Maturation, thickness and density of new bone after distraction osteogenesis of dog zygoma

Abstract:AIM: To observe the effect of distraction osteogenesis of zygoma based on the bony structure and bony density of new bone formation. METHODS: The experiment was carried out in the laboratory of Department of Experimental Surgery, the Fourth Military Medical University from October 2004 to April 2005. ①In the study side, the 10 mm segmental maxillary defects was prepared by removing part of zygomatico-orbital process of maxilla under general anesthesia, and the distraction osteogenesis of zygoma were performed on 6 dogs to simulate large defects of maxilla, the zygoma was distracted with a rate of 0.5 mm every time, twice per day for ten days with 10 mm distraction. The bone on the opposite side was preserved as control side. ②The process of bone formation was examined by X-ray. The specimen were taken in 8, 12 and 16 weeks respectively after distraction to perform the gross specimen observation, bone density analysis and scanning electron microscopy study. RESULTS:①All the 6 dogs completed the experiment, and the bone formation was smooth and rapid. ②New bone formed well in 8 weeks, but the bone structure, density and mature status of study side was lower than normal bone. The bone density of the study side was 56.9% of the control side. Then with the bone formed continuously, bone formation was almost the same to normal bone at the 16th week, the bone density of study side was 86.1% of the control side. CONCLUSION: In 8 weeks of consolidation, new bone forms with high quality and quantity, the bone defects are nearly repaired. Then, with the continuously regenerated and matured of bone, the bone density is strengthened, and at the 16th week, the density, thickness and maturation of the new bone are close to those of normal bone. At the 8th to 12th week of consolidation, the distractor can be disassembled to consider further therapy.
