Abstract:Abstract Covalent organic frameworks (COFs) as a type of porous and crystalline covalent organic polymer are built up from covalently linked and periodically arranged organic molecules. Their precise assembly, well-defined coordination network, and tunable porosity endow COFs with diverse characteristics such as low density, high crystallinity, porous structure, and large specific-surface area, as well as versatile functions and active sites that can be tuned at molecular and atomic level. These unique properties make them excellent candidate materials for biomedical applications, such as drug delivery, diagnostic imaging, and disease therapy. To realize these functions, the components, dimensions, and guest molecule loading into COFs have a great influence on their performance in various applications. In this review, we first introduce the influence of dimensions, building blocks, and synthetic conditions on the chemical stability, pore structure, and chemical interaction with guest molecules of COFs. Next, the applications of COFs in cancer diagnosis and therapy are summarized. Finally, some challenges for COFs in cancer therapy are noted and the problems to be solved in the future are proposed.

Abstract:For decades, covalent organic frameworks (COFs) have attracted wide biomedical interest due to their unique properties including ease of synthesis, porosity, and adjustable biocompatibility. Versatile COFs can easily encapsulate various therapeutic drugs due to their extremely high payload and porosity. COFs with abundant functional groups can be surface-modified to achieve active targeting and enhance biocompatibility. In this paper, the latest developments of COFs in the biomedical field are summarized. First, the classification and synthesis of COFs are discussed. Cancer diagnosis and treatment based on COFs are studied, and the advantages and limitations of each method are discussed. Second, the specific preparation methods to obtain specific therapeutic properties are summarized. Finally, based on the combination and modification of COFs with various components, this review system summarizes different combination therapies. In addition, the main challenges faced in COF research and prospects for applying COFs to cancer diagnosis and treatment are evaluated. This review provides enlightening insights into the interdisciplinary research on COFs and applications in biomedicine, which highlight the great expectations for their further clinical transformation.

Abstract:Cancer has become one of the main causes of death worldwide. Chemotherapy as one of the main therapy modalities is very unsatisfactory. The various nanocarriers have brought new opportunities for effective tumor treatment. However, most of the current nanocarriers still suffer from low efficiency and confront significant challenges in overcoming multiple biological barriers. Compared with conventional nanocarriers, covalent organic frameworks (COFs) with unique and attractive features exhibited great potential to serve as a promising platform for anticancer drug delivery. In this review, we first summarize the strategies and challenges of nanocarriers for cancer chemotherapy and then highlight the recent advances in COF-based nanocarriers for improved delivery of chemotherapeutic agents. Finally, the challenges remaining for COF-based nanocarriers for clinical applications are outlined.

Abstract:As a newly emerging crystalline porous materials, covalent organic frameworks (COFs) possess fascinating structures and some specific features such as modularity, crystallinity, porosity, stability, versatility, and biocompatibility. Besides adsorption/separation, sensing, catalysis and energy, COFs recently show a new trend in biomedical application. This contribution provides an overview of recent developments of COF-based medicines in cancer therapeutics, including drug delivery, photodynamic therapy (PDT), photothermal therapy (PTT), and combined therapy. Furthermore, major challenges and developing trend in this field are also discussed. These recent developments are summarized and discussed to help encourage further contributions in this emerging and promising field.

Abstract:<p>Nanoscale metal‐organic frameworks (NMOFs) have attracted increasing attention for biomedical applications due to their large specific surface area, good biocompatibility, adjustable structures, and diverse functions. By choosing appropriate metal ions and ligands, NMOFs can be synthesized and regulated to assist the diagnosis and treatment of cancer, acting as imaging agents, drug carriers, and cancer therapeutic agents. This review summarizes the recent advances of NMOFs in synthesis, biocompatibility, imaging, and applications in cancer therapies. Among these, the term "biocompatibility" is used to outline their various biological characteristics, and it is mainly discussed from the aspects of size and surface properties of NMOFs. The imaging section mainly emphasizes the application advantages of NMOFs as imaging agents in magnetic resonance, computed tomography, and fluorescence imaging. The applications of NMOFs in four cancer therapies, including phototherapy, radiotherapy, microwave therapy, and ultrasonic therapy, are addressed, especially for thermal and dynamic therapy. Finally, the prospects and challenges of NMOFs in imaging and cancer therapies are also discussed.</p>

Abstract:Despite consecutive efforts devoted to the establishment of innovative therapeutics for cancer control, cancer remains as a primary global public health concern. Achieving controlled release of anti-cancer agents may add great value to the field of oncology that requires the involvement of nanotechnologies. Metal organic frameworks (MOFs) hold great promise in this regard owing to their unique structural properties. MOFs can act as superior candidates for drug delivery given their porous structure and large loading area, and can be prepared into anti-cancer therapeutics by incorporating stimuli-sensitive components into the ligands or nodes of the framework. By combing through chemical and physical features of MOFs favorable for onco-therapeutic applications and current cancer treatment portfolios taking advantages of these characteristics, this review classified MOFs feasible for establishing controlled anti-cancer modalities into 6 categories, outlined the corresponding strategies currently available for each type of MOF, and identified understudied areas and future opportunities towards innovative MOF design for improved or expanded clinical anti-cancer applications.

Abstract:<p>Owing to the growing risk of cancer to human health in recent decades, various strategies to treat cancer have been developed. Traditional strategies including chemotherapy and radiotherapy have been widely employed in cancer treatment with wonderful performance. However, there is an urgent need for novel methods to address the serious side effects of traditional treatment methods. As a rising nanomaterial, nanoscale metal-organic frameworks (NMOFs) have various practical applications in cancer therapy because of their advantages, such as tunable chemical compositions, crystalline structures, and high porosity. For their nanoscale size, NMOFs have better long-term biodegradability and lower toxicity to normal tissue. These superior characteristics contributed to their applications in photodynamic therapy (PDT), photothermal therapy (PTT), and chemodynamic therapy (CDT). Herein, diversified strategies based on metal-organic frameworks for cancer therapy were reviewed. First, the preparation approaches of MOFs and the MOFs-based monofunctional strategies for cancer therapy including drug carrier, chemodynamic therapy, radiotherapy, photothermal therapy, and immunotherapy were summarized. Thereafter, the diversified MOFs-based combined strategies such as PDT/CDT, enzyme cascade catalysis in CDT, immunotherapy/CDT/PDT were described. Finally, the challenges associated with MOFs in successful clinical translation were discussed.</p>

Abstract:With large specific surface area, controllable pore size, increased active sites, and structural stability, two-dimensional metal organic frameworks (2D MOFs) have emerged as promising nanomedicines in cancer therapy. These distinctive features make 2D MOFs particularly advantageous in cancer treatment and the corresponding application has gained considerable popularity, signifying significant application potential. Herein, recent advances in various applications including drug delivery and chemotherapy, photodynamic therapy, sonodynamic therapy, chemodynamic therapy, catalytic therapy, and combined therapy were summarized, with emphasis on the latest progress of new materials and mechanisms for these processes. Owing to their ability to be constructed through covalent bonds involving light elements, such as hydrogen, boron, carbon, nitrogen, and oxygen, COFs offer greater stability and lower cytotoxicity than metal organic frameworks do, addressing critical limitations in in vivo applications. Their unique attributes, such as high surface area, customizable pore sizes, and versatile surface functionalities, make them ideal for various biomedical applications. This review aims to provide an overview of the recent advancements in modern COFs for biomedical uses. First, a variety of methods for the synthesis of COFs are outlined, which ensures their suitability for medical use. Abstract:The rapid development of nanotechnology has generated numerous ideas for cancer treatment, and a wide variety of relevant nanoparticle platforms have been reported. Metal-organic frameworks (MOFs) have been widely investigated as an anti-cancer drug delivery vehicle owing to their unique porous hybrid structure, biocompatibility, structural tunability, and multi-functionality. MOF materials with catalytic activity, known as nanozymes, have applications in photodynamic and chemodynamic therapy. Nucleic acids have also attracted increasing research attention owing to their programmability, ease of synthesis, and versatility. A variety of functional DNAs and RNAs have been applied both therapeutically (gene-targeting drugs for cancer treatment) and nontherapeutically (used as modified materials to enhance the therapeutic effects of other nanomedicines). The combined use of MOFs and functional nucleic acids have been extensively investigated and has been associated with excellent tumor-suppressor activity in various treatment methods. In this review, we summarize the progress in the research and development of tumor therapy based on MOFs and nucleic acid delivery over recent years, focusing on the combinational use of different delivery and design strategies for MOF/therapeutic nucleic acid platforms. We further summarize the strategies for combining MOFs (universal carrier, functional carrier) and nucleic acids (therapeutic nucleic acids, nontherapeutic nucleic acids) and discuss the corresponding therapeutic effects in cancer treatment. Abstract:Nanoscale metal-organic frameworks (MOFs) have been widely used as controlled drug delivery vehicles and cancer therapy agents due to their intrinsic superior properties. Scientists have made remarkable achievements in the field of nanomedicine by using the MOFs and MOF-based multifunctional nanomaterials due to the easy synthesis into nanoscale and functionalization. In this review, we highlight the recent progress of nanoscale MOFs as drug delivery vehicles for cancer theranostics. We divide the discussion into three parts. The first and second parts focus on the drug delivery of unmodified MOF and modified MOFs, respectively, while the third part focuses on porphyrin MOFs as photosensitizers for photodynamic therapy. Finally, we conclude by identifying areas of research that we believe will propel the translation and application of MOFs.

Reticular Chemistry-Enabled Sonodynamic Activity of Covalent Organic Frameworks for Nanodynamic Cancer Therapy.

Abstract:The development of covalent organic framework (COF) sonosensitizers with intrinsic sonodynamic effects is highly desirable. However, such COFs are generally constructed using small-molecule photosensitizers. Herein, we report that the reticular chemistry-based synthesis of COFs from two inert monomers yields a COF-based sonosensitizer (TPE-NN) with inherent sonodynamic activity. Subsequently, a nanoscale COF TPE-NN is fabricated and embedded with copper (Cu)-coordinated sites to obtain TPE-NN-Cu. Results show that Cu coordination can enhance the sonodynamic effect of TPE-NN, whereas ultrasound (US) irradiation for sonodynamic therapy can augment the chemodynamic efficacy of TPE-NN-Cu. Consequently, TPE-NN-Cu upon US irradiation shows high-performance anticancer effects based on mutually reinforced sono-/chemo-nanodynamic therapy. This study reveals the backbone-originated sonodynamic activity of COFs and proposes a paradigm of intrinsic COF sonosensitizers for nanodynamic therapy.

Abstract:Nanomaterials, with unique physical, chemical, and biocompatible properties, have attracted significant attention as an emerging active platform in cancer diagnosis and treatment. Amongst them, metal–organic framework (MOF) nanostructures are particularly promising as a nanomedicine due to their exceptional surface functionalities, adsorption properties, and organo-inorganic hybrid characteristics. Furthermore, when bioactive substances are integrated into the structure of MOFs, these materials can be used as anti-tumor agents with superior performance compared to traditional nanomaterials. Abstract:Recently, nanomaterials have attracted extensive attention in cancer-targeting therapy and as drug delivery vehicles owing to their unique surface and size properties. Multifunctional combinations of nanomaterials have become a research hotspot as researchers aim to provide a full understanding of their nanomaterial characteristics. In this study, metal–organic framework-capped gold nanorod hybrids were synthesized. Our research explored their ability to kill tumor cells by locally increasing the temperature via photothermal conclusion. The specific peroxidase-like activity endows the hybrids with the ability to disrupt the oxidative balance in vitro. Simultaneously, chemotherapeutic drugs are administered and delivered by loading and transportation for effective combinatorial cancer treatment, thereby enhancing the curative effect and reducing the unpredictable toxicity and side effects of large doses of chemotherapeutic drugs. These studies can improve combinatorial cancer therapy and enhance cancer treatment.
