Abstract:Now, Jason W. Chin and colleagues report tRNA display as a ribosome-independent approach to evolve orthogonal synthetases for tRNA acylation with non-canonical monomers in Escherichia coli cells. Isolated orthogonal tRNAs that were successfully acylated in vivo by the orthogonal synthetase were labelled with a biotin moiety for pulldown, whereas non-acylated orthogonal tRNAs were oxidized and excluded. Moreover, a gene fusion of the synthetase and a split tRNA was designed to provide a connection between genotype and acylation. Using tRNA display and mutant libraries of Methanosarcina mazei pyrrolysyl-tRNA synthetase, the researchers identified enzymes that can specifically acylate orthogonal tRNAs with monomers 1–9 (pictured) including β-amino acids, an α,α-disubstituted amino acid and a β-hydroxy acid. Subsequently, the evolved enzymes were employed to ribosomally incorporate the monomers 2–9 into green fluorescent protein (GFP) in vivo using the stop-codon suppression method. Indeed, GFPs with β-amino acids 2 , 4 , 7 and α,α-disubstituted amino acid 8 were expressed and several milligrams of the proteins isolated. This quantity was sufficient to successfully crystallize GFP with β-amino acid 2 . However, the researchers did not observe protein production with 3 , 5 , 6 or 9 indicating that these monomers are not well tolerated by the ribosomal translation machinery. As such, the acceptance of unusual monomers by the ribosome is still an ongoing challenge in synthetic biology. To address this issue, the evolved orthogonal aminoacyl-tRNA synthetase/tRNA pairs described in this work provide a useful starting point to engineer the ribosome for wider monomer compatibility.
