Abstract:The catalytic properties and thermodynamic kinetics of the endoglucanase from a marine Bacillus subtilis were analyzed. Optimum pH and temperature of the endoglucanase activity were 5.0 and 35 degrees C. The endoglucanase activity, melt point temperature was 1.13 folds (247.02 U . ml(-1)), 2.1 degrees C higher (392 degrees C) in 6% ethanol solution than that (218.60 U . ml(-1)), (37.1 degrees C) in free ethanol. At 40 degrees C-55 degrees C, Gibbs free energy, Delta G, and the content of alpha-helix was higher in 6% ethanol solution than that in ethanol free solution. The increasing of alpha-helix content led to higher activity and better thermostability in ethanol solution. The cold adapt ethanol tolerant endoglucanase was valuable for bioethanol product by simultaneous saccharification and fermentation process. Study on the Soluble Inducer for Cellulase Production Derived from Glucose by Glucosylase

Abstract:Cellulose in insoluble solid state is commonly used as carbon source and inducer for cellulase production,which results in some problems,such as strong mass transfer resistance,inaptitude for fed-batch fermentation and low-yield production.A soluble inducer for cellulase production was obtained from glucose by transglycosylation of glucosylase in this study.The high performance liquid chromatogram of the mixture derived from glucose by glucosylase shows that the mixture contains sophorose which is a kind of strong inducer for cellulase production.When the glucose of 300～500 mg mL 1 was used as substract and the transglycosylation reaction of glucosylase was conducted at pH of 3.0～4.5 and 50℃for 100 h,the concentration of sophorose in the mixture can rach 40 mg mL 1.Using the mixture derived from glucose by glucosylase to produce the cellulase by Trichoderma reesei in batch process,the mixture plays the roles of carbon source and inducer simultaneously;after 5 h,the cellulase produced can be detected,and after 60 h,its filter paper activity reaches 1.8 IU mL 1 which is 14 times higher than that can be produced by using glucose as carbon source.The above results suggest a new approach for the enzymatic preparation of soluable inducer for cellulase production,which is meaningful for enhancing the efficiency of cellulase production and accelerating the application of cellulase in bioethanol production.
Synthesis of galactooligosaccharides by β-galactosidase from Halomonas sp. S62

Abstract:We used the Halomonas sp. S62 β-galactosidase to synthesize galactooligosaccharides from lactose, to explore galactooligosaccharides synthesis efficiency. To improve the yield of galactooligosaccharides, the reaction conditions were optimized, and the orthogonal test was carried out by the reaction temperature, p H, enzyme dosage and substrate concentration. The optimal reaction conditions were as follows: reaction temperature 40 ℃, p H 7. 0, enzyme dosage 50 U/m L, substrate concentration 300 g/L.The maximal yield of galactooligosaccharides was (41. 91±0. 27) ％ at 6 h and the lactose consumption rate was (82. 47±0. 38) ％.Within 4-8 h, the yield of galactooligosaccharides was above 40％ and the lactose consumption rate was over80％.The yield of galactooligosaccharides was maintained above 40％ within 4-8 h of the reaction. The consumption rate of lactose was all above 80％. The utilization rate of lactose was increased while the high production of galactooligosaccharides was achieved, which was beneficial to the reduction of production cost, laying the foundation for the industrial application of low-temperature S62 β-galactosidase.
Hydrothermal Decomposition of Carbohydrates to Levulinic Acid with Catalysis by Ionic Liquids

Abstract:The hydrothermal decomposition reactions of cellulose, glucose, and fructose were catalyzed by ionic liquids (ILs) to produce levulinic acid (LA). Among 18 types of ionic liquids with different anions, the ionic liquids containing haloids and hydrogen sulfates exhibited a universal activity for the conversion of fructose, glucose, and cellulose to produce LA. Reusability tests for [PrSO(3)HMIm]Cl and [BSO(3)HMIm]HSO4 were carried out and revealed that [BSO(3)HMIm]HSO4 can be used over four cycles without a loss of activity. The effects of reactant loading, ionic liquid loading, reaction temperature, and time on the decomposition of glucose and cellulose were investigated with [BSO(3)HMIm]HSO4. The optimum reaction conditions were obtained as a [BSO(3)HMIm]HSO4 loading of 0.5 g, a reactant loading of 0.05 g, a temperature of 180 degrees C, and a reaction time of 1 h. Yields of 60.8% from glucose and of 54.S% from cellulose were obtained for the catalysis with [BSO(3)HMIm]HSO4. Kinetics studies of the decomposition of glucose and cellulose were conducted. The rate constants at each temperature were obtained. For the decomposition of glucose, the activation energies for the conversion of glucose to LA and conversion of glucose to humins were determined as 121.1 +/- 4.7 kJ/mol and 114.6 +/- 6.1 kJ/mol, respectively. For the decomposition of cellulose, the activation energies for the conversion of cellulose to glucose, conversion of glucose to LA, and conversion of glucose to humins were determined as 186.8 +/- 7.4 kJ/mol, 114.0 +/- 2.7 kJ/mol, and 148.3 +/- 11.2 kJ/mol, respectively.

Abstract:Galacto-oligosaccharides (GOS) are prebiotic compounds that are mainly used in infant formula to mimic bifidogenic effects of mother's milk. They are synthesized by β-galactosidase enzymes in a trans -glycosylation reaction with lactose. Many β-galactosidase enzymes from different sources have been studied, resulting in varying GOS product compositions and yields. The in vivo role of these enzymes is in lactose hydrolysis. Therefore, the best GOS yields were achieved at high lactose concentrations up to 60% wt , which require a relatively high temperature to dissolve. Some thermostable β-glucosidase enzymes from thermophilic bacteria are also capable of using lactose or para nitrophenyl-galactose as a substrate. Here, we describe the use of the β-glucosidase BglA from Thermotoga maritima for synthesis of oligosaccharides derived from lactose and cellobiose and their detailed structural characterization. Also, the BglA enzyme kinetics and yields were determined, showing highest productivity at higher lactose and cellobiose concentrations. The BglA trans -glycosylation/hydrolysis ratio was higher with 57% wt lactose than with a nearly saturated cellobiose (20% wt ) solution. The yield of GOS was very high, reaching 72.1% wt GOS from lactose. Structural elucidation of the products showed mainly β (1 → 3) and β (1 → 6) elongating activity, but also some β (1 → 4) elongation was observed. The β-glucosidase BglA from T. maritima was shown to be a very versatile enzyme, producing high yields of oligosaccharides, particularly GOS from lactose.

Abstract:BackgroundPretreating biomass with ionic liquids (IL) increases enzyme accessibility and cellulose is typically recovered through precipitation with an anti-solvent. An industrially feasible pretreatment and hydrolysis process requires robust cellulases that are stable and active in the presence of either small amounts of ILs co-precipitated with recovered cellulose or for saccharifications in the presence of IL. β-glucosidase (BG) hydrolyzes cellobiose into two molecules of glucose (Glc) and is the last step of biomass hydrolysis. These enzymes are prone not only to product inhibition by glucose but also to inactivation by ILs. With increasing interest in IL-based pretreatment methods, there is increasing focus toward a search for Glc-tolerant and IL-tolerant BG.ResultsWe identified a BG belonging to the GH1 family, H0HC94, encoded in Agrobacterium tumefaciens 5A, and cloned and overexpressed the protein in Escherichia coli. H0HC94 exhibited high enzymatic activity with β-glycosidic substrates (248 µmol/min/mg on pNPGlc and 262 µmol/min/mg on cellobiose) and tolerant to Glc (apparent Ki = 686 mM). Further evidence of Glc-based stabilization came from the increase in melting temperature of H0HC94, with increasing Glc concentrations. The half-life of H0HC94 also increased between 2- and 20-fold in the presence of increasing concentrations of Glc. In the presence of 0.9 M of different [C2mim]-based ionic liquids, the specific activity of H0HC94 decreased by around 20–30 %. However, the addition of 100 mM glucose to the IL-enzyme mix resulted in a more stable enzyme as evidenced by the slight recovery of H0HC94 melting temperature and up to tenfold increase in half-life. This higher stability came at a cost of 2–10 % decrease in specific activity. The steady-state kinetic analyses for a subset of the ionic liquids tested indicate that the enzyme undergoes uncompetitive inhibition by glucose and ionic liquid, indicating the possibility of binding of the ionic liquid and glucose to the enzyme–substrate complex.ConclusionsH0HC94 is a Glc-stabilized BG that is also tolerant up to 0.9 M concentrations of different IL’s and indicates the possibilities of using an IL–Glc-based cellulose solvent that displays enzyme-compatibility.

Abstract:In this study, a heterodimeric β-galactosidase (PpenLac2) with high transgalactosylation activity from Pediococcus pentosaceus was screened from 64 lactic acid bacteria, and heterologously expressed in Escherichia coli . The optimal pH and temperature of PpenLac2 were 8.0 and 45 °C, respectively. The galactooligosaccharides (GOS) produced by PpenLac2 showed a preference for β-(1,6) linkages, and the primary products were allolactose and 6'-galactosyl lactose. Based on homology modeling and molecular docking, mutation sites were designed to modify PpenLac2 to enhance GOS yield. The key residue impacting the transgalactosylation activity of the enzyme was Phe600. Saturated mutation at the site of Phe600 revealed that the majority of mutants had higher yields of 6'-galactosyl lactose. F600H gained the highest GOS yield increasing from 37.6% to 46.7%, owing to the mutation driving the rotation of Trp 922 and generating Pi-Pi stacking between the pyran ring of substrate and the aromatic ring of Trp922, which promoted the transgalactosylation reaction without affecting hydrolysis reaction. Following the optimization of the reaction conditions, F600H achieved an even higher GOS yield of 52.4%. This study provides a promising enzyme for the synthesis of GOS and serves as a guide for the rational design for the improvement of transgalactosylation efficiency of GH2 family members.

Molecular Basis of Aqueous-like Activity of Lipase Treated with Glucose-Headed Surfactant in Organic Solvent.

Abstract:Lipases are useful as catalysts, particularly for the kinetic resolution of racemic alcohols and esters. However, their industrial applications are limited by their poor activities in organic media. We recently found that a lipoprotein lipase from Burkholderia species displays dramatically enhanced activity in organic solvent if the protein is coated with glucose-headed surfactant (GHS). Here we investigate the molecular basis of this enhanced enzymatic activity in organic solvent by performing molecular dynamics simulations on Burkholderia cepacia lipase as a model enzyme. Our simulation results indicate that the enhanced activity of lipase stems from a dual function of GHSs different in water and organic solvent. GHS molecules maintain the open conformation of lipase by providing lipid-like microenvironment surrounding the active site in water and stabilize its native active conformation by providing water-like microenvironment around the surface of the lipase in the organic solvent. Our data also suggest the role of organic cosolvent that can facilitate closed-to-open conformational changes during the freeze-drying process. The computational approach in this study lays its potential for guiding the design of more effective surfactant molecules to improve the activity of lipases in organic solvent.

Abstract:β-D-Galactosidase from Kluyveromyces lactis was immobilized on glutaraldehyde-activated chitosan and used in a packed-bed reactor for the continuous hydrolysis of lactose and the synthesis of galactooligosaccharides (GOS). The biocatalyst was tested for its optima pH and temperature, thermal stability in the presence of substrate and products, and operational stability. Immobilization increased the range of operational pH and temperature, and the enzyme thermal stability was sharply increased in the presence of lactose. Almost complete lactose hydrolysis was achieved for both milk whey and lactose solution at 37 °C at flow rates up to 2.6 mL min(-1). Maximal GOS concentration of 26 g L(-1) was obtained at a flow rate of 3.1 mL min(-1), with a productivity of 186 g L(-1) h(-1). Steady-state operation for 15 days showed the reactor stability concerning lactose hydrolysis.

Abstract:α-glucosidase is an essential enzyme for the production of isomaltooligosaccharides (IMOs). Allowing α-glucosidase to operate at higher temperatures (above 60 °C) has many advantages, including reducing the viscosity of the reaction solution, enhancing the catalytic reaction rate, and achieving continuous production of IMOs. In the present study, the thermal stability of α-glucosidase was significantly improved by constructing cyclized proteins. We screened a thermotolerant α-glucosidase (AGL) with high transglycosylation activity from Thermoanaerobacter ethanolicus JW200 and heterologously expressed it in Bacillus subtilis 168. After forming the cyclized α-glucosidase by different isopeptide bonds (SpyTag/SpyCatcher, SnoopTag/SnoopCatcher, SdyTag/SdyCatcher, RIAD/RIDD), we determined the enzymatic properties of cyclized AGL. The optimal temperature of all cyclized AGL was increased by 5 °C, and their thermal stability was generally improved, with SpyTag-AGL-SpyCatcher having a 1.74-fold increase compared to the wild-type. The results of molecular dynamics simulations showed that the RMSF values of cyclized AGL decreased, indicating that the rigidity of the cyclized protein increased. This study provides an efficient method for improving the thermal stability of α-glucosidase.

Abstract:The bond selective hydrolysis of glycyrrhizin (GL) to glycyrrhetic acid 3-O-mono-β-d-glucuronide (GAMG) catalyzed by recombinant β-d-glucuronidase from Escherichia coli BL21 (PGUS-E) was successfully performed in an ionic liquid (IL)/buffer biphasic system. Five ILs were analyzed, however, a hydrophobic IL 1-butyl-3-methylimidazolium hexafluorophosphate ([BMIM]PF6) showed the best biocompatibility with PGUS-E. An obvious enhancement in the initial reaction rate, substrate conversion, GAMG yield and chemical bond selectivity (Scb) was observed using 40% (v/v) [BMIM]PF6/buffer as the reaction medium when compared to the acetate buffer medium. Under the optimized conditions (pH 6.0, temperature 50°C, substrate concentration 6mM and shaking speed 200rpm), the initial reaction rate, the GAMG yield and the Scb reached 3.15mMh−1, 74.36% and 98.12%, respectively. The recyclability of [BMIM]PF6 was also studied and found to be reusable for five batches with high recovery percentage (≥92%). Furthermore, the desired product and byproduct were easily separated since they were distributed in different phases. Additionally, higher Vmax (3.14 versus 2.24mMh−1), lower apparent Km (1.21 versus 1.80mM) and Ea (25.97 versus 32.60kJmol−1) were achieved in [BMIM]PF6/buffer biphasic system than that in monophasic buffer system.
Rhamnolipid enhanced beta-glucosidase from Paenibacillus sp. LLZ1 for in situ lignocellulose saccharification in ionic liquids

Abstract:Low β-glucosidase activity and stability are challenging limitations for the in situ saccharification of lignocellulose pretreated by ionic liquids. The addition of 0.1% (v/v) rhamnolipid, an ionic biosurfactant, was found to enhance the activity of β-glucosidase from <i>Paenibacillus</i> LLZ1 by 39% through a synergistic effect with 5% [Emim]OAc. The concentration of [Emim]OAc was further optimized to 2.5% (v/v) obtaining as high as 48% PsBGL activity increase. Fluorescence and near-UV circular dichroism spectroscopy analyses revealed looser protein structure and more hydrophilic microenvironment in the system of rhamnolipid and [Emim]OAc. Though ILs showed deactivation effect on cellulases, rhamnolipid was speculated to alleviate protein denaturation significantly. The reaction system was further applied in the hydrolysis of cellobiose and bagasse cellulose. The conversion was increased by 33% for cellobiose and by 62% for bagasse cellulose, demonstrating the application potential of the reaction system. The improvement was attributed to both reduced cellobiose inhibition and effective substrate solubilizing, and the latter was monitored by scanning electron microscopy to show the morphological change of substrates directly. This study proved the effect of surfactant in in situ enzymatic saccharification of lignocellulose and provided a practical enzymatic hydrolysis system.

Abstract:Cyclodextrin glycosyltransferase (CGTase) is a preferable biocatalyst for production of cyclodextrin and phenols glycosylated derivatives due to its cost-effective donors and broad range of acceptors. Organic solvents (OSs) are often beneficial for solubilizing hydrophobic substrates and product specificity, but often harmful towards CGTases. Herein, using Paenibacillus macerans CGTase ( Pm CGTase) as a template and DMSO as a screening solvent, semi-rational engineering was performed at 18 surface residues by a developed high throughput screening method. Using site saturation mutagenesis and iterative saturation mutagenesis, 5 OSs-resistant variants (G539I, G539V, G539I/R146F, G539V/R146A, G539I/R146F/D147N) were obtained. Compared with WT, the best variant G539I/R146F/D147N showed a 79.3 % enhanced residual activity at 30 % DMSO and a 60 % increased t 1/2 value at both 40 and 50 °C. In addition, a decreased K m value and an increased k cat value led to 41.9 % higher catalytic efficiency of G539I/R146F/D147N than WT in 15 % DMSO. This variant was also applied to glycosylation of sophoricoside in 20 % DMSO, leading to 26.2 % increased conversion. Molecular dynamics demonstrates that its enhanced OSs resistance may be attributed to the strengthened structural compactness and rigidity. Our results provide guidance for engineering OSs resistance of Pm CGTase, which would further improve its application potential.

Abstract:The stability of cellulase activity in the presence of ionic liquids (ILs) is critical for the enzymatic hydrolysis of insoluble cellulose pretreated with ILs. In this work, cellulase was incorporated in the liposomes composed of negatively charged 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphoglycerol (POPG) and zwitterionic phosphatidylcholines (PCs) with different length and degree of unsaturation of the acyl chains. The liposomal cellulase-catalyzed reaction was performed at 45°C in the acetate buffer solution (pH 4.8) with 2.0g/L CC31 as cellulosic substrate. The crystallinity of CC31 was reduced by treating with 1-butyl-3-methylimidazolium chloride ([Bmim]Cl) at 120°C for 30min. The liposomal cellulase continuously catalyzed hydrolysis of the pretreated CC31 for 48h producing glucose in the presence of 15wt% [Bmim]Cl. The charged lipid membranes were interactive with [Bmim](+), as elucidated by the [Bmim]Cl-induced alterations in fluorescence polarization of the membrane-embedded 1,6-diphenyl-1,3,5-hexatriene (DPH) molecules. The charged membranes offered the microenvironment where inhibitory effects of [Bmim]Cl on the cellulase activity was relieved. The maximum glucose productivity GP of 10.8 mmol-glucose/(hmol-lipid) was obtained at the reaction time of 48h with the cellulase incorporated in the liposomes ([lipid]=5.0mM) composed of 50mol% POPG and 1,2-dilauroyl-sn-glycero-3-phosohocholine (DLPC) with relatively short and saturated acyl chains.

Abstract:There are a variety of ways to make glucose-galactose syrup (GGS) and other products of lactose hydrolysis; therefore, research is still ongoing and will undoubtedly result in improved methods and lower costs. The aim of the study was to use a two-stage fermentation approach to increase the sweetness of glucose-galactose syrup. Comparing lactose hydrolysis with β-galactosidases, the enzyme Ha-Lactase 5200 (K. lactis) showed the highest hydrolysis yield but NOLATM Fit5500 (B. licheniformis) and GODO-YNL2 (K. lactis) hydrolysis yields varied. After the two-stage fermentation, the syrups from sweet whey permeate had shown the highest sweet taste intensity scores; the sweetest samples were 1NFS and 1HLS with a score of 9.2 and 9.3, respectively. The presence of fructose in the range of 14 ± 3 to 25 ± 1 %, significantly (p < 0.05) increased the sweetness of the syrups. Obtained syrups from whey permeates using enzymes NOLATM Fit5500 and Ha-Lactase 5200 contained less than 10% lactose. Additionally, results indicate that hydrolysis of lactose and subsequent enhancement of sweetness through glucose isomerisation may provide additional benefits through the production of galacto-oligosaccharides (GOS) in the range of 2 ± 1 to 34 ± 7%.

Abstract:Hyperthermostable endoglucanases of glycoside hydrolase family 12 from the archaeon Pyrococcus furiosus (EGPf) catalyze the hydrolysis of β-1,4-glucosidic linkages in cellulose and β-glucan structures that contain β-1,3- and β-1,4-mixed linkages. In this study, EGPf was heterologously expressed with Aspergillus niger and the recombinant enzyme was characterized. The successful expression of EGPf resulted as N-glycosylated protein in its secretion into the culture medium. The glycosylation of the recombinant EGPf positively impacted the kinetic characterization of EGPf, thereby enhancing its catalytic efficiency. Moreover, glycosylation significantly boosted the thermostability of EGPf, allowing it to retain over 80% of its activity even after exposure to 100 °C for 5 h, with the optimal temperature being above 120 °C. Glycosylation did not affect the pH stability or salt tolerance of EGPf, although the glycosylated compound exhibited a high tolerance to ionic liquids. EGPf displayed the highest specific activity in the presence of 20% (v/v) 1-butyl-3-methylimidazolium chloride ([Bmim]Cl), reaching approximately 2.4 times greater activity than that in the absence of [Bmim]Cl. The specific activity was comparable to that without the ionic liquid even in the presence of 40% (v/v) [Bmim]Cl. Glycosylated EGPf has potential as an enzyme for saccharifying cellulose under high-temperature conditions or with ionic liquid treatment due to its exceptional thermostability and ionic liquid tolerance. These results underscore the potential of N-glycosylation as an effective strategy to further enhance both the thermostability of highly thermostable archaeal enzymes and the hydrolysis of barley cellulose in the presence of [Bmim]Cl.

Abstract:Glycyrrhizin(GL),the biologically active compounds of liquorice,can be hydrolysed to glycyrrhetic acid 3-O-mono-β-D-glucuronide(GAMG)by whole cells containing β-D-glucuronidase.GAMG is widely used in the pharmaceutical and food industries due to its stronger physiological functions,higher biological activity,more favorable sweetness,and lower caloric value as compared with GL.The recombinant Pichia pastoris(r-PGUS-P)designated in this work expressing β-D-glucuronidase,was used as a whole-cell biocatalyst catalyzing GL to GAMG in non-aqueous solvents systems.The results showed that r-PGUS-P gave the highest yield of GAMG in 1-butyl-3-methylimidazolium hexafluorophosphate(PF6)/water(2∶8,vol)biphasic system.The optimal conditions of this reaction were determined as follows:optimum pH,substrate concentration,temperature and cell content were 5.4,6.0 mmol·L-1,45℃,and 8.0 g·L-1,respectively.Under these optimized conditions,69.6% yield of GAMG and 67.2% of the chemical bond selectivity was achieved after 58 h reaction time,increase by 12.4% and 12.61%,respectively against aqueous phase.Simultaneously,the ionic liquid PF6 also remained at higher recovery percentage of about 93.47% during repeated use for 7 reaction cycles.Especially,product GAMG and byproduct glycyrrhetinic acid(GA)got separated in biphasic system automatically that brought convenience for the separation and purification of GAMG.
Enhancing β-Galactosidase Performance for Galactooligosaccharides Preparation via Strategic Glucose Re-Tunneling

Abstract:This study focuses on the characterization and re-engineering of glucose transport in β-galactosidase (BglD) to enhance its catalytic efficiency. Computational prediction methods were employed to identify key residues constituting access tunnels for lactose and glucose, revealing distinct pockets for both substrates. In silico simulated saturation mutagenesis of residues T215 and T473 led to the identification of eight mutant variants exhibiting potential enhancements in glucose transport. Site-directed mutagenesis at T215 and T473 resulted in mutants with consistently enhanced specific activities, turnover rates, and catalytic efficiencies. These mutants also demonstrated improved galactooligosaccharide (GOS) synthesis, yielding an 8.1–10.6% enhancement over wild-type BglD yield. Structural analysis revealed that the mutants exhibited transformed configurations and localizations of glucose conduits, facilitating expedited glucose release. This study's findings suggest that the re-engineered mutants offer promising avenues for enhancing BglD's catalytic efficiency and glucose translocation, thereby improving GOS synthesis. By-product (glucose) re-tunneling is a viable approach for enzyme tunnel engineering and holds significant promise for the molecular evolution of enzymes.

Engineering of Cyclodextrin Glycosyltransferase Reveals pH-Regulated Mechanism of Enhanced Long-Chain Glycosylated Sophoricoside Specificity

Abstract:Sophoricoside glycosylated derivatives, especially long-chain glycosylated sophoricosides (LCGS), have greatly improved water solubility compared with sophoricoside. Here, cyclodextrin glycosyltransferase from Paenibacillus macerans (PmCGTase) was employed for sophoricoside glycosylation. Saturation mutagenesis of alanine 156, alanine 166, glycine 173, and leucine 174 was performed due to their nonconservative properties among α-, β-, and γ-CGTases with different product specificities. Variants L174P, A156V/L174P, and A156V/L174P/A166Y greatly improved the product specificity for LCGS. pH significantly affected the extent of glycosylation catalyzed by the variants. Further investigations revealed that the pH-regulated mechanism for LCGS synthesis mainly depends on a disproportionation route at a lower pH (pH 4) and a cyclization-coupling route at a higher pH (pH 8) and equivalent effects of cyclization-coupling and disproportionation routes at pH 5. Whereas short-chain glycosylated sophoricosides (SCGS) are primarily produced via disproportionation of maltodextrin at pH 4 and secondary disproportionation of LCGS at pH 8. At pH 5, SCGS synthesis mainly depends on a hydrolysis route by the wild type (WT) and a secondary disproportionation route by variant A156V/L174P/A166Y. Kinetics analysis showed a decreased Km value of variant A156V/L174P/A166Y. Dynamics simulation results demonstrated that the improved LCGS specificity of the variant is possibly attributed to the enhanced affinity to long-chain substrates, which may be caused by the changes of hydrogen bond interactions at the -5, -6, and -7 subsites. Our results reveal a pH-regulated mechanism for product specificity of CGTase and provide guidance for engineering CGTase toward products with different sugar chain lengths.IMPORTANCE The low water solubility of sophoricoside seriously limits its applications in the food and pharmaceutical industries. Long-chain glycosylated sophoricosides show greatly improved water solubility. Here, the product specificity of cyclodextrin glycosyltransferase (CGTase) for long-chain glycosylated sophoricosides was significantly affected by pH. Our results reveal the pH-regulated mechanism of the glycosylated product specificity of CGTase. This work adds to our understanding of the synthesis of long-chain glycosylated sophoricosides and provides guidance for exploring related product specificity of CGTase based on pH regulation.
Inhibition of Lactose Crystallisation in the Presence of Galacto-Oligosaccharide

Abstract:The stabilization of lactose in the form of amorphous (i.e. non-crystalline form) is the basic requirement to maintain the quality of relevant food and pharmaceutical products. The physiochemical properties of amorphous lactose mixed with galacto-oligosaccharide (GOS) were investigated. Water sorption, glass transition temperature, and crystallisation behaviour of lactose in the present of GOS (1:1 w/w) were measured at various water activity (0.11-0.75 a(w), 25 degrees C) and lactose mutarotation was also evaluated. All of them were compared with the physiochemical properties of trehalose-lactose (1:1 w/w). The addition of GOS to lactose increased the hygroscopicity of the mixture, as well as slightly increased the glass transition temperature compared to lactose alone. The critical water activity (at 0.68 a(w)) of lactose crystallisation was increased by the addition of GOS as compared to that of trehalose-lactose (1:1 w/w) (at 0.58 a(w)) or lactose alone (at 0.44 a(w)). The dramatical inhibition of lactose crystallisation with a lower crystallisation kinetic constant and the alternation of lactose crystal forms in the presence of GOS was observed as compared to the crystallisation behaviour of trehalose-lactose (1:1 w/w) and pure lactose at 0.68 and 0.75 a(w), 25 degrees C. Without affecting its T-g, the significantly delayed crystallisation of lactose in GOS-lactose mixture (1:1 w/w) was more likely due to the change of lactose mutarotation. As comparing to trehalose that is an effective inhibitor, GOS has a stronger ability to prevent lactose from crystallisation in hydrous matrices.
