Abstract:This study examined the latent structure of spontaneous social attention in 11- to 26-month-olds with autism spectrum disorder (ASD, n = 90) and typically developing (n = 79) controls. Application of the joint and individual variance explained decomposition technique revealed that attention was driven by a condition-independent tuning into the dynamic social scenes construct and context-specific constructs capturing selection of the most relevant social features for processing. Gaze behavior in ASD is characterized by a limited tuning into the social scenes and by a selection of atypical targets for processing. While the former may be due to early disruption of the reward circuitry leading to limited appreciation of the behavioral relevance of social information, the latter may represent secondary deficits reflecting limited knowledge about social partners.

Abstract:Alopecia areata is a chronic relapsing autoimmune disease characterized by a local or diffuse non-scarring hair loss. Statistically, 0.2% of people worldwide are affected by alopecia areata and 1.7% were diagnosed with it at some point in life. The incidence of alopecia areata in children has been rapidly increasing in recent years. The main role in pathogenesis of alopecia areata belongs to the loss of immune privilege due to the influence of genetics and various external factors. Follicles become vulnerable to the attack by patients own T-lymphocytes, which results in hair shedding. Some autoimmune and allergic diseases are known to be comorbidities of alopecia areata. The diagnosis is made on the basis of trichoscopy results, besides there are some manifestation features in children which are different than in adults. Currently, there are several promising drugs but the number of studies (regarding pediatric patients) is extremely limited. According to a number of authors, corticosteroids and Janus kinase inhibitors have shown their effectiveness and safety in the treatment of childhood alopecia areata, although they are not approved by the United States Food and Drug Administration (FDA) at the moment.

Abstract:Alopecia areata (AA) is a common autoimmune-mediated hair loss disorder in humans with an estimated lifetime risk of approximately 2 %. Episodes of hair loss usually begin with isolated hairless patches that may progress to complete hair loss over the entire body. A familial occurrence of AA is well established, with recurrence risks of about 6–8 % in first-degree relatives. AA is a multifactorial disorder involving both environmental and genetic risk factors. Previous research has identified 14 susceptibility loci, most of which implicate genes involved in the immune response. The following review presents a summary of the latest findings from genome-wide association, sequencing and gene expression studies of AA, as well as their contribution to the recent therapeutic developments.

Abstract:Alopecia areata is a nonscarring, inflammatory disease of hair follicles. Histological changes are limited to anagen follicles that are surrounded by mononuclear cells and inflammatory mediators. This infiltration is composed primarily of cytotoxic (CD8+) and helper (CD4+) T cells. The infiltration is markedly reduced by intralesional injection of triamcinolone acetonide with subsequent hair re-growth in the affected site. Alopecia areata is suspected to be an autoimmune disease targeting hair follicles. The disease closely resembling human is found in a large production colony of C3H/HeJ mice and DEBR rats. The traits and pathogenesis of alopecia areata in C3H/HeJ mice and DEBR rats are discussed.

Abstract:Introduction: Alopecia areata (AA) is a chronic disorder in which the immune system affects the hair follicles, nails, and occasionally, the retinal pigment epithelium. The characteristic manifestation of alopecia areata encompasses isolated, smooth, non-scarring surfaces of hair loss on the scalp or anywhere with hair growth. Objective: to detail the current information related to alopecia areata, etiology, epidemiology, pathophysiology, histopathology, manifestations, diagnosis, treatment and complications of the condition. Methodology: a total of 48 articles were analyzed in this review, including review and original articles, as well as clinical cases, of which 32 bibliographies were used because the other articles were not relevant for this study. The sources of information were PubMed, Google Scholar and Cochrane; the terms used to search for information in Spanish, Portuguese and English were: alopecia areata, hair loss, hair follicles, hair loss, Janus kinase. Results: The lifetime risk of presenting alopecia areata is approximately 2%, it is the second most frequent non-scarring alopecia, after male and female pattern alopecia. So far no study has shown whether the prevalence of alopecia areata is different between ethnic groups. The first appearance of this alteration is more common between the third and fourth decades of life, however it can occur in any age group. Recent evidence suggests that the origin of the condition is autoimmune, with a significant genetic contribution. This alteration is related to medical and psychiatric comorbidities such as depression, anxiety and different autoimmune disorders. Conclusions: Alopecia areata is an autoimmune condition that generates a sudden and uneven hair loss, affecting any area of the body. Recently, alopecia areata has been interpreted as an autoimmune disease with a genetic substrate. The hair follicles do not suffer permanent damage, which is positive because of the possibility of regrowth. It is of vital importance to know the etiology, epidemiology, pathophysiology, histopathology, manifestations, diagnosis and treatment of the condition. At the moment there is no cure for alopecia areata, however the use of some medications has contributed to the improvement of the symptomatology, there are multiple treatment options available. Abstract:Alopecia areata is a common condition causing nonscarring hair loss. It may be patchy, involve the entire scalp (alopecia totalis) or whole body (alopecia universalis). Patients may recover spontaneously but the disorder can follow a course of recurrent relapses or result in persistent hair loss. Alopecia areata can cause great psychological distress, and the most important aspect of management is counseling the patient about the unpredictable nature and course of the condition as well as the available effective treatments, with details of their side effects. Although many treatments have been shown to stimulate hair growth in alopecia areata, there are limited data on their long-term efficacy and impact on quality of life. We review the evidence for the following commonly used treatments: corticosteroids (topical, intralesional, and systemic), topical sensitizers (diphenylcyclopropenone), psoralen and ultraviolet A phototherapy (PUVA), minoxidil and dithranol.

Abstract:Alopecia areata (AA) is a common, inflammatory, nonscarring type of hair loss. Significant variations in the clinical presentation of AA have been observed, ranging from small, well-circumscribed patches of hair loss to a complete absence of body and scalp hair. Patients affected by AA encompass all age groups, sexes, and ethnicities, and may experience frustration with the unpredictable nature of their disease for which there is currently no definitive treatment. The cause of AA remains incompletely understood, though it is believed to result-at least in part-from a loss of immune privilege in the hair follicle, autoimmune-mediated hair follicle destruction, and the upregulation of inflammatory pathways. Patients with AA frequently experience marked impairment in psychological well-being, self-esteem, and may be more likely to suffer from psychiatric comorbidities. Part one of this two-part continuing medical education series describes the epidemiology, clinical evaluation, prognosis, and recent advancements in the understanding of the pathogenesis of AA.

Abstract:Alopecia areata (AA) is a complex autoimmune condition that causes nonscarring hair loss. It typically presents with sharply demarcated round patches of hair loss and may present at any age. In this article, we review the epidemiology, clinical features, pathogenesis, and new treatment options of AA, with a focus on the immunologic mechanism underlying the treatment. While traditional treatment options such as corticosteroids are moderately effective, a better understanding of the disease pathogenesis may lead to the development of new treatments that are more directed and effective against AA. Abstract:Alopecia areata (AA) is a chronic, multifactorial, polygenic, and heterogeneous disorder affecting growing hair follicles in susceptible individuals, which results in a non-scarring and reversible hair loss with a highly unpredictable course. Despite very considerable research effort, the nature of the precipitating factor(s) responsible for initiating AA in any given hair follicle remains unclear, due largely to significant gaps in our knowledge of the precise sequence of the etiopathogenic events in this dermatosis. However, disease-related changes in the immune-competence of the lower growing hair follicle, together with an active immune response (humoral and cellular) to hair follicle-associated antigens, are key associated phenomena. Confirmation of the hair follicle antigen(s) implicated in AA disease onset has remained stubbornly elusive. While it may be considered somewhat philosophical by some, it is also unclear whether immune-mediated hair loss in AA results from a) an ectopic (i.e., in an abnormal location) immune response to native (unmodified) self-antigens expressed by the healthy hair follicle, b) a normal immune response against modified self-antigens (or neoantigens), or c) a normal immune response against self-antigens (modified/non-modified) that were not previously visible to the immune system (because they were conformationally-hidden or sequestered) but become exposed and presentable in an MHC-I/-II molecule-restricted manner. While some candidate hair follicle antigen target(s) in AA are beginning to emerge, with a potential role for trichohyalin, it is not yet clear whether this represents the initial and immunodominant antigenic focus in AA or is simply one of an expanding repertoire of exposed hair follicle tissue damage-associated antigens that are secondary to the disease. Confirmation of autoantigen identity is essential for our understanding of AA etiopathogenesis, and consequently for developing a more informed therapeutic strategy. Major strides have been made in autoantigen discovery in other autoimmune conditions. In particular, some of these conditions may provide insights into how post-translational modifications (e.g., citrullination, deamidation, etc.) of hair follicle-restricted proteins may increase their antigenicity and so help drive the anti-hair follicle immune attack in AA.

Hirschsprung's disease associated with alopecia universalis congenita: a case report

Abstract:Background: Hirschsprung's disease is one of the commonest causes of intestinal obstruction in neonates because of gut motility disorder. It is characterized as a complex genetic heterogenous disorder with variable inheritance. Hirschsprung's disease occurs as an isolated phenotype in majority (70 %) of cases. In other cases it may be associated with syndromes (such as Down's syndrome, Waardenburg syndrome, congenital central hypoventilation, or cartilage-hair hypoplasia) or with a spectrum of congenital anomalies involving neurological, cardiovascular, or urological systems or with sensorineural anomalies. In our patient, Hirschsprung's disease was associated with alopecia universalis. Alopecia universalis congenita is a rare disorder of skin characterized by generalized absence of hair at or shortly after birth. The inheritance patterns range from autosomal recessive, dominant or X-linked recessive forms. The autosomal recessive form is the most common and severe type in which patients present with complete absence of hair development, affecting the entire scalp and body. Alopecia universalis congenita occurs either in isolation or as a part of congenital syndromes. Here, we report the case of a neonate who presented with Hirschsprung's disease with alopecia universalis congenita, an association which has not been reported before. Case presentation: A preterm (33 weeks' gestation) 1.4 kg Indian baby girl was born to a gravida two mother by caesarean section. At birth, clinical examination revealed total absence of scalp and body hair. On day 3, she had bilious vomiting and a barium study was suggestive of Hirschsprung's disease. An exploratory laparotomy and intestinal biopsy report revealed aganglionic muscularis propria; a skin biopsy from her scalp was suggestive of alopecia universalis. Postoperatively, she died due to multiorgan failure. Her family history revealed that her elder sibling also had alopecia universalis and esophageal atresia. This child died on day twelve. Our patient's clinical features and the biopsy reports confirmed our diagnosis of Hirschsprung's disease with alopecia universalis congenita. Conclusions: A diagnosis of Hirschsprung's disease should make treating clinicians actively investigate for any associated syndromes and anomalies. Alopecia is an unusual association with Hirschsprung's disease. Alopecia universalis congenita is the most severe form of alopecia areata. Early diagnosis and classification is essential for appropriate and timely management of such cases.
Intramuscular beta2-agonist administration enhances early regeneration and functional repair in rat skeletal muscle after myotoxic injury.

Abstract:Systemic administration of beta(2)-adrenoceptor agonists (beta(2)-agonists) can improve skeletal muscle regeneration after injury. However, therapeutic application of beta(2)-agonists for muscle injury has been limited by detrimental cardiovascular side effects. Intramuscular administration may obviate some of these side effects. To test this hypothesis, the right extensor digitorum longus (EDL) muscle from rats was injected with bupivacaine hydrochloride to cause complete muscle fiber degeneration. Five days after injury, half of the injured muscles received an intramuscular injection of formoterol (100 mug). Muscle function was assessed at 7, 10, and 14 days after injury. A single intramuscular injection of formoterol increased muscle mass and force-producing capacity at day 7 by 17 and 91%, respectively, but this effect was transient because these values were not different from control levels at day 10. A second intramuscular injection of formoterol at day 7 prolonged the increase in muscle mass and force-producing capacity. Importantly, single or multiple intramuscular injections of formoterol did not elicit cardiac hypertrophy. To characterize any potential cardiovascular effects of intramuscular formoterol administration, we instrumented a separate group of rats with indwelling radio telemeters. Following an intramuscular injection of formoterol, heart rate increased by 18%, whereas systolic and diastolic blood pressure decreased by 31 and 44%, respectively. These results indicate that intramuscular injection can enhance functional muscle recovery after injury without causing cardiac hypertrophy. Therefore, if the transient cardiovascular effects associated with intramuscular formoterol administration can be minimized, this form of treatment may have significant therapeutic potential for muscle-wasting conditions.

Abstract:The pathobiology of alopecia areata (AA), one of the most frequent autoimmune diseases and a major unsolved clinical problem, has intrigued dermatologists, hair biologists and immunologists for decades. Simultaneously, both affected patients and the physicians who take care of them are increasingly frustrated that there is still no fully satisfactory treatment. Much of this frustration results from the fact that the pathobiology of AA remains unclear, and no single AA pathogenesis concept can claim to be universally accepted. In fact, some investigators still harbour doubts whether this even is an autoimmune disease, and the relative importance of CD8(+) T cells, CD4(+) T cells and NKGD2(+) NK or NKT cells and the exact role of genetic factors in AA pathogenesis remain bones of contention. Also, is AA one disease, a spectrum of distinct disease entities or only a response pattern of normal hair follicles to immunologically mediated damage? During the past decade, substantial progress has been made in basic AA-related research, in the development of new models for translationally relevant AA research and in the identification of new therapeutic agents and targets for future AA management. This calls for a re-evaluation and public debate of currently prevalent AA pathobiology concepts. The present Controversies feature takes on this challenge, hoping to attract more skin biologists, immunologists and professional autoimmunity experts to this biologically fascinating and clinically important model disease.

Abstract:Background: Alopecia areata (AA) is a frequent, reversible kind of hair loss. It is commonly seen as patchy regions of full hair loss on the scalp and other body parts that might lead to complete loss of all body hair. The condition is characterized by localized inflammatory lesions with perifollicular T-cell infiltrates, demonstrating the importance of local cytokine generation in the formation of patchy hair loss. IL-1β is a major inhibitor of hair development in vitro. Its impact is blocked by the interleukin-1 receptor antagonist, IL-1ra. Objective: To review the epidemiology, clinical characteristics, pathogenesis, and associated genetic factors and the emerging therapeutics for AA in this review article. Greater knowledge of the disease pathophysiology may contribute to establishing novel medicines that are more targeted and effective against AA. Methodology: The databases were searched for articles published in English in 3 databases [PubMed – Google scholar science direct] and Boolean operators (AND, OR, NOT) had been used such as [Alopecia Areata AND Genetic Polymorphisms OR Overview] and in peer-reviewed articles between January 2001 and May 2022. Conclusion: Comprehensive understanding of the genetic basis of AA may mediate an efficient therapeutic approach for managing and treating this life quality affecting condition.

Abstract:Alopecia areata (AA) is a common inflammatory, non-cicatricial hair loss. At present, it is considered that its pathogenesis is an autoimmune disease specific to hair follicle organs mediated by T cells under the combined action of genetic and environmental factors. Treatment is challenging for children with severe AA who are resistant or intolerant to conventional treatment.Here, we treated a 3-year-old child with severe AA with microneedling combined with compound betamethasone. After 6 months of treatment, the patient's condition was significantly improved, and most of the primary hair loss areas had hair regeneration.

Abstract:Alopecia areata is an autoimmune form of non‐scarring hair loss. It is usually characterized by limited areas of hair loss. However, the disease may progress to complete scalp and body hair loss (alopecia totalis, alopecia universalis). In patients with alopecia areata hair loss significantly impacts the quality of life. Children and adolescents with alopecia areata often experience bullying, including physical aggression. The disease severity evaluation tools used in clinical practice are: the Severity of Alopecia Tool (SALT) score and the Alopecia Areata Scale (AAS). A SALT score equal to or greater than 20 constitutes a commonly accepted indication for systemic therapy in alopecia areata. When using the AAS, moderate to severe alopecia areata should be considered a medical indication for systemic treatment. Currently, the only two EMA‐approved medications for alopecia areata are baricitinib (JAK 1/2 inhibitor) for adults and ritlecitinib (JAK 3/TEC inhibitor) for individuals aged 12 and older. Both are EMA‐approved for patients with severe alopecia areata. Other systemic medications used off‐label in alopecia areata include glucocorticosteroids, cyclosporine, methotrexate and azathioprine. Oral minoxidil is considered an adjuvant therapy with limited data confirming its possible efficacy. This consensus statement is to outline a systemic treatment algorithm for alopecia areata, indications for systemic treatment, available therapeutic options, their efficacy and safety, as well as the duration of the therapy.

Abstract:Nest alopecia (circular baldness) ― acquired non-pubescent alopecia, which begins, as a rule, with rounded foci on the scalp. During nest alopecia, there are three stages ― active (progressive, or progressive), stationary and regressive. The study of the causes and mechanisms of the pathogenesis of nest alopecia indicates a pronounced role of neurotrophic disorders, autoimmune and genetic factors, endocrine diseases and injuries. The course of alopecia areata is often chronic, recurrent (more than one episode was found in 85% of patients), however, spontaneous remission without treatment is possible in 50% of patients. In cases where the manifestations of alopecia areata start before puberty, the risk of developing a total form of the disease is high (up to 50%). The probability of a complete cure in severe forms of alopecia is less than 10% of cases. The development of alopecia areata is often accompanied by damage to the nail plates, however, the incidence of onychodystrophies varies significantly (677%). More than ten types of changes in the nail plates have been described. According to our observations, onychodystrophies were seen in all 6 patients with alopecia areata, with thimble-type deformities in children and longitudinal lines in adults. The lack of evidence in the literature on the frequency and specificity of damage to the nail plates requires additional research. Comorbid conditions in patients (systemic lupus erythematosus, psoriasis, vitiligo, atopic dermatitis, etc.) can be trigger factors in the development of nest alopecia and complicate the diagnosis and treatment of the underlying disease and concomitant dermatoses. This determines the need for a clear distinction between therapeutic and diagnostic approaches, based on their reliable significance, which will eliminate outdated or unreasonable therapeutic procedures. The article presents two clinical observations: a combination of alopecia areata with anemia, chronic gastrointestinal diseases and celiac disease in a one-year-old child and with anemia, thyroiditis, and gastrointestinal diseases in an adult patient with post-COVID syndrome.
Time-Dependent and Organ-Specific Changes in Mitochondrial Function, Mitochondrial DNA Integrity, Oxidative Stress and Mononuclear Cell Infiltration in a Mouse Model of Burn Injury

Abstract:Severe thermal injury induces a pathophysiological response that affects most of the organs within the body; liver, heart, lung, skeletal muscle among others, with inflammation and hyper-metabolism as a hallmark of the post-burn damage. Oxidative stress has been implicated as a key component in development of inflammatory and metabolic responses induced by burn. The goal of the current study was to evaluate several critical mitochondrial functions in a mouse model of severe burn injury. Mitochondrial bioenergetics, measured by Extracellular Flux Analyzer, showed a time dependent, post-burn decrease in basal respiration and ATP-turnover but enhanced maximal respiratory capacity in mitochondria isolated from the liver and lung of animals subjected to burn injury. Moreover, we detected a tissue-specific degree of DNA damage, particularly of the mitochondrial DNA, with the most profound effect detected in lungs and hearts of mice subjected to burn injury. Increased mitochondrial biogenesis in lung tissue in response to burn injury was also observed. Burn injury also induced time dependent increases in oxidative stress (measured by amount of malondialdehyde) and neutrophil infiltration (measured by myeloperoxidase activity), particularly in lung and heart. Tissue mononuclear cell infiltration was also confirmed by immunohistochemistry. The amount of poly(ADP-ribose) polymers decreased in the liver, but increased in the heart in later time points after burn. All of these biochemical changes were also associated with histological alterations in all three organs studied. Finally, we detected a significant increase in mitochondrial DNA fragments circulating in the blood immediately post-burn. There was no evidence of systemic bacteremia, or the presence of bacterial DNA fragments at any time after burn injury. The majority of the measured parameters demonstrated a sustained elevation even at 20–40 days post injury suggesting a long-lasting effect of thermal injury on organ function. The current data show that there are marked time-dependent and tissue-specific alterations in mitochondrial function induced by thermal injury, and suggest that mitochondria-specific damage is one of the earliest responses to burn injury. Mitochondria may be potential therapeutic targets in the future experimental therapy of burns.

Abstract:Two groups of patients were studied: group with stable coronary artery disease and group without the coronary artery disease as well as glycaemia and insulinaemia were studied during the oral glucose tolerance test. The results show positive correlation between hyperinsulinemia and some classical risk factors of coronary artery disease.
Prevalence and factors associated with neonatal occipital alopecia: a retrospective study

Abstract:Background: For many years, the etiology of neonatal occipital alopecia (NOA) has been thought to be friction. It is recently clear that NOA is related to the physiological hair shedding. Objective: We sought to evaluate the prevalence and factors associated with NOA. Methods: Medical records of 240 postpartum patients who had been delivered between January 2006 and June 2007 at our institution were reviewed. Phone interviews with 193 respondents were conducted to investigate the actual conditions of NOA. Results: NOA was present in 39 babies (20.2%). Univariate analysis showed that NOA was not associated with the baby's sleeping position, but was significantly associated with maternal parturition age, the delivery method, and the gestational age (p<0.05). In multiple logistic regression analysis, the risk of NOA was higher in the group younger than 35 years at parturition (OR, 3.86; 95% CI, 1.08~13.82), in the group not undergoing a Caesarean-section delivery (2.47; 1.09~5.60), and in the group delivered after 37 weeks of gestational age (3.36; 1.22~9.26). Conclusion: The pregnancy-related factors, such as non-elderly gravida, non-Caesarean-section delivery, and enough gestational age, were associated with NOA. These findings support the recent theory that NOA is not an acquired alopecia, but a physiological condition, resulting from synchronized shedding of telogen hairs initiated in utero.
