assess the cognitive abilities of the mice. Neuroinflammation and changes in neurovascular structure in the brains were examined. We further assessed the mechanism by which AA exerts neuroprotective effects against HFD-induced pathological brain aging in vitro and in vivo. Behavioral tests showed that cognitive function was improved in AA-treated animals. AA treatment reduced microglia activation and inflammatory cytokine release induced by a HFD. Furthermore, AA treatment increased the number of hippocampal neurons, the density of dendritic spines, and the expression of tight junction proteins. We also demonstrated that AA attenuated microglial activation by targeting the SIRT3-NF-κB/MAPK pathway and ameliorated microglial activation-induced tight junction degeneration in endothelial cells and apoptosis in hippocampal neurons. The results of this study show that AA may be a promising agent for the treatment of HFD-induced brain aging.

Abstract:Rationale and Methods: Skeletal muscle derangements, potentially including mitochondrial dysfunction with altered mitochondrial dynamics and high reactive oxygen species (ROS) generation, may lead to protein catabolism and muscle wasting, resulting in low exercise capacity and reduced survival in chronic heart failure (CHF). We hypothesized that 8-week n-3-PUFA isocaloric partial dietary replacement (Fat = 5.5% total cal; EPA + DHA = 27% total fat) normalizes gastrocnemius muscle (GM) mitochondrial dynamics regulators, mitochondrial and tissue pro-oxidative changes, and catabolic derangements, resulting in preserved GM mass in rodent CHF [Myocardial infarction (MI)-induced CHF by coronary artery ligation, left-ventricular ejection fraction <50%]. Results: Compared to control animals (Sham), CHF had a higher GM mitochondrial fission-fusion protein ratio, with low ATP and high ROS production, pro-inflammatory changes, and low insulin signalling. n-3-PUFA normalized all mitochondrial derangements and the pro-oxidative state (oxidized to total glutathione ratio), associated with normalized GM cytokine profile, and enhanced muscle-anabolic insulin signalling and prevention of CHF-induced GM weight loss (all p < 0.05 vs. CHF and p = NS vs. S). Conclusions: n-3-PUFA isocaloric partial dietary replacement for 8 weeks normalizes CHF-induced derangements of muscle mitochondrial dynamics regulators, ROS production and function. n-3-PUFA mitochondrial effects result in preserved skeletal muscle mass, with potential to improve major patient outcomes in clinical settings.

Abstract:Introduction: Many studies have shown that omega-3 polyunsaturated fatty acids (ω-3 PUFAs) have beneficial effects on cardiovascular disease but little is known about the effects of ω-3 PUFAs on cardiac fibrosis, which is the major cause of cardiac remodeling leading to hear failure. Hypothesis: ω-3 PUFAs will inhibit cardiac fibrosis and improve heart function. Methods: Ten weeks old male C57/B6 mice were fed control diet (corn oil, ω-6 PUFAs; n=20) or Fish oil diet (FO) 1% energy from EPA+DHA (n=20). After 8 weeks on the assigned diet, Sham or Transverse Aortic Constriction (TAC) surgery will be randomly performed on each diet group. The groups allocated in a 2x2 factorial design into 4 groups as (1) Cont/Sham, (2) Cont/TAC, (3) FO/Sham, (4) FO/TAC. After 4 weeks of continued dietary, myocardial fibrosis was determined by picro-sirius red staining for collagen in the middle of left ventricle. The ω-3 index (the sum of red blood cell EPA + DHA) were determined by gas chromatography. Echocardiography (Echo) were also performed on mice one day before and 2- and 4-weeks post-surgery. In vitro experiments were performed in adult cardiac fibroblasts (CFs) isolated from the heart of male C57BL/6 mice. CFs collagen synthesis, differentiation to myofibroblasts and proliferation were determined by 3H-proline incorporation, α-smooth muscle actin (α-SMA) staining and MTS assay, respectively. Results: Compare to Cont/sham group, in Cont/TAC group, Fractional Shortening (FS) were decreased significantly (p<0.01) as well as myocardial fibrosis was significantly increased in (p<0.01) at 4 weeks. However, FO treatment rescued the effects of TAC stress as FS remarkably reverse (FO/ TAC 45%±10% vs Cont/TAC 28%±10%, p<0.01) and myocardial fibrosis significantly decreased (FO/ TAC 8.9%±2.8% vs Cont/TAC 14.1%±4.1%, p<0.01). In addition, the ω-3 index was notably increased in FO diet group (FO 15.17%±0.4% vs Cont 3.9%± 0.6%, p<0.01). In vitro, EPA and DHA (10 μM) significantly blocked the pro-fibrotic effects of TGF- β1 (1ng/mL) on collagen synthesis while arachidonic acid (AA) and oleic acid (OA) did not. Both EPA and DHA suppressed TGF- β1-induced differentiation of CFs to myofibroblasts. Furthermore, EPA and DHA (1 to 50 μM) inhibited CFs proliferation induce by both Angiotensin II (100 nM) and TGF- β1 (1ng/mL) more than AA (p<0.01) and OA (p<0.05) did. Conclusion: Both in vivo and in vitro. ω-3 PUFAs have the potential to inhibit the development of cardiac fibrosis.
Docosahexaenoic acid protects against palmitate-induced mitochondrial dysfunction in diabetic cardiomyopathy

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Objective</h3><p>Regular consumption of n-3 polyunsaturated fatty acids is associated with decreased cardiovascular morbidity and mortality. This study assessed the therapeutic effect of docosahexaenoic acid (DHA) in palmitic acid (PA)-induced cytotoxicity <em>in vitro</em> and in rats fed a high-fat diet (HFD).</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Methods</h3><p>H9C2 cells and rat primary cardiomyoblasts were exposed to PA or PA + DHA for 24 h. PA-induced lipotoxicity and mitochondrial dysfunction were evaluated by immunostaining, real-time PCR, cardiomyocyte contraction and transmission electron microscopy. The effects of dietary DHA on diabetic cardiomyopathy were evaluated in male Sprague-Dawley rats fed a reference diet rich in DHA, an HFD, or an HFD with added DHA for 16 weeks. Oxidative stress and lipotoxicity in rat heart tissue were assayed by Masson staining, immunohistochemistry, and TUNEL.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p><em>In vitro</em> studies showed that dietary DHA reduced the occurrence of cardiomyopathy and improved cardiac responses to PA. In the rat model, dietary DHA reduced mitochondrial oxidative stress in HFD-induced diabetic cardiomyopathy.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusion</h3><p>Dietary DHA reduced mitochondrial oxidative stress and ameliorated PA-induced lipid toxicity. DHA consumption may have had direct effects on cardiovascular risk via myocardial protection.</p>

Effects of dietary alpha linolenic acid on cholesterol metabolism in male and female hamsters of the LPN strain

Abstract:N-3 polyunsaturated fatty acids and estrogens are recognized as protective factors of atherosclerosis, however their interactions on cholesterol metabolism remain unclear. Male and female hamsters were fed for 9 weeks diets containing 12.5% lipids and rich in either alpha-linolenic acid ("linseed" diet) or saturated fatty acids ("butter" diet). Hamsters fed the "linseed" diet exhibited lower plasma concentrations of cholesterol (-29%), total LDL (-35%) and HDL (-17%), glucose (-20%), insulin (-40%) and of the LDL-cholesterol/HDL-cholesterol ratio (-27%) than those fed the "butter" diet. In the liver, cholesterol content was 2.7-fold lower in response to the "linseed" diet, whereas the concentration of HDL receptor (SR-BI) and the activities of HMGCoA reductase and cholesterol 7alpha-hydroxylase were 30 to 50% higher than with the "butter" diet. By contrast, the LDL receptor concentration did not vary with the diet. Females exhibited higher concentration of LDL (+24%), lower concentration of plasma triglycerides (-34%), total VLDL (-46%) and VLDL-cholesterol (-37%) and of biliary phospholipids (-19%). Besides, there was also an interaction between gender and diet: in males fed the "butter" diet, plasma triglycerides and VLDL concentration, were 2 to 4 fold higher than in the other groups. These data suggest that gene and/or metabolic regulations by fatty acids could interact with that of sex hormones and explain why males are more sensitive to dietary fatty acids.

Exposure to the American lifestyle-induced obesity syndrome (ALIOS) diet recapitulates many of the classic findings of metabolic syndrome in mice. Abstract:The dietary intake of ω-3 polyunsaturated fatty acids has been linked to a reduction in the incidence of aging-associated disease including cardiovascular disease and stroke. Additionally, long-lived Caenorhabditis elegans glp-1 germ line-less mutant animals show a number of changes in lipid metabolism including the increased production of the ω-3 fatty acid, α-linolenic acid (ALA). Here, we show that the treatment of C. elegans with ALA produces a dose-dependent increase in lifespan. The increased longevity of the glp-1 mutant animals is known to be dependent on both the NHR-49/PPARα and SKN-1/Nrf2 transcription factors, although the mechanisms involved are incompletely understood. We find that ALA treatment increased the lifespan of wild-type worms and that these effects required both of these transcription factors. Specifically, NHR-49 was activated by ALA to promote the expression of genes involved in the β-oxidation of lipids, whereas SKN-1 is not directly activated by ALA, but instead, the exposure of ALA to air results in the oxidation of ALA to a group of compounds termed oxylipins. At least one of the oxylipins activates SKN-1 and enhances the increased longevity resulting from ALA treatment. The results show that ω-3 fatty acids inhibit aging and that these effects could reflect the combined effects of the ω-3 fatty acid and the oxylipin metabolites. The benefits of ω-3 fatty acid consumption on human health may similarly involve the production of oxylipins, and differences in oxylipin conversion could account for at least part of the variability found between observational vs. interventional clinical trials.

Abstract:This study aims to evaluate the hepatoprotective, hypolipidemic and aortic morphometric effects of fish oil rich in omega-3 in hypercholesterolemic BALB/c mice. This is an experimental model that included 16 male BALB/c mice (Mus musculus) divided into three groups (G1 (standard commercial chow and 0.9% saline solution), G2 (hypercholesterolemic diet and 0.9% saline solution) and G3 (hypercholesterolemic diet and fish oil)) for 8 weeks. There was no significant difference in the treatment with omega-3-rich fish oil in the lipid profile (p > 0.05). In the histological analysis, group G2 detected the presence of hepatitis and liver tissue necrosis, but this was not observed in group G3. As for the morphometry in the light area of the vessel, the G1 group had a higher score (2.62 ± 0.36 mm2) when compared to G2 (2.10 ± 0.16 mm2) and G3 (2.26 ± 0.25 mm2) (p 0.05). It is concluded that supplementation with fish oil rich in omega-3 carried out in this study may have a protective effect on liver tissue, but it has not yet improved the lipid and morphometric profile. Despite this research being preliminary, it is a relevant study with future prospects for improving the doses of EPA and DHA in order to better elucidate the benefits of fish oil in models of dyslipidemia.

High fat diet ameliorates mitochondrial cardiomyopathy in CHCHD10 mutant mice

Abstract:Mutations in CHCHD10, a mitochondrial protein with undefined functions, are associated with autosomal dominant mitochondrial diseases. Chchd10 knock-in mice harboring a heterozygous S55L mutation (equivalent to human pathogenic S59L) develop a fatal mitochondrial cardiomyopathy caused by CHCHD10 aggregation and proteotoxic mitochondrial integrated stress response (mtISR). In mutant hearts, mtISR is accompanied by a metabolic rewiring characterized by increased reliance on glycolysis rather than fatty acid oxidation. To counteract this metabolic rewiring, heterozygous S55L mice were subjected to chronic high-fat diet (HFD) to decrease insulin sensitivity and glucose uptake and enhance fatty acid utilization in the heart. HFD ameliorated the ventricular dysfunction of mutant hearts and significantly extended the survival of mutant female mice affected by severe pregnancy-induced cardiomyopathy. Gene expression profiles confirmed that HFD increased fatty acid utilization and ameliorated cardiomyopathy markers. Importantly, HFD also decreased accumulation of aggregated CHCHD10 in the S55L heart, suggesting activation of quality control mechanisms. Overall, our findings indicate that metabolic therapy can be effective in mitochondrial cardiomyopathies associated with proteotoxic stress.
Abstract 408: An Essential Protective Role for Cardiomyocyte Alpha1a-adrenergic Receptors in a Mouse Model of Myocardial Infarction

Abstract:Our previous work reveals that dabuzalgron, an oral agonist of alpha-1A adrenergic receptors (α1A-ARs) protects rodents from pathological cardiac stress, a finding in agreement with genetic studies of global α1A-AR overexpression/inactivation. However, it remains largely obscure whether cardiomyocyte-specific actions of α1A-ARs alone determines these cardioprotective effects. To test this essential gap in knowledge, we generated a new mouse line lacking α1A-AR only on cardiomyocytes by crossing αMHC-cre mice with floxed α1A-KO mice (CMKO= cre+ fl/fl, CMWT= cre-fl/fl). After validating specific deletion, baseline features were measured by conscious echo. No overt structural or functional anomaly was identified in either group. Fractional Shortening (FS, %) was virtually identical in 2 groups (51.7±0.7 vs. 52.4±5.3, p=NS). Next, both groups were subjected to permanent left anterior descending (LAD) ligation. By day 14, CMKO had significantly higher mortality (9/15 vs 2/13, p=0.025). To determine early changes that might account for this difference, hearts were harvested at day 3 post-ligation from another cohort. HW did not differ between two groups (7.0±1.1 vs. 7.2±1.1 mg/g·BW, p=NS), neither did LW (8.0±2.1 vs. 7.2±1.0 mg/g·BW, p=NS). Serum troponin level was numerically higher in CMKO mice (8.4±12.4 vs 4.8±6.2 ng/mL,p=0.38). CMKO mice had 51% larger injury (infarcted and leukocyte-infiltrating area) compared to CMWT (31.0±10.5 vs. 20.5±4.9% of LV area, p=0.05). Our preliminary postmortem examination suggested a higher trend of cardiac rupture in injured CMKO (3/5 vs 0/5). To test myocardial biomechanical properties between 2 groups, ventricles from mice 3d post ligation were then subjected to biaxial mechanical stress test. Myocardium from infracted CMKO mice exhibited 70% more stiffness (dt Stress/dt Strain) CMWT (190±34.29 vs.111±42 kPa, p=0.004). Collectively, our results demonstrate that mice deficient of α1A-AR on cardiomyocytes had lower late survival, more profound early pathological injury & adverse mechanical properties after LAD ligation, suggesting that activation of cardiomyocyte α1A-ARs plays an essential role in protecting experimental myocadiac infarction, holds the promise of future clinical implications.

Abstract:Objectives: We hypothesized that systemic administration of high-mobility group box 1 fragment attenuates the progression of myocardial fibrosis and cardiac dysfunction in a hamster model of dilated cardiomyopathy by recruiting bone marrow mesenchymal stem cells thus causing enhancement of a self-regeneration system. Methods: Twenty-week-old J2N-k hamsters, which are δ-sarcoglycan-deficient, were treated with systemic injection of high-mobility group box 1 fragment (HMGB1, n = 15) or phosphate buffered saline (control, n = 11). Echocardiography for left ventricular function, cardiac histology, and molecular biology were analyzed. The life-prolonging effect was assessed separately using the HMGB1 and control groups, in addition to a monthly HMGB1 group which received monthly systemic injections of high-mobility group box 1 fragment, 3 times (HMGB1, n = 11, control, n = 9, monthly HMGB1, n = 9). Results: The HMGB1 group showed improved left ventricular ejection fraction, reduced myocardial fibrosis, and increased capillary density. The number of platelet-derived growth factor receptor-alpha and CD106 positive mesenchymal stem cells detected in the myocardium was significantly increased, and intra-myocardial expression of tumor necrosis factor α stimulating gene 6, hepatic growth factor, and vascular endothelial growth factor were significantly upregulated after high-mobility group box 1 fragment administration. Improved survival was observed in the monthly HMGB1 group compared with the control group. Conclusions: Systemic high-mobility group box 1 fragment administration attenuates the progression of left ventricular remodeling in a hamster model of dilated cardiomyopathy by enhanced homing of bone marrow mesenchymal stem cells into damaged myocardium, suggesting that high-mobility group box 1 fragment could be a new treatment for dilated cardiomyopathy.

Abstract:Background This study aimed to investigate the effects of combined alpha-lipoic acid (ALA) and mitoquinone (Mito Q) supplementation on cardiac function and the underlying mechanisms in aged rats with myocardial infarction (MI). Methods The aged rats underwent left anterior descending artery (LADA) occlusion for 30 min, followed by reperfusion for 24 h. ALA (100 mg/kg, gavage) and Mito Q (10 mg/kg, IP) were administered daily for two weeks before ischemia. Cardiac function, inflammatory, and apoptotic markers were evaluated 24 h after ischemia. Results The results of this study indicated that the administration of the combination of ALA and Mito Q significantly improved cardiac function. This improvement was linked to a reduction in the expression of pro-inflammatory cytokines TNF-α, IL-6, and IL-1β ( P < 0.001) and apoptotic markers (Bax, caspase-3, and Cyt-c), as well as a decrease in the percentage of TUNEL-positive cells (P < 0.001). Conclusion The study revealed that combined intervention synergistically mitigated cardiac dysfunction by suppressing inflammatory and apoptotic pathways in aged rats with MI. Further research is needed to validate the potential of ALA and Mito Q as therapeutic options for elderly people at risk of heart attacks.

Abstract:The effects of n3 polyunsaturated fatty acids (PUFA) on cardiovascular disease are controversial. We currently explored the effects of various ratios of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) on high-fat-induced atherosclerosis. In model apoE(-/-) mice, high-fat diets (HFD) were partially replaced with fish and algal oils (DHA/EPA 2:1, 1:1 and 1:2) and/or plant oils enriched in linoleic and alpha-linolenic acids with an n6/n3 ratio of 4:1. PUFA supplementation significantly reduced the atherosclerotic plaque area, serum lipid profile, inflammatory response, aortic ROS production, proinflammatory factors and scavenger receptor expression as compared to those in the HFD group. However, plant oils did not have a significant effect on the following: serum HDL-C level; aortic ABCA1, ABCG1 and LAL mRNA expression; and CD36 and LOX-1 protein expression. Compared to the plant-oil-treated group, the DHA/EPA 1:1 group had a smaller atherosclerotic plaque area, higher serum HDL-C levels and lesser CD36 and MSR-1 mRNA expression; the DHA/EPA 2:1 group had lower serum TC, LDL-C and TNF-α levels and lower aortic ROS levels. Our study suggested that n3 PUFA from animals had more potent atheroprotective effects than that from plants. Supplementation involving higher DHA/EPA ratios and an n6/n3 ratio of 4:1 was beneficial for reducing serum "bad cholesterol" and a 1:1 DHA/EPA ratio with an n6/n3 ratio of 4:1 was beneficial for improving serum "good cholesterol" and inhibiting ox-LDL uptake. Our results suggest that achieving an n6/n3 ratio of 4:1 in the diet is also important in addition to having an optimal DHA/EPA ratio.
