Abstract:Cold acclimation (CA) and deacclimation (DA), which are often accompanied by changes in freezing tolerance (FT), carbohydrates and hormones, are crucial for winter survival, especially under global warming. Plants with weak CA and premature DA caused by warm winters and/or unseasonal warm spells can be easily injured by adverse reactions to cold. Thus, understanding the molecular mechanisms of FT is imperative. In this study, we used high-throughput RNA-seq to profile the CA and DA of leaves of overwintering Rhododendron "Miyo-no-Sakae" over time; these leaves do not undergo dormancy but do undergo photoprotection during CA, and they do not grow during DA. Using Mfuzz and weighted gene coexpression network analysis, we identified specific transcriptional characteristics in each phase of CA and DA and proposed networks involving coexpressed genes and physiological traits. In particular, we discovered that the circadian rhythm is critical for obtaining the strongest FT, and high expression of circadian rhythm-related genes might be linked to sugar accumulation during winter. Furthermore, evergreen leaves exhibited robust photoprotection during winter, as revealed by high values of nonphotochemical quenching, high expression of transcripts annotated as "early light-induced proteins", loss of granum stacks and destacking of thylakoids, all of which were alleviated during DA. The strong requirement of photoprotection could be the reason for decreased abscisic acid (ABA) and jasmonic acid (JA) contents during CA, and decreases in ABA and JA contents may contribute to decreases in lignin content. Our data suggest that the molecular mechanisms of FT in overwintering leaves are unique, which may be due to the high requirements for photoprotection during winter.
Resource Allocation in Relation to Leaf Retention Time of the Wintergreen Rhododendron Lapponicum

Abstract:Extended leaf longevity has been explained both as an adaptation to conserve nutrients and to increase seasonal or life—time carbon gain. The relative importance of these possible adaptations was investigated in the wintergreen dwarf shrub Rhododendron lapponicum by analyses of responses of growth and nutrient allocation to early and late spring defoliation and to natural reduction of leaf biomass following flowering. Removal of old leaves at the time of budbreak had no effect on biomass accumulation and nutrient pools of the new shoots 1 mo later, although the defoliation prevented the transport of resorbed nutrients from old leaves to the new shoots. This transport accounted for by far the largest part of the increase in shoot N and P in underfoliated controls. Extra nutrients were not transported from stems and roots to compensate for the lost import from old leaves. Hence, defoliated plants compensated for the losses solely by increasing nutrient uptake. In contrast, defoliation 3 wk earlier caused a 29% reduction of annual shoot growth and a proportional decline of nutrient pools in the shoots, while the nutrient concentrations, reflecting the shoot nutrient status, did not decline. Almost identical responses were observed in unmanipulated plants the year after heavy flowering. Reproductive branches do not form leaves, resulting in a decreasing photosynthetic capacity and a reduction of resource storage and allocation to next year's leaf cohort. Although early and late spring defoliation caused equal losses of mobile nutrients, only the early defoliation decreased growth, suggesting that it was the loss of carbon rather than nutrients that affected growth. This support earlier observations that the extended retention time of leaves in wintergreen and evergreen species plays a greater role in improving the carbon balance than in supplying the new leaves with nutrients.

Abstract:Desiccation tolerant plants can survive extreme water loss in their vegetative tissues. The fern Anemia caffrorum produces desiccation tolerant (DT) fronds in the dry season and desiccation sensitive (DS) fronds in the wet season, providing a unique opportunity to explore the physiological mechanisms associated with desiccation tolerance. Anemia caffrorum plants with either DT or DS fronds were acclimated in growth chambers. Photosynthesis, frond structure and anatomy, water relations and minimum conductance to water vapour were measured under well-watered conditions. Photosynthesis, hydraulics, frond pigments, antioxidants and abscisic acid contents were monitored under water deficit. A comparison between DT and DS fronds under well-watered conditions showed that the former presented higher leaf mass per area, minimum conductance, tissue elasticity and lower CO2 assimilation. Water deficit resulted in a similar induction of abscisic acid in both frond types, but DT fronds maintained higher stomatal conductance and upregulated more prominently lipophilic antioxidants. The seasonal alternation in production of DT and DS fronds in A. caffrorum represents a mechanism by which carbon gain can be maximized during the rainy season, and a greater investment in protective mechanisms occurs during the hot dry season, enabling the exploitation of episodic water availability.
Carbon allocation in early successional tree species at elevated air humidity and different soil nitrogen sources

Abstract:Global climate change scenarios predict increasing air temperature, enhanced precipitation and air humidity for Northern latitudes. We&#8239;investigated the effects of elevated air relative humidity (RH) and different inorganic nitrogen&#8239;sources&#8239;(NO3-, NH4+) on above- and belowground traits in different tree species, with particular emphasis on rhizodeposition rates. Silver birch, hybrid aspen and Scots pine saplings were grown in PERCIVAL growth chambers with stabile temperature, light intensity and two different air humidity conditions: moderate (mRH, 65% at day and 80% at night) and elevated (eRH, 80% at day and night). The collection of fine root exudates was conducted by a culture-based cuvette method and total organic carbon content was determined by Vario TOC analyser. Fine root respiration was measured with an infra-red gas analyser CIRAS 2.&#8239;&#160;We analysed species-specific biomass allocation, water and rhizodeposition fluxes, foliar and fine root traits in response to changing environmental conditions. The&#8239;eRH&#8239;significantly decreased the transpiration flux in all species. In birch the transpiration flux was also affected by the nitrogen source. The average carbon exudation rate for aspen, birch and pine varied from 2 to 3 &#8239;&#956;g&#8239;C g-1&#8239;day&#8239;-1. The exudation rates for deciduous tree species tended to increase at&#8239;eRH, while conversely decreased for coniferous trees (p=0.045), coinciding with the changes in biomass allocation.&#8239;C flux released by fine root respiration varied more than the fine root exudation, whereas the highest root respiration was found in silver birch and lowest in aspen. At eRH the above and belowground&#8239;biomass ratio in aspen increased, at the expense of decreased root biomass and root respiration.&#8239;&#160;Moreover,&#8239;eRH&#8239;significantly affected fine root morphology, whereas the response of specific root area was reverse for deciduous and coniferous tree species. However, fine roots with lower root tissue density had higher C exudation rate. Our findings underline the importance of considering species-specific differences by&#8239;elucidating tree&#8217;s&#8239;acclimation&#8239;to environmental factors and their&#8239;interactions.&#8239;&#8239;&#160;
Overwintering evergreen oaks reverse typical relationships between leaf traits in a species spectrum

Abstract:The leaf economics spectrum has given us a fundamental understanding of the species variations in leaf variables. Across plant species, tight correlations among leaf mass per area (LMA), mass-based nitrogen ( N m ) and photosynthetic rate ( A m ) and leaf lifespan have been well known as trade-offs in leaf carbon economy. However, the regional or biome-level correlations may not be necessary to correspond with the global-scale analysis. Here, we show that almost all leaf variables in overwintering evergreen oaks in Japan were relatively well included within the evergreen-broadleaved trees in worldwide temperate forests, but N m was more consistent with that in deciduous broadleaved trees. Contrary to the universal correlations, the correlation between A m and N m among the evergreen oaks was negative and the correlation between A m and LMA disappeared. The unique performance was due to specific nitrogen allocation within leaves, i.e. the evergreen oaks with later leaf maturation had lower N m but higher nitrogen allocation to photosynthetic enzymes within leaves, to enhance carbon gain against the delayed leaf maturation and the shortened photosynthetic period due to cold winters. Our data demonstrate that correlations between leaf variables in a local scale are occasionally different from averaged global-scale datasets, because of the constraints in each biome.

Fructan Structure and Metabolism in Overwintering Plants

Abstract:In northern regions, annual and perennial overwintering plants such as wheat and temperate grasses accumulate fructan in vegetative tissues as an energy source. This is necessary for the survival of wintering tissues and degrading fructan for regeneration in spring. Other types of wintering plants, including chicory and asparagus, store fructan as a reserve carbohydrate in their roots during winter for shoot- and spear-sprouting in spring. In this review, fructan metabolism in plants during winter is discussed, with a focus on the fructan-degrading enzyme, fructan exohydrolase (FEH). Plant fructan synthase genes were isolated in the 2000s, and FEH genes have been isolated since the cloning of synthase genes. There are many types of FEH in plants with complex-structured fructan, and these FEHs control various kinds of fructan metabolism in growth and survival by different physiological responses. The results of recent studies on the fructan metabolism of plants in winter have shown that changes in fructan contents in wintering plants that are involved in freezing tolerance and snow mold resistance might be largely controlled by regulation of the expressions of genes for fructan synthesis, whereas fructan degradation by FEHs is related to constant energy consumption for survival during winter and rapid sugar supply for regeneration or sprouting of tissues in spring.
Revisiting the early light-induced protein hypothesis in the sustained thermal dissipation mechanism in yew leaves

Abstract:Overwintering evergreen trees in boreal regions continuously convert absorbed light energy into heat through a process known as 'sustained thermal dissipation'. To better understand this mechanism, this study examined the alterations in the photosynthetic apparatus and transcriptomes of yew (Taxus cuspidata) leaves throughout the year, comparing sun-exposed and shaded leaves. The Y(II) parameter, conventionally used to estimate the quantum yield of photosystem II (PSII), suggests the occurrence of temperature-dependent thermal dissipation during winter. On the other hand, the levels of photosystem subunits, including the D1 subunit of the PSII reaction center, remain relatively stable year-round, suggesting that typical photoinhibition is unlikely to occur. Time-resolved chlorophyll fluorescence analysis revealed that heat dissipation at the PSII antenna is prominent in winter. Winter transcriptomes are notably characterized by a predominance of Elip transcripts encoding early light-induced protein (ELIP), which constitute 20% of the total transcripts, as deduced from RNA-seq analysis. Furthermore, ELIP protein concentration increases to nearly half that of the major light-harvesting complexes. The predicted structure of ELIP includes potential chlorophyll a and carotenoid binding sites. Considering a previous report showing ELIP's capacity for energy dissipation, these findings lead to a reevaluation of its significant role in sustained thermal dissipation.
Photosynthetic Traits of Quercus coccifera Green Fruits: A Comparison with Corresponding Leaves during Mediterranean Summer

Abstract:Fruit photosynthesis occurs in an internal microenvironment seldom encountered by a leaf (hypoxic and extremely CO2-enriched) due to its metabolic and anatomical features. In this study, the anatomical and photosynthetic traits of fully exposed green fruits of Quercus coccifera L. were assessed during the period of fruit production (summer) and compared to their leaf counterparts. Our results indicate that leaf photosynthesis, transpiration and stomatal conductance drastically reduced during the summer drought, while they recovered significantly after the autumnal rainfalls. In acorns, gas exchange with the surrounding atmosphere is hindered by the complete absence of stomata; hence, credible CO2 uptake measurements could not be applied in the field. The linear electron transport rates (ETRs) in ambient air were similar in intact leaves and pericarps (i.e., when the physiological internal atmosphere of each tissue is maintained), while the leaf NPQ was significantly higher, indicating enhanced needs for harmless energy dissipation. The ETR measurements performed on leaf and pericarp discs at different CO2/O2 partial pressures in the supplied air mixture revealed that pericarps displayed significantly lower values at ambient gas levels, yet they increased by ~45% under high CO2/O2 ratios (i.e., at gas concentrations simulating the fruit's interior). Concomitantly, NPQ declined gradually in both tissues as the CO2/O2 ratio increased, yet the decrease was more pronounced in pericarps. Furthermore, net CO2 assimilation rates for both leaf and pericarp segments were low in ambient air and increased almost equally at high CO2, while pericarps exhibited significantly higher respiration. It is suggested that during summer, when leaves suffer from photoinhibition, acorns could contribute to the overall carbon balance, through the re-assimilation of respiratory CO2, thereby reducing the reproductive cost.

Abstract:Under drought stress, temperate forest understorey forbs prioritize biomass and leaf area retention, while graminoids enhance water‐use efficiency and photoprotection for survival. Drought stress can profoundly affect plant growth and physiological vitality, yet there is a notable scarcity of controlled drought experiments focused on herbaceous species of the forest understorey. In this study, we collected seeds from five forb and four graminoid species common in European temperate forests. Seeds were germinated under controlled glasshouse conditions and subjected to moderate drought stress for 5 weeks. We assessed biomass partitioning, stomatal and leaf morphology, leaf gas exchange, minimum leaf conductance (gmin), and chlorophyll fluorescence parameters. Comparison of the two ecological guilds revealed that graminoids had a higher R/S, improved WUE, greater carboxylation efficiency, and enhanced non‐photochemical quenching under drought conditions compared to forbs. In contrast, forbs had significantly lower gmin, with higher total biomass and total leaf area. Despite these differences in morpho‐physiological functional traits, both groups experienced a similar relative reduction in biomass after drought stress. Key predictors of biomass accumulation under drought included photochemical quenching, stomatal traits, total leaf area and gmin. A negative correlation between biomass and gmin suggests that plants with lower residual water loss after stomatal closure can accumulate more biomass under drought stress. Additionally, gmin was positively correlated with guard cell length, suggesting that larger stomata contribute to higher residual water loss. Contrasting strategies in morpho‐physiological responses to drought define the differences between the two groups. In graminoids, drought resistance suggests greater emphasis on stress tolerance as a survival strategy. In contrast, forbs were able to maintain higher biomass and total leaf area, indicating a competitive strategy for maximizing resource acquisition.

Abstract:During prolonged dry periods, non-rainfall water (NRW) plays a vital role as water input into temperate grasslands, affecting the leaf surface water balance and plant water status. Previous chamber and laboratory experiments investigated air–leaf water exchange during dew deposition, but overlooked the importance of radiative cooling on air–leaf water exchange because the chamber is a heat trap, preventing radiative cooling. To complement these previous studies, we conducted a field study, in which we investigated the effect of radiatively-induced NRW inputs on leaf water isotope signals and air–leaf water exchange in a temperate grassland during the dry-hot summers of 2018 and 2019. We carried out field measurements of the isotope composition of atmospheric water vapor, NRW droplets on foliage, leaf water, xylem water of root crown, and soil water, combined with meteorological and plant physiological measurements. We combined radiation measurements with thermal imaging to estimate leaf temperatures using different methods, and computed the corresponding leaf conductance and air–leaf water exchange. Our results indicate that radiative cooling and leaf wetting induced a switch of direction in the net water vapor exchange from leaf-to-air to air-to-leaf. The leaf conductance and air–leaf water exchange varied by species due to the species-specific biophysical controls. Our results highlight the ecological relevance of radiative cooling and leaf wetting in natural temperate grasslands, a process which is expected to influence land surface water budgets and may impact plant survival in many regions in a drier climate.

Phenology explains different storage remobilization in two congeneric temperate tree species with contrasting leaf habit

Abstract:The dependence of trees on carbon and nutrient storage is critical to predicting the forest vulnerability under climate change, but whether evergreen and deciduous species differ in their use and allocation of stored resources during spring phenology is unclear. Using a high temporal resolution, we evaluated the role of spring phenology and shoot growth as determinants of the carbon and nutrient storage dynamics in contrasting leaf habits. We recorded the phenology and shoot elongation and determined the concentrations of total non-structural carbohydrates (NSCs), starch, soluble carbohydrates, nitrogen (N) and phosphorus (P) in buds, expanding shoots and previously formed shoots of two sympatric Nothofagus species with contrasting leaf habit. Species reached similar shoot lengths, though shoot expansion started 35 days earlier and lasted c. 40 days more in the deciduous species. Thus, although the deciduous species had a relatively constant shoot growth rate, the evergreen species experienced a conspicuous growth peak for c. 20 days. In the evergreen species, the greatest decreases in NSC concentrations of previously formed shoots and leaves coincided with the maximum shoot expansion rate and fruit filling, with minimums of 63 and 65% relative to values at bud dormancy, respectively. In contrast, minimum NSC concentrations of the previously formed shoots of the deciduous species were only 73% and occurred prior to the initiation of shoot expansion. Bud N and P concentrations increased during budbreak, whereas previously formed shoots generally did not decrease their nutrient concentrations. Late spring phenology and overlapping of phenophases contributed to the greater dependence on storage of proximal tissues in the studied evergreen compared with deciduous species, suggesting that phenology is a key determinant of the contrasting patterns of storage use in evergreen and deciduous species.

Effects of Light and Soil Water Availability on Leaf Photosynthesis and Growth of Arisaema Heterophyllum , a Riparian Forest Understorey Plant

Abstract:The effects of soil-water availability on leaf light acclimation and whole-plant carbon gain were examined in Arisaema heterophyllum Blume, a riparian deciduous forest understorey plant. Photosynthesis, above-ground morphology and ramet biomass accumulation (relative growth rate: RGR of a corm for a full leaf life-span) were measured on plants raised under three light treatments combined with two soil water conditions. The two higher light treatments during growth (high: max. 550 µmol photons m–2 s–1; medium: 150 µmol photons m–2 s–1) resulted in a twofold increase in RGRs, 30% higher photosynthetic capacities and 20% less photosynthetic low-light use efficiency than those under a low light condition (50 µmol photons m–2 s–1). Leaf area was the smallest and leaf mass area ratio was the largest under the high light treatment. Water stress decreased both photosynthetic rate and leaf area and, hence, RGR in all the light regimes. However, water stress did not alter the general patterns of physiological and morphological responses to different light regimes. We estimated that higher photosynthetic low-light use efficiency and larger leaf area in the low light leaf would lead to a threefold carbon gain as compared with the high light leaf under simulated low light conditions. Both experimental and simulation results suggest that the physiological and morphological acclimations tend to be beneficial to carbon gain when light availability is low, whereas they favor increased water use efficiency when light availability is sufficiently high.

Abstract:Green stem photosynthesis has been shown to be relatively inefficient but can occasionally contribute significantly to the carbon budget of desert plants. Although the possession of green photosynthetic stems is a common trait, little is known about their photosynthetic characteristics in non‐desert species. Dianthus caryophyllus is a semi‐woody species with prominent green stems, which show similar photosynthetic anatomy with leaves. In the present study, we used a combination of gas exchange and chlorophyll fluorescence measurements, some of which were taken under varying O2 and CO2 partial pressures, to investigate whether the apparent anatomical similarities between the species' leaves and stems translate into similar photosynthetic physiology and capacity for CO2 assimilation. Both organs displayed high photosynthetic electron transport rates (ETR) and similar values of steady‐state non‐photochemical quenching (NPQ), albeit leaves could attain them faster. The analysis of OJIP transients showed that the quantum efficiencies and energy fluxes along the photosynthetic electron transport chain are largely similar between leaves and stems. Stems displayed higher total conductance to CO2 diffusion, similar biochemical properties, significantly higher photosynthetic rates and lower water use efficiency than leaves. Leaf ETR was more sensitive to sub‐ambient O2 and super‐ambient CO2 partial pressures, while leaves also displayed a higher relative rate of Rubisco oxygenation. We conclude that the highly responsive NPQ and the enhanced photorespiration and WUE in leaves represent photoprotective and water‐conserving adaptations to the high incident light intensities they experience naturally, at the expense of higher CO2 assimilation rates, which the vertically orientated stems can readily attain.

Abstract:A challenge for the designers of decision support systems (DSSs) is that the introduction of a decision aid into a complex domain often generates new, unexpected user needs. In this paper, we discuss how the iterative application of an experience-centered design methodology called DAISY (Design Aid for Intelligent Support SYstems) provides concrete methods for modeling these changes and identifying new system requirements caused by the introduction of the DSS. We illustrate the iterative use of DAISY in the design, evaluation and modification of Fox, a DSS intended to assist expert military users by helping them to generate and evaluate a broad range of plan options.
Comparison of morphological and physiological response to drought stress among temperate forest understory forbs and graminoids

Abstract:Drought stress can profoundly affect plant growth and physiological vitality, yet there is a notable scarcity of controlled drought experiments focused on herbaceous species of the forest understory. In this study, we collected seeds from five forb and four graminoid species growing in the temperate forest understory of the Czech Republic. These seeds were germinated under controlled glasshouse conditions and subjected to moderate drought stress for five weeks. We assessed biomass partitioning, stomatal and leaf morphology, leaf gas exchange, minimum leaf conductance (gmin), and chlorophyll fluorescence parameters. The comparison of two ecological guilds revealed that graminoids exhibited a higher root-to-shoot ratio, improved water-use efficiency, greater carboxylation efficiency, and enhanced non-photochemical quenching under drought conditions compared to forbs. In contrast, forbs had significantly lower gmin, along with higher total biomass and total leaf area. Despite these differences in morpho-physiological functional traits, both groups experienced a similar relative reduction in biomass during drought stress. Key predictors of biomass accumulation under drought included photochemical quenching, stomatal traits, total leaf area and gmin. A negative correlation between biomass and gmin suggests that plants with lower residual water losses after stomatal closure can accumulate more biomass under drought stress. Additionally, gmin was positively correlated with guard cell length, suggesting that larger stomata contribute to higher residual water loss. Graminoids exhibited morpho-physiological modifications that enhanced drought resistance, indicating a greater emphasis on stress tolerance as a survival strategy. In contrast, forbs maintained higher biomass and total leaf area, reflecting a competitive strategy for maximizing resource acquisition.

Fine Root Growth of Black Spruce Trees and Understory Plants in a Permafrost Forest Along a North-Facing Slope in Interior Alaska

Abstract:Permafrost forests play an important role in the global carbon budget due to the huge amounts of carbon stored below ground in these ecosystems. Although fine roots are considered to be a major pathway of belowground carbon flux, separate contributions of overstory trees and understory shrubs to fine root dynamics in these forests have not been specifically characterized in relation to permafrost conditions, such as active layer thickness. In this study, we investigated fine root growth and morphology of trees and understory shrubs using ingrowth cores with two types of moss substrates (feather- and Sphagnum mosses) in permafrost black spruce ( Picea mariana ) stands along a north-facing slope in Interior Alaska, where active layer thickness varied substantially. Aboveground biomass, litterfall production rate, and fine root mass were also examined. Results showed that aboveground biomass, fine root mass, and fine root growth of black spruce trees tended to decrease downslope, whereas those of understory Ericaceae shrubs increased. Belowground allocation (e.g., ratio of fine root growth/leaf litter production) increased downslope in both of black spruce and understory plants. These results suggested that, at a lower slope, belowground resource availability was lower than at upper slope, but higher light availability under open canopy seemed to benefit the growth of the understory shrubs. On the other hand, understory shrubs were more responsive to the moss substrates than black spruce, in which Sphagnum moss substrates increased fine root growth of the shrubs as compared with feather moss substrates, whereas the effect was unclear for black spruce. This is probably due to higher moisture contents in Sphagnum moss substrates, which benefited the growth of small diameter (high specific root length) fine roots of understory shrubs. Hence, the contribution of understory shrubs to fine root growth was greater at lower slope than at upper slope, or in Sphagnum than in feather-moss substrates in our study site. Taken together, our data show that fine roots of Ericaceae shrubs are a key component in belowground carbon flux at permafrost black spruce forests with shallow active layer and/or with Sphagnum dominated forest floor.

Abstract:Increased risk of soil waterlogging in winter and spring at northern latitudes will potentially affect forest production in the future. We studied gas exchange, chlorophyll content index, chlorophyll fluorescence, nutrient concentration and biomass accumulation in 1-year-old silver (Betula pendula Roth) and pubescent birch (Betula pubescens Ehrh.) seedlings. We hypothesized that B. pubescens has different physiological mechanisms that make it tolerate waterlogging better than B. pendula. The treatments were: (i) no waterlogging throughout the experiment; (ii) 4-week waterlogging during dormancy (dormancy waterlogging 'DW'); (iii) 4-week waterlogging during the early growing season (growth waterlogging 'GW'); and (iv) 4-week DW followed by 4-week GW during the early growing season ('DWGW'). Stomatal conductance and light-saturated net assimilation rate were reduced by GW in both species, and in B. pubescens also by DW. However, recovery was seen during the follow-up growing season. In B. pendula, DW, GW and DWGW temporarily resulted in reduced stem biomass, and GW and DWGW caused reduced leaf biomass. In B. pubescens, the stem biomass was decreased in GW and DWGW. Leaf nitrogen (N) and phosphorus (P) concentrations were generally low, and increased by GW, while potassium, calcium, magnesium and to some extent, boron and zinc concentrations decreased in both species and additionally manganese in B. pendula. The increases in N and P are mostly due to a concentration effect due to smaller leaf biomass, yet suggest that their uptake was not impaired. The decreases in cation concentrations are likely to be connected to impaired root functioning, which was not yet fully recovered from GW. We conclude that morphological acclimation to waterlogging of the leaves and roots rather than photosynthesis explains why B. pubescens is able to grow better in wetter areas than B. pendula.
