Abstract:Objective: To investigate the behavioral development in adolescent rats of perinatal hypothyroidism and its relation to androgen receptor(AR) gene expression in the hippocampus.Methods: Perinatal hypothyroidism was induced by gavages 50 mg/d of propylthiouracil solution in 48 dams starting at embryonic day 15 through the lactation period.Twenty-four pups(M∶F=1) of perinatal hypothyroidism were injected intraperitoneally with 2 μg T4/100 g BW daily from the day of birth to the age of 21 days(treatment group);24 pups(M∶F=1) without treatment were designated as hypothyroidism group.And 24 normal pups(M∶F=1) served as the control group.The effects of perinatal hypothyroidism on the abilities to learn and retain memory traces and on behavior were observed in rats of both sexes at 60 days.Experiments were performed using models of conditioned open field test and passive avoidance reflexes.Hippocampus samples were collected and AR mRNA was detected by competitive RT-PCR.Results: Perinatal hypothyroidism caused an increase of crossing number and decrease of rearing and defecation in both sexes.In treatment groups,only the crossing number in male didn't reach the normal level(P0.05).In passive avoidance test,hypothyroidism groups showed more mistakes in both sexes and shorter latencies in males,the females performed better than males(P0.01).The treatment groups performed significantly better than the age-matched hypothyroidism groups and reached the normal level(P0.05).AR mRNA levels in hippocampus of hypothyroid group were lower than those of the controls in males,and the levels in treatment groups were significantly higher in comparison with the hypothyroidism groups(P0.01).There were no significant differences among the three female groups(P0.05).In male group,there was negative correlation between the number of crossing and AR mRNA in the hippocampus(r=-0.537,P=0.001),negative correlation between the number of mistake and AR mRNA(r=-0.532,P=0.001),and positive correlation between the latency and AR mRNA(r=0.564,P=0.000).Conclusions: Perinatal hypothyroidism results in hyperactivity and anti-anxiety effects on adolescent rats,the sex difference is depleted,and also causes learning and memory impairment but the degree of influence higher in male than female.The decreased level of AR mRNA expression in hippocampus contributes to the change of behavioral ability in adolescent male.

Abstract:Oligodendrocyte development and myelination are under thyroid hormone control. In this study we analysed the effects of chronic manipulation of thyroid status on the expression of a wide spectrum of oligodendrocyte precursor cells (OPCs) markers and myelin basic protein (MBP) in the subventricular zone (SVZ), olfactory bulb and optic nerve, and on neural stem cell (NSC) lineage in adult rats. Hypo- and hyperthyroidism were induced in male rats, by propyl-thio-uracil (PTU) and L-thyroxin (T4) treatment, respectively. Hypothyroidism increased and hyperthyroidism downregulated proliferation in the SVZ and olfactory bulb (Ki67 immunohistochemistry and Western blotting, bromodeoxyuridine uptake). Platelet-derived growth factor receptor alpha (PDGFalpha-R) and MBP mRNA levels decreased in the optic nerve of hypothyroid rats; the same also occurred at the level of MBP protein. Hyperthyroidism slightly upregulates selected markers such as NG2 in the olfactory bulb. The lineage of cells derived from primary cultures of NSC prepared from the forebrain of adult hypo- and hyperthyroid also differs from those derived from control animals. Although no difference of in vitro proliferation of NSCs was observed in the presence of epidermal growth factor, maturation of oligodendrocytes (defined by process number and length) was enhanced in hyperthyroidism, suggesting a more mature state than in control animals. This difference was even greater when compared with the hypothyroid group, the morphology of which suggested a delay in differentiation. These results indicate that thyroid hormone affects NSC and OPC proliferation and maturation also in adulthood.

Effects of Hypothyroidism on Apoptosis and the Expression of Bcl-2 and Bax Gene in the Neonatal Rat Hippocampus Neurons

Abstract:OBJECTIVE:During the critical period of brain development, insufficiency of thyroid hormone results in severe mental retardation and learning deficit. This study was designed to investigate the effects of hypothyroidism on apoptosis and the expression of Bcl-2 and Bax gene in the developing rat hippocampus neurons and to explore the mechanism of brain development regulated by thyroid hormone.METHOD:Hypothyroidism was induced by administration of propylthiouracil (PTU, 50 mg/d) solution to the dams from gestational day 15 by gavage. Pups from both hypothyroid and control groups were harvested at postnatal day 1 (P1), P5, P10 and P15, respectively. Blood samples were collected at the time of death for the determination of thyroid hormone. Serum free tri-iodothyronine (FT(3)) and free thyroxine (FT(4)) were measured by using chemoluminescence. Hippocampus collected from the control and hypothyroid pups were examined under light and transmissional electron microscopy. Measurement of DNA fragmentation was carried out by agarose gel electrophoresis. The expression of Bcl-2 and Bax protein in the developing rat hippocampus neurons was performed by Western blotting.RESULTS:Significantly lower circulating FT(4) and FT(3) levels confirmed the hypothyroid status of the experimental pups. The shrunken and contracted degenerations increased in hippocampus neurons of hypothyroid pups under light microscopy. Enhanced apoptotic cells were found in hippocampus neurons of hypothyroid pups under transmission electron microscopy, especially at P10 and P15. Extensive DNA fragmentation was seen throughout development in hippocampus of hypothyroid pups, but not in the euthyroid controls except for basal level at P10. The expression of Bcl-2 in the hippocampus neurons of hypothyroid pups was significantly lower than that of euthyroid controls at all stages of development (P1: 1.95 +/- 0.27 vs. 2.59 +/- 0.19, P < 0.05, P5: 1.86 +/- 0.24 vs. 2.47 +/- 0.17, P < 0.05, P10: 1.29 +/- 0.22 vs. 1.86 +/- 0.28, P < 0.05 and P15: 1.21 +/- 0.27 vs. 2.18 +/- 0.17, P < 0.01, respectively). The relative amount of expression varied significantly with age in the control pups. The level of Bcl-2 was high in hippocampus neurons of euthyroid at P1, P5, and decreased significantly at P10, and showed a trend of recovery at P15. Similar age-related variation in the expression of Bcl-2 gene was observed in the hypothyroid group at P1, P5 and P10, but the level was maintained low at P15. The expression of Bax in the hippocampus neurons of hypothyroid pups was significantly higher than that of control pups at all stages of development (P1: 1.69 +/- 0.14 vs. 1.24 +/- 0.23, P < 0.05, P5: 1.78 +/- 0.16 vs. 1.29 +/- 0.17, P < 0.05, P10: 1.92 +/- 0.18 vs. 1.45 +/- 0.14, P < 0.05 and P15: 1.86 +/- 0.14 vs. 1.51 +/- 0.12, P < 0.05, respectively). The ratio of Bcl-2/Bax in hippocampus neurons of hypothyroid pups was lower than that of age-matched controls (P1: 1.16 +/- 0.17 vs. 2.12 +/- 0.35, P < 0.05, P5: 1.05 +/- 0.16 vs. 1.94 +/- 0.36, P < 0.05, P10: 0.68 +/- 0.17 vs. 1.29 +/- 0.16, P < 0.05 and P15: 0.67 +/- 0.19 vs. 1.45 +/- 0.22, P < 0.01, respectively).CONCLUSION:Thyroid hormone significantly prevents apoptosis of hippocampus neurons. Congenital hypothyroidism increases not only the extent but also the duration of apoptosis by down-regulation of the anti-apoptotic gene Bcl-2 and maintaining a high level of the pro-apoptotic gene Bax.
Alteration of Myocardial Sarcoplasmic Reticulum Ca2+ Transport Protein Expression in Neonatal Hypothyroid Rats

Abstract:AIM:To investigate the alteration of sarcoplasmic reticulum(SR)Ca~(2+)transport proteins including sarcoplasmic reticulum Ca~(2+)-ATPase 2a(SERCA2a)and phospholamban(PLB)mRNA expression as well as the alteration of myocardial SR Ca~(2+)-ATPase activity in neonatal hypothyroid rats,and to explore the effect of levothyroxine(L -T_4)substitution therapy on the above indexes.METHODS:Hypothyroidism was induced by the administration of propy-lthiouracil (PTU,50 mg/d)to the pregnant SD rats by gavage beginning on embryonic day 15 and continuing throughout the lactational period.A subgroup of neonatal hypothyroid rats were intraperitoneally injected with L-T_4(20μg/kg BW daily),starting from the day of birth.Other pregnant SD rats received normal saline instead of PTU.The samples of the rats in all 3 groups were harvested at postnatal day 3,5 and 7(n=10).After measurement of serum thyroid hormone levels, the hearts were removed and the ventricles were weighed(HW).The concentration of calcium in ventricular myocardium (ventricular myoCa~(2+))was detected by fluorospectrophotometry and the activity of SR Ca~(2+)-ATPase was determined by the inorganic phosphorus method.The mRNA expression of SERCA2a and PLB was also detected by real-time PCR. RESULTS:Neonatal hypothyroid rats had a significant lower level of SERCA2a mRNA(P0.05)and a higher level of PLB mRNA(P0.01),and subsequent lower SERCA2a/PLB at each postnatal day(P0.01)was observed.Compared with hypothyroid group,the mRNA expression of SERCA2a significantly increased(P0.05)and that of PLB significantly decreased(P0.05)in L-T_4 treatment group.The concentration of ventricular MyoCa~(2+)in hypothyroid group was significantly higher than that in control group(P0.01),and that in L-T_4 treatment group showed a significant decrease as compared with hypothyroid group(P0.05).The activity of sarcoplasmic reticulum Ca~(2+)-ATPase in hypothyroid group was significantly lower than that in control group(P0.01),and that in L-T_4 treatment group showed a significant increase as compared to hypothyroid group(P0.05).CONCLUSION:The deficiency of thyroid hormone,resulting in decreased expression of SERCA2a mRNA as well as increased PLB mRNA,contributes to the reduction of SR Ca~(2+)-ATPase activity in neonatal rats.This may be one of the most important mechanisms of myocardial systolic and diastolic dysfunctions.
Selective suppression of neurofilament antigen expression in the hypothyroid rat cerebral cortex

Abstract:As an integral component of the cytoskeleton neurofilaments play a central role in the establishment and maintenance of neuronal form. In particular, high neurofilament concentrations are characteristic of many classes of axons in the central nervous system. Isolated neurofilaments from rat brain consist of 3 distinct polypeptides with apparent molecular weights 210K, 160K and 68K. A murine monoclonal antibody, mabN210, has been produced which specifically recognizes an epitope associated with the high molecular weight subunit and this antibody has been used to explore the regulation of neurofilament expression during brain development. It has been shown that in the rat cerebellar cortex, the expression of mabN210-immunoreactivity in basket cell axons is severely suppressed in hypothyroidism while neurofilament antigen expression in other cerebellar axons seems not to require thyroid hormones. In view of the well-known cortical deficits in hypothyroidism, these studies have now been extended to include the developing rat cerebral cortex and selected cortical afferent and efferent axons. In hypothyroid rats there is a marked suppression of mabN210-immunoreactivity in the cerebral cortex and corpus callosum and, to a lesser extent, there is a reduction in staining in the internal capsule. By contrast, hypothyroidism did not reduce mabN210-immunoreactivity in the lateral olfactory tract or the stria medullaris. In rats, serum thyroid hormone starts to rise to adult levels on postnatal day 4. It appears that axons that have attained their mature distribution prior to the onset of thyroid hormone expression are not affected by hypothyroidism whereas mabN210-immunoreactivity is suppressed in those axonal tracts that reach a mature distribution after P4.

In the T(4) injected group hypothyroid rats were injected intraperitoneally with levothroxine (L-T(4)) 2 microg/100 g BW daily, starting from the day of birth. Cerebral cortex and hippocampus specimen were collected from controls,hypothyroid and T(4)-injected hypothyroid rats on postnatal d1, 5, 10, 15 and 20. Quantification of ARmRNA in cerebral cortex and hippocampus was performed with competitive RT-PCR using internal and external standardization.RESULT:Age-related increasing ARmRNA levels were observed in neonatal rats, and those in male animals were significantly higher. AR expression was higher in the hippocampus than in the cerebral cortex. ARmRNA levels in the hypothyroid pups were lower than those in age-matched controls. The mRNA levels in the T(4)-injected hypothyroid pups were significantly higher compared with the age-matched hypothyroid pups, but in hippocampus ARmRNA expression did not reach normal levels in male rats at d10 and d20, in female at d15 and d20.CONCLUSION:The expression of ARmRNA decreases in brain of rats with perinatal hypothyroidism. Treatment with thyroid hormone can recover ARmRNA expression in cerebral cortex, but not in hippocampus.
Effect of thyroid hormone deficiency on developmental expression of goalpha gene in the brain of neonatal rats by competitive RT-PCR and in situ hybridization histochemistry

Abstract:Goalpha is a guanine nucloetide-binding regulatory protein alpha subunit which is mainly distributed in the central nervous system, but it has not previously been reported how it is regulated by thyroid hormone in the brain of neonatal rat at transcriptional levels. In this report, we used quantitative competitive reverse transcriptional PCR to quantify the effects of TH deficiency on Goalpha gene expression in the brain of neonatal rat at mRNA levels. It was found that Goalpha mRNA levels in the brain of 14-day-old rats significantly increased over 3-fold after induction of perinatal hypothyroidism, and declined markedly after treatment of thyroxine replacement. In situ hybridization histochemistry was further employed to observe the time-course and spatial expression of Goalpha gene in the brain of neonatal rats affected by thyroid hormone deficiency during the developmental period. The data showed that perinatal hypothyroidism can enhance Goalpha mRNA levels in the temporal cortex, sensorimotor cortex, piriform cortex, amygdala, hippocampal CA1-4 subfields, dentate gyrus, arcuate nucleus (AR) and ventromedial hypothalamic nucleus (VMH) of hypothalamus, but not in the striate cortex, cingulate cortex, claustrum, caudate/putamen and thalamus in the brain of rat at 7-21 days post-partum. The results suggest that up-regulation of Goalpha gene expression may be one kind of common mechanism responsible for neurological deficits in some brain areas arising from thyroid hormone deficiency in the critical periods of neonatal rats.
Protracted expression of serotonin transporter and altered thalamocortical projections in the barrelfield of hypothyroid rats

Abstract:In humans, thyroid hormone deficiency during development causes severe neurological diseases but the underlying mechanisms are unclear. We have examined the effects of thyroid hormones on the development of somatosensory thalamocortical projections, by inducing hypothyroidism in rats by methimazole treatment at embryonic day 13 and subsequent thyroidectomy at postnatal day 6 (P6). Initial development of the thalamocortical projections and their tangential and laminar patterning were similar in normal and hypothyroid rats from birth to P4. The tangential spread of the thalamocortical arbors is reduced in hypothyroid rats after P4, paralleling the overall cortical atrophy. Anterograde tracing and single axon reconstructions indicate that thalamic afferents reached layer IV but that they had fewer and shorter branches, with a 42% reduction in the number of boutons. The transient serotonin (5-HT) immunostaining and 5-HT transporter (5-HTT) expression were both prolonged by 5 days in hypothyroid rats. This does not reflect a delayed maturation of the thalamus because other transiently expressed genes such as the vesicular monoamine transporter and the 5-HT1B receptor are not modified. Protracted 5-HTT expression also occurred in other areas with transient expression, but no changes were observed in the raphe nuclei where the 5-HTT is expressed permanently. Thus, thyroid hormones appear to be important in regulating the extinction of the 5-HTT in nonserotoninergic neurons. The transient stabilization of 5-HT reuptake in hypothyroid rats could affect the growth of thalamic axons. Our data stress the importance of maternal and foetal thyroid hormones for the normal development of sensory systems.
Radiation-induced temporal lobe necrosis.

Abstract:Because nasopharyngeal carcinoma has a high frequency of intracranial spread, adequate radiation treatment inevitably results in irradiation of the temporal lobes. A 38-year-old woman who was treated for nasopharyn-geal carcinoma 30 months previously and had been given radiation therapy (70 Gy to the primary site) presented with headaches. Computed tomography (CT) showed an enhancing lesion in the right temporal lobe, with edema extending superiorly into the parietal lobe. Biopsy revealed brain necrosis and a course of corticosteroid therapy was started. Five months later, magnetic resonance (MR) showed less edema in the right temporal lobe but an extensive lesion on the contralateral side, not seen before (Fig 1A–C). Corticosteroid therapy was again given and 9 months later, the nasopharynx was reevaluated but no tumor recurrence was seen. MR showed dilatation of the temporal horns indicating cerebral atrophy (Fig 1D). Enhancing lesions in both temporal lobes were still evident, though less extensive than before. Doses below 60 Gy at conventional 2 Gy daily appear inadequate for tumor control (1). Unfortunately, the effective dose for nasopharyngeal carcinoma (65 to 70 Gy) exceeds the quoted tolerance limits for the adjacent neural structures (2). There is, therefore, a substantial risk of radiation damage to the brain. Temporal lobe necrosis (TLN) is the most dreaded complication of radiation therapy and accounts for 65% of treatment mortality. Lee et al (1) reported a 3% cumulative incidence of TLN in a series of 4527 patients. The latent interval ranged from 1.5 to 13 years (median, 5 years). TLN is probably underdiagnosed, because in Lee et al's study 39% of patients had only vague symptoms, whereas 16% had no symptoms (3). Although the radiation dose to the brain is approximately equal on both sides, changes in the brain are often asymmetric. Half the patients with TLN will present with unilateral abnormalities; in only 10% of these patients will bilateral lesions subsequently develop (3). The earliest sign of TLN is cerebral edema, which can be extensive. Enhancing lesions can be located in the gray or white matter. On CT, TLN appears patchy but delayed scans often show less inhomogeneity and better-defined margins. On MR, the necrotic foci show patchy enhancement but demarcation from the adjacent brain is better seen. Necrotic foci within the gray matter are often associated with minimal edema. Abstract:Neonatal hypothyroidism impairs structural maturation in the brain and results in diminished electrical activities and energy metabolism. We recently found that glucose utilization (CMR(glc)) is markedly depressed throughout the brain in mice with targeted mutations in thyroid hormone receptor alpha1 (TR alpha 1), but not TR beta. Previous studies had shown that CMR(glc) increases linearly with spike frequency in the afferent pathways to synapse-rich regions in neuropil, but not in neuronal cell bodies. To determine whether the decreased CMR(glc) in mutant TR alpha 1(PV/+) mice reflected lesser synaptic density or reduced functional activity in existing synapses, we stimulated vibrissae unilaterally and measured CMR(glc) bilaterally in four stations of the whisker-to-barrel cortex pathway. Baseline CMR(glc) (unstimulated side) was markedly lower in all four stations in the TR alpha 1(PV/+) mutants than in wild-type controls, even though Northern blot and immunohistochemical examinations showed normal Na(+),K(+)-adenosine triphosphatase expression and neuronal differentiation. Despite the lower baseline CMR(glc), however, vibrissal stimulation evoked percent increases in CMR(glc) in the TR alpha 1(PV/+) mutants that were as great as those in wild-type mice. These results indicate that in the TR alpha 1(PV/+) mutants there it is a reduction in synaptic density that is responsible for the decrease in CMR(glc), but functionality of existing synapses is retained.
Comparison of iodothyronine 5'-deiodinase and other thyroid-hormone-dependent enzyme activities in the cerebral cortex of hypothyroid neonatal rat. Evidence for adaptation to hypothyroidism

Abstract:Recent studies have shown that approximately 75% of the nuclear 3,5,3'-triiodothyronine (T(3)) present in adult rat cerebral cortex (Cx) derives from 5'-deiodination of thyroxine (T(4)) within this tissue. The activity of iodothyronine 5'-deiodinase (I 5'D), the enzyme catalyzing T(4) to T(3) conversion, increases rapidly after thyroidectomy, suggesting that this could be a compensatory response to hypothyroxinemia. To evaluate this possibility during the period of central nervous system maturation, we studied several thyroid hormone-responsive enzymes (aspartic transaminase [AT], succinic dehydrogenase [S.D.], and Na/K ATPase) in the Cx of 2-, 3-, and 4-wk-old rats. The rats were made congenitally hypothyroid by placing 1, 2, 5, and 20 mg methimazole (MMI) in 100 ml of the mothers' drinking water from day 16 of gestation throughout the nursing period and to the litters after weaning. In addition, serum thyroid hormones, I 5'D, and, in some experiments, in vivo T(4) to T(3) conversion in Cx were measured in the same pups. Serum T(4) concentrations varied from <1 to 40 ng/ml and were generally inversely related to maternal MMI dose. Serum T(3) was less affected by MMI than was T(4). At 2 wk, decreases in AT, S.D., and ATPase were present in the 20-mg-MMI group but not in the 5-mg-MMI pups despite low serum T(4) (<10 ng/ml) in the latter. At 3 and 4 wk, both 5- and 20-mg-MMI groups had significant reductions in these cortical enzymes despite a normal serum T(3) in the 5-mg-MMI rats. Cortical I 5'D activity was 10-fold the control value in 5- and 20-mg-MMI animals at 2 wk but increased only three- to fivefold at 3 and 4 wk. I 5'D correlated inversely with serum T(4) (r >/= 0.96) at all ages, but the less marked elevation of this enzyme in 3- and 4-wk-old pups was not accompanied by an increase in serum T(4). Serum T(3) increased or remained the same between 2 and 3 wk. These results suggested that the 10-fold increase in I 5'D at 2 wk protected the 5-mg-MMI group from tissue hypothyroidism, but that the three- to fivefold increase at 3 and 4 wk could not. Injection of approximately 250 ng T(4)/100 g body weight to 2-wk-old, 20-mg-MMI pups (one-sixth the normal T(4) production rate) led to both a 1.8-ng/g cortical tissue increment in cortical T(3) and a significant increase in AT at 24 h, compared with a 0.38-ng/g cortical tissue T(3) increment and no change in AT in euthyroid controls. The larger increment in T(3) of the 20-mg-MMI pups was due in great part to increased fractional T(4) to T(3) conversion. Although the latter resulted in greater serum T(3) concentrations, three-fourths of the newly formed T(3) in the cortex was generated in situ, and it was blocked by iopanoic acid as was the increase in AT. We conclude that 70-80% of the T(3) in the Cx of the neonatal rat is produced locally. Serum T(4) appears to serve both as a precursor for T(3) and as a critical signal for increases in cortical I 5'D. The increased I 5'D can result in normal or near-normal cerebrocortical T(3) concentrations despite marked reductions in serum T(4). This mechanism seems to be particularly effective around 2 wk of age when many thyroid-hormone-dependent maturational changes occur in the rat Cx.
[Dynamics of postnatal histogenesis of the thyroid in normal rats and after sympathectomy]

Abstract:The changes of the thyroid gland and neurocytes of the cranial sympathetic ganglia were followed in rats of different ages after guanethidine injections with the use of radioimmunological assay, electron microscopy and morphometry. The injection of 15 mg of the drug per kg of body weight within the first two weeks after birth caused the death of over 80% of the cells in the sympathetic ganglion. In the sympathectomized 15-day- and 1-month-old rats the functional activity of the thyroid gland was markedly reduced. Later on, intrathyroid hormonogenesis somewhat increases due, apparently, to partial recovery of the organ adrenergic innervation and increase in the production of thyrotropic hypophysial hormone and calcitonin.
Free Thyroid Hormone Concentrations during Postnatal Development in the Rat

Abstract:Summary: Sprague-Dawley rats were sacrificed by decapitation at 5, 7, 12, 14, 22, 26, 32, and 40 days of age. Adult animals (175 to 225 g) were also studied. Serum-free thyroxine (FT4) concentrations rose rapidly between 5 and 12 days to levels similar to adult concentrations, whereas the percentage of FT4 was relatively high between 5 and 12 days before declining to adult values by 14 days. Serum-free triiodothyronine (FT3) concentrations rose progressively to attain peak concentrations at 26 days and subsequently declined to adult levels by 40 days. The percentage FT3 rose in parallel with the FT3 concentrations to peak values at 22 to 26 days before declining to adult levels. FT4/thyroid-stimulating hormone (TSH) and FT3/TSH ratios increased progressively through 22 days of age in parallel with the FT3/FT4 ratio. These data indicate that free thyroid hormone concentrations follow essentially the same developmental profile as do total thyroid hormone concentrations. Progressive maturation of the negative feedback control mechanism for the pituitary-thyroid axis, as assessed by the FT4/TSH and FT3/TSH ratios, occurs through 14 days. However, the continued rise in FT3 concentrations, FT3/TSH, and FT3/FT4 ratios through 26 days suggests a further resetting of the setpoint of the pituitary-thyroid axis possibly related to the stress of weaning.Speculation: Maturation of the pituitary-thyroid axis in the rat is principally a postnatal phenomenon. Careful study of the developmental profile of this axis under physiologic and pathologic states may afford considerable insight into the ontogenesis of the pituitary-thyroid axis in the human fetus.

Abstract:Thyroid hormones play an important morphogenetic role during the fetal and neonatal periods and regulate numerous metabolic processes. In the central nervous system, they control myelination and overall brain development, regional gene expression, and regulation of oxygen consumption. Their deficiency in the fetal and neonatal periods causes severe mental retardation, due to lack of thyroid function, or to iodine deficiency. At the same time, nitric oxide is an atypical neurotransmitter that also has special relevance in neuronal development and plasticity and functions as a vasodilator, regulating cerebral blood flow. Although under physiological conditions it functions as a neuroprotector, in excess it can be neurotoxic. We have studied, by immunocytochemical and Western blot techniques, the evolution of the expression of neuronal and inducible isoforms of the enzyme nitric oxide synthase, and of nitrotyrosine as a marker of protein nitration produced by the presence of nitric oxide, during the early stages of postnatal brain development. We induced hypothyroidism by administering mercaptomethylimidazole to pregnant mothers, from the seventh day of gestation until the sacrifice of the offspring. The results show a delay in the evolution of the expression of the two isoforms of the enzyme nitric oxide synthase in hypothyroid animals, followed by an anomalous overexpression in later stages. Finally, the expression of nitrotyrosine follows an evolution that is synchronized with that shown by both isoenzymes in control and hypothyroid animals.
Development and Validation of a Bioanalytical Method for Quantification of 2,6-Bis-(4-hydroxy-3-methoxybenzylidene)-cyclohexanone (BHMC) in Rat Plasma

Abstract:A sensitive and accurate high performance liquid chromatography with ultraviolet/visible light detection (HPLC-UV/VIS) method for the quantification of 2,6-bis-(4-hydroxy-3-methoxybenzylidene)-cyclohexanone (BHMC) in rat plasma was developed and validated. BHMC and the internal standard, harmaline, were extracted from plasma samples by a simple liquid–liquid extraction using 95% ethyl acetate and 5% methanol. Plasma concentration of BHMC and internal standard were analyzed by reversed phase chromatography using a C18 column (150 × 4.6 mm I.D., particle size 5 µm) and elution with a gradient mobile phase of water and methanol at a flow rate of 1.0 mL/min. Detection of BHMC and internal standard was done at a wavelength of 380 nm. The limit of quantification was 0.02 µg/mL. The calibration curves was linear (R2 > 0.999) over the concentration range of 0.02–2.5 µg/mL. Intra- and inter-day precision were less than 2% coefficient of variation. The validated method was then applied to a pharmacokinetic study in rats by intravenous administration of BHMC at a single dose of 10 mg/kg. Pharmacokinetic parameters such as half-life, maximum plasma concentration, volume of distribution, clearance and elimination rate constant for BHMC were calculated.

Abstract:Thyroid hormones play important roles in the maturation and function of the central nervous system. However, the underlying mechanism behind thyroid hormone-regulated gene expression in the adult brain is not well understood. Two genes critical for neuronal plasticity and implicated in psychiatric disorders, reelin and brain-derived neurotrophic factor (BDNF), were investigated in the present study. Triiodothyronine (T3), the active form of thyroid hormone was administered to young adult rats in two different manners: systemic injection or local brain infusion. Real time RT-PCR results revealed that T3 administration lead to a significant increase in reelin, total BDNF and exon-specific BDNF mRNA expression in the hippocampus. Furthermore, the association of transcriptional coactivators (including steroid receptor coactivator-1 (SRC-1), cAMP response element binding protein-binding protein (CBP), and thyroid hormone receptor associated protein 220 (TRAP 220)) and RNA polymerase II (RNA Pol II), with reelin and BDNF genes in the rat hippocampus displayed a distinct process following thyroid hormone administration. These findings suggest that association of transcriptional coactivators and RNA Pol II with gene promoters may be a possible mechanism explaining T3-induced reelin and BDNF expression in the hippocampus of young adult rats.
Influence of thyroid hormones on myelin proteins in the developing rat brain

Abstract:This communication describes developmental changes in myelin proteins prepared from control, hypothyroid, and hyperthyroid rat brain. In the 10- to 37-day postnatal period studied, total myelin protein was found to double, and this change mainly reflected the increase in proteolipid and basic protein constituents. Thyroid states affect differentially the various myelin proteins. Hypothyroidism decreases the proteolipid and slow-moving basic protein, but has no effect on the fast basic or minor proteins. In hyperthyroidism, an increase was observed in proteolipid as well as both slow- and fast-moving proteins. The protein alterations were correlated to the changes previously found in lipid composition of myelin consequent upon hypo- and hyperthyroidism, and the role of thyroid hormones in brain development.

Abstract:Abstract In this paper the problem of detecting and isolating faults on the actuators of a small commercial aircraft using online approximators is dealt with. In particular the capabilities of radial basis neural networks to approximate faults on the engines and elevators of an AP-68 Vulcanair commercial aircraft are investigated also in the presence of model uncertainties and external disturbances. The structure of the aircraft and of the fault models appears to be well suited for this kind of application. Although some effort has to be spent in tuning a certain number of parameters, the numerical results obtained on a full nonlinear Matlab-Simulink simulator of the aircraft seem to be encouraging.

Abstract:SUMMARY1. Alterations in the cardiac ai‐adrenoceptor and its subtypes in hypothyroid rats were studied by radioligand binding assays and reverse transcription‐polymerase chain reaction (RT‐PCR). Hypothyroidism was created by feeding rats with 0.2% 2‐thiouraciI solution instead of daily drinking water for 20 days.2. The density of cardiac α1‐adrenoceptors (Bmax) was increased from 67.5 ± 4.3 fmol/mg in control rats to 81.1 ± 7.2 fmol/mg (P < 0.05) in hypothyroid rats.3. Compared with control rats, in hypothyroid rats the percentages of high‐affinity sites for (+)‐niguldipine and 5‐methyl‐urapidil were increased from 13.8 ± 5.6 and 31.9 ± 6.3%, respectively, to 24.9 ± 73 and 45.5 ± 2.4%, respectively (both P < 0.05), while those for BMY7378 were decreased from 37.2 ± 8.9 to 23.8 ± 8.4% (P < 0.05), respectively. The percentage of high‐affinity sites for WB4101 was not significantly different in control and hypothyroid rats (43.3 ± 9.1 and 39.4 ± 3.6%, respectively).4. Reverse transcription‐PCR experiments revealed that the steady state levels of mRNA for otiA‐ and am‐adrenoceptors were increased, while those for ctiD‐adrenoceptor were decreased in the hearts of hypothyroid rats.5. The concentration‐contraction response curves for noradrenaline in the presence of a β‐adrenoceptor antagonist in control and hypothyroid rats showed that the maximal response was reduced from 344 ± 58 to 200 ± 23 mg, respectively (P < 0.05).6. The data suggest that in hypothyroid rats the total number of cardiac ai‐adrenoceptors is increased. The change is subtype‐selective, with levels of ctiA‐ and am‐adrenoceptors being increased and levels of aiD‐adrenoceptors being reduced. Furthermore, the positive inotropic response mediated by a i‐adrenoceptors is reduced in hypothyroid rats.
