Abstract:Understanding the microbiology of phosphorus (P) removal is considered essential to knowledge-based optimization of enhanced biological P removal (EBPR) systems. Biological P removal is achieved in these systems by promoting the growth of organisms collectively known as the polyphosphate accumulating organisms (PAOs). Also considered important to EBPR are the glycogen accumulating organisms (GAOs), which are theorized to compete with the PAOs for resources at the expense of P removal efficiency. Numerous studies have sought to identify the PAOs and their GAOs competitors, with several candidates proposed for each over the last few decades. The current study collectively assessed the abundance and diversity of all proposed PAOs and GAOs in 18 Danish full-scale wastewater treatment plants with well-working biological nutrient removal over a period of 9 years using 16S rRNA gene amplicon sequencing. The microbial community structure in all plants was relatively stable over time. Evidence for the role of the proposed PAOs and GAOs in EBPR varies and is critically assessed, in light of their calculated amplicon abundances, to indicate which of these are important in full-scale systems. Bacteria from the genus Tetrasphaera were the most abundant of the PAOs. The "Candidatus Accumulibacter" PAOs were in much lower abundance and appear to be biased by the amplicon-based method applied. The genera Dechloromonas, Microlunatus, and Tessaracoccus were identified as abundant putative PAO that require further research attention. Interestingly, the actinobacterial Micropruina and sbr-gs28 phylotypes were among the most abundant of the putative GAOs. Members of the genera Defluviicoccus, Propionivibrio, the family Competibacteraceae, and the spb280 group were also relatively abundant in some plants. Despite observed high abundances of GAOs (periodically exceeding 20% of the amplicon reads), P removal performance was maintained, indicating that these organisms were not outcompeting the PAOs in these EBPR systems. Phylogenetic diversity within each of the PAOs and GAOs genera was observed, which is consistent with reported metabolic diversity for these. Whether or not key traits can be assigned to sub-genus level clades requires further investigation.

Abstract:Glycogen-accumulating organisms (GAOs) are recognized as competitors of phosphorus-accumulating organisms (PAOs) in enhanced biological phosphorus removal (EBPR) systems. However, different types of GAOs exhibit unique metabolic traits in different environments. To characterize the metabolic activity of GAOs, two distinct modes were constructed. Long-term operation with the mode of anaerobic post-drainage demonstrated the successful establishment of a rapid and enduring GAOs system dominated by Candidatus Contendobacter community. Acetate was completely consumed within 60 min of the anaerobic phase. Concurrently, glycogen levels decreased by 0.062 g Glycogen/g SS, while intracellular carbon storage of poly-hydroxybutyrate (PHB) and poly-hydroxyvalerate (PHV) increased by 0.027 g PHB/g SS and 0.007 g PHV/g SS, respectively. Various concentrations of phosphorus loads illustrated the resistance of the system to high phosphorus loads, while maintaining glycogen accumulating metabolism (GAM). Under complete phosphorus limitation, no correlation between glycogen and acetate was observed during the initial 140 d. The addition of 1 mg/L phosphate to the influent preserved the typical GAM characteristics. Moreover, the pathway involved in glycogen conversion to trehalose may be pivotal in enhancing anaerobic glycogen degradation. The complete tricarboxylic acid cycle ensured efficient energy production, thereby sustaining the foundational metabolism. Notably, sucC and mdh were critical regulators of energy flux. This study provides a distinctive metabolic profile of a robust and resilient GAOs model with Ca. Contendobacter .

Fine-Scale Population Structure of Accumulibacter Phosphatis in Enhanced Biological Phosphorus Removal Sludge

Abstract:To investigate the diversities of Accumulibacter phosphatis and its polyhydroxyalkanoate (PHA) synthase gene (phaC) in enhanced biological phosphorus removal (EBPR) sludge, an acetate-fed sequencing batch reactor was operated. Analysis of microbial communities using fluorescence in situ hybridization and 16S rRNA gene clone libraries showed that the population of Accumulibacter phosphatis in the EBPR sludge comprised more than 50% of total bacteria, and was clearly divided into two subgroups with about 97.5% sequence identity of the 16S rRNA genes. PAO phaC primers targeting the phaC genes of Accumulibacter phosphatis were designed and applied to retrieve fragments of putative phaC homologs of Accumulibacter phosphatis from EBPR sludge. PAO phaC primers targeting G(1PAO), G(2PAO), and G(3PAO) groups produced PCR amplicons successfully; the resulting sequences of the phaC gene homologs were diverse, and were distantly related to metagenomic phaC sequences of Accumulibacter phosphatis with 75-98% DNA sequence identities. Degenerate NPAO (non-PAO) phaC primers targeting phaC genes of non-Accumulibacter phosphatis bacteria were also designed and applied to the EBPR sludge. Twenty-four phaC homologs retrieved from NPAO phaC primers were different from the phaC gene homologs derived from Accumulibacter phosphatis, which suggests that the PAO phaC primers were specific for the amplification of phaC gene homologs of Accumulibacter phosphatis, and the putative phaC gene homologs by PAO phaC primers were derived from Accumulibacter phosphatis in the EBPR sludge. Among 24 phaC homologs, a phaC homolog (G1NPAO-2), which was dominant in the NPAO phaC clone library, showed the strongest signal in slot hybridization and shared approximately 60% nucleotide identity with the G(4PAO) group of Accumulibacter phosphatis, which suggests that G1NPAO-2 might be derived from Accumulibacter phosphatis. In conclusion, analyses of the 16S rRNA and phaC genes showed that Accumulibacter phosphatis might be phylogenetically and metabolically diverse.
In situ identification of polyphosphate- and polyhydroxyalkanoate-accumulating traits for microbial populations in a biological phosphorus removal process

Abstract:Polyphosphate- and polyhydroxyalkanoate (PHA)-accumulating traits of predominant microorganisms in an efficient enhanced biological phosphorus removal (EBPR) process were investigated systematically using a suite of non-culture-dependent methods. Results of 16S rDNA clone library and fluorescence in situ hybridization (FISH) with rRNA-targeted, group-specific oligonucleotide probes indicated that the microbial community consisted mostly of the alpha- (9.5% of total cells), beta- (41.3%) and gamma- (6.8%) subclasses of the class Proteobacteria, Flexibacter-Cytophaga (4.5%) and the Gram-positive high G+C (HGC) group (17.9%). With individual phylogenetic groups or subgroups, members of Candidatus Accumulibacter phosphatis in the beta-2 subclass, a novel HGC group closely related to Tetrasphaera spp., and a novel gamma-proteobacterial group were the predominant populations. Furthermore, electron microscopy with energy-dispersive X-ray analysis was used to validate the staining specificity of 4,6-diamino-2-phenylindole (DAPI) for intracellular polyphosphate and revealed the composition of polyphosphate granules accumulated in predominant bacteria as mostly P, Ca and Na. As a result, DAPI and PHA staining procedures could be combined with FISH to identify directly the polyphosphate- and PHA-accumulating traits of different phylogenetic groups. Members of Accumulibacter phosphatis and the novel gamma-proteobacterial group were observed to accumulate both polyphosphate and PHA. In addition, one novel rod-shaped group, closely related to coccus-shaped Tetrasphaera, and one filamentous group resembling Candidatus Nostocoidia limicola in the HGC group were found to accumulate polyphosphate but not PHA. No cellular inclusions were detected in most members of the alpha-Proteobacteria and the Cytophaga-Flavobacterium group. The diversified functional traits observed suggested that different substrate metabolisms were used by predominant phylogenetic groups in EBPR processes.
‘Candidatus Competibacter’-lineage genomes retrieved from metagenomes reveal functional metabolic diversity

Abstract:The glycogen-accumulating organism (GAO) ‘Candidatus Competibacter’ (Competibacter) uses aerobically stored glycogen to enable anaerobic carbon uptake, which is subsequently stored as polyhydroxyalkanoates (PHAs). This biphasic metabolism is key for the Competibacter to survive under the cyclic anaerobic-‘feast’: aerobic-‘famine’ regime of enhanced biological phosphorus removal (EBPR) wastewater treatment systems. As they do not contribute to phosphorus (P) removal, but compete for resources with the polyphosphate-accumulating organisms (PAO), thought responsible for P removal, their proliferation theoretically reduces the EBPR capacity. In this study, two complete genomes from Competibacter were obtained from laboratory-scale enrichment reactors through metagenomics. Phylogenetic analysis identified the two genomes, ‘Candidatus Competibacter denitrificans’ and ‘Candidatus Contendobacter odensis’, as being affiliated with Competibacter-lineage subgroups 1 and 5, respectively. Both have genes for glycogen and PHA cycling and for the metabolism of volatile fatty acids. Marked differences were found in their potential for the Embden–Meyerhof–Parnas and Entner–Doudoroff glycolytic pathways, as well as for denitrification, nitrogen fixation, fermentation, trehalose synthesis and utilisation of glucose and lactate. Genetic comparison of P metabolism pathways with sequenced PAOs revealed the absence of the Pit phosphate transporter in the Competibacter-lineage genomes—identifying a key metabolic difference with the PAO physiology. These genomes are the first from any GAO organism and provide new insights into the complex interaction and niche competition between PAOs and GAOs in EBPR systems.
Biological Phosphorus Removal Performance and Relevant Microorganism Characteristics of Activated Sludge in Municipal Wastewater Treatment Plants, China

Abstract:Municipal wastewater discharge is threatening the ecological security of the local water environment. This study investigated the field process performance and microorganism characteristics of enhanced biological phosphorus removal (EBPR) in municipal wastewater treatment plants (WWTPs) in China. The results showed that three WWTPs met the required criteria of phosphorus for discharge (≤1 mg/L), but with a low level of Accumulibacter (6.4 to 3.8%, on average) and a relatively high level of Competibacter (3.2 to 9.1%) in sludge. The phosphorus release and uptake rates were varied from 0.224 to 7.770 mg/gVSS·h and 0.386 to 7.901 mg/gVSS·h, respectively. Denitrifying polyphosphate‐accumulating organisms were estimated to be 28.2% of the polyphosphate‐accumulating organisms. Sludge characteristics (phosphorus release and uptake rates) were positively correlated with the abundance of Accumulibacter and negatively correlated with the proportion of Competibacter. Moreover, the lower the ratio of anaerobic phosphorus/acetate (e.g., 0.496) is, the more abundant the Competibacter would be. Further discussion on an improvement strategy for these WWTPs for EBPR should be comprehensively based on the data of periodic investigations on field operation, sludge activities, and microbial populations.
Exploration of the metabolic flexibility of glycogen accumulating organisms through metatranscriptome analysis and metabolic characterization

Abstract:Glycogen accumulating organisms (GAOs) are closely related to the deterioration of enhanced biological phosphorus removal systems. However, the metabolic mechanisms that drive GAOs remain unclear. Here, the two-thirds supernatant of a reactor were decanted following the anaerobic period to enrich GAOs. Long-term monitoring demonstrated that the system was stable and exhibited typical characteristics of GAOs metabolism. Acetate was completely consumed after 60 min of the anaerobic phase. The level of glycogen decreased from 0.20 to 0.14 g/gSS during the anaerobic phase, whereas the level of glycogen significantly increased to 0.21g/gSS at the end of the aerobic period. Moreover, there was almost no phosphate release and absorption in the complete periods, thus confirming the successful construction of a GAOs enrichment system. Microbial community analysis demonstrated that Ca. Contendobacter was among the core functional genera and showed the highest activity among all of the communities. Furthermore, our study is the first to identify the involvement of the ethyl-malonyl-CoA pathway in the synthesis of polyhydroxyvalerate via croR, ccr, ecm, mcd, mch and mcl genes. The Embden-Meyerhof-Parnas (EMP) pathway was preferentially used via glgP. Furthermore, the glyoxylate cycle was the main source of ATP under anaerobic conditions, whereas the tricarboxylic acid cycle provided ATP under aerobic conditions. aceA and mdh appeared to be major modulators of the glyoxylate pathway for controlling energy flow. Collectively, our findings not only revealed the crucial metabolic mechanisms in a GAOs enrichment system but also provided insights into the potential application of Ca. Contendobacter for wastewater treatment.

Abstract:We report on magneto-oscillations in differential resistivity of a two-dimensional electron system subject to intense microwave radiation. The period of these oscillations is determined not only by microwave frequency but also by its intensity. A theoretical model based on quantum kinetics at high microwave power captures all important characteristics of this phenomenon which is strongly nonlinear in microwave intensity. Our results demonstrate a crucial role of the multiphoton processes near the cyclotron resonance and its harmonics in the presence of strong dc electric field and offer a unique way to reliably determine the intensity of microwaves acting on electrons.
Study on community structure and metabolic mechanism of dominant polyphosphate-accumulating organisms (PAOs) and glycogen-accumulating organisms (GAOs) in suspended biofilm based on phosphate recovery

Abstract:Biofilm sequencing batch reactor (BSBR) can achieve efficient phosphate (P) removal and enrichment, but its process performance and metabolic mechanisms for P removal and enrichment of municipal wastewater remain largely unclear. In the present study, we assessed the P removal and enrichment of municipal wastewater at influent P concentrations of 2.5 mg/L and 10 mg/L. The efficiency of P removal and enzyme activity in polyphosphate-accumulating organisms (PAOs) and glycogen-accumulating organisms (GAOs) were compared, and the growth and metabolic characteristics of dominant PAOs and GAOs at different influent P concentrations were studied with the macro-sequencing technology. The results showed that the P recovery efficiencies were 70.03% and 76.19% when the influent P concentration was 2.5 mg/L and 10 mg/L in BSBR, respectively, and the maximum P concentration of recovery liquid was 81.29 mg/L and 173.12 mg/L, respectively. There were no phosphate kinase (PPK) and phosphate hydrolase (PPX) in extracellular polymeric substances (EPS). The dominant PAOs were Candidatus_Contendobacter, Dechloromonas, and Flavobacterium, and the dominant GAO was Candidatus_Competibacter. The abundance of Candidatus_Contendobacter was the highest with the most potential contribution to P removal. PAOs had competitive advantages in carbon (C) source uptake, glycogen metabolism, P metabolism, and adenosine triphosphate (ATP) metabolism. HMP was unique to PAOs, EMP had the highest abundance in glycogen metabolism, and ED was contained in PAOs of BSBR. These results indicated that BSBR provided sufficient reducing power and ATP for PAOs through different glycogen decomposition pathways to promote P uptake and obtained competitive advantages in P metabolism, C source uptake, and ATP utilization to achieve efficient P removal and enrichment. Collectively, our current findings provided valuable insights into the P removal and enrichment mechanism of BSBR in municipal sewage.

Abstract:A high phosphate (P) recovery concentration was achieved in pilot-scale biofilm sequencing batch reactor (BSBR) with a low carbon source (C) cost. Especially, a high-abundance glycogen-accumulating organisms (GAOs) (13.93–31.72%) was detected that was accompanied by a high P recovery concentration of BSBR. High-abundance GAOs obtain additional C through various C compensation pathways (split tricarboxylic acid cycle (TCA cycle), glyoxylate shunt and gluconeogenesis), thus reducing the need to compete with polyphosphate-accumulating organisms (PAOs) for C and weakening the adverse effects on P recovery by PAO cells. Under the action of N -acyl homoserine lactones (AHLs)-mediated quorum sensing (QS), GAOs promoted the secretion of a large amount of extracellular polymeric substances (EPS), which helped to realize the P recovery of EPS-dominated biofilms (68.02%–96.89%). This study provides a low-carbon technology for the recovery of high concentration P from municipal wastewater, and improves the ecological theory of P recovery in collaboration with GAOs and PAOs.

Characterization of an Acetate-Degrading Sludge Without Intracellular Accumulation of Polyphosphate and Glycogen

Abstract:A sequencing batch reactor (SBR) was operated in the conventional anaerobic–aerobic mode for enhanced biological phosphate removal (EBPR) using acetate as the sole substrate. Results showed that, however, the reactor was unable to remove phosphate from wastewater. The sludge containing 1.65% of phosphate did not exhibit the typical characteristics of polyphosphate-accumulating organisms (PAO) or glycogen-accumulating organisms (GAO). Phylogenetic analysis, based on 16S rDNA sequences of individual microorganisms, showed that the microbial community of this acetate-degrading sludge was closely related to Comamonas testosteroni (43.8% of total population) of β-1-proteobacteria, Zoogloea resiniphila (25.0%) of β-2-proteobacteria, and Xanthomonas maltophilia (19.8%) of γ-proteobacteria. Results of this study imply that GAO might not be the sole group of bacteria responsible to the deterioration of phosphate removal efficiency in an EBPR reactor.

Abstract:A sequencing batch reactor fed mainly by acetate was operated to perform enhanced biological phosphorus removal (EBPR). A short-term pH shock from 7.0 to 6.0 led to a complete loss of phosphate-removing capability and a drastic change of microbial communities. 16S rRNA gene pyrosequencing showed that large proportions of glycogen accumulating organisms (GAOs) (accounted for 16% of bacteria) bloomed, including Candidatus Competibacter phosphatis and Defluviicoccus-related tetrad-forming organism, causing deteriorated EBPR performance. The EBPR performance recovered with time and the dominant Candidatus Accumulibacter (Accumulibacter) clades shifted from Clade IIC to IIA while GAOs populations shrank significantly. The Accumulibacter population variation provided a good opportunity for genome binning using a bi-dimensional coverage method, and a genome of Accumulibacter Clade IIC was well retrieved with over 90% completeness. Comparative genomic analysis demonstrated that Accumulibacter clades had different abilities in nitrogen metabolism and carbon fixation, which shed light on enriching different Accumulibacter populations selectively.
New insight into the granule formation in the reactor for enhanced biological phosphorus removal

Abstract:While granulated activated sludge exhibits high productivity, the processes of granule formation are incompletely studied. The processes of granule formation and succession of communities were investigated in a laboratory sequencing batch reactor (SBR) under conditions for enhanced biological phosphorus removal (EBPR) using microbiological and molecular techniques. Active consumption of acetate, primarily by the phosphate-accumulating organisms (PAO), commenced at day 150 of cultivation. This was indicated by the high ratio of molar P-released/acetate uptake (0.73–0.77 P-mol/C-mol), characteristic of PAO. During this period, two types of granule-like aggregates formed spontaneously out of the activated sludge flocs. The aggregates differed in morphology and microbial taxonomic composition. While both aggregate types contained phosphorus-enriched bacterial cells, PAO prevailed in those of morphotype I, and glycogen-accumulating organisms (GAOs) were predominant in the aggregates of morphotype II. After 250 days, the elimination of the morphotype II aggregates from the reactor was observed. The subsequent selection of the community was associated with the development of the morphotype I aggregates, in which the relative abundance of PAO increased significantly, resulting in higher efficiency of phosphorus removal. Metagenomic analysis revealed a predominance of the organisms closely related to Candidatus Accumulibacter IС and IIС and of Ca. Accumulibacter IIB among the PAO. Based on the content of the genes of the key metabolic pathways, the genomes of potential PAO belonging to the genera Amaricoccus, Azonexus, Thauera, Zoogloea, Pinisolibacter , and Siculibacillus were selected. The patterns of physicochemical processes and the microbiome structure associated with granule formation and succession of the microbial communities were revealed.

Abstract:Impact of marginal availability and severe deficiency of nitrogen on the growth of glycogen accumulating organisms (GAOs) and performance of anaerobic/aerobic activated sludge systems treating nutrient deficient wastewater was investigated at marginal availability and severe deficiency of phosphorus. Two continuous-flow lab-scale systems were operated simultaneously fed by model winery wastewater, one with marginal availability and the other one at severe deficiency of nitrogen. In the second experimental stage, marginal availability of P was converted into severe deficiency by interrupting external dosing.Common practice of dosing N- and P-sources to marginal availability caused enhanced proliferation of filamentous bacteria leading to poor biomass settling and instable operation. At marginal N-availability accumulation of GAOs started when conditions became deficient for phosphorus. Abstract:AbstractThe enhanced biological phosphorus removal (EBPR) has been widely applied in treating domestic wastewater, while the performance on high-strength P wastewater is less investigated and the feasibility of coupling with short-cut nitrogen removal process remains unknown. This study first achieved the simultaneous high-efficient P removal and stable nitrite accumulation in one sequencing batch reactor for treating the synthetic digested manure wastewater. The average effluent P could be down to 0.8 ± 1.0 mg P/L and the P removal efficiency was 99.5 ± 0.8%. Candidatus Accumulibacter was the dominant polyphosphate accumulating organism (PAO) with the relative abundance of 14.2-33.1% in the reactor. Examination of the micro-diversity of Candidatus Accumulibacter using 16s rRNA gene-based oligotyping analysis revealed one unique Accumulibacter oligotype that different from the conventional system, which accounted for 64.2-87.9% of the total Accumulibacter abundance. The presence of high-abundant glycogen accumulating organisms (GAO) (15.6-40.3%, Defluviicoccus and Candidatus Competibacter) did not deteriorate the EBPR performance. Moreover, nitrite accumulation happened in the system with the effluent nitrite up to 20.4 ± 6.4 mg N/L and the nitrite accumulation ratio was nearly 100% maintained for 140 days (420 cycles). Nitrosomonas was the dominant ammonia-oxidizing bacteria with relative abundance of 0.3-2.4% while nitrite-oxidizing bacteria were almost undetected (<0.1%). The introduction of extended anaerobic phase and high volatile fatty acid concentrations were proposed to be the potential selector forces to promote partial nitrification. This is the first study that combined EBPR with nitrite-accumulation for digested manure wastewater treatment, and it provided new sights in strategies to combine the EBPR and short-cut nitrogen removal via nitrite to achieve simultaneous nitrogen and phosphorus removal.

Abstract:The identification of phosphate-accumulating organisms (PAOs), denitrifying phosphate-accumulating organisms (DPAOs) and their relationship is a key pathway for optimizing nitrate and phosphate removal efficiency in activated sludge. In this study, the acclimatization of microorganisms in sequencing batch reactor were performed with anaerobic/aerobic (A/O) and anaerobic/anoxic (A/A) cycles, the biomass changes of PAOs and DPAOs and the correlations were then discussed. The results indicated that after acclimatization, the nutrient removal efficiencies reached to 85.34% (COD), 93.64% (PO43−-P) and 92.34% (NO3−-N), respectively, with NO3−-N:PO43-P of 1.5:1. The successful enrichment of PAOs and DPAOs (reached 97.9%) was verified by the change of relative metabolic activities, which was further proved by the change of bacterial diversity. The number of Candidatus Accumulibacter, Zoogloea, and Dechloromonas all increased at A/O and A/A stages while the number of Acinetobacter only increased at A/O stage. So Accumulibacter sp. was DPAO while Acinetobacter sp. was only PAO in this process, and genera Accumulibacter, Dechloromonas and Zoogloea greatly coordinated in denitrification and accumulating phosphorous though RDA and chord plot. This was worthy of attention and development to explore enhanced biological phosphorus removal (EBPR) in practical wastewater treatment via improving identification of bacterial species and symbiosis of bacteria community.

Abstract:Enhanced biological phosphorus removal (EBPR) may deteriorate or fail during low organic carbon loading periods. Polyphosphate accumulating organisms (PAOs) in EBPR were acclimated under both high and low organic carbon conditions, and then dynamics of polymers in typical cycles, anaerobic conditions with excess organic carbons, and endogenous respiration conditions were examined. After long-term acclimation, it was found that organic loading rates did not affect the yield of PAOs and the applied low organic carbon concentrations were advantageous for the enrichment of PAOs. A low influent organic carbon concentration induced a high production of extracellular carbohydrate. During both anaerobic and aerobic endogenous respirations, when glycogen decreased to around 80 ± 10 mg C per gram of volatile suspended solids, PAOs began to utilize polyphosphate significantly. Regressed by the first-order reaction model, glycogen possessed the highest degradation rate and then was followed by polyphosphate, while biomass decay had the lowest degradation rate.

Abstract:Production of polyhydroxyalkanoates (PHAs) by an open mixed culture enriched in glycogen accumulating organisms (GAOs) under alternating anaerobic-aerobic conditions with acetate as carbon source was investigated. The culture exhibited a stable enrichment performance over the 450-day operating period with regards to phenotypic behavior and microbial community structure. Candidatus Competibacter phosphatis dominated the culture at between 54% and 70% of the bacterial biomass throughout the study, as determined by fluorescence in situ hybridization. In batch experiments under anaerobic conditions, PHA containing 3-hydroxybutyrate (3HB) and 27 mol-% 3-hydroxyvalerate (3HV) was accumulated up to 49% of cell dry weight utilizing the glycogen pool stored in the SBR cycle. Under aerobic and ammonia limited conditions, PHA comprising only 3HB was accumulated to 60% of cell dry weight. Glycogen was consumed during aerobic PHA accumulation as well as under anaerobic conditions, but with different stoichiometry. Under aerobic conditions 0.31 C-mol glycogen was consumed per consumed C-mol acetate compared to 0.99 under anaerobic conditions. Both the PHA biomass content and the specific PHA production rate obtained were similar to what is typically obtained using the more commonly applied aerobic dynamic feeding strategy.

Enhanced Biological Phosphorus Removal in Aerobic Granular Sludge Reactors by Granular Activated Carbon Dosing

Abstract:This study investigated the effects of granular activated carbon (GAC) addition on the enrichment of polyphosphate accumulating organisms (PAOs), stratification of PAOs in the co-existing GAC-biofilms and granules and biological nutrient removal (BNR) in aerobic granular sludge (AGS) reactors. It was found that BNR increased in the GAC-augmented system. Establishment of enhanced biological phosphorus removal (EBPR) pathway was faster with about 1.7 to 2-fold higher P removal in GAC system than control. EBPR biomass grown in the presence of GAC was segregated into different size fractions for determining BNR and stratification of microbial groups. It was found that EBPR was majorly associated with the large biomass (>0.5 mm) fraction, corroborating with higher abundance of PAOs. Higher P removals of 60 to 70% with characteristic EBPR profiles were observed in 0.5 mm fraction. In contrast, P removals by 0.25 mm fraction were lower at 20 to 35% without EBPR profiles. EBPR biomass (>0.5 mm) fraction was segregated into granules and GAC-biofilms for determining the role of GAC in PAOs enrichment. P release (2.5-3.5 mg L-1 P) and P uptake (5-7 mg L-1 P) were higher in the P removal profiles exhibited by GAC-biofilms. In contrast, P release and P uptake were lower with the granules. These differences in P removal profiles resulted in distinct net P removal efficiencies of 70±5% and 50±6% for GAC-biofilms and granules, respectively. These differences in P removals were corroborated by higher abundance of PAOs in the GAC-biofilms than co-existing granules. PAO clade-level enrichment was found to be dependent on substrate wherein acetate feeding enriched PAO clade I, while acetate-propionate feeding caused enrichment of both PAO clade I and II. These results suggest that GAC addition to AGS reactors can aid in enrichment of PAOs, reduce the start-up period for EBPR, and increase P removal efficiencies.

Abstract:To treat gibberellin (GA) wastewater, a full-scale, multi-stage combined contact process was developed. This whole process employs three anaerobic reactors followed by micro-aerobic, anoxic/aeration and biological oxidation treatment. Pollutant removal results showed that the combined process could remove more than 98% of the chemical oxygen demand (COD), NH3-N, and SO42- pollutants because of the different microbial communities in each reactor. 16S rRNA gene sequencing was used to examine the microbial communities in the internal circulation (IC) and in the two up flow anaerobic sludge blanket (UASB) reactors, as well as to investigate the effect of sampling elevation on the microbial community. The results showed that Firmicutes and Euryarchaeota were the most dominant phyla at the bacterial and archaeal levels, respectively. High levels of Synergistaceae_uncultured were detected in IC and UASB1. Chloro-flexi_uncultured was the dominant genus of bacterial communities within UASB2, and Methanosaeta was the dominant genus of archaeal communities. Principal coordinates analysis (PCoA) revealed variations among the microbial communities in 9 samples, and Venn analysis showed different operational taxonomic units (OTUs) among samples collected at various elevations within the three anaerobic reactors. However, partial Mantel tests indicated no significant correlation between the microbial community structure and elevation in the three anaerobic reactors.
