Abstract:Chitosan/Pluronic hydrogels were prepared to develop injectable depot systems for gene therapy to enhance local transgene expression at injection sites. Water-soluble chitosan and Pluronic were separately acrylated to prepare photo-crosslinkable polymers. A mixture of acrylated polymers was mixed with plasmid DNA and temperature was elevated to 37 degrees C to physically crosslink polymers to form hydrogels. Chitosan/Pluronic hydrogels were chemically crosslinked by photo-irradiated hydrogels at 37 degrees C. Mass erosion rates and release profiles of photo-crosslinked hydrogels were determined with varying photo-irradiation periods and chitosan contents of the hydrogels. The hydrogels with short photo-irradiation times degraded fast while high chitosan content in the hydrogels accelerated degradation rates. Release rates of plasmid DNA in the hydrogel were also controlled by changing chitosan content and photo-irradiation times. Released plasmid DNA was complexed with released Pluronic or chitosan and could be dissociated by adding sodium dodecyl sulfate. Scanning electron microscopy revealed that released plasmid DNA formed nanoparticles with released Pluronic or chitosan; released chitosan formed a condensed complex with plasmid DNA compared to released Pluronic. Transfection studies employing HEK293 cells showed that released fractions from chitosan/Pluronic hydrogels showed better transfection efficiency than those from Pluronic hydrogels. This result suggested that local transfection efficiencies of plasmid DNA in hydrogels were controlled by chitosan contents in chitosan/Pluronic hydrogels.
Responsive Polypeptide-Dna Hydrogel Crosslinked by G-Quadruplex

Abstract:In the past thirty years, DNA molecule has received considerable attention as a promising building material due to its precise base-paring recognition, the programmable sequences, the predictable secondary structures, etc. Based on DNA self-assembly technique, DNA supramolecular hydrogels can be fabricated which are crosslinked networks swollen in an aqueous phase by the supramolecular interaction and also have widely applications in the biomedical field. In this paper, a new type of DNA-based supramolecular hydrogel was prepared by simply mixing two components: Polypeptide-DNA and G-quadruplex. We have successfully synthesized single-stranded DNA (ssDNA) grafted polypeptide via the copper-catalyzed click reaction. By introducing the intermolecular G-quadruplex structure, which is rich in guanine and is capable of forming a four-stranded structure in the presence of potassium, the hydrogel can be formed by DNA hybridization in a fast and in-situ manner and also obtain thermal and ionic responsive gel-sol transition, respectively. In this experiment, we firstly used polyacrylamide gel electrophoresis (PAGE), circular dichroism spectra (CD), UV-Vis spectroscopy to demonstrate the G-quadruplex structure formation, and the assembling process was efficient which are formed the parallel structure and the melting temperature (T-m) was about 43 degrees C. Then by the rheological tests, the shear-storage modulus (G') was obviously higher (up to 2.4 kPa) than the shear-loss modulus (G '') over the entire time range, and a minimum concentration of 0.5 wt% polypeptide-DNA was required to form a gel-like state, and the mechanical strength of the DNA hydrogels increased with increasing concentration of the DNA building blocks. What's more, the hydrogels can switch between the gel and the sol state when changing the temperature from 25 to 50 degrees C many times, indicating that the DNA hydrogels respond to thermal in a reversible way. At last, by using the properties of crown ether complexing potassium ion, the DNA hydrogel can obtain the ion responsiveness properties. Due to its designable responsiveness and good biocompatibility, this supramolecular hydrogel would have great potential applications in tissue engineering and 3D bioprinting field.
Release of DNA from Polyelectrolyte Multilayers Fabricated Using 'Charge-Shifting' Cationic Polymers: Tunable Temporal Control and Sequential, Multi-Agent Release.

Abstract:We report an approach to the design of multilayered polyelectrolyte thin films (or 'polyelectrolyte multilayers', PEMs) that can be used to provide tunable control over the release of plasmid DNA (or multiple different DNA constructs) from film-coated surfaces. Our approach is based upon methods for the layer-by-layer assembly of DNA-containing thin films, and exploits the properties of a new class of cationic 'charge-shifting' polymers (amine functionalized polymers that undergo gradual changes in net charge upon side chain ester hydrolysis) to provide control over the rates at which these films erode and release DNA. We synthesized two 'charge-shifting' polymers (polymers 1 and 2) containing different side chain structures by ring-opening reactions of poly(2-alkenyl azlactone)s with two different tertiary amine functionalized alcohols (3-dimethylamino-1-propanol and 2-dimethylaminoethanol, respectively). Subsequent characterization revealed large changes in the rates of side chain ester hydrolysis for these two polymers; whereas the half-life for the hydrolysis of the esters in polymer 1 was ~200 days, the half-life for polymer 2 was ~6 days. We demonstrate that these large differences in side chain hydrolysis make possible the design of PEMs that erode and promote the surface-mediated release of DNA either rapidly (e.g., over ~3 days for films fabricated using polymer 2) or slowly (e.g., over ~1 month for films fabricated using polymer 1). We demonstrate further that it is possible to design films with release profiles that are intermediate to these two extremes by fabricating films using solutions containing different mixtures of these two polymers. This approach can thus expand the usefulness of these two polymers and achieve a broader range of DNA release profiles without the need to synthesize polymers with new structures or properties. Finally, we demonstrate that polymers 1 and 2 can be used to fabricate multilayered films with hierarchical structures that promote the sequential release of two different DNA constructs with separate and distinct release profiles (e.g., the release of a first construct over a period of ~3 days, followed by the sustained release of a second for a period of ~70 days). With further development, this approach could contribute to the design of functional thin films and surface coatings that provide sophisticated control over the timing and the order of the release of two or more DNA constructs (or other agents) of interest in a range of biomedical contexts.
Sustained Release of PEG-g-chitosan Complexed DNA from Poly(lactide-Co-glycolide).

Abstract:Chitosan and its derivatives have emerged as promising gene-delivery vehicles because of their capability to form polyplexes with plasmid DNA and enhance its transport across cellular membranes through endocytosis. Evidently, polyplexes of chitosan and DNA significantly improve transfection efficiency; however, these polyplexes are not capable of sustained DNA release and, thus, prolong gene transfer. In order to achieve prolonged delivery of DNA/chitosan polyplexes, we have formulated microspheres by physically combining poly(ethylene glycol)-grafted chitosan (PEG-g-CHN) with poly(lactide-co-glycolide) (PLGA) using a modified conventional in-emulsion solvent evaporation method. Electrophoretic analysis of materials released from these microspheres suggests the presence of PEG-g-CHN complexed DNA and these microspheres are capable of sustained release of DNA/PEG-g-CHN for at least 9 weeks. The rate of DNA release can be modulated by varying the amount of PEG-g-CHN. The release products from these microspheres are bioactive and show enhanced transfection in vitro over DNA released from conventional PLGA microspheres containing no PEG-g-CHN. In vivo experiments also show that these microspheres are capable of achieving gene transfer in a rat hind limb muscle model.
Influence of cross-linker chemistry on release kinetics of PEG-co-PGA hydrogels

Abstract:An investigation of encapsulated plasmid DNA release from degradable poly(ethylene glycol)-co-poly(glycolic acid) hydrogels (PEG-co-PGA) is presented. We determined by varying the chemistry of the cross-linker group, significant variations in hydrogel degradation kinetics could be achieved to control the release profiles of plasmid DNA. We prepared three analogues of PEG-co-PGA hydrogels by a photopolymerization process and measured variation in degradation rates by monitoring mechanical properties and release of plasmid DNA. 1H 1D DOSY NMR (one-dimensional diffusion ordered nuclear magnetic resonance spectroscopy) was used to measure conversion of vinyl groups after photocross-linking. Nearly full vinyl conversion was reached after 10 min exposure under ultraviolet light. Gel electrophoresis analysis confirmed that plasmid DNA remained structurally intact after photoencapsulation and release from the gels. This approach provides an additional strategy for controlling the release of biologically active compounds from hydrogels.
Association between beta2-glycoprotein I plasma levels and the risk of myocardial infarction in older men.

Abstract:von Willebrand factor (VWF) serves as adhesive surface for platelets to adhere to the vessel wall. We have recently found that beta2-glycoprotein I is able to inhibit platelet binding to VWF, indicating a role in the pathophysiology of arterial thrombosis. In the present study, we investigated whether differences in beta2-glycoprotein I plasma levels influence the risk of myocardial infarction. We have measured beta2-glycoprotein I and VWF antigen levels in 539 men with a first myocardial infarction and in 611 control subjects. Although we did not find a profound effect of beta2-glycoprotein I plasma levels on myocardial infarction in the overall population, we found a dose-dependent protective effect of increasing beta2-glycoprotein I plasma levels on myocardial infarction in men 60 years and older. In this age group, we found an odds ratio of 0.41 (95% confidence interval, 0.22-0.74) for high beta2-glycoprotein I levels compared with low levels. High plasma levels of beta2-glycoprotein I remained protective for myocardial infarction despite high levels of VWF. To conclude, high circulating levels of beta2-glycoprotein I appeared to be associated with a reduced risk of myocardial infarction in elderly men. In vivo experiments are needed to investigate the exact contribution of beta2-glycoprotein I on the pathophysiology of myocardial infarction.
Spectrum of HTLV‐III infection in a hemophilic cohort treated with blood products from a single manufacturer

Abstract:One hundred fifty‐eight hemophilia A, B, and von Willebrand disease (VWD) patients treated with clotting factor concentrates from a single manufacturer were tested for antibody to the human T‐lymphotropic virus type III (HTLV‐III). Antibody was detected in 63% and 40% of those with severe hemophilia A and B, respectively 12% and 0% of those with mild hemophilia A and B, and two patients with recessive VWD. Forty‐two antibody‐positive and 20 antibody‐negative patients were studied for clinical and laboratory features of infection. Eleven seropositive patients had clinical signs of infection including Pneumocystis carinii pneumonia, lymphadenopathy, splenomegaly or diarrhea; however, only one patient had developed acquired immune deficiency syndrome (AIDS), and only two had significant impairment of their performance status. Thirty‐one patients remained totally asymptomatic. Eight patients had a history suggestive of acute HTLV‐III infection. Thrombocytopenia was observed in 18% of seropositive patients, lymphopenia in 60%, depressed T‐helper cells in 43%, reduced T‐helper: T‐suppressor ratios (TH: TS) in 33%, and elevated platelet‐bound immunoglobulin in 53%. The antibody‐negative group had normal T‐helper cell levels (except one patient) and TH: TS ratios, and normal platelet immunoglobulin levels. Both groups demonstrated a significant elevation of immunoglobulin levels and a high prevalence of antinuclear factor and antismooth muscle antibodies. The mean level of IgG was significantly higher in the antibody‐positive group. This study confirms the correlation between HTLV‐III infection and reduced T‐helper cells in hemophiliacs but demonstrates a low incidence of clinical symptomatology. There was evidence of polyclonal B‐cell hyperactivity in the antibody‐negative group as well as the seropositive group.
Poly(d,l-lactide-co-glycolide) Dispersions Containing Pluronics: from Particle Preparation to Temperature-Triggered Aggregation

Abstract:In this work the preparation mechanism, properties and temperature-triggered aggregation of poly(D, L-lactide- co-glycolide) (PLGA) dispersions are investigated. The dispersions were prepared by interfacial deposition in aqueous solution containing Pluronic L62 (EO(6)PO(30)EO(6)) or F127NF (EO(101)PO(56)EO(101)), where EO and PO are ethylene oxide and propylene oxide, respectively. PLGA dispersions were also prepared in the absence of added Pluronic for comparison. The PLGA particles were characterized using SEM, photon correlation spectroscopy and electrophoretic mobility measurements. It was found that the hydrodynamic diameter (d) increased with PLGA concentration used in the organic solvent phase ( C PLGA(o) ). The value for d was proportional to C(PLGA)(o) (1/3). The value for d increased upon addition of 0.04 M NaNO(3) which demonstrated the importance of electrostatic interactions during particle formation. Electrophoretic mobility measurements were conducted as a function of pH and the data used to estimate the Pluronic layer thicknesses on the PLGA particles. The layer thickness was greatest for the PLGA particles prepared in the presence of Pluronic F127NF. PLGA dispersions containing Pluronic L62 exhibited temperature-triggered aggregation in the presence of 0.15 M NaNO(3). It was found that the critical temperature for dispersion aggregation (T(crit)) was comparable to the cloud point temperature ( T(cp)) for the parent Pluronic L62 solution. Conditions were established for achieving temperature-triggered aggregation at body temperature for PLGA particle/Pluronic L62 dispersions under physiological ionic strength and pH conditions. The PLGA/Pluronic L62 mixtures studied may have potential for use as injectable biodegradable implants for controlled release applications.

Abstract:Photo-cross-linked hydrogels from thermoresponsive polymers can be used as advanced injectable biomaterials via a combination of physical interaction (in situ thermal gelation) and covalent cross-links (in situ photopolymerization). This can lead to gets with significantly enhanced mechanical properties compared to non-photo-crosslinked thermoresponsive hydrogels. Moreover, the thermally phase-separated gels have attractive advantages over non-thermoresponsive gels because thermal gelation upon injection allows easy handling and holds the shape of the gels prior to photopolymerization. In this study, water-soluble thermoresponsive copolymers containing multiple methacrylate groups were synthesized via one-step deactivation enhanced atom transfer radical polymerization (ATRP) of poly(ethylene glycol) methyl ether methacrylate (PEGMEMA, M(n) = 475), poly(propylene glycol) methacrylate (PPGMA, M(n) = 375), and ethylene glycol dimethacrylate (EGDMA) and were used to form covalent cross-linked hydrogels by photopolymerization. The cross-linking density was found to have a significant influence on the mechanical and swelling properties of the photo-cross-linked gels. Release studies using lysozyme as a model protein demonstrated a sustained release profile that varied dependent on the copolymer composition, cross-linking density, and the temperature. Mouse C2C12 myoblast cells were cultured in the presence of the copolymers at concentrations up to 1 mg/mL. It was found that the majority of the cells remained viable, as assessed by Alamar Blue, lactate dehydrogenase (LDH), and Live/Dead cell viability/cytotoxicity assays. These studies demonstrate that thermoresponsive PEGMEMA-PPGMA-EGDMA copolymers offer potential as in situ photopolyimerizable materials for tissue engineering and drug delivery applications through a combination of facile synthesis, enhanced mechanical properties, tunable cross-linking density, low cytotoxicity, and accessible functionality for further structure modifications.
Gene delivery in tissue engineering: a photopolymer platform to coencapsulate cells and plasmid DNA

Abstract:Purpose: Toward the ultimate goal of developing an engineered tissue capable of mimicking complex natural healing processes, we have designed a photopolymer platform that enables simultaneous encapsulation of cells and plasmid DNA in degradable hydrogels. Photopolymerization enables spatial and temporal control of gel formation under physiological conditions, but the presence of photoinitiator radicals poses challenges for DNA photoencapsulation. Methods: The effects of photoinitiating conditions (ultraviolet light and photoinitiator radicals) on plasmid DNA were studied. Protection methods were identified. Plasmid DNA was photoencapsulated in photocrosslinked hydrogels, and the quantity and quality of the released DNA were assessed. Plasmid DNA was simultaneously entrapped (coencapsulated) with cells in hydrogels to assess in situ transfection. Results: Experiments showed that in the absence of other species, plasmid DNA was sensitive to photoinitiator radicals, but the addition of transfection agents and/or antioxidants greatly reduced DNA damage by radicals. Encapsulated plasmid DNA was released from degradable, photocrosslinked hydrogels in active forms (supercoiled and relaxed plasmids) with an overall -60% recovery. Released DNA was capable of transfecting both plated and encapsulated cells. Encapsulated cells expressed the encoded gene of the coencapsulated plasmid as the polymer degraded. Conclusions: This photopolymerization platform allows for the creation of engineered tissues with enhanced control of cell behavior through the spatially and temporally controlled release of plasmid DNA.
Formulation and in vitro transfection efficiency of poly (D, L-lactideco-glycolide) microspheres containing plasmid DNA for gene delivery

Abstract:The stability, in vitro release, and in vitro cell transfection efficiency of plasmid DNA (pDNA) poly (D,L.-lactide-co-glycolide) (PLGA) microsphere formulations were investigated. PLGA microspheres containing free and polylysine (PLL)-complexed pDNA were prepared by a water-oil-water solvent extraction/evaporation technique. Encapsulation enhanced the retention of the supereoiled structure of pDNA as determined by gel electrophoresis. PLL complexation of pDNA prior to encapsulation increased both the stability of the supercoiled form and the encapsulation efficiency. Free pDNA was completely degraded after exposure to DNase while encapsulation protected the pDNA from enzymatic degradation. Rapid initial in vitro release of pDNA was obtained from microspheres containing free pDNA. while the release from microspheres containing PLL-complexed pDNA was sustained for more than 42 days. Bioactivity of encapsulated pDNA determined by in vitro cell transfection using Chinese hamster ovary cells (CHO) showed that the bioactivity of encapsulated pDNA was retained in both formulations but to a greater extent with PLL-complexed pDNA microspheres. These results demonstrated that PLGA microspheres could be used to formulate a controlledrelease delivery system for pDNA that can protect the pDNA from DNase degradation without loss of functional activity.

Highly Branched Poly(5-Amino-1-Pentanol-Co-1,4-Butanediol Diacrylate) for High Performance Gene Transfection

Abstract:The top-performing linear poly(β-amino ester) (LPAE), poly(5-amino-1-pentanol-co-1,4-butanediol diacrylate) (C32), has demonstrated gene transfection efficiency comparable to viral-mediated gene delivery. Herein, we report the synthesis of a series of highly branched poly(5-amino-1-pentanol-co-1,4-butanediol diacrylate) (HC32) and explore how the branching structure influences the performance of C32 in gene transfection. HC32 were synthesized by an “A2 + B3 + C2” Michal addition strategy. Gaussia luciferase (Gluciferase) and green fluorescent protein (GFP) coding plasmid DNA were used as reporter genes and the gene transfection efficiency was evaluated in human cervical cancer cell line (HeLa) and human recessive dystrophic epidermolysis bullosa keratinocyte (RDEBK) cells. We found that the optimal branching structure led to a much higher gene transfection efficiency in comparison to its linear counterpart and commercial reagents, while preserving high cell viability in both cell types. The branching strategy affected DNA binding, proton buffering capacity and degradation of polymers as well as size, zeta potential, stability, and DNA release rate of polyplexes significantly. Polymer degradation and DNA release rate played pivotal parts in achieving the high gene transfection efficiency of HC32-103 polymers, providing new insights for the development of poly(β-amino ester)s-based gene delivery vectors.
Degradable gene delivery systems based on Pluronics-modified low-molecular-weight polyethylenimine: preparation, characterization, intracellular trafficking, and cellular distribution

Abstract:Background: Cationic copolymers consisting of polycations linked to nonionic amphiphilic block polymers have been evaluated as nonviral gene delivery systems, and a large number of different polymers and copolymers of linear, branched, and dendrimeric architectures have been tested in terms of their suitability and efficacy for in vitro and in vivo transfection. However, the discovery of new potent materials still largely relies on empiric approaches rather than a rational design. The authors investigated the relationship between the polymers' structures and their biological performance, including DNA compaction, toxicity, transfection efficiency, and the effect of cellular uptake. Methods: This article reports the synthesis and characterization of a series of cationic copolymers obtained by grafting polyethyleneimine with nonionic amphiphilic surfactant polyether-Pluronic(®) consisting of hydrophilic ethylene oxide and hydrophobic propylene oxide blocks. Transgene expression, cytotoxicity, localization of plasmids, and cellular uptake of these copolymers were evaluated following in vitro transfection of HeLa cell lines with various individual components of the copolymers. Results: Pluronics can exhibit biological activity including effects on enhancing DNA cellular uptake, nuclear translocation, and gene expression. The Pluronics with a higher hydrophilic-lipophilic balance value lead to homogeneous distribution in the cytoplasm; those with a lower hydrophilic-lipophilic balance value prefer to localize in the nucleus. Conclusion: This Pluronic-polyethyleneimine system may be worth exploring as components in the cationic copolymers as the DNA or small interfering RNA/microRNA delivery system in the near future.

Abstract:A facile approach for polymer gene carriers was used to construct hyaluronic acid (HA) shielding polyplexes due to the electrostatic interaction. By adding HA to PEI/DNA complexes, the ξ-potential of ternary polyplexes was changed from positive to negative. Spherical particles with diameter about 250nm were observed. Ethidium bromide exclusion assay indicated that the electrostatic complexation was loosened after addition of HA. However, DNA disassembly did not occur. The proper reason was that the intensity of negative charges was not strong enough to release DNA from the complexes in our experiment. The stability of PEI/DNA/HA polyplexes in physiological condition was improved and the cytotoxicity was reduced. Comparing with PEI/DNA polyplexes, the uptake and transfection efficiency of HA shielding polyplexes was lower for HEK293T cells probably due to the reduced adsorptive endocytosis, whereas it was higher for HepG2 cells due to HA receptor mediated endocytosis. This facile approach to constructing HA shielding polyplexes might have great potential application in non-viral gene delivery research and tumor therapy.

Abstract:This paper investigates drug release from a novel series of mPEG-functionalised PLLA polymers whose individual components (PEG and PLLA) have regulatory FDA approval. Two processing methods were explored to understand their effect on the morphology and drug release profiles of the polymers, with and without mPEG functionalisation. In the first method the polymer and Propranolol.HCl drug powders were mixed together before injection moulding. In the second method, supercritical CO2 was used to mix the polymer and drug before injection moulding. When non-functionalised PLLA was processed through injection moulding alone, there were no signs of polymer-drug interaction, and the drug was confined to crystals on the surface. This resulted in up to 85 wt% burst release of propranolol.HCl after one day of incubation. By contrast, injection moulding of mPEG-functionalised polymers resulted in the partial dissolution of drug in the polymer matrix and a smaller burst (50 wt% drug) followed by sustained release. This initial burst release was completely eliminated from the profile of mPEG-functionalised polymers processed via supercritical CO2. The addition of mPEG facilitated the distribution of the drug into the bulk matrix of the polymer. Paired with supercritical CO2 processing, the drug release profile showed a slow, sustained release throughout the 4 months of the study.

Abstract:Whereas hydrogels created from synthetic polymers offer a high level of control over their stability and mechanical properties, their biomedical activity is typically limited. In contrast, biopolymers have evolved over billions of years to integrate a broad range of functionalities into a single design. Thus, biopolymeric hydrogels can show remarkable capabilities such as regulatory behavior, selective barrier properties, or antimicrobial effects. Still, despite their widespread use in numerous biomedical applications, achieving a meticulous control over the physical properties of macroscopic biopolymeric networks remains a challenge. Here, we present a macroscopic, DNA‐crosslinked mucin hydrogel with tunable viscoelastic properties that responds to two types of triggers: temperature alterations and DNA displacement strands. As confirmed with bulk rheology and single particle tracking, the hybridized base pairs governing the stability of the hydrogel can be opened, thus allowing for a precise control over the hydrogel stiffness and even enabling a full gel‐to‐sol transition. As those DNA‐crosslinked mucin hydrogels possess tunable mechanical properties and can be disintegrated on demand, they can not only be considered for controlled cargo release but may also serve as a role model for the development of smart biomedical materials in applications such as tissue engineering and wound healing. Abstract:Drug-loaded poly(lactic-co-glycolic) acid(PLGA) microspheres were dispersed in methylcellulose reverse thermo-sensitive hydrogel for long-term sustained release of biomacromolecular drugs.The in vitro release properties of the model drug lysozyme in the mixture formulation were examined.The influence of the hydrogel on the degradation of the PLGA was investigated using gel permeation chromatography(GPC) and a scanning electron microscope(SEM).The mechanisms for drug release from the microspheres in the hydrogel were deduced by Peppas' formula.The results show that the mixture formulation exhibits nearly zero-order release of lysozyme for over 30 days without any burst effect.The drug release is controlled by both drug diffusion and material erosion.

Abstract:A novel ultrathin enzymatically degradable multilayered film using DNA as building blocks was fabricated by the layer-by-layer (LbL) technique. The UV-vis spectrometry and AFM experiments showed that the buildup of DNA and poly-L-lysine (PLL) was a kind of "exponentially growing films". The dye adsorption experiment suggested that the DNA molecules in the multilayered films were effectively protected by PLL. The films were further cross-linked by glutaraldehyde (GA). The cross-linking density of the films was modulated through the simple controlling of the time of the GA incubation process. An in vitro enzymatic degradation was carried out to investigate the DNA release profiles. The UV-vis spectrometry and fluorescence measurements indicated that the DNA release profiles were accordingly changed with the cross-linking density of the films. The nanoscale, easily processed enzymatically biodegradable PLL/DNA film with the ability to precisely control DNA release profiles may serve as a novel DNA delivery system, which may have great potential for gene therapy applications in implantable materials and biomedical devices.

Abstract:An investigation about the robust estimation of the INS (inertial navigation system) that is mainly integrated by MEMS-based micro inertial devices (accelerometer and gyroscope) is presented in this study. The robust estimator of INS includes two parts: a regression form fuzzy system that is adopted to approximate the unknown acceleration and angular rate commands come respectively from the MEMS-based accelerometer and gyroscope devices and a robust estimator based on the above fuzzy system by robust filter design concept for eliminating the effects of device noises and modeling uncertainties is proposed. This combination of MEMS devices and robust filtering technologies will successfully offer one an opportunity to develop a more small size and high accuracy module for practical navigation applications
