Abstract:Macrobrachium rosenbergii nodavirus (MrNV) and extra small virus (XSV) have been identified as the causative agents for white tail disease (WTD) of M. rosenbergii. In this study, the gene sequences encoding MrNV and XSV capsid proteins were separately ligated into the pGEX-4T-3 expression vector and transformed into Escherichia coli. After induction, glutathione-S-transferase (GST)-tagged MrNV and XSV fusion proteins were obtained with molecular masses of 68 and 43 kDa, respectively. Specific polyclonal antibodies for MrNV and XSV against viral recombinant proteins and infected prawn tissues were verified using Western blotting. According to immunodot blot assay results, the detection sensitivities of antibodies were approximately 5 ng μL(-1) for both recombinant proteins GST-MrNV and GST-XSV. In additional, MrNV and XSV were detected at dilution levels of 1:2560 and 1:640 in the infected prawn tissues, respectively. No cross-reactions with white spot syndrome virus or grouper nervous necrosis virus were observed using immunodot blot assays. MrNV and XSV in infected muscle tissues were detected using immunohistochemistry. Although the detection limit of the immunodot blot assay was lower than that of nested reverse transcription polymerase chain reaction, these polyclonal antibodies can be useful for confirming MrNV and XSV infections in field tests.

Abstract:Whitish muscle diseases of freshwater giant prawn, Macrobrachium rosenbergii, is a new epidemic disease in the larvae and post-larvae of M. Rosenbergii in recent years. The clinical sign of the disease is a whitish or opacity appearance of the muscles in abdomen. Mortalities may reach more than 60%, sometimes even reach 100%. The pathogen of the disease is confirmed as Macrobrachium rosenbergii Nodavirus(MrNV), an icosahedral and non-enveloped virus sized from 25-26 nm with two linear ssRNA sized 3.0 kb and 1.3 kb respectively. MrNV was the first nodavirus founded in crustacean. To develop a rapid method for MrNV detection, monoclonal antibody against MrNV was prepared and used for virus detection. Briefly, mouse myeloma cells(SP2/0) were fused with spleen cells from BALB/c immunized with purified MrNV from diseased postlarvae(PL). Positive colonies were screened by indirect ELISA with the plates coated with purified MrNV and cloned 2-3 times with limited dilution method. Twelve positive hybridoma cell lines secreting monoclonal antibodies (Mabs) against MrNV were at last obtained and used for ascites preparation. The titres of positive ascites Mabs were ranged from 1∶10~5 to 1∶10~6 by indirect ELISA. These twelve Mabs were isotyped by Southern Biotechnology clonotyping system, and the results showed that six Mabs were IgG1, four IgG2a and two IgG2b, respectively. No cross reactions were found when the twelve Mabs were used for indirect ELISA with white spot syndrome virus(WSSV) and Taura syndrome virus(TSV). A triple antibody sandwich enzyme-linked immunosorbent assay(TAS-ELISA) was developed with the plates pre-coated with rabbit polyclonal antibodies against MrNV for virus capturing, followed by MrNV and samples incubation, Mabs and goat-anti-mouse-IgG antibodies labeled with HRP. Mab 2B5, characterized as Mab against 43 kD coat protein of MrNV by Western blot, was selected for TAS-ELISA. The results showed that TAS-ELISA can be used successfully for minimum virus detection with the sensitivity of 0.98 ng MrNV, or with the viral titer of 1∶40960 when typical diseased PL homogenization supernatants were used for TAS-ELISA. TAS-ELISA could be considered as a useful technique for MrNV detection as well as for epidemiological studies for its rapid and inexpensive cost with high specificity and sensitivity.
The crisis in Asian shrimp aquaculture: current status and future needs

Abstract:White‐spot disease is a relatively recent but widespread epizootic of cultured shrimp in Asia. This epizootic probably began in China in 1993 and subsequently spread from there to Japan, Taiwan, and the rest of Asia as far as India. Pond‐side losses in 1996 in Thailand alone reached approximately 70000 metric tonnes, valued at over half a billion US dollars. For all of Asia, the loss must amount to several billion dollars. The Asian science community reacted quickly to this disease outbreak. Within one year, the causative agent was identified as a bacilli‐form virus and within 3 years diagnostic DNA probes had been developed by several groups working in Japan, Taiwan, and Thailand. The virus has been included in the nonclassified virus group and is now generally referred to as white‐spot syndrome virus (WSSV). Previous experience with another serious epizootic caused by yellow‐head virus (YHV) helped to prime the science community for the advent of WSSV. Using the DNA diagnostic probes developed, many sources of WSSV were identified. The list now includes over 40 reservoir crustaceans and perhaps even aquatic insects. All of the commonly cultivated shrimp in Asia can be infected with the virus and for many, the infection can be deadly. In addition to noncultivated carriers, the postlarvae used to stock rearing ponds have also been found to be potential sources of rearing pond contaniination. Knowledge of the virus and its host species has brought about revolutionary changes in hatchery and farming practice, in order to implement preventive measures. These include the use of polymerase chain reaction (PCR) technology to screen broodstock and larvae, and the widespread adoption of closed or semiclosed cultivation. It is believed that without widespread application of these and other preventive measures, the losses in Thailand in 1996 would have been much higher. The WSSV epizootic has increased the awareness of the shrimp farming industry to the urgent need for further research and development work, if the shrimp industry is to become sustainable. Prime needs include domestication of broodstock leading to breeding programs; a better understanding of shrimp defences against disease; a better understanding of shrimp nutrition; and a better understanding of the shrimp pond environment so that growth can be optimized and negative impacts on the environment eliminated.

Abstract:The Penaeus vannamei nodavirus (PvNV), which causes muscle necrosis in Penaeus vannamei from Belize, was identified in 2005. Infected shrimp show clinical signs of white, opaque lesions in the tail muscle. Under transmission electron microscopy, the infected cells exhibit increases in various organelles, including mitochondria, Golgi stacks, and rough endoplasmic reticulum. Cytoplasmic inclusions containing para-crystalline arrays of virions were visualized. The viral particle is spherical in shape and 19 to 27 nm in diameter. A cDNA library was constructed from total RNA extracted from infected shrimp. Through nucleotide sequencing from the cDNA clones and northern blot hybridization, the PvNV genome was shown to consist of 2 segments: RNA1 (3111 bp) and RNA2 (1183 bp). RNA1 contains 2 overlapped open reading frames (ORF A and B), which may encode a RNA-dependent RNA polymerase (RdRp) and a B2 protein, respectively. RNA2 contains a single ORF that may encode the viral capsid protein. Sequence analyses showed the presence of 4 RdRp characteristic motifs and 2 conserved domains (RNA-binding B2 protein and viral coat protein) in the PvNV genome. Phylogenetic analysis based on the translated amino acid sequence of the RdRp reveals that PvNV is a member of the genus Alphanodavirus and closely related to Macrobrachium rosenbergii nodavirus (MrNV). In a study investigating potential PvNV vectors, we monitored the presence of PvNV by RT-PCR in seabird feces and various aquatic organisms collected around a shrimp farm in Belize. PvNV was detected in mosquitofish, seabird feces, barnacles, and zooplankton, suggesting that PvNV can be spread via these carriers.

Abstract:The white spot syndrome virus (WSSV) is one of the most serious diseases in the shrimp and crab farm. This virus disseminates very quickly and causes high mortality in culture pond, accompanied by great economic losses. Recently, high mortality of WSSV-infected mud crabs collected from M.G.R. Thittu, Cuddalore district, Tamil Nadu, south east coast of India during 2015 and 2016. A total of 283 samples collected from pens fattening ponds were tested for the presence of WSSV. Of these, a prevalence of 32.5% WSSV-infected mud crabs showed. Hence, the present study was carried to understand the reason for the mortality. Our examinations of crab mortality confirmed the possible infestation of white spot syndrome virus. The classical symptoms of white spot virus infections like white spots in the inner carapace and histopathological changes on gills like eosinophilic inclusion bodies and basophilic Cowdry type “A” inclusion bodies have been observed. Also, our examinations confirmed the infestation through molecular diagnosis and TEM visualization of hypertrophied cells with massive viral particles in the nucleus and cytoplasm of infected gills. In the experimental work, the fastest mortality was observed in the ingestion then in the waterborne mode of infection. The waterborne mode caused the 40% mortality on 90th days. The ingestion mode caused the 100% mortality on 90th days.

Abstract:Litopenaeus vannamei is the most widely cultured shrimp species in the world and has been impacted by the emergence of various diseases over the course of its history. Infectious myonecrosis virus (IMNV), Taura syndrome virus (TSV), and white spot syndrome virus (WSSV) are three primary pathogens having caused financial losses for shrimp farmers. The purpose of this post hoc study was to evaluate the effectiveness of laboratory viral challenges for resistance to IMNV, TSV, and WSSV for a commercial breeding population of L. vannamei. Phenotypic records for viral challenges from 20 batches, including 442 full-sib families over approximately 2.3 generations were included in the study. Independent challenges for each virus were conducted with late-stage post larvae (PLs) inoculated with viral suspensions. Binary survival data was analyzed with both logistic regression and linear animal models. Heritability estimates were low for the logistic regression models on both the underlying liability scale (0.15 to 0.17) and the observed scale (0.08 to 0.11) for resistance to the three viruses and low (0.23) to moderate (0.39) for the linear animal models for TSV and IMNV, respectively, indicating resistance as defined during challenges is under some level of additive genetic control. Heritability estimates for the linear animal model were not obtained for WSSV resistance due to non-convergence of the model. Estimated breeding values (EBVs) from the logistic regression models were regressed against time, showing significant positive genetic trends, equating to 18.7, 3.0, and 27.4% average genetic gains per generation for IMNV, TSV, and WSSV resistance, respectively. Genetic correlations between viral-resistance traits were weak (0.135 to 0.204), but positive and significant (p < 0.01).
New genotypes of white spot syndrome virus (WSSV) and Taura syndrome virus (TSV) from the Kingdom of Saudi Arabia

Abstract:White spot syndrome virus (WSSV) and Taura syndrome virus (TSV) are highly pathogenic to penaeid shrimp and have caused significant economic losses in the shrimp culture industry around the world. During 2010 and 2011, both WSSV and TSV were found in Saudi Arabia, where they caused severe mortalities in cultured Indian white shrimp Penaeus indicus. Most outbreaks of shrimp viruses in production facilities can be traced to the importation of infected stocks or commodity shrimp. In an attempt to determine the origins of these viral outbreaks in Saudi Arabia, we performed variable number of tandem repeat (VNTR) analyses for WSSV isolates and a phylogenetic analysis for TSV isolates. From the WSSV genome, the VNTR in open reading frames (ORFs) 125 and 94 were investigated with PCR followed by DNA sequence analysis. The genotypes were categorized as {N125, N94} where N is the number of repeat units in a specific ORF, and the subscript indicates the ORF (i.e. ORFs 125 and 94 in this case). From 15 Saudi Arabia WSSV isolates, we detected 3 genotypes: {6125, 794}, {7125, del94}, and {8125, 1394}. The WSSV genotype of {7125, del94} appears to be a new variant with a 1522 bp deletion encompassing complete coding regions of ORF 94 and ORF 95 and the first 82 bp of ORF 93. For TSV genotyping, we used a phylogenetic analysis based on the amino acid sequence of TSV capsid protein 2 (CP2). We analyzed 8 Saudi Arabian isolates in addition to 36 isolates from other areas: SE Asia, Mexico, Venezuela and Belize. The Saudi Arabian TSV clustered into a new, distinct group. Based on these genotyping analyses, new WSSV and TSV genotypes were found in Saudi Arabia. The data suggest that they have come from wild shrimp Penaeus indicus from the Red Sea that are used for broodstock.
Oral Vaccination Trials with Crayfish, Procambarus Clarkii, to Induce Resistance to the White Spot Syndrome Virus

Abstract:The potential of oral vaccination against white spot syndrome virus (WSSV) in crayfish Procambarus clarkii was investigated. The protective effect of binary ethylenimine (BEI)-inactivated WSSV was tested by oral vaccination, followed by an oral challenge with WSSV. The crayfish fed with feed pellets coated with BEI-inactivated WSSV showed a resistance to WSSV on the seventh day post vaccination (dpv). The relative percentage survival values were 60%, 70% and 75% for the vaccinated once, twice and thrice with inactivated WSSV. Following an intramuscular injection experiment, no mortality was recorded in the inactivated WSSV group and the negative control at 17 days post challenge. The cumulative mortalities in the heated WSSV group and WSSV group were 100%. Shrimp that survived the WSSV challenge on the seventh day after cessation of oral vaccination were positive for the presence of WSSV by a polymerase chain reaction assay specific for WSSV. This result indicated that inactivated WSSV could protect crayfish against WSSV by oral delivery.
Determination of the Infectious Agent of Translucent Post-Larva Disease (TPD) in Penaeus vannamei

Abstract:A new emerging disease called “translucent post-larvae disease” (TPD) or “glass post-larvae disease” (GPD) of Penaeus vannamei, characterized by pale or colorless hepatopancreas and digestive tract, has become an urgent threat to the shrimp farming industry. Following this clue that treatment of an antibacterial agent could alleviate the disease, systematic investigation of the potential infectious agent of TPD was conducted using bacterial identification and artificial challenge tests to fulfill Koch’s postulates. A dominant bacterial isolate, Vp-JS20200428004-2, from the moribund individuals was isolated and identified as Vibrio parahaemolyticus based on multi-locus sequence analysis. However, Vp-JS20200428004-2 differed from the V. parahaemolyticus that caused typical acute hepatopancreatic necrosis disease. Immersion challenge tests revealed that Vp-JS20200428004-2 could cause 100% mortality within 40 h at a dose of 1.83 × 106 CFU/mL, and experimental infected shrimp showed similar clinical signs of TPD. The Vp-JS20200428004-2 could be re-isolated and identified from the experimental infected individuals. Moreover, histopathological analysis of diseased samples indicated that Vp-JS20200428004-2 caused severe necrosis and sloughing of epithelial cells of the hepatopancreas and midgut in shrimp individuals both naturally and experimentally infected. Our present results indicated that Vp-JS20200428004-2 is a highly virulent infectious agent associated with the TPD and deserves further attention.

Abstract:Experimental infection of crayfish,Procambarus clarkii,were conducted by feeding white spot syndrome virus(WSSV) infected shrimp,Fenneropenaeus chinensis to determine the susceptibility of crayfish to WSSV.The results showed that a cumulative mortality of 90% was observed in the infected crayfish,while no death in control group.Crayfish that dead in oral infection was positive for the presence of WSSV by PCR.PCR detection results also showed that six tissues including hepatopancreas,mid-intestine,muscle,gills,gonad and heart of dying crayfish were positive to WSSV,while those were negative in control group.Optical microscope observation also proved that hepatopancreas,mid-intestine,muscle,gills,gonad,heart and hemolymph of dying crayfish showed pathological variation to different degree.
Differential expression of microRNAs in giant freshwater prawn (Macrobrachium rosenbergii) during the infection of Vibrio parahaemolyticus

Abstract:MicroRNAs (miRNAs) are a category of small non-coding RNAs regarded as vital regulatory factors in various biological processes, especially immune regulation. The differently expressed miRNAs in Macrobrachium rosenbergii after the challenge of Vibrio parahaemolyticus were identified using high-throughput sequencing. A total of 18 known as well as 12 novel miRNAs were markedly differently expressed during the bacterial infection. The results of the target gene prediction and enrichment analysis indicated that a total of 230 target genes involved in a large variety of signaling pathways and biological processes were mediated by the miRNAs identified in the current research. Additionally, the effects of novel-miR-56, a representative differentially expressed miRNA identified in the previous infection experiment, on the immune-related gene expression in M. rosenbergii were explored. The expression of the immune-related genes including Spätzle1(Spz1), Spz4, Toll-like receptor 1 (TLR1), TLR2, TLR3, immune deficiency (IMD), myeloid differentiation factor 88 (MyD88), anti-lipopolysaccharide factor 1 (ALF1), crustin1, as well as prophenoloxidase (proPO) was significantly repressed in the novel-miR-56-overexpressed prawns. The expression of these genes tested in the novel-miR-56-overexpressed M. rosenbergii was still signally lower than the control in the subsequent V. parahaemolyticus challenge, despite the gene expression in each treatment increased significantly after the infection. Additionally, the cumulative mortality of the agomiR-56-treated prawns was significantly higher than the other treatments post the bacterial challenge. These results suggested that novel-miR-56 might function as a negative regulator of the immune-related gene expression of M. rosenbergii in the innate immune defense against V. parahaemolyticus.
Investigation on Natural Infection of Covert Mortality Nodavirus in Large Yellow Croaker, Larimichthys crocea

Abstract:Covert mortality nodavirus (CMNV) recently became more prevalent, severely affecting farmed shrimps, and also other invertebrates and teleost fish, in the coastal areas of China. For better understanding of its prevalence and susceptibility of large yellow croaker Larimichthys crocea to CMNV, an epidemiological survey was carried out in the main producing areas (Ningbo, Zhejiang, China, and Ningde, Fujian, China) and its offshore feeding grounds in the Southern Yellow Sea. The collected cultured L. crocea individuals showed clinical symptoms such as anorexia and abnormal swimming and diagnosed for CMNV infection by using RT-nPCR assay. The positive rates for CMNV in the collected L. crocea samples from Ningbo, Ningde, and the Yellow Sea were 14.29% (4/28), 20.00% (7/35), and 16.67% (1/6), respectively. Furthermore, CMNV diagnosis using TaqMan probe-based reverse transcription quantitative PCR (TaqMan RT-qPCR) yielded positive rates of 25.00% (7/28), 22.86% (8/35), and 16.67% (1/6) respectively in the above-mentioned samples, and with a range of 12.73–3,108.33 of CMNV copy numbers/μg total RNA. Phylogenetic tree analysis based on the RNA-dependent RNA polymerase (RdRp) and capsid protein genes showed that CMNV isolates from L. crocea samples were clustered tightly with the original isolate of CMNV from Penaeus vannamei . The histopathological studies indicated that severe cellular necrosis occurred in the heart, liver, spleen, eye, and gill of naturally infected fish. Stronger positive signals for CMNV-specific probes appeared in the necrotic cells and tissues in the in situ hybridization (ISH) analysis. Transmission electron microscopy (TEM) revealed the presence of numerous 30 nm diameter CMNV-like viral particles. The purified CMNV suspension could cause a significant cytopathic effect (CPE) in epithelioma papilloma cyprini (EPC) cells and resulted in the formation of typical inclusion bodies, and also visible CMNV particles around the lysed EPC cells under the TEM. Taken together, all results of this study demonstrated that L. crocea is susceptible to CMNV. The prevalence of CMNV and its potential impacts on the wild population of L. crocea see the need for further investigations.

Insights on the virulence and genomic features of Lactococcus garvieae isolated from giant freshwater prawn Macrobrachium rosenbergii (de Man 1879)

Abstract:Giant freshwater prawn (Macrobrachium rosenbergii (MR)) is a significant aquafarm species commercially cultured in Taiwan. Intensive farming practices have led to the outbreak of Lactococcus garvieae (LG), which causes Lactococcosis in MR. Recently, LG has re-emerged and the number of mortalities in prawn farms has increased in Taiwan. However, there is no preventative strategy described and a lack of knowledge on virulence factors and pathogenesis from LG in MR. The most virulent strain of L. garvieae from M. rosenbergii was screened in vivo among seven isolates selected for infectivity testing injecting 0.1 mL of 108 CFU/mL bacterial concentration. Among the seven isolates screened, L. garvieae 109-6 resulted in 100% mortality within 3 days post-infection. Furthermore, 109-6 L. garvieae LD50 dosage from in MR was found to be 106 CFU/mL. Subsequently, the most virulent strain 109-6 was sequenced using MinIon Nanopore sequencing. Results indicated that the LG genome yielded a protein-coding of 3857 with 59 tRNA and 16 rRNA and no plasmid. Interestingly, the distribution of subsystems in the annotated genome revealed genes related to virulence, defence, and disease among LG 50 genes. Altogether, the virulent strain and its genome data revealed distinctive features of LG, which hinted toward its pathogenicity and could facilitate for better preventive strategies.

Abstract:This review aims to provide an update on the current scientific understanding of various aspects of White Spot Syndrome Virus (WSSV) formation, diagnostic procedures, transmission, ecological effects, pathophysiology and management strategies. In terms of production and financial benefits, the WSSV has been the most virulent in shrimp and several other crustacean sectors around the globe. It spreads vertically from diseased broodstock to post-larvae and horizontally by cannibalism, invertebrate vectors, freshwater and sediments. In the transfer of white spot disease (WSD) in newly stocked ponds, the survivability of WSSV in sediment is the most important variable. In typical cultural conditions, it is a highly infectious pathogen capable of inflicting total death within 3-10 days after an outbreak. Some of the current biosecurity strategies used to keep diseases out of shrimp ponds such as pond water disinfection, quarantine of new stocks before stocking and broader usage of specific pathogen-free shrimp. The sequencing and characterisation of various WSSV strains have provided details about pathogen biology, pathogenicity and disease. To develop successful control methods, knowledge of these characteristics is essential. In several shrimp-producing countries in Asia and the Americas, the infections produced by the WSSV have had disastrous socio-economic consequences. As a result of international trade or migration of diseased species, the World Animal Health Organization recognised several illnesses as posing a substantial hazard to farmed shrimp. WSD is receiving much scientific research due to the potential economic effects of the virus. Research is now being done to understand better the molecular biology and pathophysiology of WSSV, as well as how to treat and prevent the virus. However, further study should be conducted in countries with more resilient host species to understand their role in mitigating disease impacts since these revelations may aid in developing a WSD treatment.

Abstract:A nucleic acid sequence-based amplification(NASBA)assay was established for the detection of Macrobrachium rosenbergii Nodavirus(MrNV).The specific primers were designed according to the high conserved region of RNA2 sequence of MrNV.The 224 bp specific amplification product was obtained in positive sample determined with 3%agarose gel electrophoresis,while no product was generated from shrimp infected with other viruses including DNA viruses(IHHNV,WSSV)and RNA viruses(TSV,IMNV,YHV).The detecting limit of the assay was 8pg nucleic acid,which is more sensitive than that of PCR method.
Isolation and Identification of a New Strain of Nervous Necrosis Virus from the Big-belly Seahorse Hippocampus Abdominalis in China

Abstract:Abstract Background: Betanodaviruses , members of the Nodaviridae family, are the causative agents of viral nervous necrosis (VNN) in fish, resulting in great economic losses worldwide. Methods: In this study, we isolated a virus strain named seahorse nervous necrosis virus (SHNNV) from cultured big-belly seahorses Hippocampus abdominalis in Xiamen city, Fujian Province, China. Cell isolation, PCR detection, phylogenetic analysis, qRT-PCR, fluorescence in situ hybridization (FISH) and histology were used for virus identification and analysis of virus histopathology. Furthermore, an artificial infection experiment was conducted for virulence testing. Results: Brain and eye tissue homogenates of diseased big-belly seahorses were inoculated onto a grouper spleen (GS) cell monolayer at 28°C. Tissue homogenates induced obvious cytopathic effects in GS cells. PCR and sequencing analyses revealed that the virus belonged to Betanodavirus and shared high sequence identity with red-spotted grouper nervous necrosis virus (RGNNV) isolates. qRT-PCR and fluorescence in situ hybridization revealed that SHNNV mainly attacked the brain and eye. Histopathological examination revealed that the virus led to cytoplasmic vacuolation in the brain and retinal tissues. Infection experiments confirmed that SHNNV was highly infectious, causing massive death in big-belly seahorses. Conclusion: A novel seahorse betanodavirus from the big-belly seahorse in China was discovered. This finding will contribute to the development of efficient strategies for disease management in aquaculture.
Virulence factor genes and comparative pathogenicity study of tdh, trh and tlh positive Vibrio parahaemolyticus strains isolated from Whiteleg shrimp, Litopenaeus vannamei (Boone, 1931) in India

Abstract:Vibrio parahaemolyticus is a gram-negative halophilic bacterium responsible for gastrointestinal infection in human and vibriosis in aquatic animals. The thermostable direct hemolysin (tdh), tdh-related hemolysin (trh) and thermolabile hemolysin (tlh) positive strains of V. parahaemolyticus were identified from brackishwater aquaculture farms of West Bengal and Andhra Pradesh, India. Moreover, the presence of other virulent genes like vcrD1, vopD, vp1680 under type three secretion system 1 (T3SS1) and vcrD2 vopD2, vopB2, vopC2 under type three secretion system 2 (T3SS2) were detected in tdh positive strain of V. parahaemolyticus. Furthermore, the study revealed that the tdh and trh positive isolates were resistant to β-lactam antibiotics and were able to lyse more than 95% of human Red Blood Cells (RBCs). In addition, both the isolates showed high cytotoxicity in Human Embryonic Kidney (HEK) cell line compared to tlh positive strain. Additionally, intraperitoneal and oral administration of tdh and trh positive strain of V. parahaemolyticus in Indian Major Carp, Labeo rohita caused 100% mortality at the level of 2.0 × 108 CFU ml-1 and 1.6 × 108 CFU ml-1, respectively. In contrast, only 10% mortality was observed in the case of tlh positive strain at the level of 2.5× 108 CFU ml-1. The histopathological changes like infiltration of blood cells and degenerated hepatic tissue in the liver of L. rohita were observed after the experimental challenge. The changes like degeneration of glomeruli, necrosis of renal tubules and Bowman's capsule were observed in the kidney section. Ragged, irregular shaped villi and necrosis of the villus were observed in the intestinal lumen. Overall, the study demonstrates that isolated V. parahaemolyticus is a potent aquatic microbial pathogen. Additionally, as V. parahaemolyticus is also a human pathogen and might pose a threat to the human population, proper management strategies are required to prevent the possible occurrence of disease.
