Abstract:A method of high performance liquid chromatography coupled with mass spectrometry(HPLC-MS) for simultaneous determination of 23 pesticides in traditional Chinese medicine Paeoniae radix Alba was developed.P.radix was extracted with acetonitrile,and then purified using a preliminary sample treatment step based on primary-secondary amine(PSA) dispersive solid-phase extraction.Limits of quantification(LOQ) were between 0.75-12.17 μg/kg.Recovery between 70% and 120% were obtained for all pesticides at 0.01 and 0.1 mg/kg levels,except for polar pesticides such as thiamethoxam and imidacloprid.This method is accurate,reproducible and has good cleanup effect.It could be applied in pesticide residue analysis in P.Radix.
Determination of Paeonol Contents in Cortex Moutan by High Performance Liquid Chromatography (HPLC) from Different Habitats and Different Years

Abstract:Objective To establish a HPLC method for determination of paeonol in Cortex Moutan from different habitats and different years.Methods The column methods were Diamonsil C18(4.6 mm×200.0 mm,5.0 μm).The mobile phase was a combination of me-thanol,aceticacid and water in the volume ratio of 54:2:44 at the flow rate of 1 ml/min.The elvation was monitored with UV absorption at 274 nm.The system worked at room temperater.Results The lineal range of Paeonol were 5.1-102.0 μg/ml,r= 0.9996,the mean recovery was 99.4%,and RSD was 0.94%.There was significant difference in contents of paeonol in Cortex Moutan from different habitats and different years(P0.01).Conclusion This method is accurate,simple and reliable and can be used to control quality of Cortex Moutan.

Abstract:OBJECTIVE:To establish a method for the simultaneous determination of 15 pesticides residues in Radix Paeoniae Alba by large volume injection-gas chromatography-mass spectrometry(LVI-GC-MS).METHODS:The pesticides, including organochlorine pesticides, organophosphorus pesticides and pyrethroid insecticides, were analyzed by LVI-GC-MS using DB-5MS capillary column (30 m X 250 μm, 0.25 μm). The column temperature programming: initial temperature 40 degree for 1 min, with the increasing rate of 20 degree/min to 210 degree for 2 min, then with the increasing rate of 5 degree/min to 280 degree for 22 min. The flow of carrier gas was 1.0 ml/min with the injection volume of 15 μl.RESULTS:The calibration curves of the pesticides were linear in the specified concentration ranges with correlation coefficients of 0.9937-0.9995. The average recoveries of the pesticides in Radix Paeoniae Alba spiked at two spiked levels ranged from 60.4 % to 106.8 % (for pendimethalin and 4, 4'-DDE those were 53.1 % and 45.2 %) with relative standard deviation(RSD) of 3.6 % to 18.6 % and the detection limits (S/N=3) were in the range of 0.16 μg/kg to 3.59 μg/kg.CONCLUSION:The established method for determination of multi-pesticide residue in Radix Paeoniae Alba is rapid, convenient and accurate with high sensitivity and low-cost.
[Study on chemical constituents of bark of Paeonia suffruticosa]

Abstract:Objective: To study the chemical constituents of the bark of Paeonia suffruticosa. Method: The 95% ethanol extract was re-extracted with EtOAc, and then separated and purified by column chromatography using silica gel, Sephadex LH -20 and RP-18 as packing materials. The structures were identified on the basis of spectral analysis and physico-chemical characters. Result: Six compounds were isolated and identified as (+)-catechin (1), paeonidanin (2), paeoniflorigenone (3), 2, 5-dihydroxy-4-methoxyacetophenone (4), paeonol (5), gallic acid (6). Conclusion: The compound 1 was isolsted from the genus Paeonia for the first time. The compounds 2, 3 were isolated from this plant for the first time.

Abstract:AIM:This paper described a HPLC method.Paeoniflorin and puerarin in Rg soft-exact(Radix Puerariae lobatae,Redix Paeoniae alba,etc.) were simultaneously determined.METHODS:A simplified HPLC procedure was developed.A 4.6 mm×250 mm Alltima C_(18) column with a mobile phase consisting of NaH_2PO_4(20 Mm,pH 2.4)-Acetonitrile:Tetrahydrofuran(900:120:5) was used.The flow rate was 1.00 mL/min.It was detected at 320 nm.RESULTS:Paeoniflorin and puerarin were well separated.The average recoveries of paeoniflorin and puerarin were 98.10% and 101.2% respectively.CONCLUSION:The result shows that this method is convenient,rapid and accurate.

Abstract:A variety of bioactive substances may account for the recognized efficacy and wide clinical application of Psoraleae Fructus in China. A high-performance liquid chromatography-diode array detector (HPLC-DAD) fingerprint method was developed to present the comprehensive phytochemical profile of the crude drug. Thirteen major compounds were separated and identified by HPLC coupled with time-of-flight mass spectrometry (HPLC/TOF-MS), namely psoralenoside (PO), isopsoralenoside (IPO), psoralen (PS), isopsoralen (IPS), neobavaisoflavone (NBF), bavachin (BC), corylin (CN), bavachromene (BCM), psoralidin (PD), isobavachalcone (IBC), bacachinin (BCN), corylifol A (CA) and bakuchiol (BK). Then quantitative analysis of multiple components by single marker (QAMS) was applied in content determination of PO, IPO, PS, IPS, BC, IBC, BCN, CA and BK, with NBF as the internal standard. The calculation results indicated no significant difference from the traditional external standard method (p > 0.05, RSD < 2.62%), suggesting that QAMS is a reliable and convenient method for content determination of multiple chemical compositions, especially when there is a shortage of reference substances. In conclusion, simultaneous qualitative and quantitative analysis of Psoraleae Fructus may be fulfilled through the newly proposed method of QAMS combined with HPLC-DAD/TOF-MS fingerprint.

Abstract:A ultraperformance liquid chromatography (UPLC)-photodiode array (PDA) detection method was established for the simultaneous determination of seven constituents in Fructus aurantii-Magnolia bark decoction: naringin, hesperidin, neohesperidin, narirutin, meranzin hydrate, honokiol and magnolol. These were separated in <17 min using a C18 column with gradient elution using (a) acetonitrile, (b) water and (c) acetic acid at a flow rate of 0.3 mL/min, and with a PDA detector. All calibration curves showed good linear regression (r(2) > 0.9992) within the test ranges. The method was validated for speciﬁcity, accuracy, precision and limits of detection. The proposed method was found to be suitable for the quality assessment of the formula.

Abstract:A novel reinforced cloud point extraction coupled with ionic liquid (RCPE-IL) was developed for the preconcentration and extraction of five monoterpene glycosides (Oxypaeoniflorin, Albiflorin, Paeonin, Paeoniflorin, Benzoylpaeoniflorin) in Paeonia suffruticosa flower samples prior to analysis by ultra-high-performance liquid chromatographic (UHPLC). In this method, the nonionic surfactant C13E8 (Genapol X-080) was the extractant, with the hydrophilic ionic liquid 1-octyl-3-methylimidazole brominated salt ([C8MIM] Br) as cloud point reinforced reagent. The method was accomplished at room temperature (about 20 °C) in 1 min. Important factors influencing the extraction efficiency were investigated and optimized. Under the optimum conditions, the linear range of five monoterpene glucosides was from 0.1 to 100 ng mL−1. Moreover, all correlation coefficients of the calibration curves were still higher than 0.9993 and The limit of detection (LODs) was 0.1 ng mL−1 (S/N = 3). Meanwhile, satisfactory recoveries (91.4% to 98.3% for five monoterpene glucosides) in samples were also achieved. In summary, the RCPE-IL method was successfully applied for the determination of five monoterpene glycosides in Paeonia suffruticosa flower samples.

Abstract:Objective:To develop an HPLC method for simultaneous determination of the two active ingredients,named polygalaxanthone Ⅲ and 3,6’-disinapoyl sucrose in the roots of Polygala tenuifolia.Methods:The marker compounds of polygalaxanthone Ⅲ and 3,6’-disinapoyl sucrose were separated on a Kromasil C18 column(250 mm×4.6 mm,5 μm),the mobile phase was acetonitrile-0.05% phosphoric acid aqueous solution(18:82)at a flow rate of 1.0 mL·min-1,the detection wavelength was set at 320 nm and the column temperature was 30 ℃.Results:The assay was linear in the ranges of 0.5-4.0 μg(r=0.9996)for polygalaxanthone Ⅲ and 0.5-8.0 μg(r=0.9996)for 3,6’-disinapoyl sucrose.The average sample recoveries(n=6)were 97.1%(RSD=0.87%)and 99.4%(RSD=1.5%)for these two constituents,respectively.Conclusion:This accurate and reproducible method realizes the simultaneous determination of the two components in P.tenuifolia,which supplies an effective method for the quality control of P.tenuifolia.
[Simultaneous Determination of Six Components in Water Extract and Alcohol Precipitation Liquid in Astragali Radix by HPLC-UV-ELSD].

Abstract:A HPLC-UV-ELSD method was established for simultaneous determination of six components in two intermediates of Shenqi Fuzheng injection (SFI) and the feasibility of establishing quantitative analysis of multi-components by single marker (QAMS) methods on different detectors was further explored. Calycosin-7-O-β-D-glucoside and astragloside Ⅳ were selected as internal reference substances for respectively flavonoids and saponins, and relative correlation factors (RCF) of formononetin-7-O-β-D-glucoside, 9, 10-dimethoxypterocarpan-3-O-β-D-glucopyranoside, 2'-dihydroxy-3', 4'-dimethoxyisoflavan-7-O-β-D-glucopyranoside and astragloside Ⅱ were calculated. Eventually, quantitative results of the 14 samples were compared between QAMS and external standard method. The sample concentrations calculated by QAMS were similar with concentrations calculated by external standard method, and the absolute values of relative deviations were generally less than 5% according to the UV detection of flavonoids. On the basis of ELSD detection for saponins, however, the absolute values of relative deviation of the two methods ranged from 0.48% to 23.17%. The QAMS method built on ultraviolet (UV) detectors was stable and can be used as a substitute method to reduce the consumption of standard compounds; meanwhile, the accuracy of QAMS method built on evaporative light scattering detector (ELSD) was inferior to that of external standard method, and the working principle of ELSD and feasible concentration range remain to be further studied.

Abstract:A reversed phase high performance liquid chromatography method was established for the simultaneous determination of eight major constituents, namely gallic acid, paeoniflorin sulfonate, catechin, albiflorin, paeoniflorin, benzoic acid, pentagalloylglucose and benzoylpaeoniflorin in red and white peony roots, the two commonly used traditional Chinese medicinal herbs. The optimal conditions of separation and detection were achieved on a C18 analytical column with a gradient mobile phase consisting of acetonitrile and 0.015% phosphoric acid at the flow rate of 1.0 mL min−1 and detection wavelength set at 230 nm. All calibration curves showed good linear regression (r>0.9995) within test ranges. This method provided good reproducibility with overall intra-and inter-day precision of less than 5% and 4% and good accuracy with recovery of more than 93%, respectively. The method was successfully applied to determine 71 samples of red and white peony roots collected from different areas. The results indicated that the contents of eight compounds varied significantly among the samples determined, which mainly resulted from processing procedure and habitat variation. The roots of Paeonia veitchii Lynch. contained a much higher amount of gallic acid and pentagalloylglucose than that of Paeonia lactiflora Pall., which could be used to distinguish the two similar species.

Abstract:OBJECTIVE:To develop an HPLC method to determine the contents of danshensu, hydroxysafflor yellow A, rosmarinic acid, lithospermic acid, salvianolic acid B in the water extract of mixed Salviae Miltiorrhizae Radix et Rhizoma and Carthami Flos simultaneously.METHOD:The separation were carried out at 30 degrees C on a ZORBAX Eclipse Plus C18 column (4.6 mm x 100 mm, 1.8 microm) with formic acid-500 mmol x L(-1) ammonium formate-water solution (0.5:10:90) as mobile phase A and acetonitrile-formic acid solution (100: 0.5) as mobile phase B in gradient mode at a flow rate of 0.5 mL x min(-1). Detection wavelengths were 280 nm for danshensu, rosmarinic acid, lithospermic acid, salvianolic acid B, and 380 nm for hydroxysafflor yellow A.RESULT:The 5 components were separated well with a good linearity (R2 > 0.999 3) in the range of the test concentration. The average recoveries of danshensu, hydroxysafflor yellow A, rosmarinic acid, lithospermic acid, and salvianolic acid B were 99.1%, 102%, 102%, 98.5% and 101%, respectively.CONCLUSION:This method is simple, accurate, and repeatable.
Primary safety evaluation of sulfated Paeoniae Radix Alba

Abstract:The paper is to report the development of a method of quantitative analysis of multi-components by high performance liquid chromatography (HPLC) for simultaneously determining paeoniflorin sulfonate (PS), paeoniflorin (PF) and albiflorin (AF) in sulfated Paeoniae Radix Alba. Moreover, the cytotoxicity of paeoniflorin sulfonate by MTT-assay and the acute toxicity of mice by administration of paeoniflorin sulfonate were evaluated. Chromatographic separation of paeoniflorin sulfonate, PF and AF were performed on a SHISEIDO CAPCELL PAK C18 column (250 mm x 4.6 mm, 5 microm) for HPLC and a mixture of acetonitrile and 0.02% phosphoric acid solution (15 : 85) as the mobile phase. As detector a spectrophotometer set at 230 nm; column temperature 30 degrees C; flow rate 1.0 mL x min(-1). The toxicity of paeoniflorin sulfonate was evaluated by in vitro cytotoxicity carried out on mouse and human primary hepatocytes, and by acute oral toxicity test carried out on mice. The calibration curve of paeoniflorin sulfonate, PF and AF revealed linearity in the range of 0.041 8 - 1.045 0, 0.023 5 - 0.587 5, and 0.039 8 - 0.995 0 mg x mL(-1), respectively (r > 0.999 8). The average recovery was ranged from 99.11% to 101.71%, RSD < 2%. Paeoniflorin sulfonate does not have any cytotoxicity to cells at all the tested concentrations (< or = 300 micromol x L(-1)) in the in vitro cytotoxicity assay. The maximum tolerance dose of paeoniflorin sulfonate solution and extraction of Paeoniae Radix Alba to mouse is 5 g x kg(-1) and 80 g x kg(-1) respectively. The contents of these three components in the samples were determined with the developed method. It is a rapid, convenient and accurate method to determine multi-components. The content of PF in sulfated Paeoniae Radix Alba is significantly lower, and there is negative correlationship between the content of paeoniflorin sulfonate and PF. The in vitro cytotoxicity assay and in vivo mouse acute toxicity test showed that there is no obvious toxicity of paeoniflorin sulfonate and water-soluble extract of sulfated Paeoniae Radix Alba.
Determination of Active Constitutents in Folium Eucommiae by RP-HPLC

Abstract:OBJECTIVE To establish a new reversed phase high-performance liquid chromatography(RP-HPLC) method for the determination of chlorogenic acid and genipoide,the active constitutents in folium eucommiae simultaneously.METHODS YMC-pack ODS-A column(250 mm×4.6 mm,5 μm) was used and the mobile phase was methanol-water-acetic acid(20:80:1.5) at the flow rate of 1.0 mL·min-1.The detection wavelength was 237 nm and the temperature of column was 30 ℃.RESULTS The linear range of chlorogenic acid and genipoide were 0.041 6-0.624 0 μg(r=0.999 2) and 0.081 6-1.224 0 μg(r=0.998 9).The average recoveries were 102.92% and 102.35%,respectively.The RSD were 1.49% and 1.20%.The content of chlorogenic acid and genipoide in folium eucommiae were 2.801 mg·g-1 and 0.273 mg·g-1.CONCLUSION The method is simple,accurate and reliability,with good repeatability,and can be used for the quality control of folium eucommiae.
Simultaneous Determination of Paeoniflorin and Ferulic Acid in Buyanghuanwu Decoction by RP-HPLC

Abstract:Objective To establish a high performance liquid chromatography(HPLC) for simultaneous determination of paeoniflorin and ferulic acid in buyanghuanwu decoction. Methods The condition of determination was optimized.Waters Symmetry C_(18) column(4.6 mm×150 mm,5 μm) was used at 30 ℃ and mobile phase was 1%acetic acid-methanol(70∶30)(pH 3.15)with a flow rate of 1.0 mL·min~(-1).The detection wavelength for paeoniflorin and ferulic acid were 230 nm and 320 nm,respectively. Results The linearity range of paeoniflorin and ferulic were 4-24 μg·mL~(-1) and 0.4-2.0 μg·mL~(-1),respectively,the correlation coefficient were 0.999 9 and 0.999 8,respectively,the average recoveries(n=3) were 101.72%(RSD=0.27%)and 104.72%(RSD=0.46%). Conclusion This is a simple and accurate detection method used for the quality control of that prescription.
[Simultaneous Determination of Three Active Constituents in Cortex Moutan by RP-HPLC].

Abstract:OBJECTIVE:To establish a RP-HPLC method for the simultaneous determination of gallic acid, paeoniflorin and paeonol in Cortex Moutan.METHOD:The analysis was carried out on a column of YMC-C18 (4. 6 mm x 250 mm, 5 microm), and gradient elution was used for the baseline separation of these three constituents with the different detection wavelength.RESULT:The linear ranges for gallic acid, paeoniflorin and paeonol were 0.0130-0.267, 0.0880-1.76 and 0.164-3.28 microg, respectively. Their average recoveries were 104.3%, 101.7% and 99.5%, respectively.CONCLUSION:The method was successfully applied to the determination of gallic acid, paeoniflorin and paeonol in Cortex Moutan from the different drugstores. It was shown in this study that the proposed method could be used to evaluate the quality of the medicinal plants or herbal preparations containing gallic acid, paeoniflorin and paeonol.

Abstract:AIM To establish an HPLC quantitative method for simultaneously determining mangiferin,berberine hydrochloride and paeonol in Zhibo Bawei Oral Solution(Anemarrhenae Rhizoma,Phellodendri chinensis Cortex,Moutan Cortex,Rehmanniae Radix,etc.).METHODS A column Diamonsil C18(250 mm×4.6 mm,5 μm) was selected with the mobile phase consisted of acetonitrile-33 mmol/L potassium dihydrogen phosphate solution in gradient elution mode.The flow rate was 0.9 mL/min.The detection wavelength was 317 nm.The column temperature was set at 50 ℃.RESULTS Mangiferin,berberine hydrochloride and paeonol were linear within the ranges of 1.288-12.88 μg/mL(r=0.999 9),2.666~26.655 μg/mL(r=0.999 9) and 1.466-14.655 μg/mL(r=0.999 8),respectively.The average recoveries were 100.6%,99.6% and 101.2%,respectively.CONCLUSION The method is simple,reproducible,and can be used for the quality control of Zhibo Bawei Oral Solution.

Abstract:Objective An HPLC method was developed for the determination of albiflorin, paeoniflorin, and benzoylpaeoniflorin in Total Glucoside of Paeony Capsule (TGPC). Methods HPLC analysis was performed on a Zorbax SB-C_ 18 column (250 mm×4.6 mm, 5 μm) with mixture of acetonitrile (A) and 0.015% phosphoric acid solution as mobile phase in gradient mode. The concentrations of solvent A were 8%, 12%, 20%, 20%, 45%, and 45% at 0, 5, 20, 25, 35, and 40 min, respectively; the detection wavelength was 230 nm and the column temperature was 30 ℃. Results The standard curves of albiflorin, paeoniflorin, and benzoylpaeoniflorin showed a good linearity and the average recoveries were 96.99% with RSD 0.56% for albiflorin, 103.6% with RSD 0.77% for paeoniflorin, and 104.2% with RSD 1.35% for benzoylpaeoniflorin (n=5), respectively. Conclusion The method is quick, simple, and repeatable for the determination of albiflorin, paeoniflorin, and benzoylpaeoniflorin in TGPC simultaneously.
Quantitative Determination of 3 Characteristic Constituents of Cortex Moutan by HPLC under Double UV Wavelengths

Abstract:Objective:To quantify paeonol,paeoniflorin and gallic acid for Cortex Moutan of different origins. Methods:The samples were determined by HPLC under double UV wavelengths on Ahima C_(18)column(250 mm×4.6 mm,5μm)with mobile phase 0.4% formic acid and acetonitrile.The gradient condition was 0 min→50 min→60 min with acetonitrile of 0%→40%→85% ;The flow rate was 1.0 mL·min~(-1).The detecting wavelength was 274 nm and 245 nm.Column temperature was room temperature.Results:The regression equation is linear in the range of 1.3-255.0,0.4-75.0 and 0.9-175.0μg·mL~(-1)respectively.The average recovery is(99.3±3.34)%,(102.2±1.55)%,(99.0±1.64)%;limit of detecting is 24.0,36.8 and 71.0 ng·mL~(-1).Conclu- sion:The presented method is accurate,reliable and simple.It is a more reasonable and credible quality control method for Cortex Moutan.

Abstract:Objective:To improve the quality standard of Pishen Liangzhu pills,the study of HPLC methodology was carried out to determine the paeoniflorin in the pills.Method: Using HPLC.The separation was performed on Kromasil-C18 column(4.6 mm×250 mm,5 μm),using a mixture of acetonitrile-0.05 mol·L-1 potassium dihydrogen phosphate(10∶ 90) as the mobile phase;the detection wavelength was wet at 230 nm.Result: The calibration curve was linear within the range of 0.106 2-1.062 μg(r=0.999 95).The average recovery for paeoniflorin was 96.83%.Conclusion: The method is selective,accurate and simple.It is suitable for controlling the quality of Pishen Liangzhu pills.
