Annexin A2 and Its Associated Genes in Alveolar Epithelial Type II Cells

Abstract:Annexin A2, a calcium‐dependent, phospholipid‐binding protein, is a substrate for several protein kinases. It has roles in the regulation of exocytosis, endocytosis, thrombolysis and cytoskeleton organization. Annexin A2 is abundantly expressed in alveolar type II cells and is known to be involved in lung surfactant secretion. Nevertheless, relatively little is known about its related targets and cellular pathways. In an effort to identify annexin A2‐regulated or related proteins, we knocked down annexin A2 by RNAi and performed cDNA microarray analysis. Fourteen down‐regulated and three up‐regulated genes were validated by real‐time quantitative PCR. When the loss of annexin A2 was rescued by the overexpression of EGFP‐tagged human annexin A2, the expression of 9 genes (Ata1A1, Fn1, Id‐1, RAB14, Prdx6, S100A6, S100A1, Retnla, p55cdc, and Ata1A2) returned to their normal level, indicating that these genes are indeed annexin A2‐associated targets. One of the identified targets, DNA binding protein inhibitor 1 (ID1) can directly interact with annexin A2 as determined by a mammalian two‐hybrid assay. Our results provide important clues for the future study of annexin A2‐involved pathways in alveolar type II cells.
