Abstract:Apoptotic death pathways are frequently activated by death ligand induction and subsequent activation of the membrane proximal signaling module. Death receptors cluster upon binding to death ligands, leading to formation of a membrane proximal death-inducing-signaling-complex (DISC). In this membrane proximal signalosome, initiator caspases (caspase 8) are processed resulting in activation of both type 1 and type 2 pathways of apoptosis signaling. How the type 1/type 2 choice is made is an important question in the systems biology of apoptosis signaling. In this study, we utilize a Monte Carlo based in silico approach to elucidate the role of membrane proximal signaling module in the type 1/type 2 choice of apoptosis signaling. Our results provide crucial mechanistic insights into the formation of DISC signalosome and caspase 8 activation. Increased concentration of death ligands was shown to correlate with increased type 1 activation. We also study the caspase 6 mediated system level feedback activation of apoptosis signaling and its role in the type 1/type 2 choice. Our results clarify the basis of cell-to-cell stochastic variability in apoptosis activation and ramifications of this issue is further discussed in the context of therapies for cancer and neurodegenerative disorders.

What problem does this paper attempt to address?
This paper aims to explore the roles of death ligand concentration and membrane - proximal signaling modules in the type 1/type 2 selection mechanism in apoptotic signaling. Specifically, the researchers studied the following aspects through the Monte Carlo simulation method: 1. **Influence of death ligand concentration**: The paper examined how different concentrations of death ligands affect the selection of type 1 and type 2 pathways during the apoptotic process. The study shows that as the death ligand concentration increases, the proportion of type 1 activation also increases, which may trigger a transition from type 2 to type 1. 2. **Status of membrane - proximal signaling modules**: The paper also explored the influence of the expression levels of various molecules (such as death receptors, adaptor proteins, procaspase 8, and anti - apoptotic cFLIP) in the membrane - proximal signaling modules on the type 1/type 2 selection. The study found that the expression levels of these molecules are key factors in determining the type 1/type 2 selection. 3. **Caspase 6 - mediated system - level feedback mechanism**: The paper further analyzed the role of caspase 6 in apoptotic signaling, especially its mechanism of affecting the type 1/type 2 selection by feedback - activating caspase 8. 4. **Stochastic variability among single cells**: The study also discussed the stochastic variability among single cells during apoptotic activation. This variability is of great significance for understanding the regulatory mechanisms of apoptosis and its application in the treatment of cancer and neurodegenerative diseases. In summary, this paper attempts to reveal the specific mechanisms of type 1 and type 2 pathway selection in the death ligand - induced apoptotic process and the potential application value of these mechanisms in disease treatment through the combination of theoretical simulation and experimental data.
